研究了碳源、氮源对彩绒革盖菌液体发酵合成漆酶的影响。结果表明,在所选的几种碳、氮源中,麸皮为试验菌株合成漆酶的较好碳源;酵母浸膏、酒石酸铵、蛋白胨均是比较理想的氮源。不同的碳氮比对彩绒革盖菌漆酶的产量有着显著的影响,高碳高氮是其生产漆酶的最佳条件。在适宜的培养条件下该菌株能合成高活力的漆酶,其酶活力可达298 U/mL以上,产酶周期为6~7 d。  相似文献

It has been shown that the wood-rotting mushroom Pleurotus dryinus IBB 903 is able to effectively produce cellulases, xylanase, laccase, and manganese peroxidase in submerged fermentation of mandarin peels and tree leaves. Gradual increasing of lignocellulosic substrates concentration from 1 to 4–6% enhanced enzyme accumulation in culture liquid. A simple and inexpensive medium containing mandarin peels and yeast extract as sole carbon and nitrogen sources allowed simultaneous production of high levels of both hydrolases and oxidases by P. dryinus IBB 903. Supplementation of this medium by copper and manganese caused earlier and faster accumulation of laccase and manganese peroxidase increasing their yield by 1.5 and 7.5 times, respectively. In addition, by adding manganese to the medium it is possible to regulate the ratio of laccase and MnP in enzyme preparation. The presence of lignocellulosic substrate is the requisite for MnP production by P. dryinus IBB 903 since there was no production of MnP when mushroom has been cultivated in the synthetic medium with different carbon source. Among carbon source tested only utilization of glucose resulted to 21-fold increase of fungus laccase specific activity compared to control medium without carbon source. The effect of corn oil addition on mycelium growth and polysaccharide productions in the medicinal mushroom Ganoderma lucidum was studied. The results showed that when a level of 2% corn oil was added at the beginning of culture, the biomass and polysaccharide productions reached a maximum of 12.9 and 1.038 g/L, respectively, during 13-day cultivation. The pH variation along with morphology observation in culture provided an indirect inference to the promotional effect of oil addition. Moreover, a curve fitting analysis was carried out to assay the elevated effect on biomass and exopolysaccharide productions in oil added culture. The experimental data of substrates consumption and products formation in culture with oil addition were predicted through the fitting equations obtained in single carbon source culture. The numerical results further clarified the stimulatory effects of oil addition in G. lucidum culture.  相似文献

The mineral Kissiris, which is formed from the thickened foam of volcanic lava, was tested to approximate its mineral composition using energy-dispersive X-ray (EDX) analysis. The solid mineral contains silicon dioxide at about 16 (w/w). The considerable surface roughness of Kissiris along with its extensive porosity made this natural solid cell support an attractive candidate for manganese peroxidase (MnP) production for synthetic dye decolourization, at low cost. The white rot fungus Phanerochaete chrysosporium immobilized on the mineral Kissiris was grown in both stationary and agitated cultures (rotary shaker, 100 rev/min) using either carbon- or nitrogen-limited growth medium to study the ability of the fungus to degrade the synthetic dye methylene blue (MB). The value of residual dye for MB used at 60 ppm was 6% within 8 days of the incubation of the nitrogen-limited culture under the shaken conditions. Production of (MnP) occurred simultaneously in nitrogen-limited culture medium with the added MnSO4 at 100 ppm. The MnP activity was at relatively high level (170 U/l).  相似文献

To screen stimulators from Chinese medicinal insects for mycelial growth and polysaccharides production of Ganoderma lucidum, G. lucidum was inoculated into the media with and without supplementation of medicinal insect extracts. The ethyl acetate extract of Eupolyphaga sinensis at 55 mg l−1 lead to significant increase in both biomass and intracellular polysaccharides (IPS) concentration from 8.53 ± 0.41 to 14.16 ± 0.43 and 1.28 ± 0.09 to 2.13 ± 0.11 g l−1, respectively. In addition, the ethyl acetate extract of Catharsius molossus at 55 mg l−1 significantly enhanced extracellular polysaccharides (EPS) production; the EPS yield increased from 350.9 ± 14.1 to 475.1 ± 15.3 mg l−1. There were no new components in the two types of polysaccharides obtained by the addition of the insect extracts.  相似文献

