Summary The effects of intracellular K+ and Na+ (K+ c, Na+ c) on the Na+,K+,Cl+– cotransport pathway of HeLa cells were studied by measuring ouabain-insensitive, furosemide-sensitive Rb+ influx (JRb) at various intracellular concentrations of K+ and Na+ ([K+]c, [Na+]c). When [K+]c was increased and [Na+]c was decreased, keeping the sums of their concentrations almost constant, JRb as a function of the extracellular Rb+ or Na+ concentration ([Rb+]e, [Na+]e) was stimulated. However, the apparent K 0.5 for Rb+ e or Na+ e remained unchanged and the ratio of the apparent K +0.5 for K+ c and the apparent K i for Na+ c was larger than 1. When JRb was increased by hypertonicity by addition of 200 mM mannitol, the apparent maximum JRb increased without change in the apparent K 0.5 for Rb+ e. These results show that K+ c stimulates and Na+ c inhibits JRb, without change in the affinities of the pathway for Rb+ e and Na+ e. The affinity for K+ c is slightly lower than that for Na+ c. Hypertonicity enhances JRb without any change in the affinity for Rb+ e. We derived a kinetic equation for JRb with respect to K+ c and Na+ c and proposed a general and a special model of the pathway. The special model suggests that, in HeLa cells, JRb takes place when Rb+ e binds to the external K+ binding site of the pathway after the binding of K+ c to the internal regulatory site.We thank Mr. T. Membrane diffusion potentials induced by amphotericin B (AmB), amphotericin B methyl ester (AmE), N-fructosyl AmB (N FruAmB) and vacidin, an aromatic polyene antibiotic, in ergosterol- or cholesterol-containing egg yolk phosphatidylcholine large unilamellar vesicles (LUV), were measured in various media, in order to determine the relative selectivity of Na+, K+, Cl and other ions in these environments. Changes in the membrane potential were followed by fluorescence changes of 3,3-dipropylthiadicarbocyanine (diS-C3-(5)). Subtle changes in intercationic selectivity were monitored by measuring biionic potentials, using the fluorescent pH sensitive probe pyranine. In all the cases studied, the intereationic selectivity of the permeability pathways induced by the four antibiotics was weak compared to that of specific biological channels, though distinct differences were noted. With AmB the selectivity appeared to be concentration dependent. Above 5 × 10–7 M, the sequence determined for sterol-free small unilamellar vesicles (SUV) and cholesterol-containing SUV and LUV, Na+ > K+ > Rb+ Cs+ > Li+ (sulfate salts), corresponded closely to Eisenman selectivity sequence number VII. At 5 × 10–7 M and below the selectivity switched from Na+ > K+ to K+ > Na +. In contrast, Li+ was the most permeant ion for AmB channels in the presence of ergosterol. The selectivity between Na+ or K+ vs. Cl varied with the antibiotic. It was very strong with vacidin at concentrations below 5 × 10–7 M, smaller with AmB, nil with AmE and N FruAmB. Regulation of the Na+/K+-ATPase by insulin: Why and how?   总被引:4,自引:0,他引:4  
The sodium-potassium ATPase (Na+/K+-ATPase or Na+/K+-pump) is an enzyme present at the surface of all eukaryotic cells, which actively extrudes Na+ from cells in exchange for K+ at a ratio of 3:2, respectively. Its activity also provides the driving force for secondary active transport of solutes such as amino acids, phosphate, vitamins and, in epithelial cells, glucose. The enzyme consists of two subunits ( and ) each expressed in several isoforms. Many hormones regulate Na+/K+ -ATPase activity and in this review we will focus on the effects of insulin. The possible mechanisms whereby insulin controls Na+/K+-ATPase activity are discussed. These are tissue- and isoform-specific, and include reversible covalent modification of catalytic subunits, activation by a rise in intracellular Na+ concentration, altered Na+ sensitivity and changes in subunit gene or protein expression. Given the recent escalation in knowledge of insulin-stimulated signal transduction systems, it is pertinent to ask which intracellular signalling pathways are utilized by insulin in controlling Na+/K+-ATPase activity. Evidence for and against a role for the phosphatidylinositol-3-kinase and mitogen activated protein kinase arms of the insulin-stimulated intracellular signalling networks is suggested. Finally, the clinical relevance of Na+/K+-ATPase control by insulin in diabetes and related disorders is addressed.  相似文献

