Utilization of storage starch in the cells of cotyledon mesophyll and root meristem in the course of alfalfa (Medicago sativa L.) seed germination on the solutions of NaCl, Na2SO4, and mannitol at different concentrations and identical osmotic pressure was investigated using the method of transmission electron microscopy. Ultrastructural analysis showed changes in the number of starch grains and deceleration of chloroplast development depending on the osmotic component of salt influence. At low concentrations corresponding to osmotic pressure of 202.6 kPa, Na2SO4 did not affect the formation of the photosynthetic machinery and utilization of starch inclusions; mannitol contributed to the preservation of considerable reserve of starch without disturbing the development of chloroplasts; NaCl did not inhibit the development of the photosynthetic machinery and induced an increase in the number of starch grains presumably at the expense of newly produced starch. When the concentration of the investigated substances increased up to the values corresponding to the osmotic pressure of 607.8 kPa, NaCl did not suppress transformation of amyloplasts into chloroplasts and utilization of starch; Na2SO4 inhibited the development of chloroplasts and starch utilization; mannitol decelerated transformation of amyloplasts and inhibited mobilization of starch grains. The obtained results make it possible to propose a method of preliminary estimation of tolerance of dicotyledons to abiotic stresses based on the cytological analysis of utilization of starch grains and formation of photosynthetic compartments of chloroplasts in the mesophyll of cotyledons.  相似文献

Alfalfa is sensitive to waterlogging, and its yields are significantly reduced under this condition. We investigated the effects of soil flooding on free abscisic acid (ABA) accumulation in shoots and roots of alfalfa in relation to plant growth and stomatal conductance responses. The production of dry matter in alfalfa was significantly affected by flooding mainly as a result of a rapid reduction in root growth. Shoot dry matter accumulation was maintained during the first 10 d of treatment and started to decline thereafter. Foliar concentration of the major mineral elements (N, P, K) was reduced by flooding, whereas only K concentration decreased in roots of flooded plants. Regrowth declined with duration of flooding and was less than 50% of controls after 2 weeks. While no changes in ABA concentration could be detected in flooded roots, an increase was noted within a few days in leaves when compared to unflooded controls. This increase in free ABA coincided with the accumulation of large quantities of starch in leaves and a rapid decline in leaf stomatal conductance. Our results support the suggestion that leaf ABA originates from the leaf itself and may be accumulating along with starch as a result of reduced translocation to the roots. Our observation of large accumulations of sucrose in flooded roots agrees with previous reports that supply of carbohydrates is not a limiting factor to root anaerobic metabolism in flooded alfalfa.  相似文献

Sodium chloride and sodium sulfate are commonly present in extraction tailings waters produced as a result of surface mining and affect plants on reclaimed areas. Red-osier dogwood (Cornus stolonifera Michx) seedlings were demonstrated to be relatively resistant to these high salinity oil sands tailings waters. The objectives of this study were to compare the effects of Na2SO4 and NaCl, on growth, tissue ion content, water relations and gas exchange in red-osier dogwood (Cornus stolonifera Michx) seedlings. In the present study, red-osier dogwood seedlings were grown in aerated half-strength modified Hoagland's mineral solution containing 0, 25, 50 or 100 mM of NaCl or Na2SO4. After four weeks of treatment, plant dry weights decreased and the amount of Na+ in plant tissues increased with increasing salt concentration. Na+ tissue content was higher in plants treated with NaCl than Na2SO4 and it was greater in roots than shoots. However, Cl concentration in the NaCl treated plants was higher in shoots than in roots. The decrease in stomatal conductance and photosynthetic rates observed in presence of salts is likely to contribute to the growth reduction. Our results suggest that red-osier dogwood is able to control the transport of Na+ from roots to shoots when external concentrations are 50 mM or less.  相似文献

