Summary A crossflow-microscreen cultivation technique was successfully used to select and maintain an easily harvestable microbial culture with a limited number of species under non-aseptic conditions in diluted cheese whey. The microbial selective pressure exerted by the system could be manipulated by varying the hydraulic () and mean cell () residence times. The optimum system parameters were =1 h and =10 h, resulting in a selected microbial population comprising three species only, namely Geotrichum candidum, Streptococcus cremoris and Leuconostoc lactophilum. The amino acid profile of the SCP produced compared favourably with other types of protein. The crossflow-microscreen technique makes SCP production possible from dilute, waste organic effluents.  相似文献

-Toxin (perfringolysin O) binds to cell surface cholesterol and forms oligomeric pores that cause membrane damage. Both in cytotoxicity and cell survival assays, a mutant Chinese hamster ovary cell line NPC1(–) that lacked Niemann-Pick C1 showed reduced sensitivity to -toxin, compared with wild-type (wt) cells. BC is a derivative of -toxin that retains cholesterol-binding activity but lacks cytotoxicity. Confocal and electron microscopy revealed the presence of multiple vesicles which bound BC, both on the cell surface and in the extracellular space of these cells. BC binding to raft microdomains was verified by its resistance to 1% Triton X-100 at 4°C and recovery of bound BC in floating low-density fractions on sucrose density gradient fractionation. BC-labeled vesicles were abolished when NPC1(–) cells were depleted of lipoproteins and also when treated with a Rho-associated kinase inhibitor Y-27632. In addition, similar vesicles were observed in wt cells treated with progesterone. In parallel with these results, -toxin sensitivity of NPC1(–) cells was increased when cells were depleted of lipoproteins or treated with Y-27632, whereas that of wt cells was decreased by progesterone. Our findings suggest that sequestration of toxin to raft-enriched cell surface vesicles may underlie reduced sensitivity of NPC1-deficient cells to -toxin.  相似文献

Summary The effect of assortative mating on the genetic correlation between traits X and Y is considered. Assortation on trait X changes the magnitude of the genetic correlation but not its sign. There are two situations depending on the signs of the correlation between mates () and of the random mating genetic correlation (): 1) if sign () = sign (), then >, where is the genetic correlation at equilibrium after continued assortation, and 2) if sign () = sign (), then < . However, negative assortative mating is virtually powerless to alter the magnitude of the genetic correlation. The consequences of a mixed assortation model, e.g., high milk production females mated to fast growing males and lesser productive females mated to slower growing sires, were also studied. Mixed positive assortation always increases the genetic correlation, but negative assortation decreases it. The implications of assortative mating on correlated responses to selection and on the equilibrium covariances between relatives for pairs of traits are discussed.  相似文献

The nanosecond fluorescence depolarization method was applied to measure the fluorescence lifetime () and the rotational correlation time () of bovine serum albumin (BSA) labeled with 1-dimethylaminonaphthalene-5-sulfonyl chloride (dansyl-Cl). Changes of and of dansyl BSA in the guanidine denaturation and in the thermal denaturation were examined. In parallel, the secondary structural change of dansyl BSA was followed by circular dichroism measurements. The magnitude of was almost unchanged between 1 and 2 M guanidine, where the secondary structure of the protein was predominantly disrupted; whereas that of began to increase before the disruption of secondary structure in the guanidine denaturation. In the thermal denaturation, in contrast, changes of both and occurred in a temperature range where the secondary structure was predominantly disrupted. The volume of equivalent sphere (V e ) and the axial ratio () for the BSA were 3.6–3.8×10–19 cm3 and 3.6 at 2M guanidine as against 2.1×10–19 cm3 and 2.2 in the absence of guanidine (25°C), respectively. The magnitudes ofV e and were 4.9×10–19 cm3 and 4.5 at 65°C, respectively. Although the secondary structural change of dansyl BSA was irreversible in the thermal denaturation,V e and were reversible.  相似文献

