The sexual reproductive process in Pinus lambertiana has not been completely described, and previous attempts to generate hybrids with Pinus monticola and other North American pines have not been successful. The nature of incompatibility barriers between P. lambertiana and P. monticola is unknown. This needs to be understood if strategies are to be developed to overcome the said barriers. In this paper, development on interspecific crosses is compared with that on intraspecific crosses on the same parent trees. Pollen grains of both species germinated on the nucellus of both species within a week after pollination. Seed cone receptivity in P. lambertiana came approximately 2 weeks after receptivity in P. monticola, and this delay was perpetuated throughout ovule development in the first year of the reproductive process. Development of the second-year seed cones proceeded more gradually in P. lambertiana. However, seed cones reached maturity only for P. monticola x P. lambertiana. In both crosses, the barriers to hybridization occurred during the second year of the reproductive process. With the P. lambertiana as the seed parent, it was manifested through the failure of the megaspores at the free-nuclear stage to resume development. When P. monticola was used as the seed parent, the male and female gametes failed to fuse. Our results clearly show that the barriers to hybridization in these species occur before or at fertilization. However, the exact mechanisms behind these are still unknown. Based on the results of this study, we present several strategies to bypass the developmental barriers and possibly produce hybrid progenies.  相似文献

Summary Attempts were made to obtain intergeneric hybrids between Diplotaxis siifolia, a wild species, and cultivars of Brassica (B. campestris, B. juncea, and B. napus). The crosses showed unilateral incompatibility. When the wild species was used as female parent, pollen germination and pollen tube growth were normal, but hybrid seeds aborted due to post-fertilization barriers. Reciprocal crosses (cultivars as female parent) showed strong pre-fertilization barriers; although pollen grains showed germination, pollen tubes failed to enter the stigma. Hybrids were realized in two of the crosses, D. siifolia x B. juncea and D. siifolia x B. napus, through ovary culture. The hybrids were multiplied in vitro by multiplication of axillary shoots, or somatic embryogenesis. Detailed studies were carried out on the hybrid D. siifolia x B. juncea. F1 hybrids had shrivelled anthers and were pollen sterile. Amphiploids of this hybrid showed 60% pollen fertility and produced seeds upon self-pollination as well as backcross pollination with the pollen of B. juncea.  相似文献

The reproductive biology of Pringlea antiscorbutica R. Br. (Brassicaceae), an endemic species from the southern Indian Ocean islands, is investigated here. Controlled crosses were performed between plants grown in a common garden on the Kerguelen Islands. Searching for potential insect pollinators, we investigated the flying ability of all insects known from the Kerguelen Islands. We showed evidence for self-compatibility, low pollen limitation in the absence of a biotic pollen vector and slight selfing depression in P. antiscorbutica. The insects present on the islands are mostly wingless and are not likely to be efficient pollinators of this species. Since P. antiscorbutica shows morphological adaptations to insect pollination despite the absence of pollinators in its present range, we conclude that this species recently evolved from insect pollination to wind and/or autonomous pollination.  相似文献

Former controlled crosses between twelve Pinus montana var. rostrata (Pinus mugo complex) and eight P. sylvestris clones revealed that only two P. sylvestris had efficiently fertilised P. montana. Two species-diagnostic chloroplast DNA markers were applied to verify the species purity of the parental clones. All maternal P. montana were unambiguously confirmed to belong to the P. mugo complex at both chloroplast DNA marker loci. Six P. sylvestris clones carried the `sylvestris' haplotypes. However, the same two P. sylvestris clones that had efficiently fertilised P. montana displayed the chloroplast haplotypes diagnostic to the P. mugo complex. The patterns of highly polymorphic cpDNA microsatellite markers in parents and offspring ruled out contamination by foreign pollen. We concluded that the two clones successful in the crosses represent fertile hybrids between the two species with P. mugo as the pollen donor. Consequently, DNA markers are proposed for verifying or falsifying the success of artificial fertilisation in general. The existence of crossing barriers between the two Pinus species, meaningful to the postulated natural hybridisation and the evolution of their populations in sympatric stands, was indicated and is newly discussed.  相似文献

