Summary Planarians (Dugesia tigrina) were kept for 1, 2 and 3 weeks in total darkness and the fine structure of the eyes was investigated. After fixation in osmium, the photoreceptors (retinal clubs) showed a progressive disintegration of the microvilli membranes into vesicles and tubules. Although the vesicular transformation of the membranes was not found in glutaraldehyde-prefixed experimental animals, it could not be detected in the osmium-fixed controls either. Consequently the vesicular transformation must be regarded as a meaningful artefact due to membrane instability induced by prolonged deprivation of light.After 3 weeks in the darkness strong atrophy and degeneration of the retinal clubs was observed both in osmium and glutaraldehyde-fixed specimens resulting in a decrease of the club to about one fifth of its original size. As a result large empty spaces appeared between rudimentary retinal clubs and the whole eye became shrunken. When animals kept for 4 weeks in the darkness were exposed to normal light conditions, a rapid regeneration of the microvilli was observed together with an increasing stability of the membranes.  相似文献

Human blood group A antigenicity of glycoproteins is retained on epon-embedded jejunum sections after glutaraldehyde fixation and osmium treatment. The intracellular location of molecules bearing these determinants was visualized in the four types of epithelial cells of A+ rabbit jejunum sections with immuno-colloidal gold labeling. The brush border membrane and in particular the glycocalyx of absorbing cells as well as the secretory granules of goblet and Paneth cells were heavily labeled. In enteroendocrine cells, the membrane of secretory granules and not their content was lightly labeled. The differential labeling of secretory or membrane bound glycoproteins is accompanied by different labels of the Golgi complex as expected if labeling of the Golgi saccules was due to the presence of glycoproteins in transit. In all cases the label is primarily concentrated in only half the cisternae on the trans side of the Golgi stacks. In absorbing cells, structures have been revealed in the terminal web that could be related to the brush border membrane and consequently implicated in its biogenesis. The fibrillar material of the glycocalyx appears as highly labeled tangled structures which apparently proceed from densely stained "carrier" vesicles arising from the Golgi apparatus. Vesicles fusing at the lower part of microvilli could result of integration of this material into the lightly labeled vesicles strictly found in the terminal web. These last vesicles could also contain newly synthesized brush border hydrolases.  相似文献

Endogenous peroxidase activity in rat thyroid follicular cells is demonstrated cytochemically. Following perfusion fixation of the thyroid gland, small blocks of tissue are incubated in a medium containing substrate for peroxidase, before being postfixed in osmium tetroxide, and processed for electron microscopy. Peroxidase activity is found in thyroid follicular cells in the following sites: (a) the perinuclear cisternae, (b) the cisternae of the endoplasmic reticulum, (c) the inner few lamellae of the Golgi complex, (d) within vesicles, particularly those found apically, and (e) associated with the external surfaces of the microvilli that project apically from the cell into the colloid. In keeping with the radioautographic evidence of others and the postulated role of thyroid peroxidase in iodination, it is suggested that the microvillous apical cell border is the major site where iodination occurs. However, that apical vesicles also play a role in iodination cannot be excluded. The in vitro effect of cyanide, aminotriazole, and thiourea is also discussed.  相似文献

The glomus cells of the rat carotid body reveal an intense fluorescence after exposure to paraformaldehyde vapor and contain catecholamines. After initial fixation in glutaraldehyde, many granulated vesicles are seen in the glomus cells. After initial fixation in osmium tetroxide, most of the vesicles are depleted of their dense interiors and granulated vesicles occur infrequently. Administration of 6-hydroxydopamine followed by initial fixation in osmium tetroxide leads to the reappearance of dense interiors in virtually all vesicles. 6-Hydroxydopamine apparently is taken up by the membrane pump of the glomus cell and is incorporated into the amine storage granules, thereby displacing the endogenous monoamines. Osmium tetroxide does not dissolve the 6-hydroxydopamine from the vesicles, as it apparently does for the normal vesicular contents. The 6-hydroxydopamine does not fluoresce, hence 6-hydroxydopamine administration results in a decreased intensity of formaldehyde induced fluorescence in the glomus cells. Administration of reserpine after 6-hydroxydopamine treatment (and subsequent initial fixation in osmium tetroxide) depletes the previously restored dense material from the vesicles of the glomus cells. 6-Hydroxydopamine acts like a monoamine in that it is taken up by the glomus cell, incorporated into the vesicles, and can be depleted from the vesicles by reserpine.  相似文献

