Ectomycorrhizas of Boletus aereus, Boletus edulis, and Boletus reticulatus were synthesized with Cistus sp. under laboratory conditions using synthesis tubes filled with a mixture of sterilized peat-vermiculite and nutrient solution. The fungal strains isolated from sporocarps were identified by molecular techniques. The inoculated seedlings were grown for 4–5 months. The ectomycorrhizas formed were described based on standard morphological and anatomical characters. The three ectomycorrhizas described were very similar, with white monopodial-pinnate morphology, a three-layered plectenchymatous mantle on plan view and boletoid rhizomorphs.  相似文献

Ectomycorrhizal (ECM) hyphal communities have not been well characterized. Furthermore, there have been few studies where the ECM hyphal community is compared to fungi detected as sporocarps or ECM-colonized root tips. We investigated fungi present as hyphae in a well-studied California Quercus–Pinus woodland. Hyphal species present were compared to those found as sporocarps and ECM root tips at the same site. Hyphae were extracted from root-restrictive nylon mesh in-growth bags buried in the soil near mature Quercus douglasii, Quercus wislizeni, and Pinus sabiniana. Taxa were identified using PCR, cloning, and DNA sequencing of internal transcribed spacer and 28s rDNA. Among the 33 species detected, rhizomorph-forming ECM fungi dominated the hyphal community, especially species of Thelephoraceae and Boletales. Most fungi in soils near Quercus spp. and P. sabiniana were ECM basidiomycetes, but we detected two ECM ascomycetes and three non-mycorrhizal fungi. Many ECM species present as hyphae were also previously detected at this site as sporocarps (18%) or on ECM root tips (58%). However, the hyphal community was mostly dominated by different taxa than either the sporocarp or ECM root communities.  相似文献

Non-native tree species have been widely planted or have become naturalized in most forested landscapes. It is not clear if native trees species collectively differ in ectomycorrhizal fungal (EMF) diversity and communities from that of non-native tree species. Alternatively, EMF species community similarity may be more determined by host plant phylogeny than by whether the plant is native or non-native. We examined these unknowns by comparing two genera, native and non-native Quercus robur and Quercus rubra and native and non-native Pinus sylvestris and Pinus nigra in a 35-year-old common garden in Poland. Using molecular and morphological approaches, we identified EMF species from ectomycorrhizal root tips and sporocarps collected in the monoculture tree plots. A total of 69 EMF species were found, with 38 species collected only as sporocarps, 18 only as ectomycorrhizas, and 13 both as ectomycorrhizas and sporocarps. The EMF species observed were all native and commonly associated with a Holarctic range in distribution. We found that native Q. robur had ca. 120% higher total EMF species richness than the non-native Q. rubra, while native P. sylvestris had ca. 25% lower total EMF species richness than non-native P. nigra. Thus, across genera, there was no evidence that native species have higher EMF species diversity than exotic species. In addition, we found a higher similarity in EMF communities between the two Pinus species than between the two Quercus species. These results support the naturalization of non-native trees by means of mutualistic associations with cosmopolitan and novel fungi.  相似文献

Post-fire Pezizales fruit commonly in many forest types after fire. The objectives of this study were to determine which Pezizales appeared as sporocarps after a prescribed fire in the Blue Mountains of eastern Oregon, and whether species of Pezizales formed mycorrhizas on ponderosa pine, whether or not they were detected from sporocarps. Forty-two sporocarp collections in five genera (Anthracobia, Morchella, Peziza, Scutellinia, Tricharina) of post-fire Pezizales produced ten restriction fragment length polymorphism (RFLP) types. We found no root tips colonized by species of post-fire Pezizales fruiting at our site. However, 15% (6/39) of the RFLP types obtained from mycorrhizal roots within 32 soil cores were ascomycetes. Phylogenetic analyses of the 18S nuclear ribosomal DNA gene indicated that four of the six RFLP types clustered with two genera of the Pezizales, Wilcoxina and Geopora. Subsequent analyses indicated that two of these mycobionts were probably Wilcoxina rehmii, one Geopora cooperi, and one Geopora sp. The identities of two types were not successfully determined with PCR-based methods. Results contribute knowledge about the above- and below-ground ascomycete community in a ponderosa pine forest after a low intensity fire.  相似文献

