Nitrogen dynamics and microbial food web structure were characterized in subtropical, eutrophic, large (2,338 km2), shallow (1.9 m mean depth), and polymictic Lake Taihu (China) in Sept–Oct 2002 during a cyanobacterial bloom. Population growth and industrialization are factors in trophic status deterioration in Lake Taihu. Sites for investigation were selected along a transect from the Liangxihe River discharge into Meiliang Bay to the main lake. Water column nitrogen and microbial food web measurements were combined with sediment–water interface incubations to characterize and identify important processes related to system nitrogen dynamics. Results indicate a gradient from strong phosphorus limitation at the river discharge to nitrogen limitation or co-limitation in the main lake. Denitrification in Meiliang Bay may drive main lake nitrogen limitation by removing excess nitrogen before physical transport to the main lake. Five times higher nutrient mineralization rates in the water column versus sediments indicate that sediment nutrient transformations were not as important as water column processes for fueling primary production. However, sediments provide a site for denitrification, which, along with nitrogen fixation and other processes, can determine available nutrient ratios. Dissimilatory nitrate reduction to ammonium (DNRA) was important, relative to denitrification, only at the river discharge site, and nitrogen fixation was observed only in the main lake. Reflecting nitrogen cycling patterns, microbial food web structure shifted from autotrophic (phytoplankton dominated) at the river discharge to heterotrophic (bacteria dominated) in and near the main lake.  相似文献

1. The microbial metabolism of organic matter in rivers has received little study compared with that of small streams. Therefore, we investigated the rate and location of bacterial production in a sixth‐order lowland river (Spree, Germany). To estimate the contribution of various habitats (sediments, epiphyton, and the pelagic zone) to total bacterial production, we quantified the contribution of these habitats to areal production by bacteria. 2. Large areas of the river bottom were characterized by loose and shifting sands of relatively homogenous particle size distribution. Aquatic macrophytes grew on 40% of the river bottom. Leaf areas of 2.8 m2 m?2 river bottom were found in a 6.6 km river stretch. 3. The epiphyton supported a bacterial production of 5–58 ng C cm?2 h?1. Bacterial production in the pelagic zone was 0.9–3.9 μg C L?1 h?1, and abundance was 4.0–7.8 × 109 cells L?1. Bacterial production in the uppermost 2 cm of sediments ranged from 1 to 8 μg C cm?3 h?1, and abundance from 0.84 to 6.7 × 109 cells cm?3. Bacteria were larger and more active in sediments than in the pelagic zone. 4. In spite of relatively low macrophyte abundance, areal production by bacteria in the pelagic zone was only slightly higher than in the epiphyton. Bacterial biomass in the uppermost 2 cm of sediments exceeded pelagic biomass by factors of 6–22, and sedimentary bacterial production was 17–35 times higher than in the overlying water column. 5. On a square meter basis, total bacterial production in the Spree was clearly higher than primary productivity. Thus, the lowland river Spree is a heterotrophic system with benthic processes dominating. Therefore, sedimentary and epiphytic bacterial productivity form important components of ecosystem carbon metabolism in rivers and shallow lakes. 6. The sediments are focal sites of microbial degradation of organic carbon in a sand‐bottomed lowland river. The presence of a lowland river section within a river continuum probably greatly changes the geochemical fluxes within the river network. This implies that current concepts of longitudinal biogeochemical relationships within river systems have to be revised.  相似文献

Little is known on microbial activities in the sediments of large lowland rivers despite of their potentially high influence on biogeochemical budgets. Based on field measurements in a variety of sedimentary habitats typical for a large lowland river (Elbe, Germany), we present results on the abundance and production of sedimentary bacteria, the potential activity of a set of extracellular enzymes, and potential nitrification and denitrification rates. A diving bell was used to access the sediments in the central river channel, enabling us to sample down to 1 m sediment depth. Depth gradients of all measures of microbial activity were controlled by sediment structure, hydraulic conditions, as well as by the supply with organic carbon and nitrogen. Microbial heterotrophic activity was tightly coupled with the availability of carbon and nitrogen, whereas chemolithotrophic activity (nitrification rate) was related to the available surface area of particles. In the central bed of the river, bacterial production and extracellular enzyme activity remained high down to the deepest sediment layers investigated. Due to the large inner surface area and their connectivity with the surface water, the shifting sediments in the central channel of the river were microbially highly active There, vertically integrated bacterial production amounted to 0.95 g C m−3 h−1, which was 2.9 to 5.5 times higher than in the nearshore habitats. We conclude that carbon and nitrogen cycling in the river is controlled by the live sediments of the central river channel, which thus represent a “liver function” in the river's metabolism.  相似文献

