Synopsis Gastric evacuation rates (R) of white perch,Morone americana, were determined in laboratory experiments and by using field data. The resulting relationship ofR and temperature (T) for white perch wasR = 0.028e0.106T (r2 = 0.98). The high r2 of the regression indicates good agreement of the combined laboratory and field data. Our rate compares well with those available for other species; and especially for Eurasian perch, which has a similar thermal existence to white perch in Lake Erie.  相似文献

Summary R st is an unstable allele of R that exhibits a mosaic phenotype in the aleurone, consisting of heavily pigmented spots on a colorless background. This variagated phenotype is presumably caused by the frequent somatic reversion of R from an inactive to an active form. Data are presented showing that such a reversion can take place at different times during the plant ontogenesis. Various stippled derivatives have been selected that differ in the number and size of dots formed in the endosperm. The reversion rate of R st and derivatives toward self-colored (R sc) has been estimated in the germinal and somatic tissues. This analysis indicates that some of the stippled derivatives differ in their capacity to revert toward R sc in both the somatic and germ cells. The effect of internal factors on the R st reversion rate has been measured. On the basis of these data the possible mechanism causing genetic instability of R st are briefly discussed.This investigation has been partially supported by Consiglio Nazionale delle Ricerche, Roma.  相似文献

A total of 562 individuals living in four villages of two Brazilian Indian tribes (Cayapo and Krahó) was studied in relation to blood groups ABO, MNSs, P, Rh, Lewis, Duffy, Kidd and Diego; haptoglobin, Gc, acid phosphatase and phosphoglucomutase types. These results were compared with those obtained previously among the Xavante, and the inhabitants of three other Cayapo villages, all of whom speak Ge languages; the ranges in gene frequencies observed in a representative series of South American Indians from all over the continent were also compiled. The Ge Indians are characterized by low frequencies ofRz, medium frequencies ofR1,R2, R0, orr,Jka andPGM11, and high frequencies ofGc2 andACPA when compared with other South American tribes. Genetic distance analyses based on six loci indicate that the intratribal variability observed among Cayapo is of the same order of magnitude as those obtained among the Xavante and Krahó, being much less pronounced than those observed among the Yanomama and Makiritare. The intertribal differences within this linguistic group are much less pronounced than those encountered among tribes that speak more differentiated languages.  相似文献

Thermal requirements for development and life table statistics ofAphis gossypii Glover (Homoptera, Aphididae) were determined over a range of constant temperatures from 10 to 30°C. The lower development threshold and the sum of effective temperatures were 6.9°C and 90.1°C, respectively, for preimaginal development, and 5.8°C and 113.6°C from birth to the onset of reproduction. Mean total fecundity ranged from 36 larvae per female at 10°C to 76 larvae at 30°C. On a time scale of days, net reproductive rate (R o ) increased with increasing temperature while generation time (T) decreased causing the intrinsic rate of increase (r m ) to increase linearly from 0.115 to 0.465. On a day-degree scaler m only varied from 0.019 to 0.028 because the growth ofR o was compensated by an increase inT with increase in temperature. The nearly constantr m in terms of day-degrees, over a wide range of temperatures, greatly simplifies the prediction of future population numbers ofA. gossypii.  相似文献

