Submicroscopic findings of T lymphocytic subpopulations of normal human peripheral blood defined by OKT monoclonal antibodies were studied using ultrastructural immunocytochemistry (immunogold staining). The results show that OKT4-and OKT8-positive lymphocytic subpopulations have a distinct morphological pattern, although some variations in the ultrastructural details of cells in each subset are evident. The most representative features of OKT8-positive cells are regular or slightly indented nucleus with condensed chromatin, abundant cytoplasm, prominent Golgi apparatus, several granules frequently localized in the Golgi area, and numerous mitochondria, often in cluster disposition. OKT4-positive cells show generally less condensed chromatin, scanty cytoplasm and poor organule pattern. Furthermore, positive lymphocytes with prominent nuclear irregularities are found in both lymphocytic subpopulations. Ultrastructural similarities between the T cell subsets phenotypically characterized by monoclonal antibodies and other immunological criteria are also discussed.  相似文献

The ultrastructural features, characterizing the different types of spermatogonia and spermatocytes in the blue fox, have been studied within and near the reproductive season, and also in the summer and autumn. Two distinct types of spermatogonia — A and Β — are described. The A-spermatogonia often have a prominent nucleolus and numerous cytoplasmic organelles including characteristic whorls of AER. Large vacuoles containing electron dense particles are sometimes observed. In the B-spermatogonia the chromatin forms condensed areas of varying size, and the nucleolus is usually absent. The number of cytoplasmic organelles is generally small. Ultrastructural characteristics are further used to distinguish between the different stages in the prophase of the primary spermatocytes. In leptotene the nucleus contains a thread-like chromatin with electron dense peripheral areas. Towards the end of the stage the mitochondria display dilated cristae, and aggregations of a granular material can be observed in the intermitochondrial matrix. Zytogene is characterized by the appearance of syniaptinemal complexes in the nucleus, and of the chromatoid body and piles of annulate lamellae in the juxtanuclear cytoplasm. In pachytene the chromosomes become apparent as aggregations of condensed chromatin associated with the synaptinemal complexes. The Golgi complex is more prominent than in the previous stages, and the number of the other cytoplasmic organelles is increasing. In the last stages of the prophase (diplotene and diakenesis) the chromosomes become still more electron dense, the nucleolus appears as a very prominent structure, and there is a marked vesiculation of the cytoplasm. The secondary spermatocytes have a characteristic nucleus with a somewhat irregular outline and larger peripheral areas of condensed chromatin. In the cytoplasm a double Golgi complex is frequently observed. In the summer and autumn spermatocytes in zygotene seem to represent the most advanced form of spermatogenic cells.  相似文献

SYNOPSIS. The structural changes in macro and microgametocytes of Parahaemoproteus velans following removal of infected blood from the avian host were studied in the light and electron microscope. Gametocytes of both sexes round up and soon escape from their host cells. Shortly thereafter they assume a dumbbell shape. The microgametocyte undergoes exflagellation forming 8 slender microgametes. During fertilization the entire microgamete appears to enter the female. The most striking ultrastructural change in the formation of the macrogamete is the condensation and enclosure by a membrane of abundant amophorus dense material seen in the cytoplasm of the immature gametocyte. Maturation of the microgametocyte begins prior to its escape from the host cell. Axonemes are present in the cytoplasm and nuclear reorganization occurs while the parasite is intracellular. Bundles of microtubules associated with condensed chromatin are found in the peripheral cytoplasm of maturing forms and apparently participate in the formation of small compact microgamete nuclei. Each of these filiform structures consists of a dense, centrally located nucleus and a single axoneme lying in flocculent cytoplasm. The nucleus and axoneme of the microgamete are seen free in the cytoplasm of a fertilized macrogamete.  相似文献

