Diabetes mellitus (DM), a metabolic disorder, is becoming a major health problem worldwide. Insulin is the single hope for management of type 1 diabetes, but it is not always available or suitable. For finding additional bioresources, the present study was performed. ELISA‐based preliminary screening of cyanobacterial biomass using antihuman insulin antibody have detected an insulin‐like antigen in Spirulina platensis S‐5, Spirulina NCCU482, and Spirulina NCCU483. Their similarity with insulin‐like antigen was further confirmed by electrophoretic mobility using bovine insulin as marker.  相似文献

Although nucleosomes and histones are lacking in dinoflagellate nuclei, small basic histone‐like proteins have been reported, but their function(s) is unknown. In this study we cloned and sequenced a gene for a histone‐like protein from the dinoflagellate Lingulodinium polyedrum (Stein) Dodge (HLp) (formerly Gonyaulax polyedra Stein) and investigated its post‐translational modification and DNA‐binding activities. HLp appears to be acetylated in L. polyedrum, and we identified several L. polyedrum proteins that possess histone acetyltransferase activity and may be responsible for this modification. HLp binds weakly to L. polyedrum DNA but to certain specific sequences with higher affinity, consistent with its having a regulatory function.  相似文献

The transmission–virulence trade‐off hypothesis is one of the few adaptive explanations of virulence evolution, and assumes that there is an overall positive correlation between parasite transmission and virulence. The shape of the transmission–virulence relationship predicts whether virulence should evolve toward either a maximum or to an intermediate optimum. A positive correlation between each of these traits and within‐host growth is often suggested to underlie the relationship between virulence and transmission. There are few experimental tests of this hypothesis; this study reports on the first empirical test on a plant pathogen. We infected Brassica rapa plants with nine natural isolates of Cauliflower mosaic virus and then estimated three traits: transmission, virulence, and within‐host viral accumulation. As predicted by the trade‐off hypothesis, we observed a positive correlation between transmission and virulence, suggestive of the existence of an intermediate optimum. We discovered the unexpected existence of two groups of within‐host accumulation, differing by at least an order of magnitude. When accumulation groups were not accounted for, within‐host accumulation was correlated neither to virulence nor transmission, although our results suggest that within each group these correlations exist.  相似文献

The 14‐3‐3 proteins are evolutionarily conserved acidic proteins that form a family with several isoforms in many cell types of plants and animals. In invertebrates, including dipteran and lepidopteran insects, only two isoforms have been reported. 14‐3‐3 proteins are scaffold molecules that form homo‐ or heterodimeric complexes, acting as molecular adaptors mediating phosphorylation‐dependent interactions with signaling molecules involved in immunity, cell differentiation, cell cycle, proliferation, apoptosis, and cancer. Here, we describe the presence of two isoforms of 14‐3‐3 in the mosquito Aedes aegypti, the main vector of dengue, yellow fever, chikungunya, and zika viruses. Both isoforms have the conserved characteristics of the family: two protein signatures (PS1 and PS2), an annexin domain, three serine residues, targets for phosphorylation (positions 58, 184, and 233), necessary for their function, and nine alpha helix‐forming segments. By sequence alignment and phylogenetic analysis, we found that the molecules correspond to ? and ζ isoforms (Aeae14‐3‐3ε and Aeae14‐3‐3ζ). The messengers and protein products were present in all stages of the mosquito life cycle and all the tissues analyzed, with a small predominance of Aeae14‐3‐3ζ except in the midgut and ovaries of adult females. The 14‐3‐3 proteins in female midgut epithelial cells were located in the cytoplasm. Our results may provide insights to further investigate the functions of these proteins in mosquitoes.  相似文献