目的】考察密旋链霉菌Act12发酵液对石油胁迫下紫花苜蓿幼苗生长及生理特性的影响,探讨其缓解石油胁迫的作用机制。【方法】以‘中苜3号’紫花苜蓿作为供试植物,采用盆栽试验,考察3%(W/W)石油胁迫和不同浓度(0、1%、10%和100%)Act12发酵液处理下紫花苜蓿幼苗生长、根系构型、光合特性、渗透调节物质含量和抗氧化能力等指标的变化。【结果】相较于对照组(CK),3%(W/W)石油胁迫显著抑制了幼苗的光合作用和生物量的积累,同时增加了丙二醛的积累,降低了可溶性糖和可溶性蛋白的含量,改变了超氧化物歧化酶(SOD)和过氧化物酶(POD)活性。中低浓度(1%和10%)的发酵液处理可以有效缓解石油胁迫对紫花苜蓿幼苗的毒害,并以10%的Act12发酵液效果最佳;与不施用Act12发酵液处理相比,10% Act12发酵液处理幼苗的株高、地上部干重、地下部干重、茎耐受指数、根耐受指数、总根长、根表面积、根体积和根尖数均显著增加,地上部分和地下部分丙二醛(MDA)含量分别降低了42.49%和56.45%、可溶性糖含量分别增加了79.25%和89.83%、可溶性蛋白含量分别增加了167.63%和256.15%,根系SOD和POD活性分别提升了35.81%和57.33%。【结论】中低浓度的密旋链霉菌Act12发酵液能有效调节石油胁迫下紫花苜蓿幼苗的抗氧化酶活性和渗透调节能力,降低MDA含量,增强光合作用能力,增加生物量,提高抗性,有效缓解石油胁迫对幼苗生长的抑制。  相似文献

Using sand culture, we examined the responses of 6-month-old jack pine (Pinus banksiana Lamb.) seedlings to boron and salinity (sodium chloride and sodium sulfate) treatments. During 4 weeks of treatments, 60 mM NaCl and 60 mM Na2SO4 significantly decreased survival, new shoot length, number of new roots, shoot to root dry weight ratio and transpiration rates. When applied in absence of the salts, B had little effect on the measured variables. However, when applied together with salts, B decreased seedling survival, increased needle injury and altered tissue elemental concentrations in jack pine seedlings. In 2 mM B treatment, B concentration was higher in the shoots than in the roots. However, when 2 mM B was present in NaCl and Na2SO4 treatments, shoot boron concentration declined and greater proportion of B accumulated in the roots. This shift corresponded to a decline in transpiration rates. In plants treated with NaCl, Na accumulated primarily in the shoots, while in Na2SO4-treated plants Na accumulated mostly in the roots. Based on the electrolyte leakage and needle necrosis data, Cl appears to be the major factor contributing to seedling injury and B aggravates the injurious effects of NaCl. We suggest that Cl may contribute to Na and B toxicity in jack pine by altering cell membrane permeability leading to increased Na concentration in the shoots.  相似文献

Pitiuba bean [ Vigna unguiculata (L.) Walp.] seeds were sown in water or. in 0.1 M NaCl. Seedling growth and cotyledon nucleic acid mobilization were delayed by NaCl salinity. The differences in cotyledonary RNase activity between seeds sown in water and in NaCl solutions suggest that salinity delays the activation and/or de novo synthesis of the enzyme. Cotyledon extracts were subjected to gel filtration through Sephadex G-100, and RNase activity measured. Only one cotyledonary RNase appeared during germination, and salinity did not induce any change in molecular weight of the enzyme. Salinity inhibited 45% of the specific activity of the RNase on the 5th day of the experimental period. The same salt concentration (0.1 M NaCl) added in vitro inhibited only 8 % of the specific activity of the enzyme. This difference may indicate that NaCl in vivo affects mainly the de novo synthesis of the RNase.  相似文献

Abstract: Black spruce (Picea mariana), white spruce (Picea glauca), and jack pine (Pinus banksiana) seedlings were inoculated with Hebeloma crustuliniforme or Laccaria bicolor and subjected to NaCl and Na2SO4 treatments. The effects of ectomycorrhizas on salt uptake, growth, gas exchange, and needle necrosis varied depending on the tree and fungal species. In jack pine seedlings, ectomycorrhizal (ECM) fungi reduced shoot and root dry weights and in the ECM white spruce, there was a small increase in dry weights. Sodium chloride treatment reduced net photosynthesis and transpiration rates in the three studied tree species. However, NaCl-treated black spruce and jack pine colonized by H. crustuliniforme maintained relatively high photosynthetic and transpiration rates and needle necrosis of NaCl-treated black spruce seedlings was reduced by the ECM fungi. Higher concentrations of Na+ were found in shoots compared with roots of the three examined conifer species. ECM fungi reduced the concentrations of Na+ mainly in the shoots and this reduction was greater in plants treated with NaCl compared with Na2SO4. Shoots contained generally higher concentrations of Cl- compared with roots. In the NaCl-treated black spruce and white spruce, both ECM species significantly reduced Cl- concentrations. Our results point to overall greater phytotoxicity of NaCl compared with Na2SO4 and support our earlier findings which demonstrated beneficial effects of ECM fungi for woody plants exposed to NaCl stress.  相似文献

