Estrdiol-17β induced a significant increase in the uterine thymidine kinase activity with a characteristic isozyme pattern 30 h after injection into immature rats. Testosterone propionate also revealed a similar increase. Following combined injection of estradiol-17β and testosterone propionate, the overall and separate isozyme activities of thymidine kinase increased to nearly the total amount of those when each hormone was injected separately.  相似文献

The purpose of this study was to evaluate the most appropriate rat age for the start of administration, and the effect of weaning, in the immature rat uterotrophic assay using ethynylestradiol (EE). Animals weaned on postnatal day (PND) 20 were administered subcutaneously EE at doses of 0.06-6 micrograms/kg/day for 3 days beginning on PND 21, 23 or 25. EE at the same doses was also administered to rats weaned on PND 17 or 20 from PND 21 for 3 days. Although uterine weight was significantly increased in the rats given 0.6-6 micrograms/kg EE in both of the studies, the percentage increase relative to the control in each group given EE from PND 21 and weaned on PND 20 was higher than in those groups given EE from PND 23 or 25, and the group weaned on PND 17.  相似文献

The suppressive effects of anti-allotype antibody on splenic lymphocytes of adult (>3 months of age) and newborn (1-week-old) rabbits were compared in vitro. An approximately 10-fold lower concentration of anti-b4 sufficed to modulate completely membrane-bound b4 immunoglobulin (Ig) when newborn cells were pulse-treated for 2 or 24 hr than when adult cells were tested. In contrast to splenic lymphocytes from adults, which regenerated most of the original proportion of b4+ cells in culture following treatment with up to 300 μg of anti-b4 for 24 hr (4), immature lymphocytes were susceptible to irreversible modulation of membrane-bound Ig even when lower concentrations of anti-b4 were used. However, under conditions permitting reversible modulation of membrane b4, lymphocytes of newborns regenerated the membrane product more rapidly than did adult cells. Both mature and immature splenocytes were shown to be capable of some level of b4 synthesis even in the continuing presence of anti-b4. No evidence for susceptibility to complement-mediated killing of newborn spleen cells by anti-allotypic antibodies was obtained. The observations reported here support the concept that the greater sensitivity of B lymphocytes from the newborn to succumb to irreversible suppressive effects by anti-allotype antibody plays a significant role in the restriction for induction of allotype suppression to the perinatal period.  相似文献

The epididymis and male accessory sex organs (vesicular gland, prostate, and bulbourethral gland) of sexually immature rabbits contain a functional estrogen receptor system which is regulated in an organ-specific manner by various hormones. In both intact and castrated animals, acute estrogen challenge causes depletion of estrogen receptor from the cytosolic fraction and its appearance in the nuclear fraction of these tissues. A considerable amount of unoccupied nuclear receptor was detected both before and after estrogen challenge. An estrogen-activated, receptor-processing mechanism is operable in these organs since chronic treatment (daily for 14 days) with estradiol benzoate modified the levels of total estrogen receptor, and altered the relative amounts of occupied to unoccupied nuclear receptor present following estrogen challenge. Chronic treatment with estradiol benzoate, Tamoxifen, and testosterone propionate (alone and in combination) had differential, organ-specific effects on the ability of subsequent estrogen challenge to cause accumulation of nuclear receptor. The vesicular gland was the most responsive to estrogen treatment and the bulbourethral gland the least responsive.  相似文献

本文拟探讨缺血预处理(ischemic preconditioning,IP)合并低温及晶体停搏液对幼兔的离体心脏是否具有心肌保护作用.采用Langendorff离体心脏灌注模型,灌注液为Krebs-Henseleit液(K-H液).取3~4周龄幼兔心脏,在第一部分实验中分为Con、IP1、IP2、IP3组(n=6),分别给予0、1、2、3次IP,其后各组心脏均在20℃低温下停灌2 h,37℃常温下再灌注30 min.在第二部分实验中分为SConI、SCon2、SCon3、SIPl、SIP2、SIP3组(n=8),其中SIPl、SIP2、SIP3组给予2次IP后灌注St.Tho-mas Ⅱ晶体停搏液(CCS)使心脏停搏,然后分别使心脏在32℃、25℃、20℃下停灌30、90和120min,其后各组均在37℃再灌注30 min.SConl,SCon2,SCon3三组则不给予IP,继续灌注20min后灌注CCS使心脏停搏,然后分别在32℃、25℃、20℃下停灌30、90和120 min,其后各组均在37℃再灌注30 min.以Maclab/4 s生理实验系统记录平衡末、缺血前、再灌注后1、3、5、10、20、30 min时心率(HR)、左心室发展压(LVDP)以及左心室内压上升及下降最大速率(±dp/dtmax),测定再灌注末心肌组织中ATP和丙二醛(MDA)的含量,以及超氧化物歧化酶(SOD)的活性.在20℃低温停灌且停灌期间不给予CCS时,再灌注末IP2组LVDP×HR、+dp/dtmax和-dp/dtmax的恢复率分别为96%±21%、101%±19%和84%±15%,显著高于Con组和IP3组(P<0.01,P<0.05);心肌组织的ATP含量亦高于Con组(P<0.01).在不同低温停灌且停灌期间给予CCS时,再灌注末SIP1、SIP2组的LVDP×HR、+dp/dtmax分别恢复到87%±14%、99%±26%(P<0.05,vs SConl group)和87%±16%、102%±20%(P<0.05,vs SCon2 group);心肌组织的ATP含量均分别显著高于SCon1组和SCon2组(P<0.05),心肌组织MDA含量亦分别低于SCon1组和SCon2组(P<0.05).上述结果提示,IP对在20℃低温停灌的兔未成熟心脏具有一定的心肌保护作用,2次IP的保护效应优于1次或3次IP.在停灌期间应用CCS,IP的心肌保护作用随停灌期间低温程度的升高而减弱.  相似文献

