Basement membranes are layered structures of the extracellular matrix which separate cells of various kinds from the surrounding stroma. One of the frequently recurring questions about basement membranes is how these structures are formed in vivo. Up to a few years ago, it was thought that basement membranes were formed spontaneously by a process of self-assembly of their components. However, it has now become clear that cell membrane receptors for basement membrane components are essential factors for the formation and stability of basement membranes in vivo. The present review highlights the modern concepts of basement membrane formation.  相似文献

A survey of anthocyanins in the flowers and other organs of thirty-three species of three genera belonging to the Polygonaceae has been carried out. There are thirteen anthocyans. Cyanidin glycosides, especially the 3-glycoside (chrysanthemin), are present most commonly and peonidin glycosides including the arabinosylglucoside are found with low frequency. The distribution of malvidin 3,5-diglucoside (malvin) is confined to the species belonging to the sectionEchinocaulon of the genusPolygonum. It is noted that cyanidin itself occurs in the stems ofPolygonum perfoliatum andP. senticosum.  相似文献

Summary Using cuprolinic blue as a stain along with enzymic digestion, heparan sulphate has been identified as the main glycosaminoglycan in the basement membrane of human gallbladder epithelium. The amount of glycosaminoglycans was quantified by counting the number of molecular profiles cm–2 in electron micrographs of mildly, moderately and severely inflamed gallbladders. There is a significant increase (P=0.009) in the amount of glycosaminoglycans in the basement membranes of severely inflamed gallbladders compared with cases of mild chronic cholecystitis. Differences, although present, are less significant when mild and moderate or moderate and severe cholecystitis are compared. The findings suggest that there is a continuous accumulation of heparan sulphate in the basement membrane in chronic cholecystitis which increases in amount with the severity of inflammation.  相似文献

SYNOPSIS. Plasmodium japonicum and P. juxtanucleare are two very similar species of avian malaria parasites. The former was discovered in domestic fowl in Japan, and the latter in the same host species in Brazil; it has since been found in chickens in Uruguay, Mexico, and Ceylon. The present study, based on a Ceylon strain of P. juxtanucleare and slides from the Bamboo Partridge (Bambusicola thoracica sonorivox) of Taiwan indicates that the latter host is a natural reservoir of P. juxtanucleare, since this species appears to differ in no significant way from P. japonicum. Infection is common in the Bamboo Partridge (5 of 26 birds showed it), but it has not so far been found in any other avian species, although a total of 973 birds, belonging to 17 orders, 45 families, 110 genera, and 183 species, have been examined from this area. Since the younger stages of Plasmodium nucleophilum look very much like similar stages of P. juxtanucleare, they are compared. The older stages however are usually easily distinguishable, and the former species is so far known only from non-gallinaceous birds whereas the latter appears to be limited to gallinaceous ones.  相似文献

Serum amyloid P component (SAP) is a non-fibrillar glycoprotein belonging to the pentraxin family of the innate immune system. SAP is present in plasma, basement membranes, and amyloid deposits. This study demonstrates, for the first time, that the Goodpasture antigen-binding protein (GPBP) binds to human SAP. GPBP is a nonconventional Ser/Thr kinase for basement membrane type IV collagen. Also GPBP is found in plasma and in the extracellular matrix. In the present study, we demonstrate that GPBP specifically binds SAP in its physiological conformations, pentamers and decamers. The START domain in GPBP is important for this interaction. SAP and GPBP form complexes in blood and partly colocalize in amyloid plaques from Alzheimer disease patients. These data suggest the existence of complexes of SAP and GPBP under physiological and pathological conditions. These complexes are important for understanding basement membrane, blood physiology, and plaque formation in Alzheimer disease.  相似文献

The present paper deals with the taxonomy of the ostracod family Paradoxostomatidae Brady & Norman, 1889 from the coral reefs around Lizard Island, northern Great Barrier Reef, Australia. 18 species were found, all of them belonging to the genusParadoxostoma Fischer, 1855. 2 species had been previously described from Heron Island, southern Great Barrier Reef. 1 species had been described from the Red Sea. 2 species are left in open nomenclature.  相似文献