华北地区是我国玉米的主产区,玉米秸秆还田不仅可有效改善土壤理化性状、提高土壤生物有效性,还会在腐解过程中释放出目前公认的化感物质——酚酸类物质,邻苯二甲酸是玉米秸秆腐解液中的主要酚酸类物质。从玉米秸秆还田过程中主要腐解产物(邻苯二甲酸)对蔬菜作物的化感效应角度进行了研究,为量化秸秆还田量及构建粮-菜轮作制度探寻化感效应依据。试验以萝卜为蔬菜材料,通过配置4个浓度(0.05、0.5、1.0、2.0 mmol/L)的邻苯二甲酸溶液,模拟玉米秸秆还田条件,以清水为对照,研究主要腐解产物邻苯二甲酸对萝卜种子萌发、幼苗生长、膜脂过氧化作用及渗透调节物质的影响。结果表明:(1)萝卜不同生育期对邻苯二甲酸化感效应的响应程度不同。在0.05-1 mmol/L浓度范围内,邻苯二甲酸处理促进了萝卜种子萌发,但随着处理浓度的增大,促进作用减弱;浓度达到2 mmol/L时对萝卜种子萌发具有抑制效果。(2)邻苯二甲酸0.05mmol/L处理,促进了萝卜幼苗干鲜物质积累,幼苗根系生长,其中根系长度和根系表面积分别比对照提高42.03%、38.36%,显著高于清水对照;植株体内超氧化物歧化酶(Superoxide dismutase, SOD)活性增大,过氧化物酶(Peroxidase, POD)、过氧化氢酶(Catalase, CAT)活性降低,膜脂过氧化产物丙二醛(Malonaldehyde, MDA)含量与对照无显著差异。(3)当邻苯二甲酸浓度超过0.5 mmol/L时,萝卜幼苗脂质过氧化伤害加剧,体内MDA含量急剧增加,代谢与生理功能出现紊乱,正常生长及干鲜物质积累受到显著抑制。邻苯二甲酸浓度达到2 mmol/L时,叶片数较对照降低了36.51%;根系长度、根系表面积及根尖数降幅分别为64.46%、40.20%、41.28%。(4)对于渗透调节物质的影响,邻苯二甲酸处理促进了萝卜幼苗叶片可溶性糖含量的增加,但随着处理浓度的升高其促进作用逐渐减弱;可溶性蛋白含量随着邻苯二甲酸处理浓度的升高表现出逐渐减少的趋势,分别较对照降低了12.82%、14.88%、21.58%、24.73%。因此,华北地区实施玉米-萝卜轮作模式,从化感效应角度研究玉米秸秆量化还田,应将土壤中邻苯二甲酸浓度控制在0.5 mmol/L范围以内,以防止邻苯二甲酸浓度过高对萝卜幼苗生长的抑制作用。  相似文献

The study focused on the dynamics of Malondialdehyde (MDA) contents and the activities of protective enzymes in the leaves of alfalfa varieties with various resistances to Aphis medicaginis Koch. The results showed that susceptible varieties always had higher MDA contents than resistant varieties, and the MDA contents tended to rise in both susceptible and resistant varieties in period of the varieties were pierced and sucked by aphids. Superoxide dismutase (SOD), peroxidase (POD) and phenylalanine ammonia-lyase (PAL) activities in susceptible varieties were lower than those in resistant varieties, and in both susceptible and resistant varieties the SOD and POD activities tended to rise at first and then decline, and the PAL activities rose to their peaks and then tended to remain stable. In the susceptible and resistant varieties the catalase (CAT) activities appeared to rise and decline alternatively; the PPO activities in resistant varieties were lower than those in susceptible varieties in early growth, but higher than those in susceptible varieties in later growth. It follows that infested by aphids, susceptible and resistant varieties had the MDA contents, variations of SOD, POD, PAL and PPO activities were closely correlated with their aphid resistances, hence these indexes could be used as physiological indexes for testing aphid resistance of alfalfa, whereas the relations of their CAT activities to their resistances needed to be further studied.  相似文献

