A hair-baiting method was used in laboratory study of the influence on the composition of the keratinolytic and non-keratinolytic mycobiota in sewage of adding the ammonium nitrogen sources, peptone, ammonia water and urea. It was demonstrated that, independent of the nitrogen source, the ammonium nitrogen concentration was the critical factor affecting the fungal composition. Ammonium prevented or markedly inhibited the growth of Trichophyton terrestre, with its teleomorph Arthroderma quadrifidum; other geophilic dermatophytes and non-keratinolytic fungi. It also favoured growth of Chrysosporium keratinophilum and some other species, mainly in the genus Chrysosporium. The composition of the keratinolytic mycobiota can be an indicator of sludge organic matter stabilization, and used in relation to sewage sludge at wastewater treatment plants and in reclamation practice.  相似文献

In order to estimate the prevalence of dermatophytes and other fungi on healthy children hairs and nails, 92 hair samples and 85 nail samples (groups of 10 finger nails from each child) were collected from 5 nurseries (children aged 9 months up to 4 years) in Assiut city. From hair samples 22 species were collected, Trichophyton (2 species) and Microsporum (2 species) were the only recovered dermatophytes in addition to well known keratinophilic genus Chrysosporium (4 species). From nail samples, 18 species were identified, Trichophyton was represented by 4 species, Microsporum, 2 species and Chrysosporium, 4 species. Nucleotide sequences of chitin synthase 1 (CHS1) gene of dermatophytes, Arthroderma benhamiae, A. simii, A. vanbreuseghemii, Trichophyton mentagrophytes var. interdigitale (T. interdigitale), and T. rubrum were analyzed for their phylogenetic relationship. About 620-bp genomic DNA fragments of the CHS1 gene were amplified from these dermatophytes by polymerase chain reaction (PCR) and sequenced. The CHS1 nucleotide sequences of these five dermatophytes showed more than 90% similarity between the species. The phylogenetic analysis of their sequences revealed that A. benhamiae, A. simii, A. vanbreuseghemii, and T. rubrum were genetically distinct from one another, but T. interdigitale was genetically very close to A. vanbreuseghemii. On the other hand, a specific restriction endonuclease site of HinfI was present in the CHS1 gene fragment of T. rubrum but not in those of A. benhamiae, A. simii, A. vanbreuseghemii and T. interdigitale. The molecular analysis of CHS1 genes will provide useful information for the identification of these Trichophyton species and the understanding of their evolution. Keratinophilic fungi are a highly specialized, keratin-degrading ecological group. They live in natural environments, mostly in the keratinrich remains of dead animal in the soil. We investigated species diversity in four types of soils with different physico-chemical properties. The strain material was identified based on morphological characters. Different representatives of Chrysosporium and geophilic dermatophytes dominated depending on soil pH. Geophilic dermatophytes were represented by one species, Trichophyton ajelloi, and the Chrysosporium group was represented by Chrysosporium keratinophilum. The frequency of Trichophyton ajelloi increased with an increase in pH, and it reached the maximum in strongly acidic soil (podzol), unlike the Chrysosporium group. The frequency of Chrysosporium keratinophilum was positively correlated with the content of humus, nitrogen, CaCO3 and phosphorus in the soils.  相似文献

One hundred and fifty-eight soil samples were collected from various areas of four districts of Kerala and screened for prevalence of keratinophilic fungi and related dermatophytes. From the positive samples (60.75%), a total of eight genera with 15 species were isolated viz., Arthroderma simii (0.63%), Chrysosporium indicum (20.25%), C. keratinophilum (6.96%), C. lobatum (1.26%), C. pannicola (1.26%), C. tropicum (5.06%), Chrysosporium state of Arthroderma cuniculi (1.26%), Chrysosporium state of Ctenomyces serratus (2.53%), Gymnascella hyalinospora (1.26%), Malbranchea aurantiaca (0.63%) M. fulva (1.26%), Microsporum gypseum complex (12.65%), Pseudogymnoascus roseus (1.26%), Trichophyton mentragrophytes (1.26%), and T. terrestre (3.16%).This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