Enzymes produced by Ganoderma australe in solid-state fermentation and submerged cultures were evaluated. Strain A464 produced laccase activity in liquid medium and in solid-state cultures containing Drimys winteri or Eucalyptus globulus wood chips, while MnP and LiP activities were not detected. On the other hand, strain A272 cultured for 75 days on E. globulus presented MnP activity of 719 IU/kg of wood. The suitability of D. winteri wood as a substrate enabling MnP production was checked with a well-documented MnP-producing basidiomycete, Ceriporiopsis subvermispora, which produced MnP activity of 327 IU/kg of wood in 9-day-old cultures. Data from two different G. australe strains (A272 and A464) indicated that MnP secretion depended on strain origin as well as on culture conditions.  相似文献

Exopolysaccharide (EPS) production and biosynthesis were studied in Ganoderma lucidum, a fungus used in traditional Chinese medicine, grown with lactose in a bioreactor. -Galactosidase activity, which implies the existence of a lactose permease system, was induced by lactose. Lactose feeding also increased -phosphoglucomutase activity and EPS accumulation but decreased phosphoglucose isomerase activity and lactate concentration in the culture broth. A maximum cell density of 22 g l–1 and EPS at 1.25 g l–1 were obtained in fed-batch bioreactor culture.  相似文献

The synthetic characteristics of the extracellular polysaccharide (EPS) of Ganoderma lucidum in batch fermentation were studied. The result showed that the production of EPS was partially growth-associated. The cell dry weight (CDW) and EPS reached 15.56 g·L−1 and 3.02 g·L−1, respectively. The yield of EPS to cell dry weight (Yp/x) was 0.19. On the basis of the test results of batch fermentation, a kinetic model was proposed by using the Logistic equation for cell growth, the Luedeking–Piret equation for EPS production, and the Luedeking-piret-like equation for the consumption of glucose as substrate. The calculated results using these models were satisfactorily compared with the experimental data under various concentrations of glucose, and the average of relative errors was found to be not more than 5%. The kinetic model had practical guidance interesting in producing PES by Ganoderma lucidum.  相似文献

Summary The effect of various carbon and nitrogen sources on laccase, manganese-dependent peroxidase (MnP), and peroxidase production by two strains of Pleurotus ostreatus was investigated. The maximal laccase yield of P. ostreatus 98 and P. ostreatus 108 varied depending upon the carbon source from 5 to 62 U l−1 and from 55 to 390 U l−1, respectively. The highest MnP and peroxidase activities were revealed in medium supplemented by xylan. Laccase, MnP, and peroxidase activities of mushrooms decreased with supplementation of defined medium by inorganic nitrogen sources. Peptone followed by casein hydrolysate appeared to be the best nitrogen sources for laccase accumulation by both fungi. However, their positive effects on enzyme accumulation were due to a higher biomass production. The secretion of MnP and peroxidase by P. ostreatus 108 was stimulated with supplementation of casein hydrolysate to the control medium since the specific MnP and peroxidase activities increased 15-fold and 3.5-fold, respectively.  相似文献

The effects of different inoculum-loading rates and pre-treatment of wheat straw with formic acid and hot water (50 °C) on the establishment of Phanerochaete chrysosporium on unsterile straw were studied in laboratory scale and in a 1.5-m3 bioreactor. The establishment of P. chrysosporium on unsterile straw was satisfactory. Phanerochaete chrysosporium and other fungi, which developed simultaneously, were able to produce the activity necessary to degrade two herbicides, bentazon and MCPA (4-chloro-2-methylphenoxyacetic acid) in 20 days (65 and 75%, respectively). The decrease of both herbicides coincided with the presence of the activity of the lignin-degrading enzymes lignin peroxidase and manganese peroxidase/laccase. Extensive growth of P. chrysosporium or other lignin-degrading fungi on unsterile straw would be excellent for inexpensive solid substrate systems intended for degradation of pesticides.  相似文献

