The effects of 17 beta-estradiol on induction of vitellogenin synthesis and oocyte development were investigated in previtellogenic ovary of immature kuruma prawn (Marsupenaeus japonicus) incubated with Medium 199. After three days incubation of previtellogenic ovary, Vg concentrations in media containing 3.6 nM, 36.7 nM, 367 nm and 3671 nM 17 beta-estradiol were significantly (p<0.01) greater than that of Ringer solution or the pure ethanol vehicle. Furthermore, a more advanced stage of oocyte development at oil globule stage (primary vitellogenic stage), which is surrounded by round and greatly expanded follicle cells, was observed in previtellogenic ovarian pieces incubated in media containing 3.6, 36.7, 367 and 3671 nM 17 beta-estradiol. The results of these studies show that 17 beta-estradiol induces Vg synthesis and appearance of primary vitellogenic oocyte in the ovary of immature prawns.  相似文献

Starch-free fatty acid complexation in the presence of whey protein   总被引:3,自引:0,他引:3  
The effect of whey protein on starch-free fatty acid (FFA) complexation was studied in a model system composed of sorghum starch, whey protein, and different FFAs (palmitic, oleic, linoleic, and lauric acids) in a weight ratio of 20:2:1(w/w/w). Whey protein decreased the enthalpy of the melting of the starch–lipid complex by 20–30% for the FFAs except linoleic acid, and increased the reformation exothermic enthalpy by 150–350% in the DSC cooling cycle. The large difference between enthalpies upon heating and cooling in the starch–FFA sample was diminished by the addition of whey protein. X-ray diffraction data showed more pronounced crystalline order of V-type starch–FFA complexes when whey protein was present. A previously described cooling stage viscosity peak, formed due to starch–FFA–protein complexation, paralleled formation of the better defined V-type crystallite of the starch–FFA complex. Whey protein also significantly decreased the amount of starch–FFA complexation in a dilute system. The effect of protein on starch–FFA complexation was related to the formation of a three-component complex composed of starch, FFA, and protein previously identified in our laboratory.  相似文献

The aim of this study was to investigate the effects of different levels of substitution of fish oil by vegetable oils rich in oleic, linoleic and linolenic acids on gilthead seabream plasma and leukocyte fatty acid compositions and prostaglandin (PG) and leptin production. Juvenile seabream of 24 g initial body mass were fed four iso-energetic and iso-proteic experimental diets for 281 days. Fatty acid composition of plasma lipids was markedly affected by the inclusion of vegetable oils (VO). ARA (arachidonate), EPA (eicosapentaenoate) and DHA (docosahexaenoate) were preferentially incorporated into polar lipids of plasma, and DHGLA (di-homogammalinoleate) accumulated with increased vegetable oil inclusion. Dietary treatments resulted in alterations of DHGLA/ARA ratios, but not ARA/EPA. ARA-derived PGE2 production in plasma was not affected by vegetable oils, in agreement with similar eicosanoid precursor ratio (ARA/EPA) in leukocytes total lipids and plasma phospholipids among fish fed with the different dietary treatments. Feeding vegetable oils leads to a decrease in plasma EPA which in turn reduced plasma PGE3 concentration. Moreover, PGE3 was the major prostaglandin produced in plasma of fish fed fish oil based diet. Such findings point out the importance of EPA as a precursor of prostaglandins in marine fish, at least for the correct function of the blood cells, and correlates well with the predominant role of this fatty acid in immune regulation in this species. A negative correlation was found between plasma PGE2 and leptin plasma concentration, suggesting that circulating levels of leptin may act as a metabolic signal modulating PGE2 release. The present study has shown that increased inclusion of vegetable oils in diet for gilthead seabream may profoundly affect the fatty acid composition of plasma and leukocytes, specially HUFA (highly unsaturated fatty acids), and consequently the production of PGE3, which can be a major PG in plasma. Alteration in the amount and type of PG produced can be at least partially responsible for the changes in the immune system and health parameters of fish fed diets with high inclusion of VO.  相似文献