Summary Cultivated and wild Oryza species belonging to different genomic groups were studied with regard to their soluble seed-protein profiles. There is an essential uniformity in the banding patterns within various genomes and the basic patterns are not species-specific but genome-specific. O. meridionalis contains a subgenome similar to the A genome of O. rufipogon. Certain specific bands present among A genome species have been found to be useful in tracing the phylogenetic affinity between the cultivated species and their presumed wild progenitors.  相似文献

A heparan sulfate proteoglycan (HSPG) synthesized by murine parietal yolk sac (PYS-2) cells has been characterized and purified from culture supernatants. A monospecific polyclonal antiserum was raised against it which showed activity against the HSPG core protein and basement membrane specificity in immunohistochemical studies on frozen tissue sections from many rat organs. However, there was no reactivity with some basement membranes, notably those of several smooth muscle types and cardiac muscle. In addition, it was found that pancreatic acinar basement membranes also lacked the HSPG type recognized by this antiserum. Those basement membranes that lacked the HSPG strongly stained with antisera against laminin and type IV collagen. The striking distribution pattern is possibly indicative of multiple species of basement membrane HSPGs of which one type is recognized by this antiserum. Further evidence for multiple HSPGs was derived from the finding that skeletal neuromuscular junction and liver epithelia also did not contain this type of HSPG, though previous reports have indicated the presence of HSPGs at these sites. The PYS-2 HSPG was shown to be antigenically related to the large, low buoyant density HSPG from the murine Engelbreth-Holm swarm tumor. It was, however, confirmed that only a single population of antibodies was present in the serum. Despite the presence of similar epitopes on these two proteoglycans of different hydrodynamic properties, it was apparent that the PYS-2 HSPG represents a basement membrane proteoglycan of distinct properties reflected in its restricted distribution in vivo.  相似文献

In 1990, the role of basement membranes in human disease was established by the identification of COL4A5 mutations in Alport’s syndrome. Since then, the number of diseases caused by mutations in basement membrane components has steadily increased as has our understanding of the roles of basement membranes in organ development and function. However, many questions remain as to the molecular and cellular consequences of these mutations and the way in which they lead to the observed disease phenotypes. Despite this, exciting progress has recently been made with potential treatment options for some of these so far incurable diseases.  相似文献

In isolated mitochondrial membranes of Astasia longa, large and small circular substructures were observed with diameters of 1.30 and 0.35 m and thickness of 0.08 and 0.03 m, respectively. Such substructures were isolated by membrane treatment with proteolytic enzymes (proteinase K, trypsin) or by lipid solubilization with Triton X-100. After the removal of surface protein layer, we uncovered circular polyribosomes with similar diameters as those of original substructures. Polyribosomes were identified on the basis of their morphology, positive staining with uranyl acetate, a capacity for chloramphenicol-sensitive incorporation of 14C-amino acids into polypeptides, and from their buoyant density as estimated by equilibrium centrifugation in the CsCl density gradient. The conclusion is that, in mitochondrial membranes of A. longa, the translational apparatus is organized similarly to that in the membranes of chloroplasts and cyanobacteria, e.g., large and small circular polyribosomes situated within the membrane ring-shaped substructures are the basics for the formation of the latter.  相似文献

Summary The ultrastructure of the secondary lamellae has been examined in four species of free-swimming elasmobranchs, two species of Raia and five species of bony flatfishes. Microvilli which are present on the outer epithelial surface vary in form and size. It is suggested that a possible function of the microvilli is to anchor a surface mucus covering whose possible functions are discussed. Vesicles are found immediately beneath the microvilli in elasmobranchs but not in teleosts. Chloride and mucous cells are present on the secondary lamellae of all species and often have microvilli of greater length than on the other cells of the epithelium. Micropinocytotic vesicles are found on both sides of the basement membrane. The number of columns enclosed in the pillar cells varied from 5–11.The water-blood pathway showed variations in thickness not only between different species but also in different parts of individual secondary lamellae. A general trend in the mean total thickness was found, being greater in the swimming elasmobranchs (10.22 ) than in the rays (5.47 ) and bony flatfish (3.59 ). This trend is also seen in the greater length of microvilli and thickness of basement membrane in the elasmobranchs than in the teleosts.  相似文献