Summary Preliminary results from a large number of reciprocal crosses between the closely related sympatric species S. gourlayi Hawkes (2n=4x=48) and S. oplocense Hawkes (2n=6x=72) indicated that they are difficult to hybridize. Pollen-pistil incompatibility barriers were detected via fluorescent microscopy. The cross incompatibility reaction occurred at three sites in 6x×4x crosses; on the stigma, in the first one-third of the style, and in the first two-thirds of the style. In the reciprocal 4x×6x crosses the incompatibility reaction invariably occurred in the ovary. Backcrosses of interspecific pentaploid hybrids (that were occasionally formed) to both parental populations were fully compatible, partially compatible, and fully incompatible with three sites of cross-incompatibility reaction similar to those observed in 6x×4x crosses, respectively. Both polyploid species were found to be selfcompatible, whereas their F1 hybrids were found to be self-incompatible. An hypothesis based on interactions of dominant cross-incompatibility (CI) genes in pistils and dominant specific complementary genes in pollen grains is postulated to explain these observations. The cross-incompatibility system that appears to be operating in nature between 4x S. gourlayi and 6x S. oplocense provides a way for gene exchange between sympatric populations without threatening the identity of either species.  相似文献

Background and Aims

Corymbia species from different sections hybridize readily, with some of increasing economic importance to plantation forestry. This study explores the locations of reproductive barriers between interspecific Corymbia hybrids and investigates the reproductive success of a wide taxonomic range of C. torelliana hybrid crosses.

Methods

Pollen, pistil and embryo development were investigated for four C. torelliana crosses (×C. torelliana, ×C. citriodora subsp. citriodora, ×C. tessellaris and ×C. intermedia) using fluorescent and standard microscopy to identify the locations of interspecific reproductive isolating barriers. Corymbia torelliana was also crossed with 16 taxa, representing six of the seven Corymbia sections, both Corymbia subgenera and one species each from the related genera, Angophora and Eucalyptus. All crosses were assessed for capsule and seed yields.

Key Results

Interspecific C. torelliana hybridization was controlled by pre-zygotic reproductive isolating barriers inhibiting pollen adhesion to the stigma, pollen germination, pollen tube growth in the style and pollen tube penetration of the micropyle. Corymbia torelliana (subgenus Blakella, sect. Torellianae) was successfully hybridized with Corymbia species from subgenus Blakella, particularly C. citriodora subsp. citriodora, C. citriodora subsp. variegata, C. henryi (sect. Maculatae) and C. tessellaris (sect. Abbreviatae), and subgenus Corymbia, particularly C. clarksoniana and C. erythrophloia (sect. Septentrionales). Attempted intergeneric hybrids between C. torelliana and either Angophora floribunda or Eucalyptus pellita were unsuccessful.

Conclusions

Corymbia hybrids were formed between species from different sections and subgenera, but not with species from the related genera Angophora or Eucalyptus. Reproductive isolation between the interspecific Corymbia hybrid crosses was controlled by early- and late-acting pre-zygotic isolating barriers, with reproductive success generally decreasing with increasing taxonomic distance between parent species. These findings support the monophyly of Corymbia and the close relationships of infrageneric clades. The hybridizing propensity of Corymbia species provides opportunities for breeding but suggests risks of environmental gene flow.  相似文献

We carried out a demographic study and evaluated the genetic and morphological variability in five populations of the endangered Sophronitis sincorana (Orchidaceae) endemic to Northeastern Brazil, based on allozyme and morphometric analyses. Plant density was approximately 0.5 plants/m2, and the projected total number of plants was approximately 50,000 individuals. However, fruit set and recruitment of individuals are rare. The genetic variability was very high in all populations (P = 100, A = 3.0−3.5, H e = 0.33−0.48), and all populations presented similar values of morphological variability. Low genetic and morphological structuring were found in the species (F ST = 0.053, AMRPP = 0.018). The elevated coefficient of endogamy encountered in populations of S. sincorana indicates the occurrence of structuring within the populations. The lack of correlation between morphological and genetic variation in this species indicates that none of the markers examined should be used separately for either conservation purposes.  相似文献