The fruiting bodies, spores, and lipid from the spores of Ganoderma lucidum have been widely used for medicinal purpose in China. Ergosterol content may be a suitable marker for evaluating the quality of ganoderma spore and ganoderma spore lipid (GSL) products. A gradient reversed-phase high-performance liquid chromatography method was developed for the simultaneous determination of free and esterified ergosterols in G. lucidum. The contents of free and esterified ergosterols in the different parts (the stipe, pileus, tubes, and spores) of G. lucidum and GSL were determined. The results showed that total ergosterol levels in the stipe, pileus, tubes, and spores of G. lucidum were between 0.8 and 1.6 mg/g. The relative abundances of free to esterified ergosterol were different in the different parts of G. lucidum. The spores and the tubes, the hymenophore tissue that contains the spore-producing cells, have a considerably higher percentage of ergosteryl esters (41.9 and 39.7% of total ergosterol) in comparison with the pileus and stipe tissues (3.6 and 6.2%).  相似文献

Summary Candida stellata is a wine-yeast species that has been proposed for use as a starter in the wine industry in combination with selected Saccharomyces cerevisiae cultures. To improve our knowledge on the metabolic interactions between these two yeast species, we have investigated the influence of temperature and oxygen concentration on their fermentation behaviour in mixed fermentation. Trials were carried out with free and immobilized C. stellata cells followed by an inoculum of a commercial strain of S. cerevisiae, to determine the biomass evolution and the oenological profiles of the resulting wines. We show that the oxygen concentration does not influence the fermentation behaviour of the C. stellata cells, while a low temperature (16 °C) is a critical condition for the metabolic activity of C. stellata in free-cell trials. The immobilization procedures produce a significant improvement in the metabolic activity of C. stellata, with a consequent enhancement of the glycerol content also at the 16 °C fermentation temperature. High cell concentrations and the immobilization system appear to positively influence the fermentation behaviour of C. stellata. These results are of interest for the practical application of this process in winemaking.  相似文献

The effect of foliar-applied glycinebetaine (GB) on chloroplast ultrastructure, the amount of chlorophyll and proteins and ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) activity in stressed tomato leaves were analysed. Initial, and total activity and activation state state of RuBPCO were also measured. RuBPCO activities, chloroplast area, and the number of plastoglobuli remained unaffected, while the relative area of starch granules increased in GB-treated, salt-stressed tomato leaflets. Under drought-stress, the relative area of plastoglobuli increased upon GB application. The primary effect of GB was, however, the increased protein and chlorophyll content.  相似文献

The effect of different carbon and nitrogen sources on the production of toxin by Clostridium argentinense was examined. The toxin production by C. argentinense in coculture with Pseudomonas mendocina increased in all the cases in relation to that produced by monocultures independent of the nature of the source. Using dextrin as carbon source C. argentinense produced the highest levels of toxin both in monocultures (300 LD50/mL) and in cocultures with P. mendocina (5000 LD50/mL). Experiments run in a microfermenter showed that the slow growth of cocultures associated with the assimilation of dextrin and the pH and Eh profiles favoured the production of toxin. Of the nitrogen sources assayed, corn steep liquor sustained the highest levels of toxin in both monocultures and cocultures with 3 and 2.8 fold increases with respect to that obtained using proteose peptone. The toxin production by C. argentinense cultures and C. argentinenseP. mendocina cocultures was highly dependent on the nature of the carbon and nitrogen sources used in the culture media. Growth of C. argentinense on substrates slowly assimilated stimulated the production of toxin.  相似文献

