Background
A selective sweep containing the insulin-like growth factor 1 (IGF1) gene is associated with size variation in domestic dogs. Intron 2 of IGF1 contains a SINE element and single nucleotide polymorphism (SNP) found in all small dog breeds that is almost entirely absent from large breeds. In this study, we surveyed a large sample of grey wolf populations to better understand the ancestral pattern of variation at IGF1 with a particular focus on the distribution of the small dog haplotype and its relationship to the origin of the dog.Results
We present DNA sequence data that confirms the absence of the derived small SNP allele in the intron 2 region of IGF1 in a large sample of grey wolves and further establishes the absence of a small dog associated SINE element in all wild canids and most large dog breeds. Grey wolf haplotypes from the Middle East have higher nucleotide diversity suggesting an origin there. Additionally, PCA and phylogenetic analyses suggests a closer kinship of the small domestic dog IGF1 haplotype with those from Middle Eastern grey wolves.Conclusions
The absence of both the SINE element and SNP allele in grey wolves suggests that the mutation for small body size post-dates the domestication of dogs. However, because all small dogs possess these diagnostic mutations, the mutations likely arose early in the history of domestic dogs. Our results show that the small dog haplotype is closely related to those in Middle Eastern wolves and is consistent with an ancient origin of the small dog haplotype there. A cascaded life cycle: reuse of electric vehicle lithium-ion battery packs in energy storage systems

Purpose
Lithium-ion (Li-ion) battery packs recovered from end-of-life electric vehicles (EV) present potential technological, economic and environmental opportunities for improving energy systems and material efficiency. Battery packs can be reused in stationary applications as part of a “smart grid”, for example to provide energy storage systems (ESS) for load leveling, residential or commercial power. Previous work on EV battery reuse has demonstrated technical viability and shown energy efficiency benefits in energy storage systems modeled under commercial scenarios. The current analysis performs a life cycle assessment (LCA) study on a Li-ion battery pack used in an EV and then reused in a stationary ESS.Methods
A complex functional unit is used to combine energy delivered by the battery pack from the mobility function and the stationary ESS. Various scenarios of cascaded “EV mobility plus reuse in stationary clean electric power scenarios” are contrasted with “conventional system mobility with internal combustion engine vehicles plus natural gas peaking power.” Eight years are assumed for first use; with 10 years for reuse in the stationary application. Operational scenarios and environmental data are based on real time-of-day and time-of-year power use. Additional data from LCA databases are utilized. Ontario, Canada, is used as the geographic baseline; analysis includes sensitivity to the electricity mix and battery degradation. Seven environmental categories are assessed using ReCiPe.Results and discussion
Results indicate that the manufacturing phase of the Li-ion battery will still dominate environmental impacts across the extended life cycle of the pack (first use in vehicle plus reuse in stationary application). For most impact categories, the cascaded use system appears significantly beneficial compared to the conventional system. By consuming clean energy sources for both use and reuse, global and local environmental stress reductions can be supported. Greenhouse gas advantages of vehicle electrification can be doubled by extending the life of the EV batteries, and enabling better use of off-peak low-cost clean electricity or intermittent renewable capacity. However, questions remain concerning implications of long-duration use of raw material resources employed before potential recycling.Conclusions
Li-ion battery packs present opportunities for powering both mobility and stationary applications in the necessary transition to cleaner energy. Battery state-of-health is a considerable determinant in the life cycle performance of a Li-ion battery pack. The use of a complex functional unit was demonstrated in studying a component system with multiple uses in a cascaded application.4.

Our method is useful for relative quantification of THFs across different growth conditions. Absolute quantification of THFs requires the use of labelled standards.5.

The results show that 1-butyl-3-methylimidazolium acetate (IL-Ac) predominantly resulted in fatty acids, whereas 1-ethyl-3-methylimidazolium tosylate (IL-Tos) mostly yielded phenolic structures. Interestingly, bark yielded more ionic liquid soluble metabolites compared to interior wood.Conclusion
From this one can conclude that the application of ionic liquids may expand the metabolic snapshot.6.

Background
In this study, we optimized the process for enhancing amylase production from Pseudomonas balearica VITPS19 isolated from agricultural lands in Kolathur, India.Methods
Process optimization for enhancing amylase production from the isolate was carried out by Response Surface Methodology (RSM) with optimized chemical and physical sources using Design expert v.7.0. A central composite design was used to evaluate the interaction between parameters. Interaction between four factors–maltose (C-source), malt extract (Nsource), pH, and CaCl2 was studied.Results
The factors pH and CaCl2 concentration were found to affect amylase production. Validation of the experiment showed a nearly twofold increase in alpha amylase production.Conclusion
Amylase production was thus optimized and increased yield was achieved.13.

Introduction
The differences in fecal metabolome between ankylosing spondylitis (AS)/rheumatoid arthritis (RA) patients and healthy individuals could be the reason for an autoimmune disorder.Objectives

Significant differences in the fecal metabolic profiles could distinguish AS/RA patients from healthy controls but could not distinguish between AS and RA patients. Significantly increased metabolites in AS/RA patients were taurine, methanol, fumarate, and tryptophan.Conclusion

Our study revealed that the tested extracts from N. Retusa contain many useful bioactive compounds. They induced in a time-dependent manner the apoptosis the tested cancerous our cell line. This result was confirmed by ladder DNA fragmentation profile and PARP cleavage, as well as a release in caspase-3 and caspase-8 level.Conclusion
Our results indicate that the tested compounds have a significant antiproliferative effect which may be due to their involvement in the induction of the extrinsic apoptosic pathway.16.

Background and aims
Functional traits may underlie differences in niches, which promote plant species co-existence, but also differences in competitive ability, which drive competitive exclusion. Empirical evidence concerning the contribution of different traits to niche differentiation and the ability to supress and tolerate competitors is very limited, particularly when considering belowground interactions.Methods
We grew 26 temperate grassland species along a density gradient of interspecific competitors to determine which belowground traits a) explain species’ ability to suppress and tolerate neighbours and b) contribute to niche differentiation, such that species with dissimilar trait values experience reduced competition.Results
We found that having larger root systems with extensive horizontal spread and lower root tissue density enabled efficient suppression of neighbours but did not significantly contribute to the ability to tolerate competition. Species with deeper root systems, lower specific root length and less branched roots were better at tolerating competition, but these traits did not significantly affect the ability to suppress neighbours. None of the measured traits contributed significantly to niche differentiation, either individually or in combination.Conclusions
This study provides little support for belowground traits contributing to species co-existence through niche differentiation. Instead, different sets of weakly correlated traits enable plants to either suppress or tolerate their competitors.17.

Introduction
Ceramides play a key role in skin barrier function in homeostatic and pathological conditions and can be sampled non-invasively through stratum corneum collection.Objectives

More than 1000 human and pig ceramides were identified. Three human and minipig ceramide classes, CER[AS], CER[NS] and CER[EOS] have been investigated in depth.Results
Sphingoid bases were characterized by a prevalence of chain lengths with sizes from C16 to C22, whereas fatty acids were mainly observed in the range of C22–C26. Overall, the ceramide profiles between human and minipig stratum corneum were similar. Differences in the CER[AS] and CER[NS] classes included a more homogeneous distribution of fatty acids (16–30 carbon atoms) in minipig, whereas in human longer fatty acid chains (>?24 carbon atoms) predominated.Conclusion
The method will be useful for the analysis of healthy and pathological skin in various specie, and the measurement of the relative distribution of ceramides as biomarkers for pharmacodynamic studies.19.
