Ruta graveotens subshrubs, known to contain a large fractionof their furanocoumarins on the leaf surface, were used forinvestigation of salt and acid rain effects on the plant surfaceand substances occurring there. Groups of shoots on each plantcovered by plastic bags were sprayed with either sulphuric acid,pII 2.5, saturated sodium chloride solution, or a distilledwater control. Uncovered shoots, an additional control, hadthe highest concentrations of furanocoumarins both on the surfaceand in the whole leaf. The upper leaves contained less furanocoumarinsin absolute amounts but were similar to the lower leaves ona percentage basis. Spraying caused a decrease in total furanocoumarinconcentrations, slight in the case of distilled water and greaterfor acid and salt, but the percentage on the surface after sprayingincreased. The upper and lower leaves reacted differently tosalt than to sulphuric acid and water: with salt, the percentagesof furanocoumarins on the surface of both kinds of leaves showedsimilar increases; with the other two sprays, upper leaves showeda similar increase, but the lower leaves had a much higher percentageon the surface owing to a high rate of extrusion. Summary Seasonal changes in the distribution and abundance of furanocoumarins in wild parsnip, Pastinaca sativa (Umbelliferae), were examined in a population of plants in Tompkins County, New York. Xanthotoxin, imperatorin and bergapten (linear furanocoumarins) occur in all above-ground parts of the plant; in addition, angelicin and sphondin (angular furanocoumarins) occur in umbels of some individuals. Total furanocoumarin content, as measured by percent dry weight, is greatest in reproductive parts, particularly buds and seeds; variation in concentrations between plants is greatest in vegetative structures (e.g., leaves).Within the plant, the distribution of furanocoumarins is significantly correlated with nitrogen, as opposed to biomass, allocation. In that nitrogen is often a factor limiting the plant growth, furanocoumarins appear to be allocated in proportion to plant tissue value; reproductive structures, obvious contributors to plant fitness, contain over ten times the amount of nitrogen and furanocoumarin contained in vegetative structures such as senescent leaves.Stepwise multiple regression analysis revealed that generalized insect herbivores tend to feed on plants or plant parts low in furanocoumarin content and, correspondingly, low in nitrogen content. Parsnip specialists, notably Depressaria pastinacella (Lepidoptera: Oecophoridae), feed exclusively on umbels, plant parts rich in nitrogen and furanocoumarins; furanocoumarin number and content in fact account for over 60% of the variance in number of umbel feeders. These patterns conform with previous determinations of the toxicological properties of furanocoumarins. Nitrogen is known to affect growth rate, fecundity, longevity and survivorship of insect herbivores; by tolerating or detoxifying furanocoumarins, D. pastinacella can consume plant tissues containing significantly greater amounts of nitrogen than tissues consumed by generalist feeders. That the presence of D. pastinacella on individual plants is correlated with the number of furanocoumarins present is consistent with the hypothesis that parsnip specialists use angular furanocoumarins as host recognition cues.  相似文献

A study of the leaf flavonoids, furanocoumarins and essential oils of 51 species (three genera) of the tribe Psoraleeae (Fabaceae) has provided chemical data to support the recent subdivision of the large worldwide genus Psoralea into a number of segregate genera. The flavonoid patterns in most species were very similar, consisting largely of proanthocyanidins and complex mixtures of flavone O- and C-glucosides. Psoralea repens, the only maritime species to be examined, is unique in containing only rutin. Three isoflavones (daidzein, formononetin and genistein) were of widespread occurrence. The genera Psoralea and Otholobium, which could not be separated on flavonoid data, were clearly demarcated by their essential oils. Furanocoumarin patterns were useful for the characterization of species.  相似文献