Two permanganate fixation methods (using Krebs-Ringer-glucose, pH 7.0, or acetate buffer, pH 5.0) were compared. Numerous dense-cored vesicles were observed only in about one tenth of the glomus cells when neutral permanganate was used for fixation, although all glomus cells showed such vesicles after fixation with glutaraldehyde and osmium tetroxide. Numerous vesicles with a dense core were observed in about one third of the cells after fixation with acid potassium permanganate. With this fixation, small dense-cored vesicles similar to those in adrenergic nerve terminals were occasionally seen in the cytoplasm of glomus cells. It is tentatively concluded that the amine-storing vesicles of the carotid body are different from those in the small intensely fluorescent (SIF) cells and those in adrenergic nerve terminals.  相似文献

The surface of the syncytial trophoblast of the human placenta is covered by a microvillous (brush) border that is in direct contact with maternal blood. Because of this location, it is the site of a variety of transport, enzymatic and receptor activities vital to many placental functions. The organization of the brush border as well as other features of placental villus organization may well be influenced by the distribution of cytoplasmic actin filaments. In order to determine the distribution of actin filaments in human placenta, small pieces of villi were briefly fixed in glutaraldehyde, permeabilized with saponin, and incubated in solutions containing subfragment 1 of myosin (S1). After S1 decoration of actin filaments, tissue was fixed in glutaraldehyde containing tannic acid in order to better visualize the polarity of the filaments, and prepared for electron microscopic examination. The microvilli each contained a core of actin filaments running from the tip of the microvillus to the apical cytoplasm. Most of the actin filaments displayed a distinct polarity, with the S1 arrowheads pointing away from the microvillar tips. These filaments extended only a short distance into the apical cytoplasm. There appeared to be another group of actin filaments in a matlike arrangement in the apical cytoplasm. Coated pits and vesicles were often observed between the microvilli. There appeared to be no clear association between the coated pits and decorated actin filaments, but this was difficult to establish with certainty because of the close proximity of the microvilli. Bundles of actin filaments were sometimes observed near the basal cell surface of the syncytial trophoblast, and in pericytes and capillary endothelial cells in the cores of the villi.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Summary The cytoplasm of the growth zone at the tip of theLilium pollen tube contains numerous membrane-bound vesicles and an abundance of smooth endoplasmic reticulum. Covering the tip of the tube is a cap of compartmentalized wall material which arises from coalesence of vesicles with each other and with the existing cap.This organization, seen following glutaraldehyde fixation, is altered to varying degrees by different fixation procedures. Neither the cap nor the e. r. are preserved by osmium fixation. Formalin or permanganate preserve the cap but not its compartments. Preservation of the cap by permanganate is concentration-dependent but poor at best. Combined fixation by acrolein and glutaraldehyde results in better preservation of membranous elements than glutaraldehyde alone.Supported by grant RG 8827 from the National Institutes of Health, U. Summary Aberrations of photoreceptor ultrastructure resulting from carotenoid/retinoid (vitamin A) deprivation were studied in the retina of Manduca sexta. The syndrome of chromophore deficiency included hypertrophy of smooth endoplasmic reticulum, variable dilation of rhabdomeric microvilli, the insertion of endomembrane fingers into such enlarged microvilli, and the formation of rhabdomeric vacuoles, intracellular compartments containing microvilli similar to those of the rhabdomere. Retinas were processed either with conventional procedures employing preliminary aldehyde fixation followed by heavy metal postfixation, or by fixation and incubation in unbuffered OsO4. The latter method deposits osmium throughout the endomembrane system, within the rhabdomeric vacuoles, and in the extracellular space of the rhabdom. However, the intravillous fingers were rarely impregnated with osmium, despite their continuity with densely stained cisternae of the smooth endoplasmic reticulum. We suggest that the insertion of endomembrane fingers into dilated microvilli results from a cytoskeleton-mediated link between cisternae of the smooth endoplasmic reticulum and the rhabdomeric membrane, an association that may be important in the turnover of photoreceptor membrane. We interpret endomembrane hypertrophy and development of rhabdomeric vacuoles as symptoms of disturbance in the pathway leading to the assembly of the rhabdomere resulting from reduced synthesis of visual pigment.  相似文献

