Summary Restriction fragment patterns of mitochondrial DNAs (mtDNAs) from 13 carrot cultivars (Daucus carota ssp. sativus), wild carrot (ssp. carota), ssp. gummifer, and D. capillifolius were compared with each other using four restriction endonucleases. The mtDNAs of the 13 carrot cultivars could be classified into three distinct types — I, II and III — and were also clearly distinguishable from the mtDNAs of wild carrot (type IV), gummifer (V) and D. capillifolius (VI). The proportions of common restriction fragments (F values) shared by two of the three mtDNA types (I, II and III) of carrot cultivars were approximately 0.5–0.6. The F values were 0.4–0.5 for mitochondrial genomes between wild carrot, ssp. gummifer and D. capillifolius. The mitochondrial genomes between wild carrot and the carrot cultivars showed closer homologies those between wild carrot, ssp. gummifer, and D. capillifolius. The diversity of the mitochondrial genomes among the carrot cultivars is too high to presume that it was generated from the cytoplasm of only one common ancestor during the relatively short history of carrot breeding. These results suggested that the three types of cytoplasms found in the carrot cultivars might have existed in a prototype of D. carota in pre-historical times.  相似文献

Expression of the rolB gene of A. rhizogenes T-DNA triggers root differentiation in transformed plant cells. In order to study the regulation of this morphogenetic gene, the GUS reporter gene was placed under the control of several deleted fragments of the rolB 5 non-coding region: carrot disc transformations and the analysis of transgenic tobacco plants containing these constructions identified the presence of distinct regulatory domains in the rolB promoter. Two regions (located from positions –623 to –471 and from –471 to –341, from the translation start codon) control the level but not the tissue specificity of rolB expression: progressive deletions of the rolB promoter starting from position –1185 to –341, although at different levels, maintained the same pattern of GUS expression — maximal in root meristems and less pronounced in the vascular tissue of aerial organs. Further deletion of 35 bp, from –341 to –306, drastically affected tissue specificity: GUS activity was still clearly detectable in the vascular tissue of the aerial organs while expression in the root meristem was totally suppressed. Analysis of transgenic embryos and seedlings confirmed that distinct promoter domains are responsible for meristematic (root) and differentiated (vascular) expression of rolB. Finally, we present data concerning the effects of plant hormones on the expression of rolB-GUS constructions.  相似文献

Effects of enzymatic preparations--pectomacerin, hemicellulase, and Trichoderma viride 13/10 cellulase--on the plant immunological status were studied by examples of the pathosystems carrot root-white rot agent (Sclerotinia libertiana) and carrot root-black rot agent (Rhizopus nigricans). It was demonstrated that these preparations reduced the damage of infections, namely, decreased the permeability of cell membranes in infected tissue and stimulated its defense responses, which manifested itself in a stable elevation in the content of phenolic compounds and formation of tissue protective barriers.  相似文献

Dermaseptin B1 is a potent cationic antimicrobial peptide found in skin secretions of the arboreal frog Phyllomedusa bicolor. A synthetic derivative of dermaseptin B1, MsrA2 (N-Met-dermaseptin B1), elicited strong antimicrobial activities against various phytopathogenic fungi and bacteria in vitro. To assess its potential for plant protection, MsrA2 was expressed at low levels (1–5 g/g of fresh tissue) in the transgenic potato (Solanum tuberosum L.) cv. Desiree. Stringent challenges of these transgenic potato plants with a variety of highly virulent fungal phytopathogens—Alternaria, Cercospora, Fusarium, Phytophthora, Pythium, Rhizoctonia and Verticillium species—and with the bacterial pathogen Erwinia carotovora demonstrated that the plants had an unusually broad-spectrum and powerful resistance to infection. MsrA2 profoundly protected both plants and tubers from diseases such as late blight, dry rot and pink rot and markedly extended the storage life of tubers. Due to these properties in planta, MsrA2 is proposed as an ideal antimicrobial peptide candidate to significantly increase resistance to phytopathogens and improve quality in a variety of crops worldwide with the potential to obviate fungicides and facilitate storage under difficult conditions.  相似文献

