We investigated the uptake, transport, storage and defence mechanisms in the freshwater crab, Potamonautes warreni, harbouring microbial gill infestations and exposed to increasing chronic (0.2, 0.5, 1.0 mg l–1) and acute (2.0 mg l–1) cadmium (Cd) concentrations under controlled laboratory conditions over a period of 21 days. Transmission electron microscopy and X-ray microanalysis revealed that the microbial gill fauna was eliminated on exposure to 0.2 mg Cd2+ l–1 and that Cd became increasingly adsorbed and incorporated into lamellar crystal deposits and permeated the cuticle of the gills of P. warreni. Degeneration of the apical membrane infoldings and vacuolation of epithelial cells occurred concurrently with pinocytosis, endocytosis and pronounced phagocytotic activity in the epithelia and haemal canal of the gills. Elevated Cd exposures (0.5 or 1.0 mg l–1) resulted in the swelling and dissociation of mitochondrial outer membranes together with an increase in transport of Cu, Cl and S by haemocytes in the haemal canal to epithelial tissues depleted in these elements. Cd also accumulated in tightly coiled concentric membrane whorls in the haemal canal, whereas the highest concentrations of Cd were found within aggregates of lysosome-like bodies in cuticulin-secreting cells of the gill stem. Chronic exposure to Cd induced increased fatigue and mild uncoordinated motor activity. In contrast, at an acute exposure of 2.0 mg l–1 over 48 h, P. warreni showed a time-specific rapid loss of motor function, although only mild cellular lesions occurred in the gill tissues. The significance of cellular changes in the gill epithelia and altered motor activity of P. warreni with increased waterborne Cd are discussed as potential biomarker responses in monitoring aquatic pollution.  相似文献

Isolated posterior gills (no. 7) of shore crabsCarcinus maenas acclimated to brackish water of a salinity of 10 S were bathed and perfused with 50% sea water (200 mmol·l-1 Na+), and the internal perfusate collected during subsequent periods of 5 min. During a single passage through the gill the pH of the perfusion medium decreased from ca. 8.1 to ca. 7.7, a result implying that the gill possesses structures which recognize unphysiologically high pH values in the haemolymph and regulates them down to physiological values of ca. 7.7. The calculated apparent proton fluxes from the epithelial cells into the haemolymph space amounted to 17.9 mol·g fw-1·h-1, a value of only 3.8% of net Na+ fluxes observed under comparable conditions. When 0.1 mmol·l-1 KCN, an inhibitor of mitochondrial cytochrome oxidase, or 5 mmol·l-1 ouabain, a specific inhibitor of Na+/K+-ATPase were applied in the internal perfusate, down-regulation of pH was no longer observed and the gill was completely depolarized, i.e. transepithelial potential differences dropped from-7.8 to 0 mV (haemolymph space negative to bath). Regulation of pH was completely inhibited by antagonists of carbonic anhydrase (0.1 mmol·l-1 acetazolamide or 0.01 mmol·l-1 ethoxyzolamide) applied in the perfusate. Inhibitors of Na+/H+ exchange, 0.1 mmol·l-1 amiloride applied in the external bathing medium or in the internal perfusate, and symmetrical 0.01 mmol·l-1 5-(N-ethyl-N-isopropyl)amiloride, as well as inhibitors of Cl-/HCO3 - exchange and Na+/HCO3 - cotransport, 0.5 mmol·l-1 4,4-diisothiocyanatostilbene-2,2-disulphonate or 0.3 mmol·l-1 4-acetamido-4-isothiocyanatostilbene 2,2-disulphonate applied on both sides of the gill, and inhibitors of H+-ATPase, 0.05 mmol·l-1 N-ethylmaleimide and 0.1 mmol·l-1 N,N-dicyclohexylcarbodiimide —applied on both sides of the gill — did not alter the acidification of the perfusate observed in controls. Using artificial salines buffered to pH 8.1 with 0.75 mmol·l-1 tris (hydroxymethyl) aminomethane instead of 2 mmol·l-1 HCO3 -, apparent proton fluxes were reduced to 11% of controls, a result suggesting that pH regulation by crab gills needs the presence of HCO3 -. The findings obtained suggest that pH regulation by crab gills depends on the oxidative metabolism of the intact branchial epithelium and that carbonic anhydrase plays a central role in this process. Na+/H+ exchange, anion exchange or cotransport and active proton