Abstract

The study of microbial communities in river sediments contaminated by thallium (Tl) is necessary to achieve the information for in-situ microbially mediated bioremediation. However, little is known about the microbial community in Tl-contaminated river sediments. In the present study, we characterized the microbial community and their responses to Tl pollution in river sediments from the Tl-mineralized Lanmuchang area, Southwest Guizhou, China. Illumina sequencing of 16S rRNA amplicons revealed that over 40 phyla belong to the domain bacteria. In all samples, Proteobacteria, Cyanobacteria, and Actinobacteria were the most dominant phyla. Based on the UPGMA (Unweighted Pair Group Method with Arithmetic Mean) tree and PCoA (Principal Coordinates Analysis) analysis, microbial composition of each segment was distinct, indicating in-situ geochemical parameters (including Tl, sulfate, TOC, Eh, and pH) had influenced on the microbial communities. Moreover, canonical correspondence analysis (CCA) was employed to further elucidate the impact of geochemical parameters on the distribution of microbial communities in local river sediments. The results indicated that a number of microbial communities including Cyanobacteria, Spirochaete, Hydrogenophaga, and Acinetobacter were positively correlated with total Tl, suggesting potential roles of these microbes to Tl tolerance or to biogeochemical cycling of Tl. Our results suggested a reliable location for the microbial community’s diversity in the presence of high concentrations of Tl and might have a potential association for in-situ bioremediation strategies of Tl-contaminated river. Overall, in situ microbial community could provide a useful tool for monitoring and assessing geo-environmental stressors in Tl-polluted river sediments.  相似文献

In the Azores, the advanced trophic state of the lakes requires a fast intervention to achieve the good ecological status prescribed by the Water Framework Directive. Despite the considerable effort made to describe the phytoplankton growing on the water column, the lack of information regarding the microbial processes in sediments is still high. Thus, for the successful implementation of internal management actions, the present work explored the relationships between geochemical profiles and dominant members of the bacterial community in sediments from eutrophic Azorean lakes. Lake Azul geochemical profiles were quite homogeneous for all parameters, while in lake Furnas the total iron profile presented a peak below the aerobic layer. For lake Verde, the concentrations of all studied parameters (20 ± 2% loss-on-ignition; 2.10 ± 0.08 mg g?1 total phosphorus; 1.31 ± 0.50 mg g?1 total nitrogen; 8.06 ± 0.13 mg g?1 total iron) in the uppermost sediment layer were approximately two times higher than the ones in sediments from other lakes, decreasing with sediment depth. The higher amounts of phosphorus and organic matter in lake Verde suggested a higher internal contribution of phosphorus to eutrophication. The dominant members of the sediment bacterial community, investigated by denaturing gradient gel electrophoresis, were mostly affiliated to Proteobacteria phylum (Alpha-, Delta-, and Gamma-subclasses), group Bacteroidetes/Chlorobi and phylum Chloroflexi. The Cyanobacteria phylum was solely detected in sediments from lake Verde and lake Furnas that presented the highest amounts of nitrogen and phosphorus both in the water column and sediments, while the other phyla were detected in sediments from the three studied lakes. In conclusion, management measurers to achieve the good ecological status until 2015 should be distinct for the different lakes taking into account the relative magnitude of the nutrient sources and the bacterial diversity in sediments.  相似文献