Transgenic chicory plants were obtained from different explantsco-cultured with Agrobacterium tumefaciens. Among tap-root,leaf and cotyledonary tissues, etiolated cotyledons showed thegreatest competence for transformation. The Agrobacterium strainsused contained either pGSGLUC1 or pTDE4 as a vector which carryboth the neomycin phosphotransferase II gene (nptll) for kanamycinresistance and ß-glucuronidase gene (uidA) under thecontrol of different promoters. Transformation was confirmedby NPTII enzymatic assay, histochemical analysis of GUS activityand DNA hybridization. Transgenic plants expressed both markergenes in root and shoot tissues. In leaves, GUS activity wasexpressed in all tissue types, whatever the nature of the promoter.Nevertheless, variable heterogeneous patterns of expressionwere observed in the different root tissues. Eight Psoralea species (Leguminosae) were inoculated with Agrobacterium rhizogenes, strains 8196 and 9402. Hairy roots were only induced by strain 9402. Attention was focussed on Psoralea lachnostachys. Transformed roots grew very rapidly in Gamborg B5 liquid medium with a doubling time of the culture of 38 hours. Whatever the culture conditions, the two furanocoumarins usually found in roots of Psoralea plants, psoralen and angelicin, were not detected in cultured transformed and non transformed roots even when some chitosan was added to the medium. However, 669 g.g–1 dry matter of psoralen and 215 g.g–1 dry matter of angelicin were found in roots from soil grown plants. Cuts were made in the surface of the shoot apices of Euphorbialathyris, tomato (Lycopersicon esculentum), and Pea (Pisum sativum)while they were completely immersed in water or aqueous mannitolat various concentrations, or in near-saturated air. Gapingoccurred all over the apical dome of Euphorbia and on the tomatoapex at the site of emergence of the primordial bulge. Maximumgaping occurred in near-saturated air and under water, and wasprogressively reduced with increasing osmotica. It is concludedthat the gaping results from tension in the surface cells andis not caused by superficial drying out. No gaping occurred in the axil of the newly formed primordiumof the tomato nor anywhere in the apex of the pea. Histologicalevidence suggests that these tissues are under lateral compression. The mechanical stresses involved are discussed in relation tothe morphology of the apices together with existing data onthe distribution of cell division during primordia formation.  相似文献

当归属是伞形科的代表性大属,日本列岛是其重要的分布地和多样性中心之一。该研究对产自日本的9种当归属植物(特有种8种)的果实形态结构和表面微形态特征进行了观察和比较,以明确它们的分类学特征,探讨潜在的种间关系,为进一步的分类修订和系统发育研究提供资料和依据。结果表明:(1)体式显微镜观察发现,产自日本的9种当归属植物的果实形状主要为倒卵形和长圆形,表面光滑或少毛,呈不同程度的背腹压扁,背棱大多呈线状突起或狭翅状,侧棱则呈或宽或窄的翅状,油管一般为每个棱槽1条、合生面2~4条。(2)扫描电镜观察发现,大多数材料的果实具有疏密不等的平行线形初级蜡质纹饰,次级纹饰呈现鳞状、束状、索状、丘状或不明显,个别可见珊瑚状或网格状基底,一般具有颗粒状附属物。(3)通过分析比较上述形态特征和鉴别要点,编制了分种检索表,并结合中国当归属系统发育的研究成果,进一步对日本特有当归属植物的系统位置及其与东亚大陆物种的亲缘关系进行了讨论。研究认为,日本特有当归属植物的果实形态特征较丰富,能为种间分类和亲缘关系研究提供有力支持。  相似文献

In Otago shelf waters surface swarms of krill (Nyctiphanes australis),hyperiid amphipods (Parathemisto spp.) and galatheid crab larvae(Munida gregaria) provide an abundant summer food source forplanktivores. We tested the hypothesis that aggregation of avianplanktivores depends upon the spatial distribution of theirprey. Gulls (Larus scopulinus, L.bulleri, L.dominicanus), sootyshearwaters (Puffinus griseus) and white-fronted terns (Sternastriata) showed significantly aggregated distributions. Thedistribution of birds sitting on the sea surface was correlatedwith the abundance pattern of krill but was not correlated withthe distribution of smaller hyperiid amphipods. The distributionof flying red-billed gulls (L.scopulinus), black-billed gulls(L.bulleri) and sooty shearwaters was correlated with the krilldistribution but black-backed gulls (L.dominicanus) were not.Stomach contents of black-billed gulls were dominated by krill,in contrast to sooty shearwaters, which ate a higher proportionof Minida, and black-backed gulls, which contained Munida andfish. No amphipods were found in bird stomachs. There was nosignificant correlation between bird distributions and the hydrographicregime, water depth or distance offshore. The distribution ofprey rather than hydrographic regime was a more important determinantof bird distributions at this spatial scale (2.6–12 km)and location.  相似文献

Nuclei from mung bean (Vigna radiata) hypocotyls contained twoglycoproteins of 50 and 49 kDa, respectively, that reacted withconcanavalin A. The glycoproteins were released from the nuclearenvelope by treatment with 2 M KCl but not with nucleases. Theglycoproteins, tentatively named gp50 and gp49, were isolatedand characterized. Gel-permeation chromatography suggested thatgp50 and gp49 seem to exist as a complex with other components.The glycoproteins could be detected only in the nuclear fractionby immunoblot analysis with specific antibodies, and they werenot detected in endoplasmic reticulum, plasma membrane, vacuolarmembrane or mitochondria. Agglutinin I from Ulex europeaus,peanut agglutinin, soybean agglutinin and wheat germ agglutininall failed to bind to the glycoproteins. Treatment with glycopeptidaseF removed all oligosaccharides from the glycoproteins and