Summary Mycoplasmas (M. gallisepticum, chicken mycoplasmas), in concert with interferon (IFN), were effective in activating macrophages (M) to be tumoricidal. The M-activating capacity of mycoplasmas was maintained after treatment with heat, 0.1 M NaOH, 1 M HC1, or trypsin. M-activating factor was extracted from mycoplasmas with chloroform/methanol and water (Mf-B). Mf-B was also effective in activating M in the presence of IFN. The threshold dose of Mf-B for M of ordinary C3H/He mice and that for those of C3H/HeJ mice, the latter being known to be low responders to bacterial lipopolysaccharide, were actually the same. This seems to indicate that the effectiveness of Mf-B was not attributable to possibly contaminating lipopolysaccharides, and that the pathway of activity of Mf-B is different from that of lipopolysaccharides. Since the M-activating principle was only a very small part of Mf-B, we have not yet succeeded in identifying it, but there was no evidence that it was protein, nucleic acid, sugar, or lipid. The cytotoxicity of M activated by Mf-B plus IFN was dependent onl-arginine in the culture, suggesting that arginine metabolites are involved in M cytotoxicity. Mf-B induced a small amount of tumor necrosis factor in M, and this induction was markedly enhanced by IFN.  相似文献

Data are presented to evaluate further the abundance/biomass comparison method for the detection of pollution disturbance in marine benthic communities. The results suggest that this technique cannot be used in estuarine intertidal communities. The grossly polluted configurations appeared under unpolluted conditions, while undisturbed patterns were found under polluted conditions. Summary Receptive fields of individual retinular cells in the stemmata ofPapilio xuthus L. were examined electrophysiologically, and the receptive field of the complete stemmatal system was reconstructed (Fig. 8).In stemmata I-IV, proximal retinular cells have narrow receptive fields (acceptance angles of = 1.7–5 °, Fig. 5) and small inclinations of the visual axes (inclinations of = 0.7–1.5 °, Fig. 2) with respect to the axis of the stemma, while distal ones have wide fields ( =7–13 °, Fig. 5) and large inclinations of the visual axes ( = 5–10 °, Fig. 3). In stemmata V and VI, both proximal and distal retinular cells have wide receptive fields ( = 7–26 °, Fig. 6) and have large inclinations of their visual axes ( = 9–19 °) with respect to the axis of the stemma except for one proximal cell ( = 0 °) (Fig. 4).The spatial properties of distal and proximal retinular cells, combined with the finding that distal cells are homogeneous in the spectral sensitivity while proximal ones are heterogeneous (Ichikawa and Tateda 1980), suggest that the distal cells may be concerned largely with the detection of objects and proximal cells are involved with the discrimination of the color and shape of the detected objects.  相似文献

The specific features of the topology of spectral powers and coherent interregional interrelationships in the narrow, individually determined -, -, 1-, 2-, and 3-frequency bands were studied by means of high-resolution EEG (62 channels) in novice and experienced meditators (NMs and EMs) at rest and under the conditions of generation of an altered state of consciousness characterized by inactivation of cognitive activity and the occurrence of a positive emotional experience of happiness. EMs in the meditation-free state were found to be characterized by a shift in the values of the individual frequency to a lower-frequency region of the spectrum, along with higher, compared to NMs, -, 1-, 2-, and 3-band power values, which probably reflects the cumulative character of the influence of long-term meditative practice. The effective achievement of altered states of consciousness in EMs was associated with an increase in the local - and 1 powers in the anterior cortical areas, as well as long-distance coherence between the prefrontal and posterior associative cortex with the formation of a center of gravity in the left prefrontal region (lead AF 3). According to the data of the correlation analysis of the EEG power values and the data of subjective scaling of the meditation state, the -power values were positively associated with positive emotional experiences and negatively associated with the level of mental activity. The results of this study are consistent with current concepts that the and activities in narrow frequency bands reflect the activity of multifunctional neuronal networks selectively associated with processes of cognitive and affective activity.  相似文献

Spatiotemporal organization of rhythmic and EEG components was studied by means of spectral-correlation analysis in seven- to eight-year-old children (n = 18) during anticipatory selective attention to sensory tactile and auditory stimuli. The topography of changes in coherence of the oscillations suggests that functional assemblies formed on the basis of the rhythm are modality-specific. Their centers are localized in respective sensory-specific cortical regions (central areas of both hemispheres during tactile attention and temporal areas during auditory attention). The functional integration on the basis of the rhythm is represented in both hemispheres by functional associations of the temporal, frontal, and (to a lesser extent) posterior associative areas independently of the modality of a relevant signal. Both types of the functional integration are significant for a correct solution of a perceptive task. The proposition that the cooperation of the - and systems in neuronal organization of voluntary attention ensure, respectively, its informational and motivational aspects.  相似文献

An -tocopherol-binding protein has been isolated and purified from rabbit heart cytosol. The purified protein had an apparent molecular mass of 14,200, as derived from SDS-PAGE. The content of the protein in rabbit heart was around 11.8 g per g of tissue. The binding of -tocopherol to the purified protein was rapid, reversible, and saturable. Neither nor tocopherol could displace the bound -tocopherol from the protein, suggesting a high specificity for -tocopherol. -Tocopherol-binding protein did not bind oleate. Transfer of -tocopherol from liposomes to mitochodria was stimulated 8-fold in the presence of the binding protein, suggesting that this protein may be involved in the intracellular transport of -tocopherol in the heart.  相似文献

