Background

Lymphedema management programs have been shown to decrease episodes of adenolymphangitis (ADLA), but the impact on lymphedema progression and of program compliance have not been thoroughly explored. Our objectives were to determine the rate of ADLA episodes and lymphedema progression over time for patients enrolled in a community-based lymphedema management program. We explored the association between program compliance and ADLA episodes as well as lymphedema progression.

Methodology/Principal Findings

A lymphedema management program was implemented in Odisha State, India from 2007–2010 by the non-governmental organization, Church''s Auxiliary for Social Action, in consultation with the Centers for Disease Control and Prevention. A cohort of patients was followed over 24 months. The crude 30-day rate of ADLA episodes decreased from 0.35 episodes per person-month at baseline to 0.23 at 24 months. Over the study period, the percentage of patients who progressed to more severe lymphedema decreased (P-value  = 0.0004), while those whose lymphedema regressed increased over time (P-value<0.0001). Overall compliance to lymphedema management, lagged one time point, appeared to have little to no association with the frequency of ADLA episodes among those without entry lesions (RR = 0.87 (0.69, 1.10)) and was associated with an increased rate (RR = 1.44 (1.11, 1.86)) among those with entry lesions. Lagging compliance two time points, it was associated with a decrease in the rate of ADLA episodes among those with entry lesions (RR = 0.77 (95% CI: 0.59, 0.99)) and was somewhat associated among those without entry lesions (RR = 0.83 (95% CI: 0.64, 1.06)). Compliance to soap was associated with a decreased rate of ADLA episodes among those without inter-digital entry lesions.

Conclusions/Significance

These results indicate that a community-based lymphedema management program is beneficial for lymphedema patients for both ADLA episodes and lymphedema. It is one of the first studies to demonstrate an association between program compliance and rate of ADLA episodes.  相似文献

A horizon with large, massive corals in growth position was discovered in the Paleocene, probably upper Danian, part of the Maastrichtian–Paleocene Lefipán Formation of Chubut (Patagonia, Argentina). All corals belong to one species, the cosmopolitan Haimesiastraea conferta Vaughan, which survived the end-Cretaceous mass extinction. The occurrence of massive corals at this site is exceptional both because of the siliciclastic depositional regime and because of the high palaeolatitude setting. An unusual autecology of this coral and strongly reduced sedimentation rates, were probably the prerequisites for coral growth, but a link to palaeoclimate is less likely.  相似文献

Nutritional mutants (co-mutants) were scored among nitrosoguanidine-induced revertants of four mutations in Streptomyces coelicolor A3 (2). All co-mutations were due to mutations in genes linked to the revertant locus. The co-mutant loci were located in a region of about 20 map units around the revertant locus (co-mutation region). Revertants for different loci showed co-mutation patterns different from each otehr and from that of random nitroso-guanidine-induced forward mutations. Mutations appeared to be completely abolished outside the co-mutation region (mutation restriction).  相似文献

A case of massive trichuriasis in a 37-year-old female from a rural locality of the Metropolitan Region of Chile, with antecedents of alcoholism, chronic hepatic damage and portal cavernomatosis, is presented. Since 12 year ago she has had geophagia. In the last six months she has frequently presented liquid diarrhea, colic abdominal pains, tenesmus and sensation of abdominal distention. Clinical and laboratory tests confirmed her hepatic affection associated with a celiac disease with anemia and hypereosinophilia. Within a week diarrhea became worse and dysentery appeared. A colonoscopy revealed an impressive and massive trichuriasis. The patient was successfully treated with two cures of 200 mg tablets of mebendazole twice daily for three days with a week interval. After the first cure she evacuated a big amount of Tricuris trichiura, fecal evacuations became normal, geophagia disappeared and recovered 4 kg of body weight.  相似文献

We describe a massive polymorphism for shell colour and pattern in Donacilla cornea , a shallow-burrowing bivalve which inhabits the mid-littoral of sandy and stony beaches in southern and western Europe. Of the four sites studied, the one where the substrate was a mixture of small stones and sand contained the highest densities of Donacilla. At this site the densities of stones and animals and the frequencies of shell and stone colour phenotypes are both highly correlated. Although the four populations sampled comprise a similar range of phenotypes, the frequencies of most morphs and population diversity indices differ significantly between sites. At two sites the similar range of colour and pattern variation between shells and substrate suggests selection for crypsis and masquerade. On the other hand, at two other sites, one or two common phenotypes (and a number of rare ones at all sites) fail to mimic any aspect of the background. We therefore suggest that if frequency-dependent selection by predators is responsible for the maintenance of polymorphism in Donacilla , then the mechanisms involved include crypsis and masquerade in some cases and selective advantages from being different in others.  相似文献