Five isolates ofCladorrhinum foecundissimum, added to soilless mix as 10-day-old fresh bran preparations (1.0% w/w), significantly reduced (P≤ 0.05) damping-off of eggplant and pepper caused byRhizoctonia solanistrain R-23. After 4 weeks of growth, plant stands in the biocontrol-amended, pathogen-infested treatments (>80%) were comparable to those in the noninfested controls. Since plant stands were similar at 2 and 4 weeks, most of the disease was preemergence damping-off. The bran preparations also reduced saprophytic growth of the pathogen, and there was an inverse correlation (r2= −0.94) between saprophytic growth and eggplant stand. Added to soilless mix at a rate of 2.0% (w/w), alginate prill containing 20% fermentor-produced biomass of six biocontrol isolates ofC. foecundissimumreduced (P≤ 0.05) damping-off of eggplant caused byR. solani, but only the prill with biomass of isolates Cf-1 or Cf-2 yielded plant stands (>80%) comparable to that in the noninfested control. As with the bran preparations, there was also an inverse correlation (r2= −0.80) between saprophytic growth of R-23 and eggplant stand with the alginate prills. Alginate prill with biomass of Cf-1 or Cf-2 also reduced (P≤ 0.05) damping-off of eggplant and pepper caused by other isolates (195, NG-2, DPR-1) ofR. solani, but only the stands (>80%) of pepper were similar to that in the noninfested control. Alginate prill formulations ofC. foecundissimum(Cf-1, Cf-2, and Cf-3) also reduced (P≤ 0.05) populations of the pathogen and damping-off of eggplant and pepper caused byPythium ultimum(PuZ3). However, although the plant stands in the treatments were not as high as those in the noninfested controls, they were higher than those in the pathogen-infested controls. The treatments also reduced populations ofP. ultimumin the soilless mix so that there were inverse correlations between the pathogen population and eggplant stand (r2= −0.81) and pepper stand (r2= −0.78). Extruded flour/clay granules containing 5.0% biomass of Cf-1 and Cf-2, added toR. solani-infested soilless mix (2.0%), reduced (P≤ 0.05) damping-off of eggplant and pepper. However, only the Cf-2 treatments resulted in stands (>80%) equal to those in the noninfested controls for the crops after 4 weeks of growth. The influence of bran and alginate prill of Cf-1 or Cf-2 on the spatial spread ofR. solaniand its ability to incite damping-off of eggplant showed that prill with Cf-1 or Cf-2 and bran with Cf-2 were equally effective in reducing the spread of the pathogen from the point source of the inoculum to the center of the flats.  相似文献

The mechanism of the novel enzymatic dehalogenating action of (R)-3-chloro-1,2-propanediol [monochlorohydrin (MCH)] was investigated. The (R)-MCH-dechlorinating enzyme system ofAlcaligenes sp. DS-S-7G, which stereoselectively assimilated (R)-MCH from the racemate, was composed of two components (Enzyme 1 and Enzyme 2). Enzyme 1 was a flavoprotein with a relative molecular mass (M r) of 70,000 and was composed of two kinds of polypeptides (58,000 and 16,000). The enzyme exhibited activity for converting (R)-MCH to hydroxyacetone with the liberation of chloride ions under aerobic conditions. On the other hand, Enzyme 2 with anM r of 86,000, which was also composed of two kinds of polypeptides (33,000 and 53,000), showed no dechlorinating activity for (R)-MCH. However, in the presence of NAD+, when Enzyme 1 was conjugated with Enzyme 2 in the (R)-MCH-dechlorinating reaction, the co-operative dechlorinating activity was four to five times higher than that by Enzyme 1 alone. (R)-MCH was finally degraded to acetic acid and formic acid by the joint action of the two enzymes. These facts indicate that (R)-MCH is oxidatively dechlorinated by the two enzymes in the presence of NAD+ inAlcaligenes sp. DS-S-7G.  相似文献

Using DNA clones, the physical distance between the linked genesnov andstr inHaemophilus influenzae was estimated. Although none of the cloned inserts contained both the markers, pJ1-8StrR 13 (insert of 18·7 kb) includedstr gene at one end and part ofnov gene at the other end of the insert. By EcoRI restriction analysis and by Southern hybridization, the distance between the two EcoRI sites, cutting at which inactivates the two genes, was estimated to be 17·7 kb. A single continuous EcoRI fragment (containing 4EcoRI sites within it) carrying both the genes intact would need to be 20·4 kb in size. These estimates were confirmed independently using different clones ofnov r andstr r alleles as probes for hybridization with BamHI-digested chromosomal DNA.  相似文献

Several reproductive triats in plants were studied in more than 200 populations of 61 wild species from diverse ecological conditions. As a result, it was found that there occur three distinct types of plants in the energy allocation patterns to reproductive structures (RA) and the propagule output per plant (PN), i.e. (1) the number of propagules per plant increases in response to the increase in RA (Type I), (2) the number of propagules decreases in response to the increase in RA (Type II), and (3) the RA remains constant despite the great differences in the propagule number per plant. A conspicuous trade-off relationship was also discovered to occur between the RA to a single propagule (RA) and the propagule output per plant (PN), such that log RA=logC−blot PN, or RA=C/PN b =CPN b , where C is a constant. The three different ranges ofb-values were recognized, i.e.b<1.0,b>1.0, andb=1.0, which correspond to Type I, Type II, and Type III, respectively. Related problems to the concept ofr- andK-strategy are also discussed.  相似文献