secretion seem not to be involved. Cl absorption across isolated, perfused gills of freshwater adapted Chinese crabs (Eriocheir sinensis) was analysed by measuring transepithelial potential differences (PDte) and radioactive tracer fluxes across isolated, perfused posterior gills. Applying hemolymph-like NaCl salines on both sides of the epithelium PDte amounted to −30±1 mV (n=14). Undirectional Cl influxes of 470±38 and effluxes of 245±27 μmol·hr−1·g−1 wet weight (ww) (n=14) resulted in a Cl net influx of 226±31 μmol·hr−1·g−1 ww. Symmetrical substitution of Na+ by choline resulted in a substantial hyperpolarisation of the gill. Cl influx was unchanged under these conditions. However, net influx of Cl decreased by 40%, due to an increase of the Cl efflux.Nevertheless, a significant Cl net influx remained which was independent of the presence of Na+. When 2 mmol/l ouabain were added to the internal perfusion medium, PDte increased, although the fluxes remained unchanged. Following external application of 1μmol/l of the V-type H+-ATPase inhibitor bafilomycin, Al PDte and Cl effluxes were not significantly affected. However, Cl influxes decreased. These findings suggest that Cl can be taken up independently of Na+ and that active Na+ independent Cl uptake across the posterior gill of Eriocheir sinensis is probably driven by a V-type H+-ATPase localized in the apical membrane.  相似文献

The mangrove crab Ucides cordatus showed a significant reduction in its output of isosmotic urine (to 60% of submerged rates) in humid, emersed conditions when in apparent water balance. A similar, but lesser, reduction was observed in the intertidal temperature species Carcinus maenas. Some of the decrease is effected by increased water withdrawal of an isosmotic fluid in the antennal gland (inulin urine to haemolymph concentration ratio=1.27) in Ucides but most is due to the reduced filtration rate which decreases from 3.41 to 2.19 ml·100 g-1·day-1 (in Carcinus from 7.37 to 5.88). In Carcinus, inulin urine to haemolymph concentration ratios are within the range 1.34–1.41 and show significant change on emersion. Comparisons of ion and inulin clearance ratios revealed interspecific differences in renal handling of Na+ and Cl-. During emersion total Na+ efflux is markedly reduced in both species. Urinary Na+ losses are responsible for most of the efflux in Carcinus but represent only 50% of the total in Ucides. Significant Na+ uptake from interstitial water occurs in Ucides. REductions in ion and water loss appear to be mainly dependent on decreases in urine output (clearance rate). However, some released urine was found to accumulate in the branchial chambers in both species. Summary Calcium fluxes were measured simultaneously in brown trout fry maintained in an artificial soft water medium of [Ca] 20 mol·l-1 and pH 5.6, and exposed to each of six trace metals (Al, Cu, Fe, Ni, Pb, and Zn). The trace metal concentrations represented typical and maximum levels found in acid waters experiencing declining fishery status. In the absence of trace metals, evidence is presented which suggests that ca. 91% of Ca taken up from the external medium was by extraintestinal active transport. Calcium efflux was stimulated by both concentrations of Al, Cu, Fe, and Pb. Efflux was also stimulated by [Ni] 170 nmol·l-1 and [Zn] 3000 nmol·l-1. In some cases, response to increased efflux was stimulation of influx. Lack of stimulation of influx resulted in negative net Ca fluxes. Summary Groups of Arctic charr,Salvelinus alpinus, which had been acclimated to water with a salinity of 7 g·l–1 and natural temperature and photoperiod, were exposed to water with different salinities and temperatures in June, September and February. At a salinity of 15 g·l–1, plasma osmolality, plasma Na+, Cl, Mg2+ concentrations and the activity of gill Na-K-ATPase were stable, irrespective of temperature and season. In June, the charr were able to regulate blood plasma ionic levels within narrow limits when exposed to a salinity of 34 g·l–1 (sea water) and a temperature of 8°C. The hypo-osmoregulatory capacity was less, but sufficient if the temperature was only 1°C during the seawater exposure. At the start of the experiment, the gill