Summary A procedure is described for the isolation of large numbers of viable aleurone protoplasts. After treatment with Onozuka cellulase protoplasts were obtained which were surrounded by a thin, Onozuka-resistant wall. These cells were termed spheroplasts. Treatment of spheroplasts with Glusulase digested away the residual wall, yielding naked protoplasts. Measurements of respiration using a Clarke-type oxygen electrode indicated that aleurone cells isolated by this procedure were viable.Work supported by National Science Foundation grant GB-27468.  相似文献

Globo H (Fuc12Gal13GalNAc13Gal14Gal14Glc) is a carbohydrate structure that shows enhanced expression in many human carcinomas. From mice immunized with a globo H-KLH (keyhole limpet hemocyanin) synthetic conjugate an IgG3 monoclonal antibody (mAb VK-9) was derived that recognizes the globo H structure. Serological analysis showed that the minimal structure recognized by this mAb was the tetrasaccharide sequence Fuc12Gal13GalNAc13Gal. An isomeric structure with an internal GalNAc linkage was also recognized but less efficiently. mAb VK-9 did not react with many related structures, such as galactosylgloboside, globoside, H type 1, H type 2 blood group structures or fucosyl-gangliotetraosyl ceramide, but did react weakly with globo A ceramide. Not only did mAb VK-9 react with carbohydrate-protein conjugates but it could also recognize globo H-ceramide and human tumor cells expressing globo H. These results suggest that globo H-KLH could be explored as a vaccine in the treatment of carcinoma patients.  相似文献

Summary In this paper a steady-state kinetic study on the system lactate dehydrogenase--thiopyruvate--thiolactate is presented and the possible mechanistic and physiological implications are discussed.At pH 7.4 the equilibrium between -thiopyruvate and -thiolactate, in the presence of NADH and lactate dehydrogenase is largerly shifted towards the formation of -thiolactate as in the case of pyruvate and lactate. This can be relevant in connection with the mixed disulfide between cysteine and -thiolactate that is observed to be present in the mammalian body fluids.The catalytic mechanism is of the bi-bi compulsory order type, and rapid equilibrium conditions for the binding of the first substrate (NADH) are shown to apply. Summary (1) A method is described for the production of acute deficiencies of copper, zinc or molybdenum in large scale sand cultures in which an adequate and satisfactory supply of water was obtained by treating rain water with ion-exchange resins. Results compared favourably with the use of glass distilled water.(2) Visual symptoms of copper, zinc and molybdenum deficiencies in large seeded leguminous crops, and in other species are summarised.(3) Seed reserves played an important part in the incidence of copper and molybdenum deficiencies and to a lesser extent of zinc deficiency in pea, runner bean, dwarf bean and broad bean.(4) The deficient seed was obtained by harvesting seed from plants grown in the respective deficiency sand cultures, and was compared with seed obtained similarly from complete nutrient sand cultures or commercially from field-grown plants.(5) Seed reserves of molybdenum were usually sufficient for a complete generation but reserves of copper and zinc were not adequate for the whole copper or zinc requirements of the plants.(6) Analytical data showed a marked decrease in the copper or molybdenum content of deficient seed, and a less striking decrease in zinc.(7) Foliage of deficient plants showed decreased content of copper and molybdenum and often of zinc compared with plants given the complete nutrient treatment, but use of deficient seed only decreased yields without further appreciably decreasing the mineral content of the foliage.  相似文献

