Statistical techniques known as the analysis of variance make it possible for the morphologist to plan work in such a way as to get quantitative data with the greatest possible economy of effort. This paper explains how to decide how many measurements to make per micrograph, how many micrographs per tissue block or organ, and how many organs or individuals are necessary for getting an exactness of sufficient quality of the results. The examples furnished have been taken from measuring volume densities of mitochondria in heart muscle cells and from cell counting in lymph nodes. Finally we show, how to determine sample sizes, if we are interested in demonstration of significant differences between mean values.  相似文献

The problem of the kinetic depth effect is revisited. We study how many points in how many views are necessary and sufficient to recover structure. The constraints in the cases where the velocities of the image points are known, and the positions of the image points are known with the correspondence between them established, are different and they have to be studied separately. In the case of two projections of any number of points there are infinitely many solutions, but if we regularize the problem we get a unique solution under some assumptions. Finally, an algorithm is discussed for learning this particular kind of regularization.  相似文献

Much has been written about how law as an institution has developed to solve many problems that human societies face. Inherent in all of these explanations are models of how humans make decisions. This article discusses what current neuroscience research tells us about the mechanisms of human decision making of particular relevance to law. This research indicates that humans are both more capable of solving many problems than standard economic models predict, but also limited in ways those models ignore. This article discusses how law is both shaped by our cognitive processes and also shapes them. The article considers some of the implications of this research for improving our understanding of how our current legal regimes operate and how the law can be structured to take advantage of our neural mechanisms to improve social welfare.  相似文献

The use of databases for the conservation of biodiversity is increasing. During the last decade, such a database has been created for European stream macroinvertebrates. Today, it includes 527 sites that are the least human-impacted representatives of many stream types across many European regions. It includes data on the abundance of 312 invertebrate genera, several environmental site characteristics, collection methods, bibliographic data sources, and 11 biological traits of the genera (e.g. size, life cycle, food and feeding habits, described in 61 categories). The database will be useful in addressing many topics that are potentially relevant to biodiversity conservation. To illustrate this potential, we provide examples of how the data could be exploited. First, we describe the frequency of some taxonomic and biological characteristics (e.g. richness and diversity of genera and traits) of the macroinvertebrate communities and assess how these characteristics are related (e.g. how trait richness increases with genus richness). Second, we describe the frequency of some characteristics of the genera and traits (e.g. occurrence frequency, abundance, dispersion index) and again assess how these characteristics are related (e.g. how occurrence increases with abundance). Finally, we suggest how the database could be developed into a collective, publicly accessible database that covers stream types and regions of Europe more comprehensively.  相似文献

Eukaryotes use a tiny protein called ubiquitin to send a variety of signals, most often by post-translationally attaching ubiquitins to substrate proteins and to each other, thereby forming polyubiquitin chains. A combination of biophysical, biochemical, and biological studies has shown that complex macromolecular dynamics are central to many aspects of ubiquitin signaling. This review focuses on how equilibrium fluctuations and coordinated motions of ubiquitin itself, the ubiquitin conjugation machinery, and deubiquitinating enzymes enable activity and regulation on many levels, with implications for how such a tiny protein can send so many signals.  相似文献

The research agenda of paleoanthropology involves many topics and methodologies. Fossil specimens are allocated to species, and those species are assigned to the hominin clade. After that we want to know how they are related to each other, what they ate, how much they weighed, how smart they were, etc. We also want to know about the origin of particular attributes of hominins, such as our delayed growth and development, bipedalism, and language. The data available to answer these complex questions are confounded by fragmentary fossil specimens, small sample sizes, limited opportunities for controlled experimentation, and the inherent limitations of historical data. Also, because many traits are effectively unique to hominins, even observational comparative studies are inevitably limited in what they can tell us, if not impossible to conduct. We explore how these limitations should, but often do not, constrain the questions that paleoanthropologists should attempt to answer.  相似文献

Cancer is the subject of intense research around the world, but many questions about how the disease works remain unanswered. How exactly does cancer start and how do tumours grow? In fact, at present there are ten times more anticancer drugs being tested in clinical trials than there were 15 years ago. However, many of the new anticancer agents are predicted to show clinical benefit in only small subpopulations of patients. The cancer stem cell model could explain not only how some cancers work but also why patients suffer relapses, providing a good opportunity to gain insight into the reasons why agents work or, more commonly, don't work, before going into a clinical trial.  相似文献

