Proliferation and migration of the scleroblasts and their precursor cells during ethisterone-induced anal-fin process formation in adult females of the medaka, Oryzias latipes , were investigated using a radioautographic technique with 3H-thymidine. In the posterior margin of the joint plate, rapid increases in labeling indexes and scleroblast population began simultaneously in the central portion on the third day of ethisterone treatment, and the population became approximately ten times that in the control fish on the fifth day. However, nomarked increase in scleroblast population occurred in the peripheral portion, although the scleroblasts showed nearly the same labeling index as those in the central portion throughout the experiment. These observations indicate that the scleroblasts and their precursor cells migrated from the peripheral to the central portion along the proxi-distal axis of the joint plate. Some cells in the inner and hind spaces also migrated to the posterior margins and subsequently developed into scleroblasts. It may be concluded, therefore, that the scleroblasts are derived from precursor cells situated not only along the posterior margins but also in the adjacent spaces. These precursor cells proliferate and/or migrate to the central portion of the posterior margins and aggregate to form the scleroblast mass.  相似文献

The ultrastructure stages of Blastocystis galli were studied in chicken's intestine and in laboratory cultures. There were found morphological structures: surface coat (cell from chickens' intestine showed a very thick surface coat); cell membrane--there were some small electron-opaque deepening "pockets" on the membrane; inner membrane; endoplasmic reticulum with attached ribosomes, which present in the cytoplasm; all cells contained numerous of small vacuoles and large glycogen inclusions in cytoplasm; mitochondria with tubular cristae; nucleus with granules condensed chromatin; central vacuole; Golgi complex was represented by number of plates grouped in a pite; the cyst-like forms were surrounded by multilayered wall.  相似文献

In the CNS of the Patinopecten yessoensis (Jay) two types of cells have been revealed. The I type cells are typical unipolar neurons with a developed granular endoplasmic reticulum and Golgi compex, with a nucleus containing small amount of chromatin. They possess elementary peptidergic granules. The II type cells have in their cytoplasm and processes a large amount of electron-opague granules, specific for adrenergic systems. The nucleus is rich in clustered chromatin, the granular endoplasmic reticulum is poorly developed, cytosomes are absent. According to their ultrastructural organization the latter correspond to small granular cells of the mammalian autonomic nervous system.  相似文献

应用透射电子显微镜技术 ,观察了水稻 (OryzasativaL .)淀粉胚乳细胞编程性死亡过程中核的变化特征。伴随胚乳的发育进程 ,淀粉胚乳细胞核表现出衰退特征 :核变形、染色质凝缩、核膜多处被降解破坏、核基质外泄等。DNALadder显示核内大片段DNA呈严重的弥散状拖尾现象 ,而核内和胞质中在 14 0~ 180bp处有明显的条带。在核衰退的同时 ,其胞质中的粗面内质网、淀粉质体和线粒体等细胞器具有正常的代谢功能 ,细胞仍在合成并积累营养物质 ,淀粉胚乳细胞一边衰退一边行使其功能 ,直至死亡。这些结果表明 ,水稻淀粉胚乳在核衰退的同时 ,细胞仍在积极合成与积累贮藏产物 ,表现为一种特殊形式的植物细胞编程性死亡现象。此外 ,对淀粉胚乳细胞特有的核质关系、植物细胞编程性死亡过程中细胞核的变化等问题进行了讨论。  相似文献

Examination of the coenenchyme tissues of Renilla reniformis revealed two regions of crystal formation: the endoderm containing small oval deposits of unknown composition and the mesoglea containing larger elongated spicules composed of calcite. Spicule formation takes place intracellularly in scleroblasts and may be explained by the following sequential processes: an organic matrix consisting of a homogeneous ground substance and fibers is formed in a large vacuole. Calcite needles 0.4 μ in diameter develop in close association with matrix fibers and vesicles, and grow to form the central core of the spicule. Large electron dense bodies dominate the scleroblast cytoplasm during these early stages of growth. In a later stage, smaller needles 0.2 μ in diameter develop surrounding the core to form the distal lobes of the spicule. ‘Lollipop’-shaped vesicles containing fibers appear in the scleroblast cytoplasm at the onset of this stage. This material is released at the calcification front and presumably is incorporated into the spicule as an organic matrix of crystals in the distal lobes.  相似文献