Summary Attempts at the reciprocal cross between Petunia parodii and P. inflata using standard emasculation and pollination techniques failed. Limited pollen tube growth down the style in reciprocal crosses led to reproductive isolation between the self-compatible P. parodii and self-incompatible P. inflata. The interspecific hybrid was successfully produced by bud-pollination of P. parodii with P. inflata as the male parent in 22 percent of attempts, but not in the opposite direction. In vitro pollination of P. parodii ovaries with P. inflata pollen also produced hybrids. The small size of the ovary made it technically impossible to use P. inflata as the female parent for in vitro pollination. The interspecific hybrids were intermediate, as compared to the two parents, for six of the seven plant and flower characters measured. Furthermore, the hybrids had high pollen fertility, set abundant seed upon self-pollination, and readily inter-crossed with the parental species. The results are consistent with a high degree of chromosomal homology in the parental species and with minor genetic divergency leading to reproductive isolation that is pre-zygotic in nature. Overcoming the barriers to cross-incompatibility by practical techniques resulted in fertile interspecific hybrids that segregated for parental characters. The localized hybridization between Orchis mascula and O. provincialis in Sardinia is investigated. The two parental species are closely related and share the same pollinators but do not hybridize in any other areas of sympatry. Artificial crosses between the two species in areas of sympatry on the continent revealed that, in absence of an ethological isolation, a strong post-pollination barrier exists and preserves species integrity in the main part of their distribution. However, on Sardinia, morphological observations, karyological and molecular analyses of intermediate phenotypes confirmed the occurrence of hybridization between the insular O. mascula ssp. ichnusae and O. provincialis and the bi-directionality of pollen flow. The occurrence of hybrids on the island indicates a failure of otherwise strong post-pollination barriers between O. mascula and O. provincialis.  相似文献

The reproductive system was studied inCrocus genus (Iridaceae) following intra- and interspecific pollination, by using light and scanning electron microscopy. The results suggest that the stigma-style tract of theCrocus pistil is a mere promoter of pollen tube growth, while intra- and interspecific discrimination of compatible and incompatible pollen occurs in the ovarian tract. Here, the transmitting tissue consists of special epidermal cells, whose granular or floccular secretions provide the selective medium for the growth of pollen tubes. The ovarian self-incompatibility (SI) is widespread within the genus, resulting in a partial or complete suppression of self-fertilization. Moreover, postzygotic SI mechanisms, as well as postzygotic mechanisms of unknown nature, seem to be recurrent and both are responsible for seed abortion. The interspecific ovarian incompatibility concerns only unrelated crosses; crosses between related fertile species succeed both in fertilization and seed-set.  相似文献

Background and Aims

Silene dioica and S. latifolia experience only limited introgression despite overlapping flowering phenologies, geographical distributions, and some pollinator sharing. Conspecific pollen precedence and other reproductive barriers operating between pollination and seed germination may limit hybridization. This study investigates whether barriers at this stage contribute to reproductive isolation between these species and, if so, which mechanisms are responsible.

Methods

Pollen-tube lengths for pollen of both species in styles of both species were compared. Additionally, both species were pollinated with majority S. latifolia and majority S. dioica pollen mixes; then seed set, seed germination rates and hybridity of the resulting seedlings were determined using species-specific molecular markers.

Key Results

The longest pollen tubes were significantly longer for conspecific than heterospecific pollen in both species, indicating conspecific pollen precedence. Seed set but not seed germination was lower for flowers pollinated with pure heterospecific versus pure conspecific pollen. Mixed-species pollinations resulted in disproportionately high representation of nonhybrid offspring for pollinations of S. latifolia but not S. dioica flowers.

