Determination of volatile fatty acids in plasma after ethanolic extraction   总被引:5,自引:1,他引:5  
1. A new rapid micro-method for measuring plasma volatile fatty acids is described. The volatile fatty acids are extracted from plasma with ethanol in the presence of a known quantity of internal standard (sodium isobutyrate). After evaporation of the ethanolic solution of the sodium salts, the residue is dissolved in a dilute solution of orthophosphoric acid to permit analysis by g.l.c. 2. A technique of g.l.c. analysis is described which permits the separation of all the volatile fatty acids from the other plasma constituents at temperatures below 100 degrees C in 5 min. 3. Steam-distillation techniques are unsatisfactory when the acetic acid concentrations in the plasma are below 0.2mm. Heating of a number of plasma constituents in acid conditions gives rise to acetic acid. 4. The binding of volatile fatty acids to plasma proteins was studied; this binding is negligible for acetic acid, but increases with the length of the fatty acid carbon chain. 5. The limits of use of the method and the physiological implications are discussed.  相似文献

It is known that volatile fatty acids can inhibit growth of species of the family Enterobacteriaceae in vitro. However, whether these volatile fatty acids affect bacterial populations in the ceca of chickens is unknown. Therefore, a study was conducted to investigate if changes in volatile fatty acids in ceca of broiler chickens during growth affect bacterial populations. Results showed that members of the Enterobacteriaceae and enterococci are present in large numbers in 3-day-old broilers and start to decrease when broilers grow older. Lactobacilli are present in large numbers as well in 3-day-old broilers, but they remain stable during the growth of broilers. Acetate, butyrate, and propionate increase from undetectable levels in 1-day-old broilers to high concentrations in 15-day-old broilers, after which they stabilize. Significant negative correlations could be calculated between numbers of Enterobacteriaceae and concentrations of undissociated acetate, propionate, and butyrate. Furthermore, pure cultures of Enterobacteriaceae isolated from the ceca were grown in the presence of volatile fatty acids. Growth rates and maximal optical density decreased when these strains grew in the presence of increasing volatile fatty acid concentrations. It is concluded that volatile fatty acids are responsible for the reduction in numbers of Enterobacteriaceae in the ceca of broiler chickens during growth.  相似文献

The effects of concentrations of volatile fatty acids on an anaerobic, glucose-limited, and pH-controlled growing culture of Salmonella enterica serovar Enteritidis were studied. Suddenly increasing volatile fatty acids to the concentrations representative of the ceca of 15-day-old broiler chickens caused washout of serovar Enteritidis. In contrast, a sudden increase to the volatile fatty acid concentrations representative of the ceca of younger broiler chickens caused a reduction in the biomass but not washout. Gradually increasing volatile fatty acids caused a gradual decrease in the biomass of serovar Enteritidis. We conclude that the concentrations of volatile fatty acids present in the ceca of broilers with a mature microflora can cause washout of serovar Enteritidis in an in vitro system mimicking cecal ecophysiology.  相似文献

Growth curves were plotted for Shigella flexneri and Klebsiella (Aerobacter aerogenes) multiplying in pure and mixed culture. In mixed culture, Klebsiella inhibited Shigella. Exponential growth was interrupted and Shigella entered into a logarithmic death phase. An analysis of cultures at the time inhibition occurred revealed that formic and acetic acids produced by Klebsiella were responsible for the inhibition of Shigella. Klebsiella strongly reduced the culture medium. The volatile fatty acids, operating under reduced conditions, exerted a bactericidal effect on Shigella. Results are discussed with reference to the possible role of volatile fatty acids as factors responsible for Shigella inhibition in vivo.  相似文献

The use of the quantitative content of volatile fatty acids as marker for the indication of the causative agents of pyo-inflammatory diseases in material taken from wounds is proposed. Thus, in 71% of cases the detection of acetic acid in a purulent wound coincided with the bacteriological isolation of monocultures, as well as associations, of facultative anaerobic bacteria. The analysis of isobutyric, butyric, isovaleric, valeric and acetic acids permitted the rapid detection of associations of obligate and facultative anaerobic bacteria in an individual patient. Changes in the quantitative content of volatile fatty acids in pathological material integrally reflected the dynamics of the causative agents of purulent infections and the effectiveness of the prescribed therapy.  相似文献