Isulin is capable of regulating cellular and metabolic processes as well as gene expression. In recent years, enthusiasm has surfaced for using insulin mimetics to study the mechanism of action of insulin. Vanadata and selenate are two compounds that have been found to mimic the action of insulin on control to blood glucose levels in vivo. Vanadata has also been shown to regulate the expression of several enzymes both in vivo, however, studies concerning selenate's ability to regulate expression have not been reported. In his study we show that administration of vanadate or selenate to streptozotocin-induced diabetic rats not only normalizes blood glucose levels similarly to insulin but also positively affects the expression of two key metabolic enzymes, glucose-6-phosphate dehydrogenase (G6PDH) and fatty acid synthase (FAS). Both G6PDH and FAS activity are significantly decreased in diabetic animals compared to non-diabetic control. Treatment of the diabetic animals with either insulin, vanadate or selenate restored both activities to about 80–90% of control. All treatment conditions exhibited activities significantly higher than those determined for the diabetic group but did not differ significantly from each other. Increases in GPDH or FAS activity are due to increases in mRNA level. Increase in both G6PDH and FAS mRNA was comparable to the observed increase in activity suggesting that regulation of expression by the mimetics occurs pretranslationally.  相似文献

This study has been undertaken to determine the effect of a diet enriched with olive oil (OO) and high-oleic sunflower oil (HOSO) on fatty acid composition of erythrocyte membrane phospholipids and blood pressure in healthy women. OO and HOSO were used as natural sources of monounsaturated fatty acids (MUFAs) in a random-order sequence over two 4-week periods with a 4-week washout period between both MUFA diets. HOSO diet resulted in significant increases in oleic [(18:1n-9) 8.6%, P < 0.001], eicosenoic [(20:1n-9) 33.3%, P < 0.05], arachidonic [(20:4n-6) 6.2%, P < 0.05], and docosapentaenoic [(22:5n-6) 56.0%, P < 0.001] acids, whereas OO diet besides increased the content of stearic acid [(18:0) 13.6%, P < 0.01] and long-chain polyunsaturated fatty acids (PUFAs) of the n-3 family (22:5n-3 and 22:6n-3), when compared with the baseline [a diet high in saturated fatty acids (SFAs) and low in MUFAs]. In contrast, there was a significant decrease in linoleic acid [(18:2n-6) 21.8%, P < 0.001] for both MUFA diets. Consistent with these data, dietary intake of OO significantly raised total PUFAs (7.2%, P < 0.05), the n-3 fatty acids (22.2%, P < 0.01) and the PUFAs/SFAs ratio (9.3%, P < 0.01), as well as decreased the ratio of cholesterol to phospholipids (26,1%, P < 0.001) versus HOSO-based diet. Interestingly, dietary OO, but not HOSO, was able to significantly reduce the systolic (3%, P < 0.05) and diastolic (4%, P < 0.05) blood pressures. Although both vegetable oils provided a similar content of MUFAs (mainly oleic acid), our findings rather indicate that OO has important benefits to modulate the fatty acid composition of membranes and the mechanisms involved in the regulation of blood pressure in human.  相似文献

落叶松毛虫Dendrolimus superans (Butler)蛹个体较大, 具有很高利用价值。为明确东北落叶松毛虫蛹中脂肪酸成分, 探讨最佳提取溶剂和提取方法的组合, 分别以正己烷、 石油醚和乙醚为提取溶剂, 结合超声波振荡萃取法、 索氏萃取法及溶剂萃取方法热浸和冷浸4种提取方法提取落叶松毛虫蛹油, 并采用毛细管色谱-质谱法分析提取物的脂肪酸种类和相对含量。结果表明: 正己烷溶剂与4种提取方法的组合中, 溶剂萃取热浸法提取率最高, 为25.60%。索氏萃取及溶剂萃取方法热浸和冷浸均检测到10种脂肪酸, 正己烷-超声波振荡萃取组合检测到9种脂肪酸。石油醚溶剂与4种提取方法的组合中, 索氏萃取提取率最高, 为29.31%, 均检测到10种脂肪酸。乙醚溶剂与4种提取方法的组合中, 溶剂萃取冷浸法提取率最高, 为29.11%, 检测到的脂肪酸种类为溶剂萃取冷浸法(13种)>索氏萃取法(12种)>溶剂萃取热浸法(11种)>超声波振荡萃取法(9种)。在检测到的总脂肪酸中, 63%以上为不饱和脂肪酸, 其含量受提取溶剂和方法的影响不大。因此, 适合东北落叶松毛虫蛹中脂肪酸提取的最佳组合为石油醚溶剂 索氏萃取法。  相似文献

