A vibrating probe was used to measure extracellular electrical currents around developing somatic embryos in two lines (RCC27, RCC48) of cultured cells of Daucus carota L. at the heart and torpedo stages. At pH 5.5, an inward current of 1.2±0.1 A·cm-2 (n=23) was detected at the cotyledon, and an outward current of 1.0±0.1 A·cm-2 (n=22) was found at the radicle in torpedostage embryos from the RCC27 line. At a pH of 5.75 the currents increased by 0.2–0.3 A·cm-2 (n=60–62). In a few cases an additional small inward current was detected at the tip of the radicle in toepedo-stage embryos from RCC27 line. Such an inward current at the radicle seemed to appear earlier, some time after the heart stage, in embryos from the RCC48 line.Both extracellular pH measurements (using microelectrodes filled with ion-sensitive resin) and ion-substitution studies were carried out in order to ascertain the ionic composition of the currents in torpedo-stage embryos from the RCC27 line. Regions adjacent to the cotyledon and radicle, at the points of current entry and exit, were found to be more acidic by 0.02±0.01 (n=14) and 0.07±0.01 (n=12) pH units, respectively, than the bulk medium. Removal of K+ from the medium reversibly reduced the currents to about 25% of their original value at both cotyledon and radicle. Deletion of Cl- decreased the currents slightly. Removal of Ca2+ resulted in a rapid doubling of currents. Addition of either N,N-dicyclohexylcarbodiimide or tetraethyl ammonium chloride substantially reduced overall currents, and their removal resulted in partial recovery of the currents. It is suggested that the inward current at the cotyledon is comprised largely of K+ influx and the outward current at the radicle is mainly the result of active H+ efflux.Abbreviations 2,4-D 2,4-dichlorophenoxyacetic acid - MS Murashige and Skoog  相似文献

Summary When Ca2+, K+ or Cl was injected iontophoretically into the cytoplasm of intactNitella cell, only Ca2+ reversibly inhibited the cytoplasmic streaming. However, when an extremely large current was used, the cytoplasmic streaming was reversibly inhibited irrespective of the ion species. A striped-leaf mutant has been identified in a Taiwanese maize line, Tainan White. The striped area is pale green at the seedling stage, and turns yellow and white at maturity, extending from the leaf blade to the sheath and the internode below the sheath. The mutant does not breed true and following self-pollination, produces three different types of progeny: green, striped and completely affected. The green type permanently loses the mutant character, while the completely affected type is lethal, without any green tissue. Only the striped type transmits the mutant phenotype to progeny, and it has a clonal distribution on the ear. Its phenotype is inherited maternally and its expression is unaffected by the paternal genotype. It cannot be inherited through the paternal parent. This pattern of inheritance, which persists after three cycles of backcrossing with the affected types as the recurrent pistillate parent, suggests that the responsible gene is located in the cytoplamic genome.  相似文献

Summary Variation in mitochondrial protein synthesis and genome organization was investigated. Three different alloplasmic cytoplasmic male-sterile Nicotiana tabacum cultivars, carrying N. repanda, N. suaveolens or N. debneyi cytoplasm, were analysed together with corresponding male-fertile parental and restored material. Although several differences were detected in the proteins synthesized by isolated mitochondria from the male-sterile and male-fertile plants, most of these were related to the origin of the mitochondria. However, a 23 kD protein was synthesized in the male-sterile cultivar carrying N. debneyi mitochondria, but not in other lines containing this cytoplasm. This protein was also present in the male-fertile parent containing N. tabacum mitochondria. Only the enhanced production of a 30 kD protein in the lines carrying mitochondria from N. repanda or N. debneyi was exclusively correlated with CMS. This protein was not present in any of the corresponding male-fertile parental and restored lines. Restriction enzyme analysis of mitochondrial DNA revealed a difference in abundance of a 5.6 kb XhoI fragment between lines containing N. debneyi mitochondria. No rearrangements of mitochondrial DNA was found between male-fertile and male-sterile lines carrying N. repanda or N. suaveolens cytoplasm. These results might indicate that CMS in alloplasmic Nicotiana cultivars is caused by alterations in the expression of mitochondrial genes, rather than by induced changes in the genome.  相似文献