Spermiogenesis, in particular the head differentiation of Diplometopon zarudnyi, was studied at the ultrastructural level by Transmission Electron Microscope (TEM). The process includes acrosomal vesicle development, nuclear elongation, chromatin condensation and exclusion of excess cytoplasm. In stage I, the proacrosomal vesicle occurs next to a shallow fossa of the nucleus, and a dense acrosomal granule forms beneath it. This step commences with an acrosome vesicle forming from Golgi transport vesicles; simultaneously, the nucleus begins to move eccentrically. In stage II, the round proacrosomal vesicle is flattened by projection of the nuclear fossa, and the dense acrosomal granule diffuses into the vesicle as the fibrous layer forms the subacrosomal cone. Circular manchettes surrounded by mitochondria develop around the nucleus, and the chromatin coagulates into small granules. The movement of the nucleus causes rearrangement of the cytoplasm. The nucleus has uniform diffuse chromatin with small indices of heterochromatin. The subacrosome space develops early, enlarges during elongation, and accumulates a thick layer of dark staining granules. In stage III, the front of the elongating nucleus protrudes out of the spermatid and is covered by the flat acrosome; coarse granules replace the small ones within the nucleus. One endonuclear canal is present where the perforatorium resides. In stage IV, the chromatin concentrates to dense homogeneous phase. The circular manchette is reorganized longitudinally. The Sertoli process covers the acrosome and the residues of the cytoplasmic lobes are removed. In stage V, the sperm head matures.  相似文献

The ultrastructural modifications of the epithelial cells of rat corpus epididymis stimulated with gonadotropic hormone were studied. The structural variety of the cells depending on functional conditions becomes more prominent 6 h after the injection of gonadotropic hormone. Light large cells have one or often two nucleus-containing bing nucleoli, in their cytoplasm there are numerous vesicles, a well-developed Golgi apparatus, other organelles and lysosomal bodies. Some other cells are filled with many large vacuoles of different density, dense bodies and vesicles. Cells of another type which are in the majority show an unusually active structure reflecting the function of synthesis. The more prominent nucleolus is associated to clumps of chromatin. Their apical cytoplasm is filled by a structure related to absorption. The whole remaining part of their cytoplasm is covered with a very extensive Golgi apparatus and a very well developed granular endoplasmic reticulum. The extremely enlarged cisternae of this reticulum were found to be very closely applied to the basal cell membrane. There is a flocculent material inside the cisternae. Similar material is observed in the extracellular medium under the basal membrane. The epithelium seems normal 10 h after the injection of hormone, but large light cells make up the majority of them.  相似文献

Summary An electron cytochemical technique is described for the localization of GABA-T, the enzyme which degrades the neurotransmitter GABA, in rat cerebellar cortex. The technique allows ultrastructural demonstration of GABA-T activity by the final deposition of an electron dense formazan precipitate at reaction sites, whilst maintaining adequate ultrastructural preservation for recognition of cellular and subcellular structures. Numerous electron dense precipitates are evident as discrete punctate deposits situated mainly in mitochondria of stellate cells, basket cells and astrocytic glial cells; they are also seen in axonal or dendritic profiles at some synaptic junctions. The technique enables the first cytochemical demonstration of the mitochondrial localization of GABA-T activity in nervous tissue to be presented. It establishes that GABA-T is present in supposed GABA neurones, in pre- or post-synaptic endings, or both, of presumed inhibitory synapses and in glial cells which may be associated with these synapses. From this seemingly ubiquitous distribution, functional aspects of GABA-T in these cells is considered.  相似文献

Although lymphocytes are never present in 'normal ' seminiferous epithelium, they are found in the terminal portions of the seminiferous tubles near their junctions with the tubuli recti. Intraepithelia lymphocytes are also found in the tubuli recti testis, ductuli efferentes, epididymis and ductus deferens. The ultrastructural morphology of these cells closely resembles that of the intraepithelial lymphocytes in the intestinal mucosa and those obtained from the lymph nodes, spleen blood and thoracic duct. The mucleus is spherical and is characterized by clumps of chromatin near the nuclear membrane. A thin rim of cytoplasm is usually found, and is remarkably free of most cell organelles except for free ribosomes. Frequently, a blunt cytoplasmic process can be seen extending from one end of the cell. Membrane-bounded granules and other dense bodies are occasionally encountered in the cytoplasm. The possible functional significance of intraepithelial lymphocytes in the male reproductive tract is discussed.  相似文献

