对我国泡桐属15个植物种类作了叶绿体DNA的RFLP分析,根据估算的相似系系,用平均链锁聚类方法构建权 图,结果可将研究材料分为南方泡桐组、毛泡桐组和白花泡桐组,种间相似系数多在0.70以上,说明各种类间亲缘关系较近,尤其是台湾泡桐和海岛泡桐,相似系数接近1.00,最后讲座了一些泡桐种类分类问题。  相似文献

陕棉抗病种质及其衍生品种的遗传多样性与群体结构研究   总被引:1,自引:0,他引:1  
通过72个分布于棉花全基因组的SSR标记,对54份陕棉抗病种质及其衍生品种的遗传多样性与群体结构进行了分析。结果表明:(1)54份陕棉种质间的遗传相似系数在0.733 3~0.987 2之间,其中材料间相似系数≤0.90的占11.1%,相似系数≥0.95的占55.6%,相似系数在0.90~0.95的占33.3%。(2)72个SSR标记等位基因变异的多态性信息含量(PIC值)在0.04~0.68,平均为0.33。(3)基于遗传距离的UPGMA聚类分析显示,在遗传相似系数为0.877时将54个品种分为5类,第Ⅰ类44个品种,第Ⅲ类7个品种,第Ⅱ、Ⅳ、Ⅴ类各1个品种。(4)基于数学模型的聚类和群体结构分析显示,54份种质归属于4个组群。研究认为,54份材料间遗传相似系数较大,遗传基础比较狭窄,多样性很低。  相似文献

为探讨COⅠ基因作为条形码在胡椒鲷亚科鱼类物种鉴定的可行性, 研究测定了胡椒鲷亚科8种鱼类51个个体线粒体COⅠ基因长度为651 bp的序列, 利用MEGA 5.0计算胡椒鲷亚科种内与种间的遗传距离, 基于最大似然法与贝叶斯法构建了系统进化树。结果显示: 胡椒鲷亚科鱼类种间平均遗传距离(0.142)显著大于种内平均遗传距离(0.003), 物种间遗传距离均大于Hebert推荐的物种鉴定最小种间遗传距离0.020(2%)。系统进化树上, 同一物种不同个体间均能形成独立的单系分支, 表明COⅠ基因可作为胡椒鲷亚科物种鉴定的有效条形码基因。研究同时揭示, 在形态分类上被认为是同种异名的两种胡椒鲷(条纹胡椒鲷与东方胡椒鲷)的COⅠ基因序列差异达到0.070, 有可能是两个独立的物种。此外, 在属级水平, 少棘胡椒鲷属与胡椒鲷属种类之间的平均遗传距离小于胡椒鲷属内部种间的遗传距离, 支持少棘胡椒鲷归属于胡椒鲷属的观点。    相似文献

The phylogeny of butterflies, Parnassius stubbendorfii and P. glacialis, collected at various localities in the Japan archipelago and the eastern part of the Asian continent was analyzed using mitochondrial DNA sequences coding for NADH dehydrogenase subunit 5 (805 bp). The molecular phylogenetic trees revealed that P. glacialis and P. stubbendorfii diverged from a common ancestor, and then the populations inhabiting the Japan archipelago and the Asian continent diverged in each species. The reliability of these divergences was supported by high bootstrap values. The divergences within the Japan archipelago and within the Asian continent in each species were unclear because of low bootstrap values. The genetic distance and a rough time-estimation in the UPGMA tree suggest that the both populations of P. glacialis and P. stubbendorfii may have been isolated in the Japan archipelago at the early time (about 1.7-2.0 Mya) of the glacial period in the Pleistocene. The genetic distance between the Japanese and the continental subspecies may be large enough that they can be classified as different species, in comparison with the genetic distances among some other parnassian species.  相似文献

云南松及其近缘种的遗传变异与亲缘关系   总被引:4,自引:0,他引:4  
The authors investigated the genetic diversity of 29 natural populations representing Pinus yunnanensis Franch. and its two close relatives, P. densata Mast. and P. kesiya Royle ex Gordn. var. langbianensis (A Chey.) Gaussen. Horizontal starch gel electrophoresis was performed for macrogametophytes collected from populations in Yunnan, Sichuan and Guangxi. Allozyme data for 33 loci of 14 enzymes demonstrated high levels of genetic variation at both population and species levels in comparison with other conifers, with the mean values for populations being P=0.694, A=2.0 and He=0.145 for P. yunnanensis; P=0.714, A=2.0 and He=0.174 for P. densata; and P=0.758, A=2.1 and He=0.184 for P. kesiya var. langbianensis. Based on Wright's F-statistics, the fixation index of P. yunnanensis, P. densata and P. kesiya var.langbianensis were 0.101, 0.054 and 0.143, respectively, indicating that the populations were largely under random mating. Based on Nei's genetic distance, the genetic differentiation was not obvious among the three species (i.e. the genetic distance was less than 0.075). Because of the wider distribution of P. yunnanensis with greater variety of habitats, it was shown that the genetic differentiation among the P. yunnanensis populations was larger than that of the populations of the other two species. According to morphological, geographic and allozymic evidences, the authors suggested that the three species be better treated as varieties under a single species. In addition, the extensive gene flow among the three pine species resulted in great genetic diversity and evolutionary potential. Also, high level of genetic variation of P. yunnanensis provides important basis for its genetic improvement and breeding in future.  相似文献

