The mode of inheritance in tetraploid cut roses   总被引:2,自引:0,他引:2  
Tetraploid hybrid tea roses (Rosa hybrida) represent most of the commercial cultivars of cut roses and form the basis for breeding programmes. Due to intensive interspecific hybridizations, modern cut roses are complex tetraploids for which the mode of inheritance is not exactly known. The segregation patterns of molecular markers in a tetraploid mapping population of 184 genotypes, an F(1) progeny from a cross of two heterozygous parents, were investigated for disomic and tetrasomic inheritance. The possible occurrence of double reduction was studied as well. We can exclude disomic inheritance, but while our observations are more in line with a tetrasomic inheritance, we cannot exclude that there is a mixture of both inheritance modes. Two novel parental tetraploid linkage maps were constructed using markers known from literature, combined with newly generated markers. Comparison with the integrated consensus diploid map (ICM) of Spiller et al. (Theor Appl Genet 122:489-500, 2010) allowed assigning numbers to each of the linkage groups of both maps and including small linkage groups. So far, the possibility of using marker-assisted selection in breeding of tetraploid cut roses and of other species with a tetrasomic or partly tetrasomic inheritance, is still limited due to the difficulties in establishing marker-trait associations. We used these tetraploid linkage maps to determine associations between markers, two morphological traits and powdery mildew resistance. The knowledge on inheritance and marker-trait associations in tetraploid cut roses will be of direct use to cut rose breeding.  相似文献

Abstract

Rose, one of the world’s most-loved and commercially important ornamental plants, is predominantly tetraploid, possessing four rather than two copies of each chromosome. This condition complicates genetic analysis, and so the majority of previous genetic studies in rose have been performed at the diploid level. However, there may be advantages to performing genetic analyses at the tetraploid level, not least because this is the ploidy level of most breeding germplasm. Here, we apply recently developed methods for quantitative trait loci (QTL) detection in a segregating tetraploid rose population (F1?=?151) to unravel the genetic control of a number of key morphological traits. These traits were measured both in the Netherlands and Kenya. Since ornamental plant breeding and selection are increasingly being performed at locations other than the production sites, environment-neutral QTL are required to maximise the effectiveness of breeding programmes. We detected a number of robust, multi-environment QTL for such traits as stem and petiole prickles, petal number and stem length that were localised on the recently developed high-density SNP linkage map for rose. Our work explores the complex genetic architecture of these important morphological traits at the tetraploid level, while helping to advance the methods for marker–trait exploration in polyploid species.

Flower color is a useful morphological marker in chickpea (Cicer arietinum L.). Inheritance of this trait was studied using two white-flowered chickpea genotypes, P 9623 and RS 11, and one blue-flowered genotype, T 39-1. The genetic constitutions of the white flower colors of P 9623 and RS 11 were different, for in an earlier study their F1 produced pink flowers. The two F1s of the crosses P 9623 x T 39-1 and RS 11 x T 39-1 also produced pink flowers. Each of the two F2 populations segregated in 9 pink:3 blue:4 white-flowered plants. These results can be explained by a three-gene model. These three independently segregating genes are probably the same as C, B, and P reported in the literature earlier. Allelic tests could not be undertaken, as the genetic stocks used in the earlier studies are not available. The genetic constitutions of the three parents and their F1s are proposed. These accessions should be useful for conducting allelic tests for determining flower color loci in chickpea and for comparative studies with field pea. The seeds of these genetic stocks are maintained at the Genetic Resources and Enhancement Program at ICRISAT and are available for research purposes on request.  相似文献