Nucleolytic activities from two plants of Leguminosae family were determined in order to consider if the nucleases of plants which belong to the same family or to the same species responded in similar ways to stress conditions during growth. Growth parameters of both plants were examined in parallel. In detail, seedlings from two plants, alfalfa (Medicago sativa L. cv. Luzerne Euver) and lentil (Lens culinaris cv. Thessalia), showed significant differences in response to iso-osmotic solutions of NaCl (100 mmol · L−1 solution equivalent to conductivity 8.0 dS m−1) and mannitol (190 mmol · kg−1). Plant height and dry weight of mannitol/NaCl-treated seeds in both plants were lower in comparison to controls (water). Mannitol stress reduced height and dry weight in alfalfa seedlings more than did NaCl. By contrast, lentil seedling growth was inhibited more by NaCl stress than mannitol. In addition, DNase and RNase response to mannitol stress differed in each plant compared to the controls. Mannitol stress induced a sharp increase in DNase- and RNase-specific activity during the initial stages of alfalfa seedlings' growth, followed by a decrease during subsequent days; in lentil seedlings, these activities were inhibited throughout the entire growth period. NaCl stress inhibited the above activities in both plants. After native electrophoresis on gels polymerized in the presence of DNA/RNA, the overall band intensities confirmed the above quantitative results of alfalfa RNase and DNase activity. In addition, the active gel analysis revealed that the decrease of nucleolytic activities in mannitol-treated alfalfa seedlings was mainly due to the strong reduction of acid nucleases. This is the first report of different non-ionic osmotic response of type I plant nucleases during seedlings' growth. In vitro, the addition of up to 300 mmol/L mannitol did not affect acid and neutral nuclease activity in enzyme preparations extracted, purified, and separated from control and mannitol-treated alfalfa seedlings.Our results suggest that plant nucleases responded in a different way to osmotic stress and ionic stress conditions during seedlings' growth.  相似文献

在甘肃省定西市九华沟系统研究了喷施蚜虱净对苜蓿主要害虫及天敌种群数量动态的影响。结果表明,施药田蚜虫和蓟马的季节平均数量极显著高于对照田,而盲蝽和瓢虫、小花蝽、蜘蛛等大多数天敌的季节平均数量则极显著低于对照田,寄生蜂与对照田没有湿著差异。喷施蚜虱净对种群数量时序动态的影响在不同害虫和天敌种类中表现不同,5月底施药后7d对照田蚜虫和蓟马的种群数量分别为施药田的4.13倍和5.2倍,此后施药田害虫的种群数量开始迅速增加,到6月底2种害虫的种群数量均与对照田无显著差异,到7月中旬以后反而显著高于对照田。施药对盲蝽种群数量时序动态的影响与蚜虫和蓟马不同,5月底施药后盲蝽的种群数量一直显著低于对照田,直到7月下旬以后,种群数量才恢复到对照田的水平。喷施蚜虱净对天敌种群数量的时序动态有极显著的影响,大多数天敌的种群数量在5月底施药后到7月底都一直显著低于对照田,直到8月上旬才恢复到对照田的水平。施用杀虫剂在防治害虫的同时杀伤大量天敌,使害虫的种群数量迅速增加,甚至造成更严重的为害。因此在菖蓿上使用对天敌杀伤力小的杀虫剂对保持天敌的自然控制能力有重要作用。  相似文献

Phaseolus aureus Roxb. was exposed to HgCl2 and Cd(NO3)2 either at the germination stage in concentration 0.5, 5 and 25 μM for 48 and 96 h, or at the seedling stage (5th day of germination) in concentration 0.5, 5 and 20 μM for 6, 24 and 48 h. The germination and the growth of roots (germination stage treatment) were less in Hg than in Cd treatment. The seedlings (seedling stage treatment) were, however, more susceptible to Cd than Hg. Both root and leaf tissues of the plant treated at the germination stage showed enhanced lipid peroxidation and activities of the antioxidative enzymes (catalase, guaiacol peroxidase and ascorbate peroxidase), except the catalase in leaf in 25 μM Cd treatment. At seedling stage the content of malondialdehyde increased significantly only in the leaf tissue, during 6 h exposure. The activities of all the enzymes exhibited an increasing trend in both the tissue of the seedlings, particularly the leaf, at least after 24 and 48 h, except the catalase whose activity declined in response to Cd. Active involvement of the guaiacol and ascorbate peroxidases, rather than catalase, in scavenging cellular H2O2 was indicated. It was concluded that the two metals had little primary damaging effect on membranes.  相似文献

The spores of the fern Anemia phyllitidis contain abundant quantities of lipid as reserve material. Germination of these spores can be induced either by red light or, even in the dark, by gibberellic acid. The effects of both these factors on lipid degradation, lipase and isocitrate lyase activities, and on the fatty acid composition have been studied in the course of the germination process. During germination in darkness with gibberellic acid, the fatty acid composition remained similar to that in the ungerminated spore. In contrast, when spores were germinated in red light, α-linolenic acid was synthesized. Little activity of lipase and isocitrate lyase could be detected in the dry spore. Red light or gibberellic acid affected a dramatic increase of the activities of these enzymes. Lipid breakdown and lipase activity were more active in red light, however. Permanent stimuli were necessary for growth and complete lipid degradation. Induction of germination simultaneously with both factors revealed an additivity of the effects of red light and gibberellic acid.  相似文献