It was observed that in the female gametophyte of Undaria pinnatifida (Harv.) Suringar (Phaeophyta, Laminariales) gametangial initials and maturing oogonia demonstrated different levels of alkaline‐phosphatase activity (APA). The oogonia exhibited a higher level of APA than in its initials. Electron‐dense granular ovoid structures ~0.5–0.6 μm were present in the cytoplasm of oogonia. These inclusions were not membrane bound and do not appear to be associated with any particular organelles. The number of the inclusions was 1 to 2 in a single section of the cell. In essential details, the specific APA and subcellular germinal body‐like structure of the developing female gamete in U. pinnatifida were very similar to those in metazoan oocytes.  相似文献

When the chlorophyte alga Dunaliella tertiolecta Butcher is placed in darkness, a form of programmed cell death with many similarities to apoptosis is induced, including the induction of caspase‐like proteases. Many uncertainties about the regulation and mediators that participate in the process remain. To examine the relationship between caspase‐like activities and different apoptotic events (i.e., phosphatidylserine [PS] translocation), increases in membrane permeability and numbers of dead cells revealed by SYTOX‐green staining, and the generation of reactive oxygen species (ROS), we used the broad‐range caspase inhibitor Boc‐D‐FMK to block the activity of the whole class of caspase‐like proteins simultaneously. In the presence of the inhibitor, ROS were not produced, and cells did not die. Loss of membrane asymmetry, indicated by external labeling of PS by annexin V, was apparent at midstages of light deprivation, although it did not conform to the typical pattern for PS exposure observed in metazoans or vascular plants, which occurs at early stages of the apoptotic event. Thus, we have evidence for a link between ROS and cell death involving caspase‐like enzymes in an alga. The fact that caspase‐like inhibitors prevent not only cell death, but also ROS and loss of cell membrane integrity and asymmetry, suggests that caspase‐like proteases might have regulatory roles early in cell death, in addition to dismantling functions.  相似文献

Impairment of glucose‐stimulated insulin secretion (GSIS) caused by glucolipotoxicity is an essential feature in type 2 diabetes mellitus (T2DM). Palmitate and eicosapentaenoate (EPA), because of their lipotoxicity and protection effect, were found to impair or restore the GSIS in beta cells. Furthermore, palmitate was found to up‐regulate the expression level of sterol regulatory element‐binding protein (SREBP)‐1c and down‐regulate the levels of pancreatic and duodenal homeobox (Pdx)‐1 and glucagon‐like peptide (GLP)‐1 receptor (GLP‐1R) in INS‐1 cells. To investigate the underlying mechanism, the lentiviral system was used to knock‐down or over‐express SREBP‐1c and Pdx‐1, respectively. It was found that palmitate failed to suppress the expression of Pdx‐1 and GLP‐1R in SREBP‐1c‐deficient INS‐1 cells. Moreover, down‐regulation of Pdx‐1 could cause the low expression of GLP‐1R with/without palmitate treatment. Additionally, either SREBP‐1c down‐regulation or Pdx‐1 over‐expression could partially alleviate palmitate‐induced GSIS impairment. These results suggested that sequent SREBP‐1c‐Pdx‐1‐GLP‐1R signal pathway was involved in the palmitate‐caused GSIS impairment in beta cells. J. Cell. The combined effects of ammonium concentration and UV radiation on the red alga Porphyra columbina (Montagne) from the Patagonian coast (Chubut, Argentina) was determined using short‐term (less than a week) experimentation. Discs of P. columbina were incubated with three ammonium concentrations (0, 50, and 300 μM NH4Cl) in anilluminated chamber (PAR=300 μmol photons·m?2·s?1, UVA=15 W·m?2, UVB=0.7 W·m?2) at 15°C. Algae incubated at 300 μM ammonium showed a significant increase (P<0.05) in the concentration of mycosporine‐like amino acids (MAAs) compared with the initial value or with the other ammonium treatments. The increase of MAAs was, however, a function of the quality of irradiance received, with a higher increase in samples exposed to UVA compared with UVB (29% and 5% increase, respectively). However, UVB radiation was more effective in inducing MAA synthesis per unit energy received by the algae. Samples exposed to PAR only had an intermediate increase in MAA concentration of 16%. Chl a concentration decreased through the incubation with the greatest decrease at high ammonium concentration. Phycobiliprotein (BP) decreased through time with the smallest decrease occurring at high ammonium concentration. Photoinhibition (as a decrease of optimal quantum yield) was significantly greater under nitrogen‐deprived conditions than that under replete ammonium levels. Maximal gross photosynthesis (GPmax), as oxygen evolution, and maximal electron transport rate (ETRmax), as chl fluorescence, increased with the ammonium concentration. Positive relationships between maximal GP or ETR and pigment ratios (BP/chl a and MAAs/chl a) and negative relationships with chl a concentration were found.  相似文献

