The chemical modifications induced in polycrystalline cycloamylose hydrates during γ-irradiation have been investigated by using g.l.c-m.s. to analyse the monosaccharide mixtures formed on hydrolysis. Unchanged substrate and material retaining the original cyclic structure were removed by precipitation prior to hydrolysis, and the products therefore reflect the effect of the radical-induced opening of the cycloamylose ring structure. The following products were identified: glucose and glucono-1, 5-lactone (1), 4-deoxy-xylo-hexose (2), arabinose (3), ribose (4), 2-deoxy-erythro-pentose (5), 3-deoxy-erythro-hexos-4-ulose (6), xylo-hexos-5-ulose (7), 6-deoxy-xylo-hexos-5-ulose (8), 5-deoxy-xylo-hexodialdose (9), 2,6-dideoxyhexos-5-ulose (10), xylose (11), 5-deoxypentose (12), 3-deoxypentulose (13), erythrose (14), and threose (15). Products 1-9 appear to be terminals of the “anhydroglucose” chain. Established free-radical reactions, typical for carbohydrates. are invoked to account for these products.  相似文献

Methyl 2,3-dideoxy-DL-pent-2-enopyranosid-4-ulose (2) and 1-O-benzoyl-2,3- dideoxy-DL-pent-2-enopyranos-4-ulose (3), obtained from furfuryl alcohol, gave methyl β-DL-erythro-pentopyranosid-4-ulose (6) and 1-O-benzoyl-β-DL-erythro-pentopyranos-4-ulose (7), respectively, on cis-hydroxylation with silver chlorate- osmium tetroxide. Reduction of the isopropylidene derivatives (8 and 9) of 6 and 7 with lithium aluminium hydride and sodium borohydride, respectively, afforded DL-ribose derivatives.  相似文献

The following primary sulphonates have been converted into the corresponding deoxyfluoro derivatives by reaction with potassium fluoride in ethylene glycol:1,2:3,4-di-O-isopropylidene-6-O-tosyl α-D-galactopyranose (1), methyl 2,3-O2-isopropyliden-5-O-tosyl-α,β-D-ribofuranoside (2), 1,2:3,4-di-O-methylene-6-O-tosyl-α-D-glucofuranose (3), 3,5-di-O-benzylidene-1,2-O-isopropylidene-6-O-tosyl-α-D-glucofuranose (4), and 1,2:3,5-di-O-isopropylidene-6-O-tosyl-α-D-glucofuranose (5). The yields were generally poor; in the reaction of 1, a major by-product was 6-O-(2-hydroxyethyl)-1,2:3,4-di-O-isopropylidene-α-D-galactopyranose (11). The reaction of the primary hydroxyl precursor of each of the above tosylates with N2-(2-chloro- 1,1,2-trifluoroethyl)-N,N-diethylamine generally yielded the O-chlorofluoroacetyl derivative; however, 1,2:3,5-di-O-methylene-α-D-glucofuranose (12) was converted into the 6-deoxy-6-fluoro derivative (8). The 19F resonances of compounds containing the CH2F moiety fall between φC +213 and φC +235 p.p.m. The differences between the vicinal19F-1H couplings of compounds having the D-gluco and D-galacto configurations clearly reflect the influence of the C-4O-4 substitutents on the populations of the C-5C-6 rotamers. A novel type of noise-modulated, heteronuclear decoupling experiment is described.  相似文献

Methyl 2,3-O-benzylidene-6-deoxy-α-L-mannopyranoside (2) reacted with butyllithium to give a mixture of 1,5-anhydro-3-C-butyl-1,2,6-trideoxy-L-ribo-hex-1-enitol (3) and its L-arabino analogue (4), together with methyl 2,3,6-trideoxy-α-L-erythro-hex-2-enopyranoside (5). In contrast, the 4-O-methyl analogue (8) of 2 was converted by butyllithium into methyl 2,6-dideoxy-4-O-methyl-α-L-erythro-hexo-pyranosid-3-ulose (9), which was further characterized as its oxime 10. The 4-O-benzyl analogue of 8, obtained as two separate diastereoisomers (6 and 7) differing in configuration at C-2 of the dioxolane ring, gave a complex mixture of products on treatment with butyllithium.  相似文献