The locus for Tyrosinase-Positive Oculocutaneous Albinism (ty-pos OCA) has not yet been localised. The search for the ty-pos OCA locus has included a search for linkage to candidate pigment loci and a candidate chromosomal region, as well as a random search using highly polymorphic markers in 42 families, including 271 individuals of whom 79 are affected. The lod scores for the tyrosinase (TYR) locus (11q14–q21), homologous to the albino locus, c, in the mouse and the CAS2/TRP1 locus (9p22-pter), homologous to the brown locus, b, in the mouse were -5.89 and -7.22, respectively, at a recombination fraction of =0.01, thus excluding them from being the ty-pos OCA locus. In the candidate chromosomal region, 11p, four loci (probes) were tested, SAA (pSAA82), CALC (pHC36), HBB (Gamma-globin haplotype) and an AC repeat polymorphism at the Wilm's Tumour locus (WT1). A portion of 11p was excluded with the following lod scores: pSAA82 lod=-2.05 at =0.10; pHC36 lod=-3.87 at =0.05; gamma-globin haplotype lod=-2.80 at =0.10; and WT1 lod=-2.34 at =0.10. Thirty-three polymorphic markers randomly distributed on 13 different chromosomes were all excluded from close linkage to ty-pos OCA.  相似文献

Newborn rat adrenocortical cells were successfully cultured in a serum free carrier protein free medium (SPFM) by using -cyclodextrin as a cholesterol carrier and have expressed corticosteroid biosynthesis in this medium. A stable inclusion complex of cholesterol--cyclodextrin with a molar ratio of almost 1 was obtained for a 5 × 10–5 mol/1 -cyclodextrin concentration. Cell cultures incubated with [4-14C] cholesterol--cyclodextrin in SPFM produced, under ACTH stimulation, various 14C labeled steroids with a predominance of corticosterone and 18-hydroxy-11-deoxycorticosterone. As measured by gas chromatography and mass spectrometry, the ratio between corticosteroids (21-hydroxylated steroids) and 20-reduced steroids produced in SPFM with cholesterol--cyclodextrin was equal to 1.8. This corresponds to a value of 3.6 times higher than that found in the serum free medium with cholesterol-albumin. Morniga M is a jacalin-related and mannose-specific lectin isolated from the bark of the mulberry (Morus nigra). In order to understand the function and application of this novel lectin, the binding property of Morniga M was studied in detail using an enzyme-linked lectinosorbent assay and lectin-glycan inhibition assay with extended glycan/ligand collection. From the results, it was found that the di-, tri-, and oligomannosyl structural units of N-glycans such as those of the bovine 1-acid glycoprotein (gp) and lactoferrin were the most active gps, but not the O-glycans or polysaccharides including mannan from yeast. The binding affinity of Morniga M for ligands can be ranked in decreasing order as follows: gps carrying multiple N-glycans with oligomannosyl residues >> N-glycopeptide with a single trimannosyl core > Tri-Man oligomer [Man1 6(Man 1 3) Man], Penta-Man oligomer [Man1 6(Man1 3)Man1 6(Man1 3) Man] Man 1 2, 3 or 6 Man > Man > GlcNAc, Glc >> L-Fuc, Gal, GalNAc (inactive), demonstrating the unique specificity of this lectin that may not only assist in our understanding of cell surface carbohydrate ligand-lectin recognition, but also provide informative guidelines for the application of this structural probe in biotechnological and clinical regimens, especially in the detection and purification of N-linked glycans.  相似文献

Summary Linkage data on human peptidase C (PEPC), human factor H (HF), and coagulation factor XIIIB (F13B) are presented. The results confirm linkage between HF and F13B (lod=5.32 at =0.10 in males), and give strong evidence for linkage between PEPC and HF (lod=5.14 at =0.10 in males) and between PEPC and F13B (lod=3.55 at =0.10 in males). A human gene previously identified as a partial cDNA homologous to the gene of RET finger protein was characterized. Northern hybridization detected three messages of 3.3, 4.2, and 7.5kb. The coding sequences of the more abundant of the three messages, the 4.2 and the 3.3kb, were determined. The former encodes a 630 amino acid protein (TRIM41) and the latter a 518 amino acid protein (TRIM41). Green fluorescent protein (GFP) fusions of full-length TRIM41 and TRIM41 were both observed as speckles in the cytoplasm and the nucleus. The result was corroborated by Western analysis of cellular fractions. Results with GFP fusions of various segments of the TRIM41 proteins indicated that the nuclear transport of the proteins is mediated by an N-terminal segment common to both isoforms, but independent of a classical nuclear localization signal sequence.  相似文献

