Parallel observations on synaptic and neurohaemal terminals in the polychaete annelids Nereis diversicolor and Harmothoe imbricata have revealed a remarkable identity of ultrastructure. Even features peculiar to the synaptic vesicles of polychaetes are mirrored by those of synaptoid inclusions. A wide range of terminal types show a clear duality of secretory inclusions, featuring both ‘storage granules’ and synaptic/synaptoid vesicles. The inclusions exhibit a marked zonation. Vesicles form tight clusters with interstitial dense material in many terminals and these make contact with release sites. Terminals containing larger, typically peptidergic granules often have mainly dense-cored synaptic/synaptoid vesicles, although some such inclusions are present in other endings also. A variety of synaptic associations are present, and ‘serial synaptoids’ are formed by neurohaemal terminals. The results are interpreted to suggest that synaptic and synaptoid vesicles have a common functional significance.  相似文献

An in vitro preparation from the pedal ganglia of the marine bivalve, Mytilus edulis, was used to examine the modulation of transmitter release by adenosine and its analogs from invertebrate nervous tissue. The ganglia of this organism contain the monoamines dopamine (DA), serotonin (5-HT), and norepinephrine (NE), and the presynaptic release of these substances is known to be calcium-dependent. This organism also contains a DA-sensitive adenylate cyclase system which resembles that seen in mammals. Neural tissue from the pedal ganglia was incubated with labeled monoamines, and release studies were then conducted in superfusion chambers; release of monoamines was evoked by the addition of 50 mM KCl. Addition to the superfusion medium of the adenosine analog, 5'-N-ethylcarboxamidoadenosine (NECA; 10 nM), inhibited the release of 5-HT and DA, and to a lesser extent NE, whereas 100-fold higher concentrations of adenosine itself and the adenosine analog, R-N6-phenylisopropyladenosine, were required to achieve comparable levels of inhibition. The inhibitory effects of NECA on neurotransmitter release were blocked by the adenosine receptor antagonist, theophylline (IC50 = 10-14 microM). The results from this study indicate for the first time the possible role of adenosine as a modulator of neurotransmitter release in the invertebrate nervous system.  相似文献

Ultrastructural observations of the feeding sites of soybean cyst nematode juveniles 3 days after inoculation of soybean roots revealed the presence of feeding tubes in the host cell syncytium. Feeding tubes, which were extruded from the stylet tips, were formed by products of secretory granules that originated in the dorsal esophageal gland and accumulated in the ampulla of the gland extension. Granules traversing the space between the gland cell and the ampulla were regulated in their movement by two sets of sphincter-like muscles located anterior and posterior to the metacorpus pump chamber. Sections through the sphincter muscles revealed obliquely arranged fibers, which in a contracted mode caused microtubules in the gland extension to be tightly packed and devoid of granules.  相似文献

The secretory cells arc a prominent component of the phloemtissue of Mimosa pudica L. They form longitudinal files in whichthe common end walls of the cells are perforated. The originof the perforations in relation to protoplast differentiationwas the objectof the study. As the cells emerge from the meristematicstate, portions of cytoplasm including some organelles aretransferredinto the central vacuole and are apparently lysed. This phenomenonis accompanied by an accumulation of flocculent material, possiblythe secretory product referred to in the literature as a carbohydrate.The tonoplast ceases to be definable so that the flocculentmaterialand degraded and non-degraded protoplast componentsare intermingled inthe cell lumen. The perforation of the endwall OCCUR while the tonoplast is still intact. Theplasmalemmasof the contiguous cells withdraw from the part of the wall undergoingdegradation. The space becomes filled with fibrous debris ofthe apparently lysed wall. The degraded wallpart becomes brokenpossibly by a unidirectional pressure flow in the cells, forthe remnants of the wall are pushed into one of the two superimposedcells. At this presumably mature stage, the cells contain someintactprotoplast components among which ribosomes are especially conspicuous.  相似文献

Hydrogen Release Compound (HRC) has been a commercially available product for engineered bioremediation of anaerobically biodegradable contaminants since 1999. HRC is a polylactate ester that, upon hydration or microbial cleavage of its ester bonds, slowly releases lactic acid. Lactic acid serves as an electron donor for microbial reductive biodegradation, while also providing hydrogen and carbon where required. HRC is a viscous amber-colored liquid that is typically injected into a contaminated aquifer using direct push technology or backfill injection into boreholes created by traditional drilling methods. Once in place, HRC creates a plume of lactic acid and its fermentation products (other organic acids and hydrogen) downgradient of the injection area and serves to accelerate anaerobic bioremediation processes. In this review of HRC field application results, the authors summarize application types, contaminants treated, site types, application locations, injection methods, site lithology and hydrology, and concentration ranges of geochemical species. The source of this information is a database of more than 850 HRC field applications, a series of 80 HRC publications that are publicly available, and 44 detailed site case histories that are available electronically.  相似文献