Conclusions

The finding of conspecific pollen precedence for pollen-tube growth but not seed siring in S. dioica flowers may be explained by variation in pollen-tube growth rates, either at different locations in the style or between leading and trailing pollen tubes. Additionally, this study finds a barrier to hybridization operating between pollination and seed germination against S. dioica but not S. latifolia pollen. The results are consistent with the underlying cause of this barrier being attrition of S. dioica pollen tubes or reduced success of heterospecifically fertilized ovules, rather than time-variant mechanisms. Post-pollination, pre-germination barriers to hybridization thus play a partial role in limiting introgression between these species.  相似文献

Four supposedly closely related taxa were crossed artificially to test for hybridization barriers. Intra- as well as inter-specific crossings were made. The intra-specific crossings resulted in a high portion of viable hybrids, whereas hybridization barriers were found in all inter-specific combinations except one. The hybrids were non-viable at different levels, viz., (1) no seeds germinated, (2) seeds formed lethal chlorotic seedings, or (3) seeds formed chlorophyllous seedlings which survived the cotyledonary stage, but developed into intermediate forms with low pollen production and/or a strongly reduced pollen viability. Crossings betweenCampanula afra and any of the other three taxa resulted in a non-viable progeny. The crossing capacity amongC. occidentalis, C. kremeri, andC. dichotoma varied. Combinations with large-flowered maternal taxa and small-flowered paternal ones usually failed, whereas a high number of the reciprocal crossings produced hybrids which survived the cotyledonary stage. The hybrid plants varied in pollen viability and pollen production, however. It is concluded that theCampanula dichotoma group should be treated as comprising four distinct species.  相似文献

以新疆准噶尔盆地藜科猪毛菜属植物紫翅猪毛菜(Salsola affinis C.A.Mey)、钠猪毛菜(Salsola nitraria Pall)为研究对象,用繁殖分配比例的方法对比分析了两种猪毛菜不同海拔同一种群内不同个体大小繁殖分配的特点,并用异速生长模型分析了不同海拔繁殖生物量与营养生物量之间分配与个体大小的依赖关系。结果发现:1)不同海拔繁殖生物量(R)与营养生物量(V)呈不同程度的异速生长。紫翅猪毛菜随海拔的升高R-V的异速生长斜率显著升高,截距随海拔的升高没有显著增加;而钠猪毛菜的斜率随海拔升高显著降低,截距则显著升高。2)紫翅猪毛菜在较低海拔个体大小与繁殖分配呈负相关,在较高海拔呈正相关;钠猪毛菜在较低海拔个体大小与繁殖分配呈正相关,在较高海拔呈负相关;两种猪毛菜繁殖分配的适应对策相反。3)将同一种群个体大小分成大、中、小3种类型,多重比较发现紫翅猪毛菜在较低海拔,中小个体的繁殖分配显著高于大个体的繁殖分配;在较高海拔,大个体的繁殖分配显著高于中小个体的繁殖分配。钠猪毛菜在较低海拔,大个体的繁殖分配显著高于中、小个体的繁殖分配;在较高海拔,小个体的繁殖分配显著高于大、中个体的繁殖分配。综合分析认为:两个物种随海拔变化产生不同的繁殖分配策略,除遗传效应外,环境和个体大小对钠猪毛菜繁殖分配的变化均产生重要影响,而紫翅猪毛菜繁殖分配的变化主要由海拔差异导致。由于微生境对同一种群的个体大小产生影响,进而产生不同的繁殖分配模式,所以在干旱区更应重视个体大小对繁殖分配的影响。  相似文献

One fundamental signature of reinforcement is elevated prezygotic reproductive isolation between related species in sympatry relative to allopatry. However, this alone is inadequate evidence for reinforcement, as traits conferring reproductive isolation can occur as a by‐product of other forces. We conducted crosses between Silene latifolia and S. diclinis, two closely related dioecious flowering plant species. Crosses with S. latifolia mothers from sympatry exhibited lower seed set than mothers from five allopatric populations when S. diclinis was the father. However, two other allopatric populations also exhibited low seed set. A significant interaction between style length and sire species revealed that seed set declined as style length increased when interspecific, but not intraspecific, fathers where used. Moreover, by varying the distance pollen tubes had to traverse, we found interspecific pollen placement close to the ovary resulted in seed set in both long‐ and short‐styled S. latifolia mothers. Our results reveal that the long styles of S. latifolia in sympatry with S. diclinis contribute to the prevention of hybrid formation. We argue that forces other than reinforcing selection are likely to be responsible for the differences in style length seen in sympatry.  相似文献