Two genetic linkage maps of tetraploid roses   总被引:11,自引:0,他引:11  
A tetraploid F2 progeny segregating for resistance to black spot, growth habit, and absence of prickles on the stem and petioles was used to construct genetic linkage maps of rose. The F1 of the progeny, 90–69, was created by crossing a black spot-resistant amphidiploid, 86–7, with a susceptible tetraploid, 82–1134. The F1 was open-pollinated to obtain 115 seedlings. AFLP and SSR markers were used to eliminate seedlings produced through cross-fertilization. The remaining progeny set of 52 F2 plants was used to study the inheritance of 675 AFLPs, one isozyme, three morphological and six SSR markers. AFLP markers were developed with three combinations of restriction enzymes, EcoRI/MseI, KpnI/MseI and PstI/MseI. Most of the markers appear to be in simplex or single-dose and segregated 3:1 in the progeny. One linkage map was constructed for each parent using only the single-dose markers. The map of 86–7 consists of 171 markers assigned to 15 linkage groups and covering more than 902 cM of the genome. The map of 82–1134 consists of 167 markers assigned to 14 linkage groups and covering more than 682 cM of the genome. In the AFLP analysis, EcoRI/MseI generated nearly twice as many markers per run than PstI/MseI. Markers developed with three restriction enzyme combinations showed a mixed distribution throughout the maps. A gene controlling the prickles on the petiole was located at the end of linkage group 7 on the map of 86–7. A gene for malate dehydrogenase locus 2 was located in the middle of linkage group 4 on the map of 86–7. These first-generation maps provide initial tools for marker- assisted selection and gene introgression for the improvement of modern tetraploid roses. Summary Vegetatively propagated plantlets of six rose cultivars were induced to flower in vitro on media containing full-strength Murashige and Skoog (MS) inorganic salts, Gamborg's B5 organic elements with 400 mg l−1 myo-inositol, and different phytohormone combinations of 6-benzyladenine (BA) with α-naphthaleneacetic acid (NAA); thidiazuron (TDZ) with NAA; and zeatin (ZT) with NAA. The most efficient flower bud induction (49.1% and 44.1%) was obtained on media supplemented with 0.5 mg l−1 (2.27 μM) TDZ and 0.1 mg l−1 (0.54 μM) NAA or 0.5 mg l−1 (2.28 μM) ZT and 0.1 mg l−1 (0.54 μM) NAA for cultivar Orange Parade. Scanning electron microscopy (SEM) showed that in vitro flower bud induction occurred mostly between 15 and 30 d in induction medium through the normal flower development processes. With TDZ and ZT as the best choice for flower induction in all six cultivars tested, different rose cultivars varied in their responses to phytohormone treatments. Our study also revealed that the total time from original culture and subculture time before flower induction were two very important factors for in vitro flower induction. Plantlets 156–561 d from original culture and subcultured for 45 d were the best for flower induction. These authors contributed equally to this work.  相似文献

The commonly found flower colors in periwinkle (Catharanthus roseus)--pink, white, red-eyed, and pale pink center--are reported to be governed by the epistatic interaction between four genes--A, R, W, and I. The mode of inheritance of an uncommon flower color, orange-red corolla and white eye, was studied by crossing an accession possessing this corolla color with a white flowered variety (Nirmal). The phenotype of the F(1) plants and segregation data of F(2) and backcross generations suggested the involvement of two more interacting and independently inherited genes, one (proposed symbol E) determining the presence or absence of red eye and another (proposed symbol O) determining orange-red corolla.  相似文献

Pickerelweed (Pontederia cordata L.) is a diploid (2n = 2x = 16), erect, emergent, herbaceous aquatic perennial. The showy inflorescences of pickerelweed make this species a prime candidate for inclusion in water gardens and aquascapes. The objective of this experiment was to determine the number of loci, number of alleles, and gene action controlling flower color (blue vs. white) in pickerelweed. Two blue-flowered and one white-flowered parental lines were used in this experiment to create S(1) and F(1) populations. F(2) populations were produced through self-pollination of F(1) plants. Evaluation of S(1), F(1), and F(2) generations revealed that flower color in these populations was controlled by 2 alleles at one locus with blue flower color completely dominant to white. Diplocarpon rosae is the causal agent of rose blackspot, one of the most severe diseases of field-grown roses. The genetics of resistance to this pathogen was investigated in crosses between tetraploid rose genotypes. The hybrid breeding line 91/100-5, which exhibits a broad resistance to all isolates tested so far, was selfed to produce an F2 population, backcrossed to the susceptible tetraploid variety ‘Caramba’ and crossed to the susceptible varieties ‘Heckenzauber’, ‘Pariser Charme’ and ‘Elina’. Infection experiments resulted in segregation ratios consistent with the presence of a single dominant resistance locus in the duplex configuration in the hybrid 91/100-5. This suggests, together with previous data on the race structure of the fungus, a “gene-for-gene” type of interaction in the pathosystem Diplocarpon/Rosa. We propose to designate this gene Rdr1, which is the first resistance gene described in the genus Rosa. The advantages and limitations of such an interaction type for future rose breeding programmes and for marker-assisted selection strategies are discussed. QTL analysis of flower and fruit traits in sour cherry   总被引:2,自引:0,他引:2  
The map locations and effects of quantitative trait loci (QTLs) were estimated for eight flower and fruit traits in sour cherry (Prunus cerasus L.) using a restriction fragment length polymorphism (RFLP) genetic linkage map constructed from a double pseudo-testcross. The mapping population consisted of 86 progeny from the cross between two sour cherry cultivars, Rheinische Schattenmorelle (RS)×Erdi Botermo (EB). The genetic linkage maps for RS and EB were 398.2 cM and 222.2 cM, respectively, with an average interval length of 9.8 cM. The RS/EB linkage map that was generated with shared segregating markers consisted of 17 linkage groups covering 272.9 cM with an average interval length of 4.8 cM. Eleven putatively significant QTLs (LOD >2.4) were detected for six characters (bloom time, ripening time, % pistil death, % pollen germination, fruit weight, and soluble solids concentration). The percentage of phenotypic variation explained by a single QTL ranged from 12.9% to 25.9%. Of the QTLs identified for the traits in which the two parents differed significantly, 50% had allelic effects opposite to those predicted from the parental phenotype. Three QTLs affecting flower traits (bloom time, % pistil death, and % pollen germination) mapped to a single linkage group, EB 1. The RFLP closest to the bloom time QTL on EB 1 was detected by a sweet cherry cDNA clone pS141 whose partial amino acid sequence was 81% identical to that of a Japanese pear stylar RNase. Safflower (Carthamus tinctorius L.) flowers are used for coloring and flavoring food and also as fresh-cut and dried flowers. The most important characteristics which contribute to the ornamental value of safflower are flower color and spinelessness. The objective of this study was to determine the inheritance mode and the number of genes controlling spininess and flower color in some Iranian genotypes of safflower. The results indicated that the existence of spines on the leaves and bracts of safflower is controlled by a single dominant gene in which the spiny phenotype was completely dominant to spineless. In some crosses, flower color was controlled by two epistatic loci each with two alleles, resulting in a ratio of 13:3 in the segregating F2 population for plants with orange and yellow flowers. Also, other mechanisms of genetic control, such as duplicate dominance and duplicate recessive types of epistasis, were observed for flower color in other crosses that led to ratios of 7:9 and 15:1 for plants with orange and yellow flowers, respectively. The results suggest that for ornamental use or in the food dying industry, genotypes with orange or yellow flowers and without spines on the leaves and bracts can be produced.  相似文献