Columnar cells of the peritoneal epithelium in slender cords of the testes were examined in normal and estradiol benzoate-treated Japanese red-bellied newt, Cynops pyrrhogaster pyrrhogaster, by light and electron microscopy. In normal newts, the peritoneal epithelium covering the slender cord consists of columnar cells, which contain extraordinarily large, oval or spindle-shaped nuclei with conspicuous indentations. The nucleus contains chromatin granules and the cytoplasm is filled with numerous tonofilaments. The primordial germ cells are scattered throughout the slender cord, and each cell is surrounded by a few follicle cells. Between the germ cells and follicle cells there are microvilli-like processes. The nucleus of primordial germ cells is multilobate and has electron lucent areas, dispersed chromatin and several electron-dense nucleoli. In the lighter cytoplasm, the nuage material is found very near to nuclear pores, and is frequently seen among the mitochondria. The nucleolus-like body is not associated with other organelles. The primary spermatogonia have bilobate nuclei. It is remarkable that most of the cytoplasmic organelles are found in the deep nuclear indentations. The nuage material and nucleolus-like body are well developed in the cytoplasm. After treatment of newts with estradiol benzoate for one year, four types of cells can be distinguished in the peritoneal epithelium. One type is quite different from the columnar cells. These newly appeared cells are large and light in appearance. Their nucleus is highly lobate, and contains dispersed chromatin and several nucleoli with compact electron dense material in its periphery. The cells are characterized by the presence of nuage material and nucleolus-like bodies in the cytoplasm. There are microvilli-like processes between these cells and adjacent elongated cells. These ultrastructural characteristics of the light cells are very similar to those of primordial germ cells and/or primary spermatogonia in normal testes. These findings suggest that the light cells which appear in the peritoneal epithelium of the testes on administration of estrogen may be germ line cells.  相似文献

1. The location, distribution and morphological characteristics of the pigment cells found in the frog gut are described. 2. The pigment cells show long and large protoplasmic projections. At the ultrastructural level, the nucleus is elongated with prominent nucleolus and dense marginal chromatin. The cytoplasm is full with pigment granules (2500-7500 A) and typical premelanosome structures have been observed. 3. The pigment cells number is higher in the esophagus and large intestine than in the stomach or small intestine and the pigment cells are always located in close contact with blood vessels and nervous structures (ganglia and fibres). 4. We have observed that the pigment content depends upon seasonal variations, increasing during the cold months. 5. We have demonstrated by histological methods that the cells pigment content is melanin. 6. According to their morphological and tinctorial characteristics the anuran gut melanin storing cells are similar to the skin epidermal melanocytes.  相似文献

The cytologic features of a case of recurrent adamantinoma of the tibia diagnosed by fine needle aspiration (FNA) in a 31-year-old patient are described. The FNA smears contained single-lying cells and cells arranged in small clusters and Indian files, with some nuclear molding. Three cell types were present: (1) large polygonal cells with chromatin clearing, (2) smaller cells with poorly defined cytoplasm and dense chromatin and (3) fusiform cells. The first two were interpreted as epithelial in nature and the third one as being derived from the stroma. These cytologic features appear to be sufficiently characteristic to suggest an FNA diagnosis of adamantinoma, especially in the case of a recurrent tumor.  相似文献