The ion channel of the nicotinic acetylcholine receptor (nAChR) is believed to be lined by transmembrane M2 helices. A 4-8-12 sequence motif, comprising serine (S) or threonine (T) residues at positions 4, 8 and 12 of M2, is conserved between different members, anion and cation selective, of the nAChR superfamily. Parallel bundles of 4-8-12 motif-containing helices are considered as simplified models of ion channels. The relationship between S and T sidechain conformations and channelion interactions is explored via evaluation of interaction energies of K+ and of Cl ions with channel models. Energy calculations are used to determine optimal 2 (C-C\-O-H) values in the presence of K+ or Cl ions. 4-8-12 motif-containing bundles may form favourable interactions with either cations or anions, dependent upon the 2 values adopted. Parallel-helix and tilted-helix bundles are considered, as are heteromeric models designed to mimic the Torpedo nAChR. The main conclusion of the study is that conformational flexibility at 2 enables both S and T residues to form favourable interactions with anions or cations. Consequently, there is apparently no difference between S and T residues in their interactions with permeant ions, which suggests that the presence of T vs. S residues within the 4-8-12 motif is not a major mechanism whereby anion/cation selectivity may be generated. Summary A female Pied Flycatcher (Ficedula hypoleuca) built over a Coal Tits' (Parus ater) nest containing five chicks. Nevertheless, the nestlings survived on account of their parents' feedings, and because they were probably lifted up in the nest-box sitting on top of the material brought into the box by the flycatcher-. After nest-completion the Pied Flycatcher- layed four eggs, but refused breeding. Instead, and probably due to the intense begging of the five Coal Tit chicks, the Flycatcher-pair immediatly started to rear its young. Based on the good care the chicks were given by their parents and foster-parents they came along nicely and fledged some days later.  相似文献

Summary The nonlinear membrane current-voltage relationship (I–V curve) for intact hyphae ofNeurospora crassa has been determined by means of a 3-electrode voltage-clamp technique, plus quasi-linear cable theory. Under normal conditions of growth and respiration, the membraneI–V curve is best described as a parabolic segement convex in the direction of depolarizing current. At the average resting potential of –174 mV, the membrane conductance is 190 mhos/cm2; conductance increases to 240 mhos/cm2 at –300 mV, and decreases to 130 mhos/cm2 at 0 mV. Irreversible membrane breakdown occurs at potentials beyond this range.Inhibition of the primary electrogenic pump inNeurospora by ATP withdrawal (with 1mm KCN) depolarizes the membrane to the range of –40 to –70 mV and reduces the slope of theI–V curve by a fixed scaling factor of approximately 0.8. For wild-typeNeurospora, compared under control conditions and during steady-state inhibition by cyanide, theI–V difference curve — presumed to define the current-voltage curve for the electrogenic pump — is a saturation function with maximal current of 20 A/cm2, a half-saturation potential near –300 mV, and a projected reversal potential of ca. –400 mV. This value is close to the maximal free energy available to the pump from ATP hydrolysis, so that pump stoichiometry must be close to 1 H+ extruded:1 ATP split.The time-courses of change in membrane potential and resistance with cyanide are compatible with the steady-stateI–V curves, under the assumption that cyanide has no major effects other than ATP withdrawal. Other inhibitors, uncouplers, and lowered temperature all have more complicated effects.The detailed temporal analysis of voltage-clamp data showed three time-constants in the clamping currents: one of 10 msec, for charging the membrane capacitance (0.9 F/cm2) a second of 50–75 msec; and a third of 20–30 sec, perhaps representing changes of intracellular composition.  相似文献

Culturing a non-habituated cell line of Fagara zanthoxyloides Lam. (Rutaceae) in either auxin-free medium or cytokinin-free medium led to opposite effects on furoquinoline accumulation. It appeared that production of skimmianine and -fagarine in the cells was strongly correlated with the presence of exogenous BAP: the levels of both alkaloids were 9 times lower when cells were cultured without cytokinin than in the control culture. NAA removal induced a slight stimulation of skimmianine and -fagarine accumulations, 1.2 and 1.9 times respectively. Culturing the cells in a PGR-free medium generated skimmianine and -fagarine levels that were 3.5 and 2.1 times lower, respectively, confirming the opposite effects of BAP and NAA on furoquinoline accumulation. Summary To investigate the impact of SOS induction on the distribution of spontaneous mutation, 111 recA441-mediated mutations were characterized at the DNA sequence level in the lacI gene of Escherichia coli. A 2.6-fold enhancement in lacI mutation frequency was observed after induction of the SOS system in the absence of mutagenic treatment, and specific classes of mutational events were induced. G : C C : G, G : C T : A and A : T T : A transversion events were specifically enhanced after SOS induction. A preferential 5-Y-Purine-3 neighbouring base specificity for these transversion events is reported here (normalised for mutation of the purine residue). In addition, a preference for transversion events at 5-C/GTGG-3 sequences is also observed. Fifty events were recovered at the lacI frameshift hotspot site and were equally represented by 4 bp addition and deletion events. This 1:1 ratio deviates significantly from the 4:1 distribution characteristic of spontaneous frameshift mutation in the RecA+ background and is a consequence of the fourfold induction of the (–)4 event. This abberrant distribution was confirmed by oligomeric probing of 474 independent recA441-mediated spontaneous lacI mutations.  相似文献