Heterothermic mammals increase the proportion of polyunsaturated fatty acids (PUFA) in their body fats prior to entering torpor. Because PUFA have low melting points, it is thought that they play an important role in maintaining the fluidity of depot fats and membrane phospholipids at low body temperatures. However, PUFA are more prone to autoxidation when exposed to reactive oxygen species (ROS) during torpor and during the periodic arousals that characterize hibernation. A lack of PUFA or an excess of PUFA may constrain the use of torpor by heterothermic mammals. We performed a mixed model meta-analysis of 17 controlled-feeding studies to test the effect of dietary PUFA on the depth and expression of torpor by daily heterotherms and hibernators. We also reviewed the literature on the PUFA content of the diet and depot fats of heterothermic mammals to address two principal topics: (1) Do low dietary levels of PUFA reduce the expression of torpor under laboratory conditions and, if so, are free-ranging animals constrained by a lack of PUFA? (2) Do high dietary levels of PUFA result in a reduction in the use, depth, and duration of torpor and, if so, do free-ranging animals seek to optimize rather than maximize PUFA intake? Low-PUFA diets consistently increase the lower setpoint for body temperature and minimum metabolic rate for both hibernators and daily heterotherms. Above the lower setpoint, low-PUFA diets usually increase body temperature and metabolic rate and decrease the duration of torpor bouts and this effect is similar for hibernators and daily heterotherms. Free-ranging rodent hibernators have dietary PUFA intakes that are far higher than those of the low-PUFA diets offered in controlled-feeding experiments, so these hibernators may never experience the constraints associated with a lack of PUFA. Diets of free-ranging insectivorous bats and echidnas have PUFA levels that are less than half as high as those offered in experimental low-PUFA diets, yet they exhibit deep and extended bouts of torpor. We argue that alternate mechanisms exist for maintaining the fluidity of body fats and that high-PUFA intake may not be a prerequisite for deep and extended bouts of torpor. Four studies indicate that animals that were fed high-PUFA diets are reluctant to enter torpor and show shallower and shorter torpor bouts. Although authors attribute this response to autoxidation, these animals did not have a higher PUFA content in their depot fats than animals where PUFA was shown to enhance torpor. We suggest that these contradictory results indicate inter-specific or inter-individual variation in the ability to control ROS and limit autoxidation of PUFA. High dietary levels of PUFA will constrain the expression of torpor only when the oxidative challenge exceeds the capacity of the antioxidant defence system. Studies of diet selection indicate that insectivorous species with low dietary PUFA levels seek to maximize PUFA intake. However, herbivorous species that have access to plants and plant parts of high-PUFA content do not appear to maximize PUFA intake. These data suggest that animals attempt to optimize rather than maximize PUFA intake. The effect of PUFA should be viewed in the light of a cost-benefit trade-off, where the benefit of high-PUFA intake is an easier access to low body temperatures and the cost is increased risk of autoxidation.  相似文献

Diurnal temperature fluctuations affect ectothermic species more than endothermic taxa. We tested the effect of three fixed temperatures (20, 25 and 30 °C) and a 24 h variable (20–30 °C) on the population growth and fatty acid profiles of the common rotifer Brachionus calyciflorus. Depending on the temperature treatment, the peak population abundances of B. calyciflorus varied from 65 to 80 ind./ml, the lowest being on variable temperature range. The rate of population increase varied from 0.31 to 0.52 per day, highest being at 30 °C. There was a curvilinear relationship between the population density and the egg ratio (number of eggs/female) in all the tested temperature regimes. The egg ratio was higher (>0.6) for treatments involving fixed temperatures, but for variable temperature regime, the egg ratios were lower (<0.5). Temperature also induced changes in the fatty acid content of B. calyciflorus. While the total saturated fatty acids increased, both mono-unsaturated and poly-unsaturated fatty acids decreased with increasing temperature regime. These results have been interpreted in relation to the role of temperature (both intensity and mode of exposure) on the population growth characteristics of rotifers.  相似文献