A soluble protein was isolated from Mougeotia by chloropromazine-sepharose 4 B affinity chromatography. The protein matches the properties of calmodulin in terms of heat stability, Ca2+-dependent electrophoretic mobility in sodium-dodecyl-sulfate polyacrylamide gels, and its ability to activate cyclic nucleotide phosphodiesterase in a Ca2+-dependent manner. Phytochrome-mediated chloroplast reorientational movement in Mougeotia was inhibited by the calmodulin antagonist trifluoperazine, a hydrophobic compound, or N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7), a hydrophilic compound; 50% inhibition (IC50) of chloroplast movement is caused by 20–50 mol l-1 trifluoperazine or 100 mol l-1 W-7. Summary Hyphae ofNeurospora crassa, like many other tipgrowing organisms, drive endogenous electric currents through themselves such that positive charges flow into the apical region and exit from the trunk. In order to identify the ions that carry the current, the complete growth medium was replaced by media lacking various constituents. Omission of K+ or of phosphate diminished the zone of inward current, effectively shifting the current pattern towards the apex. Omission of glucose markedly reduced both inward and outward currents; addition of sodium azide virtually abolished the flow of electric current. Growing hyphae also generate a longitudinal pH gradient: the medium surrounding the apex is slightly more alkaline than the bulk phase, while medium adjacent to the trunk turns acid. The results suggest thatNeurospora hyphae generate a proton current; protons are expelled distally by the H+-ATPase and return into the apical region by a number of pathways, including the symport of protons with phosphate and potassium ions. Calcium influx may also contribute to the electric current that enters the apical region. There seems to be no simple obligatory linkage between the intensity of the transcellular electric current and the rate of hyphal extension. Calcium ions, however, are required in micromolar concentrations for extensions and morphogenesis of hyphal tips.  相似文献

Summary The effect of the intracellular concentration of Ca2+ on the cytoplasmic streaming of tonoplast-free cells ofChara australis was studied by intracellular perfusion. The perfusion media contained 1 mM Mg · ATP. Both cell ends were cut and left open. Media of different Ca2+ concentrations were perfused through the cell and the rate of the cytoplasmic streaming just after perfusion was measured. The critical concentration of Ca2+ for inhibiting the streaming was 5 × 10–4M, which was substantially higher than that found earlier byWilliamson (1975) andHayama et al. (1979). Recovery from the inhibition occurred, though not completely, by removing Ca2+.In tonoplast-free cells the Ca2+ sensitivity differed according to the culture conditions. Cells cultured indoors exhibited a higher sensitivity than those cultured outdoors. Theformer cells contained granule-rich endoplasm aggregates after loss of the tonoplast, while the latter cells did no such aggregates. The aggregates were fixed to the cortical gel with a high dosage of Ca2+ and freed by removing it.  相似文献

The pharynx of Caenorhabditis elegans consists of a syncytium of radially orientated muscle cells that contract synchronously and rhythmically to ingest and crush bacteria and pump them into the intestine of the animal. The action potentials that support this activity are superficially similar to vertebrate cardiac action potentials in appearance with a long, calcium-dependent plateau phase. Although the pharyngeal muscle can generate action potentials in the absence of external calcium ions, action potentials are absent when sodium is removed from the extracellullar solution (Franks et al. 2002). Here we have used whole cell patch clamp recordings from the pharynx and show low voltage-activated inward currents that are present in zero external calcium and reduced in zero external sodium ions. Whilst the lack of effect of zero calcium when sodium ions are present is not surprising in view of the known permeability of voltage-gated calcium channels to sodium ions, the reduction in current in zero sodium when calcium ions are present is harder to explain in terms of a conventional voltage-gated calcium channel. Inward currents were also recorded from egl-19 (n582) which has a loss of function mutation in the pharyngeal L-type calcium channel and these were also markedly reduced in zero external sodium. Despite this apparent dependence on external sodium ions, the current was partially blocked by the divalent cations, cadmium, barium and nickel. Using single-channel recordings we identified a cation channel for which the open-time duration was increased by depolarisation. In inside-out patches, the single-channel conductance was highest in symmetrical sodium solution. Further studies are required to determine the contribution of these channels to the pharyngeal action potential.  相似文献