This paper discusses the widespread use of heritability calculations in recent behaviour research including behaviour genetics. In the sequel, a radical criticism concerning the basic axioms of the underlying, more general concept itself is presented. The starting point for testing the proclaimed universal validity of this concept stems from a fictitious yet realistic example taken from learning research. The theoretical result, based on the application of the conventional reasoning in this field, states that developmental processes — and learning is only one specific case out of an immense number of similar behavioural mechanisms — can neither be adequately described nor causally explained with sufficient reliability within the context of the heredity paradigm. On the contrary, an inherent inconsistency of the concept itself when applied to behaviour processes is demonstrated. Finally, a conceptual alternative involving a systems-theoretical approach to the problem is presented: In such a perspective it is the concept of cognition which represents the adequate explanatory theorem - a theorem in which quantitative processing of information from the environment is clearly revealed to belong to a subordinate level of living organization.  相似文献

In this paper, we offer a stability analysis of the cue interaction model of depth perception (House (1984)). Depth estimation using stereopsis suffers from the matching problem, the problem of correctly matching the retinal image of a feature in one eye, to its retinal image in the other eye. The Cue Interaction Model overcomes this by using monocular cues to disambiguate between the correct matches and the incorrect matches. Its decision making is based on the concept of cooperation and competition in a neural network. A general class of cooperative and competitive models has been mathematically analysed by Amari and Arbib (1977), with special attention given to equilibrium states and stability. In this paper we adapt their methods to study the above model. In particular, we prove that if the parameters are correctly tuned, the model successfully achieves its goals by suppressing the cues which represent the incorrect matches.Preparation of this paper was supported in part by NIH grant number NS-1 R01 NS24926 from NINCDS  相似文献

We study an age-dependent population equation with a nonlinear death rate of logistic type. The global asymptotic stability of the null solution is investigated when R(0)<1. If R(0)>1 we get the existence of a nontrivial steady state that becomes asymptotically stable itself, while the null solution is unstable. Residual toxicities of registered and selected experimental pesticides used on citrus against Agistemus industani Gonzalez (Acari: Stigmaeidae) were compared. Pesticides considered highly toxic to A. industani were: abamectin 0.15 EC at 731ml/ha+FC 435-66 petroleum oil at 46.8l/ha, pyridaben 75WP at 469g/ha, ethion 4EC at 7.01l/ha+FC 435-66 petroleum oil at 46.8l/ha, propargite 6.55 EC at 3.51l/ha, chlorfenapyr 2SC at 1.46l/ha applied alone or in combination with FC 435-66 petroleum oil at 46.8l/ha, sulphur 80DF at 16.81kg/ha, dicofol 4EC at 7.01l/ha, fenbutatin oxide 50WP at 2.24kg/ha, benomyl 50WP at 2.24kg/ha, benomyl 50WP at 1.68kg/ha+ferbam 76 GF at 5.60kg/ha, ferbam 76GF at 11.21kg/ha, neem oil 90EC at 46.8l/ha, and copper hydroxide DF (40% metallic copper) at 4.48kg metallic copper/ha+FC 435-66 petroleum oil at 46.8l/ha. Pesticides that were moderately to slightly toxic included: copper sulphate 98% at 4.48kg metallic copper/ha+FC 435-66 petroleum oil at 46.8l/ha, fenbuconazole 2F at 280ml/ha+FC 435-66 petroleum oil at 46.8l/ha, FC 435-66 petroleum oil applied alone at 46.8l/ha or 23.4l/ha, and diflubenzuron 25WP at 1.40kg/ha. Pesticides that were non-toxic included: fenbuconazole 2F at 585ml/ha, malathion 57EC at 5.85l/ha, FC 435-66 petroleum oil at 46.8l/ha, carbaryl 80S at 3.36kg/ha, chlorpyrifos 4EC at 4.68l/ha, and formetanate 92SP at 1.12kg/ha. Understanding the toxic effects of field weathered pesticides against key predacious mite species is important for effective IPM. The results of this study provide a comparison of direct and indirect toxic effects of various pesticides to A. industani under field conditions.  相似文献