Transmission of B chromosomes in tetraploid (2n=28) garden roses was studied. A total of 1 304 seeds were used in the experiment which were obtained from various crossings and selfings made between two 1B and four 0B parents. B chromosomes were inherited in only 51 plants out of a progeny of 143. In the progeny 1B was more common in occurrence than 2Bs. About 17% of the plants were aneuploids. In general, the b chromosomes adversely affected survival, vigour and pollen fertility of the progeny plants.This work is a part of a Ph.D. thesis of the University of London (1980).  相似文献

QTL analysis of leaf morphology in tetraploid Gossypium (cotton)   总被引:13,自引:0,他引:13  
Molecular markers were used to map and characterize quantitative trait loci (QTLs) determining cotton leaf morphology and other traits, in 180 F2 plants from an interspecific cross between a Gossypium hirsutum genotype carrying four morphological mutants, and a wild-type Gossypium barbadense. The prominent effects of a single region of chromosome 15, presumably the classical ”Okra-leaf” locus, were modified by QTLs on several other chromosomes affecting leaf size and shape. For most traits, each parent contained some alleles with positive effects and others with negative effects, suggesting a large potential for adapting leaf size and shape to the needs of particular production regimes. Twenty one QTLs/loci were found for the morphological traits at LOD≥3.0 and P≤0.001, among which 14 (63.6%) mapped to D-subgenome chromosomes. Forty one more possible QTLs/loci were suggested with 2.0≤LOD<3.0 and 0.001<P≤0.01. Among all of the 62 possible QTLs (found at LOD≥2.0 and P≤0.01) for the 14 morphological traits in this study, 38 (61.3%) mapped to D-subgenome chromosomes. This reinforces the findings of several other studies in suggesting that the D-subgenome of tetraploid cotton has been subject to a relatively greater rate of evolution than the A-subgenome, subsequent to polyploid formation. Purpose  

There is a need to assess social impacts of products along the full life cycle, not only to be able to address the “social dimension” in sustainability, but also for potentially improving the circumstances of affected stakeholders. This paper presents a case study for a social life cycle assessment (S-LCA) based on the recently published “Guidelines for Social Life Cycle Assessment of Products” developed by the United Nations Environment Programme/Society of Environmental Toxicology and Chemistry (UNEP/SETAC) working group. General aim is to “try out” the proposed method. The case study itself compares the impacts of rose production in Ecuador with the Netherlands. Furthermore, the objective is to identify differences and similarities in environmental and social life cycle modelling and both social and environmental hot spots in each of the life cycles.  相似文献