Abstract We inserted the mosquito esterase B1 gene into the expression vector pRL‐439, which possesses the strong promoter PpsbA. The recombinant plasmid pRL‐Bl was used to transform E. coli HBlOl, and the positive clones were screened on LB medium plate containing 100 mg/mL ampicillin. The results of dot blotting and Southern hybridization demonstrated that these positive clones were transformed bacteria. Western blotting indicated that esterase B1 gene had been successfully expressed under the control of the PpsbA promoter in E. coli. A shuttle verruction‐B1 (pDGBl) was constructed by inserting B1‐cDNA from pRL‐Bl into polycloning site of plasmid pDc8. PDGBl was transferred into Synechoccus sp. PCC7942 through triparental conjugal transfer. Transformed single Synechococcus sp. PCC 7942 clone was obtained by neomycin screening, and large‐scale culture in liquid medium was carried out. Results of Southern blotting proved that pWB1 was transferred into Synechococcus sp. PCC 7942.  相似文献

The photoprotective response in the dinoflagellate Glenodinium foliaceum F. Stein exposed to ultraviolet‐A (UVA) radiation (320–400 nm; 1.7 W · m2) and the effect of nitrate and phosphate availability on that response have been studied. Parameters measured over a 14 d growth period in control (PAR) and experimental (PAR + UVA) cultures included cellular mycosporine‐like amino acids (MAAs), chls, carotenoids, and culture growth rates. Although there were no significant effects of UVA on growth rate, there was significant induction of MAA compounds (28 ± 2 pg · cell?1) and a reduction in chl a (9.6 ± 0.1 pg · cell?1) and fucoxanthin (4.4 ± 0.1 pg · cell?1) compared to the control cultures (3 ± 1 pg · cell?1, 13.3 ± 3.2 pg · cell?1, and 7.4 ± 0.3 pg · cell?1, respectively). In a second investigation, MAA concentrations in UVA‐exposed cultures were lower when nitrate was limited (P < 0.05) but were higher when phosphate was limiting. Nitrate limitation led to significant decreases (P < 0.05) in cellular concentration of chls (chl c1, chl c2, and chl a), but other pigments were not affected. Phosphate availability had no effect on final pigment concentrations. Results suggest that nutrient availability significantly affects cellular accumulation of photoprotective compounds in G. foliaceum exposed to UVA.  相似文献

The cell wall of the green alga Micrasterias denticulata Bréb. ex Ralfs (Desmidiaceae, Zygnematophyceae, Streptophyta) was investigated to obtain information on the composition of component polysaccharides and proteoglycans to allow comparison with higher plants and to understand cell wall functions during development. Various epitopes currently assigned to arabinogalactan‐proteins (AGPs) of higher plants could be detected in Micrasterias by immuno TEM and immunofluorescence methods, but the walls did not bind the β‐d ‐glycosyl‐Yariv (β‐GlcY) reagent. Secretory vesicles and the primary wall were labeled by antibodies against AGPs (JIM8, JIM13, JIM14). Dot and Western blot experiments indicated a proteoglycan nature of the epitopes recognized, which consisted of galactose and xylose as major sugars by high performance anion exchange chromatography with pulsed amperometric detection (HPAEC‐PAD). Epitopes of alkali‐soluble polysaccharides assigned to noncellulosic polysaccharides in higher plants could be detected and located in the wall during its formation. The polyclonal anti‐xyloglucan (anti‐XG) antibody labeled primary and secondary wall of Micrasterias, whereas the monoclonal antibody CCRC‐M1, directed against the fucose/galactose side chain of xyloglucan (XyG), did not recognize any structures. Labeling by anti‐XG antibody at the trans‐sites of the dictyosomes and at wall material containing vesicles indicated that secretion of the epitopes occurred similar to higher plants. The presence of (1→3, 1→4)‐β‐glucan (mixed linked glucan) in the secondary cell wall but not in the primary cell wall of Micrasterias could be demonstrated by an antibody recognizing this glucan type, whereas (1→3)‐β‐glucan (callose) could not be detected. The analytical results revealed that alkali‐soluble polysaccharides in the secondary wall of Micrasterias consist mostly of (1→3, 1→4)‐β‐d ‐glucan.  相似文献