Summary The effect of assortative mating on the genetic correlation between traits X and Y is considered. Assortation on trait X changes the magnitude of the genetic correlation but not its sign. There are two situations depending on the signs of the correlation between mates () and of the random mating genetic correlation (): 1) if sign () = sign (), then >, where is the genetic correlation at equilibrium after continued assortation, and 2) if sign () = sign (), then < . However, negative assortative mating is virtually powerless to alter the magnitude of the genetic correlation. The consequences of a mixed assortation model, e.g., high milk production females mated to fast growing males and lesser productive females mated to slower growing sires, were also studied. Mixed positive assortation always increases the genetic correlation, but negative assortation decreases it. The implications of assortative mating on correlated responses to selection and on the equilibrium covariances between relatives for pairs of traits are discussed.  相似文献

Summary Darwin's phrase place in natural economy, andSpencer's term correspondence can be regarded as first attempts to express the organism-environment relationships. The same concept has more recently been approached from the point of view of (1) life-form, (2) external activities, and (3) habitat. Though all these points are interlocking, they have been stressed differently in the writings of American and European ecologists. It is proposed that the term niche would be most useful and rational if applied to the total of relationships between a living organism (population, species) and its complete environment, both biotic and abiotic.  相似文献

Paddy levees form networks of narrow linear habitats and play various roles in cultural landscapes. Traditional landscapes on the west side of Lake Biwa consist of paddy field terraces and both stone and soil levees that have been maintained by paddy field management using local resources. Paddy levees in this study site are principally classified into five different types. Our study points out how differences in paddy levee structure as well as in management practices influence the plant species. Seventeen paddy levee transects were split into four habitat types based on their species components by TWINSPAN. Spatial characteristics and physical structures of paddy levees depended on natural conditions and human activities. The species–area curves of each levee type showed a clear distinction: the soil, stone and abandoned curves were steep, while the concrete and consolidated ones were gentle. The vegetation on consolidated levees was utterly different from the vegetation on traditional levee types from the aspect of species richness and species components. Soil type levees contained various woody plant species and included more diverse and indigenous plant species than abandoned type levees.  相似文献

The carbohydrate-binding specificity ofPseudomonas aeruginosa lectin I (PA-I) in iodinated or biotinylated form was studied. A large number of glycosphingolipids, as well as some glycoproteins and neoglycoproteins were used as ligands. Also, inhibition by free saccharides of PA-I binding to glycosphingolipids was tested. Mild cerebral anoxic/ischemic/stress insults promote tolerance and thereby protect the brain from subsequent lethal anoxic/ischemic insults. We examined whether specific activation of PKC , , , or isoforms is associated with ischemic preconditioning (IPC) in rat brain. IPC was produced by a 2-minute global cerebral ischemia. Membrane and cytosolic fractions of the hippocampi were immunoblotted using specific antibodies for PKC, , , and . PKC showed a significant translocation to the membrane fraction from 30 min to 4 h and PKC at 4 h following IPC. In contrast, the membrane/cytosol ratio of PKC showed a tendency to decrease at 30 min and 8 h, and the membrane/cytosol ratio of PKC was significantly decreased from 30 min to 24 h following IPC. These findings indicate PKC isoform-specific membrane translocations in the hippocampus after brief global brain ischemia and suggest that activation of PKC and PKC may be associated with IPC-induced tolerance in the rat hippocampus.  相似文献

We consider a general class of Markov population models formulated as stochastic difference equations. The population density is shown to converge either to 0, to +, or to a unique stationary distribution concentrated on (0, +), depending on the signs of the mean log growth rates near 0 and +. These results are applied to the Watkinson-MacDonald bottleneck model of annual plants with a seedbank, extended to allow for random environmental fluctuations and competition among co-occurring species. We obtain criteria for long-term persistence of single-species populations, and for coexistence of two competing species, and the biological significance of the criteria is discussed. The lamentably few applications to the problem at hand of classical limit-theory for Markov chains are surveyed.  相似文献
