Disappearance ofred light-requirement for the germination of upper seeds subjectto anoxia, chilling, cyanide or azide pretreatmenL—J.exp. Bot. 37: 1652–1662. The effects on the germination of positively photoblastic uppercocklebur (X anthium pennsylvanicum Wallr.) seeds by pretreatingwith anoxia, chilling, cyanide or azide, which stimulates theirdark germination, were examined in relation to light actions.Prior to experiments, seeds were pre-soaked at 23 °C inthe dark for 1 or 2 weeks to remove the pre-existing Pfr. Whenthe prctreatment conditions were suboptimal for germinationinduction, the stimulating effects of the pretreatments on germinationduring a subsequent dark period at 23 °C were manifest onlywhen seeds were irradiated with red light before or after thepretreatment Red light promotion was reversed by blue or far-redlight treatment. However, both prc-chilling for 6 d at 8 °Cand prctreatment with 1· 5 mol m – 3 NaN3 for 2d could induce full germination without red light exposure.On the other hand, both pre-exposure to anoxia for 8 d and pretreatmentwith 30 mol m–3 KCN could induce the dark germinationonly when germination occurred at 33 °C which is known toaugment the ratio of an alternative respiration flux to a cytochromeone. Moreover, the dark germination in response to these inductionswere strongly inhibited by the inhibitors of alternative respiration,propyl gallate and benzohydroxamic acid, applied during a subsequentdark period. It was thus suggested that Pfr has some relationto the operation of two respiration systems of cocklebur seeds,but it is not indispensable to germination of this positivelyphotoblastic seed. Cyclamen persicum Mill, seeds germinate in a narrow range oftemperature and germination is strongly inhibited by continuousirradiation with white light. The thermal optimum is approx.15 °C in both darkness and light. The effects of storage conditions on the germination of developingmuskmelon (Cucumis melo L.) seeds were tested to determine whetherafter-ripening is required to obtain maximum seed vigour. Seedswere harvested at 5 d intervals from 35 (immature) to 60 (fullymature) days after anthesis (DAA), washed, dried, and storedat water contents of 3·3 to 19% (dry weight basis) at6, 20, or 30°C for up to one year. Germination was testedin water and in polyethylene glycol 8000 solutions ( –0·2to –1·2 MPa osmotic potential) at 15, 20, 25 or30°C. Germination percentages and rates (inverse of meantimes to radicle emergence) were compared to those of newlyharvested, washed and dried seeds. For 40 and 60 DAA seeds,one year of storage at 20°C and water contents <6·5%significantly increased germination percentages and rates at20°C, but had little effect on germination at 25 and 30°C.Storage reduced the estimated base temperature (Tb) and meanbase water potential (b) for germination of both 40 and 60 DAAseeds by approximately 5°C and 0·3 MPa, respectively.Immature 35 DAA seeds showed the greatest benefit from storageat 3 to 5% water content and 30°C, as germination percentagesand rates increased at all water potentials (). Storage underthese same conditions had little effect on the germination ofmature seeds in water, but increased germination percentagesand rates at reduced 's. Accelerated ageing for one month at30°C and water contents from 15 to 19° increased germinationrates and percentages of mature seeds at reduced 's, but longerdurations resulted in sharp declines in both parameters. Immatureseeds lost viability within one month under accelerated ageingconditions. An after-ripening period is required at all stagesof muskmelon seed development to expand the temperature andwater potential ranges allowing germination and to achieve maximumgerminability and vigour. Post-harvest dormancy is deepest atthe point of maximum seed dry weight accumulation and declinesthereafter, both in situ within the ripening fruit and duringdry storage. Seed germination in Talinum triangulare as affected by photoperiod,with or without previous incubation in the dark in water at25 or 4 °C, was studied. The time course and quantity ofseed germination in photoperiods of 1 h and above were similarwith or without dark pretreatment, but the time to half maximumgermination was reduced from 12 days in non-dark pretreatedseeds to 4 days in seeds given 20 days in the dark at 25°C.A photoperiod of 0·25 h gave a lower rate and total germinationthan photoperiods of 1 h and above. Un-pretreated seeds required17 cycles of 24 h photoperiod for maximum germination as comparedwith 7 or less cycles if the seeds received more than 10 daysdark pretreatment at 25 °C. Both the rate and total germinationin light increased as the length of dark pretreatment at 25°C was increased from zero to 30 days. Incubation of theseeds in water in the dark at 4 °C for 5 to 30 days priorto illumination at 21 °C, reduced both the rate and quantityof seed germination in light as compared with those similarlyincubated