To investigate the net tissue fatty acid deposition in response to graded levels of energy restriction and modification of diet fatty acid composition, rats were randomly assigned into four dietary groups and fed for 10 weeks diets containing 40% as energy of either fish, safflower, or olive oil, or beef tallow, consumed ad libitum or energy restricted to 85% or 68% of ad libitum intake by reducing diet carbohydrate content. An additional eight rats were killed before the diet regimen, to provide baseline data from which fatty acid deposition rates were calculated. Body weight, and heart, liver and fat mass gains were decreased with energy restriction (P<0.001). Olive oil feeding resulted in higher body weight gain (P < 0.03) than tallow feeding, whereas fish oil feeding was associated with highest (P < 0.007) liver weight and lowest (P < 0.03) fat mass gains. Energy deficit-related differences in the deposition of stearic, linoleic, arachidonic, and docosahexaenoic acids in heart and palmitic and docosahexaenoic acids in liver were dependent on the dietary oil consumed (P < 0.03). Similarly, interactive effects of restricted food intake and dietary oil type were found in the gain of palmitic, stearic, oleic, and linoleic acids in adipose tissue (P < 0.01) when expressed in relation to the amount of each fatty acid consumed. These data suggest that energy intake level can influence the deposition pattern, as well as oxidation rate, of tissue fatty acids as a function of tissue type, fatty acid structure, and dietary fatty acid composition.  相似文献

The changes in fatty acid compositions during nutritional interaction among almond Amygdalus communis Linnaeus (Rosales: Rosaceae) (host plant), lacebug Monosteria unicostata (Mulsant and Rey) (Heteroptera: Tingidae) and its predator Piocoris luridus Fieber (Heteroptera: Lygaeidae) were determined by gas chromatography and gas chromatography-mass spectrometry analyses. The fatty acid profiles of phospholipids and triacylglycerols were substantially different. Unlike the general observations for virtually most terrestrial insects, arachidonic and eicosapentaenoic acids were detected in high proportions of phospholipid fractions in both insects, especially in P. luridus. Also the almond tissues provide very little oleic acid to the herbivore diet, yet both insect species developed high proportions of this component. Our data reveals instances of specific accumulation of fatty acid biosynthesis, elongation/desaturation, and not incorporating selected fatty acids into cellular lipids.  相似文献

We have hypothesized that among algae of alpine environment there could be strains particularly rich in long chain polyunsaturated fatty acids (LC-PUFA). Indeed, the chlorophyte (Trebuxiophyceae) Parietochloris incisa isolated from Mt. Tateyama, Japan, was found to be the richest plant source of the pharmaceutically valuable LC-PUFA, arachidonic acid (AA, 20:4omega6). The alga is also extremely rich in triacylglycerols (TAG), which reaches 43% (of total fatty acids) in the logarithmic phase and up to 77% in the stationary phase. In contrast to most algae whose TAG are made of mainly saturated and monounsaturated fatty acids, TAG of P. incisa are the major lipid class where AA is deposited, reaching up to 47% in the stationary phase. Except for the presence of AA, the PUFA composition of the chloroplastic lipids resembled that of green algae, consisting predominantly of C(16) and C(18) PUFAs. The composition of the extrachloroplastic lipids is rare, including phosphatidylcholine (PC), phosphatidylethanolamine (PE) as well as diacylglyceryltrimethylhomoserine (DGTS). PC and PE are particularly rich in AA and are also the major depots of the presumed precursors of AA, l8:3omega6 and 20:3omega6, respectively.  相似文献

Rats depleted in long-chain polyunsaturated omega3 fatty acids (omega3-depleted rats) display several features of the metabolic syndrome including hypertension and cardiac hypertrophy. This coincides with alteration of the cardiac muscle phospholipid and triacylglycerol fatty acid content and/or pattern. In the present study, the latter variables were measured in the cardiac endothelium of normal and omega3-depleted rats. Samples derived from four rats each were obtained from 16 female normal fed rats and three groups of 36-40 female fed omega3-depleted rats each aged 8-9, 15-16 and 22-23 weeks. At comparable mean age, the ratio between the square root of the total fatty acid content of phospholipids and cubic root of the total fatty acid content of triacylglycerols was lower in omega3-depleted rats than in control animals. The total fatty acid content of triacylglycerols was inversely related to their relative content in C20:4omega6. Other differences between omega3-depleted rats and control animals consisted in a lower content of long-chain polyunsaturated omega3 fatty acids in both phospholipids and triacylglycerols, higher content of long-chain polyunsaturated omega6 fatty acids in phospholipids, higher activity of delta9-desaturase (C16:0/C16:1omega7 and C18:0/C18:1omega9 ratios) and elongase [(C16:0 + C16:1omega7)/(C18:0 + C18:1omega9) and C20:4omega6/C22:4omega6 ratios], but impaired generation of C22:6omega3 from C22:5omega3 in the former rats. These findings support the view that cardiovascular perturbations previously documented in the omega3-depleted rats may involve impaired heart endothelial function.  相似文献