Summary Sounds produced during prefeeding, feeding and postfeeding behaviour of the tsetse, Glossina m. morsitans Westw. were investigated. Males and virgin very seldom sang before feeding but mated sang in more than 20% of the cases before taking a meal. Sound production during engorgement was negligible among all 3 groups. Postfeeding sounds were the most commonly produced, mated singing significantly more than virgin and both these groups more than . The mean duration of the postfeeding sounds did not differ between the sexes. Within each group, postfeeding sound production did not significantly change with age. There were, however, significant differences in the patterns of postfeeding sound production when were compared to over a period of 8 weeks.Oscillograms of these sounds indicated that the songs differed between the sexes and also varied from time to time. The sounds were composed of frequencies up to 50 kHz with the exception of the prefeeding sounds which did not contain any ultrasonic components. The most dominant frequency was centered between 1.5–2.5 kHz. Peaks were also observed between 0.5–0.8 kHz and around 5 kHz and 9 kHz. Prefeeding songs were of a weaker intensity than postfeeding songs.It is suggested that the sounds associated with feeding behaviour play a role in attracting hungry flies to suitable hosts and that, the low frequency components having higher intensities (30–40 dB) than the ultrasonic frequencies (10 dB) carry the most important part of the acoustic information.
Résumé L'étude a porté sur les sons produits pendant les comportements préalimentaire, alimentaire et postalimentaire de la mouche Tsétsé, Glossina morsitans morsitans.Les mâles et les femelles vierges chantent très rarement avant l'alimentation, mais les femelles fécondées chantent dans plus de 20% des cas avant de prendre un repas. La production de sons pendant la prise de nourriture était négligeable pour les trois catégories. Les sons postalimentaires étaient plus fréquents, les femelles fécondées chantant plus que les femelles vierges et l'ensemble des femelles plus que les mâles. La durée des sons postalimentaires ne différait pas suivant les sexes. Dans chaque catégorie, les sons postalimentaires ne changaient pas significativement avec l'âge. Il y a eu, cependant, des différences significatives dans les caractéristiques de la production de son postalimentaire lorsque les mâles étaient comparés aux femelles pendant une période de 8 semaines.Les oscillogrammes de ces sons différaient suivant le sexe et variaient de temps en temps. Les sons comportaient des fréquences jusqu' à 50 kHz, à l'exception des sons préalimentaires des mâles qui ne contenaient pas d'ultrasons. Les fréquences les plus dominantes étaient concentrées entre 1,5–2,5 kHz. Des pics étaient observés entre 0,5–0,8 kHz et autour de 5 kHz et 9 kHz. Les sons préalimentaires étaient plus faibles que les sons postalimentaires. On en suggère que les sons associés au comportement alimentaire jouent un rôle en attirant les mouches affamées vers un hôte convenable et que la plus grande partie de l'information acoustique est portée par les composantes à basse fréquence qui ont une intensité plus élevée (30–40 dB) que les fréquences ultrasoniques (10 dB).