Spores of selected strains of Bacillus megaterium were prepared by various methods and examined with the electron microscope. An exosporium like that of B. cereus, with a nap and basal layer, was found in spores of a B. megaterium strain that reportedly contains a capsule-like exosporium. The exosporium occasionally appeared to be doubled or have an apical opening. Pili-like filaments were discerned on the surface. Beneath the exosporium were found large deposits of planar inclusions, which in cross section appeared laminated and in surface views consisted of a patchwork of striated packets with a periodicity of approximately 5 nm. The inclusions were usually attached to the exosporium, but in ultrastructure they differed from both the exosporium and coat. In two other strains of B. megaterium, one or two coats occurred but a typical exosporium was not present.  相似文献

Secretory phospholipase A2 (sPLA2s) are small secreted proteins (14–18 kDa) and require submillimolar levels of Ca2+ for liberating arachidonic acid from cell membrane lipids. In addition to the enzymatic function, sPLA2 can exert various biological responses by binding to specific receptors. Physiologically, sPLA2s play important roles on the neurotransmission in the central nervous system and the neuritogenesis in the peripheral nervous system. Pathologically, sPLA2s are involved in the neurodegenerative diseases (e.g., Alzheimer’s disease) and cerebrovascular diseases (e.g., stoke). The common pathology (e.g., neuronal apoptosis) of Alzheimer’s disease and stroke coexists in the mixed dementia, suggesting common pathogenic mechanisms of the two neurological diseases. Among mammalian sPLA2s, sPLA2-IB and sPLA2-IIA induce neuronal apoptosis in rat cortical neurons. The excess influx of calcium into neurons via l-type voltage-dependent Ca2+ channels mediates the two sPLA2-induced apoptosis. The elevated concentration of intracellular calcium activates PKC, MAPK and cytosolic PLA2. Moreover, it is linked with the production of reactive oxygen species and apoptosis through activation of the superoxide producing enzyme NADPH oxidase. NADPH oxidase is involved in the neurotoxicity of amyloid β peptide, which impairs synaptic plasticity long before its deposition in the form of amyloid plaques of Alzheimer’s disease. In turn, reactive oxygen species from NADPH oxidase can stimulate ERK1/2 phosphorylation and activation of cPLA2 and result in a release of arachidonic acid. sPLA2 is up-regulated in both Alzheimer’s disease and cerebrovascular disease, suggesting the involvement of sPLA2 in the common pathogenic mechanisms of the two diseases. Thus, our review presents evidences for pathophysiological roles of sPLA2 in the central nervous system and neurological diseases.  相似文献

Bees were captured as they left from or arrived at a feeding place (F), or took off from the hive (H) (leave-F, arrive-F, and leave-H, respectively). They were displaced and released (one or more times) at initially unfamiliar sites. In some tests, sight conditions prior to release were restricted. Searching behaviour performed prior to leaving the release site was recorded as well as the final vanishing bearing (VB). The VBs obtained are distributed in one or more clusters, depending on the type of experiment. As a rule, one of the peaks coincides with the compass bearing that bees would have selected had they not been displaced. Another peak is in accordance with a ‘fleeing’ course in the direction of the hive, probably initiated by the stress situation caused by the experimental manipulations. Prior to selecting the final course, leave-H bees searched longer than did leave-F or arrive-F bees, first-time flyers searched longer than repeaters and bees limited to sky-vision searched longer than bees with free sight. There is a significant positive correlation between searching behaviour and scatter of vanishing bearings. Function and origins of the searching behaviour, as well as the effects of sight conditions and of repeated releases, are discussed.  相似文献

Electron microscopy of the photoreceptors in the marine nematode, Deontostoma californicum, revealed numerous lamellated inclusions in the Schwann cells ensheathing the lateral cephalic nerves. Immediately after the axons from the modified bipolar neurons of the photoreceptors enter the lateral nerves, these spherical-to-oval lamellated bodies are observed in the surrounding Schwann cell cytoplasm. These previously undescribed Schwann cell inclusions, approximately 500 nm long and 320 nm in diameter, are lamellated and characterized by the presence of an electron-dense stalk-like process, 80-280 nm long. The lamellated inclusions are bound by a single limiting membrane, 6-7 nm thick, which shows occasional interruptions. The internal structure of the inclusions is characterized by the presence of electron-dense lamellae or bands, 11-16 nm thick, which assume various complex patterns ranging from arrays of parallel linear densities to a reticulate appearance. In addition, the nematode Schwann cell cytoplasm contains the usual organelles, gliosome- and lysosome-like inclusions. Their relationship with lipofuscin pigments is briefly discussed.  相似文献