The effects on fertility of the immunoglobulins, IgG (similar to serum IgG) and secretoy IgA (the predominant IgA in uterine fluid, different from serum IgA) and the fluids from the uteri of semen-immunized rabbits were studied. Female rabbits were immunized either by transvaginal (TV) injections (every 4 or 5 days for 3 weeks) with adjusted semen added to buffered saline containing .5 mg each polyadenylic acid )Poly-A) and polyuridylic acid or a combination of transvaginal injections and systemic injections with rabbit semen mixed with adjuvant (each once weekly for 3 weeks). 2 weeks following the last injection of semen each rabbit received a TV injection followed in 1 week by an instillation of semen and Poly A and U into uterine horns ligated at both ends. 2 weeks following intrauterine (IU) instillation uterine contents were withdrawn. Uteri were reinstilled. Passive hemagglutinating (PHA) and sperm-immobilizing antibody titers were determined on both serum and adjusted uterine fluid (UF) samples. Sperm immobilizin tests and PHA were positive in all serum samples from the contained immunization group, and PHA were positive in 4 out of 7 UF samples of this group. 2 out of 10 and 3 out of 10 samples from the TV group were positive for SI and PHA, respectively, and 4 out of 10 UF samples were PHA positive. Treatment of rabbit semen with UF samples prior to insemination of estrous rabbits resulted in an implantation rate of 2.8% for the combined immunization group and 24.2% for the TV group compared to 79.2% for controls. Some IUF samples without detectable antibodies appeared to inhibit fertility. The implantation rate of sperm treated with immune UF from the TV group absorbed with goat normal serum, anti-rabbit gamma-globulin serum, anti-rabbit-secretory IgA serum, anti-rabbit IgG serum, or sperm, were 3.4%, 88.9%, 42.9%, 64%, and 73. 3% respectively. SIgA and IgG antibodies were seperated by gel filtration. 2 of 4 pooled IgA samples and 3 of 4 pooled IgG samples depressed fertility, with 24.3% and 22.6% implantation and 25.8% 5, and 0% implantation, respectively. To assure that the antifertility effects observed were due to sperm antibodies of each immunoglobin class, SIgA fractions of 2 IUF pools were subjected to anion-exchange chromatography, and both SIgA and IgG fraction were absorbed with goat NS, anti-SIgA, and anti-IgG. IgG fractions absorbed with anti-SIgA or anti-IgG resulted in implantation rates of 0% and 56.8%, respectively, compared to a control of 89.5%. SIgA fractions absorbed with anti-SIgA or anti-IgG resulted in implantation rates of 81.8% and 19.4%. Thus IgG and secretory IgA of the uterine fluids are active in inhibition of fertility.  相似文献

Progesterone induced effect on rabbit uterus follows a log dosis endometrial glandular changes linear relationship and log dosis-organ weight one. It appears a strong correlation between endometrial changes and organ weight. Thus organ weight may be looked like a pseudogestative activity test, histological investigation remaining the actual proof of specific activity. Experiments on rabbits carried out by the food-procuring conditioned reflex method have shown that conditioned reflex systems to "pure" time can be elaborated, provided that conditioned stimulus is presented at regular, though different time intervals, following one another in a fixed sequence. The complexity of elaborating such systems to time is determined by the number of signal time intervals in the system, their different length, and their sequence. Our laboratory has previously demonstrated that maximal bronchoconstriction produces a greater degree of airway narrowing in immature than in mature rabbit lungs (33). To determine whether these maturational differences could be related to airway structure, we compared the fraction of the airway wall occupied by airway smooth muscle (ASM) and cartilage, the proportion of wall area internal to ASM, and the number of alveolar attachments to the airways, from mature and immature (6-mo- and 4-wk-old, respectively) rabbit lungs that were formalin fixed at total lung capacity. The results demonstrate that the airway walls of immature rabbits had a greater percentage of smooth muscle, a lower percentage of cartilage, and fewer alveolar attachments compared with mature rabbit airways; however, we did not find maturational differences in the airway wall thickness relative to airway size. We conclude that structural differences in the airway wall may contribute to the greater airway narrowing observed in immature rabbits during bronchoconstriction.  相似文献