in the dark at 25 °C. However, previous incubationin the dark for 30 days at 4 °C partially substituted forthe light requirement. The germination of the seeds of Rhinanthus Crista-galli hasbeen induced by exposing them to moisture at 2° C. for periodsof from 17 weeks to over a year, depending on the amount ofdry storage to which the seeds had previously been subjected.Germination could not be brought about by moisture-treatmentat 20° C. During moisture-treatment at 2° C. the respirationrate falls significantly after an initial increase, and thengradually rises, after which germination takes place. Generally,respiration is significantly lower during moisture-treatmentat 20° C. Analyses of the treated seeds suggest that therespiratory substrate might be protein. It was found by paperchromatography that during moisture-treatment at 2° C. thenumber of amino-acids in the alcohol extract first diminishesand then increases, reaching a maximum in the ungerminated seedjust before germination becomes apparent. Differences were detectedin the amino-acids found in the alcohol extracts of seeds treatedat 2° C. compared with those moisture-treated at 20°C. Reasons are given which suggest that the limiting factorin the germination process may be the nature and rate of thehydrolysis of the reserve proteins of the seeds.  相似文献

Seed conditioning and germination in witchweed (Striga asiatica(L.) Kuntze) were temperature-dependent. With higher conditioningtemperatures, shorter conditioning time was required for germinationwith terminal dl-strigol (strigol) treatment at 30 °C. Maximumgermination (80–100%) was obtained by conditioning inwater at 20, 25, 30 and 35 °C for 14, 7, 5 and 3 d, respectively,and terminally treating with 10–6 M strigol at 30 °C.Seeds conditioned in 10–8 M strigol instead of water germinatedmuch less with the same terminal strigol treatment. Generally,conditioning was slower when seeds were conditioned in strigolrather than water. The reduction in germination rate by pretreatmentin strigol or pretreatment at low temperatures could be overcomeby increasing the terminal strigol concentration in the germinationtest. Conditioned seeds did not germinate at 10 and 15 °Cwith a terminal 10–6 M strigol treatment but yielded closeto maximum germination at 25, 30 and 35 °C with the sameterminal strigol treatment. To obtain maximum germination, boththe minimum conditioning temperature and the minimum germinationtemperature for conditioned seeds were 20 °C. The effects of osmoconditioning on the germination at 15 and25 °C of pepper (Capsicum annuum L.) seeds were studiedover a 3-year period with respect to temperature of storage.Untreated seeds stored at 5 °C showed high germinabilitythroughout the entire storage period, whereas untreated seedsstored at 25 °C showed a progressive decline in germinability,especially when assayed at 15 °C. Seeds that had been osmoconditionedprior to storage retained a high level of germinability irrespectiveof either storage or germination temperatures. Flow cytometric analyses of nuclear DNA levels were carriedout during development, stratification and germination of dormantseeds from three tree species with contrasting characteristics.Norway maple (Acer platanoides) and sycamore (Acer pseudoplatanus)have orthodox (desiccation-tolerant) and recalcitrant (desiccation-sensitive)storage behaviours, respectively, and require only a periodof cold to break dormancy, whereas, orthodox cherry (Prunusavium) seeds require an initial warm period before cold stratificationto fully stimulate germination. Whole embryos and radicle tipsof both Norway maple and sycamore were found to have stablehigh levels of 4C DNA during the latter stages of developmentand both contained nuclei arrested at the 2C and 4C levels atmaturity. Mature cherry embryos had nuclei predominantly arrestedat the 2C level. This suggests that the acquisition of desiccationtolerance is not correlated with the arrest of the cell cycleat any particular nuclear DNA level. Neither DNA replicationin radicle cells nor germination occurred when seeds were maintainedmoist at a constant 20 °C. However, in the late stages ofcold treatment during stratification, nuclear DNA levels inradicle cells changed in advance of radicle emergence in theorthodox Norway maple and cherry, whereas in the recalcitrantsycamore, change was not recorded until after radicle emergence.These results show that DNA replication has potential use asan indicator of the progress of tree seeds through stratificationtreatments used to break some types of dormancy. Effects of dehydration, storage temperature and humidificationon germination of Salix alba andS. matsudana seeds were studied.Newly released seeds showed 100% germination before and afterdehydration to 11–12% moisture content. Germination ofthe high vigour lot (100% initial normal germination) was notaffected by dehydration to 6.7% moisture content but germinationdecreased