The impact of a biocontrol agent spreading from a point source on crop–weed interactions was modeled. The model encompassed: (i) severity of crop–weed competition as affected by a rust pathogen, (ii) velocity of spread of the rust pathogen, and (iii) density of infected plants introduced in the weed population as starting points (inoculum sources) for an epidemic. The model was parameterized for a study system encompassing the crop Daucus carota (carrot), the weed Senecio vulgaris (common groundsel), and its antagonist Puccinia lagenophorae. The parameters of (i) were estimated in a greenhouse study using a response-surface design. Estimates of the parameter of (ii) were obtained from the literature. The density of infected plants (iii) was varied to simulate crop loss as function of density. Simulations were run for various densities of the weed and various velocities of rust pathogen spread. The results of the simulations indicated a crop-loss ranging from 5 to 10% at levels of relatively weak D. carotaS. vulgaris competition. Density of inoculum sources and velocity of P. lagenophorae spread had only minor effects on crop loss. In contrast, density of inoculum sources and velocity of spread had major effects on crop loss at levels of intermediate (range of 10–35% loss) and severe competition (range of 30–70% loss). The results are discussed both with respect to biological control of S. vulgaris using P. lagenophorae as biocontrol agent and as a general model describing the impact of the spatial dynamics of a pathogen (natural enemy) on plant competition.  相似文献

Diseased carrot seeds were treated with selected micro-organisms isolated from soils, carrot seeds and tap roots. The effects of those antagonists on the control of Alternaria radicina were evaluated by growing-on tests on water agar, filter paper, vermiculite and in a potting medium (BVB no. 4). The germination percentage, emergence percentage and the disease severity of those carrot seeds treated with Burkholderia (Pseudomonas) cepacia no.229 were significantly (P=0.05) differed from the non-treated seeds and the seed treated with other antagonists. The effects of B. cepacia no.229 in promoting seed emergence and controlling disease were as good as those seeds treated with iprodione (100 p.p.m.). Black rot lesions on carrot tap roots were significantly reduced (P=0.05) in size when roots were treated with B. cepacia no 229 or Bacillus amyloliquefaciens no. 224 compared to the nontreated roots. Also, B. cepacia no. 229 significantly (P=0.05) reduced black rot on the foliage of carrot compared to check.  相似文献

The effect of growth retardants on anthocyanin production was studied in wild carrot (Daucus carota) cell suspension cultures. Paclobutrazol [(2RS,3RS) — 1 — (4-chlorophenyl) — 4,4 —dimethyl-2-(1,2,4-triazol-1-yl) pentan-3-ol], uniconazole [(E)-1-(4-chlorophenyl-4,4 —) dimethyl-2-(1,2,4-triazol-1-yl)-1-penten-3-ol], tetcyclacis [5-(4-chloro-phenyl) -3,4,5,9,10-pentaaza-tetracyclo-5, 4, 102,6, O8,11 — dodeca-3, 9-diene], ancymidol [-cyclopropyl — 4 — methoxy-(pyrimidine-5-yl)benzyl alcohol] and CCC (2-chloro-ethyltrimethylammonium chloride) increased anthocyanin accumulation. AMO-1618 [(2-isopropyl-5-methyl-4-trimethyl-ammonium-chloride)-phenyl-1-piperidinium carboxylate] did not increase anthocyanin accumulation in the first passage but did increase it during the second passage on medium for improved anthocyanin accumulation. Prohexadione (3,5-dioxo-4-propionylcyclohexane carboxylic acid) decreased anthocyanin accumulation by 10%–12.5%.The inhibitory effect of gibberellin on anthocyanin accumulation was reversed by paclobutrazol. Paclobutrazol together with 10–6M GA3 increased anthocyanin level from 33% of control in GA3 treated cell suspension to 76%. These results are consistent growth retardants increasing anthocyanin accumulation in carrot cell suspension cultures by inhibiting gibberellin biosynthesis.  相似文献

