The effects of meat-curing agents on germination and outgrowth of putrefactive anaerobe 3679h (PA 3679h) spores were studied in microcultures. Nitrite concentrations up to 0.06% at pH 6.0 or between 0.8 and 1% at pH 7.0 allowed emergence and elongation of vegetative cells but blocked cell division. The newly emerged cells then lysed. With more than 0.06% nitrite at pH 6.0 or more than 0.8 to 1% at pH 7.0, the spores lost refractility and swelled, but vegetative cells did not emerge. Even as much as 4% nitrite failed to prevent germination (complete loss of refractility) and swelling of the spores. Sodium chloride concentrations above 6% prevented complete germination (i.e., the spores retained a refractile core). In the presence of 3 to 6% sodium chloride, most of the spores germinated and produced vegetative cells, but cell division was often blocked. Sodium nitrate had no apparent effect on germination and outgrowth at concentrations up to 2%.  相似文献

Bean plants were grown under constant levels of sodium chlorideand sodium sulphate salinity, and under changing levels of sodiumsulphate salinity. Although growth was suppressed similarly by the two types ofsalinity when expressed on an osmotic basis, other parametersshowed different responses according to salinity type. Chloride-salinatedplants have thicker leaves with higher water content than thesulphate and control plants. The relative water content of thesulphate plants was somewhat lower. Transpiration rates weresuppressed more by chloride salinity. Osmotic adjustment seemsto be faster under chloride salinity and was of a differentnature. Chloride accumulated to much higher levels than sulphate.Increase in potassium ion concentration and decrease in calciumion concentration was more pronounced under sulphate salinity,but the total cation concentration in the sap was similar forall treatments. As a result, the inorganic ions' electricalbalance was more negative under chloride salinity. Leaf expansion in the changing-level treatment reflected rapidlythe variation m substrate salinity. However, total yield wassuppressed by the changing treatments to a similar extent asby constant salinity. According to other parameters the changingregime may be considered as a shorter period of exposure tosalinity.  相似文献

Unidirectional Na fluxes from frog''s striated muscle were measured in the presence of 0 to 5 mM sodium azide. With azide concentrations of 2 and 5 mM the Na efflux was markedly stimulated; the Na efflux with 5 mM azide was about 300 per cent greater than normal. A similar increase was present when all but the 5.0 mM sodium added with azide was replaced by choline. 10-5 M strophanthidin abolished the azide effect on Na24 efflux. Concentrations of azide of 1.0 mM or less had no effect on Na efflux. The Na influx, on the other hand, was only increased by 41 per cent in the presence of 5 mM NaN3. From these findings it is concluded that the active transport of Na is stimulated by the higher concentrations of azide. The hypothesis is advanced that the active transport of Na is controlled by the transmembrane potential and that the stimulation of Na efflux is produced as a consequence of the membrane depolarization caused by the azide.  相似文献

Enterococcus hirae grows in a broad pH range from 5 to 11. An E. hirae mutant 7683 lacking the activities of two sodium pumps, Na+-ATPase and Na+/H+ antiporter, does not grow in high Na+ medium at pH above 7.5. We found that 7683 grew normally in high Na+ medium at pH 5.5. Although an energy-dependent sodium extrusion at pH 5.5 was missing, the intracellular levels of Na+ and K+ were normal in this mutant. The Na+ influx rates of 7683 and two other strains at pH 5.5 were much slower than those at pH 7.5. These results suggest that Na+ elimination of this bacterium at acid pH is achieved by a decrease in Na+ entry and a normal K+ uptake.  相似文献

The use of a slowly released oral preparation of sodium chloride is described. It was given to patients and athletes to treat or prevent acute and chronic sodium chloride deficiency. The effect of NaNO2 and NaCl on the growth of 24 lactic acid bacteria strains isolated from vacuum-packed cooked ring sausages were examined by analyzing different growth parameters with Bioscreen. NaNO2 had a very limited effect on the growth of lactic acid bacteria at 50 and 100 mg/l but at 400 mg/l a more pronounced inhibitory effect was found. Bacterial growth was enhanced by 1–2% (w/v) of added NaCl, while NaCl concentrations above 3% (w/v) had a clear inhibitory effect. Leuconostoc isolates seemed to be more sensitive to sodium nitrite and sodium chloride than homofermentative lactobacilli strains. Among homofermentative lactobacilli, the strains resembling Lactobacillus curvatus were more sensitive to NaCl than those resembling Lactobacillus sake.  相似文献

