The present report concerns our attempts to further study the effect of quinolone coccidiostats on the sporulation of Eimeria tenella oocysts by analyzing the meiotic behaviour of the chromosomes. To that end, synaptonemal complexes were analyzed by TEM applied to intact meiotic chromosomes. These were isolated after disruption of oocysts, which were harvested from decoquinate-medicated and non-medicated (control) birds. In oocysts from control birds, synaptonemal complexes appeared as the 14 bivalents of the normal karyotype. However, in oocysts from medicated birds, our synaptonemal complex analysis revealed a reciprocal translocation, which was observed as an irregular pairing of chromosome axes 5 and 12 resulting in quadrivalent and trivalent configurations. This finding suggests breakage points in chromosomes 5 and 12 and exchange of chromosomal segments. Furthermore, breakpoints in chromosome 12 resulted in telomere deletion. The chromosomal aberrations described in the present study may result in reduced sporulation since chromosomes involved in translocations segregate abnormally during meiosis. In addition, the results reported provide new evidence of the inhibitory effect of quinolones on the sporulation of E. tenella oocysts, since sporocysts were not formed.  相似文献

用电子显微镜技术观察鲻鱼早期卵子发生进入第一次成熟分裂前期联会丝复合体期、粗线期、双线期和网状期卵原细胞和初级卵母细胞生发泡和胸质的超微结构特点。在联会丝复合体期,生发泡内同源染色体配对,联会丝复合体中央出现重组节,胞质中不同发育类型的核仁样及其相关线粒体的分布及其数量可作为划分鲻鱼早期卵子发生各个时期的依据。另外,首次观察到靠近膜细胞有一种不规则形细胞,推测是分泌成熟抑制肽细胞。  相似文献

Hemipteran chromosomes are holocentric and show regular, special behavior at meiosis. While the autosomes pair at pachytene, have synaptonemal complexes (SCs) and recombination nodules (RNs) and segregate at anaphase I, the sex chromosomes do not form an SC or RNs, divide equationally at anaphase I, and their chromatids segregate at anaphase II. Here we show that this behavior is shared by the X and Y chromosomes of Triatoma infestans and the X(1)X(2)Y chromosomes of Triatoma pallidipennis. As Rec8p is a widely occurring component of meiotic cohesin, involved in meiotic homolog segregation, we used an antibody against Rec8p of Caenorhabditis elegans for immunolocalization in these triatomines. We show that while Rec8p is colocalized with SCs in the autosomes, no Rec8p can be found by immunolabeling in the sex chromosomes at any stage of meiosis. Furthermore, Rec8p labeling is lost from autosomal bivalents prior to metaphase I. In both triatomine species the sex chromosomes conjoin with each other during prophase I, and lack any SC, but they form "fuzzy cores", which are observed with silver staining and with light and electron microscopy during pachytene. Thin, serial sectioning and electron microscopy of spermatocytes at metaphases I and II reveals differential behavior of the sex chromosomes. At metaphase I the sex chromosomes form separate entities, each surrounded by a membranous sheath. On the other hand, at metaphase II the sex chromatids are closely tied and surrounded by a shared membranous sheath. The peculiar features of meiosis in these hemipterans suggest that they depart from the standard meiotic mechanisms proposed for other organisms.  相似文献

Recombination nodules in the oocytes of the chicken, Gallus domesticus   总被引:2,自引:0,他引:2  
Chicken oocytes at pachytene were processed with the microspreading technique (Moses, 1977), and their synaptonemal complex (SC) complements were analyzed by electron microscopy. Ellipsoidal nodules, 140 X 120 nm in diameter, were associated with the central space of synaptonemal complexes. The average number of nodules per pachytene oocyte was 57.5. The number of nodules per bivalent showed a clear linear relationship with SC length, except for the microchromosomes, which showed a single obligatory nodule. The distribution of nodules along the 10 longest SCs was nonrandom, with low frequencies in the vicinity of kinetochores and high frequencies near the telomeres. The microchromosomes showed a single nodule whose average location was 1.21 micron from the kinetochore. In the ZW pair there was a single nodule whose average location was 0.31 micron from the paired telomeres and not more than 0.65 micron from them. The total number of nodules per cell and the number of nodules in each of the five major bivalents showed good agreement with the total number of chiasmata and the number of chiasmata of the major bivalents of roosters. Thus, these nodules share the characteristics of recombination nodules described in other organisms. The single, obligatory, strictly localized recombination nodule found in the pairing end of the ZW pair strongly suggests that recombination between the Z and W chromosomes in the female chicken is a regular process that may be similar to the obligatory recombination between the pairing ends of the human X and Y chromosomes that was recently described in studies using DNA probes.  相似文献