The Lake Maggiore bondella Coregonus macrophthalmus use perivisceral adipose tissue for gonadal development (with particular evidence in females), and muscle fat as an energy source. Females use mainly saturated fatty acids (SFA) for energy purposes, while males use monounsaturates (MUFA) also. During vitellogenesis, females can also mobilize fatty acid precursors of gonadal highly unsaturated fatty acids of the ω3 family from muscle tissue, but in both sexes specific retention of docosahexaenoic acid (DHA=22: 6ω3) in muscle is evident. Landlocked shad, unlike several freshwater fishes, can utilize the liver as well as body cavity and muscle as a fat reserve depot. The perivisceral fat stored during the feeding period appears to be utilized mainly for winter energy maintenance requirements. Lipid reserves from liver and muscle are also mobilized to meet the requirements resulting from gonadal development. In particular, females seem to use liver reserves, especially 18: 1ω9, more than males, to meet the demands of ovogenesis. In the muscle, SFA are used mainly for energy purposes. During gonadal development, specific muscular and hepatic ω3 highly unsaturated fatty acids (ω3 HUFA=20: 5 + 22: 5 + 22: 6) and ω6 HUFA (20: 4+22: 4+22: 5) retention is evident in both sexes, while fatty acid precursors of ω3 and ω6 HUFA are mobilized, especially from muscle tissue.  相似文献

The objective was to characterize vitellogenin expression in the ovary and hepatopancreas, and to describe the morphometry and ultrastructure of oocytes, in the freshwater prawn Macrobrachium amazonicum at various stages of ovarian development. Five ovarian stages were defined: (I) immature, (II) maturing, (III) mature, (IV) spawned, and (V) reorganized. Ovaries and hepatopancreas were analyzed by immunohistochemistry for vitellogenin expression. Vitellogenin expression in both ovary and hepatopancreas was predominantly widespread, beginning at Stage I, peaking at Stage III, and decreasing in Stages IV and V. Characterization of the ovary included measurement of the following germ cell types: oogonia (OG), and previtellogenic (PV), early vitellogenesis (EV), advanced vitellogenesis (AV), and mature (M) oocytes. Mean ± SD diameter of OG and EV oocytes in Stages I (14.2 ± 5.5 and 119.8 ± 15.7 μm) and II (17.9 ± 4.8 and 114.3 ± 34.6 μm), respectively, were significantly different from that in Stages IV (16.6 ± 4.7 and 107.0 ± 24.6 μm) and V (14.4 ± 4.1 and 101.0 ± 25.2 μm). Both scanning and transmission electron microscopy enabled identification of EV, AV and M oocytes based on the presence of a nucleus, and the organization and distribution of yolk in the cytoplasm. In summary, vitellogenesis occurred both in the hepatopancreas and ovary, with the ovary expressing vitellogenin starting as early as Stage I. This process promoted accumulation of yolk and growth of oocytes, thus favoring sexual maturation of females. This knowledge may be applied to improve production of farmed M. amazonicum.  相似文献

