A double-blind, randomized control clinical trial was performed to investigate the effectiveness of teflubenzuron in controlling sea lice Lepeophtheirus salmonis on farmed Atlantic salmon Salmo salar. A total of 40 sea cages from 3 commercial cage sites in Atlantic Canada were used in this Good Clinical Practice (GCP) trial. The teflubenzuron was administered in the feed at a dosage of 10 mg kg(-1) biomass d(-1) for 7 d. Medicated and control cages were matched by site, cage size, and pre-treatment mean lice counts using cages as the unit of concern. Post-treatment lice counts and staging of developmental stages were performed at 1 and 2 wk after the end of treatment. Chalimus stages in medicated cages were significantly lower than in control cages at 1 wk (79% reduction in mean lice counts, p < 0.001), and at 2 wk (53% reduction, p < 0.001). Mobile (pre-adult and adult) stages were also significantly reduced in medicated cages at 1 wk (69% reduction, p < 0.01), and at 2 wk (40% reduction, p < 0.01) post-treatment, respectively. Teflubenzuron was proven effective for reducing lice burdens on salmon despite the low parasite levels experienced during the trial and the recruitment of lice from the untreated cages. The use of cage as the unit of concern was an important design component of this trial.  相似文献

Two trials were conducted at commercial salmon farms to evaluate the efficacy of emamectin benzoate (Slice, 0.2% aquaculture pre-mix, Schering-Plough Animal Health) as a treatment for sea lice Lepeophtheirus salmonis (Kr?yer) and Caligus elongatus Nordmann infestations in Atlantic salmon Salmo salar L. Trials were carried out in 15 m2 commercial sea pens, at temperatures of 5.5 to 7.5 degrees C and 10.8 to 13.8 degrees C. Each pen was stocked with 14,000 to 17,500 fish with mean weights of 0.44 to 0.74 and 1.33 to 1.83 kg. Fish were naturally infested with sea lice at the start of each trial. At Day -1, samples of 10 or 15 fish were taken from each pen to determine pre-treatment numbers of lice. Emamectin benzoate was administered in feed, to 4 replicate pens, at a dose of 50 micrograms kg-1 biomass d-1 for 7 consecutive days (Days 0 to 6). Sea lice were counted again, between Days 7 and 77, and comparisons made with untreated control fish. Despite adverse weather conditions, wide variations in fish weights and exposure to new infestations, treatment was effective against chalimus and motile stages of L. salmonis. In the autumn trial, efficacy at Day 27 was 89%, and lice numbers remained lower on treated fish than on control fish 64 d from the start of treatment. In the winter trial, reductions in lice numbers at low temperatures were slower but good efficacy was achieved by Day 35. Although control fish had to be treated with hydrogen peroxide at Day 21, fish treated only with emamectin benzoate on Days 0 to 6 still had 89% fewer lice than control fish at Day 35. There were very few C. elongatus present, but at the end of both trials numbers were lower on treated fish. No adverse effects were associated with treatment of fish with emamectin benzoate.  相似文献

Effects of artificial salmon lice infection and pharmaceutical salmon lice prophylaxis on survival and rate of progression of Atlantic salmon (n = 72) and brown trout post-smolts (n = 72) during their fjord migration, were studied by telemetry. The infected groups were artificially exposed to infective salmon lice larvae in the laboratory immediately before release in the inner part of the fjord to simulate a naturally high infection pressure. Groups of infected Atlantic salmon (n = 20) and brown trout (n = 12) were also retained in the hatchery to control the infection intensity and lice development during the study period. Neither salmon lice infection nor pharmaceutical prophylaxis had any effects on survival and rate of progression of fjord migrating Atlantic salmon post-smolts compared to control fish. Atlantic salmon spent on average only 151.2 h (maximum 207.3 h) in passing the 80 km fjord system and had, thus, entered the ocean when the more pathogenic pre-adult and adult lice stages developed. The brown trout, in comparison to Atlantic salmon, remained to a larger extent than Atlantic salmon in the inner part of the fjord system. No effect of salmon lice infection, or protection, was found in brown trout during the first weeks of their fjord migration. Brown trout will, to a larger extent than Atlantic salmon, stay in the fjord areas when salmon lice infections reach the more pathogenic pre-adult and adult stages. In contrast to Atlantic salmon, they will thereby possess the practical capability of returning to freshwater when encountering severe salmon lice attacks.  相似文献