A diffusion chamber is described which concentrates short-chain, volatile fatty acids from seawater while simultaneously separating them from interfering salts. The procedure relies on the passive diffusion of volatile compounds from acidified seawater samples and their subsequent absorption onto a base-impregnated filter. The method is simple, efficient, and adaptable to most commonly used methods of volatile acid analysis.  相似文献

The secretory response of the exocrine pancreas to volatile fatty acids was investigated in the isolated preparations of mice, Japanese field voles and goats. The amylase content in pancreases of the voles was half that of the mice, and the trypsin content was four times higher than that of the mice. Amylase release to volatile fatty acids in the goats showed a dose-dependent relationship. The response in the voles was obtained by acetate and propionate, but preparations in the mice were ineffective in response to volatile fatty acids. The results indicate that the pancreatic digestive enzyme secretory function in voles resembles that of ruminants.  相似文献

Medium-chain-length fatty acids, such as nonanoic (9:0) and octanoic (8:0) acids, are more toxic to Ralstonia eutropha than volatile fatty acids such as acetic, propionic and butyric acids. Nonanoic acid was degraded to acetic and propionic acids via -oxidation by Ralstonia eutropha for cell growth and synthesis of polyhydroxyalkanoates (PHAs). In a mixture of the fatty acids, utilization of nonanoic acid was depressed by acetic and propionic acids, and vice versa. The PHA accumulation from the volatile fatty acids was decreased from 53% (w/w) of dry cell mass to 23% due to the nonanoic acid. Similar phenomena were also observed with octanoic acid and its metabolic intermediates, acetic and butyric acids.  相似文献

Candida ingens, a pellicle-forming yeast utilizing volatile fatty acids, grew over a pH range of 4.1 to 6.0 on nonsterile supernatants from anaerobically fermented pig wastes; growth was inconsistent between pH 4.1 and 4.6. When ambient temperature above the pellicle was 21°C and the temperature of the medium was 29 to 32°C, a pH range of 4.8 to 5.0 gave yields of 1.90 to 3.31 g of dry matter per liter, and 0.059 to 0.065 mol of volatile fatty acids was utilized per liter. There was no advantage in utilization of volatile fatty acids and yield of dry matter in keeping the pH constant during a 24-h growth period. C. ingens grew at pH 4.8 and 5.0 when both ambient and medium temperatures were 30°C. When ambient temperature was 10°C, maximum yield and utilization of volatile fatty acids occurred at a medium temperature of 28 to 30°C.  相似文献

应用毛细管气相色谱-质谱联用技术对红树植物木榄胚轴中挥发性成分和脂肪酸成分进行了分析检测,检测到挥发性成分30个,鉴定了其中的27种成分,占所检测挥发性成分总量的97.01%,以烷烃类为主,占鉴定成分的67.43%,还有酸、酯及萜类化合物等;检测到脂肪酸组分23种,确定了其中的19种,脂肪酸检出量为97.81%,其中饱和脂肪酸占64.87%,不饱和脂肪酸占35.13%。  相似文献

It is known that the presence of volatile fatty acids may play a role in the inactivation of pathogens for systems that employ an acid phase reactor. This study was conducted to investigate the influence of volatile fatty acids on the inactivation of Salmonella spp. over a range of digestion temperatures. In this study, digesters that were treating municipal wastewater treatment plant sludges were operated at temperatures that ranged from 35 to 49 degrees C and had a solids residence time of 15 days. Samples collected from the effluent of the digesters were dosed with solutions containing acetic, propionic, and butyric acids alone and in mixtures, and the dosed effluents were analyzed for Salmonella spp. over time. In the first round of testing, the digester effluents were dosed with individual organic acids and also a mixture containing all three volatile fatty acids over a range of concentrations from 750 to 6000 mg/L, and the pH of the samples was fixed at a value of 5.5. In the second round of testing, the sample sludges were spiked with a fixed amount of organic acid mixture, and the pH was varied from 4.5 to 7.5. The reduction of Salmonella spp. in digester effluents, when dosed with volatile organic acids, was found to depend on pH, temperature, the chain length of the acids, and the concentration and composition of the acids present. Increases in temperature appeared to increase the inhibitory effects of the volatile organic acids. At mesophilic temperatures, acidic pHs resulted in a greater inhibition of Salmonella spp.; whereas at higher temperatures neutral pHs were found to be more inhibitory. The results suggest that acid phase digesters that operate at elevated temperatures and low pH can achieve substantial reduction of Salmonella spp.  相似文献

