A simple two-step synthesis of bufadienolides is reported. It consists in the addition of the dimethyl acetal of chloroketene to a steroidal 20-methylene 21-aldehyde and in the treatment of the resulting 2,2-dimethoxy 3-chloro 2,3-dihydropyran with sodium methoxide in dimethylsulfoxide. This method is exemplified by the synthesis of 3β-hydroxy-5α,14α-bufadienolide from 3β-acetoxy-20-methylene-5α-pregnan-21-al, prepared from 3β-acetoxy-5α-androstan-17-one. The new procedure represents the most efficient bufadienolide synthesis yet known.  相似文献

A simple method was developed for the preparation of acrylamide derivatives containing thioglycosides. The synthetic scheme involves the preparation of an O-acetylated 1-thiosugar via a pseudothiourea derivative, the controlled addition of the thiosugar to N,N′-bismethyleneacrylamide to form a monoaddition product in which one of the acrylamide groups remains unchanged, and finally de-O-acetylation. Similar reaction schemes using bisacrylamido derivatives of 1,2-diaminoethane and 1,6-diaminohexane lead to analogous compounds with longer aglycon chain lengths. The thioglycoside derivatives of acrylamide thus prepared could be copolymerized with acrylamide to form polymers or gels containing thioglycosides as ligands. These gels were successfully used as affinity materials for the purification of peanut lectin and in cell-adhesion studies.  相似文献

A method for the separation of benzene metabolites using reverse-phase high-pressure liquid chromatography is described. The antoxidant, ascorbic acid is added to an aqueous mixture of 1,2,4-benzenetriol, hydroquinone, catechol, and phenol, to prevent autooxidation. The eluting solvents are equilibrated with nitrogen, degassed, and maintained under a nitrogen atmosphere during the analysis. A highly resolved and reproducible profile of the metabolites is achieved under these conditions. This method should prove useful in a number of pharmacokinetic studies where the biotransformation of the parent compound to autooxidizable species such as polyphenols and quinones precludes analysis under aerobic conditions.  相似文献

The heterocyclic amino-compounds 11a, 13a, 13b, and 17 reacted with formaldehyde and p-thiocresol (14) in alcoholic solution to give the corresponding N-methylphenylthiomethyl derivatives (16, 15a, 15b, and 18a, respectively) in satisfactory to good yields. The reactions were catalyzed by acetic acid. 2-N-Methylguanosine (6a) was obtained in good yield by treatment of 15b with sodium borohydride followed by acidic hydrolysis, or alternatively by Raney nickel desulfurization of 15a followed by ammonolysis of the product. Sodium borohydride reduction of 18a gave 21 in good yield. 2-N,2-N-Dimethylguanosine (6b) was obtained from 19a in three steps.  相似文献

Glucocorticoids will enhance the growth of cultured human skin fibroblasts in serum-containing medium. In serum-free cultures hydrocortisone (5 X 10(-6) M) will enhance insulin stimulation of sugar transport and DNA synthesis (as measured by thymidine incorporation into trichloroacetic acid-precipitable material). The optimal concentration for the glucocorticoid effect on DNA synthesis was 5 X 10(-8) M for dexamethasone and 5 X 10(-7) M for hydrocortisone. In dexamethasone-treated cells, concentrations of insulin as low as 250 microU/ml (10 ng/ml) were effective in stimulating DNA synthesis. Further, hydrocortisone and dexamethasone (both at 5 X 10(-6) M) exhibited potentiating effects on insulin-stimulated sugar transport. These effects appeared to be mediated via inhibitory actions on the hexose transport system with the preservation of a functional insulin-receptor interaction resulting in insulin stimulation of deoxy-D-glucose transport at physiological insulin concentrations, 250 microU/ml (10 ng/ml). Hydrocortisone also enhanced specific [125I]insulin binding in these cells. The data indicate that the mechanism(s) of glucocorticoid enhancement of two actions of insulin may be different.  相似文献