Na-K-ATPase activity was significantly higher in June than corresponding enzyme activities in September and February. Furthermore, an increase in gill Na-K-ATPase activity during the seawater exposure (8°C) was seen in June. Irrespective of ambient temperature and salinity, no fish died during the June experiments. In September and February, exposure to sea water produced marked increases in plasma osmolality and plasma ion concentrations. There were no changes in gill Na-K-ATPase activity. Consequently, the fish became dehydrated and were moribund after a short period of seawater exposure. Highest mortality was recorded when charr were exposed to winter sea conditions (34 g·l–1 and 1°C) in February. The results indicate that an increase in daylength induce a hypo-osmoregulatory capacity in the Arctic charr during summer. In fall and winter, however, reduced daylength are accompanied by poor hypo-osmoregulatory capacity. This leads to high mortality as a result of increased electrolyte levels and dehydration.  相似文献

Isolated perfused gills of stenohaline crabs Cancer pagurus adapted to seawater, brackish water-adapted euryhaline shore crabs Carcinus maenas and freshwater-adapted extremely euryhaline Chinese crabs Eriocheir sinensis were tested for their capacity to excrete ammonia. Gills were perfused with haemolymph-like salines and bathed with salines equal in adaptation osmolality. Applying 100 μmol · l−1 NH4Cl in the perfusion saline and concentrations of NH4Cl in the bath that were stepwise increased from 0 to 4000 μmol · l−1 allowed us to measure transbranchial fluxes of ammonia along an outwardly as well as various inwardly directed gradients. The gills of all three crab species were capable – to different extents – of active excretion of ammonia against an inwardly directed gradient. Of the three crab species, the gills of Cancer pagurus revealed the highest capacity for active excretion of ammonia, being able to excrete it from the haemolymph (100 μmol · l−1 NH+ 4) through the gill epithelium against ambient concentrations of up to 800 μmol · l−1, i.e. against an eightfold gradient. Carcinus maenas and E. sinensis were able to actively excrete ammonia against approximately fourfold gradients. Within the three crab species, the gills of E. sinensis exhibited the greatest capacity to resist influx at very high external concentrations of up to 4000 μmol · l−1. We consider the observed capacities for excretion of ammonia against the gradient as ecologically meaningful. These benthic crustaceans protect themselves by burying themselves in the sediment, where, in contrast to the water column, concentrations of ammonia have previously been reported that greatly increase haemolymph levels. Electrophysiological results indicate that the permeabilities of the gill epithelia are a clue to understanding the species-specific differences in active excretion of ammonia. During the invasion of brackish water and freshwater, the permeabilities of the body surfaces greatly decreased. The gills of marine Cancer pagurus exibited the greatest permeability (ca. 250 mS cm−2), thus representing practically no influx barrier for ions including NH+ 4. We therefore assume that C. pagurus had to develop the strongest mechanism of active excretion of ammonia to counteract influx. On the other hand, freshwater-adapted E. sinensis exhibited the lowest ion permeability (ca. 4 mS cm−2) which may reduce passive NH+ 4 influxes at high ambient levels. Summary Two primarily granivorous rodents of Old World deserts,Gerbillus allenbyi (mean adult body mass=26 g) andG. pyramidum (mean adult body mass=40 g), coexist in sandy habitats in the northwestern Negev desert. Both species are burrow dwellers and are nocturnal; however, in their overall distributions,G. pyramidum occurs in more extreme deserts than doesG. allenbyi. In comparing field metabolic rate (FMR) and water influx of the twoGerbillus species, we considered two alternative hypotheses: (1) given the difference in their overall distributions,G. pyramidum has a lower FMR and water influx thanG. allenbyi, and (2) given the similarity in their diets, and that we worked with sympatric populations, FMR and water influx are similar. The latter alternative proved to be correct. Field metabolic rates in summer were 7.29 kJ · g-0.51 · day-1 forG. allenbyi and 7.74 kJ · g-0.51 · day-1 forG. pyramidum, values that were 69.3% and 74.5%, respectively, of those predicted for rodents of their body masses. Summer water influx ofG. allenbyi was 0.167 ml · g-0.90 · day-1 and that ofG. pyramidum was 0.144 ml · g-0.90 · day-1; these values were 79.4% and 68.6%, respectively, of water influxes predicted for rodents of their body masses. When compared allometrically, there were no interspecific differences in any of the measurements.  相似文献

