Observations, both experimental and microscopic, indicate thatzoospores of Rhizophydium planktonicum Canter emend., can remainalive, but rarely become adherent upon cells of Asterionellaformosa Hassall under conditions of very low light or in completedarkness. The behaviour patterns of zoospores towards host cells underdarkened conditions were compared with those which took placeunder well illuminated conditions leading to the settlementof zoospores on host cells. The differences noted may help toexplain the lack of zoospores found upon inadequately illuminateddiatom cells. Some evidence suggests that young zoospores lack the abilityto adhere to host cells. After the encystment of zoospores uponAsterionella cells in the light, their further growth and developmentcan continue in darkness. Background and Aims Dwarf mistletoes (Arceuthobium; Viscaceae)are highly specialized dioecious angiosperms parasitic on manygymnosperm hosts in the northern hemisphere. Several dwarf mistletoespecies are capable of inducing an unusual form of isophasicinfection in which the internal (endophytic) system proliferateseven into the apical buds of its hosts. Studies of the internalendophytic system have, for the most part, focused on the parasitewithin secondary host tissues. The present anatomical and ultrastructuralstudy characterizes the growth pattern of the isophasic endophyticsystem of Arceuthobium douglasii within the dormant apical budsof Pseudotsuga menziesii. • Methods Semi-thin serial sections from dwarf mistletoe-infectedhost apical buds were mounted, stained and micrographed. Graphicfiles were created from the serial micrographs and these fileswere stacked. These stacked files were utilized to describethe pattern of growth of the endophyte within the host tissue.The interface between cells of the mistletoe and host was alsoexamined at the ultrastructural level by transmission electronmicroscopy. • Key Results By utilizing a novel technique of superimposedgraphics, the current study reveals an organized pattern ofmistletoe distribution that penetrates further into host tissuesthan previously known. A consistent pattern of growth occurringeven into the preformed leaves of the host is documented. • Conclusions The apparently non-intrusive growth of theparasite appears to be developmentally synchronized with thatof the host. No symplastic connections were observed in theultrastructural examination of the parasite/host interface withinthe apical buds of Pseudotsuga menziesii parasitized by A. douglasiior of Pinus contorta parasitized by A. americanum.  相似文献

The possible involvement of enzymes in the penetration of intrusivecells of the parasitic angiospermOrobancheinto host root tissueswas studied using cytochemical and immunocytochemical methods.Pectin methyl esterase (PME) was detected, with specific antibodies,in the cytoplasm and cell walls ofOrobancheintrusive cells andin adjacent host apoplast. Depletion and chemical changes ofpectins in host cell walls were shown by histochemical stainingwith PATAg, which detects carbohydrates that are sensitive toperiodic acid, especially pectins, and with the monoclonal antibodiesJIM 5 and JIM 7 that label pectins with low and high rates ofesterification, respectively. Galacturonic sequences with lowrates of esterification were more abundant in host cell wallsadjacent to the parasite, which is consistent with pectin de-methylationby PME release from the parasite. Pectins were absent in middlelamellae and in host cell walls neighbouring mature intrusivecells of the parasite, consistent with further degradation ofpectins by other enzymes. Anatomical studies were carried out on initiation of the secondaryhaustorium in Alectra vogelii, a root parasite of leguminouscrops in Nigeria. In both the normal and self-haustorium, theformation of the haustorial initial on the parasite root soonafter initial contact between the host and parasite roots isfollowed by the penetration of the host root by the haustorium.Specialized penetrating cells (intrusive cells) at the haustorialfront prise apart and loosen the host root cortical cells, whichlater become digested. Through the same processes, a few ofthese columnar intrusive cells at the haustorial front piercethe endodermis to make contact with the xylem of the host root.Thereafter, a true conductive bridge consisting of short, isodiametric,reticulate vessel elements is established between the parasiteand host roots through the secondary haustorium. No pholem tissuewas observed in the connection. There is a close similaritybetween the mode of initiation of the secondary haustorium ofAlectra vogelii and that previously described for its primaryhaustorium. The microscopy and ultrastructure of the interaction of Pucciniastriiformis with a susceptible wheat cultivar was examined atintervals from the time of first haustorium formation to theonset of sporulation. At any particular point in the radiallyexpanding area of infection a sequence of morphological changesoccurred in the infected host cells and the fungus which werecorrelated with successive phases of active fungal growth, accumulationof reserves and finally export of reserves to the developingreproductive structures. Certain species of Chlorella have exploited an intracellularhabitat and occur naturally as cytobionts in Hydra viridissima.The algae evoke phagocytosis by Hydra digestive cells and aresequestered in individual phagosomes that migrate to the baseof the host cells and resist fusion with lysosomes. The abilityto resist digestion is closely correlated with release of extracellularcarbohydrate (maltose) by the algae. The established populationof algae grows at an average rate equal to or greater than thatof the host and a constant population density is maintained.The host regulates algal population density by expelling ordigesting excess algae, or by controlling algal cell division.The control mechanism is unknown but can be breached by additionof inorganic ions to the Hydra culture medium with the resultthat the algae overgrow the Hydra. The Hydra-Chlorella symbiosis is probably mutually beneficial,but conditions such as prolonged darkness (with or without feeding)can reduce the competitive fitness of the host since this conditionresults in heterotrophy by the algae at the expense of selectedhost substrates. The evolution of selective permeability toorganic substrates is a major feature of the successful colonizationof the intracellular habitat by symbiotic Chlorella.  相似文献