decreasedtheir molecular masses by about one thousand daltons each. Theseresults suggest that the glycoproteins contained N-linked, high-mannose-typeoligosaccharides with six or seven hexose residues. gp50 andgp49 seemed to be isoforms of a single glycoprotein becausethe two proteins had some common properties. Nuclear fractionsfrom azuki bean (Phaseolus angularis) and soybean (Glycine max)contained proteins that were immunologically similar to gp50and gp49. Plantlets were regenerated from somatic embryos originatingfrom cotyledonary tissues of pomegranate (Punica granatum) throughmultiple somatic embryogenesis. Embryogenic cell clusters proliferatedvigorously with regular sub-culturing after 20 d on RBM-II mediumcontaining 1 µM kinetin (KN), 2 µM benzylamino purine(BAP) and 5 µM 2,4-dichlorophenoxyacetic acid (2, 4-D).Developmental stages of somatic embryos were expressed on sub-culturingwith a low level of 2, 4-D (2.5 µM). Embryogenic initialscells were small, round to oval, thick-walled, contained densecytoplasm which stained with acetocarmine and were usually attachedto non-embryogenic cells. It is not knownif, in polarized cells, desensitization events can be influenced by thedomain on which the receptor resides. Desensitization was induced by5'-(N-ethylcarboxamido)adenosine (NECA) and wasquantitated by measurement of short-circuit current (Isc) in response to adenosine. NECA addedto either the apical or basolateral compartments rapidly desensitizedreceptors on these respective domains. Although apical NECA had noeffect on the basolateral receptor stimulation, basolateral NECAinduced a complete desensitization of the apical receptor. Wehypothesized that desensitization of apical receptor by basolateraldesensitization could relate to a trafficking step in which A2breceptor is first targeted basolaterally upon synthesis and transportedto the apical surface via vesicular transport/microtubules. Becausedesensitization is associated with downregulation of receptors, apicaladenosine receptor can thus be affected by basolateral desensitization. Both low temperature and nocodazole inhibited Iscinduced by apical and not basolateral adenosine. In conclusion:1) a single receptor subtype, here modeled by the A2b receptor,differentially desensitizes based on the membrane domain on which it isexpressed, 2) agonist exposure on one domain can result indesensitization of receptors on the opposite domain, 3)cross-domain desensitization can display strict polarity, and4) receptor trafficking may play a role in thecross-desensitization process.

Inoculation of the potato cut surfaces with an incompatiblerace of Phytophthora infestans induced an accumulation of rishitin,but only a trace occurred when infected by a compatible race.When the tuber was cut, a large amount of steroid-glycoalkaloids(solanine) accumulated in the cut surface tissue, although onlya trace was found in intact tissue. Only a small amount of solanine,if any, was contained in the wound periderm tissue. Most ofthe solanine seemed to be distributed in tissue neighbouringthe newly formed meristematic tissue zone. Distribution of solanineas a function of distance from the cut surface was exponential.Infection of the cut surface by an incompatible race of Phytophthorainfestans reduced the accumulation of solanine. The higher theconcentration of zoospore used, the less the solanine content.It has been reported that the higher the concentration of zoosporesused for inoculation of the cut surface, the less the numberof renewed meristematic cells in the wound tissue. In experimentsusing fresh and aged tubers, a good correlation between thenumber of renewed meristematic cells and solanine content wasfound. The accumulation of solanine in the wound tissue andits reduction due to infection by an incompatible race may berelated to renewed meristematic cells formation and its reductioncaused by the infection. The relationship between coleoptile elongation and alcoholicfermentation of rice under anoxia is examined using seeds either:(a) N2 flushed during submergence, (b) incubated in stagnantdeoxygenated agar at 0·1% w/v to simulate the stagnantconditions of waterlogged soil, or (c) incubated in waterloggedsoil. Coleoptile elongation growth was greater for N2 flushing> stagnant agar > soil; seed survival was also greatestin this order over 1-5 d. Ethanol concentrations in coleoptiles and intact seeds (cv.IR42) were approximately 300 and 100 mol m-3 respectively whenseeds were grown 3 d in stagnant agar, however 92% of the ethanolin seeds diffused into the external medium when solutions weremixed for 5-10 s. Coleoptile growth under anoxia was relatedto rates of ethanol synthesis (RE) in different