The large GTPase dynamin is the first protein shown to catalyze membrane fission. Dynamin and its related proteins are essential to many cell functions, from endocytosis to organelle division and fusion, and it plays a critical role in many physiological functions such as synaptic transmission and muscle contraction. Research of the past three decades has focused on understanding how dynamin works. In this review, we present the basis for an emerging consensus on how dynamin functions. Three properties of dynamin are strongly supported by experimental data: first, dynamin oligomerizes into a helical polymer; second, dynamin oligomer constricts in the presence of GTP; and third, dynamin catalyzes membrane fission upon GTP hydrolysis. We present the two current models for fission, essentially diverging in how GTP energy is spent. We further discuss how future research might solve the remaining open questions presently under discussion.  相似文献

Abstract: Statistics is one of the most important yet difficult subjects for many ecology and wildlife graduate students to learn. Insufficient knowledge about how to conduct quality science and the ongoing debate about the relative value of competing statistical ideologies contribute to uncertainties among graduate students regarding which statistical tests are most appropriate. Herein, we argue that increased education of the available statistical tests alone is unlikely to ameliorate the problem. Instead, we suggest that statistical uncertainties among graduate students are a secondary symptom of a larger problem. We believe the root cause lies in the lack of education on how to conduct science as an integrated process from hypothesis creation through statistical analysis. We argue that if students are taught to think about how each step of the process will affect all other steps, many statistical uncertainties will be avoided.  相似文献

Although scientists have studied and touted the importance of insects to many ecosystem services for decades, insects and insect science are often poorly perceived by the public and by policy makers. Because insects do have important influences on many ecosystem services, they have even greater potential to be used to solve some anthropogenically-caused current global problems. We give several examples where insects are currently being used, or are being considered for future use, as a part of the solution to a global problem, including their potential roles as a part of the solution to sustainable fuel and food systems, deforestation and other environmental degradation, and global inequities. Ecologists have an important role to play in assessing how many of the proposed uses of insects will alter ecological processes and ecosystem services, or how their use may be effectively implemented.  相似文献

To bridge these vast difference in scales, biologists construct hierarchies of organization of cellular structures. HIV-1 and parasitic infections co-circulate in many populations, and in a few well-studied examples HIV-1 co-infection is known to amplify parasite transmission. There are indications that HIV-1 interacts significantly with many other parasitic infections within individual hosts, but the population-level impacts of co-infection are not well-characterized. Here we consider how alteration of host immune status due to HIV-1 infection may influence the emergence of novel parasite strains. We review clinical and epidemiological evidence from five parasitic diseases (malaria, leishmaniasis, schistosomiasis, trypanosomiasis and strongyloidiasis) with emphasis on how HIV-1 co-infection alters individual susceptibility and infectiousness for the parasites. We then introduce a simple modelling framework that allows us to project how these individual-level properties might influence population-level dynamics. We find that HIV-1 can facilitate invasion by parasite strains in many circumstances and we identify threshold values of HIV-1 prevalence that allow otherwise unsustainable parasite strains to invade successfully. Definitive evidence to test these predicted effects is largely lacking, and we conclude by discussing challenges in interpreting available data and priorities for future studies.  相似文献

Living cells maintain a delicate balance between oxidizing and reducing species, and many disorders such as rheumatoid arthritis and Alzheimer's disease have been associated with a disturbed intracellular 'redox equilibrium'. The past few years have witnessed accelerated research into how natural redox responses and antioxidant defence systems are activated and how they restore a healthy redox balance. To function properly, many of these processes rely on a powerful sulfur redox chemistry, which is best exemplified by the complex, newly emerging cysteine-based redox regulation of the glutathione and thioredoxin pathways. Other redox systems based on oxidatively activated amino acid side chains in proteins are also becoming increasingly important, but are still barely understood or explored.  相似文献

Several countries spanning the equatorial forest regions of Africa have had outbreaks of Ebola hemorrhagic fever over the last three decades. This article is an overview of the many published investigations of how Ebola virus circulates in its natural environment, focusing on the viral reservoir, susceptible animal species, environmental conditions favoring inter-species transmission, and how the infection is transmitted to humans. Major breakthroughs have been made in recent years but many outstanding questions must be dealt with if we are to prevent human outbreaks by interfering with the viral life cycle.  相似文献

Common signaling pathways such as those for Wnts and BMPs are used many times during embryogenesis. During the development of the neural crest, Wnt and BMP signals are used repeatedly at different stages to influence initial induction, segregation from the neuroepithelium and cell fate determination. This review considers how specificity is generated within the neural crest for these reiterated signals, discussing examples of how the outcomes of signaling events are modulated by context.  相似文献