The global increase in the production of domestic farmed fish in open net pens has created concerns about the resilience of wild populations owing to shifts in host–parasite systems in coastal ecosystems. However, little is known about the effects of increased parasite abundance on life-history traits in wild fish populations. Here, we report the results of two separate studies in which 379 779 hatchery-reared Atlantic salmon smolts were treated (or not) against salmon lice, marked and released. Adults were later recaptured, and we specifically tested whether the age distribution of the returning spawners was affected by the treatment. The estimates of parasite-induced mortality were 31.9% and 0.6% in the River Vosso and River Dale stock experiments, respectively. Age of returning salmon was on average higher in treated versus untreated fish. The percentages of fish returning after one winter at sea were 37.5% and 29.9% for the treated and untreated groups, respectively. We conclude that salmon lice increase the age of returning salmon, either by affecting their age at maturity or by disproportionately increasing mortality in fish that mature early.  相似文献

Marine salmon farming has been correlated with parasitic sea lice infestations and concurrent declines of wild salmonids. Here, we report a quantitative analysis of how a single salmon farm altered the natural transmission dynamics of sea lice to juvenile Pacific salmon. We studied infections of sea lice (Lepeophtheirus salmonis and Caligus clemensi) on juvenile pink salmon (Oncorhynchus gorbuscha) and chum salmon (Oncorhynchus keta) as they passed an isolated salmon farm during their seaward migration down two long and narrow corridors. Our calculations suggest the infection pressure imposed by the farm was four orders of magnitude greater than ambient levels, resulting in a maximum infection pressure near the farm that was 73 times greater than ambient levels and exceeded ambient levels for 30 km along the two wild salmon migration corridors. The farm-produced cohort of lice parasitizing the wild juvenile hosts reached reproductive maturity and produced a second generation of lice that re-infected the juvenile salmon. This raises the infection pressure from the farm by an additional order of magnitude, with a composite infection pressure that exceeds ambient levels for 75 km of the two migration routes. Amplified sea lice infestations due to salmon farms are a potential limiting factor to wild salmonid conservation.  相似文献

Physiological, immunological and biochemical parameters of blood and mucus, as well as skin histology, were compared in 3 salmonid species (rainbow trout Oncorhynchus mykiss, Atlantic salmon Salmo salar and coho salmon O. kisutch) following experimental infection with sea lice Lepeophtheirus salmonis. The 3 salmonid species were cohabited in order to standardize initial infection conditions. Lice density was significantly reduced on coho salmon within 7 to 14 d, while lice persisted in higher numbers on rainbow trout and Atlantic salmon. Lice matured more slowly on coho salmon than on the other 2 species, and maturation was slightly slower on rainbow trout than on Atlantic salmon. Head kidney macrophages from infected Atlantic salmon had diminished respiratory burst and phagocytic capacity at 14 and 21 d post-infection (dpi), while infected rainbow trout macrophages had reduced respiratory burst and phagocytic capacities at 21 dpi, compared to controls. The slower development of lice, coupled with delayed suppression of immune parameters, suggests that rainbow trout are slightly more resistant to lice than Atlantic salmon. Infected rainbow trout and Atlantic salmon showed increases in mucus lysozyme activities at 1 dpi, which decreased over the rest of the study. Mucus lysozyme activities of infected rainbow trout, however, remained higher than controls over the entire period. Coho salmon lysozyme activities did not increase in infected fish until 21 dpi. Mucus alkaline phosphatase levels were also higher in infected Atlantic salmon compared to controls at 3 and 21 dpi. Low molecular weight (LMW) proteases increased in infected rainbow trout and Atlantic salmon between 14 and 21 dpi. Histological analysis of the outer epithelium revealed mucus cell hypertrophy in rainbow trout and Atlantic salmon following infection. Plasma cortisol, glucose, electrolyte and protein concentrations and hematocrit all remained within physiological limits for each species, with no differences occurring between infected and control fish. Our results demonstrate that significant differences in mucus biochemistry and numbers of L. salmonis occur between these species.  相似文献

The treatment of sea lice, Lepeophtheirus salmonis infestations on farmed Atlantic salmon, Salmo salar requires regular bathing of the fish in diachlorvos (DDVP), 2,2-dichlorovinyl dimethyl phosphate, with subsequent release to the marine environment of spent pesticide. The toxicity of a possible alternative compound, carbaryl, l-naphthyl N-methylcarbamate) to kill sea lice and Atlantic salmon was examined. Preliminary studies with carbaryl indicate the potential advantage of a reduced treatment dose was outweighed by the greater persistence of both the parent compound and of its toxic degradation product. Hence this carbamate insecticide is unlikely to be considered by regulatory authorities as an alternative treatment for sea lice infestations.  相似文献