Thioredoxin and NADP-thioredoxin reductase from cultured carrot cells   总被引:7,自引:0,他引:7  
Dark-grown carrot (Daucus carota L.) tissue cultures were found to contain both protein components of the NADP/thioredoxin system—NADP—thioredoxin reductase and the thioredoxin characteristic of heterotrophic systems, thioredoxin h. Thioredoxin h was purified to apparent homogeneity and, like typical bacterial counterparts, was a 12-kdalton (kDa) acidic protein capable of activating chloroplast NADP-malate dehydrogenase (EC 1.1.1.82) more effectively than fructose-1,6-bisphosphatase (EC 3.1.3.11). NADP-thioredoxin reductase (EC 1.6.4.5) was partially purified and found to be an arsenite-sensitive enzyme composed of two 34-kDa subunits. Carrot NADP-thioredoxin reductase resembled more closely its counterpart from bacteria rather than animal cells in acceptor (thioredoxin) specificity. Upon greening of the cells, the content of NADP-thioredoxin-reductase activity, and, to a lesser extent, thioredoxin h decreased. Black spot leads to great marigold losses worldwide. The disease is characterized by black spots on leaves and stems in its early stages, and the whole plant has black rot at the advanced stage. In this report, 6 of 217 Alternaria strains isolated from lesions of marigold plants in Beijing were randomly selected. The morphological characteristics and a pathogenic tree based on two protein‐coding genes (gpd and alt a 1) indicated that Alternaria tagetica is the causal agent of marigold black spot in Beijing. All six Alternaria strains could successfully re‐infect marigold, but they could not infect carrot or zinnia by either spore spray in a greenhouse or planting experiments in the epidemic area. This is the first report of the A. tagetica pathogen being isolated from marigold in Beijing.  相似文献

Radopholus spp. were reared in carrot tissue culture via established procedures, with slight modification. Several plant tissue maceration enzymes and flotation media (salts and sucrose) were evaluated with regard to nematode toxicity and extraction efficiency. Best extraction of viable nematodes and eggs was attained when carrot tissue infested with Radopholus citrophilus or R. similis was macerated with a mixture of 0.50% driselase and 0.50% cellulysin, w/v each, with 2.5 ml of enzyme solution based for each gram of carrot tissue. Maceration slurries containing carrot tissue and nematodes were maintained in open flasks on a rotary shaker (175 rpm) at 26 C for 24 hours. Nematodes and eggs were extracted from resultant culture slurries by flotation with MgSO₄-7H₂0 (sp gr 1.1). A protocol is presented to extract large quantities of viable burrowing nematodes and their eggs from carrot disk cultures.  相似文献

A binary competition experiment between carrot (Daucus carota L.) and Chenopodium album L. was conducted in a greenhouse at seven population densities and eight nutrient concentrations to investigate the effects of a nutrient gradient on plant competition in both monocultures and mixtures. The patterns of carrot biomass allocation (measured as root : shoot ratio) in monocultures and mixtures with C. album were affected by both nutrient availability and population density. Chenopodium album had a broader response to nutrient concentrations than carrot. The maximum yield of carrot in both monocultures and mixtures occurred at fourfold the standard concentration of nutrients, while C. album in both monocultures and mixtures had the maximum yield at 16-fold the standard nutrient concentration. The yield–density relationship of carrot tended to be increasing or asymptotic at lower nutrient concentrations but parabolic at higher concentrations, whereas that of C. album was little affected by nutrient availability. Nutrient availability had a profound influence on the competitive relationships between the two species: at both low and high nutrient concentrations, C. album tended to be more competitive than carrot, while at intermediate levels of nutrients, carrot was more competitive than C. album. Our results suggest that in relation to competitive performance, the weed has a greater ability to adapt itself to varying environments than does the crop. Additionally, the relative merits of the quantitative measures of competitive ability are briefly discussed.  相似文献