This present study includes twelve species that represent the Ficus genus, namely; aspera, carica, tinctoria subsp. gibbosa, hirta, hispida, neriifolia, palmata, pumila, racemosa, septica, sur, and sycomorus, belonging to the Moraceae family. The species samples were collected from various locations in Egypt. The study focused on the anatomical and molecular characteristics of mature foliage leaves. Since the identification and classification of taxa are highly dependent on the anatomical features of leaves, the anatomical characteristics were recorded in the form of a comparison between the examined plants in the data matrix. This study aims to contribute to the identification of the studied species based on the anatomical details of the matured leaves. Anatomical characterization includes the variations in upper and lower epidermal layers that are covered by a thin or thick cuticle; the number of palisade and spongy layers; crystals; secretory elements; lithocysts; the midrib zone has parenchyma associated with mechanical tissue, vascular system, and investigation of trichomes; on the other hand, in the current study, the phylogenetic analysis was conducted by using the ITS and 5.8 S sequences. From the analysis of all the available data, it could be stated that there is an overall agreement with the anatomical character dendrogram.  相似文献

Seven isolates ofTorulomyces from Asian and Australian soil samples were studied in comparison with known taxa of the genus and withMonocillium indicum, the type species ofMonocillium. Three new species,Torulomyces parviverrucosus, T. laevis, andT. ovatus, are described, andT. brunneus is described as a new combination. Conidial characteristics, especially their shape and surface structure, are useful taxonomic criteria for distinguishing species ofTorulomyces. In most ectomycorrhizal (EM) community studies involving molecular identification methods there is a poor correspondence between fungi that appear dominant as sporocarps and those that appear dominant on EM roots and the species richness belowground is higher than that above ground. As a consequence, many fungi from root tip samples remain unidentified. In most studies, genetic data from multiple samples of an EM morphotype collected from various sampling locations are compared to genetic data from one to a few sporocarps of each species for identification purposes. The mismatch between above- and belowground species richness may be influenced by these different sampling efforts. To address this, intra-specific variation in the ITS region first investigated in Kårén et al. (1997) is revisited here, but at a spatial scale in which variation is expected to be low. Sporocarps were collected across a 7 km region of the Oregon Dunes National Recreation Area in western North America. ITS–RFLP data are presented for 3-18 sporocarps from each of 44 EM species in 18 genera. A total of 311 sporocarps were analyzed. Fifty-three ITS–RFLP types were observed. Of the 44 species, 38 (86% of total) yielded a single, species specific, RFLP type. No 2 species had the same RFLP type. Polymorphic ITS–RFLP types were observed in six species (14%). The following three species had two ITS-RFLP types with one type dominating: Inocybe lacera, Laccaria proxima, and Rhizopogon subcaerulescens. The following three species had three RFLP types with no type dominating: Laccaria laccata, Lactarius deliciosus, and Tricholoma flavovirens. A phylogenetic analysis of ITS sequences in Tricholoma revealed that two of the RFLP types in T. flavovirenswere apparently the result of intra-specific variation, while the third RFLP type was likely a cryptic species. All the other RFLP types observed in Tricholoma represented unique phylogenetic species. These results suggest that ITS–RFLP data from single samples (sporocarp or EM) are robust for characterizing most of the species at this scale. However, restriction endonucleases detect a limited amount of existing nucleotide variation and thus have limited value to detect cryptic species. Therefore, additional analyses of sequence data should be added to the RFLP matching technique to identify unknown RFLP types. These data also suggest that polymorphic RFLP types within species do not adequately explain the mismatch between above- and belowground views of EM species richness.  相似文献

Identification of the fungi forming ectomycorrhiza is still a great challenge. Ectomycorrhizae of Picea abies, collected in southwest Germany during several years and described as morphotypes, were identified using LSU and ITS sequences. To this the fungal sequences amplified from the mycorrhizae were compared with sequences from identified sporocarps. The fungal partner of Piceirhiza gelatinosa was identified as Hygrophorus olivaceoalbus, the fungal partner of Piceirhiza rosa-nigrescens was identified as Dermocybe cf. semisanguinea, and the fungal partner of a white mycorrhiza, described here for the first time, belongs to the Hebeloma velutipes group (Hebeloma crustuliniforme complex). Identification to genus level was possible for Piceirhiza lanuginosa where a Cortinarius-species is the fungal partner. A Tomentella-species forms a newly described light-brown mycorrhiza. Identification to family and to order-level was achieved for a milky-dull silvery mycorrhiza (Cortinariaceae), and Piceirhiza globulifera (Aphyllophorales), respectively. Ten samples of white, slightly bent mycorrhizae were formed by 8 different Cortinarius-species, including among others Cortinarius traganus, C. delibutus, and C. brunneus. The fungal partners of several brown, smooth mycorrhizae displaying only a Hartig net are Ascomycetes, among these are Wilcoxina cf. mikolae and Hymenoscyphus cf. ericae.  相似文献