Aureobasidin A (AbA), a cyclic depsipeptide produced byAureobasidium pullulans R106, is highly toxic to fungi includingSaccharomyces cerevisiae. We isolated several dominant mutants ofS. cerevisiae which are resistant to more than 25 µg/ml of AbA. From a genomic library of one suchAUR1 mutant, theAUR1 R (foraureobasidinresistant) mutant gene was isolated as a gene that confers resistance to AbA on wild-type cells. Its nucleotide sequence showed that the predicted polypeptide is a hydrophobic protein composed of 401 amino acids, which contains several possible transmembrane domains and at least one predicted N-linked glycosylation site. Comparison of the mutant gene with the wild-typeaur1 + gene revealed that the substitution of Phe at position 158 by Tyr is responsible for acquisition of AbA resistance. We suggest that the gene product of the wild-typeaur1 + is a target for AbA on the basis of following results. Firstly, cells that overexpress the wild-typeaur1 + gene become resistant to AbA, just as cells with anAUR1 R mutation do. Secondly, disruption of theaur1 + gene demonstrated that it is essential for growth. Thirdly, in the cells with a disruptedaur1 locus, pleiotropic morphological changes including disappearance of microtubules, degradation of tubulin and abnormal deposition of chitin were observed. Some of these abnormalities are also observed when wild-type cells are treated with AbA. The abnormality in microtubules suggests that the Aur1 protein is involved in microtubule organization and stabilization.  相似文献

Knowledge of root respiration is a prerequisite for a better understanding of ecosystem carbon budget and carbon allocation. However, there are not many relevant data in the literature on direct measurements of in situ root respiration by root chamber method. Furthermore, few studies have been focused on the effects of root diameter (D r) and root nitrogen concentration (N r) on in situ root respiration among different seasons and tree species. To address these goals, we used a simplified root-chamber system to measure in situ root respiration rates of Acacia crassicarpa and Eucalyptus urophylla in subtropical plantations of south China. We found that the species and season variation in root respiration were affected by D r and N r. Also, the root respiration per unit dry mass (R r, nmol CO2 g−1 s−1) and root respiration per unit N (R n, nmol CO2 g N−1 s−1) were affected by D r and N r. The R r, R n, N r and soil temperature sensitivity (Q 10) of R r for the two species significantly decreased with an increase of D r. The R r of the two species showed significant an inter-seasonal and diurnal pattern, and this trend decreased with increasing D r. Both the R r and Q 10 of the two species increased with increasing N r. The D r and N r explained 54 and 52% of the observed variation in R r for A. crassicarpa, and 65 and 70% for E. urophylla. The R r, N r, and Q 10 of A. crassicarpa were significantly higher than those of E. urophylla. Our results indicated that root respiration was dependent on D r and N r, and this dependence varied with season and plant species.  相似文献

In the Belgian Blue Cattle breed, coat color variation is mainly under the influence of a single autosomal locus, the roan locus, characterized by a pair of codominant alleles: r + (black) and R (white). Heterozygous r + R animals have intermingled black and white hairs, yielding the ``blue' phenotype typical of the breed. Major interest for the roan locus stems from its pleiotropic effect on fertility, owing to the critical role of the R allele in the determinism of White Heifer Disease. We describe the linkage mapping of the roan locus to bovine Chromosome (Chr) 5, in the interval between microsatellite markers BPI and AGLA293, with an associated lodscore of 11.2. A trenching method was used to determine the contribution of root respiration to soil respiration. Soil respiration rates in a trenched plot (R trench) and in a control plot (R control) were measured from May 2000 to September 2001 by using an open-flow gas exchange system with an infrared gas analyser. The decomposition rate of dead roots (R D) was estimated by using a root-bag method to correct the soil respiration measured from the trenched plots for the additional decaying root biomass. The soil respiration rates in the control plot increased from May (240–320 mg CO2 m–2 h–1) to August (840–1150 mg CO2 m–2 h–1) and then decreased during autumn (200–650 mg CO2 m–2 h–1). The soil respiration rates in the trenched plot showed a similar pattern of seasonal change, but the rates were lower than in the control plot except during the 2 months following the trenching. Root respiration rate (R r) and heterotrophic respiration rate (R h) were estimated from R control, R trench, and R D. We estimated that the contribution of R r to total soil respiration in the growing season ranged from 27 to 71%. There was a significant relationship between R h and soil temperature, whereas R r had no significant correlation with soil temperature. The results suggest that the factors controlling the seasonal change of respiration differ between the two components of soil respiration, R r and R h.  相似文献

