Eight weeks of heat exposure (34±0.5°C) in sham-orchiectomized rats leads to an increase of body temperature, slowing of body growth rate, and decrease of serum corticosterone level, as compared with animals maintained at 21±2°C. Orchiectomy decreases body temperature, slows growth rate, and increases plasma corticosterone concentration both in control and heat exposed animals. Testosterone administration reverts these parameters to initial values. We conclude that testosterone plays a role in the regulation of heat balance in male rats.  相似文献

Airways inflammation and epithelial injury induced by chronic ozone (O(3)) in genetically mast cell-deficient mice (Kit(W)/Kit(W-v)) were compared with those in mast cell-sufficient mice (+/+) and Kit(W)/Kit(W-v) mice repleted of mast cells (Kit(W)/Kit(W-v)-BMT). Mice were exposed to 0.26 ppm O(3) 8 h/day, 5 days/wk, for 1-90 days. Background was 0.06 ppm O(3). Age-matched mice were exposed to filtered air for O(3) controls. Reversibility of lesions was evaluated 35 days after exposure. Compared with Kit(W)/Kit(W-v), O(3) caused greater increases in lavageable macrophages, epithelial cells, and polymorphonuclear leukocytes in +/+ and Kit(W)/Kit(W-v)-BMT mice. O(3) also caused lung hyperpermeability, but the genotypic groups were not different. Cells and permeability returned to air control levels after O(3). O(3) induced lung cell proliferation only in +/+ and Kit(W)/Kit(W-v)-BMT mice; proliferation remained elevated or increased in +/+ and Kit(W)/Kit(W-v)-BMT mice after O(3). Greater O(3)-induced cell proliferation was found in nasal epithelium of +/+ and Kit(W)/Kit(W-v)-BMT mice compared with Kit(W)/Kit(W-v) mice. Results are consistent with the hypothesis that mast cells affect airway responses induced by chronic O(3) exposure.  相似文献

The effects of high helium pressure on the subsequent acquisition of spatial memory were studied in male rats. Thirty-two rats were exposed to 65 ATA helium-oxygen pressure for 4.2 days, decompressed (total time in chamber 5 days), and then tested in an eight-arm radial maze. Thirty-two control rats were exposed in the chamber to 1 ATA air. Each rat had 20 sessions in the maze (2 sessions/day for 10 days), and the number of correct (visiting an arm not previously visited to obtain the reward pellet) and incorrect choices (visiting a previously visited arm) were recorded. Statistical analysis showed that the rats exposed to 65 ATA performed significantly better than 1-ATA controls during the first 8 of 20 sessions. This effect was most pronounced in sessions 5-8. Results for sessions 9-20 showed that the pressure-treated rats still made more correct choices but to an extent that did not always reach statistical significance. Possible explanations include the pressure-treated rats performing better because of hunger after a lower food consumption at pressure. Alternatively, pressure itself may enhance proposed mechanisms of spatial memory such as long-term potentiation.  相似文献

Cholesterol, 1, which is present in both the lung lining fluid and cell membranes of lung tissue, reacts with ozone in aqueous systems to give 3 beta-hydroxy-5-oxo-5,6-secocholestan-6-al (2) as the major product. Reaction of 2 with 2,4-dinitrophenyl hydrazine (DNPH) in aqueous solutions, liposomes or lung extracts affords the anti and syn DNPH derivatives of 2 (3b and 3c) and of the rearrangement product 3,5-dihydroxy-B-norcholestane-6-carboxaldehyde (3a). These derivatives also are detected in lung tissue extracts from rats exposed to 1.3 ppm ozone for 12 hr.  相似文献