Intraspecific variability in root colonization, extraradical growth pattern, and survival after cold storage of Lactarius deliciosus isolates was determined in pure culture conditions using Pinus pinaster as a host plant. The ectomycorrhizal ability of L. deliciosus at 30, 45, and 60 days from inoculation was highly variable among isolates and was negatively correlated to the age of the culture (time elapsed from isolation). The formation of rhizomorphs was related to colonization ability, but no relationship was found between colonization and formation of extraradical mycelium. The final colonization achieved at 60 days from inoculation was not related to the tree species under which the sporocarps were collected. However, isolates from sporocarps collected under P. pinaster colonized more rapidly the seedlings than those collected under other pine species. The climatic range of the sporocarps from which the isolates were obtained (maritime vs. continental) was not related to the formation of mycorrhizas at 60 days from inoculation. However, isolates from sporocarps collected from a maritime climate area colonized more rapidly the P. pinaster seedlings than those collected from a continental zone. Tolerance to cold water storage of L. deliciosus was also isolate dependent. Growth revival in agar was obtained from most of the isolates after 28 months of cold storage at 4°C, but only 10 out of 29 isolates showed unaffected growth. The ITS rDNA alignment of all the L. deliciosus isolates showed a low variability with identities over 99%. Most of the variation was detected in the ITS1 region and consisted in single nucleotide changes and/or punctual indel mutations. The number of base differences per sequence from averaging over all sequence pairs was 1.329, which is in the low range when compared with other ectomycorrhizal species. No ITS pattern due to geographical origin of the isolates could be discerned.  相似文献

Three new species of Architectonicidae Heliacinae,Heliacus (Torinista) chonos, Heliacus (Grandeliacus) antu andSolatisonax bieleri, are described. These include the first reports ofGrandeliacus andSolatisonax from Chile.Solarium australe Philippi, 1887 is an earlier name forHeliacus (Torinista) bahamondei Frassinetti & Covacevich, 1981, but is a junior homonym ofS. australe Philippi, 1849. Type material and new specimens of the five previously described architectonicid species from Chile are described and figured for comparison. Together with other gastropod taxa from the same deposits, Architectonicidae provide evidence for tropical to subtropical water temperatures in central Chile during the early Miocene.  相似文献