Previously, we observed that sustained activation of P2Y1 leads to inhibition of Na+,K+,Cl cotransport (NKCC) in C11 cells resembling intercalated cells from collecting ducts of the Madin-Darby canine kidney. This study examined the role of stress-activated protein kinases (SAPK) in NKCC inhibition triggered by purinergic receptors. Treatment of C11 cells with ATP led to sustained phosphorylation of SAPK such as JNK and p38. Activation of these kinases also occurred in anisomycin-treated cells. Surprisingly, we observed that compounds SP600125 and SB202190, known as potent inhibitors of JNK and p38 in cell-free systems, activated rather than inhibited phosphorylation of the kinases in C11 cells. Importantly, similarly to ATP, all the above-listed activators of JNK and p38 phosphorylation inhibited NKCC. Thus, our results suggest that activation of JNK and/or p38 contributes to NKCC suppression detected in intercalated-like cells from distal tubules after their exposure to P2Y1 agonists.  相似文献

Abstract

We report results of molecular dynamics simulations of the limiting conductance of Na2+, Cl2?, Na°, and Cl° in supercritical water using the SPC/E model for water in conjuction with our previous study (Lee et al., Chem. Phys. Lett. 293, 289 (1998)). The behavior of the limiting conductances of Na2+ and Cl2? in the whole range of water density shows almost the same trend as those of Na+ and Cl?, but the deviation from the assumed linear dependence of limiting conductances of Na2+ and Cl2? on the water density is smaller than that of Na+ and Cl?. The ratio of the limiting conductance of the divalentions to that of the corresponding monovalentions over the whole range of water density is almost constant. In the cases of Na2+ and Cl2?, the dominating factor of the number of hydration water molecules around ions in the higher-density region and the dominating factor of the interaction strength between the ions and the hydration water molecules in the lower-density region are also found as was the cases for Na+ and Cl?. These factors, however, are not so strong as for the corresponding monovalent ions because the change in the energetics, structure, and dynamics are very small mainly due to the strong Coulomb interaction of the divalent ions with the hydration water molecules. The diffusion coefficient of Na° and Cl° monotonically increases with decreasing water density over the whole range of water density. The increase of the diffusion coefficient with decreasing water density is attributed only to the dramatic decrease of the hydration number of water in the first solvation shell around the uncharged species. Among the two important competing factors in the limiting conductance of Na+ and Cl?, the effect of the number of hydration water molecules around the uncharged species is the only existing factor over the whole range of water density since the interaction strength between the uncharged species and the hydration water molecules very small through the LJ interaction. This result has confirmed the dominating factor of the number of hydration water molecules around ions in the higher-density region in the explanation of the limiting conductance of Na+ and Cl? in supercritical water at 673 K.  相似文献

Transport of 86Rb+/K+, 22Na+, 36Cl?, and [3H]indole acetic acid (IAA) has been studied on suspension-cultured cells of the parsley, Petroselinum crispum (Mill) Nym. By compartmental analysis two intracellular compartments of K+, Na+, and Cl? have been identified and ascribed to the cytoplasm and vacuole; half-times of exchange were around 200 s and 5 h, respectively. According to the Ussing-Teorell flux equation, active transport is required for the influx into the cytoplasm at the plasmalemma (K+, Cl?) and the tonoplast (K+, Na+, Cl?). The plasmalemma permeability pattern, PK:PNa:PCl=1.00:0.24:0.38, features an increased chloride permeability compared with cells from higher plant tissues. IAA uptake showed an exponential timecourse, was half-maximal after 10 min, and a linear function of the IAA concentration from 10?9 to 10?5 M. IAA and 2,4-dichlorophenoxy acetic acid reduce the apparent influx of K+, Na+, Cl? during the initial 30 min after addition and subsequently accelerate both in- and efflux of these ions. We discuss that auxins could affect the ion fluxes in a complex way, e.g. by protonophorous activity and by control of the hypothetical proton pump.  相似文献