Plant species and genotypes within the same species may differ in phosphorus efficiency. The objective of this research was to study phosphorus efficiency of cabbage (Brassica oleraceae L.), carrot (Daucus carota L.), and potato (Solanum tuberosum L.) and to quantify the contribution of morphological root characteristics to P uptake of the plant species. An experiment was conducted in a glasshouse with six P levels: 0, 12, 27, 73, 124 and 234 mg P kg–1 soil, and with six replications. Cabbage attained 80% of its maximum yield already at the level of no P supply, whereas carrot and potato reached only 4 and 16% of their highest yields respectively at this level of P supply. This indicated that cabbage was P-efficient compared to carrot and potato. Root/shoot ratio (cm root g–1 shoot d. m.) increased in the order of cabbage < carrot < potato, and was enhanced at lower P levels. Root hair length was not affected by P level, and averaged 0.22, 0.03 and 0.18 mm for cabbage, carrot, and potato, respectively. Predicting P uptake by a mechanistic simulation model revealed that root hairs contributed about 50% to the total P uptake of cabbage and potato, but only 0.3% to that of carrot. The relationship between the observed P uptake and the predicted P uptake of the plants revealed that model parameters explained nearly 4/5th of the total P uptake of carrot and potato, but only 2/5th of that of cabbage. This showed that the P uptake of cabbage was strongly under-predicted, whereas that of carrot and potato was predicted well. Therefore, it was hypothesised that cabbage may have the ability to mobilise and take up soil P additionally by other root mechanisms such as exudation of organic acids.  相似文献

Field trials were carried out in upstate New York in 1990, 1992, 1993, and 1994 and in Chile in 1992–1993 and 1993–1994 in order to evaluate the ability of various strains ofTrichodermaspp. to control bunch rot of grape, to assess the compatibility and possible additive effects of selected biocontrol fungi and dicarboximide fungicides, and to determine factors affecting biocontrol efficacy. In 1990, three strains ofTrichodermaspp. were evaluated for their biocontrol ability, and all provided significant control ofBotrytis cinerea.As few as two late applications of the biocontrol fungi were nearly as effective as up to five applications throughout bloom and fruit development. Trials in New York in 1992 and in Chile in 1992–1993 indicated thatTrichoderma harzianumcould replace some applications of iprodione or vinclozolin with little reduction in efficacy. In New York in 1993, we found that applications ofT. harzianumat bloom and early fruit development followed by a tank-mix application ofT. harzianumand half rates of iprodione gave extremely effective control of bunch rot. In 1994, less effective control was obtained than in earlier years. Addition of a nutritive adhesive (Pelgel, a mixture of carboxymethyl cellulose and gum arabic) applied with the biocontrol agent tended to improve results. Thus, biological control of bunch rot of grape withT. harzianumcan be an effective method of management of this disease.  相似文献

Comparative analysis of two optical methods—optical coherence tomography (OCT) and optical coherence microscopy (OCM)—was made for vital visualization of plant tissues in tomato (Lycopersicon esculentum Mill), spiderwort (Tradescantia pallida (Rose) D. Hunt), orach (Atriplex sp.), and leaves and seeds of medium starwort (Stellaria media L.). The obtained OCT- and OCM-images allowed the morphological and functional state of plant tissues to be assessed in vivo. A higher spatial resolution of the OCM method, as compared to OCT method, allowed plant morphological structures to be identified with greater confidence. Bacteria were isolated from corn plants and efficacy of a selected isolate for control of corn stalk rot caused by Fusarium graminearum was evaluated. Abstract