Akinetes are dormancy cells commonly found among filamentous cyanobacteria, many of which are toxic and/or nuisance, bloom-forming species. Development of akinetes from vegetative cells is a process that involves morphological and biochemical modifications. Here, we applied a single-cell approach to quantify genome and ribosome content of akinetes and vegetative cells in Aphanizomenon ovalisporum (Cyanobacteria). Vegetative cells of A. ovalisporum were naturally polyploid and contained, on average, eight genome copies per cell. However, the chromosomal content of akinetes increased up to 450 copies, with an average value of 119 genome copies per akinete, 15-fold higher than that in vegetative cells. On the basis of fluorescence in situ hybridization, with a probe targeting 16S rRNA, and detection with confocal laser scanning microscopy, we conclude that ribosomes accumulated in akinetes to a higher level than that found in vegetative cells. We further present evidence that this massive accumulation of nucleic acids in akinetes is likely supported by phosphate supplied from inorganic polyphosphate bodies that were abundantly present in vegetative cells, but notably absent from akinetes. These results are interpreted in the context of cellular investments for proliferation following a long-term dormancy, as the high nucleic acid content would provide the basis for extended survival, rapid resumption of metabolic activity and cell division upon germination.  相似文献

Egg yolk phosphatidylcholine (PC) liposomes were prepared by ultrasonic irradiation. At least 25 mol% of coenzyme Q-10 (CoQ-10) can be incorporated nonstoichiometrically into PC liposomes. Electron microscopy showed no visible influence of CoQ-10 on the membrane structure. Nuclear magnetic resonance spectra of sonicated PC liposomes containing CoQ-10 showed two peaks (3.82 and 3.98 ppm) due to CoQ-10 methoxyl protons and a 'high-field component' (1.52 and 1.58 ppm). The areas of these peaks were inversely related and influenced by the time of ultrasonic irradiation. After short sonication the low-field positions (3.98/1.58 ppm) are favoured, after long sonication the high-field positions (3.82/1.52 ppm). No gradual shift of the two peaks is observed. The 'critical' liposome diameter was found to be between 500 to 700 A. Lanthanide induced pseudocontact shift on CoQ-10 resonances ('high-field component' and methoxyl) does not lead to a split of the peaks and the difference between them remains constant. It is concluded that CoQ-10 is incorporated into the membrane core, beyond C-2 of the PC acyl chains, with two bilayer curvature-dependent resonance positions.  相似文献

Aptamers are DNA or RNA single-stranded molecules that bind specifically to target molecules with high affinity. Function of nucleic acid aptamers is based on organized tertiary structure of them that is related to primary sequence, length of nucleic acid molecule, and environmental conditions. Herein, a localized surface plasmon resonance (LSPR) nanobioprobe has been developed based on specific aptamer-conjugated gold nanoparticles for rapid detection of methamphetamine. Detection of methamphetamine was studied via monitoring the gold nanoparticles (GNPs) LSPR band alterations in the presence of different concentrations. The covalent conjugation has been confirmed with FT-IR spectroscopy, and size alterations of gold nanoparticles before and after the conjugation state were monitored using dynamic light scattering (DLS) technique. The results show high affinity of aptamer to methamphetamine. Moreover, the results show conjugated aptamer with GNP in different concentrations of methamphetamine that contribute to color changes that is visible with unaided eye. Also, 14 nm LSPR shift was seen after conjugation of aptamer with GNP. Nanoparticle diameter after conjugation with aptamer was increased from 30 to 91 nm and decreased after incubation with methamphetamine (due to folding) from 91 to 84 nm. Detection limit of this designed nanoprobe is 500 nM. Plasmonic nanoparticle-based nanobioprobe is a new field for development of sensitive detection systems.