The ultrastructural distribution of specific connective-tissue components in the normal rat lung was studied by electron immunohistochemistry. Three of these components were localized: type I collagen, fibronectin and laminin. Type I collagen was present not only in major airways and vascular structures, but also in alveolar septa. Laminin was found in all basement membranes, and only in basement membranes, demonstrating once more that this glycoprotein is an intrinsic component of the basement membrane. Fibronectin was found free in the interstitium and on the surfaces of collagen fibers. The basement membranes of bronchial, glandular and endothelial cells of large vessels lacked fibronectin; however, capillary endothelial and occasionally epithelial alveolar basement membranes contained some fibronectin in an irregular, spotty distribution. This localization suggests that in the lung, as in other tissues, fibronectin is not an intrinsic component of the basement membrane, but rather a stromal and plasma protein. Only basement membranes in the alveolar parenchyma contained "trapped" plasma fibronectin.  相似文献

Nidogen 1 is a highly conserved protein in mammals, Drosophila melanogaster, Caenorhabditis elegans, and ascidians and is found in all basement membranes. It has been proposed that nidogen 1 connects the laminin and collagen IV networks, so stabilizing the basement membrane, and integrates other proteins, including perlecan, into the basement membrane. To define the role of nidogen 1 in basement membranes in vivo, we produced a null mutation of the NID-1 gene in embryonic stem cells and used these to derive mouse lines. Homozygous animals produce neither nidogen 1 mRNA nor protein. Surprisingly, they show no overt abnormalities and are fertile, their basement membrane structures appearing normal. Nidogen 2 staining is increased in certain basement membranes, where it is normally only found in scant amounts. This occurs by either redistribution from other extracellular matrices or unmasking of nidogen 2 epitopes, as its production does not appear to be upregulated. The results show that nidogen 1 is not required for basement membrane formation or maintenance.  相似文献

Mushroom bodies are prominent brain neuropils present in most arthropod representatives. Similar structures in the brain of certain polychaete species are possibly homologous to these structures. Using three-dimensional reconstruction techniques, we investigated the structural composition of the mushroom body neuropils in the polychaete species Nereis diversicolor and Harmothoe areolata. Comparative analysis revealed a common organization of neuropil substructures in both species that closely matches the basic assembly of arthropod mushroom bodies. Concurring with earlier homology assessments, these neuroarchitectural similarities provide support for a common origin of mushroom body neuropils in polychaetes and arthropods. Beyond that, differences in the morphological differentiation of neuropil substructures indicate polychaete mushroom bodies to show a high degree of morphological variability, thus impeding the quest for a common ground pattern of these brain centers.  相似文献

The authors showed stabilizing effect of rhenium binuclear cluster compounds with organic ligands on erythrocytic membranes. Estimation of functional condition of cells was studied by a chemical fragility method. Biphase type of rhenium complexes concentration curves stabilization has been found out; the influence of organic radicals and axial substructures (Cl- and Br-) on degree of stabilization has been showed. Different mechanisms of interaction of the investigated complexes with erythrocytic are suggested.  相似文献