An electromagnet was modified for measurements of the magnetic circular dichroism of samples held at cryogenic temperatures using a standard laboratory cryostat. The external dimensions of the cryostat are too great to permit its insertion in the air gap between the poles of the magnet without an unacceptable reduction in the strength of the magnetic field at the sample. This problem was overcome by designing new pole caps which become an integral part of the vacuum system of the cryostat. The ends of the new pole caps project into the body of the cryostat so that the gap between them is 1 in. or less, thus achieving a magnetic field exceeding one Tesla at the sample. No permanent alterations of the cryostat are required. The chief advantages of this design are economy and flexibility since a general purpose cryostat is used instead of a special unit designed to fit in the small space between the poles of an unmodified magnet. The cryostat used in this design cools the sample by conduction; thus the problem of optical distortions resulting from bubbling of liquid nitrogen or other cryogen is avoided and the temperature can be varied continuously using standard auxiliary equipment. Extra windows at 90° with respect to the optical beam permit inspection of the sample in situ and could be used for experiments such as fluorescence-detected magnetic circular dichroism which require optical access perpendicular to the direction of the magnetic field.  相似文献

A novel, automated method is described for the determination of Nτ-methylhistidine in human urine. The method uses a modification (H. Nakamura and J. J. Pisano (1976) Arch. Biochem. Biophys.177, 334–335) of the reaction of fluorescamine with amines, which renders it specific for certain imidazoles. Interference due to histidine and histamine is selectively removed by prior reaction with aldehydes. The fluorescence yield for Nτ-methylhistamine is 280, 440, 50, and 1.7, respectively. The concentrations of Nτ-methylhistidine in human urine as determined by this technique correlate well (r = 0.99) with those determined by ion-exchange chromatography. Furthermore, the technique is rapid (6–7 samples/h net throughout), is reproducible (coefficient of variation 1.8%), requires no prior treatment of the sample, and is implemented with widely available equipment.  相似文献

Phosphatidylcholine phosphatidohydrolase (EC 3.1.4.4, phospholipase D) catalyzes the hydrolysis of phosphatidylcholine to phosphatidic acid and choline. We have developed a spectrophotometric assay for phospholipase D using choline kinase, pyruvate kinase, and lactate dehydrogenase to couple the release of choline with the oxidation of NADH. The assay was linear both with time and with enzyme concentration. The assay should prove useful for continuous monitoring of enzyme activity, determination of initial rates of reaction, and detailed kinetic studies of phospholipase D. The method is limited to analysis of purified preparations of phospholipase D lacking competing activities to the coupled system.  相似文献

Studies were carried out on cultures of human skin fibroblasts to explore the effects of culture medium glucose levels on insulin binding and action. Cell cultures in 5.55 mm glucose-containing medium depleted their medium glucose within 3 days, and at that time exhibited elevated deoxy-d-glucose (2-DG) transport (84% greater than control cultures fed 22.2 mm glucose) and failure of insulin to stimulate 2-DG transport (an insulin:control transport ratio of 1.02). There was also a significant negative correlation between basal 2-DG transport and insulin binding (r = ?0.621; n = 29; P < 0.01), while insulin binding exhibited a significant positive correlation with insulin action (r = 0.816; n = 12; P < 0.01). Glucose starvation of cultures for 18 h resulted in several changes: (i) a 49% decrease in specific 125I-insulin binding due to a reduction in binding capacity; (ii) elevated basal 2-DG transport; and (iii) an absence of insulin stimulation of 2-DG transport. Exposure to increasing concentrations of glucose for 18 h led to a glucose concentration-dependent increase in specific insulin binding. Additionally, the various changes in the glucose-starved group were reversed after as little as 6 h of glucose refeeding. The results indicate that basal sugar transport, and insulin binding and action can be regulated by the amount of glucose in the medium.  相似文献