In this paper, we investigate the ability of the gold nanorods (GNRs) to detect some proteins and demonstrate their potential to be used as plasmonic nanobiosensors. The GNRs were synthesized by a two-step seed-mediated growth procedure at room temperature. Firstly, a seed solution of gold nanoparticles was synthesized in the presence of cetyltrimethylammonium bromide surfactant and, subsequently, incorporated with appropriate amount of silver nitrate and tetrachloroauric acid solutions to grow GNRs with average length of 50 nm and diameter of 14 nm. We study the interaction of GNRs with proteins whose molecular weight varies from 6.5 up to 75 kDa. We investigate the resulting solutions by means of UV–vis absorption spectroscopy to determine the effect of the proteins characteristics on the shift of the localized surface plasmon resonance (LSPR). We show that for the case when proteins are in large excess compared to the GNRs concentration, whatever the protein is, the LSPR shift is constant and does not depend on the protein molecular weight. Moreover, we have been able to demonstrate that the sensitivity of such LSPR sensor is around 10–9 M/nm on a concentration range from 10–10 to 10–8 M. Some comparison with finite-difference time-domain simulations have also shown that the number of proteins adsorbed at the GNRs surface is around 40.  相似文献

Variation in the vertebrate major histocompatibility complex (MHC) genes is crucial for fighting pathogen assault. Because new alleles confer a selective advantage, MHC should readily introgress between species, even under limited hybridization. Using replicated transects through two hybrid zones between strongly reproductively isolated European newts, Lissotriton montandoni and L. vulgaris, we demonstrated recent and ongoing MHC class I and II introgression in the Carpathian region. The extent of introgression correlated with the age of contact. In the older zone, MHC similarity between species within transects exceeded similarity between transects within species, implying pervasive introgression ‐ a massive exchange of MHC genes, not limited to specific variants. In simulations, the observed pattern emerged under the combined action of balancing selection and hybridization, but not when these processes acted separately. Thus, massive introgression at advanced stages of divergence can introduce novel and restore previously lost MHC variation, boosting the adaptive potential of hybridizing taxa. In consequence, MHC genes may be the last to stop introgressing between incipient species.  相似文献

The phosphorylase kinase deficiency (Phk) locus has been located in the mouse X chromosome, the order of genes being centromere-Bn-Phk-Ta-jp. Allogenic stem cell transplantation can reduce lysosomal storage of heparan sulfate-derived oligosaccharides by up to 27 % in Sanfilippo MPS3a brain, but does not reduce the abnormal storage of sialolactosylceramide (GM3) or improve neurological symptoms, suggesting that ganglioside storage is in a non-lysosomal compartment. To investigate this further we isolated the Triton X100-insoluble at 4 °C, lipid raft (LR) fraction from a sucrose-density gradient from cerebral hemispheres of a 7 month old mouse model of Sanfilippo MPS3a and age-matched control mouse brain. HPLC/MS/MS analysis revealed the expected enrichment of normal complex gangliosides, ceramides, galatosylceramides and sphingomyelin enrichment in this LR fraction. The abnormal HS-derived oligosaccharide storage material was in the Triton X100-soluble at 4 °C fractions (8–12),whereas both GM3 and sialo[GalNAc]lactosylceramide (GM2) were found exclusively in the LR fraction (fractions 3 and 4) and were >90 % C18:0 fatty acid, suggesting a neuronal origin. Further analysis also revealed a >threefold increase in the late-endosome marker bis (monoacylglycerol) phosphate (>70 % as C22:6/22:6-BMP) in non-LR fractions 8–12 whereas different forms of the proposed BMP precursor, phosphatidylglycerol (PG) were in both LR and non-LR fractions and were less elevated in MPS3a brain. Thus heparan sulfate-derived oligosaccharide storage is associated with abnormal lipid accumulation in both lysosomal (BMP) and non-lysosomal (GM3 and GM2) compartments.  相似文献

Background

Ultrasound-targeted microbubble destruction (UTMD) is a type of ultrasound therapy, in which low frequency moderate power ultrasound is combined with microbubbles to trigger cavitation. Cavitation is the process of oscillation of gas bubbles causing biophysical effects such as pushing and pulling or shock waves that permeabilize biological barriers. In vivo, cavitation results in tissue permeabilization and is used to enable local delivery of nanomedicine. While cavitation can occur in biological liquids when high pressure ultrasound is applied, the use of microbubbles as cavitation nuclei in UTMD largely facilitates the induction of cavitation. UTMD is intensively studied for drug delivery into tumor tissue, but also for the activation of anti-tumor immune responses. The first clinical studies of UTMD-mediated chemotherapy delivery confirmed safety and efficacy of this approach.