Cytological and genetical studies are reported on the hybrid and later generations ofGossypium herbaceum (A1 genome) xG. triphyllum (Hg genome). Chromosome pairing between the two genomes was high, and their chromosomes are considered, with minor exceptions, to be strueturally similar. Studies of the F1 and segregating families showed thatG. triphyllum carries the complementary factorsR inf2 supΘS R infa supGO for petal spot,Y a(Yb) Y infc supP for cream petal,pa for cream pollen, andY ap for depression of yellow petal pigment. Leaf ahape ofG. triphyllum was dominant to the leaf shape ofG. herbaceum. The blue petal color ofG. triphyllum was recovered only in the backeross of the F1 to this species. Summary Mutants, designated tamA r, have been isolated on the basis of simultaneous resistance to toxic analogues thiourea, aspartate hydroxamate and chlorate with L-alanine as the sole nitrogen source. tamA r mutants are also resistant to methylammonium. This resistance of tamA r mutants is correlated with partially repressed activity of a number of enzyme and transport systems regulated by ammonium. Furthermore, tamA r mutants have low NADP-glutamate dehydrogenase (NADP-GDH) activity and also efflux ammonium under certain growth conditions.Mutants at the areA locus (areA r) have also been isolated on the basis of resistance to these analogues, with nitrate or L-aspartate as the nitrogen source. These, similar to tamA r lesions, result in resistance to methylammonium and are partially repressed for ammonium repressible systems, but in contrast to tamA r, areA r alleles have wild-type NADP-GDH activity and normal ammonium efflux. tamA r and areA r mutants grow as wild type on all nitrogen or carbon sources tested, are recessive, and appear to be epistatic to all other mutations (gdhA1, meaA8 and meaB6) which result in derepressed levels of ammonium regulated system. Summary The X-linkedrudimentary (r) mutants ofDrosophila melanogaster are pyrimidine auxotrophs and require exogenous pyrimidines (Nørby, 1970; Falk, 1976). We have established a set ofrudimentary cell lines that are derived from embryos, homozygous for eitherr 1 orr 36. The enzymatic activities of the pyrimidine synthesizing enzymes were measured in the mutant lines. We have further investigated the nutritional requirements of the mutant cells in vitro by using a pyrimidine free culture medium.Ther 1 cell lines were found to express 3–7%dihydroorotase (DHOase) activity as compared to a wildtype cell line. Reducedaspartate transcarbamylase (ATCase) activity was measured in somer 1 cell lines whereas wildtypecarbamylphosphate synthetase (CPSase) activity is expressed in allr 1 cell lines. Ther 36 cell line expresses wildtype activity ofDHOase andCPSase. ATCase activity was found to be reduced to 10% of the wildtype activity.The mutant cell lines do not proliferate in pyrimidine free minimal medium and cell proliferation is obtained by the addition of crude RNA. Proliferation of ther 1 cells is restored by the supplementation of the minimal medium withdihydroorotate whereas proliferation of ther 36 cells is restored by supplementation with eitherdihydroorotate orcarbamylaspartate.The results demonstrate that therudimentary phenotypesr 1 andr 36 are expressed at the cellular level and that the two mutant cell types behave as cellular pyrimidine auxotrophs in vitro.  相似文献