The aerobic polyaromatic hydrocarbon (PAH) degrading microbial communities of two petroleum-impacted Spartina-dominated salt marshes in the New York/New Jersey Harbor were examined using a combination of microbiological, molecular and chemical techniques. Microbial isolation studies resulted in the identification of 48 aromatic hydrocarbon-degrading bacterial strains from both vegetated and non-vegetated marsh sediments. The majority of the isolates were from the genera Paenibacillus and Pseudomonas. Radiotracer studies using 14C-phenanthrene and 14C-pyrene were used to measure the PAH-mineralization activity in salt marsh sediments. The results suggested a trend towards increased PAH mineralization in vegetated sediments relative to non-vegetated sediments. This trend was supported by the enumeration of PAH-degrading bacteria in non-vegetated and vegetated sediment using a Most Probable Numbers (MPN) technique, which demonstrated that PAH-degrading bacteria existed in non-vegetated and vegetated sediments at levels ranging from 102 to 105 cells/g sediment respectively. No difference between microbial communities present in vegetated versus non-vegetated sediments was found using terminal restriction fragment length polymorphism (of the 16S rRNA gene) or phospholipid fatty acid analysis. These studies provide information on the specific members and activity of the PAH-degrading aerobic bacterial communities present in Spartina-dominated salt marshes in the New York/New Jersey Harbor estuary.  相似文献

The distribution of microorganisms in deep subsurface profiles was determined at three sites at the Savannah River Plant, Aiken, South Carolina. Acridine orange direct counts (AODC) of bacteria were highest in surface soil samples and declined to the 106 to 107 per gram range in the subsurface, but then did not decline further with depth. In the subsurface, AODC values varied from layer to layer, the highest being found in samples from sandy aquifer formations and the lowest in clayey interbed layers. Sandy aquifer sediments also contained the highest numbers of viable bacteria as determined by aerobic spread plate counts (CFU) on a dilute heterotrophic medium. In some of these samples bacterial CFU values approached 100% of the AODC values. Viable protozoa (amoebae and flagellates, but no ciliates) were found in samples with high bacterial CFU values. A variety of green algae, phytoflagellates, diatoms, and a few cyanobacteria were found at low population densities in samples from two of the three boreholes. Low numbers of fungi were evenly distributed throughout the profiles at all three sites. Microbial population density estimates correlated positively with sand content and pore‐water pH, and negatively with clay content and pore‐water metal concentration. A large diversity of prokaryotic and eukaryotic microorganisms was found in samples with high population densities. A survey of bacterial strains isolated from subsurface samples revealed associations of gram‐positive bacteria with high clay sediments and gram‐negative bacteria with sandy sediments. The ability to deposit lipophilic storage material (presumably poly‐ß‐hydroxybutyrate) was found in a high proportion of isolates from sandy sediments, but only rarely in isolates from high clay sediments.  相似文献

Viruses were found to be very abundant in the top layer of the sediments of Lac Gilbert, Québec. Viruses were extracted from the sediments using pyrophosphate buffer, and viruses from the diluted extracts were pelleted onto grids and enumerated using transmission electron microscopy. Viral abundance in the sediments ranged from 6.5 × 108 to 1.83 × 1010 ml–1, which is 10- to 1,000-fold greater than the number observed in the water column. This increase corresponds well with the 100- to 1,000-fold increase in bacterial abundance in the sediments. Viral abundance differed significantly among the surface sediment samples taken at different bottom depths and among samples taken at different depths of the water column. Viral abundance also varied significantly between the oxic and anoxic zones of the water column and the sediments. The virus-to-bacteria ratio varied greatly among the different sediment sites but not among depths in the water column. Viral abundance in the water column was related to bacterial abundance and chlorophyll concentration, whereas viruses in the sediments were most abundant in sediments with high organic matter content. Freshwater fouling invertebrate zebra mussels (Dreissena polymorpha) harbor a diverse population of microorganisms in the Great Lakes of North America. Among the indigenous microorganisms, selective species are opportunistic pathogens to zebra mussels. Pathogenicity to zebra mussels by opportunistic bacteria isolated from the mussels was investigated in this study. Among the more than 30 bacteria isolated from temperature-stressed mussels, Aeromonas media, A. veronii, A. salmonicida subsp. salmonicida, and Shewanella putrefaciens are virulent pathogens to juvenile zebra mussels. Inoculation of a bacterial concentration of A. media, A. salmonicida subsp. salmonicida and S. putrefaciens at 107 cells per zebra mussel resulted in 100% mortality within 5 days, and only 64.9% for A. veronii. In contrast, mortality was less than 12.3% following inoculation of a sterile phosphate buffer solution as a control. In addition, mortality was dependent on the size of the pathogen population used in inoculation and the incubation temperature, indicating the close relationship between the bacterial population and subsequent death. On the mussel tissue, a dense microbial population was evident from the moribund mussels viewed with Scanning Electron Microscope (SEM). Opportunistic bacteria invaded and destroyed the D. polymorpha tissue after 7 days of incubation when the bacterial inoculation was larger than 105 per zebra mussel. Our results suggest that mussels are reservoirs of opportunistic pathogenic microorganisms to aquatic organisms and humans and a better understanding of the microbial ecology of the mussels will provide insights to the possible health hazards from these microorganisms.  相似文献