Reduction of 1,6-anhydro-3,4-dideoxy-β-D-glycero-hex-3-enopyranos-2-ulose (levoglucosenone) with lithium aluminium hydride afforded principally 1,6-anhydro-3,4-dideoxy-β-D-threo-hex-3-enopyranose (3), which was converted into 3,4-dihydro-2(S)-hydroxymethyl-2H-pyran (8) following acid-catalysed methanolysis and reductive rearrangement of the resulting α-glycoside 4 with lithium aluminium hydride. 1,6-Anhydro-3,4-dideoxy-2-O-toluene-p-sulphonyl-β-D-threo-hexopyranose, prepared from 3, reacted slowly with sodium azide in hot dimethyl sulphoxide to give 1,6-anhydro-2-azido-2,3,4-trideoxy-β-D-erythro-hexopyranose, which was transformed into a mixture of methyl 2-acetamido-6-O-acetyl-2,3,4-trideoxy-α-D-erythro-hexopyranoside (10) and the corresponding β anomer following acid-catalysed methanolysis, catalytic reduction, and acetylation. Acid treatment of methyl 4,6-O-benzylidene-3-deoxy-α-D-erythro-hexopyranosid-2-ulose yielded the enone 15, which was readily transformed into methyl 6-O-acetyl-3,4-dideoxy-α-D-glycero-hexopyranosid-2-ulose (19). Procedures for the conversions of DL-8, 10, and 19 into methyl 2,6-diacetamido-2,3,4,6-tetradeoxy-α-D-erythro-hexopyranoside (methyl N,N′-di-acetyl-α-purpurosaminide C) have already been described.  相似文献

Methyl 4,6-O-benzylidene-2-deoxy-α-d-erythro-hexopyranosid-3-ulose reacted with potassium cyanide under equilibrating conditions to give, initially, methyl 4,6-O-benzylidene-3-C-cyano-2-deoxy-α-d-ribo-hexopyranoside (7), which, because it reverted slowly to the thermodynamically stable d-arabino isomer, could be crystallised directly from the reaction mixture. The mesylate derived from the kinetic product 7 could be converted by published procedures into methyl 3-acetamido-2,3,6-trideoxy-3-C-methyl-α-d-arabino-hexopyranoside, which was transformed into methyl N-acetyl-α-d-vancosaminide on inversion of the configuration at C-4. A related approach employing methyl 2,6-dideoxy-4-O-methoxymethyl-α-l-erythro-hexopyranosid-3-ulose gave the kinetic cyanohydrin and thence, via the spiro-aziridine 27, methyl 3-acetamido-2,3,6-trideoxy-3-C-methyl-α-l-arabino-hexopyranoside, a known precursor of methyl N-acetyl-α-l-vancosaminide.  相似文献

The mechanism of the radiation-induced dephosphorylation reaction was investigated by studying the γ-radiolysis of 10mM solutions of D-glucopyranosyl phosphate, D-glucose 6-phosphate (Glc-6-P), and d-ribose 5-phosphate (Rib-5-P). Dephosphorylation occurred with OH-radical participation, since G(H3PO4) values for nitrous oxide-saturated solutions were 4.1, 1.7, and 2.3, and for nitrogen-saturated solutions 2.6, 1.1, and 1.6, respectively. The formation of phosphate-free compounds accompanied the release of inorganic phosphate. The main, neutral products of the radiolysis of Glc-6-P were 6-deoxyhexos-5-ulose (G = 0.2) and D-gluco-hexodialdose (G = 0.3). Irradiation of Rib-5-P gave ribo-pentodialdose and 5-deoxypentos-4-ulose as the main, neutral products. A scheme for the dephosphorylation process is proposed.  相似文献