Suillus grevillei in two Larix kaempferi stands was determined over two years by inter-simple sequence repeat (ISSR) polymorphism analysis using primers, (GTG)5, (GCC)5 and (GACA)4. Thirty-five genets were identified from 67 sporocarps at the older stand (stand A in which the distribution of S. grevillei genet in 1997 was analyzed previously) in 1998, and 14 genets from 52 sporocarps at the younger stand (stand B) in 1997 and 1998. The characteristics of S. grevillei genets in stand A in 1998 were similar to those in 1997. A single genet was represented by 1.8 and 3.7 sporocarps on average in stands A and B, respectively. In stand A, 42 out of 61 genets, i.e., about 70% were represented by individual sporocarps compared to five out of 14 genets, i.e., about 35% in stand B. The largest and the average genet sizes was 6.8 m and less than 1 m in stand A, and 11 m and 2.3 m in stand B, respectively. A t-test showed the genet size in stand A to be significantly smaller than that in stand B. The above results indicate that the smaller genets of the S. grevillei population in stand A might be due to environmental conditions not genetic traits specific to this species. Observations over two years showed that although some genets formed sporocarps in both 1997 and 1998, many formed sporocarps only in one of the two years. Emerging positions of sporocarps in 1997 and 1998, which belonged to the same genet, were similar but not identical, about 2 m apart, suggesting mobility in the subterranean parts of ECM fungal genets. A new caprellid species, Caprellinoides singularis, is described and illustrated based on the material collected on the Polarstern Cruise ANT XVII/3 from the Branfield Strait. The most striking characteristic of this species is the presence of bilobed gills on pereonites 3 and 4. The genus Caprellinoides is revised. Caprellinoides antarctica Schellengerg, 1926 and Caprellinoides spinosus Barnard, 1930 are considered junior synonyms of Caprellinoides tristanensis Stebbing, 1888 and Caprellinoides mayeri (Pfeffer, 1888), respectively. The new species, C. singularis, is compared with the remaining species in the genus Caprellinoides: C. tristanensis and C. mayeri, which are illustrated in detail.  Ectomycorrhizal types of black alder [Alnus glutinosa (L.) Gaertn.] collected over a 3-year period within an alder forest were characterised by morphological and anatomical features. Of the total of 16 types, 14 are described for the first time in this paper. Eight identified types belong to the genera Russula, Lactarius, Naucoria, and Cortinarius, while eight further types remained unidentified. In some cases, similarities of mantle features indicate relationships to identified mycorrhizas. Mycorrhizas of Naucoria escharoides and N. subconspersa were not distinguished. Two unidentified mycorrhizal types exhibited hyphal mantle structures very similar to these Naucoria species. Within the genus Cortinarius, mycorrhizas of C. cf. helvelloides were easily distinguished from all other Cortinarius-like mycorrhizas described on Alnus, which in general showed little anatomical variation. Two further unidentified mycorrhizas, "Alnirhiza lilacina" and "A. violacea", probably also belong to Cortinarius. The ectomycorrhiza of Russula pumila was the only identified type within the genus Russula, but the unidentified type "Alnirhiza cremicolor" also likely belongs to this genus. Three Lactarius species were present in the experimental plot. Two species (L. obscuratus and L. omphaliformis) had indistinguishable mycorrhizal types, but were easily differentiated from the mycorrhizas of L. lilacinus, which caused intracellular penetration of Hartig net hyphae into epidermal and cortical cells. All other mycorrhizal types of black alder exhibited a paraepidermal Hartig net without penetration of root cells. Two unidentified mycorrhizal types "Alnirhiza atroverrucosa" and "A. cystidiobrunnea", already described from North American Alnus rubra as unnamed morphotypes, showed no similarity to identified mycorrhizas. The beetle fauna of 299 sporocarps of the bracket fungus Fomitopsis pinicola in a 200 ha spruce forest in southeastern Norway was investigated in relation to sporocarp, tree and forest variables. The sporocarps contained 36 species of beetles, of which six species are on the Norwegian Red List. Of 12,373 individual beetles collected, 91% were Cis glabratus. Plots of species accumulation curves suggested that there may be more than 60 beetle species present in F. pinicola in the area, but that probably all the specialist Ciidae were found. The major factor influencing beetle diversity turned out to be the level of dead wood at and in the vicinity of the sampling site, with a higher number of species per unit volume of sporocarps in areas with high levels of dead wood. There were also significantly more red-listed species in those areas. Analysing the species occurrence with stepwise logistic regression, we show preferential habitat selection of the six most abundant species of Ciidae. Conservation of beetles associated with bracket fungi using amount of dead wood as a surrogate measurement is discussed.  相似文献

Trichodesmium Ehrenberg species were collected in the Caribbean Sea, Sargasso Sea, and coastal areas of Tanzania (Indian Ocean). The specimens were divided into five species on the basis of morphometric characters such as cell dimensions and colony formation: T. tenue Wille, T. erythraeum Ehrenberg, T. thiebautii Gomont, T. hildebrandtii Gomont, and T. contortum Wille. In addition, Trichodesmium sp., a spherical colony of uncertain taxonomic position was examined. The cell structure of each species was investigated by means of light, scanning electron, and transvnission electron microscopy. Particular attention was paid to the presence and ultrastructural arrangement of gas vacuoles and glycogen fiber clusters (GFCs). This resulted in identification of two major groups of species: 1) T. tenue, Trichodesmium sp. with spherical-shaped colonies, and T. erythraeum with GFCs and more or less localized gas vacuoles; and 2) T. thiebautii, T. hildebrandtii, and T. contortum lacking GFCs and with gas vacuoles spread at random. The species within each group were further characterized with respect to the dimension of the gas vesicles, cylindrical bodies, scroll bodies, and a new cellular inclusion body, Differences in colony formation and cell dimensions correlated with specific ultrastructural characters in five of the six forms. This is the first ultrastructural study comparing different forms of Trichodesmium sampled at geographically remote areas and shows that one species appears identical regardless of the sampling site. Some of the species had not been investigated earlier, and probably more species are to be identified and analyzed.  相似文献