In this paper, four fungi collected in Nansei Islands are reported. One is Strasseria garciniae, which must be transferred to the genus Phyllosticta. This fungus was collected in Okinawa Main Island and Iriomote Island. The other three fungi are newly added to the Japanese fungal flora. Phyllosticta ghaesembillae on Codiaeum variegatum and Cercospora asplenii on Asplenium antiquum were collected in Yoron Island. The last one, Coniella australiensis on Eucalyptus robusta was collected in Okinawa Main Island. Their morphology and symptoms on the host plant are described, with some mycological notes.  相似文献

Recently, reef-building coral populations have been decreasing worldwide due to various disturbances. Population genetic studies are helpful for estimating the genetic connectivity among populations of marine sessile organisms with metapopulation structures such as corals. Moreover, the relationship between latitude and genetic diversity is informative when evaluating the fragility of populations. In this study, using highly variable markers, we examined the population genetics of the broadcast-spawning coral Acropora digitifera at 19 sites in seven regions along the 1,000 km long island chain of Nansei Islands, Japan. This area includes both subtropical and temperate habitats. Thus, the coral populations around the Nansei Islands in Japan are northern peripheral populations that would be subjected to environmental stresses different from those in tropical areas. The existence of high genetic connectivity across this large geographic area was suggested for all sites (F ST≤0.033) although small but significant genetic differentiation was detected among populations in geographically close sites and regions. In addition, A. digitifera appears to be distributed throughout the Nansei Islands without losing genetic diversity. Therefore, A. digitifera populations in the Nansei Islands may be able to recover relatively rapidly even when high disturbances of coral communities occur locally if populations on other reefs are properly maintained.  相似文献

Among 75 camels showing skin lesions, 48% were positive for fungal infection. The younger individuals were more susceptible to this infection. Sixteen species belonging to nine genera of keratinophilic and cycloheximide-resistant fungi were recovered from diseased camels.Trichophyton, Microsporum andChrysosporium were the most common genera.T. verrucosum appeared to be the main causer of ringworm in small camels whileT. mentagrophytes infected older ones. Camel skin presents a suitable habitat for the growth of some dermatophytes and other potentially pathogenic fungi.  相似文献

One of the basic tasks in the field of biodeterioration is to recognise the microbial species responsible for the destruction of particular substrates, and to identify factors impacting the level of damage caused by microorganisms. Even in 1839, it was known that there existed some fungi capable of attacking keratinized tissues, although, at that time, only dermatophytes were recognised. The relevant literature pertaining to microbiological deterioration of keratinous substrates includes 299 fungi belonging to 100 genera. Representatives of the genera Aspergillus, Penicillium, Chrysosporium, Fusarium, Microsporum, Trichophyton, and Acremonium appear to be the most common. Of the 299 species collected, 107 belonging primarily to the Onygenales and Eurotiales are pathogenic to humans. The research focusing on microbial ability to colonize and destroy keratinous materials has been carried out mainly on sheep fleece, hairs, and feathers, but only a few authors have dealt with woollen fabric biodeterioration, which is of particular significance for the preservation of antique textiles.  相似文献

Résumé Les auteurs rapportent l'observation d'une dermatomycose humaine dûe àTrichophyton gallinae, ce cas étant le troisième en Roumanie, dont on a pu isoler ce dermatophyte. Avec cette souche et une autre antérieure (isolée deux ans auparavant), les auteurs ont fait des investigations cliniques et mycologiques, afin de préciser l'identité probable entre leTrichophyton gallinae et leTr. quinckeanum. Basés sur la ressemblance macro- et microscopique des leurs colonies, sur les lésions expérimentales obtenues par l'inoculation à l'homme, au cobaye et à la souris blanche, de même que sur l'aspect variable des lésions humaines, dont ces deux expèces sont responsables, les auteurs arrivent à la conclusion qu'il serait possible que les deux dermatophytes soient un et le même champignon. Le caractère du pigment diffusible — dit on — particulier pourTr. gallinae — n'est pas un argument assez puissant, selon l'opinion des auteurs, pour faire de ce dermatophyte une espèce indépendente dans la classification des dermatophytes.
Summary Having the opportunity to observe a human ringworm infection, caused byTrichophyton gallinae (the third in Rumania up to now), the authors report their conclusions concerning the position of this fungus in the classification of dermatophytes. Based on their mycological research made with two stains ofTrichophyton gallinae and others belonging toTr. quinckeanum and taking into consideration the resemblance between the macro- and microscopical features of their colonies, the same as for the clinical aspect of the human and experimental lesions it provoked, the authors point out the possible identity of the two species. The red pigment diffusing through the medium, characteristic (?) — one says — forTr. gallinae (but not current, as the authors observed) could not be a strong proof, in the authors' opinion, to classifyTr. gallinae as a single and different species fromTr. quinckeanum.