d-erythro-2,3-Hexodiulosono-1,4-lactone 2-arylhydrazones (2) were prepared by condensation of dehydro-d-arabino-ascorbic acid with the desired arylhydrazine. Reaction of 2 with hydroxylamine gave the 2-arylhydrazone 3-oximes (3). On boiling with acetic anhydride, 3 gave 2-aryl-4-(2,3-di-O-acetyl-d-erythro-glycerol-1-yl)-1,2,3-triazole-5-carboxylic acid 5,11-lactone (5), whereas the unacetylated triazole derivatives were obtained upon reaction of 3 with bromine in water. On treatment of 5 with hydrazine hydrate, 2-aryl-4-(d-erythro-glycerol-1-yl)-1,2,3-triazole-5-carboxylic acid 5-hydrazides (6) were obtained. Acetylation of 6 gave the hexaacetyl derivatives. Similarly, treatment of 5 with liquid ammonia gave the triazolecarboxamides (12). Vigorous acetylation of 12 with boiling acetic anhydride gave tetraacetates, whereas acetylation with acetic anhydride-pyridine gave triacetates. Periodate oxidation of 6 gave the 2-aryl-4-formyl-1,2,3-triazole-5-carboxylic acid 5-hydrazides (8), and, on reduction, 8 gave the 2-aryl-4-(hydroxymethyl)-1,2,3-triazole-5-carboxylic acid 5-hydrazides, characterized as acetates. Similarly, periodate oxidation of 12 gave the triazolealdehyde (15), and reduction of 15 gave the hydroxymethyl derivatives (16). Acetylation of 16 gave the mono- and di-acetates, and, on reaction with o-phenylenediamine, 15 afforded the triazoleimidazole. Controlled reaction of 3 with sodium hydroxide, followed by neutralization, gave 3-(d-erythro-glycerol-1-yl)-4,5-isoxazolinedione 4-arylhydrazones. Reaction of 3 with HBr-HOAc gave 5-O-acetyl-6-bromo-6-deoxy-d-erythro-2,3-hexodiulosono-1,4-lactone 2-arylhydrazone 3-oximes (21). Compounds 21 were converted into 4-(2-O-acetyl-3-bromo-3-deoxy-d-erythro-glycerol-1-yl)-2-aryl-1,2,3-triazole-5-carboxylic acid 5,11-lactone on treatment with acetic anhydride.  相似文献

Acetonation of dimeric 1,6-anhydro-β-D-arabino-hexopyranos-3-ulose yields, besides a monomeric di-O-isopropylidene compound, the dimer 2, which crystallizes in space group P212121 with a  1.3680 (9), b  1.0686 (7), and c  1.0319 (7) nm, Z  4. The crystal and molecular structure of 2 have been determined by X-ray analysis with direct methods and was refined to a final Rw of 5.55% for 2468 reflections. Compound 2 has not the same dimeric structure as the parent compound with a central 1,4-dioxane ring, but contains instead a central 1,3-dioxolane ring. The pyranose ring bearing the isopropylidene group adopts an almost ideal sofa conformation, with a nearly planar arrangement of C-1, C-2, C-3, C-4, and C-5. By analogy, it was concluded that the dimeric mono-O-isopropylidene derivative 7 of 1,6-anhydro-β-D-xylo-hexopyranos-3-ulose has the same asymmetric structure. The 360-MHz 1H-n.m.r. spectra of both compounds are in full agreement with the proposed structures.  相似文献

3-Deoxy-4-O-β-D-galactopyranosyl-D-erythro-hexos-2-ulose (1) was obtained from lactose by reaction with benzoylhydrazine in the presence of a slightly acidic solution of p-toluidine, followed by hydrazinolysis of the product, 3-deoxy-4-O-β-D-galactopyranosyl-D-erythro-hexos-2-ulose bis(benzoylhydrazone) (3), with benzaldehyde. A variety of derivatives of 1 and 3 was prepared. Lactose aroylhydrazones were also prepared. Quantitative determination of the oxidant during the periodate oxidation of 3 was studied. Periodate oxidation of monosaccharide arylhydrazones gave glyoxal mono(arylhydrazones) which afforded the corresponding, mixed bis(hydrazones).  相似文献