with further dehydration to 4.3%. The lower vigourlot (75% initial normal germination) was more susceptible todehydration and germination decreased following dehydrationto 6.7% moisture content. Dry seeds of both species survivedimmersion in liquid nitrogen without loss of viability. Thegermination of seeds stored with 9% moisture content decreasedto 35–40% in 5 months at -20°C or in 2 months at 5°C.However, at 25°C seeds entirely lost viability within 2weeks. Seeds showed improved performance when stored at -70°C> - 20°C > 5°C > 25°C and tolerated dehydrationto a moisture content in equilibrium with 15% relative humidity.Results suggest that they are orthodox in storage behaviouralthough they are short-lived. Humidification treatment of lowvigour seed lots resulted in a remarkable increase in germinationpercentage. Positive linear relationships were shown between constant temperaturesand the rates of progress of germination to different percentiles,G, for single populations of each of five genotypes of chickpea(Cicer anetinum L.). The base temperature, Tb, at which therate of germination is zero, was 0·0°C for all germinationpercentiles of all genotypes. The optimum temperature, To(G),at which rate of germination is most rapid, varied between thefive genotypes and also between percentiles within at leastone population. Over the sub-optimal temperature range, i.e.from Tb to To(G), the distribution of thermal times within eachpopulation was normal. Consequently a single equation was appliedto describe the influence of sub-optimal temperatures on rateof germination of all seeds within each population of each genotype.The precision with which optimum temperature, Tb(G), could bedefined varied between populations. In each of three genotypesthere was a negative linear relationship between temperatureabove Tb(G) and rate of germination. For all seeds within anyof these three populations thermal time at supra-optimal temperatureswas constant. Variation in the time taken to germinate at supra-optimaltemperatures was a consequence of normal variation in the ceilingtemperature, To(G)—the temperature at or above which rateof progress to germination percentile G is zero. A new approachto defining the response of seed germination rate to temperatureis proposed for use in germplasm screening programmes. In two populations final percentage germination was influencedby temperature. The optimum constant temperature for maximumfinal germination was between 10°C and 15°C in thesepopulations; approximately 15°C cooler than the optimumtemperature for rate of germination. Thermal analyses of freezing events in hydrated lettuce (LactucasativaL.) seeds show a correlation between low temperature exotherms(LTEs) (evidence of ice crystal formation) and seed death. Yet,weather patterns common to the Northern Great Plains of NorthAmerica regularly create conditions where non-dormant seedsof native plants hydrate with snow melt and are subsequentlyexposed to -30 °C or colder conditions. To determine ifsuch weather patterns decimate dispersed seeds, we measuredthe effects of freezing on fully hydrated winterfat (Eurotialanata(Pursh) Moq.) seeds harvested from the Northern Plainsat two USA and one Canadian location. Survival of hydrated seedsto -30 °C at a cooling rate of 2.5 °C h-1was similarto that of seeds not subjected to cooling, even though botha high temperature exotherm (HTE) and an LTE were observed.Although the LTE was not related to winterfat seed survival,freeze-stressed seeds had reduced germination rates and reducedseedling vigour, particularly for the collection with the lightestseeds. The temperature of LTEs was similar among seed collectionswith a mean of -17.6 °C, but was warmer when the seeds wereimbibed at 0 °C compared to 5, 10 or 20 °C. We founda significant correlation between the HTE and LTE temperatures.The difference and the correlation may be due to the highermoisture content of seeds imbibed at 0 °C. After pericarpremoval, only one exotherm in the range of the LTE was observed.This was also true for the naked embryo. The influence of prehydration in water or priming in –1.5 MPa polyethylene glycol 8000 solution for various periods,followed by redrying, on germination rate and longevity of lettuce(Lactuca sativa L.) seeds (achenes) was determined during controlleddeterioration at 10% moisture content (fresh weight basis) and40°C. Short prehydration treatments (up to 1 h) had littleeffect on either germination rate or longevity, but significantlyimproved root growth rates. Increasing durations of prehydrationor priming reduced the mean time to germination by up to 61%relative to untreated seeds, but also reduced mean seed longevityby as much as 84% Prehydration and priming altered the relationshipsbetween germination rate and viability and between normal andabnormal seedlings during ageing. Prehydration in abscisic acidor at a temperature inhibitory to germination did