The Indian dwarf mistletoe, Arceuthobium minutissimum Hook f.is the most diminutive dicotyledonous stem parasite on Pinusexcelsa. The endophytic system is well developed, having a largenumber of anastomosing strands in the cortex and sinkers penetratingthe medullary rays in wood. The cortical strand is protostelicwith the central tracheary elements, the vessels, surroundedby paren-chymatous cells. An earlier report of absence of vesselsseems to be erroneous. The growth of the cortical strands iseffected by an apical cell. The sinkers typically associatedwith the rays of host, are composed of parenchymatous cellsand tracheary elements including vessels. They make contactswith the cells of the ray through pits present in the trachearyelements. The sinkers cause hypertrophy and even fusion of twoor more rays to form a composite medullary ray. The tracheidsof the host tissue also become stunted and contorted in shape.These observations are in agreement with those of other investigatorson American host species for Arceuthobium.  相似文献

The occurrence of species of the cyanophytes Nostoc and Anabaenain the cortex near the algal zone is reported for apogeotropicroots of Macrozamia communis L. Johnson. Algae were found tooccur both intercellularly and intracellularly in cells of theinner and outer cortex. This is the first record of intracellularalgae in the cycads and only the second report of this phenomenonin vascular plants. By examination of cells at various stagesof invasion by algae, it is interpreted that algal invasionof cortical cells and intercellular spaces is preceded by mucusapparently secreted by algal zone cells of the host, and depositedin intercellular spaces of cortical parenchyma cells nearby.Also algal penetration of cortical cells is preceded by an algalinvasion front of finely granular mucal material which bypassesmucus already deposited in intercellular spaces and may eitherlyse part of the host cell wall or enter through the plasmo-desmata,filling much of the cell cavity. Subsequently, large numbersof the algal symbionts enter the cell and may be enclosed withinhost wall material. Electron microscopic techniques are nowbeing employed to further clarify these invasion processes.  相似文献

Infection and Root-Nodule Development in Stylosanthes Species by Rhizobium   总被引:4,自引:0,他引:4  
Root nodules of the tropical forage legume Stylosanthes occurredonly at lateral root junctions and resulted from direct invasionby rhizobia through spaces between epidermal cells. No infectionthreads were present in either the root hairs or nodules. Invasionof the host cortical cells was through structurally alteredcell walls. The bacteria reached the site of nodule initiationin the lateral root cortex by progressive collapse of the initiallyinvaded cells which were compressed by neighbouring cells toform intercellular thread-like infection zones. The bacteriamultiplied in the invaded cells of the nodule initial whichdivided repeatedly to form the nodule. Bacteroids formed onlywhen the host cells ceased to divide. Some abnormal associations occurred in S. capltata and S. hamata40264A. Division of invaded cells was restricted in S. capitataand the bacteria became enlarged and grossly deformed. In S.hamata restricted cell division was immediastely followed bythe brcakdown of the host cells and, although the bacteria multiplied,no bacteroids were formed. Bacteria isolated from these nodulesformed both effective and abnormal nodules when inoculated ontothe same host.  相似文献