Our previously published whole-cell patch-clamp studies on the cells of the intralobular (granular) ducts of the mandibular glands of male mice revealed the presence of an amiloride-sensitive Na+ conductance in the plasma membrane. In this study we demonstrate the presence also of a Cl conductance and we show that the sizes of both conductances vary with the Cl concentration of the fluid bathing the cytosolic surface of the plasma membrane. As the cytosolic Cl concentration rises from 5 to 150 mmol/liter, the size of the inward Na+ current declines, the decline being half-maximal when the Cl concentration is approximately 50 mmol/liter. In contrast, as cytosolic Cl concentration increases, the inward Cl current remains at a constant low level until the Cl concentration exceeds 80 mmol/liter, when it begins to increase. Studies in which Cl in the pipette solution was replaced by other anions indicate that the Na+ current is suppressed by intracellular Br-, Cl and NO 3 - but not by intracellular I-, glutamate or gluconate. Our studies also show that the Cl conductance allows passage of Cl and Br- equally well, I-less well, and NO 3 - , glutamate and gluconate poorly, if at all. The findings with NO 3 - are of particular interest because they show that suppression of the Na+ current by a high intracellular concentration of a particular anion does not depend on actual passage of that anion through the Cl conductance. In mouse granular duct cells there is, thus, a reciprocal regulation of Na+ and Cl conductances by the cytosolic Cl concentration. Since the cytosolic Cl concentration is closely correlated with cell volume in many epithelia, this reciprocal regulation of Na+ and Cl conductances may provide a mechanism by which ductal Na+ and Cl transport rates are adjusted so as to maintain a stable cell volume.This project was supported by the National Health and Medical Research Council of Australia. Summary Na–K–Cl cotransport stoichiometry and affinities for Na, K and Cl were determined in flounder intestine. Measurement of simultaneous NaCl and RbCl influxes resulted in ratios of 2.2 for Cl/Na and 1.8 for Cl/Rb. The effect of Na and Rb on Rb influx showed first order kinetics withK 1/2 values of 5 and 4.5mm and Hill coefficients of 0.9 and 1.2, respectively. The effect of Cl on rubidium influx showed a sigmoidal relationship withK 1/2 of 20mm and a Hill coefficient of 2.0. The effects of variations in Na and Cl concentration on short-circuit current (I sc) were also determined. TheK 1/2 for Na was 7mm with a Hill coefficient of 0.9 and theK 1/2 for Cl was 46mm with a Hill coefficient of 1.9. Based on the simultaneous influx measurements, a cotransport stoichiometry of 1Na1K2Cl is concluded. The Hill coefficients for Cl suggest a high degree of cooperativity between Cl binding sites. Measurements of the ratio of net Na and Cl transepithelial fluxes under short-circuit conditions (using a low Na Ringer solution to minimize the passive Na flux) indicate that the Cl/Na flux ratio is approximately 21. Therefore Na recycling from serosa to mucosa does not significantly contribute to theI sc. Addition of serosal ouabain (100 m) inhibited Rb influx, indicating that Na–K–Cl cotransport is inhibited by ouabain. This finding suggests that a feedback mechanism exists between the Na–K-ATPase on the basolateral membrane and the apical Na–K–2Cl cotransporter.  相似文献

Summary Experiments were performed usingin vitro perfused medullary thick ascending limbs of Henle (MTAL) and in suspensions of MTAL tubules isolated from mouse kidney to evaluate the effects of arginine vasopressin (AVP) on the K+ dependence of the apical, furosemide-sensitive Na+:Cl cotransporter and on transport-related oxygen consumption (QO2). In isolated perfused MTAL segments, the rate of cell swelling induced by removing K+ from, and adding onemm ouabain to, the basolateral solution [ouabain(zero-K+)] provided an index to apical cotransporter activity and was used to evaluated the ionic requirements of the apical cotransporter in the presence and absence of AVP. In the absence of AVP cotransporter activity required Na+ and Cl, but not K+, while in the presence of AVP the apical cotransporter required all three ions.86Rb+ uptake into MTAL tubules in suspension was significant only after exposure of tubules to AVP. Moreover,22Na+ uptake was unaffected by extracellular K+ in the absence of AVP while after AVP exposure22Na+ uptake was strictly K+-dependent. The AVP-induced coupling of K+ to the Na+:Cl cotransporter resulted in a doubling in the rate of NaCl absorption without a parallel increase in the rate of cellular22Na+ uptake or transport-related oxygen consumption. These results indicate that arginine vasopressin alters the mode of a loop diuretic-sensitive transporter from Na+:Cl cotransport to Na+:K+:2Cl cotransport in the mouse MTAL with the latter providing a distinct metabolic advantage for sodium transport. A model for AVP action on NaCl absorption by the MTAL is presented and the physiological significance of the coupling of K+ to the apical Na+:Cl cotransporter in the MTAL and of the enhanced metabolic efficiency are discussed.  相似文献