Twelve natural populations of four cedar pine species,Pinus sibirica, P. cembra, P. pumila, andP. koraiensis, occurring in the Soviet Union were investigated by starch-gel electrophoresis. Frequencies of 55 alleles at 19 loci were determined. Interpopulation genetic diversity inP. sibirica andP. pumila was only 2–4 per cent of the total genetic diversity. Nei's distance coefficient (Dn) was used to estimate the level of genetic differentiation among conspecific populations and among species. Dn values among populations ranged from 0.006 to 0.038. A dendrogram constructed using Dn values divided cedar pines species into 2 clusters:sibirica-cembra (Dn = 0.030) andpumila-koraiensis (Dn = 0.143). Nei's distance between these clusters was 0.232. On the basis of the data obtained it was possible to draw the following conclusion:P. sibirica, P. pumila, andP. koraiensis are distinct species, whileP. cembra should apparently be regarded as geographicalP. sibirica race.  相似文献

Starch gel electrophoresis was employed to survey the allozyme polymorphism and phylogenetic relationships among 35 populations covering six closely related Western Australian endemic Pterostylis species (series Grandiflorae); viz P. rogersii, P. aspera, P. angusta, P. hamiltonii, P. scabra and P. aff. alata. The aim of this study was to determine intraspecific and interspecific genetic diversity and species relationships based on allozyme analysis. The frequencies of 56 alleles at 12 enzyme systems coded by 15 loci were determined along with a mean intraspecific genetic identity value. Allozyme markers clearly discriminated populations belonging to different species. Nei's genetic distance/identity co-efficient was used to measure the level of genetic differentiation among populations and species. Based on these values, a dendrogram was constructed which revealed that all the populations clustered into groups corresponding to the respective species. Gene diversity analysis among all the species revealed total genetic diversity H(t) of 0.23 with co-efficient of gene differentiation 10% (G(st)=0.10). Mean genetic variability (H(e)=0.136, P=40%) was also higher than for other outbreeding plant species. Mean genetic identity coefficient of populations of all species was 0.859 which increased to 0.877 upon exclusion of P. aff. alata, indicating a high degree of similarity among all species except P. aff. alata which segregated distinctively from the rest. Overall, the investigation provided independent support for the morphological segregation of these taxa.  相似文献

The mitochondrial DNA D-Loop region was sequenced, analyzed and used as a molecular marker for populations of chimpanzee (Pan troglodytes), bonobo (P. paniscus) and gorilla (Gorilla gorilla and G. beringei), and also compared with data previously reported for these taxa. In Gorilla, a main disjunction between western (G. gorilla) and eastern (G. beringei, including G. b. graueri) species was observed, as well as high mitochondrial diversity within the western species. The genetic distance values within G. gorilla (0.14) were higher than those between subspecies (eastern lowland and mountain 0.12). Likewise, values of genetic diversity within this species (0.05) were higher than those between species (western and eastern lowland gorilla 0.04). Similarly, in genus Pan a main differentiation between western (P. t. verus) and central forms (P. t. troglodytes and P. t. schweinfurthii) was observed. The obtained values of genetic distance and genetic diversity revealed that the central subspecies are closer to each other than either of them is to the western one, while bonobos composed a distinct clade that expresses a well-defined specific identity. The current distribution, phylogeny and levels of genetic diversity in African great ape populations are consistent with the hypothesis that Pleistocene climatic events led to cyclical periods of isolation in forest refugia followed by expansion and dispersal. The implications of this high level of genetic diversity for taxonomic classification, wildlife management and conservation are discussed.  相似文献

Allozymic variation at 21-23 loci was studied in 28 populations of Talitrus saltator, 23 populations of Orchestia montagui, 13 populations of O. stephenseni, and five populations of Platorchestia platensis from the Mediterranean Basin. Different levels of gene flow (Nmtheta) were detected within each species at the scale of the whole Mediterranean: O. montagui and P. platensis had low population structure, with levels of Nmtheta > or = 1, whereas the T. saltator and 0. stephenseni populations have values of Nmtheta < 1. The relationship between Nmtheta and geographic distance was analyzed to test for the presence of an isolation by distance pattern in the spatial genetic variation within each species. A model of isolation by distance is useful to describe the pattern of genetic structuring of study species at the scale of the whole Mediterranean: geographic distance explained from 28% to 70% of the variation in gene flow. In the Aegean area all species showed an island model of genetic structuring regardless of the levels of gene flow.  相似文献