not preventthe reduction in longevity under controlled deterioration conditions.While prehydration or priming treatments effectively acceleratesubsequent germination rates of lettuce seeds, the redried seedsare nonetheless highly susceptible to deterioration in storage. Biochemical properties, i.e. endogenous abscisic acid, proline,sugars, respiration, adenosine phosphates and adenylate energycharge, and growth and moisture content were measured duringthe development of seeds of Machilus thunbergii. As dry matteraccumulated in the embryo during development, moisture content,ABA, proline, respiration and sugars all declined. At maturity,the dry mass of the seeds failed to attain a plateau beforethe period of natural seed shedding; the axis and cotyledonsreached moisture contents of 58 and 45%, respectively. Dryingof immature seeds at 73% relative humidity and 25 °C for30 d resulted in a complete loss of viability at all developmentalstages tested with the exception of mature seeds that were ableto tolerate a 5% decrease in moisture content before germinationdeclined. ABA was detected in all embryos tested, with a maximum value16.·16 µg g-1 d. wt about midway through development.Although the presence of ABA induced no tolerance to desiccationof mature seeds, it did coincide with decreased content of waterin the developing seeds and decreased respiration. Desiccationdamage of M. thunbergii seeds occurred when moisture contentwas still high (45%) and this damage was not related to theabsence of oligosaccharides in the mature seeds. We concludethat developing embryos and mature seeds of M. thunbergii haveproperties common to many recalcitrant seeds, with seeds beingsensitive to desiccation at all stages, having a prominent ABApeak, little proline, lacking oligosaccharides, and specifically,little dormancy and a moderate rate of respiration of matureseeds (0·9 µmol O2 min-1 g-1 f. wt). Adenosinetriphosphate content and energy charge decreased from stagefour to stage eight of seed development, then increased againto 103 nmol g-1 d. wt and 0·73, respectively, in matureseeds. Seeds of Araucaria hunsteinii K. Schum. dried quicker at 29°Cthan at 19°C and quicker with the seed-coat removed thanwhen intact; seeds enclosed in polyethylene bags increased inmoisture content. At 15°C, seeds in a flow of air driedquicker than seeds in a box with silica gel, which in turn driedquicker than seeds in a box with no desiccant. No loss of germinationability occurred on drying fresh seed from 53 to about 32 percent moisture content (fresh weight basis); during further desiccationthe percentage germination was related to percentage moisturecontent in the form of a sigmoid curve, culminating in a completefailure to germinate at approximately 14 per cent moisture content.A consistent relationship was observed for all treatments andthe mean critical moisture content for seed death (failure togerminate) was near 23 per cent. Excised embryos grew on 1 percent agar but died if previously subjected to 14 h of desiccationat 15°C. In contrast, no relationship was found between germination andmoisture content of A. cunninghamii D. Don on desiccation from21 to 7 per cent moisture content. Possible causes for the observeddifference in response to desiccation are discussed and methodsfor seed storage are considered. Seed growth characteristics of Aesculus hippocastanum were examinedin detail during development from about 70 to 140 d after anthesis(DAA), mainly in 1988 and 1989. Mean fresh and dry weights increasedfor both the axis and the whole seed up to the time of peakseed fall at 135 DAA with no cessation before fruit abscission.Water per seed increased up to 100 DAA, after which no furtherincrease occurred; moisture content declined for the embryonicaxis and whole seed respectively from above 75 and 65% at 95DAA to 65 and 50% at 130 DAA. At fruit shedding in 1990 waterpotential values of -1·2, -2·6 and -1·1MPa were observed for the testa, cotyledon and axis tissuesrespectively; relevant sorption isotherms are presented. Decreases in seed moisture content during development were accompaniedby increases in desiccation tolerance and in germinability,both reaching their maximum at the time of peak seed fall. Atmaturity, only about 10% viability was retained on drying seedto 20% moisture content; it is confirmed that the seeds are'recalcitrant'. The exact relationship between moisture contentand germination during development was dependent on the deptof dormancy, as judged by the period of chilling required; eachduration of chilling at 2°C within the range 3-12 weeksyields a curve of sigmoid shape. No germination occurred at26°C without chilling, but nearly full germination can beobserved for samples collected at 6 weeks before maximum seedfall with 12 weeks chilling. The effect of ethylene on quantitative and qualitative aspectsof respiration in Spergula arvensis L. seeds was studied. Oxygenconsumption and