A study of ovarian structure in adult Alligator Lizards (Gerrhonotus coeruleus) was conducted by light microscopy and transmission electron microscopy. Particular attention was directed to characterizing the ultrastructure of germ-line cells, prior to follicle formation. General ovarian structure in this lizard is similar to that of other lizards. The paired organs are hollow, thin-walled sacs containing follicles in roughly 3 to 4 size classes. Ovarian germinal tissue consists of oogonia (diploid cells which divide mitotically) and oocytes (meiotic cells), intermixed with ovarian surface epithelial cells. Germ cells reside in two dorsal patches of epithelium per ovary (germinal beds), as is common in lizards. Oogonia in interphase show a highly dispersed chromatin pattern. Within oogonia cytoplasm, Golgi complexes are scarce, rough endoplasmic reticulum is absent, and lipid droplets are rare. Ribosomes are scattered in small clusters. Small, round vesicles are common in all oogonia; glycogen-like granules are present in some. Mitochondria form a juxtanuclear mass within which groups of several mitochondria surround a dense granule. “Nuage” granules also are found unassociated with mitochondria. Oocytes are present in stages of meiotic prophase up to diplotene. Synaptinemal complexes are seen in several (pachytene) cells. The cytoplasm of oocytes differs from that of oogonia in that mitochondria do not form groups, and nuage and glycogen are absent, whereas small round vesicles and large irregular vesicles are common. The ultrastructural similarities in germ cells of a reptile as compared to those of other vertebrates strengthens the notion that germ-line cells possess (or lack) qualities related to the undifferentiated state of these cells.  相似文献

Testis of the Teleostean fish Trisopterus minutus capelanus has been examined to study the organization of the seminiferous tubules and the ultrastructural features of the germ cells. The testis is shown to be composed of seminiferous tubules full of cells: only few of them have just a very narrow lumen. Each tubule is divided by thin septa of connective tissue in zones containing homogeneous cells; such an organization is confirmed by ultrastructural images showing groups of synchronously developing germ cells. By morphological characterization of the germ cells found in each zone, 6 maturation stages have been identified. During spermiogenesis, a progressive shrinkage of germ cells and a nuclear chromatin condensation have been observed. Intercellular bridges, homogeneously dispersed granules of glycogen and groups of mitochondria associated with dense granular material have been described. Such features are present in the earlier stages of spermiogenesis and are retained until the later stages of spermatid differentiation. The spermatozoon shows a lack of acrosome as in many other teleosts previously studied.  相似文献

The epidermis of Ostariophysi fish is composed of 4 main cell types: epidermal cells (or filament containing cells), mucous cells, granular cells and club cells. The morphological analysis of the epidermis of the catfish Pimelodella lateristriga revealed the presence of only two types of cells: epidermal and club cells. The latter were evident in the middle layer of the epidermis, being the largest cells within the epithelium. Few organelles were located in the perinuclear region, while the rest of the cytoplasm was filled with a non-vesicular fibrillar substance. Club cells contained two irregular nuclei with evident nucleoli and high compacted peripheral chromatin. Histochemical analysis detected prevalence of protein within the cytoplasm other than carbohydrates, which were absent. These characteristics are similar to those described to most Ostariophysi studied so far. On the other hand, the epidermal cells differ from what is found in the literature. The present study described three distinct types, as follows: superficial, abundant and dense cells. Differences among them were restricted to their cytoplasm and nucleus morphology. Mucous cells were found in all Ostariophysi studied so far, although they were absent in P. lateristriga, along with granular cells, also typical of other catfish epidermis. The preset study corroborates the observations on club cells'' morphology in Siluriformes specimens, and shows important differences in epidermis composition and cell structure of P. lateristriga regarding the literature data.  相似文献

Ethidium bromide (EB) is widely used for investigating the DNA conformation in chromatin both with conventional and cytofluorimetric techniques. Since the interaction of the dye with DNA should result in structural deformations which can be different in isolated or in situ chromatin, a study has been performed on the effects caused by different amounts of EB and the analogous propidium iodide on isolated nuclei, in which chromatin maintains its native relationships with the other nuclear structures (envelope, nucleolus, interchromatin RNP, nuclear matrix). The results obtained by comparing ultrastructural observations in thin sections and in freeze-fracturing with conformational analysis in multiparameter flow cytometry indicate that the phenanthridinic fluorochromes, especially at the high concentrations used for cytofluorimetric analyses, cause deep rearrangements of the chromatin in situ. These effects consist both in aggregation and condensation of the fibers into the dense chromatin domains, and in an increase of the supernucleosomal configuration associated with an enlargement of interchromatin spaces in which the RNP particles appear particularly evident. These results, discussed with those available on isolated chromatin, suggest that any unwinding effect of the intercalating dyes on the DNA cause a general condensation of chromatin as a consequence of the constraints which characterize the organization of the chromatin inside the nucleus.  相似文献