Atlantic salmon Salmo salar L. artificially infected with salmon lice Lepeophtheirus salmonis (Kr?yer 1837) recovered from detrimental physiological changes and skin damage induced by preadult lice as the parasites matured. Growth rates of Atlantic salmon remained unaffected by lice infection, but food consumption decreased with increasing feeding and movement of the lice prior to and post-mating, correlating with the appearance of head erosions and detrimental changes in physiological integrity. Food consumption of the fish increased as the lice moulted to the adult stage and gravid female lice settled in a posterior location on the fish, subsequently reducing the impact of infection and allowing recovery of the skin damage. However, the impact of preadults was limited, as the decrease in food consumption of fish at 21 d post-infection had no effect on either the specific growth rate or condition factor of the fish. Furthermore, the intensity of lice infections at each of the sample days was not correlated with food consumption, specific growth rate or any of the haematological or physiological parameters measured, either before or after infection, indicating that lice intensity was independent of social dominance/subordinance. This work has provided the first evidence that infected fish can recover from the detrimental changes caused by lice infection, even when they are still infected with lice. If fish can survive the preadult stage of lice, then the mortal impact of lice infections is greatly reduced.  相似文献

Animal migrations can affect disease dynamics. One consequence of migration common to marine fish and invertebrates is migratory allopatry-a period of spatial separation between adult and juvenile hosts, which is caused by host migration and which prevents parasite transmission from adult to juvenile hosts. We studied this characteristic for sea lice (Lepeophtheirus salmonis and Caligus clemensi) and pink salmon (Oncorhynchus gorbuscha) from one of the Canada's largest salmon stocks. Migratory allopatry protects juvenile salmon from L. salmonis for two to three months of early marine life (2-3% prevalence). In contrast, host diversity facilitates access for C. clemensi to juvenile salmon (8-20% prevalence) but infections appear ephemeral. Aquaculture can augment host abundance and diversity and increase parasite exposure of wild juvenile fish. An empirically parametrized model shows high sensitivity of salmon populations to increased L. salmonis exposure, predicting population collapse at one to five motile L. salmonis per juvenile pink salmon. These results characterize parasite threats of salmon aquaculture to wild salmon populations and show how host migration and diversity are important factors affecting parasite transmission in the oceans.  相似文献

The river Ingdalselva, which drains to the Trondheims fjord, has no local salmon population due to an impassable waterfall 500 m upstream from the outlet. In the period 1994–97, a total of 31 mature Atlantic salmon (19 females and 12 males) from the rivers Orkla (1994–96) and Vigda (1997) were radio-tagged and released in the river Ingdalselva before spawning. The main goal of the project was to find out whether the fish would stay and spawn in the river, and if the observations during the spawning period could be used to indicate where spawning had taken place. Some fish left the river shortly after release, but 77% of the fish stayed in the river during the spawning period in October. Most of the females (74%) spawned in the river, including multi-sea winter salmon of approximately 10 kg. Some fish remained at the site of the release, while others migrated downstream to hiding places where they stayed until spawning. Long distance upstream migrations were not observed. The observations of the fish during the spawning period were used to identify spawning areas. Electrofishing for salmon fry in the spawning areas the following year in all cases produced fry, and in two of the years salmon fry were also found in areas where no spawning activity had been registered. Radio-tracking was an efficient method for determining whether transplanted salmon would remain in a `new'n river during the spawning season and for locating spawning areas, particularly when the fish were tracked daily.  相似文献

Four commercial salmon farms on the West coast of Norway were recruited to a programme of field trials in which the efficacy of SLICE (0.2% emamectin benzoate; Schering-Plough Animal Health) was compared with a commercially available product, EKTOBANN (teflubenzuron 2 g kg(-1); Skretting A/S) in treating natural sea lice Lepeophtheirus salmonis infections in Atlantic salmon Salmo salmar L. At each test site, 3 fish pens were treated with each product. In total, nearly 1.2 million first-year-class fish were included in the trial, of which approximately 561,000 received emamectin benzoate at a dosage of 50 microg kg(-1) body wt d(-1), while approximately 610,000 received teflubenzuron at a dosage of 10 mg kg(-1) body wt d(-1). Medicated feed was provided at 0.5% body wt d(-1) over 7 consecutive days. Feed containing emamectin benzoate was generally well accepted by the fish and no problems were encountered in feeding the medicated diet at the desired dose. Lice numbers were counted 2 d before and 1, 7, 14 and 21 d after commencement of treatment. While treatment with both substances rapidly reduced lice numbers, pens treated with emamectin benzoate were found to harbour significantly fewer lice 14 and 21 d post-treatment. Twenty-one days following treatment with emamectin benzoate the lice abundance was reduced on average by 94%. Limited sampling outside the main study period indicated that emamectin benzoate protects against sea-lice infestation over longer periods.  相似文献

The efficacy of teflubenzuron (Calicide) for the treatment of farmed Atlantic salmon Salmo salar L. infested with sea lice Lepeophtheirus salmonis (Kr?yer, 1838), was investigated at low water temperatures in 2 commercial salmon farms. Calicide, coated on commercial feed pellets, was administered orally at 10 mg kg(-1) d(-1) for 7 consecutive days. Fish were randomly sampled and lice numbers recorded from both treated and control groups on 3 or 4 sampling occasions post-medication. Statistically significant reductions in the number of L. salmonis per fish were recorded. Maximum efficacy was observed toward chalimus and preadult stages of L. salmonis, and was achieved approximately 26 d post-medication. No adverse drug reactions or palatability problems were associated with the treatments.  相似文献