The gypsy moth, Lymantria dispar, produces two structurally and genetically distinct types of spermatozoa. The eupyrene spermatozoa are genetically haploid and structurally typical. The apyrene spermatozoa are anucleate and structurally different from eupyrene spermatozoa. To understand further the events contributing to meiotic chromosome missegregation in apyrene spermatocytes, we examined the progression of meiosis in these cells with respect to their eupyrene counterparts. Chromosomal bouquet formation and fusion of nucleolar organizing regions are disrupted in apyrene nuclei. In addition, the chromatin of apyrene nuclei is prematurely and extremely condensed compared with that of eupyrene nuclei. An antibody to the conserved synaptonemal complex protein 3 (SCP3) labeled eupyrene pachytene chromosomes, but not apyrene pachytene chromosomes. In addition, apyrene meiotic spindles are missing a subset of microtubules, which likely include kinetochore microtubules. Because the condensation behavior of meiotic chromatin in apyrene spermatocytes deviates from that of eupyrene spermatocytes, we examined the appearance and distribution of the phosphorylated form of histone H3, but no significant differences in histone H3 phosphorylation were found between apyrene and eupyrene spermatocytes. We argue that because a pachytene checkpoint is not initiated in apyrene spermatocytes, this system may provide a way to understand better the underlying biochemical connections between pairing, recombination, synapsis, kinetochore assembly and segregation of chromosomes during meiosis in a higher eukaryote.  相似文献

Although the processes of mitosis and meiosis are similar, there is evidence for fundamental regulatory differences between the two. To examine these differences, we have compared the meiotic phenotype of DNA topoisomerase II mutants with their previously described mitotic phenotype (C. Holm, T. Goto, J. Wang, and D. Botstein, Cell 41:553-563, 1985). top2 mutants in meiosis show no defects in the latest detectable stages of recombination, yet they arrest prior to spindle establishment at meiosis I. Fluorescence and electron microscopy reveal that top2 mutants exhibit wild-type levels of meiotic chromosome condensation and form morphologically normal synaptonemal complex but are delayed in the exit from pachytene. Arrested cells retain viability and form colonies if transferred to mitotic medium. Our results suggest that the top2 meiotic arrest is regulatory in nature. This arrest may have evolved to ensure the resolution of fortuitous tangles between nonhomologous chromosomes.  相似文献

Summary InSaprolegnia, kinetochore microtubules persist throughout the mitotic nuclear cycle but, whilst present at leptotene, they disappear coincidently with the formation of synaptonemal complexes at pachytene and reform at metaphase I. In some other fungi chromosomal segregation is random in meiosis and non-random in mitosis. The attachment of chromosomes to persistent kinetochore microtubules in mitosis, but not meiosis, inSaprolegnia provides a plausible explanation for such behaviour. At metaphase I each bivalent is connected to the spindle by 2 laterally paired kinetochore microtubules whereas at metaphase II (as in mitosis) each univalent bears only one kinetochore microtubule, thus showing that all kinetochores are fully active at all stages of meiosis.  相似文献

Electron microscopy of whole-mount surface-spread synaptonemal complex complements and conventional light microscopy of chromosomes at first metaphase of meiosis were used to compare the relative frequencies of pairing configurations at the two stages in inbred autotetraploid rye (Secale cereale L.). Statistical tests showed significantly fewer multivalents at first metaphase than expectations based on random initiation of synapsis at each telomeric site within each group of four homologues. Direct observations of synaptic behaviour of chromosomes showed that this deviation is due primarily to a preponderance of bivalents during zygotene and pachytene. It is also the result of a significant drop in multivalent frequency from meiotic prophase to metaphase I, which is attributable both to a lack of chiasmata with which to consolidate multivalents and inhibition of chiasma formation in synaptonemal complex segments of multivalents that are nonhomologous.  相似文献

In organisms with chiasmatic meiosis two different relationships have been described between crossing over and synapsis: in one group of organisms synapsis depends on the initiation of meiotic recombination while in the other group it is independent of this initiation. These patterns have been observed mainly in organisms where all meiotic bivalents in the set have similar behaviors. In some heteropteran insects a pair of chromosomes named m chromosomes is known to behave differently from autosomes regarding synapsis and recombination. Here we used immunodetection of a synaptonemal complex component and acid-fixed squashes to investigate the conduct of the small m chromosome pair during the male meiosis in the coreid bug Holhymenia rubiginosa. We found that the m chromosomes form a synaptonemal complex during pachytene, but they are not attached by a chiasma in diakinesis. On the other hand, the autosomal bivalents synapse and recombine regularly. The co-existence of these variant chromosome behaviors during meiosis I add further evidence to the absence of unique patterns regarding the interdependence of synapsis and recombination.  相似文献