Methyl 2,3-anhydro-4,6-O-benzylidene-3-C-nitro-β-d-allopyranoside (1), as well as its β-d-manno (2) and α- d-manno (3) isomers, reacted with dimethylamine to give the same, crystalline 3-(dimethylamino) adduct (4) of 1,5-anhydro-4,6-O-benzylidene-2-deoxy-2-(dimethylamino)-d-erythro-hex-1-en-3-ulose (5). The enulose 5 was obtained from 4 by the action of silica gel. Similarly, the β-d-gulo (6) and α-d-talo (7) stereoisomers of 13 afforded a 3-(dimethylamino) adduct (8) of the d-threo isomer (9) of 5. Reaction of dimethylamine with 5,6-anhydro-1,2-O-isopropylidene-6-C-nitro-α-d-glucofuranose (10) yielded a mixture of two diastereoisomeric (possibly anometic at C-6) 5-deoxy-5-(dimethylamino)-1,2-O-isopropylideric-α-d-hexodialdo-1,4:6,3-difuranoses (11). The β-glycoside 1 and the α-glycoside 3 reacted with methylmagnesium iodide to produce methyl 4,6-O-benzylidene-3-deoxy-3-C-methyl-3-(N-hydroxy-N-methylamino)-β- and -α-d-hexopyranosides (12) and (13), respectively; both products had the 1,2-trans configuration, but their configurations at the quaternary center C-3 have not been determined.  相似文献

Isolevoglucosenone (1,6-anhydro-2,3-dideoxy-β-d-glycero-hex-2-enopyranos-4-ulose, 3) has been synthesized from levoglucosenone (2) in six steps. Thus, 1,6-anhydro-4-O-benzyl-3-deoxy-β-d-erythro-hexopyranos-2-ulose, obtained by Michael addition of benzyl alcohol to 2, was reduced with sodium borohydride to yield a separable mixture of the C-2 epimeric alcohols 1,6-anhydro-4-O-benzyl-3-deoxy-β-d-arabino- and -ribo-hexopyranose, both of which displayed intramolecular hydrogen-bonding. Acetylation, hydrogenolytic debenzylation, and pyridinium chlorochromate oxidation then led to the 2-O-acetyl-1,6-anhydro-hexos-4-uloses, from which 3 was obtained by tetraethylammonium acetate-catalyzed β-elimination of acetic acid. On sealed-tube thermolysis in the range of 210–260°, 3 generated 3-oxidopyrylium by loss of formaldehyde; this ylide was efficiently trapped by unreacted 3, to yield the [4π + 2π]-1,3-dipolar cycloadducts 14 and 15. The structure of 14 was fully elucidated by an X-ray crystallographic study. Neither 3 was, nor the adducts 14 and 15 were, detected among the products from acid-catalyzed pyrolysis of cellulose.  相似文献

1,4:3,6-dianhydro-α-d-glucopyranose (1) was formed, together with 1,6-anhydro-3,4-dideoxy-β-d-glycero-hex-3-enopyranos-2-ulose (levoglycosenone, 2) and levoglucosan (4), on acid-catalyzed pyrolysis of d-glucose, amylopectin, and cellulose. Pyrolysis of 1 in the presence of acid provided significant quantities of 2, indicating that 1 can act as a pyrolytic precursor of 2. A pyrolysis product from cellulose previously considered to be 1,6-anhydro-3-deoxy-β-d-erythro-hex-3-enopyranose (12) was shown to be dianhydride 1.  相似文献