Methods are described for extracting the rate constants, governing the transitions between states, for a generalized, linear three discrete state model for the Na conductance,g Na. It is shown that the problem cannot be solved given only the time course ofg Na during some test potential step with fixed initial conditions. However, if the effects of any two durations of some conditioning potential step on the peakg Na during the test step are taken into account, and the physically required assumption that the steady state is an equilibrium state is made, then the rate constants for both conditioning and test potentials can be solved for simultaneously. These methods were applied to thev 5 model of Goldman (5) in which theg Na(t) was described in terms of a generalized linear second order variable. After extensive computations involving some 20,000 combinations of experimentally determined input values, it was not found possible to extract a set of rate constants that were all real and positive as is required. It was concluded that if the transitions between states are first order and there are no subunit interactions, then the Na gating unit displays more than three states.  相似文献

The review is concerned with three Na(+)-dependent biotin-containing decarboxylases, which catalyse the substitution of CO(2) by H(+) with retention of configuration (DeltaG degrees '=-30 kJ/mol): oxaloacetate decarboxylase from enterobacteria, methylmalonyl-CoA decarboxylase from Veillonella parvula and Propiogenium modestum, and glutaconyl-CoA decarboxylase from Acidaminococcus fermentans. The enzymes represent complexes of four functional domains or subunits, a carboxytransferase, a mobile alanine- and proline-rich biotin carrier, a 9-11 membrane-spanning helix-containing Na(+)-dependent carboxybiotin decarboxylase and a membrane anchor. In the first catalytic step the carboxyl group of the substrate is converted to a kinetically activated carboxylate in N-carboxybiotin. After swing-over to the decarboxylase, an electrochemical Na(+) gradient is generated; the free energy of the decarboxylation is used to translocate 1-2 Na(+) from the inside to the outside, whereas the proton comes from the outside. At high [Na(+)], however, the decarboxylases appear to catalyse a mere Na(+)/Na(+) exchange. This finding has implications for the life of P. modestum in sea water, which relies on the synthesis of ATP via Delta(mu)Na(+) generated by decarboxylation. In many sequenced genomes from Bacteria and Archaea homologues of the carboxybiotin decarboxylase from A. fermentans with up to 80% sequence identity have been detected.  相似文献

In cardiac myocytes, action potentials are initiated by an influx of sodium (Na+) ions via voltage-gated Na+ channels. Na+ channel gain of function (GOF), arising in both inherited conditions associated with mutation in the gene encoding the Na+ channel and acquired conditions associated with heart failure, ischemia, and atrial fibrillation, enhance Na+ influx, generating a late Na+ current that prolongs action potential duration (APD) and triggering proarrhythmic early afterdepolarizations (EADs). Recent studies have shown that Na+ channels are highly clustered at the myocyte intercalated disk, facilitating formation of Na+ nanodomains in the intercellular cleft between cells. Simulations from our group have recently predicted that narrowing the width of the intercellular cleft can suppress APD prolongation and EADs in the presence of Na+ channel mutations because of increased intercellular cleft Na+ ion depletion. In this study, we investigate the effects of modulating multiple extracellular spaces, specifically the intercellular cleft and bulk interstitial space, in a novel computational model and experimentally via osmotic agents albumin, dextran 70, and mannitol. We perform optical mapping and transmission electron microscopy in a drug-induced (sea anemone toxin, ATXII) Na+ channel GOF isolated heart model and modulate extracellular spaces via osmotic agents. Single-cell patch-clamp experiments confirmed that the osmotic agents individually do not enhance late Na+ current. Both experiments and simulations are consistent with the conclusion that intercellular cleft narrowing or expansion regulates APD prolongation; in contrast, modulating the bulk interstitial space has negligible effects on repolarization. Thus, we predict that intercellular cleft Na+ nanodomain formation and collapse critically regulates cardiac repolarization in the setting of Na+ channel GOF.  相似文献