Wild Mexican potato species are an important untapped source of useful variation for potato improvement. Introgression methods such as 2n gametes, chromosome doubling, and crossing with disomic 4x 2 endosperm balance number (EBN) bridge species have been used to overcome post-zygotic endosperm failure according to the EBN hypothesis. Stylar barriers can prevent zygote formation, bilaterally when zygote formation is blocked in both directions of the cross or unilaterally when zygote formation is blocked in self incompatible (SI) × self compatible (SC) crosses. In several Solanaceae species, the S-locus for SI has been implicated in interspecific incompatibility. The objectives of this research were to determine if: (1) disomic 4x 2EBN Solanum stoloniferum can be used as a bridge species for introgression of the Mexican 2x 1EBN species Solanum cardiophyllum and Solanum pinnatisectum, (2) pre- and/or post-zygotic barriers limit hybridization among EBN compatible Solanum inter-series crosses, and (3) reproductive barriers act unilaterally or bilaterally. Fruit formation and seed set was recorded for inter-pollinations of S. stoloniferum, 4x 2EBN chromosome doubled S. cardiophyllum and S. pinnatisectum, and 2x 2EBN S. tuberosum haploids (HAP) or haploid-species hybrids (H-S). In vivo pollen tube growth was analyzed for each cross combination with fluorescence microscopy. Attempts to create bridge hybrids between S. stoloniferum, and S. cardiophyllum or S. pinnatisectum were not successful. Pre- and post-zygotic barriers prevented seed formation in crosses involving S. cardiophyllum and S. pinnatisectum. Self compatibility in S. stoloniferum and S. pinnatisectum suggests that the S-locus does not contribute to the stylar barriers observed with these species. Alternatively, the presence of functional and nonfunctional (SC) S-alleles may explain interspecific incompatibility in intra- and inter-ploidy crosses. A non-stylar unilateral incongruity was discovered in H-S/HAP × S. stoloniferum crosses, indicating either a post-zygotic barrier, or a pre-zygotic barrier acting at or within the ovary. Furthermore, lack of S. stoloniferum pollen rejection may occur through absence of S. stoloniferum pollen-active genes needed to initiate pollen rejection, or through competitive interaction in S-locus heterozygous S. stoloniferum pollen. Introgression strategies using these species would benefit potato breeding by introducing genetic diversity for several traits simultaneously through co-current introgression.  相似文献

Background and Aims

Heteromorphy in flowers has a profound effect on breeding patterns within a species, but little is known about how it affects reproductive barriers between species. The heterostylous genus Primula is very diverse in the Himalaya region, but hybrids there have been little researched. This study examines in detail a natural hybrid zone between P. beesiana and P. bulleyana.

Methods

Chloroplast sequencing, AFLP (amplified fragment length polymorphism) markers and morphological comparisons were employed to characterize putative hybrids in the field, using synthetic F1s from hand pollination as controls. Pollinator visits to parent species and hybrids were observed in the field. Hand pollinations were conducted to compare pollen tube growth, seed production and seed viability for crosses involving different morphs, species and directions of crossing.

Key Results

Molecular data revealed all hybrid derivatives examined to be backcrosses of first or later generations towards P. bulleyana: all had the chloroplast DNA (cpDNA) of this species. Some individuals had morphological traits suggesting they were hybrids, but they were genetically similar to P. bulleyana; they might have been advanced generation backcrosses. Viable F1s could not be produced with P. bulleyana pollen on P. beesiana females, irrespective of the flower morphs used. Within-morph crosses for each species had very low (<10 %) seed viability, whereas crosses between pin P. bulleyana (female) and pin P. beesiana had a higher seed viability of 30 %. Thus genetic incompatibility mechanisms back up mechanical barriers to within-morph crosses in each species, but are not the same between the two species. The two species share their main pollinators, and pollinators were observed to fly between P. bulleyana and hybrids, suggesting that pollinator behaviour may not be an important isolating factor.

