A European strain of viral haemorrhagic septicaemia virus (VHSV) isolated from wild-caught cod Gadus morhua (H16/7/95) was shown to cause clinical disease and mortality in excess of 80% in juvenile Atlantic cod when administered by the intra-peritoneal (i.p.) route. No virus was recovered from cod cohabiting with experimentally infected fish at a ratio of 1:1, and no VHSV-associated mortality was demonstrated following immersion infection. External signs of disease in cod were the presence of exophthalmia and ascites. Virus was identified as VHSV by enzyme-linked immunosorbent assay (ELISA) and was recovered from both brain and organ pools (kidney, liver and spleen) of 100% of i.p. infected cod mortalities. Virus was also detected using an indirect immunofluorescence test on tissue imprints of kidney, liver, spleen and brain taken from moribund fish. The fact that cod were not susceptible to VHSV following waterborne exposure raises important questions surrounding the propagation, maintenance and impact of a naturally occurring reservoir of virus in the marine environment.  相似文献

A polyclonal rabbit antiserum directed against the conserved region of the Atlantic salmon antiviral Mx1 protein was used to detect the putative Atlantic cod Mx protein using Western and dot blotting. A doublet band at about 75kDa and 65kDa was detected by Western blotting in kidney and spleen extracts of cod 3 and 4 days after i.p. injection with poly I:C but not in control fish injected with PBS. In blood leucocyte lysates, similar immunostaining could also be detected in Atlantic cod weakly after injection with PBS and more intensely after injection with poly I:C, suggesting some constitutive expression of Mx protein by leucocytes. Dot blot analysis showed that the Mx protein level was significantly higher in spleen, kidney, liver and gill of cod at least up to 4 days after injection with poly I:C when compared with the PBS-injected controls.  相似文献

The recapture rate and survival of reared cod Gadus morhua L. (15–45 cm) released in fjords and open coastal areas of Norway increased with increasing size at release, while release area and season also had a significant influence on the results. High mortality soon after release reduced the number of released fish by more than two–thirds within 6 months, and reduced the potential commercial yield to a low level. In most cases the recapture yield was lower than the released biomass and the net yield increased little or not at all with increasing size at release. In western Norway high fishing mortality of young fish reduced the numbers of surviving fish rapidly and the growth overfishing reduced the potential yield still further. Natural mortality decreased with increasing size at release, and became low when the fish approached their size refuge, i.e. when they could not be eaten by their usual predators.  相似文献

This study was carried out to reveal some characteristics of cationic proteins from Atlantic cod (Gadus morhua) milt chromatin and to investigate their ability to activate Atlantic salmon (Salmo salar) macrophages. Cationic proteins extracted from cod milt chromatin were fractionated on a cation exchange chromatography column. SDS-PAGE and amino acid analyses of the resulting fractions indicated that these proteins are similar to calf thymus histones. Two cationic protein fractions were used to stimulate leucocytes from Atlantic salmon in vitro and in vivo. Increased production of superoxide, measured as reduction of nitroblue tetrazolium (NBT), was used as indication of macrophage activation. Both fractions induced elevated superoxide anion production in the macrophages after 3 and 6 days of in vitro stimulation. Intraperitoneal injection of the cationic protein fractions in Atlantic salmon (100 mg kg(-1)) four days prior to slaughtering stimulated superoxide production when assayed after one and two days of cell cultivation. In macrophages from fish slaughtered two days after injection, activation could first be seen after two days of cell cultivation.  相似文献

The present study assessed differences in fecundity and egg quality from Atlantic cod Gadus morhua fed isoproteic diets containing 13% fat (low fat, LF) or 20% fat (high fat, HF) and either stressed or left unstressed as a control over the spawning season. Each diet was fed to triplicate groups of G. morhua from June 2009, through to first maturation and spawning. In January 2010 sub-groups of G. morhua were moved to land-based spawning tanks where the experimental trial was carried out. At the start of the experiment, G. morhua fed the high-fat diet were significantly larger than G. morhua fed low-fat diet. These differences were maintained through the spawning season, although with a loss of mass in both dietary groups. Relative fecundity through the season was significantly lower in stressed G. morhua fed LF compared to unstressed G. morhua fed the same diet. Stressed G. morhua had a higher variability in weekly amount of eggs spawned, spawning occurred more irregularly, and the spawning period lasted longer than in unstressed G. morhua. Several egg quality variables were also affected: eggs from G. morhua fed LF and exposed to stress had lower fertilization and hatching rates compared to the unstressed G. morhua fed the same diet as well as all G. morhua fed HF. Gadus morhua fed a low-fat diet appeared less tolerant to stress than fish fed a high-fat diet.  相似文献

Egg specific gravity is a function of egg volume, chorion volume, perivitelline space (PVS) and the specific gravity of chorion and ovoplasm. Experimental studies on cod ( Gadus morhua L.), a marine multiple batch spawner, showed that approx. forty per cent of the chorion volume is incorporated between batches and that the chorionic material has a mean specific gravity of 1·20 (range: 1·14–1·35). The chorion volume and specific gravity are low at the end of the spawning period in extremely fecund fish. Specific gravity of ovoplasm was observed to be 1·017 in marine eggs and calculated to be about 1·008 in brackish water eggs. Inherent variation in PVS is found to have little influence on egg specific gravity. With the other variables constant, egg specific gravity is inversely related to egg volume supported by studies on single batches of eggs. Measurements on every batch of eggs reveal that the production of large eggs (>1·3 mm) gives low density eggs initially and high density eggs towards the end of spawning, while the production of small eggs (<1·3mm) gives moderate specific gravity. These observations are discussed in relation to vertical distribution of the eggs and reproductive success of recruit and repeat spawners. There are indications that repeat spawners produce larger eggs.  相似文献