Summary Genetic analysis for leaf rust reaction of two widely adapted cultivars, Sonalika and WL 711, has been done using 21 near isogenic Lr lines and rust culture IL004 — avirulent on the two cultivars and all the Lr lines used. The segregation pattern in the F2 generation indicated the presence of a recessive gene in Sonalika and of a dominant gene in WL 711. These genes in cultivars Sonalika and WL 711 have been identified as Lr 11 and Lr 13, respectively. Gene Lr 13 is no longer effective in WL 711 but it continues to give field resistance in the backgrounds of Chris, Prelude and Thatcher. There has been no significant change in the virulence spectrum of the leaf rust pathogen in India with the release of WL 711. High susceptibility of WL 711 seems to be due to the evolution of more aggressive forms of the pathogen to this cultivar. The gene Lr 11, which behaves as a recessive in Sonalika, was effective against leaf rust when this cultivar was released. The high susceptibility of Sonalika is probably due to an increase in the frequency of race 77 virulent on Lr 11. Lr 11 has shown a dominance reversal in the background of Sonalika. Present results suggest that interaction of resistance genes with the background genotype must be studied for their effective use in breeding programme.  相似文献

Hydraulic lift, the transport of water from deep in the soil through plant root systems into the drier upper soil layers, has been demonstrated in several woody plant species. Here the volume of water involved in hydraulic lift by a mature sugar maple tree is estimated. Twenty-four intact soil cores were collected from the vicinity of a sugar maple tree at the same positions at which thermocouple psychrometers had been placed. Desorption measurements were made on the soil cores and the data were fitted to the Campbell relation for soil matric potential versus soil water content . The psychrometer data were filtered to obtain the diurnal component contributed by hydraulic lift. The diurnal component in was combined with the Campbell relation for each soil core to obtain the increase in soil water content due to hydraulic lift. The additional water contents were numerically integrated to obtain a volume of 102±54 1 of water which was hydraulically lifted each night. The volume of hydraulically lifted water (HLW) is sufficiently great that in ecosystems where hydraulic lift occurs it should be included in models for calculating the water balance. However, a previous analysis of the stable hydrogen isotope composition (D) of water in understory plants around trees conducting hydraulic lift implies a much greater volume of HLW than that calculated from the analysis performed above. To reconcile these differences, it is hypothesized that some understory plants preferentially extract HLW due to its higher matric potential and that the proportion of this water source within the xylem sap of at least some understory plants that use HLW was so great that the roots of these plants must therefore be in close proximity to the tree roots from which the HLW comes. The results of this study have implications for studies of plant competition where positive associations may exist as well as for ion uptake, nutrient cycling and the design of agroforestry systems.  相似文献

Summary Estimates of recombination frequency among enzyme loci of pitch pine revealed two new linkages, Mdh3:Pgm2 (=0.01) and Pep1:Mdh4 (=0.38), and confirmed two previously established linkages. Tighter linkage (=0.30) was ruled out for nearly all gene pairs examined. Summary Confidence interval estimators have not been defined for dominance to additive genetic variance () and average degree of dominance () for the nested, factorial, and backcross mating designs. The objective of this paper was to describe interval estimators for these parameters. Approximate F random variables were defined for expected mean square (EMS) ratios for linear models with one environmental effect. Approximate 1– parametric interval estimators were defined for and using these random variables. Random variables defined for linear models with no environmental effects are not approximately distributed as F random variables because common EMS are involved in the numerators and denominators of the EMS ratios. The EEG correlates of the performance of examination tests and special cognitive tests under the conditions of everyday study (common studying conditions, CSCs) and immediately before examination (examination conditions, ECs) were analyzed in 39 male students aged 18–20 years. The results of the examinations strongly correlated with the relative spectral power (SP) of the EEG rhythm before the examination. Therefore, the students were divided into two groups with different directions of changes in the -range SP under the ECs compared to the CSCs. Students from group 1 were characterized by increased relative - and -rhythm SPs under the ECs and a good examination performance; students of group 2, by decreased - and -rhythm SPs, an increased index of the EEG of the central and frontal cortical regions under the ECs, and a poor examination performance.  相似文献