A procedure utilizing a reverse-phase semipreparative high-performance liquid chromatography column and a binary solvent system consisting of trifluoroacetic acid and 1-propanol has been developed for the semipreparative scale purification and analytical identification of four newly synthesized analogs of methotrexate. The methotrexate analogs containing a lysine or an ornithine residue in place of a terminal glutamate residue together with their respective dansyl derivatives were purified in milligram quantities by the procedures described.  相似文献

A method is described for the isolation of basement membranes from rabbit renal cortex in which the detergent N-lauroyl sarcosine is used as the disruptive agent. The isolated membranes have been compared with membranes prepared using ultrasonication and they were comparable both in terms of purity and gross chemical composition. Glomerular and tubular basement membranes were isolated by first separating glomeruli from tubules by density gradient centrifugation followed by detergent treatment of the separated tissues.The detergent method has the advantage that the basement membranes retained their native structure to a large degree, whereas sonicated membranes were severely fragmented. Collagen fibres were a significant contaminant in both preparations and were revealed more clearly by negative staining than by examination of thin sections. Studies with the detergent-treated membrane revealed that a few proteins, which seemed to be membrane components, were extracted with 1 M NaCl and that these proteins were lost from the basement membranes during sonication used in the conventional isolation procedure.  相似文献

A new method is described for the determination of the cytolytic activity of extremely low levels of stable as well as very labile cytotoxins. The method involves the application of the cytotoxin to a column of immunobilized erythrocytes or other suitable cells and a continuous monitoring of the column eluate for the presence of hemoglobin or other cell constituents. The cytotoxic activity of horseradish peroxidase at concentrations as low as 10?12, m can be measured with this technique. The column hemolytic assay is compared with a static (batch) hemolytic assay with respect to sensitivity and reproducibility. Furthermore, a method is described to determine the true rates of lysis, i.e., the number of cells lysed per minute.  相似文献

Trifluoroacetolysis of d-glucuronic acid and methyl α-d-glucopyranosiduronic acid resulted in an initial phase of degradation followed by stabilisation of the compounds as their 6,3-lactones. The methyl ester of methyl 4-O-methyl-α-d-glucopyranosiduronic acid was largely stable towards trifluoroacetolysis. Aldonic acids substituted at O-3 or O-6 were stable towards trifluoroacetolysis because of the formation of γ-lactones. Aldonic acids substituted at O-4, and incapable of forming γ-lactones, were converted into the trifluoroacetylated enol of 3-deoxy-2-hexulosonic acid. Treatment of the 3-deoxy-2-hexulosonic acid with mild base eliminated the substituent at O-4.  相似文献

In the seeds of Tropaeolum and Tamarindus, which have been found to have the highest content of amyloids, these polysaccharides are associated with sucrose and O-d-galactosylsucroses of the raffinose-stachyose series. Balsamine seeds have a low content of amyloid and do not contain the aforementioned galactosides, which are otherwise widely distributed in plant seeds. They contain sucrose and a mono-O-D-galactosylsucrose that has been crystallized and identified as planteose. Planteose has previously been isolated from some Sympetalous plants. As far as we know, this is the first report of its occurrence in an Archichlamydeaeous plant seed.  相似文献

An affinity-label chelate for the enzyme trypsin was synthesized by a novel synthetic technique which takes advantage of the presence of a dangling carboxylate arm in the [Co(EDTA)Cl]2- complex anion. The dangling carboxylate group was coupled to the amino group of p-aminobenzamidine, an effective inhibitor of trypsin activity, via the carbodiimmide reaction to produce a trypsin affinity label at one end and a strong EDTA-like chelating agent at the other, coupled through an amide bond. The cobalt ion can be removed if desired by reduction with Fe2+ + ascorbate, and alternate metal ions inserted in its place. The reaction is general, and affinity labels which contain amino groups can be easily coupled via this procedure, allowing the introduction of a paramagnetic or fluorescent probe into a protein or nucleotide system. The same method has been used to prepare a highly effective chelating gel which is capable of removing calcium and lanthanide ions from the binding protein parvalbumin.  相似文献