The synergistic effect of plant growth retardants, such as daminozide, paclobutrazol and triadimefon, and of indole-3-butyric acid (IBA) on the formation of adventitious roots in hypocotyl cuttings of mung bean was studied. The three retardants and IBA all stimulated adventitious root growth, but IBA was the most effective. However, mixtures of the retardants with IBA have proven generally more effective than IBA alone in promoting adventitious root formation. When IBA was applied to the hypocotyls one day after cutting preparation followed by the growth retardant on the second day, there were even more adventitious roots produced than if applied in the reverse order. OBJECTIVE: The purpose of this study was to investigate the effects of progesterone and the most commonly prescribed synthetic progestogen, norethisterone, on regional immune-like reactivity of neuropeptide Y (NPY), substance P (SP), neurokinin A (NKA) and neurotensin (NT) in brains of female ovariectomized estradiol-substituted rats. Results: Norethisterone+estradiol-treated rats had 44% lower SP levels compared with estradiol-only-treated in frontal cortex and 20% lower NKA levels in comparison with progesterone+estradiol-treated in frontal cortex. Progesterone+estradiol-treated rats had 66% lower SP levels in striatum in comparison with both estradiol-only-treated and norethisterone+estradiol-treated. No significant results were found for NPY and NT. CONCLUSION: Progesterone and the synthetic progestogen, norethisterone, have different effects on SP- and NKA-like immunoreactivity in rat cortex and striatum.The effects of NET on SP- and NKA-like immunoreactivity in frontal cortex may contribute to the mood effects ascribed to this progestogen in clinical usage.  相似文献

Increased capillary permeability at implantation sites was demonstrated in rabbits by extravasation of intravascular blue dye on day 7 of pregnancy. Subcutaneous administration of indomethacin (Id, 8 mg/kg twice daily) on days 4–6 of pregnancy inhibited this uterine blueing response and appeared to reduce the size of implantation swellings. To test the latter observation blastocyst diameter and development of the embryonic disk were assessed at 144 hr . In females receiving indomethacin at the dose level which inhibited uterine blueing, blastocysts were significantly smaller than those from control females. Developmental staging of embryonic disks revealed only slight differences between the smaller (Id-treated) blastocysts and control blastocysts. No effect of Id was seen on ovarian function as judged by luteal weights and plasma progesterone and estradiol levels. Since the major biological effects of indomethacin are due to its inhibition of prostaglandin synthesis, it appears that prostaglandins may play a role in the uterine response to blastocyst stimulation and in the expansion of blastocysts in the rabbit.  相似文献

To investigate the biological effects of exposure to feeble high frequency electromagnetism, skin surface temperature, blood vessel (arterioles and venules) diameter were examined, using infrared thermography, a laser doppler flowmeter, and a video microscope, respectively, in the ear of rabbits. After exposing the ear of rabbits to high frequency electromagnetism value of 9 MHz for 15 minutes, continued rising of local temperature was demonstrated. Though dilatation of arterioles was not seen. In addition, venules tended to dilate and blood flow also to increase, and microcirculation was accelerated at the site where electromagnetism was exposed. Hazardous effects of long term exposures of high frequency electromagnetism (9 MHz for 30 days, 8 hours/day) on guinea pigs were not observed in their behavior, food consumption, body and organ weights, hematological and biochemical values, macroscopic and microscopic findings on autopsy.  相似文献

An immunological demonstration of alpha-fetoprotein (AFP) in uterine cytosol from immature rats is reported. The identity between serum AFP and the estrogen binding 4.5 S macromolecular complex of uterine cytosol was demonstrated by the use of a specific immunoadsorbent to AFP. Analysis of the sedimentation profile in glycerol gradients of uterine cytosol incubated with tritiated estrone or estradiol suggests that the total estrogen binding capacity of the 4.5 S complex is provided by AFP. Changes of AFP content in rat uterus with the age of the animals suggest that this protein is probably present in the cytosol as a serum contaminant.  相似文献