Tobacco plants (Nicotiana tabacum‘Samsun’) havexylem cell walls which are more sensitive to changes in theproperties of the matrix than was predicted using current cellwall mechanical models. A model is proposed which can accountfor the importance of the cell wall matrix in determining themodulus of the material. This model is based on a helical springsystem, with micromechanisms operating at the molecular scale.Xylem tissue fibre cells can behave as helical springs whenthe crosslink density of lignin is low, or they can behave likea composite sheet with fibres at an angle to the applied loadwhen the crosslink density is high. The structure of effective nodules on Trifolium subterraneumL. grown at a root temperature favourable for nitrogen fixationwas examined by light microscopy. Rhizobium were released intohost cells from vesicles on infection threads which were closelyassociated with the host cell nucleus. Enlargement into bacteroidforms was rapid and synchronous in similarly-aged host cells,but groups of rhizobia close to the nucleus sometimes did notdevelop into bacteroids. Rhizobium changed in shape from smallrods to more elongate and then to irregularly swollen formsbefore finally becoming coccoid. On degeneration clusters ofbacteroids became un evenly stained before coalescing at thecentre of the host cell and finally disintegrating. Small vegetativerods multiplied amongst the degenerating bacteroids and in thegreatly enlarged intercellular spaces; host cell walls becamefolded and distorted and sometimes broke before collapsing ontoeach other.  相似文献

The parasitoid nanoflagellate (PNF) Pirsonia diadema is hostspecific for the marine centric diatom Coscinodiscus spp. Experimentsshowed that flagellates significantly prefer C. wailesii overC.granii as host species (interspecific selectivity). This preferencewas independent of light conditions (dark, irradiance of 10and 70 µmol m–2 s–1) and temperature (10 and15C). 活体微注射测定结果表明,将0.5毒囊当量(venom reservoir equivalent, VRE)的蝶蛹金小蜂毒液注射于其寄主菜粉蝶蛹体内,注射后4~24 h寄主浆血细胞和颗粒血细胞的延展、存活和对Sephadex A50微珠的包囊能力显著下降;以0.002~0.02 VRE/μL的该蜂毒液处理其离体寄主血细胞均能产生同样的生理效应。该毒液抑制90%菜粉蝶蛹浆血细胞和颗粒血细胞延展的浓度各为0.00076 VRE/μL和0.00804 VRE/μL。可见,蝶蛹金小蜂毒液能显著抑制其寄主血细胞的延展和包囊作用,并导致血细胞的死亡。然而,同样条件下丽蝇蛹集金小蜂毒液对其非自然寄主菜粉蝶蛹的血细胞延展、存活和包囊作用则无任何效应。可见,寄生蜂毒液的生理作用具有明显的寄主特异性。  相似文献