Addition of ethyl isocyanoacetate in strongly basic medium to the glycosuloses 1,2:5,6-di-O-isopropylidene-α-d-ribo-hexofuranos-3-ulose (1) and 1,2-O-isopropylidene-5-O-trityl-d-erythro-pentos-3-ulose (2) gave the unsaturated derivatives (E)- and (Z)-3-deoxy-3-C-ethoxycarbonyl(formylamino)methylene-1,2:5,6-di-O-isopropylidene-α-d-glucofuranose (3 and 4), and (E)-3-deoxy-3-C-ethoxycarbonyl(formylamino)methylene-1,2-O-isopropylidene-5-O-trityl-α-d-ribofuranose (5). In weakly basic medium, ethyl isocyanoacetate and 1 gave 3-C-ethoxycarbonyl(formylamino)methyl-1,2:5,6-di-O-isopropylidene-α-d-allofuranose (12) in good yield. The oxidation of 3 and 4 with osmium tetraoxide to 3-C-ethoxalyl-1,2:5,6-di-O-isopropylidene-α-d-glucofuranose (17), and its subsequent reduction to 3-C-(R)-1′,2′-dihydroxyethyl-1,2:5,6-di-O-isopropylidene-α-d-glucofuranose (18) and its (S) epimer (19) and to 3-C-(R)-ethoxycarbonyl(hydroxy)methyl-1,2:5,6-di-O-isopropylidene-α-d-glucofuranose (21) and its (S) epimer (22) are described. Hydride reductions of 12 yielded the corresponding 3-C-(1-formylamino-2-hydroxyethyl), 3-C-(2-hydroxy-1-methylaminoethyl), and 3-C-(R)-ethoxycarbonyl(methylamino)methyl derivatives (13, 14 and 16). Catalytic reduction of 3 and 4 yielded the 3-deoxy-3-C-(R)-ethoxycarbonyl-(formylamino)methyl derivative 6 and its 3-C-(S) epimer. Further reduction of 6 gave 3-deoxy-3-C-(R)-(1-formylamino-2-hydroxyethyl)-1,2:5,6-di-O-isopropylidene-α-d-allofuranose (23) which was deformylated with hydrazine acetate to 3-C-(R)-(1-amino-2-hydroxyethyl)-3-deoxy-1,2:5,6-di-O-isopropylidene-α-d-allofuranose (24). The reaction between 2-amino-2-deoxyaldoses and β-dicarbonyl compounds yields polyhydroxyalkylpyrroles. Thus, 6,6-dimethyl-2-(D-galacto-pentitol-1-yl)-4,5,6,7-tetrahydroindol-4-one (4a), 6,6-dimethyl-2-(D-gluco-pentitol-1-yl)-4,5,6,7-tetrahydroindol-4-one (4b), and 6,6-dimethyl-2-(D-manno-pentitol-1-yl)-4,5,6,7-tetrahydroindol-4-one (4c) have been obtained from 5,5-dimethylcyclohexane-1,3-dione (2) and 2-amino-2-deoxyheptoses having D-glycero-L-gluco (1a), D-glycero-D-ido (1b), and D-glycero-D-talo (1c) configurations, respectively. 2-Amino-2-deoxy-D-glycero-L-manno-heptose (1d), the epimer of 1a, also reacts with 2, to yield 4a. In a similar way, 1a, 1b, and 1c react with cyclohexane-1,3-dione (3), to give 2-(D-galacto-pentitol-1-yl)-4,5,6,7-tetrahydroindol-4-one (5a), 2-D-gluco-pentitol-1-yl)-4,5,6,7-tetrahydroindol-4-one (5b), and 2-(D-manno-pentitol-1-yl)-4,5,6,7-tetrahydroindol-4-one (5c), respectively.  相似文献

Amylose (1) was tritylated at O-6, the ether p-toluenesulfonylated at O-2 and O-3, and the product (3) treated with sodium iodide and zinc dust in N,N-dimethyl-formamide, to give 2,3-dideoxy-6-O-trityl-α-D-erythro-hex-2-enopyranoglycan (4). This 2,3-unsaturated polysaccharide could be converted into a 2,3-dibromo derivative (5), and hydrogenated with concomitant detritylation to the saturated analogue (6), and, on treatment with aqueous acetic acid, it gave 2-(D-glycero-1,2-dihydroxyethyl)-furan (8). The 2,3-bis(p-toluenesulfonate) (10) of β-D-xylan (9) was similarly converted into the 2,3-unsaturated polysaccharide, 2,3-dideoxy-β-D-glycero-pent-2-enopyranoglycan (11), which, with aqueous acetic acid, gave 2-(hydroxymethyl)furan (12a).  相似文献