In this study, an antagonistic yeast isolate, Wickerhamiella versatilis was considered as a promising biocontrol agent against Pectobacterium carotovorum subsp. Carotovorum (Pcc) the causal agent of soft rot disease of potato. Antagonistic yeast inhibited the growth of Pcc in vitro, and reducing the soft rot severity of infected potato tubers (cv. Diamant) under greenhouse conditions. Consequently, cellulase and pectinase hydrolytic activities in infected potato tubers with yeast?+?Pcc were decreased compared with infected tubers with Pcc. The histological characterization of treated potato tubers with antagonistic yeast W. versatilis using scanning electron microscope showed the accumulation of extracellular substances that may induce plant resistant and protects potato tubers from hydrolysis and damages. This study may introduce the possibility of using the antagonistic yeast isolate, as a biocontrol agent against soft rot of potato tubers.  相似文献

As an alternative to the use of synthetic chemical fungicides to control plant disease, aluminium‐containing salts were evaluated for their effects on the mycelial growth of various fungal or fungus‐like pathogens and their ability to control carrot cavity spot (Pythium sulcatum) and potato dry rot (Fusarium sambucinum). Results showed that various aluminium‐containing salts provided strong inhibition of all the tested pathogens (Alternaria solani, Botrytis cinerea, F. sambucinum, P. sulcatum and Rhizopus stolonifer) with minimal inhibitory concentration of 1–10 mM. Aluminium chloride and aluminium sulphate were generally the most effective, inhibiting mycelial growth of pathogens by as much as 47% and 100%, respectively, at a salt concentration of 1 mM. Applied at 5 mM, aluminium sulphate also provided 28% and 100% inhibition of dry rot and cavity spot, respectively. Aluminium chloride (5 mM) reduced dry rot by 25% whereas aluminium lactate (5 mM) decreased cavity spot lesions by 86%. These results indicate that various aluminium‐containing salts may provide an alternative to the use of synthetic fungicides to control these pathogens.  相似文献

ECP31, an embryogenic-cell protein from carrot (Daucus carota L.), was purified by sequential column-chromatographic steps and digested by V8 protease on a nitrocellulose membrane. The resultant peptides were separated by reverse-phased column chromatography and sequenced. The sequences obtained were 70–80% homologous to those of a late-embryogenesis-abundant protein (D34) from cotton (Baker et al, 1988, Plant Mol. Biol. 11, 227–291). The level of ECP31 in somatic embryos of carrot was increased by treatment of the embryos with 3.7 · 10–6 M abscisic acid (ABA) for 48 h, and there was no change in this enhanced level for up to 192 h in the presence of ABA. No similar enhancing effect of ABA was observed on the level of ECP31 in embryogenic callus or segments of carrot hypocotyls. Measuring uncoupled oxygen uptake rate (OUR) could provide a convenient method for quantifying changes in the metabolic activity of plant cultures caused by hydrodynamic shear. Experiments on Daucus carota (carrot) cells were performed in a novel O2-permeable Couette viscometer at varying levels of laminar shear (6 to 100 N m–2). When the uncoupled OUR of the cells was compared with mitochondrial activity (determined by 2,3,5-triphenyl tetrazolium chloride assay), a significant correlation was observed (R=0.91 by linear regression).  相似文献

The application of a specific species of willow—Salix amygdalina L., marked by high transpiration ability—is a cheap and effective method of landfill leachate disposal. A 2-year study examined the effectiveness of leachate evapotranspiration from soil–plant systems with willow species S. amygdalina L. Evapotranspiration from soil–plant systems planted with willow was from 1.28 up to 5.12 times higher than evaporation from soil surface barren of vegetation. This proves the usefulness of soil–plant systems with willow in landfill leachate treatment through vaporization. Evapotranspiration efficiency, as opposed to total amount of water added into the lysimeter, was not strong enough to vaporize all input of the landfill leachate in the lysimeters. This may indicate that the ground water requires isolation when soil systems remain under landfill leachate irrigation. Linear dependence between willow biomass growth and transpiration was observed to be significant (p < 0.05). Additionally, the research showed that the application of sewage sludge into the soil caused an increase in vaporization efficiency.  相似文献