The phylogenetic group stramenopiles refers to the systematic groups that possess tripartite tubular hairs (stramenopiles) on their flagella. There have been a number of studies describing the fine structure of these mastigonemes and a few studies isolating the component proteins; however, these proteins and their gene sequences have not yet been identified. In the present study, we identified a mastigoneme protein (Ocm1) of the chrysophycean alga Ochromonas danica Pringsh. (UTEX LB1298). Its corresponding gene, Ocm1, was identified by using degenerate primers that correspond to the partial amino acid sequences of a protein (85 kDa) obtained from a mastigoneme‐rich fraction of isolated flagella. The polypeptide encoded by Ocm1 has four cysteine‐rich, epithelial growth factor (EGF)–like motifs, potentially involved in protein–protein interactions. It lacks obvious hydrophobic regions characteristic of transmembrane domains, suggesting that this polypeptide is not likely a protein for anchoring the mastigoneme. In addition, a polyclonal antibody against Ocm1 labeled the area where the tubular shafts of the mastigonemes are located, but not the basal portion or the terminal filaments.  相似文献

Research Objective: This study focuses on ADs in the Netherlands and introduces a cross‐cultural perspective by comparing it with other countries. Methods: A questionnaire was sent to a panel comprising 1621 people representative of the Dutch population. The response was 86%. Results: 95% of the respondents didn't have an AD, and 24% of these were not familiar with the idea of drawing up an AD. Most of those familiar with ADs knew about the Advanced Euthanasia Directive (AED, 64%). Both low education and the presence of a religious conviction that plays an important role in one's life increase the chance of not wanting to draw up an AD. Also not having experienced a request for euthanasia from someone else, and the inconceivability of asking for euthanasia yourself, increase the chance of not wanting to draw up an AD. Discussion: This study shows that the subjects of palliative care and end‐of‐life‐decision‐making were very much dominated by the issue of euthanasia in the Netherlands. The AED was the best known AD; and factors that can be linked to euthanasia play an important role in whether or not people choose to draw up an AD. This differentiates the Netherlands from other countries and, when it comes to ADs, the global differences between countries and cultures are still so large that the highest possible goals, at this moment in time, are observing and possibly learning from other cultural settings.  相似文献

The coccolithophores Emiliania and Pleurochrysis demonstrate increased coccolith production when growth is reduced by nitrate or phosphate limitation. The function of enhanced coccolith production under these conditions and its regulation have not been resolved. Studies at the molecular level are ideally suited to determine the exact relationship between calcification and other cellular functions. In a previous study we provided evidence for the presence of a vacuolar H+‐ATPase on coccolith vesicle membranes of P. carterae. These trans‐Golgi–derived vesicles are the sites of coccolith production, with each vesicle containing one coccolith. In this study, expression of the vap gene, encoding subunit c of the vacuolar H+‐ATPase, was investigated. Our objective was to explore potential relationships between vap expression, nutrient‐dependent growth, and calcification. Specifically, we monitored vap expression relative to two genes, fcp and pcna, whose expression was previously shown to vary with growth conditions; fcp encodes a fucoxanthin chl a/c‐binding protein, and pcna encodes the proliferating cell nuclear antigen. Relative to the expression of pcna and fcp, vap expression was highest at nutrient concentrations where growth curves and chl a patterns indicated arrest of cell division. Our results indicate that the level of vap expression does not decrease when cell growth diminishes.  相似文献