Conclusions

Hybridization is strongly asymmetric, with P. bulleyana the only possible mother and all detected hybrids being backcrosses in this direction. Partial ecological isolation and inhibition of heterospecific pollen, and possibly complete barriers to F1 formation on P. beesiana, may be enough to make F1 formation very rare in these species. Therefore, with no F1 detected, this hybrid zone may have a finite life span as successive generations become more similar to P. bulleyana.  相似文献

Hybridization is a common phenomenon in Daphnia (Cladocera; Anomopoda); interspecific hybrids have been found between several species and hybrids are found in many European lakes. Although much information on the morphology, ecology and genetics of hybrids is available, little is known about the level of reproductive isolation among species or about the relative fitness of hybrids and parental species. In order to facilitate studies on differentiation and speciation processes and comparative experimental studies on hybrids and recombinant genotypes, we present the first successful laboratory crossing experiments of two different Daphnia species, D. galeata and D. cucullata. Males and sexual females from two D. galeata and two D. cucullata clones were reciprocally crossed, juveniles hatched from resting eggs and reared until maturity. Hatching and juvenile survival rates of hybrids were relatively low (12.1% and 24%, respectively). D. galeata and D. cucullata clones vary in their level of successful interspecific matings and in the number of subsequent offspring. In general, hybrid crosses between D. cucullata females and D. galeata males were more successful than reciprocal crosses.  相似文献

Ten species in the genusSilene sectt.Siphonomorpha andAuriculatae were crossed artificially involving 612 crosses to test inter- and infraspecific, intervarietal and intersectional crossability. In sect.Siphonomorpha all interspecific crosses (between diploids) failed due to cross- or seed-incompatibility; however, intervarietal crosses betweenS. gigantea var.gigantea andS. gigantea var.incana produced hybrids. In sect.Auriculatae hybrids were produced betweenS. vallesia andS. boryi, both tetraploids, but crosses between these and the diploid species were unsuccessful. The biological containment of the potato (Solanum tuberosum) was assessed by establishing the crossability of this tuberous crop with the related wild non-tuberous species in The Netherlands, black nightshade (S. nigrum) and bittersweet (S. dulcamara). To circumvent crossability barriers, genotypes with different ploidy number were employed and crosses were performed under different environmental conditions. S. dulcamara was shown to be incongruent with potato at all ploidy levels, while S. nigrum displayed unilateral incompatibility. If S. nigrum was emasculated and used as female, fertilization by potato pollen resulted in berry set and seed development. Emasculation of S. nigrum was essential in this cross, because analysis of the fertilization process demonstrated that this species is highly self-compatible and potato pollen was outcompeted by pollen of S. nigrum. The hybrid seeds derived from this cross did not mature and appeared not to be viable. By application of the technique of embryo rescue of immature embryos, hybrid plants could be obtained. However, these hybrid plants proved to be sterile. These data demonstrate that gene flow by pollen dispersal from potato to its most common wild relatives in Western Europe is highly unlikely. The potato is thus a naturally contained species in this part of the world.  相似文献

In closely related plant species that display strong similarities in phenology and pollinator communities, differences in breeding system and associated shifts in floral traits may have important effects on the magnitude and direction of heterospecific pollen flow and hybridization. Here, we quantified the strength of several pre‐ and postzygotic barriers acting between the facultatively outcrossing Centaurium erythraea and the predominantly selfing C. littorale via a suite of experiments, and estimated the frequency of hybridization in the field using molecular markers. The reproductive barriers primarily responsible for preventing hybridization were essentially prezygotic and these acted asymmetrically. Due to differences in floral display, pollen production, and pollen transfer rates, heterospecific pollen flow occurred predominantly from C. erythraea to C. littorale. In C. littorale, on the other hand, close anther–stigma positioning and resulting higher capacity for autonomous selfing functioned as an efficient barrier to counterbalance the higher risk for hybrid mating. In both species the action of all reproductive barriers resulted in a small opportunity for hybrid establishment, which was confirmed by the occurrence of only ~1% putative hybrids in the field. Our findings confirm that differences in breeding system affect heterospecific pollen transfer patterns and that autonomous selfing may efficiently prevent hybridization.  相似文献