Mitochondria-rich cells have been separated from other epithelial cells of tilapia (Oreochromis mossambicus) gills by density gradient centrifugation on Percoll. During centrifugation two main bands of cells formed. The viability of the cells in both bands was high (>90%). In one band, 45–47% of the total cell number was mitochondria-rich cells. The other band contained at least 80% pavement cells, representing the majority of other gill epithelial cell types. A comparison of the activities of four enzymes involved in major metabolic and ion regulatory functions was made between these two different fractions of cells. Furthermore, the separation of gill epithelial cells and determination of enzymatic activity was carried out in tilapia after the fish were acclimated to fresh water or hyperhaline sea water (60 mg·ml-1 S) to gain an indication of the relative contribution of mitochondria-rich cells and pavement cells to both NaCl excretion and absorption. Regardless of acclimation salinity, the activities of Na+/K+-ATPase, glutamate dehydrogenase and glucose-6-phosphate dehydrogenase were significantly higher in mitochondria-rich cells than in pavement cells. However, tilapia acclimated to hyperhaline sea water possessed significantly lower carbonic anhydrase activity in mitochondria-rich cells than in pavement cells. Summary The isolated gills of Carcinus maenas, perfused at pressure drops of 1–10 cm of water, exhibited flow rectification, the resistance to perfusion via the afferent vessel being many times lower than that for efferent perfusion. The asymmetry was greater at the lower end of this pressure range.The overall afferent branchial resistance for Carcinus of weight 65 g, and with no ventilatory component in the transmural pressure difference, was estimated to be 0.05 cm of water. l–1 · sec. The corresponding overall reverse (efferent) branchial resistance was 0.36 cm of water · l–1 · sec.LM, TEM and SEM examination of the gills indicated that haemolymph leaves each gill lamella via several discrete parallel efferent channels which drain different regions of the lamella, and that each efferent channel is nearly closed, at its junction with the efferent branchial vessel, by a diaphragm of loosely interwoven and very elongated cells. It is concluded that these cells may constitute efferent valves and that narrow apertures between them may contribute a major component to the branchial resistance and be primarily responsible for the rectification of flow. Relatively wide apertures lead directly from the afferent vessel into the lamellae and are not asociated with valves of any kind.The valves may be important in enabling changes in transmural pressure associated with ventilatory reversals to pump haemolymph unidirectionally through the lamellae. Similarly valves may allow the oscillating venous pressures associated with locomotor activity to improve gill perfusion during exercise.The elongated tails of the cells of the efferent valve contain numerous microtubules. The wider cell bodies contain the nucleus and many mitochondria. Unusual organelles composed of many short (about 0.25 m long) microtubules and often lying close to the nuclear membrane may be microtubule organising centres. It is speculated that, in addition to their simple mechanical function, the valve cells may play a more dynamic role in regulating flow of haemolymph through different lamellar routes, or that they may monitor composition, pressure or flow of the efferent lamellar circulation.  相似文献

