The characteristics of the flow of culture medium significantly affects the photosynthetic productivity of bioreactors incorporating microalgae. Therefore, in order to optimize the performance of a conical helical tubular photobioreactor (CHTP) designed to be useful in practical applications, we characterized the flow pattern of the culture medium through the reactor. The effects of medium flow conditions on the photosynthetic productivity of Chlorella sp. were investigated using several different CHTP units with 0.50-m2 installation areas which were designed to vary the direction and rate of flow driven by airlift. In addition, the performance of two- and four-unit systems constructed by combining individual CHTP units was evaluated. We found that when medium flowed from the bottom to the top of the photostage, it exhibited smoother flow of culture medium than when flowing from top to bottom, which led to higher photosynthetic productivity by the former. Consistent with theoretical calculations, varying the lengths of vertical flow passages caused flow rates to vary, and higher flow rates meant smoother circulation of medium and better photosynthetic performance. Flow of medium through a four-unit CHTP system was similar to that in single units, enabling a photosynthetic productivity of 31.0 g-dry biomass per m2-installation area per day to be achieved, which corresponded to a photosynthetic efficiency of 7.50% (photosynthetically active radiation (PAR; 400-700 nm)). This high photosynthetic performance was possible because smoother medium flow attained in single units was also attained in the four-unit system.

As photosynthetic efficiencies are relatively high at irradiation levels of <500 micromol m(-2) s(-1), photosynthetic productivity could be increased by redistributing strong light over a larger photo-receiving area using conical, helical, tubular photobioreactors (HTP). When Chlorella were exposed to light irradiation of 980 micromol m(-2) s(-1), the ratio of productivities was 1.00:1.13:1.23:1.66 for conical HTPs with cone angles of 180 degrees (flat type), 120 degrees, 90 degrees, and 60 degrees, respectively. This suggests that photo-redistribution technology is a highly effective and convenient approach for increasing the photosynthetic productivity of microalgae.

An especial method was applied for calculating the photosynthetic productivity of oak (Quercus robur L.) stand. The methodological basis is that the efficiency of photosynthetic utilization of intercepted PAR over the daylight period depends on environmental conditions and distribution of PAR within the canopy. This method was also used to determine the photosynthetic productivity of individual trees within the stand and to estimate the contribution of trees with various extents of growth retardment to the total photosynthetic productivity. For the oak stand in the forest-steppe region, the photosynthetic efficiency was expressed as a function of climatic variables such as solar irradiance day sum, day average temperature, air humidity, and water availability (quantified as the predawn leaf water potential, PLWP). This functional dependence differed for sun-exposed and shade leaves. Under optimum water availability (PLWP = −0.5 MPa), the photosynthetic productivity of a 60-year-old oak stand amounted 44.5 t C/(ha year) but it declined with the decrease in water availability. Under severe water deficit, when PLWP dropped to −2.1 MPa, the photosynthetic productivity decreased 17-fold and equaled 2.6 t C/(ha year). The strongest decline in photosynthetic productivity occurred on sunny days under moisture deficit in dominant and codominant trees belonging to growth classes I and II.__________Translated from Fiziologiya Rastenii, Vol. 52, No. 4, 2005, pp. 522–531.Original Russian Text Copyright © 2005 by Molchanov.

Microalgal photosynthesis requires appropriate culture medium temperatures to achieve high photosynthetic performance and to maintain production of a high-quality biomass product. Enclosed systems, such as our conical, helical tubular photobioreactor (HTP), can accomplish high photosynthetic efficiency and the small amount of culture medium used by these systems means that the culture medium temperature may be effectively controlled. On the other hand, because a high ratio of surface area to culture medium volume leads to rapid heating under the illumination condition and substantial heat loss at night, maintaining a suitable culture medium temperature is necessary to achieve efficient, commercially practical biomass production. In order to predict changes in the culture medium temperature caused by changes in solar irradiance and ambient temperature, it is necessary to understand the heat balance within the photobioreactor. We therefore investigated the heat balance in three major parts (photostage, degasser, and helical heat exchanger) of our conical HTP, analyzed the time-dependent changes in medium temperature at various room temperatures and radiant energy inputs, and predicted changes in the culture medium temperature based on the characteristics of heat transfer among the three parts. Using this model, the predicted changes in culture medium temperature were very similar to the changes observed experimentally in the laboratory and under field conditions. This means that by calculating the time-dependent changes in the culture medium temperature, based on measurements of solar energy input and ambient temperature, we should be able to estimate the energy required to maintain the culture medium temperature within a range where photosynthetic performance of microalgae is high.