Summary Primary fixation of the frog oocyte with glutaraldehyde, compared to osmium tetroxide, alters the appearance of components involved in the morphogenesis of annulate lamellae. With glutaraldehyde, the outer layer of the nuclear envelope is connected with membranous laminae of variable length. Rounded blebs of the outer layer of the nuclear envelope are infrequently observed. Further, rather than clusters or rows of cytoplasmic vesicles as are observed in osmium tetroxide-fixed cells; numerous and long, smooth-surfaced lamellae are present in the ooplasm after glutaraldehyde fixation. The long membranous laminae then become concentrated in several ooplasmic packets. This is followed by the progressive alignment or orientation of the laminae within the packet. Summary Rat pineal nerve endings contain a population of small and of large synaptic vesicles that are either electron lucent or have electron-dense cores. It has been reported that their osmiophilia is elminated when collidine buffer is used in the fixation procedure. We investigated this effect and found that osmium tetroxide and potassium dichromate reactivity were abolished when excised pineal glands were briefly incubated with collidine buffer before glutaraldehyde-cacodylate fixation. Such an effect was not observed when collidine was applied after fixation. Glands that had been fixed in glutaraldehyde or osmium tetroxide buffered with collidine exhibited a peripheral zone containing reactive synaptic vesicles and a deeper, central zone where such reactivity was absent. These results indicate that the effect of collidine is due to depletion of monoamines rather than to chemical blockage of their reactivity, and further suggest that collidine has a higher rate of penetration into tissues than the tested fixatives.  相似文献

The isolated rabbit retina was studied electron microscopically after fixation with a 3% solution of glutaraldehyde in a 0.05 M S?rensen's phosphate buffer. In radial sections, the inner segments, nuclei, and synapses of the photoreceptor cells seemed similar in size to those from retinas that had been fixed in an isotonic solution containing 1 % crystalline osmium tetroxide in the incubating medium used for the isolation procedure. However, when the number of comparable structures was greatly increased by viewing them in tangential sections, the cellular shrinkage and mitochondrial swelling produced by this widely used, hypertonic, glutaraldehyde fixative were obvious.  相似文献

Mouse photoreceptor cells exhibit local accumulations of osmium deposits after prolonged osmic staining at slightly elevated temperatures. Deposits were evident along the membranes of outer segment lamellae, Golgi cisternae and vesicles, nuclear envelopes, and synaptic vesicles. Other membranes within the photoreceptor cells were unreactive. No osmium reactivity was seen in other cells of the retina except for osmiophilic outer segment material which had been phagocytized by the pigment epithelium. In the outer segments, inner segments, and synaptic regions of the photoreceptor cells, the amount of osmium reactivity was increased by light stimulation and decreased following extended dark adaptation. The possible significance of the localized osmium reactivity is discussed.  相似文献