We examined transepithelial transport of Ca2+ across the isolated opercular epithelium of the euryhaline killifish adapted to fresh water. The opercular epithelium, mounted in vitro with saline on the serosal side and fresh water (0.1 mmol·l–1 Ca2+) bathing the mucosal side, actively transported Ca2+ in the uptake direction; net flux averaged 20–30 nmol·cm–2·h–1. The rate of Ca2+ uptake varied linearly with the density of mitochondria-rich cells in the preparations. Ca2+ uptake was saturable, apparent K 1/2 of 0.348 mmol·l–1, indicative of a multistep transcellular pathway. Ca2+ uptake was inhibited partially by apically added 0.1 mmol·l–1 La3+ and 1.0 mmol·l–1 Mg2+. Addition of dibutyryl-cyclic adenosine monophosphate (0.5 mmol·l–1)+0.1 mmol·l–1 3-isobutyl-l-methylxanthine inhibited Ca2+ uptake by 54%, but epinephrine, clonidine and isoproterenol were without effect. Agents that increase intracellular Ca2+, thapsigargin (1.0 mol·l–1, serosal side), ionomycin (1.0 mol·l–1, serosal side) and the calmodulin blocker trifluoperazine (50 mol·l–1, mucosal side) all partially inhibited Ca2+ uptake. In contrast, apically added ionomycin increased mucosal to serosal unidirectional Ca2+ flux, indicating Ca2+ entry across the apical membrane is rate limiting in the transport. Verapamil (10–100 mol·l–1, mucosal side), a Ca2+ channel blocker, had no effect. Results are consistent with a model of Ca2+ uptake by mitochondria rich cells that involves passive Ca2+ entry across the apical membrane via verapamil-insensitive Ca2+ channels, intracellular complexing of Ca2+ by calmodulin and basolateral exit via an active transport process. When intact crayfish are in an ion-poor medium (KCl, 0.1 mmol·l-1+KHCO3, 0.1 mmol·l-1) there is a large potential difference (transepithelial potential difference),-20 to-40 mV (hemolymph negative), across the gills. Addition of Ca2+ to the medium is followed by a rapid change in transepithelial potential difference to near 0 mV. The transepithelial potential difference showed a non-linear dependence on [Ca2+]out with a limiting value of+2 to+10 mV at>1 mmol·l-1. The concentration generating a half-maximum transepithelial potential difference change (15–20 mV) was 0.1 to 0.2 mmol·l-1. Three other alkaline earth ions were also electrogenic; Ba2+ caused slightly larger transepithelial potential difference changes, Sr2+ and Mg2+ were a little less effective. It has been suggested that the transepithelial potential difference in ion-poor medium (in fish) is due to the diffusive efflux of NaCl across the gills, with a Cl-/Na+ permeability ratio of <1. Evidence is presented that this might be the case in crayfish. The electrogenic effect of Ca2+ might then be due to its effect on gill permeability to Na+ and Cl- such that the permeability ratio increased and approached unity as the transepithelial potential difference approached 0. However, this was shown to be unlikely. Summary Chloride extrusion is examined in the isolated perfused gill of the pinfish,Lagodon rhomboides. In both sea water and Ringer's baths, the Cl efflux from the isolated gill is 45% that of the intact animal. The transepithelial electrical potential (TEP) across the isolated gill in sea water is equal to that in vivo, in Ringer's the gill TEP is slightly less than in vivo. Cl efflux is linearly dependent upon afferent flow of the perfusate. Furosemide, added to the perfusate inhibits 57% of the Cl efflux in gills bathed bilaterally by Ringer's. Ouabain causes a marked vasoconstriction and increase in afferent pressure. Removal of Na from the perfusate produces an inhibition of the Cl efflux that is not potential mediated. Net extrusion of Cl is inhibited in isolated gills bathed bilaterally by sodium free Ringer's.  相似文献