The community structures of anammox bacteria in sediments along an anthropogenic inorganic nitrogen input gradient were further delineated with the newly available information incorporated. Anammox bacterial 16S rRNA gene-amplified sequences retrieved from riparian sediments of the Pearl River, Mai Po coastal wetland, and the South China Sea (SCS) sediments were compiled, compared and analyzed. Results indicated that the community structures of anammox bacteria varied from the upstream of the Pearl River to deep-ocean sediment of the SCS along the anthropogenic input grandient. Mai Po wetland had the most diverse anammox bacteria, followed by the shallow SCS, deep SCS and the Pearl River. Genera of the anammox bacteria Kuenenia and Brocadia showed higher proportion in the riparian sediments of the Pearl River, while those of Kuenenia and Scalindua dominated the Mai Po coastal wetland. The Scalindua subclusters showed apparent segregation in coastal wetland (S. zhenghei-III and S. wagneri), shallow SCS (S. zhenghei-I and S3) and deep SCS (S. zhenghei-I, S2 and S. arabica). Pearson correlation analysis indicated nitrogen species [NH4+ and ∑(NO2?+NO3? )] negatively correlated with the diversity indices of anammox bacteria. Canonical correspondence analysis (CCA) showed that salinity, inorganic nitrogen [NH4+, ∑(NO2?+NO3?)], and ratio of NH4+/∑(NO2? +NO3?) significantly affected the bacterial community compositions. Results collectively support that the community composition of anammox bacteria can serve as a bio-indicator to the anthropogenic terrestrial N input or pollution.  相似文献

The Scheldt river drains a densely populated and industrialized area in northern France, western Belgium and the south-west Netherlands. Mineralization of the high organic load carried by the river leads to oxygen depletion in the water column and high concentrations of dissolved nitrogen and phosphorus compounds. Upon estuarine mixing, dissolved oxygen concentrations are gradually restored due to reaeration and dilution with sea water. The longitudinal redox gradient present in the Scheldt estuary strongly affects the geochemistry of nutrients. Dissolved nutrients in the water column and dissolved nitrogen species in sediment porewaters were determined for a typical summer and winter situation. Water column concentration-salinity plots showed conservative behaviour of dissolved Si during winter. During summer (and spring) dissolved Si may be completely removed from solution due to uptake by diatoms. The geochemistry of phosphorus was governed by inorganic and biological processes. The behaviour of nitrogen was controlled by denitrification in the anoxic fluvial estuary, followed by nitrification in the upper estuary (prior to oxygen regeneration). In addition, nitrogen was taken up during phytoplankton blooms in the lower estuary. Dissolved inorganic nitrogen species in porewaters from the upper 20 cm of sediments were obtained from a subtidal site in the middle of the lower estuary. Dissolved nutrient concentrations were low in the upper 10–15 cm of the sandy and organic poor (<1% POC) sediments mainly as a result of strong sediment mixing. The porewater profiles of ammonium and nitrate were evaluated quantitatively, using a one-dimensional steady-state diagenetic model. This coupled ammonium-nitrate model showed ammonification of organic matter to be restricted to the upper 4 to 7 cm of the sediments. Total nitrification ranged from 3.7–18.1 mmol m?2 d?1, converting all ammonium produced by ammonification. The net balance between nitrification and denitrification depended on the season. Nitrate was released from the sediments during winter but is taken up from the water column during summer. These results are in good agreement with data obtained from the independently calibrated water column model for the Scheldt Estuary (VAN GILSet al., 1993).  相似文献