In the last 5 years a role for β‐catenin in the skeleton has been cemented. Beginning with mutations in the Lrp5 receptor that control β‐catenin canonical downstream signals, and progressing to transgenic models with bone‐specific alteration of β‐catenin, research has shown that β‐catenin is required for normal bone development. A cell critical to bone in which β‐catenin activity determines function is the marrow‐derived mesenchymal stem cell (MSC), where sustained β‐catenin prevents its distribution into adipogenic lineage. β‐Catenin actions are less well understood in mature osteoblasts: while β‐catenin contributes to control of osteoclastic bone resorption via alteration of the osteoprotegerin/RANKL ratio, a specific regulatory role during osteoblast bone synthesis has not yet been determined. The proven ability of mechanical factors to prevent β‐catenin degradation and induce nuclear translocation through Lrp‐independent mechanisms suggests processes by which exercise might modulate bone mass via control of lineage allocation, in particular, by preventing precursor distribution into the adipocyte pool. Effects resulting from mechanical activation of β‐catenin in mature osteoblasts and osteocytes likely modulate bone resorption, but whether β‐catenin is involved in osteoblast synthetic function remains to be proven for both mechanical and soluble mediators. As β‐catenin appears to support the downstream effects of multiple osteogenic factors, studies clarifying when and where β‐catenin effects occur will be relevant for translational approaches aimed at preventing bone loss and terminal adipogenic conversion. Microencapsulated ferrous sulfate with soy lecithin (SFE-171) has been used as an iron source for the fortification of milk and dairy products. With the purpose to extend the use of this agent to other kind of foods or even to pharmaceutical preparations for oral administration, the SFE-171 was turned into a fluid powder (SFE-171-P) by means of vacuum drying. The iron bioavailability (BioFe) of SFE-171-P was evaluated in this work by means of the prophylactic-preventive method in rats, using ferrous sulfate as reference standard. Both iron sources were separately added to a basal diet of low iron content in a concentration of 10 mg iron/kg diet. Two groups of 10 weaned rats 25 days old received the fortified diets during 28 days, while a third group of the same size received the basal diet without iron additions. The weights and haemoglobin concentrations (HbC) of every animal were determined before and after the treatment, thus allowing the calculation of the mass of iron incorporated into haemoglobin (HbFe) during this period. The BioFe of the iron sources were obtained as the percentage ratio between the HbFe and the mass of iron consumed by each animal. The results were also given as Relative Biological Value (RBV), which relates the BioFe of the studied source with that of the reference standard. The liver iron concentration (LIC) of each animal was determined at the end of the experiment in order to evaluate the influence of the studied iron sources on the liver iron stores. SFE-171-P presented BioFe, RBV and LIC values of (47 ± 7) %, 109% and (46.6 ± 3.4) mg/kg respectively, while the corresponding values for the reference standard were of (43 ± 7)%, 100% and (45.0 ± 4.7) mg/kg. These results show that the drying process used to produce the SFE-171-P does not affect its bioavailability, which is also adequate for the potential use of this product in food fortification or with pharmaceutical purposes.  相似文献

SYNOPSIS. Monolayers of bovine kidney cells were overlaid with Eimeria magna sporozoites and observed with phase-contrast optics until penetration of the cells by the parasites had begun. Cells and penetrating parasites were fixed with glutaraldehyde and OsO4-containing ruthenium red, dehydrated, and embedded in situ. Cells being penetrated were selected for study in the electron microscope. The lack of intracellular staining with ruthenium red and intact plasmalemmas of cells being penetrated, was accepted as evidence that the sporozoites did not disrupt the plasma membranes. The sporozoite caused invagination of the host cell plasmalemma until the parasite was entirely within the cell, after which the invagination was sealed off by short pseudopodia enclosing the sporozoite within a membrane-lined vacuole inside the cell. Often myelin-forms, apparently of host cell origin, were seen in the space between the sporozoite and the cell.  相似文献

Each glandular hair of Nyrnphoides peltaturn (Gmel.) O. Kuntz consisted of only one row of cylindar cells with secretory function. The hairs originated from the protoderm cells on the adaxial surface of the second leaf primordium from the shoot apex. Cells of the glandular hairs prossessed dense cytoplast during the secretory period, but the vacuoles were very small. There were not only abundant mitochondria, Golgi bodies and endoplasmic reticulum in the glandular hair cells, but also many plasmodesmata. The authors' research indicated that the mucilage was carried to the edge of the cells by the membranous multilamellar bodies and the vesicles from both Golgi bodies and endoplasmic reticulum. The mucilage was secreted extracellularly by either exocytosis or ecrine secretion. The side walls of the glandular hairs swelled because of mucilage mass accumulation in the walls. The mucilage, being tested to be composed of polysaccharides and a trace of protein, played an important role in protecting the development of the vegetative buds of N. peltatum.  相似文献