There was no direct effect of copper on the ontogeny or function of the heart of the brine shrimp Artemia franciscana in sea water (salinity= 36 mg·ml-1, 25°C). There was, however, an indirect effect as an increase in copper concentration resulted in a reduced growth rate. There was no difference between the critical O2 tensions of newly hatched (stage 0/1) nauplii of control and treated (<0.32 and 10.11 mol·l-1 copper, respectively) individuals. However by developmental stages 4–6, when both the heart and thoracic gills are in the process of differentiating, respiratory performance had improved (i.e. critical O2 tension decreased from 6.27±0.45 to 4.69±0.24 kPa) in control but not in copper-treated individuals. It is suggested that respiratory impairment of stages 4–6 individuals is unlikely to be related to differences in cardiac performance or cellular respiration. Instead it may be related to metal-related damage to newly differentiating gill tissue and/or by copper in some way compromising the normal ontogenic shift in haemoglobin O2 affinity. Summary Daily intramuscular injection of cortisol (4 mg kg–1 body weight) in rainbow trout,Salmo gairdneri, for 10 days caused significant increases in the number and individual apical surface area of gill chloride cells per mm2 of filament epithelium. Concomitantly, whole body influxes of sodium (Na+) and chloride (Cl) increased. Acute (3 h) intra-arterial infusion of cortisol did not affect whole body Na+ or Cl influx. A significant correlation was observed between both Na+ and Cl influxes and the fractional apical surface area of filament chloride cells in control, sham (saline-injected) and experimental (cortisol-injected) fish. The chloride cells displayed similar ultrastructural modifications in trout undergoing cortisol treatment as in trout transferred to ion-deficient water. These findings suggest the existence of structure/function relationships in which branchial chloride cell morphology is an important determinant of Na+ and Cl transport capacity. We conclude that chronic cortisol treatment enhances whole body Na+ and Cl influxes by promoting proliferation of branchial chloride cells. The results of correlation analysis indicate that the chloride cell is an important site of NaCl uptake in freshwater rainbow trout.  相似文献

Within 1 min of transfer from coelomic fluid to fresh water, eggs of rainbow trout (Oncorhynchus mykiss) underwent a transient loss of Na+ and K+ coupled with an elevation of the chorionic envelope. Both mechanisms were blocked by adding a monovalent cation Li+ or K+ (140 mmol·l-1) to the fresh water, but the divalent ion Mg2+ (100 mmol MgCl2·l-1) or elevating the osmotic pressure to 300 mOsmol·l-1 with glycine had no inhibitory effect. The blocking of Na+ loss occurred at external monovalent cation (LiCl) concentrations above 70 mmol·l-1. A 20-s exposure of eggs to fresh water was sufficient to trigger Na+ loss and chorion elevation, even when the eggs were subsequently transferred to fresh water containing 140 mmol LiCl·l-1. Eggs placed in a medium containing 140 mmol LiCl·l-1 and 2 mmol Ca(NO3)2·l-1 showed chorion elevation and associated Na+ loss after addition of calcium ionophore (20 mol·l-1 A.23187). Summary An isolated and perfused fish gill filament preparation has been developed to facilitate investigations on the fish gill microclimate. Measurements have been made of the concentration of sodium at the lamellar surface and in the mixed-exit water, and of the maximum interlamellar velocity. When the external water has a sodium concentration of 0.10 mmol · 1-1, water at the gill surface has a mean sodium concentration of 0.146 mmol · 1-1. Mixed-exit water, 50 m behind the perfused filament, has a mean sodium concentration of 0.118 mmol · 1-1. When the water flowing over the filament has a maximum velocity of 18 cm · s-1, the maximum velocity between secondary lamellae is 1.5 cm · s-1. These measurements have been used to support the development of a velocity-modified diffusion model which describes ion movement in the interlamellar space. The analysis and observations on the preparation suggest that substantial unstirred layers do not exist at the lamellar surfaces in this preparation. Simple diffusion to and from the tissue surfaces, in conjunction with parabolic velocity profiles within the interlamellar spaces adequately explain net sodium movements.  相似文献