Cortical F‐actin reorganization during the cell cycle was observed in Pyrenomonas helgolandii U. J. Santore (SAG 28.87) for the first time in Cryptophyta using fluorescein‐isothiocyanate (FITC)–phalloidin staining. In interphase, a number of F‐actin bundles were observed as straight lines running parallel to the long axis of the cell on the cell cortical region. They extended from an F‐actin bundle that runs along the margin of the vestibulum. Although the F‐actin bundles running parallel to the long axis of the cell disappeared during anaphase, they gradually reappeared in telophase. By contrast, the F‐actin bundle along the vestibulum margin remained visible during cytokinesis and dynamically changed following the enlargement of the vestibulum, suggesting that F‐actin was involved in the mechanism of vestibulum enlargement. F‐actins were not found in the cytoplasmic and nucleoplasmic regions throughout the cell cycle. In addition, a contractile ring‐like structure appeared at the cleavage furrow during cytokinesis. Treatment with cytochalasin B and latrunculin B significantly inhibited the formation of cleavage furrow, resulting in forming an abnormal cell with two nuclei, suggesting that cytokinesis in P. helgolandii is controlled by the contractile ring‐like structure constituted of F‐actin.  相似文献

Few members of the well‐studied marine phytoplankton taxa have such a complex and polymorphic life cycle as the genus Phaeocystis. However, despite the ecological and biogeochemical importance of Phaeocystis blooms, the life cycle of the major bloom‐forming species of this genus remains illusive and poorly resolved. At least six different life stages and up to 15 different functional components of the life cycle have been proposed. Our culture and field observations indicate that there is a previously unrecognized stage in the life cycle of P. antarctica G. Karst. This stage comprises nonmotile cells that range in size from ~4.2 to 9.8 μm in diameter and form aggregates in which interstitial spaces between cells are small or absent. The aggregates (hereafter called attached aggregates, AAs) adhere to available surfaces. In field samples, small AAs, surrounded by a colony skin, adopt an epiphytic lifestyle and adhere in most cases to setae or spines of diatoms. These AAs, either directly or via other life stages, produce the colonial life stage. Culture studies indicate that bloom‐forming, colonial stages release flagellates (microzoospores) that fuse and form AAs, which can proliferate on the bottom of culture vessels and can eventually reform free‐floating colonies. We propose that these AAs are a new stage in the life cycle of P. antarctica, which we believe to be the zygote, thus documenting sexual reproduction in this species for the first time.  相似文献

The sterile insect technique (SIT) was developed to eradicate the new world screwworm from the southern United States and Mexico, and became a component of many area‐wide integrated pest management programs, particularly useful in managing tephritid fruit flies. SIT is based on the idea of rearing and sterilizing male pests, originally by ionizing radiation, and then releasing into field, where they compete for and mate with wild females. Mating with sterile males leads to reduced fecundity to lower pest populations. There are concerns with the use and distribution of radioisotopes for SIT programs, which have led to developing X‐ray irradiation protocols to sterilize insects. We considered the possibility that X‐ray irradiation exerts sublethal impacts aside form sterilizing insects. Such effects may not be directly observable, which led us to the hypothesis that X‐ray irradiation in one life stage creates alterations in biological fitness and protein expression in the subsequent stage. We tested our hypothesis by irradiating larvae of Bactrocera dorsalis. There are two major points. One, exposing larvae to X‐ray treatments led to reduced adult emergence, fecundity, fertility, and flight capacity from the corresponding pupae and emerged adults. Two, the X‐ray treatments led to substantial expression changes in 27 pupal proteins. We assorted the 67 spots representing these proteins into three groups, metabolism, development, and structure. Our interpretation is these X‐ray induced changes in biological performance and protein expression indicate their adult counterparts may be disabled in their abilities to successfully compete for and mate wild females in native habitats.  相似文献