Understanding responses of marine species to temperature variability is essential to predict impacts of future climate change in the oceans. Most ectotherms are expected to adjust their behavior to avoid extreme temperatures and minimize acute changes in body temperature. However, measuring such behavioral plasticity in the wild is challenging. Combining 4 years of telemetry‐derived behavioral data on juvenile and adult (30–80 cm) Atlantic cod (Gadus morhua), and in situ ocean temperature measurements, we found a significant effect of sea temperature on cod depth use and activity level in coastal Skagerrak. During summer, cod were found in deeper waters when sea surface temperature increased. Further, this effect of temperature was stronger on larger cod. Diel vertical migration, which consists in a nighttime rise to shallow feeding habitats, was stronger among smaller cod. As surface temperature increased beyond ~15°C, their vertical migration was limited to deeper waters. In addition to larger diel vertical migrations, smaller cod were more active and travelled larger distances compared to larger specimens. Cold temperatures during winter tended, however, to reduce the magnitude of diel vertical migrations, as well as the activity level and distance moved by those smaller individuals. Our findings suggest that future and ongoing rises in sea surface temperature may increasingly deprive cod in this region from shallow feeding areas during summer, which may be detrimental for local populations of the species.  相似文献

The genomic architecture underlying ecological divergence and ecological speciation with gene flow is still largely unknown for most organisms. One central question is whether divergence is genome‐wide or localized in ‘genomic mosaics’ during early stages when gene flow is still pronounced. Empirical work has so far been limited, and the relative impacts of gene flow and natural selection on genomic patterns have not been fully explored. Here, we use ecotypes of Atlantic cod to investigate genomic patterns of diversity and population differentiation in a natural system characterized by high gene flow and large effective population sizes, properties which theoretically could restrict divergence in local genomic regions. We identify a genomic region of strong population differentiation, extending over approximately 20 cM, between pairs of migratory and stationary ecotypes examined at two different localities. Furthermore, the region is characterized by markedly reduced levels of genetic diversity in migratory ecotype samples. The results highlight the genomic region, or ‘genomic island’, as potentially associated with ecological divergence and suggest the involvement of a selective sweep. Finally, we also confirm earlier findings of localized genomic differentiation in three other linkage groups associated with divergence among eastern Atlantic populations. Thus, although the underlying mechanisms are still unknown, the results suggest that ‘genomic mosaics’ of differentiation may even be found under high levels of gene flow and that marine fishes may provide insightful model systems for studying and identifying initial targets of selection during ecological divergence.  相似文献

Large eggs (1·38 mm) of Atlantic cod reared in the laboratory produced large larvae. However, large larvae had low survival rates. Results also indicated that the first few days' of growth of cod larvae mainly resulted in an increase in mass. Exogenous feeding tended to result in faster growth than endogenous feeding. In the delayed feeding groups (larvae not fed until 67 degree-days), larvae from large eggs grew faster than those from small eggs (1·28 mm) after feeding commenced, while there was no significant difference in growth rate within feeding groups. Final thermal preferendum ( T ) experiments were conducted in a horizontal thermal gradient tank from the beginning of August 2001 to mid‐November 2001 using Atlantic cod Gadus morhua from 6·5 to 79·0 cm fork length ( L F). The value of T varied significantly ( P  < 0·005) with L F( T  = 7·23–0·054 L F), with smaller (younger) fish choosing higher temperatures than larger (older) fish. The preferendum varied from 6·9° C for fish of 6·5 cm to 3·0° C for those of 79·0 cm. Experiments comparing fish positions in the gradient tank between thermal gradients of 0·5–11·0 and 4·5–14·5° C demonstrated that fish positions were determined by temperature selection instead of undesirable tank effects. This study is the first to demonstrate the effect of ontogeny on temperature preferences of a marine fish species.  相似文献

Metal binding and conformational stability characteristics of psychrophilic elastase (ACE) from Atlantic cod (Gadus morhua) has been investigated. Chelation to Ca(2+) was found to be important for maintaining the biologically active conformation and for the thermal stability of the enzyme. However, presence of metal ions such as Zn(2+), Fe(3+) and Cu(2+) was found to inhibit its hydrolytic activity and so did the chelating agent EDTA. Both pH and guanidinium chloride induced denaturation of the enzyme was followed by monitoring the changes in the tryptophan fluorescence. ACE exhibited a simple two-state unfolding pattern in both acidic and basic conditions with the midpoint of transition at pH values 4.08 and 10.29, respectively. Guanidinium chloride and heat induced denaturation of the enzyme was investigated at two pH values, 5.50 and 8.00, wherein the enzyme possesses similar tertiary structure but differ in its hydrolytic activity. Guanidinium chloride induced denaturation indicated that the enzyme unfolds with a C(m) of 1.53 M at pH 8.0 and a DeltaG(H2O) of 6.91 kJ mol(-1) (28.65 J mol(-1) residue(-1)) which is the lowest reported for psychrophilic enzymes investigated till-date. However, at pH 5.50, DeltaG(H2O) value is slightly lowered by 0.65 kJ mol(-1) consistent with the observed increase in the apparent quenching constant obtained with acrylamide. On the other hand, increase in T(m) by 38.45 degrees C was observed for the enzyme at acid pH (5.50) in comparison to the heat induced unfolding at pH 8.0. The increase in the apparent T(m) has been attributed to the possible weak intermolecular association of the enzyme molecules at moderately high temperatures that is favoured by the increase in the accessible surface area / dynamics under acidic conditions. The stability characteristics of ACE have been compared with the available data for mesophilic porcine pancreatic elastase and possible mechanism for the low temperature adaptation of ACE has been proposed.  相似文献