Internal heat necrosis (IHN) is a physiological disorder of potato tubers. We developed a linkage map of tetraploid potato using AFLP and SSR markers, and mapped QTL for mean severity and percent incidence of IHN. Phenotypic data indicated that the distribution of IHN is skewed toward resistance. Late foliage maturity was slightly but significantly correlated with increased IHN symptoms. The linkage map for ‘Atlantic’, the IHN-susceptible parent, covered 1034.4 cM and included 13 linkage groups, and the map for B1829-5, the IHN-resistant parent, covered 940.2 cM and contained 14 linkage groups. QTL for increased resistance to IHN were located on chromosomes IV, V, and groups VII and X of ‘Atlantic’, and on group VII of B1829-5 in at least 2 of 3 years. The QTL explained between 4.5 and 29.4% of the variation for mean severity, and from 3.7 to 14.5% of the variation for percent incidence. Most QTL detected were dominant, and associated with decreased IHN symptoms. One SSR and 13 AFLP markers that were linked to IHN were tested in a second population. One AFLP marker was associated with decreased symptoms in both populations. The SSR marker was not associated with IHN in the second population, but was closely linked in repulsion to another marker that was associated with IHN, and had the same (negative) effect on the trait as the SSR marker did in the first population. The correlation between maturity and IHN may be partially explained by the presence of markers on chromosome V that are linked to both traits. This research represents the first molecular genetic research of IHN in potato.  相似文献

The chilling and heat requirements and flowering time were studied, for 2 years, in an almond progeny from the cross between the late-flowering French selection “R1000” and the early-flowering Spanish “Desmayo Largueta”. These three temperature-dependent traits showed quantitative inheritance, although for chilling requirements and flowering time a major gene could be involved, modified by other minor genes. The results indicate that flowering time is mainly a consequence of the chilling requirements; heat requirements having a smaller effect. In agreement with the genetic findings, a significant Quantitative Trait Loci (QTL) for chilling requirements was found in G4 together with other minor QTLs in G1, G3, and G7. For heat requirements, two QTLs in G2 and G7 were identified. The results also show the high influence of temperature in the expression of the three traits and their QTL analyses. In addition, QTL analysis for flowering time allowed the identification of one significant QTL in linkage group 4 (G4) that explained most of the phenotypic variation together with other minor QTLs located in G1, G6, and G7.  相似文献

Three progenies of sour cherry (Prunus cerasus) were analysed to correlate self-(in)compatibility status with S-RNase phenotype in this allotetraploid hybrid of sweet and ground cherry. Self-(in)compatibility was assessed in the field and by monitoring pollen tube growth after selfing. The S-RNase phenotypes were determined by isoelectric focusing of stylar proteins and staining for RNase activity and, for the parents, confirmed by PCR. Seedling phenotypes were generally consistent with disomic segregation of S-RNase alleles. The genetic arrangements of the parents were deduced to be ‘Köröser’ (self-incompatible) S 1 S 4 .S B S D , ‘Schattenmorelle’ (self-compatible) S 6 S 13 .S B S B , and clone 43.87 (self-compatible) S 4 S 13 .S B S B , where “.” separates the two homoeologous genomes. The presence of S 4 and S 6 alleles at the same locus led to self-incompatibility, whereas S 13 and S B at homoeologous loci led to self-compatibility. The failure of certain heteroallelic genotypes in the three crosses or in the self-incompatible seedlings indicates that S 4 and S 6 are dominant to S B . However, the success of S 13 S B pollen on styles expressing corresponding S-RNases indicates competitive interaction or lack of pollen-S components. In general, the universal compatibility of S 13 S B pollen may explain the frequent occurrence of S 13 and S B together in sour cherry cultivars. Alleles S B and S D , that are presumed to derive from ground cherry, and S 13 , presumably from sweet cherry, were sequenced. Our findings contribute to an understanding of inheritance of self-(in)compatibility, facilitate screening of progenies for self-compatibility and provide a basis for studying molecular interactions in heteroallelic pollen.  相似文献

A homologous series of long-chain secondary alcohols in which each member is a mixture of three isomers with a hydroxyl group at 4, 5 and 6 positions has been identified for the first time in a plant wax. The isomer, having an OH-group at C5 is the prevalent one. Summary Complete flower heads of cut roses (cv. Baccara) were exposed to 14CO2 for 1–4 h. The flower tissue was able to fix CO2 via PEP carboxylase (E.C. 4.1.1.31) in the dark; various TCA products were identified in petals, ovary and anthers, including malate, aspartate, citrate, serine/glycine, glutamate and asparagine. The concentrations of these labelled products were similar in the petals and anthers, but lower in the ovary. After removal of the petals the amounts of these components were reduced in the anthers to a relatively high extent (to 1/6), whereas the amounts in the ovary increased slightly.