The photosynthetic performance of a helical tubular photobioreactor ("Biocoil"), incorporating the filamentous cyanobacterium Spirulina platensis, was investigated. The photobioreactor was constructed in a cylindrical shape (0.9 m high) with a 0.25-m(2)basal area and a photostage comprising 60 m of transparent PVC tubing of 1.6-cm inner diameter (volume = 12.1 L). The inner surface of the cylinder (area = 1.32 m(2)) was illuminated with cool white fluorescent lamps; the energy input of photosynthetically active radiation(PAR, 400 to 700 nm) into the photobioreactor was 2920 kJ per day. An air-lift system ncorporating 4%CO(2) was used to circulate the growth medium in the tubing. The maximum productivity achieved in batch culture was 7.18 g dry biomass per day [0.51 g . d biomass/L . day, or 5.44 g . d biomass/m(2)(inner surface of cylindrical shape)/day] which corresponded to a photosynthetic (PAR) efficiency of 5.45%. The CO(2) was efficiently removed from the gaseous stream; monitoring the CO(2) the outlet and inlet gas streams showed a 70% removal of CO(2) from the inlet gas over an 8-h period with almost maximum growth rate. Chlorella zofingiensis was grown in semi-continuous culture in an outdoor enclosed tubular photobioreactor. At the quasi-steady state, the response of the culture to changes in photosynthetically active radiance (PAR) was studied by following closely the dissolved O2 concentration, biomass concentration and the carbohydrate and protein content of the biomass. Generally, O2 production and the output of carbon and nitrogen of the biomass showed a linear correlation with incident PAR, suggesting that the cultures were light-limited. Photoinhibition was not observed in high light adapted cultures (on a sunny day), but was observed in shade adapted cultures(cloudy days) when there was a sudden four-fold increase in PAR. The output rate of biomass nitrogen observed on sunny days was, however, lower than that measured on cloudy days. On sunny days, a rapid increase in the C/N ratio of the biomass was seen. We attribute the increase in C/N ratio on sunny days to a slower response of protein synthesis to big increases in PAR, compared to carbohydrate synthesis. The possible influence of this C and N response pattern on the productivity of outdoor algal cultures is discussed. This revised version was published online in August 2006 with corrections to the Cover Date.

The production of microalga Phaeodactylum tricornutum in an outdoor helical reactor was analysed. The influence of temperature, solar irradiance and air flow rate on the yield of the culture was evaluated. Biomass productivities up to 1.5 g l(-1) per day and photosynthetic efficiency up to 14% were obtained by maintaining the cultures below 30 degrees C, dissolved oxygen levels less than 400% Sat. (with respect to air saturated culture) and controlling the cell density in order to achieve an average irradiance within the culture below 250 microE m(-2) s(-1). Under these conditions, the fluorescence parameter, Fv/Fm, which reflects the maximal efficiency of PSII photochemistry, remained roughly 0.6-0.7 and growth rates up to 0.050 h(-1) were achieved. The average irradiance and the light/dark cycle frequency, were the variables determining the behaviour of the cultures. A hyperbolic relationship between growth rate and biomass productivity with the average irradiance was observed, whereas both biomass productivity and photosynthetic efficiency linearly increased with the light/dark cycle frequencies. Optimum design and operational conditions which maximise the production of P. tricornutum biomass in outdoor helical reactors were determined.