The ubiquinone (coenzyme Q: Q) system of 17 strains of the form-genusChrysosporium was analyzed by high performance liquid chromatography (HPLC) and found to show a heterogeneous distribution of the major ubiquinone. Q-9, Q-10 or Q-10(H2) was found to be the major ubiquinone in 3, 9 and 5 strains, respectively. It was further demonstrated that the teleomorphs of the species characterized by Q-9 and Q-10 could be classified into two separate families, Arthrodermataceae (Q-9) and Onygenaceae (Q-10), which were defined within the revised order Onygenales by Currah. Teleomorphs ofChrysosporium species having Q-10(H2) have not been found. This paper also includes the ubiquinone system of dermatophytes which relate to the form-genusChrysosporium morphologically.  相似文献

Hair and scalp mycobiota of 1389 clinically normal children aged 6–12 years attending 12 schools in the Nablus District, Palestinian Authority, was assessed on three occasions over 8-month period (October 1998–May 1999) using the hair brush technique. One hundred and one fungal species belonging to 33 genera were recovered: 6 dermatophytes, 16 dermatophyte-like keratinophilic fungi, and 79 other keratinophilic fungal species. Species varied considerably in their frequency of occurrence and abundance based on their relative importance values (RIVs). The most frequent and abundant species were: Cladosporium cladosporioides, Cl. herbarum, Penicillium chrysogenum and Aspergillus flavus. Microsporum canis, Aphanoascus fulvescence and Chrysosporum sulfureum were the most frequent and abundant species of all dermatophytes and dermatophyte-like keratinophilic fungi recovered. The most frequent and abundant dermatophytes in different communities were M. canis in rural (RIV 0.87) and urban children (0.45), and Trichophyton violaceum (1.41) in refugee camp children. Chrysosporium species were the most frequent and abundant dermatophyte-like keratinophilic fungus in children from all localities followed by Aphanoascus fulvescence. Comparable results on the frequency and abundance of human hair and scalp mycobiota component fungi were obtained based on age group and sex of children. Higher number of species was recovered in spring months (73 species) than in autumn (57) and winter (44) months. Similar occurrence pattern was also noted for dermatophyte-like keratinophilic species and dermatophytes. Higher percentages of children with moderate (11–50) and heavy (≤ 50) spore loads (7.54 and 0.73, respectively) were found in urban school children community than in rural and refugee camp school children (4.7 and 0.1, respectively). Also significantly higher light (1–10) spore load percentages were found in rural (63.67) and refugee camp (62.9) than in urban children (52.6). Of all localities, school children with light spore load comprised the highest percentage of the children examined (37.4), followed by moderate (6.13), and heavy (0.41) spore load categories. However, children with undetected spore load comprised 36.05% of all children. Spore load distribution did not show clear seasonal variations in the study period. Higher percentages of moderate and heavy spore loads were found in male children (8.72 and 0.69, respectively) than in female children (3.4 and 0.1, respectively). However, higher percentages of undetected (38.3) or light spore loads (58.4) were found in females than in males (34.04 and 56.53, respectively). In the first report of the present series, six species collected from Nansei Islands were discussed. Of these, one found onElaeocarpus japonica was described as a new species,Pseudocercospora elaeocarpicola. Pseudocercospora plumeriae onPlumeria rubra was transferred from the genusCercospora and added to the Japanese fungous flora, together withCercosporidium bougainvilleae onBougainvillea spectabilis. One of the three remaining species,Pseudcercospora punicae onPunica granatum was newly recorded from Nansei Islands. On the other two species, namelyP. circumscissa andP. fukuokaensis, some additional notes concerning distribution and host range were described.  相似文献