Methyl 3-azido-2-O-benzoyl-3,4-dideoxy-β-dl-erythro-pentopyranoside (6) was synthesized through two routes in five steps from methyl 2,3-anhydro-4-deoxy-β-dl-erythro-pentopyranoside (1). The first route proceeded via selective azide displacement of the 3-tosyloxy group of methyl 4-deoxy-2,3-di-O-tosyl-α-dl-threo-pentopyranoside, followed by detosylation and benzoylation. The second route consisted, with a better overall yield, in the azide displacement of the mesyloxy group of methyl O-benzoyl-4-deoxy-3-O-methylsulfonyl-α-dl-threo-pentopyranoside (10), obtained by benzylate opening of 1, followed by benzoylation, debenzylation, and mesylation. Compound 6 was transformed into its glycosyl chloride, further treated by 6-chloropurine to give the nucleoside 9-(3-azido-2-O-benzoyl-3,4-dideoxy-β-dl-erythro-pentopyranosyl)-6-chloropurine (13). When treated with propanolic ammonia, 13 yielded 9-(3-azido-3,4-dideoxy-β-dl-erythro-pentopyranosyl)adenine.  相似文献

Methyl 4-O-benzoyl-6-bromo-6-deoxy-α-d-glucopyranoside, obtainable from methyl 4,6-O-benzylidene-α-d-glucopyranoside (1), was converted into the 2,3-unsaturated 4-benzoate (3) by application of the triiodoimidazole method. Debenzoylation of 3, followed by acetylation, afforded crystalline methyl 4-O-acetyl-6-bromo-2,3,6-trideoxy-α-d-erythro-hex-2-enopyranoside (5). Treatment of 5 with benzylmethylamine under conditions of palladium-catalyzed, allylic substitution gave a separable mixture of the corresponding 4-(N-benzyl)methylamino-6-bromo-2-enoside (37%) and the 4,6-di-[(N-benzyl)methylamino]-2-enoside (55%). Debromination of 5 with lithium triethylborohydride, proceeding with simultaneous deacetylation, readily yielded methyl 2,3,6-trideoxy-α-d-erythro-hex-2-enopyranoside (8). The 4-acetate of 8 (obtained by reacetylation), and also its 4-benzoate (prepared by a different synthetic route), furnished high yields (~80%) of methyl 4-[(N-benzyl)-methylamino]-2,3,4,6-tetradeoxy-α-d-erythro-hex-2-enopyranoside (13) upon palladium-catalyzed animation with benzylmethylamine. Catalytic hydrogenation of 13 effected saturation of the alkenic double bond and removal of the N-benzyl group, to afford methyl 2,3,4,6-tetradeoxy-4-methylamino-α-d-erythro-hexopyranoside, which was subsequently N-methylated with formaldehyde and sodium borohydride, to give its N,N-dimethyl analog, methyl α-d-forosaminide (15). The overall yield of 15 from 1 was 24%. Hydrolysis of 15 to the free sugar has been described previously.  相似文献

The syntheses of three classes of C-3 perfluoroalkyl substituted steroids are described. They are the 3β-hydroxy-3α-perfluoroalkylandrost-4-en-17-ones (5a-c), 3-perfluoroalkylandrosta-3,5-dien-3-ones (8a-c) and 3β-hydroxy-3α-perfluoroalkylandrost-5-en-17-ones (12a-c). Addition of a series of perfluroalkylorganometallic reagents (RFLi; RF = C2F5, C3F7, or C4F9) to the 3 position of silylated testosterone 2b afforded Δ4 perfluoroalkyl carbinols 3. In Scheme 1, deprotection with HF and oxidation at the C-17 carbon with PCC produced Δ4 ketones 5. In Scheme 2 dehydration of 3 with 1,2-phenylenephosphorochloridite and iodine afforded Δ3,5 dienes 6 which were deprotected and oxidized as above to the C-17 ketones 8. In Scheme 3 isomerization of the double bond of 3 from the C-4 to the C-5 position using the allylic halogenation followed by treatment with lithium aluminum hydride led to the synthesis of the double bond isomer series 12. A new method for dehydration was developed. On average and within experimental error, 3β-hydroxy-3α-perfluoroalkylandrost-5-en-17 ones (12a-c) were better than the 3-perfluoroalkylandrosta-3,5-dien-17-ones (8a-c) and 3β-hydroxy-3α-perfluoroalkylandrost-4-en-17-ones (5a-c) at inhibiting glucose-6-phosphate dehydrogenase.  相似文献