In mammals, 5,8,11-eicosatrienoic acid (Mead acid, 20:3n − 9) is synthesized from oleic acid during a state of essential fatty acid deficiency (EFAD). Mead acid is thought to be produced by the same enzymes that synthesize arachidonic acid and eicosapentaenoic acid, but the genes and the pathways involved in the conversion of oleic acid to Mead acid have not been fully elucidated. The levels of polyunsaturated fatty acids in cultured cells are generally very low compared to those in mammalian tissues. In this study, we found that cultured cells, such as NIH3T3 and Hepa1–6 cells, have significant levels of Mead acid, indicating that cells in culture are in an EFAD state under normal culture conditions. We then examined the effect of siRNA-mediated knockdown of fatty acid desaturases and elongases on the level of Mead acid, and found that knockdown of Elovl5, Fads1, or Fads2 decreased the level of Mead acid. This and the measured levels of possible intermediate products for the synthesis of Mead acid such as 18:2n − 9, 20:1n − 9 and 20:2n − 9 in the knocked down cells indicate two pathways for the synthesis of Mead acid: pathway 1) 18:1n − 9 → (Fads2) → 18:2n − 9 → (Elovl5) → 20:2n − 9 → (Fads1) → 20:3n − 9 and pathway 2) 18:1n − 9 → (Elovl5) → 20:1n − 9 → (Fads2) → 20:2n − 9 → (Fads1) → 20:3n − 9.  相似文献

The heat shock response has been studied extensively, yet the molecular signals that trigger the response remain elusive. The dogma of the heat shock response contends that denatured proteins initiate the response, but evidence is accumulating to point to a more complex system in which at least more than one signal is involved in this process. Thermal stress initiates changes in cellular phospholipid membrane physical state, which when acted upon by phospholipases may release lipid mediators that could serve as triggering signals during the heat shock response. We have examined the heat shock response in freshly isolated leukocytes from the pronephros of rainbow trout (Oncorhynchus mykiss). In this study, we show that leukocytes isolated from rainbow trout acclimated to 5 or 19°C express elevated levels of heat shock protein 70 (hsp70) mRNA when heat shocked at 5°C above their respective acclimation temperature and supplementation with exogenous docosahexaenoic acid or arachidonic acid followed by heat shock enhanced levels of hsp70 mRNA. The time course for docosahexaenoic acid induced enhancement of hsp70 mRNA was accelerated compared with heat shock alone, and staurosporine inhibited the docosahexaenoic acid induced increase of hsp70 mRNA. We also provide evidence that phospholipase A2 is involved in the heat shock response.  相似文献

The potential combined effects of oleic, linoleic and linolenic acids supplementation on lactation performance and the milk fatty acid (FA) profile in dairy cows have not been well investigated. Our objective was to examine the effects of supplementation with a combination of these FA as well as the effects of removing each from the combination on lactation performance and the milk FA profile in dairy cows. Eight Holstein cows (101±11 days in milk) received four intravenously infused treatments in a 4×4 Latin square design, and each period lasted for 12 days which consisted of 5 days of infusion and 7 days of recovery. The control treatment (CTL) contained 58.30, 58.17 and 39.96 g/day of C18: 1 cis-9; C18: 2 cis-9, cis-12; and C18: 3 cis-9, cis-12, cis-15, respectively. The other three treatments were designated −C18: 1 (20.68, 61.17 and 41.72 g/day of C18: 1 cis-9; C18: 2 cis-9, cis-12; and C18: 3 cis-9, cis-12, cis-15, respectively), −C18: 2 (61.49, 19.55 and 42.13 g/day of C18: 1 cis-9; C18: 2 cis-9, cis-12; and C18: 3 cis-9, cis-12, cis-15, respectively) and −C18: 3 (60.89, 60.16 and 1.53 g/day of C18: 1 cis-9; C18: 2 cis-9, cis-12; and C18: 3 cis-9, cis-12, cis-15, respectively). Dry matter intake and lactose content were not affected by the treatments, but the milk protein content was lower in cows treated with −C18: 2 than that in CTL-treated cows. Milk yield as well as milk fat, protein and lactose yields were higher in cows treated with −C18: 3 than the yields in CTL-treated cows, and these yields increased linearly as the unsaturation degree of the supplemental FA decreased. Compared with the CTL treatment, the −C18: 2 treatment decreased milk C18: 2 cis-9 content (by 2.80%) and yield (by 22.12 g/day), and the −C18: 3 treatment decreased milk C18: 3 cis-9, cis-12, cis-15 content (by 2.72%) and yield (by 22.33 g/day). In contrast, removing C18: 1 cis-9 did not affect the milk content or yield of C18: 1 cis-9. The −C18: 2-treated cows had a higher C18: 1 cis-9 content and tended to have a higher C18: 1 cis-9 yield than CTL-treated cows. The yields of C8: 0, C14: 0 and C16: 0 as well as <C16: 0 tended to increase linearly as the unsaturation degree of the supplemental FA decreased (P=0.06, 0.07, 0.07 and 0.09, respectively). These results indicated that supplementation with C18 unsaturated FA might not independently affect the lactation performance and the milk FA profile of dairy cows.  相似文献