The cytogenetic study performed has shown that karyotyping of meiotic cells can be based on the synaptonemal complexes (SC) of spreading pachytene spermatocytes of bull and of horse. The horse SC karyotype has not been previously described. A comparison of the relative length of SC with metaphase chromosomes of bull and horse somatic cells has revealed the correspondence of the chromosome length in pachytene of meiosis and metaphase, which is in agreement with the data on house mouse and Chinese hamster. The method of spreading pachytene cells may be of great practical importance in studies of the fertility disturbances in farm animals.  相似文献

During meiosis, sister chromatid cohesion is required for normal levels of homologous recombination, although how cohesion regulates exchange is not understood. Null mutations in orientation disruptor (ord) ablate arm and centromeric cohesion during Drosophila meiosis and severely reduce homologous crossovers in mutant oocytes. We show that ORD protein localizes along oocyte chromosomes during the stages in which recombination occurs. Although synaptonemal complex (SC) components initially associate with synapsed homologues in ord mutants, their localization is severely disrupted during pachytene progression, and normal tripartite SC is not visible by electron microscopy. In ord germaria, meiotic double strand breaks appear and disappear with frequency and timing indistinguishable from wild type. However, Ring chromosome recovery is dramatically reduced in ord oocytes compared with wild type, which is consistent with the model that defects in meiotic cohesion remove the constraints that normally limit recombination between sisters. We conclude that ORD activity suppresses sister chromatid exchange and stimulates inter-homologue crossovers, thereby promoting homologue bias during meiotic recombination in Drosophila.  相似文献

When the resulting protoplasts are exposed to distilled water, they burst, and complete sets of synaptonemal complexes are released to settle on plastic coated slides. After drying and formalin fixation, the synaptonemal complexes can be stained with silver or phosphotungstic acid and observed in the light and/or electron microscope. Silver staining gives better contrast for both light and electron microscopy but stains only lateral elements and kinetochores. Phosphotungstic acid staining gives little or no contrast for light microscopy, but stains both the lateral and central elements of the synaptonemal complex, kinetochores, and structures that are probably recombination nodules for electron microscopy. This technique offers a powerful tool for genome analysis by allowing (1) the determination of relative and absolute lengths of synaptonemal complexes and chromosome arm ratios at pachytene, (2) the analysis of complex patterns of synapsis, and (3) the location of what are probably recombination nodules along the length of synaptonemal complexes.  相似文献

Pigeon spermatocytes were processed with a drying-down technique and their synaptonemal complex (SC) complements were analyzed by electron microscopy. The synaptonemal complex karyotype of the macrobivalents shows an excellent correspondence with the mitotic karyotype. The number and distribution of recombination nodules (RNs) were scored in complete nuclei stained with phosphotungstic acid. The average number of RNs per nucleus is 64.7. The number of nodules per bivalent shows a clear linear relationship with SC length in the 10 longest synaptonemal complexes, while the microbivalents usually bear a single RN. The location of RNs has a non-random distribution along the largest synaptonemal complexes, with lower frequencies near kinetochores and higher frequencies toward the telomeres. The ZZ bivalent is the fourth in size and shows free recombination, having on average 3.8 RNs. The mean number of nodules per cell and the mean number of nodules in the largest bivalents show very good agreement with the corresponding number of chiasmata scored in metaphase-I spermatocytes. It is concluded that the recombination nodules provide a good check for reciprocal exchanges in this and other species of birds. Additionally, a new morphology for the recombination nodules is presented, consisting of groups of electron-dense particles measuring 43 nm in diameter.  相似文献

The prophase of the first meiotic division was studied in field mice of the species Apodemus (Sylvaemus) flavicollis, A. (S.) ponticus, and A. (S.) uralensis by light and electron microscopy. The karyotypes of the species were described on the base of electron microscopy of synaptonemal complexes in spermatocytes I. The axial elements of the sex chromosomes at early-middle pachytene can synapse along the major portion of the Y axis; at late pachytene-early diplotene, the synapsis region shrinks; and at diakinesis-metaphase I, X and Y chromosomes associate tail-to-tail in all species studied. The behavior of sex chromosomes in the synapsis in the species studied was quite uniform. The results are discussed in the context of earlier data on the behavior of sex chromosomes in various rodent species in meiosis prophase I and their banding.  相似文献