CO2 evolution from seeds increased 60 h afteraddition of ethylene to the dry seed; these increases were consideredto be a function of the number of seeds that were germination,rather than a direct effect of ethylene. The respiratory quotient(RQ) on day 1 (0.5) was lower than on subsequent days, indicatingan absence of anaerobic processes in the early stages of germinationand the use of a different respiratory substrate to that oxidizedon days 2–4. Measurements of C4/C1 ratios showed thatethylene did not break dormancy by initiating a switch fromthe Embden-Meyerhof-Parnas pathway (EMPP) to the pentose phosphatepathway (PPP). On the contrary, some evidence was obtained fora switch from the PPP to the EMPP after 3–6 h in pre-imbibedand non pre-imbibed seeds treated with ethylene. Effects of 2 °C chilling on the threshold moisture contentsand water potentials for various physiological processes wereestimated forAesculus hippocastanumL. seed. Seed harvested atthe time of maximum seed fall exhibited a dual response to drying:partial drying from near 50% to 32–40% moisture contentprogressively increased germination percentage (at 16 °C)up to various peak values; further desiccation was detrimental,confirming that the seeds are ‘recalcitrant’. Themoisture content for optimum germination was increased by atleast 10% as the chilling period was raised from 0 to 9 weeks.A negative linear relationship was found between log10mean timeto germinate and probit final germination, regardless of pre-treatment,indicating that partial desiccation and chilling are interchangeablein promoting germination of hydrated seed. For nearly fullyhydrated seeds, increasing the chilling period from 6 to 26weeks increased the viability-loss onset point for desiccationinjury from near 40% to about 48% moisture content without alteringthe drying rates of seed tissues. Extending moist chilling invarious seed lots from 0 to 26 weeks decreased subsequent longevityat 16 °C. For 26-week-chilled seeds longevity (the periodto lose one probit of germination) differed above and belowa threshold moisture content of 48%. It remained constant inthe moisture-content range 48–38%, but increased progressivelyas moisture content was raised above 48%. This threshold moisturecontent coincided with the value above which chilled seed pre-germinatedin storage. Sweet pepper seeds were osmotically conditioned in 0.4 M mannitolsolution for 4 d (at 25 °C, in darkness) before or afterstorage at 35 °C for up to six months, and their germinationand viability was compared with that of untreated seeds storedunder the same conditions. Seeds that had been osmoconditionedprior to storage retained a high rate of germination and germinatedto a high final percentage (from 80 to 50 per cent) at both15 and 25 °C throughout the storage period. By contrast,both the rate and total level of germination of untreated pepperseeds declined rapidly at both germination temperatures, andby three months of storage the total level of seed viabilitywas already less than 10 per cent. Seeds that were first storedat 35 °C, and then osmoconditioned just prior to germination,showed a decline in germinability which when tested at 25 °Cwas the same as for untreated seeds, while tested at 15 °Coccurred at a slightly slower rate than for untreated seeds. Moist chilling (cold stratification) is well-known as a simpleand effective means of overcoming physiological seed dormancyand enhancing germination of many temperate tree and shrub species.However, the apparent activation of intracellular repair mechanismsin non-dormant black spruce (Picea mariana) seeds followingmoist chilling, has not previously been reported. The presentcontribution records the beneficial effects of moist chillingafter accelerated ageing (40°C/98% relative humidity) for3–10 d of black spruce seeds. This treatment was maximallyeffective after 7 d of accelerated ageing, increasing germinationfrom 43 to 61%. Ultrastructural examination of embryo cellsindicated that intracellular activation occurred during moistchilling even when seeds had not been subjected to acceleratedageing, and that the increasing levels of damage accumulatingover the 3–7 d period of ageing were substantially reversedduring subsequent moist chilling. The effects of temperature, 40–85 °C, on the permeabilityand germinability of the hard seeds of the pioneer tree Rhusjavanica L. with a fire syndrome were studied. The temperatureeffective for removal of the water-impermeable coat dormancyof the seeds was 55 ± 7·4 °C. With increasingtemperature, shorter exposure became sufficient to render theseeds permeable, but at temperatures above 75 °C, heat impairmentof germinability resulted in less than 60% germinability, evenwith long exposure. The most favourable regimes among thosetested were temperatures of 65–75 °C for durationsof 30–120 min, which frequently occur on denuded groundduring the midday hours of clear spring or summer days.