Disposal of fecally contaminated poultry litter by land application can deliver pathogens and fecal indicator bacteria (FIB) into receiving waters via runoff. While water quality is regulated by FIB enumeration, FIB testing provides inadequate information about contamination source and health risk. This microbial source tracking (MST) study compared the persistence of the Brevibacterium sp. strain LA35 16S rRNA gene (marker) for poultry litter with that of pathogens and FIB under outdoor, environmentally relevant conditions in freshwater, marine water, and sediments over 7 days. Salmonella enterica, Campylobacter jejuni, Campylobacter coli, Bacteroidales, and LA35 were enumerated by quantitative PCR (qPCR), and Enterococcus spp. and E. coli were quantified by culture and qPCR. Unlike the other bacteria, C. jejuni was not detectable after 48 h. Bacterial levels in the water column consistently declined over time and were highly correlated among species. Survival in sediments ranged from a slow decrease over time to growth, particularly in marine microcosms and for Bacteroidales. S. enterica also grew in marine sediments. Linear decay rates in water (k) ranged from −0.17 day−1 for LA35 to −3.12 day−1 for C. coli. LA35 levels correlated well with those of other bacteria in the water column but not in sediments. These observations suggest that, particularly in the water column, the fate of LA35 in aquatic environments is similar to that of FIB, C. coli, and Salmonella, supporting the hypothesis that the LA35 marker gene can be a useful tool for evaluating the impact of poultry litter on water quality and human health risk.  相似文献

We report the isolation of nitrogen fixing, phytohormone producing bacteria from sugarcane and their beneficial effects on the growth of micropropagated sugarcane plantlets. Detection of the nitrogen fixing bacteria by ARA-based MPN (acetylene reduction assay-based most probable number) method indicated the presence of up to 106 bacteria per gram dry weight of stem and 107 bacteria per gram dry weight of root of field-grown sugarcane. Two nitrogen fixing bacterial isolates were obtained from stem (SC11, SC20) and two from the roots (SR12, SR13) of field-grown plants. These isolates were identified as Enterobacter sp. strains on the basis of their morphological characteristics and biochemical tests. The isolate SC20 was further characterized by 16S rRNA sequence analysis, which showed high sequence similarity to the sequence of Enterobacter cloacae and Klebsiella oxytoca. All the isolates produced the phytohormone indoleacetic acid (IAA) in pure culture and this IAA production was enhanced in growth medium containing tryptophan. The bacterial isolates were used to inoculate micro-propagated sugarcane in vitro where maximum increase in the root and shoot weight over control was observed in the plantlets inoculated with strain SC20. By using the15N isotope dilution technique, maximum nitrogen fixation contribution (28% of total plant nitrogen) was detected in plantlets inoculated with isolate SC20.  相似文献

大气氮沉降增加作为全球气候变化的重要因素,其对土壤生态系统影响的研究受到了生态学家的广泛关注。土壤微生物是有机物分解和养分循环的主要参与者,在维持土壤的功能多样性和可持续发展方面发挥着重要的作用。氮沉降的激增会引起土壤微生物群落结构和功能的改变。土壤中营养物质在不同团聚体组分中分布的不均匀,为微生物提供了空间异质微生境。为揭示草原土壤不同粒径团聚体中微生物群落分布及其对氮素添加响应特征。自2010年起,在内蒙古贝加尔针茅草原典型地段设置N0(0 kg hm-2 a-1)、N15(15 kg hm-2 a-1)、N30(30 kg hm-2 a-1)、N50(50 kg hm-2 a-1)、N100(100 kg hm-2 a-1)、N150(150 kg hm-2 a-1)6个氮素添加处理模拟氮沉降野外控制试验。采用磷脂脂肪酸(phospholipid fatty acid,PLFA)法测定>2 mm、0.25-2 mm和<0.25 mm 3个粒径土壤团聚体中微生物PLFA含量,探讨氮素添加对土壤团聚体微生物群落结构的影响。结果表明:氮素添加提高了土壤碳、氮含量,降低了土壤pH。氮素添加显著提高了0.25-2 mm土壤团聚体微生物群落磷脂脂肪酸总量、真菌磷脂脂肪酸含量和真菌/细菌(Fungi/bacteria,F/B)、革兰氏阳性菌/革兰氏阴性菌(Gram-positive bacteria/gram-negative bacteria,G+/G-)的比值(P<0.05),降低了土壤团聚体微生物Margalef丰富度指数(P<0.05)。相关性分析表明,土壤团聚体微生物总PLFAs、真菌PLFAs含量、G+/G-、F/B与土壤有机碳、全氮含量呈显著正相关关系,与C/N值负相关。综合研究表明,连续8年氮素添加显著提高了土壤有机碳和全氮含量、降低了土壤pH;提高了0.25-2 mm土壤团聚体真菌群落,土壤有机碳、全氮的固持与真菌群落的增加有关。  相似文献