Adult longevity, developmental time and juvenile mortality ofEncarsia formosa Gahan (Hymenoptera:Aphelinidae) parasitizing the Poinsettia-strain ofBemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) on Poinsettia (Euphorbia pulcherrima Willd.) were investigated in laboratory experiments at three temperatures: 16 °C, 22 °C and 28 °C. Furthermore, the parasitoid's preference for different larval stages of the whitefly was determined at 24.5 °C. The lifespan ofE. formosa decreased with temperature from one month at 16 °C to nine days at 28 °C. A lower temperature threshold of 11 °C for adult development was found. The development of juvenile parasitoids inB. tabaci lasted more than two months at the lowest temperature, but was only 14 days when temperature was 28 °C. The lower temperature threshold for immature development was 13.3 °C, yielding an average of 207 day-degrees for the completion of development into adults. Juvenile mortality was high, varying from about 50% at 16 °C to about 30% at 22 °C and 28 °C.E. formosa preferred to oviposit in the 4th instar and prepupal stages ofB. tabaci followed by the 2nd and 3rd instars. The preference for the pupal stage was low. The parasitoid used all instars of the whitefly for hostfeeding, with no apparent differences between the stages. The average duration of the oviposition posture was four minutes. Demographic parameters were calculated from life tables constructed from the data. The intrinsic rate of increase (r m) and the net reproductive rate (R 0) increased with temperature from 0.0279 day−1 at 16 °C to 0.2388 day−1 at 28 °C and from about 12 at 16 °C to about 66 at 28 °C, respectively.  相似文献

Of the 130 clinical isolates of Mycoplasma hominisfrom patients with nonspecific inflammatory diseases of the urogenital tract (UGT), approximately 10% contained the tet(M) gene after the course of treatment with tetracyclines. This gene was found in nine (25%) of the 36 Ureaplasma urealyticum clinical isolates. The nucleotide sequence of 13 tet(M) genes in TcR clinical isolates ofM. hominis and five genes in U. urealyticum TcR clinical isolates was determined. A comparison of nucleotide sequences of eight tetM genes of different origin and tet(M) genes ofGardnerella vaginalis and M. hominis and U. urealyticumclinical isolates showed that the mosaic structure of thetet(M) gene is completely identical in 11 of 13 M. hominis TcRisolates but belongs to an unidentified allele different from those described earlier. Another new allelic variant oftet(M) was found in two isolates. In three of five TcR clinical isolates of U. urealyticum, a tet(M) gene, whose mosaic structure was identical to that of tet(M) reported previously for ureaplasmas, and also two new allelic variants, which have not been described so far, were found.  相似文献

A chimeric gene composed of the coding sequence of theble gene fromStreptoalloteichus hindustanus fused to the 5 and 3 untranslated regions of theChlamydomonas reinhardtii nuclear geneRBCS2 has been constructed. Introduction of this chimeric gene into the nuclear genome ofC. reinhardtii by co-transformation with theARG7 marker yields Arg+ transformants of which approximately 80% possess theble gene. Of these co-transformants, approximately 3% display a phleomycin-resistant (PmR) phenotype. Western blot analysis using antibodies against theble gene product confirms the presence of the protein in the PmR transformants and genetic analysis demonstrates the co-segregation of theble gene with the phenotype in progeny arising from the mating of a PmR transformant to wild-type strains. Direct selection of PmR transformants was achieved by allowing an 18-h period for recovery and growth of transformed cells prior to selection. This work represents the first demonstration of stable expression and inheritance of a foreign gene in the nuclear genome ofC. reinhardtii and provides a useful dominant marker for nuclear transformation.  相似文献

TheSchizosaccharomyces pombe rhp51 + gene encodes a recombinational repair protein that shares significant sequence identities with the bacterial RecA and theSaccharomyces cerevisiae RAD51 protein. Levels ofrhp51 + mRNA increase following several types of DNA damage or inhibition of DNA synthesis. Anrhp51::ura4 fusion gene was used to identify the cis-acting promoter elements involved in regulatingrhp51 + expression in response to DNA damage. Two elements, designated DRE1 and DRE2 (fordamage-responsiveelement), match a decamer consensus URS (upstream repressing sequence) found in the promoters of many other DNA repair and metabolism genes fromS. cerevisiae. However, our results show that DRE1 and DRE2 each function as a UAS (upstream activating sequence) rather than a URS and are also required for DNA-damage inducibility of the gene. A 20-bp fragment located downstream of both DRE1 and DRE2 is responsible for URS function. The DRE1 and DRE2 elements cross-competed for binding to two proteins of 45 and 59 kDa. DNase I footprint analysis suggests that DRE1 and DRE2 bind to the same DNA-binding proteins. These results suggest that the DRE-binding proteins may play an important role in the DNA-damage inducibility ofrhp51 + expression.  相似文献