NADP-Glutamate dehydrogenase (NADP-GDH) located at the interface of carbon and nitrogen metabolism has the potential to dictate fungal carbon flux. NADP-GDH from Aspergillus terreus, itaconate producer and an opportunistic pathogen, was purified to homogeneity using novel reactive dye-affinity resins. The pure enzyme was extensively characterized for its biochemical and kinetic properties and compared with its well studied Aspergillus niger counterpart. The A. terreus NADP-GDH was more stable and showed non-competitive ammonium inhibition with respect to glutamate. It exhibited hyperbolic 2-oxoglutarate saturation albeit with a weak substrate inhibition. This is in contrast to the allosteric nature of the enzyme from other Aspergilli. Differential susceptibility to chymotrypsin is also consistent with the absence of substrate cooperativity and conformational changes associated with A. terreus NADP-GDH. The non-allosteric nature of A. terreus NADP-GDH provides a unique opportunity to assess the contribution of allostery in metabolic regulation.  相似文献

Bacterial strains (93 isolates) capable of growth on full-strength nutrient media were isolated from 86 fungal fruit bodies collected in the Moscow region. Antimicrobial activity of the endobiont isolates against 12 bacterial and fungal test strains (including drug-resistant ones) was studied in submerged cultures. Most of the strains (84.9%) were found to produce antibiotic compounds with different antimicrobial properties, including antifungal activity in 18.3% of the strains. Morphological characteristics and analysis of the 16S rRNA gene sequences were used to determine the taxonomic position of 16 bacterial strains of the following 10 species: Bacillus subtilis, Ewingella americana, Pseudomonas sp., Stenotrophomonas maltophilia, as well as Achromobacter spanius, B. licheniformis, Hafnia paralvei, Micrococcus terreus, Nocardia coeliaca, and St. rhizophila, which have not been previously known to be endobionts of basidiomycete fruit bodies. Antimicrobial activity of A. spanius, E. americana, H. paralvei, M. terreus, N. coeliaca, and St. rhizophila has not been reported previously. Complex mechanisms of symbiotic relations between fungi and bacteria, including those associated with antibiotic formation, probably developed in the course of co-evolution.  相似文献

为了解黑龙江省药用植物根际土壤真菌的种群结构和区系分布特点,于2010年7月份和10月份,在黑龙江省的伊春、铁力、绥化、哈尔滨、牡丹江和佳木斯6个中药材产区共采集土壤样品220份,所采集的药用植物种类主要有五味子、平贝母、刺五加、党参、防风、柴胡、桔梗、黄芩等14种.经稀释平板法和土壤颗粒平板法分离共获得1016株真菌,经形态鉴定归为35属86种,其中接合菌7属10种,占7.78%;子囊菌1属2种,占0.69%;无性型真菌27属74种,占70.76%,其余20.77%的菌株为不产孢真菌.试验结果表明,黑龙江省药用植物根际土壤真菌的种群多样性丰富,其中青霉属Penicillium、曲霉属Aspergillus、木霉属Trichoderma、镰孢菌属Fusarium是优势种群,粘帚霉属Gliocladium、金孢属Chrysosporium、毛霉属Mucor、枝孢属Cladosporium、枝顶孢属Acremonium、根霉属Rhizopus是亚优势种群.不同药用植物根际土壤真菌区系的结构和组成存在一定的差异.除无孢类群外,青霉属Penicillium、曲霉属Aspergillus、木霉属Trichoderma和镰孢菌属Fusarium是14种药用植物根际土壤真菌的优势菌群.五味子、平贝母和柴胡是黑龙江省种植的主要中药材,它们在6个采样地点间的真菌种群的多样性水平存在差异,其中伊春地区的多样性指数(H’=2.9574)和丰富度指数(R=5.6683)最高,而佳木斯地区的均匀度指数(J=0.9200)最高.不同地区的相似性水平也存在差异,其中牡丹江与绥化的药用植物根际土壤真菌种群组成之间的相似性系数最高(Cj=0.6315),牡丹江与哈尔滨的相似性最低(Cj=0.3704).  相似文献