Outdoor open thin-layer microalgal photobioreactor: potential productivity
We have previously estimated the productivity and photosynthetic efficiency of the microalga Chlorella sp. grown in an outdoor open thin-layer photobioreactor under climate conditions typical of the Middle European region, i.e. with many days unsuitable for intensive growth of algae (cloudy and rainy days, low air temperature, low solar PAR input). To estimate the real potential productivity of the bioreactor, we collected data on algae yields obtained during clear summer day periods. Cultivation was performed in fed-batch cycles in a bioreactor with a 224 m2 culture area (length 28 m, slope 1.7%), and a 6–7 mm-thick layer of algal culture. The suspension volume in the bioreactor was 2,000 L. The mean values found for Třeboň (49°N), Czech Republic, as an average of several sunny summer cultivation periods in July, were: net areal productivity, P net = 38.2 g dry weight (DW) m-2 day-1; net volumetric productivity, Pvol, = 4.3 g algal DW L-1 day-1, photosynthetic efficiency (based on PAR), ηnet = 7.05%. The peak values were: P net about 50 g (DW) m-2 day-1, ηnet about 9%. Algal growth rate was practically linear up to high biomass densities (40–50 g DW L-1, corresponding to an areal density of 240–300 g DW m-2), at which point the culture was harvested. The concentration of dissolved oxygen increased from about 10 mg L-1 at the beginning to about 23 mg L-1 at the end of culture area at noon. Use of the above-described technology for economical production of bioethanol is proposed.

High annual microalgae productivities can only be achieved if solar light is efficiently used through the different seasons. During winter the productivity is low because of the light and temperature conditions. The productivity and photosynthetic efficiency of Chlorella sorokiniana were assessed under the worst-case scenario found during winter time in Huelva, south of Spain. The maximum light intensity (800?μmol photons m-2 s-1) and temperature (20°C) during winter were simulated in a lab-scale photobioreactor with a short light-path of 14?mm. Chemostat conditions were applied and the results were compared with a temperature-controlled situation at 38°C (optimal growth temperature for C. sorokiniana). When temperature was optimal the highest productivity was found at a dilution rate of 0.18 h-1 (P v?=?0.28?g Kg-1 h-1), and the biomass yield on light energy was high (Y x,E?=?1.2?g?mol-1 photons supplied). However, at suboptimal temperature, the specific growth rate of C. sorokiniana was surprisingly low, not being able to support continuous operation at a dilution rate higher than 0.02 h-1. The slow metabolism under suboptimal temperature resulted in a decline of the light energy requirements of the cells. Consequently, the maximum winter irradiance was experienced as excessive, leading to a low photosynthetic efficiency and productivity (Y x,E?=?0.5?g mol-1 photons supplied, P v?=?0.1?g Kg-1 h-1). At suboptimal temperature a higher carotenoid-to-chlorophyll ratio was observed indicating the activation of light-dissipating processes. We conclude that temperature control and/or light dilution during winter time will enhance the productivity.

The biomass productivity of Scenedesmus obliquus was investigated outdoors during all seasons in solar tracked flat panel photobioreactors (PBR) to evaluate key parameters for process optimization. CO2 was supplied by flue gas from an attached combined block heat and power plant. Waste heat from the power plant was used to heat the culture during winter. The parameters pH, CO2, and inorganic salt concentrations were automatically adjusted to nonlimiting levels. The optimum biomass concentration increased directly with the photosynthetic active radiation (PAR) from 3 to 5 g dry weight (DW)?L?1 for a low PAR of 10 mol photons m?2 day?1 and high PAR of 40–60 mol photons m?2 day?1, respectively. The annual average biomass yield (photosynthetic efficiency) was 0.4?±?0.5 g DW mol?1 photons. However, biomass yields of 1.5 g DW mol?1 photons close to the theoretical maximum were obtained at low PAR. The productivity (including the night biomass losses) ranged during all seasons from ?5 up to 30 g DW m?2 day?1 with a mean productivity of 9?±?7 g DW m?2 day?1. Low night temperatures of the culture medium and elevated day temperatures to the species-specific optimum increased the productivity. Thus, continuous regulation of the biomass concentration and the culture temperature with regard to the fluctuating weather conditions is essential for process optimization of outdoor microalgal production systems in temperate climates.

