Abstract SDS-PAGE and zymographic analysis of protein extracts from Bacillus subtilis AN8, which is deficient in the major 50-kDa amidase (CwlB[LytC]), revealed another distinct but relatively weak 50-kDa protein and its strong activity band. As well as the 50-kDa protein (designated as CwIE), a 35-kDa protein (designated as CwlF) and its activity were also found. In contrast to CwlE production, CwlF production was unaffected by a flaDl ( sinR ) point mutation which represses other vegetative phase autolysins. These newly identified autolysin activities quickly disappeared when cell growth entered stationary phase. The introduction of a sigD -null mutation caused the disappearance of Cw1E activity but Cw1F activity was unaffected by the mutation, as judged on zymography. The possible roles of CwlE and CwlF during vegetative growth are discussed.  相似文献

The small acid-soluble spore proteins alpha and beta were not detected during stationary-phase growth of asporogenous Bacillus subtilis mutants blocked in stages 0, II, or III, but mutants blocked in stages IV or V accumulated nearly wild-type levels of these small acid-soluble spore proteins. Similar results were obtained when production of Bacillus megaterium C protein (also a small acid-soluble spore protein), as well as alpha and beta, were monitored in these mutants containing a recombinant plasmid carrying the B. megaterium C protein gene. The only exception was a spo0H mutant which synthesized a small amount of C protein, but no alpha or beta.  相似文献

Peptidoglycan hydrolase, LytF (CwlE), was determined to be identical to YhdD (deduced cell wall binding protein) by zymography after insertional inactivation of the yhdD gene. YhdD exhibits high sequence similarity with CwlF (PapQ, LytE) and p60 of Listeria monocytogenes. The N-terminal region of YhdD has a signal sequence followed by five tandem repeated regions containing polyserine residues. The C-terminal region corresponds to the catalytic domain, because a truncated protein without the N-terminal region retained cell wall hydrolase activity. The histidine-tagged LytF protein produced in Escherichia coli cells hydrolyzed the linkage of D-gamma-glutamyl-meso-diaminopimelic acid in murein peptides, indicating that it is a D,L-endopeptidase. Northern hybridization and primer extension analyses indicated that the lytF gene was transcribed by EsigmaD RNA polymerase. Disruption of lytF led to slightly filamentous cells, and a lytF cwlF double mutant exhibited extraordinary microfiber formation, which is similar to the cell morphology of the cwlF sigD mutant.  相似文献

The change of motility and the presence of flagella were followed throughout growth and sporulation in a standard sporulating strain and in 19 cacogenic sporulation mutants of Bacillus subtilis. For the standard strain, the fraction of motile cells decreased during the developmental period to less than 10% at T4. Motility was lost well before the cells lose their flagella. Conditions reducing the decrease of motility also reduced sporulation: motile cells never contained spores. The decrease of motility was not coupled with a decrease in the cellular concentration of adenosine 5'-triphosphate or a decline in oxygen consumption, but an uncoupling agent immediately destroyed motility at any time. Apparently, motility decreased during development because it became increasingly uncoupled from the energy generating systems of the cell. The motility of sporulation mutants decreased after the end of growth at the same time as or earlier than the motility of the standard strain; the early decrease of motility in an aconitase mutant, but not that in an alpha-ketoglurate dehydrogenase mutant, could be avoided by addition of L-glutamate. Sporulation or related events such as extracellular antibiotic or protease production were not needed for the motility decline.  相似文献

DNA-dependent RNA polymerase has been purified from Bacillus subtilis at various stages of vegetative cell growth. Polymerase isolated from cultures approaching the end of the logarithmic growth phase was associated with a 60,000-dalton polypeptide and was only 10–20% as active as polymerase isolated from rapidly growing cells. Appearance of this new polypeptide and the change in template activity occur prior to stage 0 of sporulation.  相似文献

The autolysins of Bacillus subtilis 168 were analyzed by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis with substrate-containing gels. Four bands of vegetative autolytic activity of 90, 50, 34, and 30 kDa (bands A1 to A4) were detected in SDS and LiCl extracts and in native cell walls by using B. subtilis 168 vegetative cell walls as the substrate incorporated in the gel. The four enzyme activities showed different substrate specificities and sensitivities to various chemical treatments. The autolysin profile was not medium dependent and remained constant during vegetative growth. During sporulation, band A4 greatly increased in activity just prior to mother-cell lysis. No germination-associated changes in the profile were observed, although a soluble 41-kDa endospore-associated cortex-lytic enzyme was found. By using insertionally inactivated mutants, bands A1 and A2 were positively identified as the previously characterized 90-kDa glucosaminidase and 50-kDa amidase, respectively. The common filamentous phenotype of various regulatory mutants could not be correlated to specific changes in the autolysin profile.  相似文献