treatments;there was greater coleoptile growth and RE for seeds in N2 flushedsolutions than in stagnant deoxygenated agar. Coleoptile growthof individual seeds was also related to the RE of each seedat 2-3 d after anoxia (r2 = 0·46). Analysis of different tissues was important in evaluating growthand metabolism of coleoptiles. Although the coleoptile onlyaccounted for 0·7% of seed dry weight at 3 d after anoxia,it contained 21% of the ethanol produced by rice seeds. Therewere also three-fold higher rates of RE on a fresh weight basisin expanding tissues in the base of the coleoptile relativeto the elongated tissues at the apex. The in vitro reversion of Pfr to Pr in the dark was preventedby adding the low molecular weight fraction obtained from aSephadex G-50 column chromatogram to the crude homogenate ofetiolated pea epicotyl tissues, and by the methanolic and boilingaquous extracts of the tissue. The degree of inhibition of darkPfr reversion was related to the logarithmic concentrationsof the methanol-extractable material. The active substance washighly soluble in water-saturated n-butanol; but not in chloroform,ethyl acetate, n-hexane, benzene and pure n-butanol. A comparative study has been made of the penetration of solarradiation through leaf canopies of contrasting structure. Forthis purpose experiments were undertaken on (a) Gladiolus withnearly vertical leaves, (b) Raphanus sativus with its rosettehabit, (c) two varieties of Linum usitatissimum, one an unbranchedflax type and the other a multi-stem linseed type, and (d) Agrostemmagithago where the apical leaves are sharply inclined and thebasal leaves disposed in a horizontal plane. Light gradientswere evaluated at different densities and at varying stagesof development by means of probes of silicon cells. Using afilter (cut off point 72-5 µ) simultaneous measurementswere made of the infra-red and ‘visible’ radiation.The distributions of the leaf and stem area through the canopywere recorded so that light interception per unit surface couldbe assessed. The shape of the light gradient between the top and bottom ofthe canopy was sigmoidal for Gladiolus but much less so forthe other species. Attenuation of the infra-red radiation withdepth was markedly smaller. For valid estimates of the extinction coefficient (K) it isessential to include stem areas; that is on theoretical groundshalf the total stem surface. The values of K were low for allspecies and in particular for Gladiolus. The relationship betweenlight attenuation and leaf area was approximately exponential.For Gladiolus it was established that K can decrease with increasingdensity but no changes with depth in the canopy were evident.The implications of these observations in terms of canopy structureare discussed.  相似文献

NADP-malic enzyme (NADP-ME) and phosphoenolpyruvate carboxykinase(PCK) are specifically expressed in bundle sheath cells (BSCs)in NADP-ME-type and PCK-type C4 plants, respectively. Unlikethe high activities of these enzymes in the green leaves ofC4 plants, their low activities have been detected in the leavesof C3 plants. In order to elucidate the differences in the geneexpression system between C3 and C4 plants, we have producedchimeric constructs with the ß-glucuronidase (GUS)reporter gene under the control of the maize NADP-Me (ZmMe)or Zoysia japonica Pck (ZjPck) promoter and introduced theseconstructs into rice. In leaves of transgenic rice, the ZmMepromoter directed GUS expression not only in mesophyll cells(MCs) but also in BSCs and vascular cells, whereas the ZjPckpromoter directed GUS expression only in BSCs and vascular cells.Neither the ZjPck nor ZmMe promoters induced GUS expressiondue to light. In rice leaves, the endogenous NADP-Me (OsMe1)was expressed in MCs, BSCs and vascular cells, whereas the ricePck (OsPck1) was expressed only in BSCs and vascular cells.Taken together, the results obtained from transgenic rice demonstratethat the expression pattern of ZmMe or ZjPck in transgenic ricewas reflected by that of its counterpart gene in rice. The basic chromatin structure of higher plants (Vicia faba andTrillium kamtschaticum) was examined biochemically. After digestionwith micrococcal nuclease, the chromatins of these species yieldedDNA-protein components which sedimented as discrete peaks at11S, 15S, 19S, and so on in a sucrose gradient. The buoyantdensity of Vicia chromatin subunits was about 1.44 g?cm–3in CsCl. Polyacrylamide gel electrophoresis of histone fromthese subunits of Vicia and Trillium chromatins indicated thatthe 11S monomer contained very little histone H1 but a fullcomplement of all other histones, whereas the oligomers containedH1 as in the case of undigested