Optimization of conditions for outdoor production of the nitrogen-fixing cyanobacterium Anabaena sp. ATCC 33047 has been pursued. In open ponds operated under semi-continuous regime biomass productivity values achieved ranged from 9 g (dry weight) m(-2) per day, in winter, to over 20 g m(-2) per day, in summer, provided that key operation parameters, including cell density, were optimized. Under these conditions the efficiency of solar energy conversion by the cells was fairly constant throughout the year, with photosynthetic efficiency values higher than 2%. The cyanobacterial biomass was rich in high-value phycobiliproteins, namely allophycocyanin and phycocyanin, for which open cultures of marine Anabaena represent a most interesting production system. The performance of Anabaena cultures operated under continuous regime in a closed tubular reactor has also been assessed outdoors, in winter. Biomass productivity values similar to those obtained in the ponds have been recorded for the closed system. Additionally, under these conditions, the cells excreted to the medium large amounts of a previously characterized exopolysaccharide, at production rates as high as 35 g m(-2) per day (1.4 g l(-1) per day). Properly operated closed cultures of this strain of Anabaena appear most suitable for outdoor mass production of valuable extracellular polysaccharides.

The light saturation effect imposes a serious limitation on the efficiency with which solar energy can be utilized in outdoor algal cultures. One solution proposed to reduce the intensity of incident solar radiation and overcome the light saturation effect is "spatial dilution of light" (i.e., distribution of the impinging photon flux on a greater photosynthetic surface area), but consistent experimental data supporting a significant positive influence of spatial light dilution on the productivity and the photosynthetic efficiency of outdoor algal cultures have never been reported. We used a coiled tubular reactor and compared a near-horizontal straight tubular reactor and a near-horizontal flat panel in outdoor cultivation of the cyanobacterium Arthrospira (Spirulina) platensis under defined operating conditions for optimum productivity. The photosynthetic efficiency achieved in the tubular systems was significantly higher because their curved surface "diluted" the impinging solar radiation and thus reduced the light saturation effect. This interpretation was supported by the results of experiments carried out in the laboratory under continuous artificial illumination using both a flat and a curved chamber reactor. The study also showed that, when the effect of light saturation is eliminated or reduced, productivity and solar irradiance are linearly correlated even at very high diurnal irradiance values, and supported findings that outdoor algal cultures are light-limited even during bright summer days. It was also observed that, besides improving the photosynthetic efficiency of the culture, spatial dilution of light also leads to higher growth rates and lowers the cellular content of accessory pigments; that is, it reduces mutual shading in the culture. The inadequacy of using volumetric productivity as the sole criterion for comparing reactors of different surface-to-volume ratio and of the areal productivity for evaluating the performance of elevated photobioreactors operated outdoors is stressed; it is furthermore suggested that the photosynthetic efficiency achieved by the culture also be calculated to provide a suitable parameter for comparison of different algal cultivation systems operated under similar climatic conditions.

Lutein production by Muriellopsis sp. in an outdoor tubular photobioreactor
The effect of dilution rate, mixing and daily solar cycles on lutein and biomass productivity of the green unicellular alga Muriellopsis sp. has been studied, throughout the year, in an outdoor tubular photobioreactor. Highest productivity values, for both lutein (about 180 mg m(-2) per day) and biomass (about 40 g (dry weight) m(-2) per day) were achieved on May and July. Values for the optimal dilution rate varied, being lower in May (0.06 h(-1)) than in November (0.09 h(-1)). Similar values for photosynthetic efficiency (about 4%) were recorded throughout the year, indicating that optimization of culture conditions was achieved for each experimental period. Along the daily solar cycle, there was a fast increase of lutein content of Muriellopsis sp. in response to irradiance during the early hours of daytime, with maximal lutein content (about 6 mg (g dry weight)(-1)) being recorded at noon, and decreasing slowly, thereafter. An increase in cell growth was observed following the establishment of maximum lutein/chlorophyll ratio, which might indicate a role for lutein in protecting cells from photodamage.