Activity of antioxidant enzymes was evaluated during somatic embryogenesis and shoot organogenesis from cultured leaf segments of Gladiolus hybridus Hort. The effect of exogenous antioxidants on somatic embryogenesis and shoot organogenesis has also been monitored. Activity of superoxide dismutase (SOD) gradually increased during somatic embryogenesis. while activities of catalase (CAT) and peroxidase (POX) decreased. In contrast, increase in CAT and POX activity and a concomitant decrease in SOD activity were noted during shoot organogenesis. Exogenous application of antioxidants such as glutathione (GSH), α-tocopherol and ascorbate (AA) inhibited somatic embryogenesis but stimulated shoot organogenesis. The frequency of somatic embryogenesis increased with the addition of H2O2. However, H2O2 inhibited shoot organogenesis. The enzyme activities of Clostridium La 1 and Clostridium kluyveri involved in the stereospecific hydrogenation of ,-unsaturated carbonyl compounds with hydrogen gas were measured. In C. La 1 the specific activities of hydrogenase and enoate reductase depended heavily on the growth phase and the composition of the medium. During growth in batch cultures on 70 mM crotonate the specific activity of hydrogenase increased and then dropped to about 10% of its maximum value, whereas the activity of enoate reductase reached its maximum in cells of the stationary phase. Under certain conditions during growth the activity ratio hydrogenase: enoate reductase changed from 120 to 1. Thus, the rate limiting enzyme for the hydrogenation can be either the hydrogenase or the enoate reductase, depending on the growth conditions of the cells.The specific activities of ferredoxin-NAD reductase and butyryl-CoA dehydrogenase increased 3-4-fold during growth on crotonate. By turbidostatic experiments it was shown that at constant input of high crotonate concentrations (200 mM) the enoate reductase activity was almost completely suppressed; it increased steadily with decreasing crotonate down to an input concentration of 35 mM.Glucose as carbon source led to high hydrogenase and negligible enoate reductase activities. The latter could be induced by changing the carbon source of the medium from glucose to crotonate. Tetracycline inhibited the formation of enoate reductase.A series of other carbon sources was tested. They can be divided into ones which result in high hydrogenase and rather low enoate reductase activities and others which cause the reverse effect.When the Fe2+ concentration in crotonate medium was growth limiting, cells with relatively high hydrogenase activity and very low enoate reductase activity in the stationary phase were obtained. At Fe2+ concentrations above 3·10-7 M enoate reductase increased and hydrogenase activity reached its minimum. The ratio of activities changes by a factor of about 200. 为探讨花榈木体胚发生过程中不同碳氮源处理对胚性愈伤组织诱导、发育和有机物积累的影响,并筛选出有利于花榈木体胚发生的碳氮源,优化体胚发生体系,该研究以成熟胚为外植体,通过单因素试验分析3种碳源、4种蔗糖浓度和6种氮源处理下胚性愈伤组织诱导、发育和有机物积累的差异。结果表明:(1)蔗糖中胚性愈伤组织诱导率显著高于葡萄糖和麦芽糖,但其体胚诱导率、体胚分化率、胚性愈伤组织可溶性糖、淀粉和可溶性蛋白含量差异不显著。(2)随着蔗糖浓度的升高,胚性愈伤组织、体细胞胚(体胚)诱导率、体胚分化率、胚性愈伤组织重量和可溶性蛋白含量呈先升高后降低的趋势,均以添加30 g·L-1蔗糖最高,而胚性愈伤组织可溶性糖和淀粉含量呈增加的趋势。(3)在6种氮源处理中,胚性愈伤组织诱导率以添加500 mg·L-1谷氨酰胺的处理最高,体胚诱导率则以添加谷氨酰胺和水解酪蛋白的处理较高,但不同氮源处理间体胚分化率无差异;添加有机氮源的处理其胚性愈伤组织可溶性蛋白含量显著高于无氮源处理。总之,不同的碳氮源通过影响花榈木胚性愈伤组织的诱导、发育和有机物的积累,从而影响其体胚诱导率,但对体...  相似文献

Essentiality of selenium (Se) for Japanese quail,Coturnix coturnix japonica, was confirmed using a formulated semipurified low-Se diet (basal) (0.05 ppm). Selenium-deficiency symptoms appeared in quails on this diet within 15 d, which corresponded to low levels of hemolysate glutathione peroxidase (GSH-Px) activity. Selenium administration at 0.05 and 2.0 ppm levels resulted in an increase of hemolysate GSH-Px activity by 64 and 116%, respectively, in both short- and long-term experiments. Growth over a 2-mo period increased the hemolysate GSH-Px activity by 120% at each level of dietary Se. A differential response was exhibited by hepatic mitochondrial and soluble GSH-Px activity to Se supplementation, the former increasing progressively with increments of Se at 0.05, 2.0, and 4.0 ppm by 45, 70 and 150%, respectively. The soluble GSH-Px activities of tissues, such as liver, kidney, and testis, and RBC membrane-bound activity remained unchanged in long-term studies at different levels of Se. Replenishment of Se to quails maintained on low-Se diets reflected no change in RBC membrane-bound and liver-soluble GSH-Px activities, although the activity in hemolysate increased consistently with Se. The GSH-Px activity in hemolysate was restored to the levels comparable to those of long-term studies only at Se administration at the 2.0-ppm level. The differential response of mitochondrial and soluble GSH-Px activities to Se and other related observations on mitochondrial functions suggest an additional role for Se in mitochondrial membrane processes and glutathione-related metabolic regulations.  相似文献