Summary Scleroblasts were separated from fragmented tissue of growing tips ofLeptogorgia virgulata and cultured using a modification of the technique of Rannou. Replacement of fetal bovine serum with horse serum seemed to increase scleroblast viability. Cell adhesion occurred from 14 to 43 d. Cultured scleroblasts demonstrated cell aggregation, spicule formation, and extrusion of spicules into the external medium. Cells showing spicules in the process of being extruded appeared on the average after 24 d of culture. Variability among cultures was marked with respect to both division and spicule formation. 应用电镜技术研究了东方扁虾(Thenus orientalis)精子发生的全过程,精原细胞呈椭圆形,其染色质分布较均匀,线粒体集中于细胞一端形成“线粒体区”。初级精母细胞较大,染色质凝聚成块,次级精母细胞核质间常出现大的囊泡,胞质内囊泡丰富而线粒体数量却明显减少,早期精细胞核发生极化、解聚,部分胞质被抛弃。中期精细胞外观呈金字塔形,分为三区;正在形成的顶体位于塔顶,核位于塔基部,居间的细胞质基质内富含膜复合物,后期精细胞顶体进一步分化。形成顶体帽和内、外顶体物质等三个结构组份。成熟精子核呈盘状或碗状,具有5-6条内部充满微管的辐射臂。  相似文献

四种淡水鱼的血细胞形态基本相似。红血球形态与其他低等脊椎动物基本相似。淋巴球绝大部分是小淋巴球:单核球数量较少;四种鱼的嗜中性白血球形态结构差不多,胞核多为蚕豆形,很少见分叶核,分叶一般也只有二叶,这与哺乳类显然不同;嗜酸性白血球的形态结构与其他脊椎动物基本相似;在少数血涂片中看到了嗜碱性白血球。    相似文献

The effect of insulin (I), cortisol (F) and prolactin (P) on the ultrastructural morphology of epithelial cells of cultured mammary explants from virgin ovariectomized (OV-X) goats were studied. The epithelial cells showed little structural organization and were devoid of fat droplets and secretory protein granules at zero time of culture. The cytoplasm contained few profiles of smooth and rough endoplasmic reticulum and the Golgi apparatus was rudimentary. After being cultured in Waymouth's medium without added hormones the epithelial cells were indistinguishable from epithelial cells of uncultured explants. The addition of I induced changes mainly in the appearance of nucleoli. The nucleoli were enlarged and fibrillogranular areas with light spaces were observed. The most obvious cytological changes of epithelial cells of explants cultured in the presence of I and F are translocation of the nucleus into the basal cytoplasm, increase of rough endoplasmic reticulum, an increase in the size of the Golgi apparatus, presence of one or two lipid droplets and in some cells vacuoles with protein granules were present. Mitochondria were more abundant. The epithelial cells of explants cultured in the presence of I, F and P were characterized by the polarization of organelles within the cytoplasm and by the formation and release of protein granules and small and large fat droplets. The cell nucleus was in the basal cytoplasm, the Golgi apparatus was supranuclear. The rough endoplasmic reticulum was extensively developed and formed large sacs. Ultrastructural changes in secondary wall formation of Phyllostachys pubescens Mazel fiber were investigated with transmission electron microscopy. Fiber developed initially with the elongation of cells containing ribosomes, mitochondria and Golgi bodies in the dense cytoplasm. During the wall thickening, the number of rough endoplasmic reticulum and Golgi bodies increased apparently. There were two kinds of Golgi vesicles, together with the ones from endoplasmic reticulum formed transport vesicles. Many microtubules were arranged parallel to the long axis of the cell adjacent to the plasmalemma. Along with the further development of fiber, polylamellate structure of the secondary wall appeared, with concurrent agglutination of chromatin in the nucleus, swelling and disintegration of organelles, while cortical microtubules were still arranged neatly against the inner side of plasmalemma. Lomasomes could be observed between the wall and plasmalemma. The results indicated that the organelles, such as Golgi bodies together with small vesicles, rough endoplasmic reticulum and lomasomes, played the key role in the thickening and lignification of the secondary wall of bamboo fiber, though cortical microtubules were correlative with the process as well.  相似文献