Cation–Cl? cotransporters (CCCs) are integral membrane proteins which catalyze the coordinated symport of Cl? with Na+ and/or K+ ions in plant and mammalian cells. Here we describe the first Saccharomyces cerevisiae CCC protein, encoded by the YBR235w open reading frame. Subcellular localization studies showed that this yeast CCC is targeted to the vacuolar membrane. Deletion of the YBR235w gene in a salt-sensitive strain (lacking the plasma-membrane cation exporters) resulted in an increased sensitivity to high KCl, altered vacuolar morphology control and decreased survival upon hyperosmotic shock. In addition, deletion of the YBR235w gene in a mutant strain deficient in K+ uptake produced a significant growth advantage over the parental strain under K+-limiting conditions, and a hypersensitivity to the exogenous K+/H+ exchanger nigericin. These results strongly suggest that we have identified a novel yeast vacuolar ion transporter mediating a K+–Cl? cotransport and playing a role in vacuolar osmoregulation. Considering its identified function, we propose to refer to the yeast YBR235w gene as VHC1 (vacuolar protein homologous to CCC family 1).  相似文献

The kccDHS1 allele of kazachoc (kcc) was identified as a seizure-enhancer mutation exacerbating the bang-sensitive (BS) paralytic behavioral phenotypes of several seizure-sensitive Drosophila mutants. On their own, young kccDHS1 flies also display seizure-like behavior and demonstrate a reduced threshold for seizures induced by electroconvulsive shock. The product of kcc shows substantial homology to KCC2, the mammalian neuronal K+–Cl cotransporter. The kccDHS1 allele is a hypomorph, and its seizure-like phenotype reflects reduced expression of the kcc gene. We report here that kcc functions as a K+–Cl cotransporter when expressed heterologously in Xenopus laevis oocytes: under hypotonic conditions that induce oocyte swelling, oocytes that express Drosophila kcc display robust ion transport activity observed as a Cl-dependent uptake of the K+ congener 86Rb+. Ectopic, spatially restricted expression of a UAS-kcc+ transgene was used to determine where cotransporter function is required in order to rescue the kccDHS1 BS paralytic phenotype. Interestingly, phenotypic rescue is largely accounted for by targeted, circumscribed expression in the mushroom bodies (MBs) and the ellipsoid body (EB) of the central complex. Intriguingly, we observed that MB induction of kcc+ functioned as a general seizure suppressor in Drosophila. Drosophila MBs have generated considerable interest especially for their role as the neural substrate for olfactory learning and memory; they have not been previously implicated in seizure susceptibility. We show that kccDHS1 seizure sensitivity in MB neurons acts via a weakening of chemical synaptic inhibition by GABAergic transmission and suggest that this is due to disruption of intracellular Cl gradients in MB neurons.Mushroom body (MB) expression of the kazachoc (kcc) K+–Cl cotransporter is shown here to rescue seizure-sensitive phenotypes in Drosophila through an effect on GABAergic fast synaptic inhibition. Heretofore, considerable interest has focused on the MB because of its essential role in olfactory learning and memory (Heisenberg 2003; Davis 2005; Keene and Waddell 2007; Berry et al. 2008). The MB occupies a central position in the fly nervous system, integrating incoming olfactory, mechanical, taste, and visual sensory signals and then sorting the distribution of outgoing motor signals (Heisenberg 2003). Short- and long-term alteration of individual nerve cell physiology in the MB is thought to form the basis of learning and memory (Davis 2005; Keene and Waddell 2007; Berry et al. 2008). A role for the MB in seizure susceptibility has not previously been suspected. Here we suggest that the orderly arrangements of axons and neuropile of MB Kenyon cells (KCs) not only facilitate learning and memory, but also provide the type of anatomical substrate in flies that is thought to be essential for seizure spread in the mammalian brain (Hauser and Hesdorffer 1990; Traub and Miles 1991).Inhibitory synaptic transmission in Drosophila is thought to be mediated primarily by GABAergic neurons found throughout the CNS at all stages of development (Buchner et al. 1988; Jackson et al. 1990; Harrison et al. 1996; Yasuyama et al. 2002). γ-aminobutyric acid (GABA) is synthesized from glutamate via a conserved glutamic acid decarboxylase encoded by the Drosophila Gad1 gene (Jackson et al. 1990; Buchner 1991). GABAergic activity is limited by sequestering extracellular GABA back into presynaptic neurons by GABA transporters that are sensitive to inhibition by dl-2,4-diaminobutyric acid, nipecotic acid, and valproic acid (Neckameyer and Cooper 1998; Leal et al. 2004). Three ionotropic GABAA receptor subunits have been identified in Drosophila and are encoded by the Rdl, LCCH3, and GRD loci (Hosie et al. 1997). When expressed heterotopically in Xenopus oocytes, the best studied of these, Rdl, forms GABA-gated Cl channels that are sensitive to block by picrotoxin (ffrench-Constant et al. 1991, 1993; Zhang et al. 1995). Inhibitory Cl currents are dependent on maintenance of Cl gradients, particularly in low intracellular Cl concentrations. In the fly, Cl gradients appear to be maintained by the kcc K+–Cl cotransporter (Hekmat-Scafe et al. 2006).Chemical synaptic transmission onto MB neurons has been examined in dissociated KCs in primary culture (Su and O''dowd 2003). Spontaneous miniature excitatory postsynaptic currents (EPSCs) are mediated mainly by nicotinic acetylcholine (ACh) receptors. Miniature inhibitory postsynaptic currents (IPSCs) appear to be mediated primarily by picrotoxin-sensitive GABAA receptors, probably encoded by Rdl (Su and O''dowd 2003; Harrison et al. 1996). In vivo, cholinergic inputs to the MB are thought to arise primarily from antennal lobe projection neurons (Yasuyama et al. 2002). Two antennal lobe neurons that project to the MB, the anterior paired lateral (APL) neurons, were recently shown to be GABAergic (Liu and Davis, 2009). Additional GABAergic inputs to the Drosophila MB seem likely; in locust they appear to come from a poorly understood region of the brain called the lateral horn, which is itself also driven by antennal lobe projection neurons (Perez-Orive et al. 2002).Previously, we identified the kccDHS1 partial loss-of-function mutation as a seizure enhancer that also causes increased seizure sensitivity in young flies (Hekmat-Scafe et al. 2006). The kcc product shows homology to the mammalian KCC2 K+–Cl cotransporter, and we inferred that a decrease in inhibitory synaptic strength is responsible for causing the seizure phenotypes. In this study, we describe our search for identifying the source of these vulnerable inhibitory synapses and report that they appear to lie primarily in the MBs of the Drosophila brain. Further, we speculate on the possibility of their involvement in synaptic plasticity functions of the MB.  相似文献