The hemodynamic response to maximal exercise was determined in sedentary and trained rats with a chronic myocardial infarction (MI) produced by coronary artery ligation and in rats that underwent sham operations (SHAM). Infarct size in the MI groups of rats comprised 28-29% of the total left ventricle and resulted in both metabolic and hemodynamic changes that suggested that these animals had moderate compensated heart failure. The training regimen used in the present study produced significant increases in maximal O2 uptake (VO2max) when expressed in absolute terms (ml/min) or when normalized for body weight (ml.min-1.kg-1) and consisted of treadmill running at work loads that were equivalent to 70-80% of the animal's VO2max for a period of 60 min/day, 5 days/wk over an 8- to 10-wk interval. This training paradigm produced two major cardiocirculatory adaptations in the MI rat that had not been elicited previously when using a training paradigm of a lower intensity. First, the decrement in the maximal heart rate response to exercise (known as "chronotropic incompetence") found in the sedentary MI rat was completely reversed by endurance training. Second, the downregulation of cardiac myosin isozyme composition from the fast ATPase V1 isoform toward the slower ATPase (V2 and V3) isoforms in the MI rat was partially reversed by endurance training. These cardiac adaptations occurred without a significant increase in left ventricular pump function as an increase in maximal cardiac output (Qmax) and maximal stroke volume (SVmax) did not occur in the trained MI rat.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

This study was the first to investigate the direct effects of anomalous concentrations of ozone mediated by summer heat waves on seed germination in alpine plants. During germination, the seeds were exposed to three peaks of O3 concentration (125 ppb for 5 and 10 days; 185 ppb for 5 days), derived from measurements taken close to the species growing site. High O3 concentration delayed the first germination time, increased the mean germination time, and reduced the germination percentage during and immediately after the treatment, but, in most cases, effects were weak and had almost vanished three weeks after the treatments. In few cases, chronic exposure to O3 (125 for 10 days’ treatment) enhanced seed germination compared to the control, suggesting that ozone may induce antioxidant and DNA-repair mechanisms or dormancy-breaking effects in hydrated seeds. Although seed mortality increased during O3 treatments in four species, the effect of O3 on seed germination is mostly limited to the period of exposure, indicating that it is unlikely to produce permanent negative effects on seeds, during the germination phase. Our results show that the direct effect of O3 on seeds of alpine plants may have minor impacts on plant reproductive performance during seed germination.  相似文献

The facility consists of a 12 × 11.5 × 2.4 m high room containing six sets of exposure apparatus and the other equipment necessary to maintain a pathogen-free system. The apparatus sets produced 5 mT (rms), 0.5 mT, or a sham exposure. The apparatus was arranged in the room to minimize the fringing field of the 5 mT set at the sham position. Each set was 3.85 × 1.80 × 0.66 m in outside dimension, containing 24 cages in the magnetically homogeneous region. The apparatus was designed using Harvey's figure-eight-configuration and generated a horizontal sinusoidal alternating field. In order to save electric power, the coil of the apparatus constituted a 50 Hz LC resonance circuit with a condensor bank to which electric power was supplied to compensate losses. Magnetic flux density was kept constant by controlling the coil current. Although mild steel was used in the skeleton of the building, the fringing flux at the sham was as low as 0.1 to 1 μT. Stainless steel was used for ventilating ducts, racks for the cages, cage covers, feeder baskets, and watering nozzles. The homogeneity of the field was measured to be ± 10% in the animal residence area, and food and water consumption was found to be unaffected by the field. At 5 mT, the coil current was 370 A, and the hollow coil was cooled by a stream of 20°C water to prevent both heat and dew on the coil surface. Vibration and acoustic noise was prevented by fiber reinforced plastic framework of the coil. High harmonic distortion was not observed at the output terminal of the coil driver. 目的: 探讨不同浓度臭氧急性暴露对大鼠肺部细胞的遗传毒性的影响。方法: 36只wistar大鼠随机分为对照组(过滤空气暴露)、臭氧暴露组(0.12 ppm、0.5 ppm、1.0 ppm、2.0 ppm、4.0 ppm)共6组,每组6只。以不同浓度的臭氧对大鼠进行动态染毒4 h后,取肺组织并分离单细胞,采用酶联免疫吸附法检测8-羟基脱氧鸟苷(8-OHdG),利用彗星实验、微核试验和DNA-蛋白质交联实验进行DNA和染色体损伤分析。结果: 与对照组相比,肺组织中8-OHdG含量从臭氧暴露浓度为0.12 ppm起即显著增加,在0.5 ppm时达到最高值。随着臭氧暴露浓度升高,彗星拖尾率逐渐上升,且存在明显的剂量-效应关系;DNA-蛋白质交联率有先升高后下降的趋势,且在2.0 ppm时达到最大值;而肺部细胞微核率尽管呈现出上升趋势,但与对照组相比无显著性差异。结论: 急性臭氧暴露在较低浓度(0.12 ppm)时即可导致大鼠肺部细胞的DNA损伤;而在较高浓度(4 ppm)时却未见显著的染色体损伤。  相似文献