We examined nucleus pulposus notochordal cells of individuals ranging in age from the eighth week of fetal life to 32 years of age. With increasing age, notochordal cell structure changed, as did the cell-to-cell relationships and the cell-to-matrix relationships. All notochordal cells contained normal organelles, including welldeveloped endoplasmic reticulum, but, in addition, fetal notochordal cells demonstrated an unusual relationship between rough endoplasmic reticulum and mitochondria: elements of the rough endoplasmic reticulum encircled almost every mitochondrion. Fetal notochordal cells contained large amounts of glycogen, while older cells had much smaller glycogen deposits. Cytoplasmic filaments were observed in cells of all ages. The cells formed tightly packed clusters in the fetus with little, if any, extracellular matrix between individual cells. Cells separated from each other with age and by the twenty-first week of fetal life, only slender strands of cytoplasm connected them. Previous light microscopic studies described notochordal cells as ‘physaliphorus’ cells since they appeared to contain large cytoplasmic vacuoles. However, electron microscopy showed that these apparent vacuoles consist of extracellular matrix surrounded by cells or cell processes. The structure of notochordal cells and their persistence in the nucleus pulposus after fetal life suggest that they may have a significant role in the formation and maintenance of the nucleus pulposus.  相似文献

Taste buds in foliate papillae of the rhesus monkey were examined by electron microscopy. Three distinct cell types were identified. Type I cells were narrow elongated cells containing an oval nucleus, bundles of intermediate filaments, several Golgi bodies, and characteristic apical membrane-bounded dense granules. These cells exhibited morphological variations: some had a moderately dense cytoplasm, perinuclear free ribosomes, and flattened sacs of rough endoplasmic reticulum; others had a more lucent cytoplasm, dilated irregular rough endoplasmic reticulum, lysosome-like dense bodies, and lipid droplets. Type II cells typically contained a spherical, pale nucleus, a prominent nucleolus, supranuclear and infranuclear Golgi bodies, mitochondria with tubular cristae, and one or two centrioles. This cell type, too, showed some variation in the relative amounts of ribosomes and smooth endoplasmic reticulum, which varied inversely with each other. Type III cells were characterized by a clear apical cytoplasm essentially devoid of ribosomes and containing microtubules. In a few type III cells, the peri- and infranuclear regions contained many ribosomes and some rough endoplasmic reticulum. In most Type III cells, there were large numbers of dense and clear vesicles in the peri- and infranuclear regions; some of the vesicles were grouped in synapse-like arrangements with adjacent nerves. The morphological variations exhibited by all three cell types could be accounted for by age differences in each of the cells. This would be consistent with the notion that cell renewal occurs in each of the three cell populations.  相似文献

用常规电镜方法观察了锦橙[Citrussinensis (L.) Osb.]汁囊从原始细胞到发育为一个具柄的成熟汁囊的过程中,汁囊构成细胞超微结构的变化。锦橙汁囊原始细胞及发育为球状体时的构成细胞以及柱状结构顶端的细胞都是一种典型的分生组织细胞。在细胞质中有包括线粒体、质体、内质网、核糖体等丰富的细胞器,但没有观察到高尔基体。这些分生细胞分裂一段时期后就停止活动,逐渐分化为适应贮藏功能的液泡化薄壁细胞。分生细胞开始分化时,在细胞中出现许多小液泡和高尔基体。这些小液泡逐渐地融合,同时细胞质变少,最后形成一个有中央大液泡的薄壁细胞,在紧贴细胞膜的薄薄的一层细胞质中有线粒体、质体、高尔基体以及含有许多脂滴的杂色体。但成熟果实中汁囊的薄壁细胞中几乎没有任何细胞器。  相似文献

The structural changes in the granulosa lutein cells in the pregnant cows between 60 and 245 days, were observed. The polyhedral or pump cells has an acidophilic cytoplasm around the spherical nucleus. The glycogen granules increased in number up to 230 days of pregnancy. The lipid globules increased in size and amount during advanced pregnancy. The mitochondria showed a wide range of variations in shape and size. The matrix of many mitochondria contained dense inclusions which replaced the entire matrix in late pregnancy. The endoplasmic reticulum was chiefly of smooth type but a few stacks of the rough variety could be seen in the early days of pregnancy. Sometimes, whorls of smooth-surfaced endoplasmic reticulum enclosing mitochondria were seen. The Golgi complex was fully developed in the granulosa cells of all cases studied. The dense granules and lipid globules increased in size and number in the older corpora lutea. The cell surface showed numerous infolding of the plasma membrane.  相似文献