Some members of the ABC-transporter superfamily, such as P-glycoprotein and the multidrug resistance associated protein, may confer resistance to the avermectin subclass of macrocyclic lactones. The aim of this study was to examine the presence of ABC transporters in both sea lice (Lepeophtheirus salmonis) and its Atlantic salmon host (Salmo salar) using monoclonal antibodies (C219 and JSB-1, with high selectivity for P-gp) and a new polyclonal antibody (SL0525) generated against a putative sea louse ABC transporter. The antibody raised to SL0525 did not react with rat P-gp, suggesting that an ABC transporter, not necessarily P-gp, was isolated. C219 was the only antibody to localize P-gp in all 3 salmon tissues (intestine, kidney and liver). American lobster (Homarus americanus) was used as a reference crustacean for L. salmonis immunostaining reactions and showed positive staining in the hepatopancreatic and intestinal tissues with all 3 antibodies. The L. salmonis showed positive staining in the intestinal epithelial lining with all antibodies. This report represents the first documented evidence for the expression of ABC transporters in L. salmonis, its Atlantic salmon host, and the American lobster.  相似文献

The behaviour of lumpfish, Cyclopterus lumpus L., in sea pens, with and without Atlantic salmon, Salmo salar L., present, was assessed by underwater camera technology. Behaviour was classified by recording the principal activity of individual fish for 30-s intervals. The majority of daylight time was spent actively foraging for food. Antagonistic behaviour between Atlantic salmon and lumpfish was not observed during the whole experimental period and no mortality was seen in either species. Cleaning behaviour, but at low frequency, was observed as lumpfish cleaned sea lice off Atlantic salmon. Significantly lower sea lice infection levels were seen on Atlantic salmon when reared together with lumpfish compared to the control group without lumpfish. Feeding behaviour can be classified as strongly opportunistic.  相似文献

During the period 1968–1991, certain morphological traits of Atlantic salmon Salmo salar and sea trout S. trutta have been recorded regularly at the hatchery at Älvkarleby, central Sweden. Total body length, weight (for females both before and after stripping), number of eggs, egg size and date of ovulation. A smaller data set for fish marked and released as smolts, providing information about total body length of released smolts, time spent at sea and body size of the recovered adults, was also available for analysis. According to theory and empirical data, the process of artificial breeding results in an evolutionary divergence of the cultured strain from the wild phenotypic norm. The reason for such a divergence is that both natural and sexual selection pressures are altered or relaxed during the process of artificial breeding, as well as random genetic processes, such as founder effects and in- and outbreeding. A path analysis of both species and sexes revealed that the size of the released smolts had increased during the study period. Time spent in sea has decreased for both female and male sea trout, but not for Atlantic salmon. Adult body size for female and male trout have increased as well as female trout condition factor. The increase found in egg size of both species was greater for the salmon than for the trout, indicating that female salmon invest more in egg size with increasing body size. These results support the view that domestication probably has a significant and selective impact on the life history traits of the two salmonid species studied.  相似文献

Drinking in Atlantic salmon (Salmo salar) juveniles was investigated in fresh water and following transfer to sea water. There was a significant effect of fish size on drinking, and smolts (20–30 g) imbibed about ten times less water than alevins of 0.2–0.3 g. Freshwater smolts drank at a rate of 0.15 ± 0.03 ml · kg−1 · h−1 and administration of doses of 10 or 20 mg · kg−1 of papaverine (stimulator of the renin- angiotensin system RAS) or [Asn1, Val5]-Angiotensin II (0.4 μmol · kg−1) resulted in significant increases in drinking, while administration of the angiotensin converting enzyme inhibitor, enalapril (50 mg · kg−1) had no effect on drinking. Transfer of Atlantic salmon smolts to 1/3, 2/3 and full strength sea water resulted in significant increases in drinking to 1.06 ± 0.12, 1.24 ± 0.0.16 and 3.89 ± 0.28 ml · kg−1 · h−1, respectively. In sea water, stimulation of the endogenous RAS by administration of papaverine (20 mg · kg−1) resulted in a 20% increase in drinking, while administration of enalapril to doses of 50 and 200 mg · kg−1 lowered drinking to 1.99 ± 0.48 and 0.32 ± 0.06 ml · kg−1 · h−1, respectively. All treatments were without effect on blood plasma levels of Na+ and Cl in fresh water, while in sea water smolts both stimulation and inhibition of drinking resulted in hemoconcentration of Na+ and Cl. The role of the renin angiotensin system in control of drinking and hydromineral balance in Atlantic salmon is discussed. Accepted: 27 February 1997  相似文献