Using ectomycorrhizal root tip morphotyping (anatomical and morphological identification), molecular analysis (internal transcribed spacer region amplification and sequencing), and fruitbody sampling, we assessed diversity and composition of the ectomycorrhizal fungal community colonizing juvenile Pinus pinaster Ait. under natural conditions in NW Spain. Overall, we found 15 Basidiomycetes and two Ascomycetes. Members of the family Thelephoraceae represented up to 59.4% of the samples. The most frequent species was Tomentella sublilacina followed by Thelephora terrestris, Russula drimeia, Suillus bovinus, and Paxillus involutus, while the less frequent were Pseudotomentella tristis, Lactarius subdulcis, Russula ochroleuca, and Entoloma conferendum. From October 2007 to June 2008, we sampled 208 sporocarps belonging to seven genera and nine species: Thelephora terrestris, Paxillus involutus, Suillus bovinus, Xerocomus badius, Scleroderma verrucosum, Amanita gemmata, A. rubescens, Amanita sp., and Russula sp. The species belonging to the genus Amanita, X. badius and S. verrucosum were not found on root samples. By comparing our results with a bibliographic review of papers published from 1922 to 2006, we found five genera and six species which have not been previously reported in symbiosis with P. pinaster. This is the first time that the diversity of the ectomycorrhizal fungal community associated with P. pinaster was investigated using molecular techniques. Considering that only 38% of the genera found by sequencing were found as fruitbodies, we conclude that integrating morphotyping and sporocarps surveys with molecular analysis of ectomycorrhizas is important to documenting the ectomycorrhizal fungus community. We evaluated the nutritional value of sporocarps of Rhizopogon vinicolor, a common hypogeous fungus in the coniferous forests of North America, for two small mammal species: the Californian red-backed vole (Clethrionomys californicus) and the northern flying squirrel (Glaucomys sabrinus). Although the nitrogen concentration of sporocarps was high, much of it was in non-protein form or associated with cell walls, suggesting that it may be of low nutritional value or protected from mammalian digestive enzymes. Sporocarps also had high concentrations of cell wall constituents, indicating low availability of digestible energy. When fed a diet of this fungus alone in a controlled feeding experiment both mammal species lost a small amount of body mass. Digestibilities of dry matter, nitrogen, cell wall constituents and energy from sporocarps by both species were lower than the digestibilities of other food types by other similarly sized small mammals. Red-backed voles digested the various components of sporocarps at least as well as the flying squirrels, even though they were almost six-fold smaller in body mass. This observation supports the notion that red-backed voles, like other microtine rodents, have morphological and physiological adaptations of the digestive system that are postulated to permit greater digestion of fibrous diets than predicted on the basis of body size. Despite this, our results re-affirm previous conclusions that hypogeous fungi are only of moderate nutritional value for most small, hindgut-fermenting mammals. The content of 19 metals (Al, V, Cr, Co, Ni, Cu, Zn, Ga, Rb, Sr, Zr, Nb, Cs, Ba, Ce, Pb, Th, U and Nd) was investigated in 15 edible species of phylum Basidiomycota collected in an area with quartzite acidic soils in a province of the central Spain. The study explores the differences in metal accumulation in relation to fungal species, and the results were related to metal content in soil through the determination of bioconcentration factors. Regarding the highest concentrations, Zn, Al, Cu and Rb were the metals more accumulated in the sporocarps. Notable concentrations were also found in Sr, Zr, Ba, Cs and Ce. The major bioconcentration factors were found for Cu and Zn in sporocarps of Agaricus silvicola and Lepista nuda. Regarding the different species, Tricholoma equestre and Cantharellus cibarius were those with the greatest capacity to absorb trace elements, and in contrast, Amanita caesarea and Agaricus campestris showed the lowest values. The cluster analysis shows that there are some species with the same nutritive physiology that share similarities in the absorptive behaviour. Lactarius sanguifluus and Lactarius deliciosus, both ectomycorrhizas of the genus Pinus, are closely related, and Clitocybe gibba, L. nuda and Marasmius oreades, all of them saprobes on soil organic matter, are very close too.  相似文献