chromatin. Therefore, the modeof organization of basic chromatin structure in higher plantsis identical with that reported with various other eukasyotes,although two plant histone components are different from thecorresponding mammalian histones, H2A and H2B, in molecularweight and amino acid composition. The results indicated alsothat chromosomes prepared from Trillium meiotic cells do notdiffer from chromatins of Trillium or Vicia somatic cells inthe sensitivity to nuclease digestion or in the size of theirsubunits. Transfer of algal cells of Chlorella regularis from 3% CO2 inair into ordinary air in the light increased external carbonicanhydrase (CA) activity as well as photosynthetic affinity forCO2 by several-fold within 2 h. Since no noticeable differencewas observed in CA activity between intact cells and cell homogenates,CA seemed to be mainly localized on the cell surface. Changesin CA activity and K?(CO2) of photosynthesis were not observedin the dark. CA induction was 50%-inhibited by incubation with10 µM DCMU during adaptation of high-CO2 cells to air,whereas it was considerably suppressed when high-CO2 cells preincubatedwith DCMU in the light for 6 h or without DCMU in the dark for24 h were used. The change in K?(CO2) of photosynthesis wasonly slightly affected by DCMU. Uncoupler like carbonylcyanide-m-chlorophenyl-hydrazone(CCCP) and inhibitors of mitochondrial respiration (KCN plussalicylhydroxamic acid) suppressed CA induction during adaptationof high-CO2 cells to low CO2 conditions. These results suggest that photosynthesis is not essential forCA induction in Chlorella regularis when some amounts of photosyntheticproducts are previously stored in the cells and respirationis active. A decrease in K?(CO2) of photosynthesis during adaptationfrom high to low CO2 was mostly independent on photosynthesis.However, light is essential for both phenomena. Cellular behaviour was examined during embryogenesis in Brassicanapus to test whether or not polyploidy occurs in the cotyledonsduring the phase of oil deposition. Nuclear DNA content, nuclearand cell size, and the mitotic index were measured in the cotyledonson various days post anthesis (dpa). In squashed monolayersfrom 15 dpa cotyledons, a polyploid (>5C) population wasdetected together with a substantial number of cells in G2 (4C).Nuclear volume was measured on sectioned tissues and, at 15dpa, the range of values from the cotyledons (40–500 *m3)contrasted with that in the vestigial suspensor and endosperm(50–> 600 µm3). At 15 dpa the nuclear volumedata suggest that whilst cells in the cotyledons were in Gland G2 many endosperm and suspensor cells were polyploid. Thus,polyploidy observed in the squashed monolayers was probablydue to contaminating endosperm/suspensor cells. At 25 and 35dpa, polyploidy was not detected; all cells were in Gl (2C)and cell area increased. The mitotic index peaked at 20 dpabefore declining and given the narrower distribution of nuclearvolumes at 25 and 35 dpa (50–300 µm3), these dataare consistent with cell arrest in Gl. Thus, polyploidy wasnot detected in the cotyledons of B. napus which differs fromwhat is known about cellular development in legume cotyledons. Our objective was to determine whether starch degradation intaproots of alfalfa (Medicago sativa L.) after defoliation wasassociated with activity and isoform complement of endo- andexo-amylases. Taproots of genetically comparable diploid (2x)and tetraploid (4x) populations and the tetraploid cultivarHi-Phy were recovered immediately after defoliation and at approx.4-d intervals thereafter. Taproot tissues were analysed forstarch concentrations and activities of endo- and exo-amylases.An electrophoretic blotting technique was used to examine amylaseisoforms. Starch degradation was most rapid in taproots of Hi-Phy,slowest in taproots of the 2x population, with the 4x populationbeing intermediate. The 4x population had a greater initialincrease in endo-amylase activity compared to the 2x population;however, Hi-Phy averaged eightfold greater endo-amylase activitythan either 2x or 4x populations. Although exo-amylase specificactivity was at least 500-fold greater than endo-amylase specificactivity in all populations, changes in endo-amylase activitywere more closely associated with trends in starch degradation.Multiple isoforms of endo- and exo-amylase were observed intaproots of all populations. Taproots of Hi-Phy contained anendo-amylase isoform that was not apparent in the 2x or 4x populationsthat may contribute to the high activity of this amylase intaproots of this cultivar. These results, although correlative,suggest an important role for endo-amylase in taproot starchhydrolysis after defoliation.