Quercus seedlings have hypogeal cotyledons and tap roots, both of which act as storage organs. The importance of the storage function in the two organs may change as the seedling develops. Therefore, changes in carbohydrate reserves in cotyledons and roots of Q. crispula grown under 40 % and 3 % of full light from shoot emergence to the completion of the first leaf flush were monitored. In addition, a shoot-clipping treatment was performed to examine the relative contribution of the cotyledons and tap roots to resprouting. Cotyledons maintained large amounts of nonstructural carbohydrates during shoot development, and carbohydrates were still present in the cotyledons during the final phase of leaf flush. In addition, a notable increase in the amount of carbohydrates was observed in tap roots before leaf flush at both light levels. Since root development occurred before leaf flush, even in plants grown under 3 % light, the carbohydrate found in them presumably originated from seed reserves and was translocated to roots as storage reserves. When shoots were clipped at the leaf flushing stage, the amount of carbohydrate decreased only in the cotyledons after resprouting, suggesting that cotyledons act as the main storage organs during shoot development stages. However, it could be advantageous as a 'risk avoidance strategy' for the seedlings to store reserves in both cotyledons and roots, since cotyledons may be removed by predators during shoot development.  相似文献

Centaurea ragusina L. is a Croatian endemic plant species growing on cliffs above the Adriatic Sea, but there is no information about its physiological behavior or stress tolerance. To investigate the response of C. ragusina plants to salinity and drought, we have analysed soluble peroxidase (POD; EC 1.11.1.7) activity, anionic isoperoxidase pattern, levels of malondialdehyde (MDA) and hydrogen peroxide in C. ragusina plants exposed to these stresses. Rooted plantlets grown on MS 1/2 nutrient medium supplemented with mannitol (300 mM) and different concentrations of NaCl (150, 300, 450 or 600 mM) were harvested after 5, 10 and 15 days. Both osmotic treatments significantly increased MDA and hydrogen peroxide contents in C. ragusina shoots after 10 days of stress, while in roots these parameters showed no significant difference compared to control in overall. POD activity of salt-stressed plants changed with respect to different saline treatments and plant organs - in shoots enzymatic activity markedly increased in response to lower saline treatments, especially 300 mM NaCl; otherwise it was similar as in control plants while in roots of plants grown under 450 and 600 mM NaCl it significantly decreased. Drought increased POD activity of both shoots and roots especially after 10 days of experiment. Generally, change in the POD isoenzyme pattern of treated plants was in accordance with the activity change in time. Several POD isoforms (P3, P4 and P9) were specifically induced by salinity and drought.  相似文献

Transpiration and water absorption rates, stomatal and cuticular resistances to water vapour diffusion, were measured onPlantago maritima (halophyte) andPlantago lanceolata (glycophyte) grown in the presence of NaCl or Na2SO4. Water absorption was reduced in the presence of Na2SO4 and transpiration rate was increased when NaCl was added to the nutrient solution. BACKGROUND AND AIMS: The influence of initial residual leaf area and initial N reserves on N uptake, final N distribution, and yield in alfalfa regrowing after cutting, were studied. METHODS: The effects of two levels of initial residual leaf area (plants cut to 15 cm, with (L+) or without (L-) their leaves) and two initial levels of N status [high N (HN) or low N (LN)] on growth, N uptake and N partitioning, allocation and storage after 29 d of post-cutting regrowth were analysed. KEY RESULTS: During most of the regrowth period (8-29 d after the initial harvest), HN and L+ plants had higher net N uptake rates than LN and L- plants, respectively, resulting in a greater final mineral N uptake for these treatments. However, the final partitioning of exogenous N to the regrowing shoots was the same for all treatments (67 % of total exogenous N on average). Final shoot growth, total plant N content, and N allocation to the different taproot N pools were significantly lower in plants with reduced initial leaf area and initial N reserve status. CONCLUSIONS: Although both initial residual leaf area and initial N reserves influenced alfalfa regrowth, the residual leaf area had a greater effect on final forage production and N composition in the taproot, whereas the N uptake rate and final total N content in plant were more affected by the initial N reserve status than by the residual leaf area.