A method previously used for the analysis of organic acids in silage has been applied to the detection and quantification of acidic fermentation products (C1 to C6 volatile fatty acids, lactic and succinic acid) of rumen bacteria and anaerobic fungi grown in pure culture. The acids were converted to tertiary butyldimethylsilyl derivatives prior to separation on a 30 m DB1 capillary gas chromatographic column. The quantitative recoveries of formic and succinic acids were found to be comparable to the recoveries of other acids reported in the original study. The quantitative recovery of lactic acid was found to be dependent on storage of the samples at ambient temperature for at least 24 h following derivatization. The simultaneous determination of a wide range of volatile and non-volatile acidic products is an important feature of this method.  相似文献

An extraction method coupled with gas-chromatographic analysis has been devised for the determination of small amounts of C1-C7 branched and straight-chain saturated fatty acids as well as 18 mono-, di- and tricarboxylic acids and related compounds, the latter being analyzed either as their methylesters or as their trimethylsilyl-derivatives. Methods for extracting non-volatile acids or both volatile and non-volatile acids are described for large and small volumes of culture media. The proposed methods have been successfully applied to the determination of non-acidic fermentation products, known amounts of alcohols, and to volatile and non-volatile acids in bacterial fermentation solutions, namely a batch culture ofLactobacillus casei subsp.rhamnosus ATCC 7469. A carbon balance sheet of this organism's metabolic behaviour throughout growth is proposed.  相似文献

A method for the preparation and gas chromatographic analysis of the butyl esters of volatile (C-1-C-7) and nonvolatile (lactic, succinic, and fumaric) acids in microbial fermentation media is presented. Butyl esters were prepared from the dry salts of the acids. The esters were separated by temperature programming on a column of Chromosorb W coated with Dexsil 300 GC liquid phase and analyzed with a flame ionization detector. Apparent recoveries with butanol-HCl or butanol-H2SO4 as butylating agents were 80 to 90% for most acids. Chromatographic profiles of the butyl esters demonstrated that both volatile and nonvolatile acids can be detected and separated in 24 min on a single column. Standard calibration curves (peak area versus concentration) of the butyl esters were linear in the range of 5 to 40 mumol of acid per ml. The advantages of using an internal standard (heptanoic acid) for quantitating fatty acids in a mixture are given. Chromatograms of butylated fermentation media in which rumen anaerobic bacteria were grown illustrated that this method is useful for determining short-chain volatile and nonvolatile acids of toxonomic significance.  相似文献

A rapid and simple procedure is described for analysis of fermentation products from anaerobic bacteria grown in glucose broth. A 1-ml sample of the culture is drained through cation-exchange resin in a Pasteur pipette. The effluent fluid is directly analyzed isothermally in a gas chromatograph for volatile fatty acids (C(2) to C(6)) as well as for lactic, pyruvic, and succinic acids. This procedure is considered to be suitable for routine use in clinical bacteriology.  相似文献

Granular sludge from an upflow anaerobic sludge blanket reactor treating synthetic waste water containing a mixture of volatile fatty acids and nitrate showed a removal efficiency of nearly 100% for both nitrogen and carbon. This activity was achieved by a combined process of denitrification and methanogenesis under conditions of surplus carbon. Under batch conditions the two processes proceeded clearly separated in time with first denitrification dominating and excluding methanogenesis. However, as soon as nitrate was depleted, methane production was initiated, showing that the inhibition of methanogenesis by nitrate was reversible. Of the volatile fatty acids supplied to the reactor, i.e. acetate, propionate, and butyrate, the denitrifying population clearly preferred butyrate and propionate even though acetate could also be metabolized. Consequently, growth of syntrophic volatile fatty acid degraders was suppressed by the denitrifiers in cases of low C:N ratios in the medium, leaving acetate as the major substrate for methanogenesis.Abbreviations UASB upflow anaerobic sludge blanket - COD chemical oxygen demand - VFA volatile fatty  相似文献