甘肃省鱼类资源现状及DNA条形码在鱼类物种鉴定中的应用   总被引:2,自引:0,他引:2  
为了摸清甘肃省土著鱼类资源与分布现状, 探索DNA条形码在鱼类辅助物种鉴定中的适用性, 2012年6-9月对甘肃境内黄河水系、嘉陵江水系和河西内陆河水系进行了较全面的鱼类调查。共采集鱼类标本3,087尾, 隶属于5目10科38属64种, 以鲤科种类最多, 为30种, 占总种数的46.88%。物种多样性分析表明, 在黄河水系的夏河和庄浪河多样性指数是所有调查点中最低的, 分别为1.38和1.09。嘉陵江水系各河段的多样性指数较高(H = 2.15-3.27), 其次为河西内陆河水系(H = 2.01-2.83)。在河西内陆河水系中, 疏勒河的均匀度指数最高, 为1.10, 黑河最低(0.68)。庄浪河的优势度指数最高, 为0.34, 而嘉陵江干流两当段的优势度指数在所有调查点中最低, 为0.04。利用DNA条形码分析了49种662尾标本的COI基因部分序列, 大部分种类在neighbor-joining系统树中形成各自的单系, 种内平均遗传距离0.88%, 种间平均遗传距离为9.99%, 在种内和种间COI序列遗传距离之间形成明显的条形码间隙, 斯氏高原鳅(Triplophysa stoliczkae)与达里湖高原鳅(T. dalaica), 甘肃高原鳅(T. robusta)与似鲇高原鳅(T. siluroides), 嘉陵裸裂尻鱼(Schizopygopsis kialingensis)与黄河裸裂尻鱼(S. pylzovi)之间的遗传距离低于2%, 甘肃高原鳅与似鲇高原鳅不能通过COI基因片段区分开, 其他两对物种可以采用核苷酸诊断法来进一步区分。斯氏高原鳅和拉氏鱼岁(Phoxinus lagowskii)种内遗传分歧较大, 揭示种内可能存在隐存种。结果表明, 对某些近缘种和不同地理种群差异较大的物种, 要将分子、形态和地理分布特点结合起来才能准确鉴定。  相似文献

使用20个随机引物研究了4种黄颡鱼的RAPD图谱,其中6个可以用来准确鉴别4个种,分别是S1、S2、S5、S7、S8、S17。同时也分析了4种黄颡鱼之间的亲缘关系,发现种间遗传距离D值在0.5000左右波动,黄颡鱼-叉尾黄颡鱼、瓦氏黄颡鱼-光泽黄颡鱼之间的D值最小,分别是0.4816和0.4017。聚类图显示,黄颡鱼和叉尾黄颡鱼属同一分支;瓦氏黄颡鱼和光泽黄颡鱼属另一分支,说明它们之间分别有更近的亲缘关系。另外,采用黄颡鱼作母本,瓦氏黄颡鱼作父本,获得了杂交F1代,对F1代进行RAPD分析,发现了杂种遗传图谱的三种变化情况:叠加、叠加-变异、叠加-弱化,表明F1代DNA多态性增强,杂合性提高,预示着杂种优势的可能性。    相似文献

The Agave deserti complex, comprising A. deserti, A. cerulata and A. subsimplex, represents a group of species and subspecies with a near allopatric distribution and clear differences in morphology. Genetic differentiation and taxonomic status with respect to spatial distribution of 14 populations of the complex were analyzed in an effort to understand the evolution and speciation process within the genus. Allelic frequencies, levels of genetic variation, expected heterozygosity (H(S)), proportion of polymorphic loci (P), and genetic differentiation (theta and Nei's genetic distance) were estimated using 41 putative RAPD loci. All three species show high levels of genetic variation (H(S)=0.12-0.29, P=63.4-95.1), and low genetic differentiation between populations and species (theta populations=0.14+/-0.02 (SE); G(st)=0.11+/-0.02). Accordingly, gene flow among populations was estimated as high by three different methods (N(m)=2.91-6.14). Nei's genetic distances between the three species were low compared to the values obtained from other Agavaceae, and there was no clear correlation with taxonomic divisions. In a UPGMA analysis, A. subsimplex and A. cerulata formed exclusive monospecific clusters, whereas the A. deserti populations appear in more than one cluster together with other species. The results were consistent with a pattern of genetic isolation by distance.  相似文献