Ultrastructural observations indicate that the primary spermatocyte of Trichuris muris is larger than the spermatogonial stage with an increased cytoplasm to nucleus ratio. The cytoplasm contains an extensive reticular system, mitochondria, numerous free ribosomes and prominent Golgi complexes which may contribute to the formation of a sub-surface, vesicular complex. Although only two spermatocytes were seen to be linked by a cytoplasmic bridge it is suggested that the number of conjoined cells is probably greater. The rearrangement of mitochondria in a ring around the nucleus and the indentation and vesiculation of the nuclear envelope preceeded its disappearance and indicated the onset of meiosis. Centrioles were frequently resolved at this stage. They were composed of nine peripheral doublets surrounded by a dense pericentriolar sheath. Three dense chromatin areas indicative of haploid chromosomes were present in later meiotic stages. Each chromosome was surrounded by a number of mitochondria and there was a clear separation of the chromosome-mitochondrial clusters from the remainder of the cytoplasm. This was particularly evident at telophase when two daughter cells were partially separated by membrane infoldings. This reflects incomplete cytokinesis in the dividing spermatocyte of T. muris and is similar to that described in other trichuroid species. A close association with processes of the non-germinal, sustentacular cells was noted throughout the spermatocyte stage.  相似文献

The ultrastructural investigation of the root cells ofAllium cepa L. exposed to 1 mM and 10 mM cadmium (Cd) for 48 and 72 h was carried out. The results indicated that Cd induced several obvious ultrastructural changes such as increased vacuolation, condensed cytoplasm with increased density of the matrix, reduction of mitochondrial cristae, severe plasmolysis and highly condensed nuclear chromatin. Electron dense granules appeared between the cell wall and plasmalemma. In vacuoles, electron dense granules encircled by the membrane were aggregated and formed into larger precipitates, which increase in number and volume as a consequence of excessive Cd exposure. Data from electron energy loss spectroscopy (EELS) confirmed that these granules contained Cd and showed that significantly higher level of Cd in vacuoles existed in the vacuolar precipitates of meristematic or cortical parenchyma cells of the differentiating and mature roots treated with 1 mM and 10 mM Cd. High levels of Cd were also observed in the crowded electron dense granules of nucleoli. However, no Cd was found in cell walls or in cells of the vascular cylinder. A positive Gomori-Swift reaction showed that small metallic silver grains were abundantly localized in the vesicles, which were distributed in the cytoplasm along the cell wall.  相似文献

The ultrastructural distribution of poly(ADP-ribose)polymerase has been studied using a specific antibody and immunocytochemistry with immunogold markers. In situ localization in synchronized Chinese hamster ovary cells reveals the antibody associated with mitotic chromosomes, and later with condensed chromatin and perichromatin regions in G1 phase. During S and G2, the label occurs mostly on perichromatin regions where perichromatin fibrils are also observed. In the nucleolus, the label appears especially on the dense fibrillar component and to a minor extent on the granular component. Immunolabeled spread active chromatin preparations from exponentially growing cultured mouse P815 cells indicate preferential association of the antibody with nascent nonribosomal RNP fibrils compared to inactive chromatin. The results, suggesting a relationship between the poly(ADP-ribose)polymerase occurrence and RNA (or RNP) formation, are discussed in view of the present knowledge about possible relations between poly(ADP-ribosylation) and synthesis of RNA and DNA.  相似文献

Ultrastructural studies were performed on the blood cells of Planorbarius corneus. Spreading and round cell forms were observed. The spreading form can be distinguished from the round form by its abundant cytoplasm, pseudopods and irregularly shaped nucleus. Electron dense granules in the mitochondrial matrix are found in the round form and are lacking in the spreading form. Moreover, cellular apoptosis is observed in the round form. The ultrastructural findings and the previously described characteristics support the idea that the two cell forms belong to two basic cell types.  相似文献