The purpose of this investigation was to examine cardiac function and biochemistry in spontaneously diabetic BB rats, a strain in which diabetes occurs spontaneously and closely resembles insulin-dependent diabetes in humans. The study involved two groups: nondiabetic littermates of BB rats and BB diabetic rats treated daily with a very low insulin dose such that the rats were severely hyperglycemic and hyperlipidemic. The hearts from these two groups were isolated and heart function (using isolated perfused working hearts) and biochemistry were examined 6 weeks after the onset of diabetes. BB diabetic rats exhibited a lower calcium-stimulated myosin ATPase activity and depressed left ventricular developed pressure, cardiac contractility, and ventricular relaxation rates compared with BB nondiabetic littermates. These results suggest that the chronically diabetic state in the BB rat produces cardiac changes similar to those demonstrable after chemical diabetes induced by alloxan or STZ, or that seen during human diabetes mellitus.  相似文献

Summary Cottonwood saplings were exposed to ozone or charcoal-filtered air in a closed chamber. After leaf abscission, decomposition of individual leaf discs was measured in containers of stream water. Exposure of plants to 200 ppb ozone for 5 h caused early leaf abscission and changes in the chemical composition of leaves at time of abscission. Early-abscised leaves from O3-exposed plants had higher nitrogen, but decomposed more slowly than leaves from control plants. Leaves from O3-exposed plants that abscised at the normal time had lower nitrogen content and lower specific leaf mass than control leaves, but decomposed at the same rate as leaves from control plants. The results imply that O3 exposure can alter fundamental processes important to the functioning of detritus-based aquatic ecosystems.  相似文献

Endosulfan was studied for its effect on rat testicular toxicity in relation to the enzymes of androgen biosynthesis, viz. 3 beta-hydroxysteroid dehydrogenase (EC 1.1.1.145, 3 beta-HSD) and 17 beta-hydroxysteroid dehydrogenase (EC 1.1.1.64, 17 beta-HSD); cytosolic conjugation enzyme, glutathione-S-transferase (EC 2.5.1.18); and testicular as well as serum testosterone levels at the dose levels of 2.5, 5.0, 7.5 and 10 mg/kg body weight fed orally for 7 and 15 days. Organ and body weights of the treated animals did not change significantly, however, the testicular protein contents were found to be increased appreciably after 7 days treatments. The activity profile of cytosolic conjugation enzyme showed much remained low during 7 days treatment, however, the two steroidogenic enzymes showed much individual variations in response to endosulfan treatments. An overall varied response with respect to testosterone biosynthesis and its secretion to serum was observed suggesting nevertheless, a profound hormonal imbalance caused by this insecticide to male gonads on short term chronic exposures.  相似文献

Adult and sexually naive male rats were individually, in repeated sessions, put into a box the floor of which had beforehand been scent-marked by oestrous females. The males of the control group were put into a box which had not been scent-marked. Although the males exposed to scent-marks showed, in two performed tests, significantly more precopulatory activities towards the passively receptive female, they were not able to initiating copulation with her. They initiated copulatory behaviour only in the test when their partner was a female exhibiting ritualized dartings.  相似文献