The photosynthetic oxygen evolution of Chlorella vulgaris (Beijer.) cells taken from phosphate-deficient (-P) and control cultures was measured during 8 days of culture growth. Under inorganic carbon concentration (50 microM) in the measuring cell suspension and irradiance (150 micromol m(-2) s(-1)), the same as during culture growth, there were no marked differences in the photosynthetic O2 evolution rate between the -P cells and the controls. The much slower growth of -P cultures indicated that the utilization of absorbed photosynthetically active radiation (PAR) in the CO2 assimilation and biomass production were in -P cells less efficient than in the controls. Alga cells under the phosphorus stress utilized more of the absorbed PAR in the nitrate reduction than the control cells. However, under conditions of more efficient CO2 supply (inorganic carbon concentration 150 microM, introducing of exogenous carbonic anhydrase to the measuring cell suspension) and under increased irradiance (500 micromol m(-2) s(-1)), the photosynthetic O2 evolution in -P cells reached a higher rate than in the controls. The results suggest that in -P cells the restricted CO2 availability limits the total photosynthetic process. But under conditions more favorable for the CO2 uptake and under high irradiance, the -P cells may reveal a higher photosynthetic oxygen evolution rate than the controls. It is concluded that an increased potential activity of the photosynthetic light energy absorption and conversion in the C. vulgaris cells from -P cultures is a sign of acclimation to phosphorus stress by a sun-type like adaptation response of the photosynthetic apparatus.

The feasibility of improving mass transfer characteristics of inclined tubular photobioreactors by installation of static mixers was investigated. The mass transfer characteristics of the tubular photobioreactor varied depending on the type (shape) and the number of static mixers. The volumetric oxygen transfer coefficient ( k(L)a) and gas hold up of the photobioreactor with internal static mixers were significantly higher than those of the photobioreactor without static mixers. The k(L)a and gas hold up increased with the number of static mixers but the mixing time became longer due to restricted liquid flow through the static mixers. By installing the static mixers, the liquid flow changed from plug flow to turbulent mixing so that cells were moved between the surface and bottom of the photobioreactor. In outdoor culture of Chlorella sorokiniana, the photobioreactor with static mixers gave higher biomass productivities irrespective of the standing biomass concentration and solar radiation. The effectiveness of the static mixers (average percentage increase in the productivities of the photobioreactor with static mixers over the productivities obtained without static mixers) was higher at higher standing biomass concentrations and on cloudy days (solar radiation below 6 MJ m(-2) day(-1)).

To be able to study the effect of mixing as well as any other parameter on productivity of algal cultures, we designed a lab‐scale photobioreactor in which a short light path (SLP) of (12 mm) is combined with controlled mixing and aeration. Mixing is provided by rotating an inner tube in the cylindrical cultivation vessel creating Taylor vortex flow and as such mixing can be uncoupled from aeration. Gas exchange is monitored on‐line to gain insight in growth and productivity. The maximal productivity, hence photosynthetic efficiency, of Chlorella sorokiniana cultures at high light intensities (1,500 μmol m?1 s?1) was investigated in this Taylor vortex flow SLP photobioreactor. We performed duplicate batch experiments at three different mixing rates: 70, 110, and 140 rpm, all in the turbulent Taylor vortex flow regime. For the mixing rate of 140 rpm, we calculated a quantum requirement for oxygen evolution of 21.2 mol PAR photons per mol O2 and a yield of biomass on light energy of 0.8 g biomass per mol PAR photons. The maximal photosynthetic efficiency was found at relatively low biomass densities (2.3 g L?1) at which light was just attenuated before reaching the rear of the culture. When increasing the mixing rate twofold, we only found a small increase in productivity. On the basis of these results, we conclude that the maximal productivity and photosynthetic efficiency for C. sorokiniana can be found at that biomass concentration where no significant dark zone can develop and that the influence of mixing‐induced light/dark fluctuations is marginal. © 2010 American Institute of Chemical Engineers Biotechnol.