Rainbow trout, Oncorhynchus mykiss, acclimated to 33% sea water (12 mg·ml-1 salinity) experienced significant (10 meq·1-1) increases in plasma [Na+] and [Cl-] within 5 h of exposure to 6.3 mol copper·1-1 indicating severe impairment of branchial ionoregulatory capacity. All plasma ion levels subsequently stabilised once the transbranchial [Na+] gradient was reduced to zero. The similar ionic strength of the external medium and their body fluids appeared to protect trout maintained in 33% sea water from further ionoregulatory stress and any secondary physiological disturbances during exposure to copper. Despite three- and fourfold greater transbranchial [Na+] and [Cl-] gradients, trout acclimated to full-strength sea water (35 mg·ml-1 salinity) suffered no major changes in plasma Na+, Cl-, K+, or Ca2+, blood gases or haematology during 24 h exposure to 6.3 mol copper·1-1. This reduction in toxicity in full strength sea water cannot be explained by differences in copper speciation. We suggest that during acute exposure to waterborne copper, active NaCl extrusion is unaffected due to the basolateral location of the gill Na+/K+-ATPase, but that ionoregulatory disturbances can occur due to gill permeability changes secondary to the displacement of surface-bound Ca2+. However, in full strength sea water the three-fold higher ambient [Ca2+] and [Mg2+] appear to be sufficient to prevent any detrimental permeability changes in the presence of 6.3 mol copper·1-1. The acclimation of the clawed toad Xenopus laevis to hyperosmotic solutions of NaCl (balanced solution of sea salt), urea or mannitol was studied. The animals could not be acclimated to salt solutions more concentrated centrated than 400 mosm·l-1. Urea was tolerated till 500 mmol·l-1. Plasma osmolality was always hyperosmotic to the environmental solution, but with diminished osmotic gradient at the highest tolerated solutions. Plasma urea concentration approached 90 mmol·l-1, similar in the three solutions of acclimation. Urine volume was very small under all conditions. Serum aldosterone and corticosterone did not differ significantly, although there was a slight tendency towards lower aldosterone in the NaCl solution. In vivo water uptake in tap water acclimated animals was very small, and was higher in the other groups. Only the salt- and urea-acclimated, but not the tap water and mannitol-acclimated groups responded with a clear increase following injection of oxytocin or theophylline. In vitro urea fluxes were similar and invariable in both directions under all conditions. No significant effect of theophylline was observed. Sodium transport measured by the short-circuit technique in vitro was lower in salt- and mannitol-acclimation conditions, and was stimulated significantly under all conditions in response to serosal oxytocin or theopylline. It is concluded that Xenopus laevis can osmoregulate at a limited range of external solutions. Summary Intermoult crayfish (Astacus astacus) were exposed to acid (pH 4), soft water ([Ca++]=100 mol·l–1) in the absence and presence of aluminium (25 mol·l–1) for variable time periods (up to 21 days) in order to assess the consequences for acid-base and electrolyte balance and haemolymph gas transport. Haemolymph osmolality and concentration of major ions decreased drastically and to a similar extent in acid and acid-aluminium water. Muscle tissue ion concentrations were, however, regulated at an almost constant level. A severe metabolic acidosis was gradually developed, attaining a haemolymph metabolic acid load of 6–7 mequiv·l–1 after 12–21 days. The acidosis was partially compensated by ventilatory means, with the postbranchial haemolymph PCO2 decreasing earlier in acidaluminium-exposed than in acid-exposed specimens. Hyperventilation seemed to be a direct acid-base regulatory response, since the rise in postbranchial PCO2 had only minimal influence on haemolymph O2 transport. The Bohr effect of Astacus astacus haemocyanin was low (log P50/GdpH=-0.24), and the mean P50 only increased from 15 to 19 mmHg after 21 days of acid exposure. The decrease in O2 affinity with decreasing pH was accompanied by a decrease in the cooperativity of O2 binding. The haemolymph haemocyanin concentration was not affected by acid and acid-aluminium exposure, but decreased after 21 days due to starvation. Muscle tissue aluminium concentrations were unaffected, whereas gill tissue concentrations increased in acid-aluminium exposed crayfish, most likely due to accumulation of aluminium on the gill surface. Mortality was low, and an internal hypoxia and lactacidosis was not developed in either of the experimental groups.