IntroductionLong chain omega-3 polyunsaturated fatty acids (LC n-3 PUFA) such as EPA and DHA have been shown to possess beneficial health effects, and it is believed that many of their effects are mediated by their oxygenated products (oxylipins). Recently, we have shown that serum levels of several hydroxy, epoxy, and dihydroxy FAs are dependent on the individual status of the parent FAs in a cohort of normo- and hyperlipidemic subjects. So far, the effect of an increased dietary LC n-3 PUFA intake on hydroxy, epoxy, and dihydroxy FA levels has not been investigated in subjects with mild combined hyperlipidemia.Subjects and methodsIn the present study, we compared oxylipin patterns of 10 hyperlipidemic (cholesterol >200 mg/dl; triglyceride >150 mg/ml) and 10 normolipidemic men in response to twelve weeks of LC n-3 PUFA intake (1.14 g DHA and 1.56 g EPA). Levels of 44 free hydroxy, epoxy and dihydroxy FAs were analyzed in serum by LC-MS. Additionally, oxylipin levels were compared with their parent PUFA levels in erythrocyte membranes; a biomarker for the individual PUFA status.ResultsDifferences in the oxylipin pattern between normo- and hyperlipidemic subjects were minor before and after treatment. In all subjects, levels of EPA-derived oxylipins (170–4800 pM) were considerably elevated after LC n-3 PUFA intake (150–1400%), the increase of DHA-derived oxylipins (360–3900 pM) was less pronounced (30–130%). The relative change of EPA in erythrocyte membranes is strongly correlated (r≥0.5; p<0.05) with the relative change of corresponding epoxy and dihydroxy FA serum levels. The effect on arachidonic acid (AA)-derived oxylipin levels (140–27,100 pM) was inconsistent.Discussion and conclusionsThe dietary LC PUFA composition has a direct influence on the endogenous oxylipin profile, including several highly biological active EPA- and DHA-derived lipid mediators. The shift in oxylipin pattern appears to be dependent on the initial LC PUFA status particularly for EPA. The finding that also levels of other oxylipins derived from ALA, LA or AA are modified by LC n-3 PUFA intake might suggest that at least some of the effects of EPA and DHA could be mediated by a shift in the entire oxylipin profile.  相似文献

Genes encoding Δ6 desaturase, Δ6 fatty acid elongase, and Δ5 desaturase from the alga, Phaeodactylum tricornutum, were co-expressed in Pichia pastoris to produce arachidonic acid (ARA; 20:4 Δ5, 8, 11, 14) and eicosapentaenoic acid (EPA; 20:5 Δ5, 8, 11, 14, 17). A panel of Pichia clones carrying progressively increasing copies of the heterologous gene expression cassette was created using an in vitro multimerization approach. ARA and EPA accumulated up to 0.3 and 0.1% of total fatty acids, respectively, in the recombinant P. pastoris carrying with double copies of these three heterologous genes, as compared to 0.1 and 0.05%, respectively, in the recombinant P. pastoris with single copy. Delta-6 Desaturase (D6D) catalyzes the first step of the synthesis of highly unsaturated fatty acids (HUFA) that play pivotal roles in many biological functions. The D6D expression is under feedback regulation by dietary HUFA. We co-transfected D6D promoter-reporter constructs to HepG2 cells with an expression vector of nuclear form sterol regulatory element binding protein-1c (SREBP-1c). A 90-bp region of the D6D promoter was required for the activation by SREBP-1c as well as for the suppression of the promoter activity by HUFA. The region contained two candidates of sterol regulatory element (SRE). Mutation analysis identified E-box like SRE (SRE-2) as essential for both SREBP-1c activation and HUFA suppression. SRE-2 has a core sequence of CAGCAG, and is also conserved in stearoyl CoA desatruases. Because HUFA are primarily incorporated into phospholipids (PL), our results suggest that the primary role of SREBP-1c in liver is the regulation of fatty acid supply for PL rather than for triglycerides.  相似文献