The present review summarizes ultrasound settings, cavitation approaches, biophysical mechanisms of drug delivery, drug carriers, and pre-clinical and clinical applications of UTMD for drug delivery into tumors.  相似文献

Major effect genes are often used for germplasm identification, for diversity analyses and as selection targets in breeding. To date, only a few morphological characters have been mapped as major effect genes across a range of genetic linkage maps based on different types of molecular markers in sorghum (Sorghum bicolor (L.) Moench). This study aims to integrate all available previously mapped major effect genes onto a complete genome map, linked to the whole genome sequence, allowing sorghum breeders and researchers to link this information to QTL studies and to be aware of the consequences of selection for major genes. This provides new opportunities for breeders to take advantage of readily scorable morphological traits and to develop more effective breeding strategies. We also provide examples of the impact of selection for major effect genes on quantitative traits in sorghum. The concepts described in this paper have particular application to breeding programmes in developing countries where molecular markers are expensive or impossible to access.  相似文献

The aquatic filamentous cyanobacteria Anabaena oscillarioides and Trichodesmium sp. reveal specific cellular regions of tetrazolium salt reduction. The effects of localized reduction of five tetrazolium salts on N(2) fixation (acetylene reduction), CO(2) fixation, and H(2) utilization were examined. During short-term (within 30 min) exposures in A. oscillarioides, salt reduction in heterocysts occurred simultaneously with inhibition of acetylene reduction. Conversely, when salts failed to either penetrate or be reduced in heterocysts, no inhibition of acetylene reduction occurred. When salts were rapidly reduced in vegetative cells, CO(2) fixation and H(2) utilization rates decreased, whereas salts exclusively reduced in heterocysts were not linked to blockage of these processes. In the nonheterocystous genus Trichodesmium, the deposition of reduced 2,3,5-triphenyl-2-tetrazolium chloride (TTC) in the internal cores of trichomes occurs simultaneously with a lowering of acetylene reduction rates. Since TTC deposition in heterocysts of A. oscillarioides occurs contemporaneously with inhibition of acetylene reduction, we conclude that the cellular reduction of this salt is of use in locating potential N(2)-fixing sites in cyanobacteria. The possible applications and problems associated with interpreting localized reduction of tetrazolium salts in cyanobacteria are presented.  相似文献

Southeast Florida witnesses an enormous seasonal influx of upper trophic level marine predators each year as massive aggregations of migrating blacktip sharks (Carcharhinus limbatus) overwinter in nearshore waters. The narrow shelf and close proximity of the Gulf Stream current to the Palm Beach County shoreline drive tens of thousands of sharks to the shallow, coastal environment. This natural bottleneck provides a unique opportunity to estimate relative abundance. Over a four year period from 2011–2014, an aerial survey was flown approximately biweekly along the length of Palm Beach County. A high definition video camera and digital still camera mounted out of the airplane window provided a continuous record of the belt transect which extended 200 m seaward from the shoreline between Boca Raton Inlet and Jupiter Inlet. The number of sharks within the survey transect was directly counted from the video. Shark abundance peaked in the winter (January-March) with a maximum in 2011 of 12,128 individuals counted within the 75.6 km-2 belt transect. This resulted in a maximum density of 803.2 sharks km-2. By the late spring (April-May), shark abundance had sharply declined to 1.1% of its peak, where it remained until spiking again in January of the following year. Shark abundance was inversely correlated with water temperature and large numbers of sharks were found only when water temperatures were less than 25°C. Shark abundance was also correlated with day of the year but not with barometric pressure. Although shark abundance was not correlated with photoperiod, the departure of the sharks from southeast Florida occurred around the vernal equinox. The shark migration along the United States eastern seaboard corresponds spatially and temporally with the spawning aggregations of various baitfish species. These baseline abundance data can be compared to future studies to determine if shark population size is changing and if sharks are restricting their southward migration as global water temperatures increase.  相似文献