Pollen of some species of the genus Quercus shows rod-shaped substructures in fresh or acetolysed exines, while in other species rod substructure is mostly masked by sporopollenin. Oxidation with potassium permanganate removes exine substance (sporopollenin) from between the rod substructures. We propose that the rods include receptors for sporopollenin. The sporopollenin between rods we refer to as ‘receptor-independent sporopollenin’. Pollen of Typha, when mature, has tectal surfaces with concave tops and sides, whereas during development the tectal surfaces are smoothly rounded. After acetolysis treatment followed by potassium permanganate the tectum surfaces again appear rounded. When these exines are subsequently eroded by a fast atom source, rod-shaped substructures are seen to protrude from the tectum. These structures are equivalent in size and shape to the rods of the exine of Quercus. Sporopollenin that accumulates over and masks rod substrucutre is less resistant to our degradative methods than the sporopollenin in rod structures of exines. We suggest that the exine material we call “receptor-independent sporopollenin” be given a simple positive name, such as masking-sporopollenin or abbreviated to masking-spn.  相似文献

Basement membranes are thin sheets of extracellular proteins situated in close contact with cells at various locations in the body. They have a great influence on tissue compartmentalization and cellular phenotypes from early embryonic development onwards. The major constituents of all basement membranes are collagen IV and laminin, which both exist as multiple isoforms and each form a huge irregular network by self assembly. These networks are connected by nidogen, which also binds to several other components (proteoglycans, fibulins). Basement membranes are connected to cells by several receptors of the integrin family, which bind preferentially to laminins and collagen IV, and via some lectin-type interactions. The formation of basement membranes requires cooperation between different cell types since nidogen, for example, is usually synthesized by cells other than those exposed to the basement membranes. Thus many molecular interactions, of variable affinities, determine the final shape of basement membranes and their preferred subanatomical localization.  相似文献

 Fossil vegetative and reproductive structures from deposits of the Raritan Formation in New Jersey (Turonian, Upper Cretaceous, ∼90 MYBP) include ferns, gymnosperms, and angiosperms. Gymnosperms collected from this formation have been known since the beginning of the 20th century. Fossil leaves, wood and seed cones have been are identified as belonging to the Cupressaceae, Pinaceae, and Taxodiaceae. In the present contribution, we describe a series of fossil dwarf shoots, leaves and a pollen cone belonging to the family Pinaceae. Fossils are charcoalified with preserved three-dimensional structure and excellent cellular detail. The dwarf shoots are assigned to a new species Prepinus crossmanensis sp. nov. and to the previously described Pinus quinquefolia Jeffrey. The new species Prepinus crossmanensis differs in size, shape, presence of hypodermis, sclerenchyma and stomata in the cataphylls, and number and shape of needle leaves from previously known species. Also, isolated leaves were found that were assigned to the new species, Prepinus raritanensis sp. nov. The new species is differentiated by the size and shape of stomata, the presence of number of layers of the hypodermis; and the cell-shape and number of layers of the mesophyll and transfusion tissue. A previously undescribed male cone, Amboystrobus cretacicum gen. and sp. nov., has an axis with spirally attached microsporophylls, each bearing two abaxial ovoid microsporangia. The pollen grains are monosulcate and bisaccate (eusaccate), with an elliptical corpus, granulate exine sculpture, and honeycomb-like wall structure. We have isolated sequences belonging to Tyl-copia group retrotransposons from the genomes of an amphibian (Pyxicephalus adspersa) and three reptiles (Conolophus subscristatus, Amblyrynchus cristatus and Pytas mucosus). Two different seqences were found in the amphibian (Tpa1 and Tpa2). Each is present in several copies per genome and absent from the genomes of two other amphibian species. The C. subcristatus sequence Tcs1 is present in multiple copies in both its host genome (Galapagos land iguana) and the genome of the related Galapagos marine iguana (A. cristatus). There is little or no polymorphism in Tcs1 insertions between different individual animals, suggesting that this sequence is not transposing rapidly in either iguana genome. The P. mucosus sequence Tpm1 shows a discontinuous distribution in snake species, suggesting that it has either been lost from many lineages during vertical germline transmission or has been transferred horizontally in some snake species. Phylogenetic comparisons of all these sequences with each other and with other members of this retrotransposon group from other animals and plants show that sequences within a particular vertebrate species are most closely related to each other, consistent with a vertical transmission model for their evolution.  相似文献