Chrysosporium species were isolated from soil and keratinized material. Primary isolation was performed following the general method of hair baiting on modified Czapek-agar media with washed, defated and sterilized human hair fragments added. Strains were maintained in test tubes of potato dextrose agar at 29 °C and cultivated on phytone yeast extract agar at 28 °C for 14 days for identification. Isolates were characterized using Van Oorschot's key. Keratinolytic activity was expressed following a subjective scale representing degree/severity of attack upon hair surface and presence of fungal structures observed in substrate. Culture results and characterization methods were effective for soil Chrysosporium strain isolation. A new hair attack mode is described. Of 71 keratinolytic fungal isolates, eight (12%) Chrysosporium species were identified. Island syndrome, previously established for isolation process of insular vertebrates' populations, have been adapted to insular parasites communities, termed parasite island syndromes. In this work, were studied for the first time the insular syndromes for nidicolous ectoparasites of the bird species, Turdus merula, Sylvia atricapilla, Fringilla coelebs and Erithacus rubecula from Azores and the mainland Portugal. Flea species were only recorded on Azorean birds, namely Dasypsyllus gallinulae and Ctenocephalides felis felis, known as not host-specific parasites. In the absence of shared flea species between mainland and islands birds, a comparison among our fleas prevalence to Azores Islands and mainland fleas prevalence, recorded to others European studies, showed that Azorean host populations undergo higher prevalence than the mainland one. This result was consistent with parasite island syndromes predictions recorded to ectoparasites, hippoboscid flies and chewing lice, that fleas have higher prevalence on the Azores Islands compared to mainland Portugal. However, our results provide a new perspective to parasite island syndromes assumptions, namely in the context of nidicolous ectoparasites that spend only brief periods on the hosts' body.  相似文献

To establish effective locations and sizes of potential protected areas for reef ecosystems, detailed information about source and sink relationships between populations is critical, especially in archipelagic regions. Therefore, we assessed population structure and genetic diversity of Acropora tenuis, one of the dominant stony coral species in the Pacific, using 13 microsatellite markers to investigate 298 colonies from 15 locations across the Nansei Islands in southwestern Japan. Genetic diversity was not significant among sampling locations, even in possibly peripheral locations. In addition, our results showed that there are at least two populations of A. tenuis in the study area. The level of genetic differentiation between these populations was relatively low, but significant between many pairs of sampling locations. Directions of gene flow, which were estimated using a coalescence‐based approach, suggest that gene flow not only occurs from south to north, but also from north to south in various locations. Consequently, the Yaeyama Islands and the Amami Islands are potential northern and southern sources of corals. On the other hand, the Miyako Islands and west central Okinawa Island are potential sink populations. The Kerama Islands and the vicinity of Taketomi Island are potential contact points of genetic subdivision of coral populations in the Nansei Islands. We found that genetic population structure of A. tenuis in the Nansei Islands is more complex than previously thought. These cryptic populations are very important for preserving genetic diversity and should be maintained.  相似文献

A simple and novel procedure for the acceleration of fungal spore production was developed. A net of hydrophobic polymer such as polypropylene (PP) and polytetrafluoroethylene (PTFE) was embedded in a nutrient agar plate, and effect of the polymer net on spore production by 6 fungal strains, such as Aspergillus terreus, Penicillium multicolor, and Trichoderma virens were estimated. The effect of hydrophobic polymer net was insufficient in a liquid-surface immobilization (LSI) system with fungal cells immobilized on a ballooned microsphere layer formed on a liquid medium surface. On the other hand, the embedding of a PTFE net in an agar plate remarkably enhanced the spore production in all 6 strains tested to produce 2.0–8.5 × 107 spores/cm2-agar plate surface. Especially, the spore production by A. terreus ATCC 20542 in the presence of a PTFE net was 7.7 times as much than that in no net. Positive correlations between the hydrophobicity of net and the spore production were observed in all 6 strains (R2, 0.653–0.999).  相似文献