The oxidation of NH4+ by Nitrosomonas europaea was insensitive to 10 mM NaClO3 (sodium chlorate) but was strongly inhibited by NaClO2 (sodium chlorite; Ki, 2 μM). The oxidation of NO2 by Nitrobacter winogradskyi was inhibited by both ClO3 and ClO2 (Ki for ClO2, 100 μM). N. winogradskyi reduced ClO3 to ClO2 under both aerobic and anaerobic conditions, and as much as 0.25 mM ClO2 was detected in the culture filtrate. In mixed N. europaea-N. winogradskyi cell suspensions, the oxidation of both NH4+ and NO2 was inhibited in the presence of 10 mM ClO3 after a 2-h lag period, despite the fact that, under these conditions, ClO2 was not detected in the filtrate. The data are consistent with the hypothesis that, in mixed culture, NH4+ oxidation is inhibited by ClO2 produced by reduction of ClO3 by the NO2 oxidizer. The use of ClO3 inhibition of NO2 oxidation in assays of nitrification by mixed populations necessitates cautious interpretation unless it can be shown that the oxidation of NH4+ is not affected.  相似文献

In brain grey matter, excitatory synaptic transmission activates glutamate uptake into astrocytes, inducing sodium signals which propagate into neighboring astrocytes through gap junctions. These sodium signals have been suggested to serve an important role in neuro-metabolic coupling. So far, it is unknown if astrocytes in white matter—that is in brain regions devoid of synapses—are also able to undergo such intra- and intercellular sodium signalling. In the present study, we have addressed this question by performing quantitative sodium imaging in acute tissue slices of mouse corpus callosum. Focal application of glutamate induced sodium transients in SR101-positive astrocytes. These were largely unaltered in the presence of ionotropic glutamate receptors blockers, but strongly dampened upon pharmacological inhibition of glutamate uptake. Sodium signals induced in individual astrocytes readily spread into neighboring SR101-positive cells with peak amplitudes decaying monoexponentially with distance from the stimulated cell. In addition, spread of sodium was largely unaltered during pharmacological inhibition of purinergic and glutamate receptors, indicating gap junction-mediated, passive diffusion of sodium between astrocytes. Using cell-type-specific, transgenic reporter mice, we found that sodium signals also propagated, albeit less effectively, from astrocytes to neighboring oligodendrocytes and NG2 cells. Again, panglial spread was unaltered with purinergic and glutamate receptors blocked. Taken together, our results demonstrate that activation of sodium-dependent glutamate transporters induces sodium signals in white matter astrocytes, which spread within the astrocyte syncytium. In addition, we found a panglial passage of sodium signals from astrocytes to NG2 cells and oligodendrocytes, indicating functional coupling between these macroglial cells in white matter.  相似文献

“Anode-free” solid-state battery concepts are explored extensively as they promise a higher energy density with less material consumption and simple anode processing. Here, the homogeneous and uniform electrochemical deposition of alkali metal at the interface between current collector and solid electrolyte plays the central role to form a metal anode within the first cycle. While the cathodic deposition of lithium has been studied intensively, knowledge on sodium deposition is scarce. In this work, dense and uniform sodium layers of several microns thickness are deposited at the Cu|Na3.4Zr2Si2.4P0.6O12 interface with high reproducibility. At current densities of ≈1 mA∙cm−2, relatively uniform coverage is achieved underneath the current collector, as shown by electrochemical impedance spectroscopy and 3D confocal microscopy. In contrast, only slight variations of the coverage are observed at different stack pressures. Early stages of the sodium metal growth are analyzed by in situ transmission electron microscopy revealing oriented growth of sodium. The results demonstrate that reservoir-free (“anode-free”) sodium-based batteries are feasible and may stimulate further research efforts in sodium-based solid-state batteries.  相似文献

The activities are reported of sodium salts of several n-alkylsalicylic acids against Staphylococcus aureus, Pseudomonas aeruginosa, Mycobacterium phlei, and Candida albicans. The acids had alkyl substituents of 1 to 18 carbon atoms in the 3, 4, or 5 positions relative to the carboxyl group of salicylic acid. Generally, the antimicrobial properties were typical of anionic surface-active agents although the salicylates are more potent than most of these, particularly against S. aureus.  相似文献