微生物对外界环境变化较为敏感,常被作为指示生物用于监测和反映水质情况。为满足延庆世园会和冬奥会举办对妫水河水质的调控要求,探讨妫水河底泥微生物群落结构特征及环境因子对其分布的影响。基于妫水河12个不同断面的水样和底泥样品,进行了水质、底泥理化性质分析,并对底泥的微生物群落结构特征进行了研究。结果表明,妫水河中、下游水体水质COD、NH~+_4-N、TN超标,其中上覆水TN含量与底泥TN含量呈极显著正相关(P=0.914);MiSeq高通量测序发现,妫水河底泥微生物共检出70门228纲1168属,变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)、酸杆菌门(Acidobacteria)、放线菌门(Actinobacteria)、绿弯菌门(Chloroflexi)、厚壁菌门(Firmicutes)、蓝细菌门(Cyanobacteria)、绿菌门(Chlorobi)、疣微菌门(Verrucomicrobia)和硝化螺旋菌门(Nitrospirae)是妫水河底泥微生物群落结构中的主要菌门,在各个样品中相对丰度之和均占84%以上,其中变形菌门为第一优势门,占比达到45.3%—69.1%,而不同断面样品的优势菌属有所不同。妫水河底泥微生物群落丰度总体较高,多样性也相对较高,其中世园段D7点Ace丰富度指数和Shannon多样性指数均较其他点位低,分别为2673和6.56。RDA(redundancy analysis)分析表明,底泥氨氮和温度是影响其微生物群落结构的主要因子(F=2.92,P=0.038;F=2.81,P=0.014),妫水河底泥的优势反硝化菌属为脱氮单孢菌属和硫杆状菌属,其丰度与NH~+_4-N、水温呈正相关,同时与DO呈负相关。研究结果对妫水河水生态环境保护和水质管理提供数据支撑及理论指导意义。  相似文献

The riparian zone is an active interface for nitrogen removal, in which nitrogen transformations by microorganisms have not been valued. In this study, a three-stage system was constructed to simulate the riparian zone environments, and nitrogen removal as well as the microbial community was investigated in this ‘engineered riparian system’. The results demonstrated that stage 1 of this system accounted for 41–51 % of total nitrogen removal. Initial ammonium loading and redox potential significantly impacted the nitrogen removal performances. Stages 1 and 2 were both composed of an anoxic/oxic (A/O) zone and an anaerobic column. The A/O zone removed most of the ammonium load (6.8 g/m2/day), while the anaerobic column showed a significant nitrate removal rate (11.1 g/m2/day). Molecular biological analysis demonstrated that bacterial diversity was high in the A/O zones, where ammonium-oxidizing bacteria and nitrite-oxidizing bacteria accounted for 8.42 and 3.32 % of the bacterial population, respectively. The denitrifying bacteria Acidovorax sp. and the nitrifying bacteria Nitrosospira/Nitrosomonas were the predominant microorganisms in this engineered riparian system. This three-stage system was established to achieve favorable nitrogen removal and the microbial community in the system was also retained. This investigation should deepen our understanding of biological nitrogen removal in engineered riparian zones.  相似文献