On emergence the radicle showsa strong curvature which is brought about by a failure of cellextension on one side. The inhibited cells generally remainun-extended throughout the life of the seedling. Endosperm development in Solanum nigrum was ab initio cellular.During early seed development, a Zone of Separation and Secretion(ZSS) differentiated within the endosperm. There were threephases in the formation of the ZSS. Phase I—stainabilityof the cell walls and middle lamella increased followed by numeroussmall plasmalemma invaginations (blebs) which became filledwith a fibrillar material. Phase II—the plasmalemma withdrewfrom the cell wall as a fibrillar lipo-carbohydrate matrix accumulatedoutside the plasmalemma. The middle lamella was gradually removedfrom between the cells forming the central axis of the cone.Phase III—the lipo-carbohydrate matrix continued to accumulateoutside the plasmalemma and also within the developing intercellularspaces. Some axial cells were completely separated from theremaining ZSS cells and became embedded in the matrix. The formationof the ZSS did not entail the destruction of the endosperm cellsand cell divisions were frequent. The ZSS was initially cone-shaped,capping the globular embryo. As the embryo sac enlarged, theZSS continued to differentiate. This resulted in a narrow curvedcorridor through the peripheral region of the endosperm whichterminated above the vascular trace. During germination of the ‘seed’ of Balanophora,endosperm cells at the radicular pole grow out as tubular structuresand anchor the ‘seed’ to the host rootlet. The radiculartier of cells of the embryo elongate as primary haustorial tubesand establish contact with the host root vasculature. Anatomical investigations were carried out on the structureand development of the mature secondary haustorium in Alectravogelii growing on Arachis hypogaea or Vigna unguiculata. Followingthe formation of the young secondary haustorium, both the cambiumand pericycle of the host root directly opposite the young secondaryhaustorium are stimulated to divide and form new tissues andorgans including haustorial roots. Further proliferations ofthe host root pericycle and the haustorial cortex give riseto a large, tuberous and complex mature secondary haustoriumwithin which the tissues of the host and parasite remain inintimate contact forming a perfect graft union with a wide zoneof contact. Apart from the haustorial axial xylcm strand whichnormally connects the xylem of the parasite secondary root withthat of the host, direct xylary connections are also establishedbetween the axial xylem of the haustorium and the xylem of thehaustorial roots. The entire surface of the mature secondaryhaustorium of Alectrais covered with these haustorial rootsas was previously observed in its mature primary haustorium. To assess the role of viruses in the bloom dynamics of Micromonaspusilla in the Gulf of Naples (Mediterranean Sea), variationsof host and virus abundance were followed over one annual cycleand in late winter–spring of three consecutive years.Micromonas pusilla was recorded from autumn to spring, withpeak values up to 6.6 x 103 cells ml–1, but was undetectablein summer. Free M.pusilla viruses were detectable in all seasons,with concentrations from 0.02 viruses ml–1 to 1.9 x 103viruses ml–1, exceeding host abundances only in one case.We found a great intraspecific variability in host susceptibilityto viruses present in natural samples, with viral titres rangingover one or two orders of magnitude for the same samples incubatedon different M.pusilla strains. Over the winter–springperiods, a highly dynamic situation was evident, with wide fluctuationsfor both host and virus abundances from one week to another.In some cases, peak host concentrations were accompanied byan increase in viral numbers, whereas in other cases the respectivefluctuations were uncoupled. Although fluctuations of M.pusillaabundance could be influenced by viral infection, there wasno evidence that viruses were able to terminate host blooms.The summer decline of M.pusilla populations did not appear tobe related to the impact of viral infection.  相似文献

Nodulated plants of Acacia littorea were pot cultured singlyin minus nitrogen sand culture in the presence or absence ofa transplanted seedling of the root hemiparasite Olax phyllanthiand harvests of cultures made 4 and 8 months after introducingthe parasite. Parasitism decreased host shoot growth while increasingroot growth to a similar extent. Final shoot:root dry weightratio was 2.2 for parasitized versus 4.3 for unparasitized Acacia.Partitioning of fixed N showed 4-fold larger N increments inshoots than roots of unparasitized plants, whereas parasitizedplants lost a small amount of shoot N, made a root gain of Ndouble that of unparasitized plants and lost over half of theirN to Olax. The increment of fixed N in the host:parasite associationwas similar to that of unparasitized Acacia. Data on dry mattergain per unit foliage area and mean CO2 assimilation rates pershoot of Olax and Acacia (parasitized or unparasitized) werediscussed in relation to an estimated heterotrophic gain ofxylem C from the host equivalent to 40% of the increment ofdry matter C made by the parasite. Growth of Olax was accompaniedby large increases in numbers of haustoria, 9% of which wereattached to root nodules as opposed to roots. Structural andnutritional features of direct parasitism of nodules are described.Models of flow and utilization of C and N in the Acacia:Olaxassociation and unparasitized Acacia are discussed in relationto published data for other host:parasite associations. Anatomical observations were made on the structure and developmentof the primary haustorium of Alectra vogelii. Its developmentinvolves a mutual aggressive growth of both the host and parasitetissues resulting in the formation of a very large and complextuberous organ. One of the host tissues whose growth is stimulatedby parasite infection is the pericycle whose cells divide repeatedlyand grow around and within the parasite haustorial cortex. Fromvarious points of the proliferating host pericycle, roots becomeinitiated and eventually the entire surface of the haustoriumbecomes covered with these roots. We have referred to them as‘haustorial roots’, a term which we have re-examinedand redefined. True xylary connections are established not onlybetween the parasite and the host root but also between theparasite and these ‘haustorial roots’. The uniquedevelopment of primary haustorium and ‘haustorial roots’in A. vogelii is discussed in relation to the development ofprimary haustoria in other root parasites.  相似文献