枯草芽胞杆菌Bacillus subtilis常被用于表达杀虫和抗菌蛋白.为了探讨苏云金芽胞杆菌B. thuringiensis营养期杀虫蛋白基因(vip3A)在枯草芽胞杆菌中的表达情况,促进杀虫防病工程菌构建,将枯草芽胞杆菌168菌株核糖体小亚基S4蛋白基因的启动子与苏云金芽胞杆菌WB7菌株vip3A基因的编码序列连接,插入大肠杆菌Escherichia coli与枯草芽胞杆菌穿梭载体pAD123,得到重组原核表达质粒pADpvip,将重组质粒转化枯草芽胞杆菌标准菌株168和分离自辣椒体内的生防内生枯草芽胞杆菌BS-2菌株中,获得工程菌株.SDS-PAGE分析表明在枯草芽胞杆菌168菌株的部分工程菌株中有约88 kDa大小的VIP条带,而BS-2的工程菌株中未见相应的条带,表明Vip3A蛋白仅在168菌株中表达.生物测定表明有5株168的工程菌株(168vip1-4,6)表现较高的杀虫活性,工程菌株发酵稀释液(约107CFU/mL)处理的小白菜叶片饲喂斜纹夜蛾2龄幼虫72 h的杀虫效果可达87.64%~92.13%,但vip3A基因转入内生枯草芽胞杆菌BS-2中不表现杀虫作用.毒力测定表明168vip2菌株对斜纹夜蛾2龄幼虫72 h的LC50为0.0194 mL/mL.这些结果为进一步研究基因在枯草芽胞杆菌中的表达构建杀虫防病工程菌打下了基础.  相似文献

The gene for the Bacillus megaterium spore C protein, a sporulation-specific gene, has been transferred into Bacillus subtilis. The B. megaterium gene was expressed little, if at all, during log-phase and early-stationary-phase growth, but was expressed during sporulation with the same kinetics as and at a level similar to that of the analogous B. subtilis genes. This finding is most consistent with the regulation of this class of genes by a mechanism of positive control.  相似文献

A simple experimental system for detection of sporulation promoting factors was presented. This system showed that there was a sporulation promoting factor in the vegetative cells of Bacillus subtilis cultivated on nutrient agar for 9 hr (at stage T0). The factor was partially purified from the sonicate of vegetative cells by ethanol fractionation, gel filtration, chromatography and preparative gel electrophoresis, and it was identified as manganese-containing protein.  相似文献

Four major heat-shock proteins (hsps) with apparent molecular masses of 84, 69, 32 and 22 kDa were detected in exponentially growing stationary phase and sporulating cells of Bacillus subtilis heat-shocked from 30 to 43 degrees C. The most abundant, hsp69, is probably analogous to the E. coli groEL protein. These proteins were transiently inducible by heat-shock. Partial purification of RNA polymerase revealed several other minor hsps. One of these, a 48 kDa polypeptide probably corresponds to sigma 43. The synthesis of this polypeptide and at least two other proteins appeared to be under sporulation and heat-shock regulation and was affected by the SpoOA mutation.  相似文献

The steady-state concentration of cell wall turnover products in the medium of Bacillus subtilis 168 growing exponentially on a casein hydrolysate-supplemented medium is equivalent to an overall rate of turnover of less than 10% per generation. After transfer of a steady-labeled culture to nonradioactive medium, the rate of release of labeled turnover products increased exponentially for up to two generations. The rate of turnover finally attained by this culture reached an apparently first-order rate of about 50% per generation. The addition of soluble autolytic activity to growing cultures of a mutant possessing a reduced rate of wall turnover resulted in a marked stimulation in the rate of solubilization of the cell wall fraction. The increased rate of solubilization produced was proportional to the concentration of added enzyme and remained constant until less than 20% of the wall originally present was left. Autolytic activity added under these conditions was bound entirely to wall at least one generation old. The results are interpreted in terms of a model for cell wall growth in which wall two or more generations old covers a total surface area at least four times larger than that occupied at the time of synthesis, forming a shallow outer layer (overlying newer wall) from which all turnover takes place. The model is discussed in relation to previous attempts to determine the pattern of surface expansion in bacilli.  相似文献