A new type of cell has been identified in cercariae of Schistosoma mansoni. The perikarya (cell bodies) of these cells were located in the body (midsegment), in an area oral to the acetabulum (ventral sucker). Cytoplasmic processes extending from the perikarya ramified throughout the parenchyma of the anterior organ (oral sucker), body, and tail segments by following the path of the nerve processes from the neuropile. The perikarya of these cells had heterochromatic nuclei and a predominance of particulate material and granules (240-360 nm) in their cytoplasm. Aggregates of granules (240-360 nm) and associated vesicles (34 nm) were scattered throughout the cytoplasmic processes of the cells and formed distinct varicosed areas. These processes often connected to the tegument in the midsegment (body) of the cercariae. The granules and associated vesicles reacted (became electron dense) with fixatives reported to be detectors of biogenic amines: The glutaraldehyde/osmium tetroxide fixation procedure rendered the granules electron dense while the glutaraldehyde/chromate/osmium tetroxide fixation procedure rendered the granules and the associated vesicles electron dense. The chromate solution of the latter procedure was responsible for the electron density of the associated vesicles. The morphology of these cells (their long ramifying cytoplasmic processes) and their reaction to chromium suggests that they are probably biogenic aminergic sensory cells.  相似文献

Pinocytic vesicles, brush border microvilli, lysosomes and basolateral plasma membranes were isolated from rat kidney cortex and their biochemical composition and membrane turnover compared. Pinocytic vesicles are devoid of marker enzymes of brush border microvilli, such as alkaline phosphatase and 5′-nucleotidase, and of lysosomes, such as acid phosphatase and β-glucuronidase. The protein pattern as revealed by polyacrylamide gel electrophoresis differs for all four membranes. Analysis of the phospholipid composition shows that pinocytic vesicles are rich in the negatively charged phospholipid phosphatidylserine and have a low content of sphingomyelin and phosphatidylethanolamine.[14C]guanido-arginine, [3H]fucose and myo-[3H]inositol were preferentially incorporated into the pinocytic vesicles. Using a double label technique with leucine also, evidence of a more rapid turnover of the pinocytic vesicle membrane proteins was obtained.The results suggest that pinocytic vesicles are not derived from the brush border microvillous membrane but are independent entities that are newly synthesized during the pinocytic process.  相似文献

Various methods of chemical fixation and freeze-drying of Candida krusei were compared to determine the most appropriate method for the ultrastructural investigation of the thick walled organisms of this genus. Freeze-drying without chemical fixation was of little value because of insufficient variation in electron density. Potassium permanganate was able to penetrate the intact cell but failed to show cytoplasmic glycogen and lipid and some details of the cell wall. While normal glutaraldehyde, formaldehyde and osmium tetroxide treatment failed to permeate and preserve intracellular structures, several cycles of rapid freezing (–155°C) and thawing followed by glutaraldehyde fixation and osmium tetroxide post-fixation demonstrated the intracellular details of the majority of cells so treated.  相似文献

Summary Each pigment-cup eye of Polycystis naegelii consists of two retinal clubs and a single pigmented cell. The latter is divided into two cavities by a septum. Under bright illumination the photoreceptor process appears as a disk containing membranous laminar whorls; under faint illumination the latter are replaced by numerous straight, closely packed, microvilli. This morphological variation is correlated with the intensity of the photoreceptor's exposure to light. The ventral photoreceptor cells of Limulus polyphemus resemble the retinular cells of the lateral eyes both in electrical behavior and in morphology. Because of the great size of the ventral photoreceptor cells they are easy to impale with glass capillary micropipettes. Their location along the length of the ventral eye nerve makes them easy to dissect out and fix for electron microscopy. Each cell has a large, ellipsoidal soma that tapers into an axon whose length depends upon the distance of the cell from the brain. The cell body contains a rich variety of cytoplasmic organelles with an especially abundant endoplasmic reticulum. The most prominent structural feature is the microvillous rhabdomere, a highly modified infolding of the plasmalemma. The microvilli are tightly packed together within the rhabdomere, and quintuple-layered junctions are encountered wherever microvillar membranes touch each other. Glial cells cover the surface of the photoreceptor cell and send long, sheet-like projections of their cytoplasm into the cell body of the photoreceptor cell. Some of these projections penetrate the rhabdomere deep within the cell and form quintuple-layered junctions with the microvilli. Junctions between glial cells and the photoreceptor cell and between adjacent glial cells are rarely encountered elsewhere, indicating that there is an open pathway between the intermicrovillous space and the extracellular medium. The axon has a normal morphology but it is electrically inexcitable.  相似文献