The circadian movement of the lamina of primary leaves ofPhaseolus coccineus L. depends on circadian changes of the K+, Cl- and (depending on the Cl- availability) malate content in the swelling and shrinking motor cells of the laminar pulvinus. After sowing in soil, the laminar pulvinus develops within about 26 days. When the leaves emerge from the soil (about 6 days after sowing) and the pulvinus starts with the diurnal movement (about 9 days after sowing) the pulvinar dimensions are about half of those of the mature pulvinus. The anatomical structure, however, is basically the same as in the developed pulvinus. In soil-grown plants, the K+, Cl- and malate content as well as the period length of the circadian leaf movement rhythm change in the developing pulvinus. In the embryo of the dry seed, the Cl- content is low (0.03 mmol g-1 DW), the K+ content, however, 22-fold higher than the Cl- content. When the leaves emerge from the soil, the pulvinar K+ and Cl- content is the same as in the whole embryo of the dry seed. In the developing pulvinus the K+ content increases by a factor of 2 and the Cl- content by a factor of 41 in the mature pulvinus. The pulvinar malate content increases between the 6th and 10th day from about 40 to 180Μmol g-1 DW, then decreases until the 17th day and remains thereafter on a low level (around 80 Μmol g-1 DW). These results indicate that the Cl- availability increases in the developing pulvinus with age. It explains furthermore why in young leaves malate was found as counterion to K+ in the osmotic leaf movement motor, in older ones, however, Cl-. The circadian leaf movement starts 9 days after sowing. The period length decreases during the development of the pulvinus from 31.3 to 28.6 h in leaves of intact soil-grown plants. In leaves which were cut from the plants and immersed with their petioles in distilled water, the age dependent decrease of the period length is also found. However, the period lengths are shorter by more than 1 h than in the leaves of intact plants. The increasing Cl- availability in the developing pulvinus does not seem to be the cause for the age dependent shortening of the period length, because the period length in 22 days old Cl- deprived pulvini is the same as in 22 days old pulvini with a high Cl- content.  相似文献

Summary The temperature dependence of cytoplasmic streaming in intact and tonoplast-free cells ofNitellopsis obtusa was studied using a cryomicroscope. The streaming velocity decreases linearly with decrease in the temperature in well-buffered tonoplast-free cells but non-linearly in some intact cells. Summary Latex beads coated with rabbit skeletal muscle myosin were introduced by intracellular perfusion intoChara cells from which the tonoplasts had been removed. Mg · ATP dependent movement of the beads along files ofChara chloroplast layers was observed. The movement was in opposite directions on the two sides of the indifferent line, indicating that the movement was dependent on the polarity of the actin bundles. This suggests that the unknown factor responsible for generating the motive force for cytoplasmic streaming inChara endoplasm is myosin. When the individual Japanese macaques of the Koshima troop feed on natural food, they usually feed alone. In situations where animals usually feed without other animals, there is a possibility that subordinate animals may avoid feeding sites at which dominant animals are feeding. This paper examines whether social relationships such as kinship or dominance exert any influence on an animal's choice of feeding sites, by analyzing episodes in which an animal approached and climbed into a tree where other animals were. As a result, it was found that social relationships did not influence whether an animal climbed into a tree where other animals were feeding, and that no particular age-sex pair co-fed. Agonistic interactions frequently occurred when the inter-individual distance was less than 1 m. From these findings, the feeding sites were divided into two spaces: (1) a tolerance feeding space, and (2) an intolerance feeding space. It is presumed that animals can feed without entering others' intolerance feeding spaces when food is abundant, as it was in the present study period. Thus social relationships do not influence an animal's choice of feeding sites in such a situation.  相似文献

During the wet season, two sympatric species of primates,Alouatta palliata (mantled howlers) andCebus capucinus (white-faced capuchins), were assayed for feeding niche differences through behavioral and habitat use patterns at Refugio de Fauna Silvestre Curu in Costa Rica. Differences in the use of relative diameter and thickness of branches and five different modes of feeding were compared between the species. White-faced capuchins used more manipulative modes of obtaining food, a wider range of arboreal habitat, and had a more diverse diet than mantled howlers. Mantled howlers may be more restricted than white-faced capuchins in arboreal microhabitat use due to their possible need for large support branches during feeding bouts and resting periods. We report that differences in feeding behaviors, diet, and arboreal habitat use seem to play a large role in separating these species niches.  相似文献