Summary The intracellular distribution of Na+, K+, Cl and water has been studied in the Ehrlich ascites tumor cell. Comparison of the ion and water contents of whole cells with those of cells exposed to La3+ and mechanical stress indicated that La3+ treatment results in selective damage to the cell membrane and permits evaluation of cytoplasmic and nuclear ion concentrations. The results show that Na+ is sequestered within the nucleus, while K+ and Cl are more highly concentrated in the cell cytoplasm. Reduction of the [Na+] of the incubation medium by replacement with K+ results in reduced cytoplasmic [Na+], increased [Cl] and no change in [K+]. Nuclear concentrations of these ions are virtually insensitive to the cation composition of the medium. Concomitant measurements of the membrane potential were made. The potential in control cells was –13.7 mV. Reduction of [Na+] in the medium caused significant depolarization. The measured potential is describable by the Cl equilibrium potential and can be accounted for in terms of cation distributions and permeabilities. The energetic implications of the intracellular compartmentation of ions are discussed.  相似文献

Summary In the epithelium of rabbit gallbladder, in the nominal absence of bicarbonate, intracellular Cl activity is about 25mm, about 4 times higher than intracellular Cl activity at the electrochemical equilibrium. It is essentially not affected by 10–4 m acetazolamide and 10–4 m 4-acetamido-4-isothiocyanostilbene-2,2-disulfonate (SITS) even during prolonged exposures; it falls to the equilibrium value by removal of Na+ from the lumen without significant changes of the apical membrane potential difference. Both intracellular Cl and Na+ activities are decreased by luminal treatment with 25mm SCN; the initial rates of change are not significantly different. In addition, the initial rates of change of intracellular Cl activity are not significantly different upon Na+ or Cl entry block by the appropriate reduction of the concentration of either ion in the luminal solution. Luminal K+ removal or 10–5 m bumetanide do not affect intracellular Cl and Na+ activities or Cl influx through the apical membrane. It is concluded that in the absence of bicarbonate NaCl entry is entirely due to a Na+–Cl symport on a single carrier which, at least under the conditions tested, does not cotransport K+.  相似文献