Immune lymphocytes sorbed on the surface of the target cells were characterized during the period of the first three hours of combined incubation by the presence of the electron-dense matrix, abundance of mitochondria and lipids; small lymphocytes had disseminated ribosome organized into polysomes in the medium lymphocytes forming individual cysterns of the granular endoplasmic reticulum in the large lymphocytes, this indicating active protein synthesis by these cells. There were also revealed cells of plasmatic type. Cells incubated with the PHA for one hour represented a homogenous population of small lymphocytes of the same size as the clear cytoplasm containing free ribosomes and individual mitochondria. The proportion of the medium lymphocytes and the blasts increased with increase of the incubation period. These are cells with the clear cytoplasm freely disseminated polyribosomes in which no developed granular endoplasmic reticulum was sometimes revealed. The presence of two types of cells whose ultrastructure reflected their functional characteristics is discussed.  相似文献

It was established that the hepatocytes of least shrew (Sorex minutissimus) contained 1-2 nucleus with wide nuclear pores and big nucleolus. There are abundant mitochondria with numerous crists in their matrix (many organelles were divided along the crists). Each mitochondrion was surrounded by the cistern of granular endoplasmic reticulum. The ultrastructure of Golgi complex (1-2 flat cisterns, small vesicles] indicated that the bile was secreted often, by small portions, that was the adaptation to repeated fractional feeding. There were many peroxisomes in the cytoplasm of hepatocytes. Lipid droplets were absent, glycogen granules were not at all hepatocytes. Sinusoidal cells had the usual structure. All sinusoids were open and consisted of the erythrocytes, fragments of hepatocyte cytoplasm and myelin figures in its lumen.  相似文献

In preparation for winter low temperatures, larvae of the gall fly, Eurosta solidaginis, accumulate the cryoprotectants glycerol, sorbitol, and trehalose. The fat body cells of these freeze-tolerant larvae can survive intracellular freezing to -80 degrees C for 48 h even though no whole larvae survive this treatment. We hypothesized that some other tissue was more susceptible to freezing and therefore may be responsible for larval death. This paper compares the ultrastructure of brain, muscle, and Malpighian tubules between non-lethally frozen and lethally frozen freeze-tolerant larvae. The nuclei of cortical brain cells from lethally frozen larvae exhibited clumped chromatin and nuclear membranes with occasional expansions or 'blebs' of the intermembranous space, while the cytoplasm contained swollen spheres of endoplasmic reticulum. In contrast, non-lethally frozen brain contained nuclei with evenly dispersed chromatin, smooth nuclear membranes and a cytoplasm free of swollen endoplasmic reticulum. Muscle tissue of lethally frozen larvae contained disrupted myofilaments surrounding the Z-line in comparison to non-lethally frozen muscle which had myofilaments extending all the way to the Z-line. Alterations of Malpighian tubule cells from lethally frozen larvae included an extracted cytoplasm with swollen and rounded mitochondria. In contrast, Malpighian tubule cells from non-lethally frozen larvae had a more concentrated cytoplasm with many rod-shaped mitochondria. Results show alterations to all three tissue types due to lethal freezing. The brain tissue contained the most observable alterations and therefore may be the most susceptible to lethal freeze damage.  相似文献

Summary The fine structure of the main excretory duct epithelium of the male mouse submandibular glands was investigated by scanning and transmission electron microscopy. Three principal cell-types were observed: type I and II, and basal cells. This epithelium was characterized by the presence of intercellular canaliculi. Type-I cells were the most numerous. They had an abundance of mitochondria, well-developed Golgi apparatus, a few electron-lucent lipid-containing granules and poorly developed basal infoldings. These cells were also characterized by many glycogen granules throughout the cytoplasm and abundant smooth endoplasmic reticulum in the apical cytoplasm. Type-II cells were the second most numerous. Their most characteristic feature was the presence of abundant heterogeneous lipid-containing granules having acid phosphatase activity at the periphery. They were concentrated in the infra- and supranuclear cytoplasm. The granules may be derived from mitochondrial transformation and seem to be a special kind of secondary autolysosome. Type-II cells also contained abundant mitochondria throughout the cytoplasm, much smooth endoplasmic reticulum in the apical cytoplasm, a well developed Golgi apparatus adjacent to the heterogeneous lipid-containing granules and no basal infoldings. Basal cells were situated adjacent to the basal lamina. They had a large nucleus and the cytoplasm was filled with glycogen granules.  相似文献