Large blooms of opportunistic green macroalgae such as Enteromorpha intestinalis are of ecological concern in estuaries worldwide. Macroalgae derive their nutrients from the water column but estuarine sediments may also be an important nutrient source. We hypothesized that the importance of these nutrient sources to E. intestinalis varies along a nutrient-resource gradient within an estuary. We tested this in experimental units constructed with water and sediments collected from 3 sites in Upper Newport Bay estuary, California, US, that varied greatly in water column nutrient concentrations. For each site there were three treatments: sediments + water; sediments + water + Enteromorpha intestinalis (algae); inert sand + water + algae. Water in units was exchanged weekly simulating low turnover characteristic of poorly flushed estuaries. The importance of the water column versus sediments as a source of nutrients to E. intestinalis varied with the magnitude of the different sources. When initial water column levels of dissolved inorganic nitrogen (DIN) and soluble reactive phosphorus (SRP) were low, estuarine sediments increased E. intestinalis growth and tissue nutrient content. In experimental units from sites where initial water column DIN was high, there was no effect of estuarine sediments on E. intestinalis growth or tissue N content. Salinity, however, was low in these units and may have inhibited growth. E. intestinalis growth and tissue P content were highest in units from the site with highest initial sediment nutrient content. Water column DIN was depleted each week of the experiment. Thus, the water column was a primary source of nutrients to the algae when water column nutrient supply was high, and the sediments supplemented nutrient supply to the algae when water column nutrient sources were low. Depletion of water column DIN in sediment + water units indicated that the sediments may have acted as a nutrient sink in the absence of macroalgae. Our data provide direct experimental evidence that macroalgae utilize and ecologically benefit from nutrients stored in estuarine sediments.  相似文献

The phylogenetic diversity of a microbial community involved in anaerobic oxidation of ammonium nitrogen in the DEAMOX process was studied. Analysis of clone libraries containing 16S rRNA gene inserts of Bacteria, (including Planctomycetes) and Archaea revealed the presence of nucleotide sequences of the microorganisms involved in the main reactions of the carbon, nitrogen, and sulfur cycles, including nitrifying, denitrifying, and ANAMMOX bacteria. In the bacterial clone library, 16S rRNA gene sequences of representatives of the phyla Proteobacteria, Bacteroidetes, Chloroflexi, Firmicutes, Verrucomicrobia, Lentisphaerae, Spirochaetales, and Planctomycetes, as well as of some new groups, were detected. In the archaeal clone library, nucleotide sequences of methanogens belonging to the orders Methanomicrobiales, Methanobacteriales, and Methanosarcinales were found. It is possible that both ANAMMOX bacteria and bacteria of the genus Nitrosomonas are involved in anaerobic ammonium oxidation in the DEAMOX reactor. Many sequences were similar to those from the clone libraries obtained previously from the ANAMMOX community of marine sediments. It is also probable that the DEAMOX reactions occur in natural ecosystems (in marine and freshwater sediments and the oceanic water column), thereby providing for the coupling of the nitrogen and sulfur cycles.  相似文献

Lake Velencei (Hungary) is one of the westernmost shallow soda lakes, extending from Eastern Europe to the Carpatian basin. The spatial and temporal distribution of the sediment microbiota, the metabolic potential of bacterial communities and the species composition of the genera Bacillus and Clostridium, as well as sulphate-reducing bacteria (SRB) were investigated regarding the close interactions between the lake sediment and the overlaying water column, the special water chemical parameters, and the extensive reed coverage of the lake. Aerobic microbial activities were tested with community-level physiological profiling (CLPP) using BIOLOG microplates. The quantification of the anaerobic fermentative and sulphate-reducing bacteria was done by the MPN (Most Probable Number) method. The cultivation of bacteria adapted to the special physico-chemical characteristics of the lake was carried out employing selective media. Multivariate analysis of CLPP data indicated that the microbial communities of the sediment separated from that of the water and showed seasonal variations of the utilised carbon sources. The results of the MPN demonstrated that the counts of the fermentative and sulphate-reducing bacteria in the reed rhizosphere were about one order higher than in the sediment. Among the isolated bacterial strains, a large number were characterised as facultative or obligate alkaliphilic and also moderately halophilic. The partial sequencing of 16S rDNA of the selected representatives resulted in species of aerobic bacteria, such as Bacillus pseudofirmus, B. halmapalus, B. cohnii, B. (Marinibacillus) marinus, and anaerobes, such as Clostridium putrificum – sporogenes, C. scatologenes, C. bifermentans, Desulfotomaculum guttoideum, Desulfovibrio alcoholivorans, and Desulfovibrio burkinensis.
