To explore the cause of difference in photosynthetic performance between different cultivars of crops, leaf net photosynt rate (P N) and photosystem 2 (PS2) photochemical efficiency (Fv/Fm), apparent quantum yield of carbon assimilation (c), electron transport rate, photophosphorylation activity, etc. were measured in two soybean cultivars, Heinong 42 and Heinong 37. At pod setting and filling, significant differences in P N between them were observed. The former with a higher P N (from 7 to 38 %) had a significantly higher leaf thickness, leaf dry mass/area (LMA), chlorophyll content, soluble protein content, apparent quantum yield of electron transport through PS2 (e), carboxylation efficiency (CE), and ribulose-1,5-bisphosphate carboxylase (RuBPC) activity. The significantly higher P N of Heinong 42 is mainly due to its higher content and activity of RuBPC.  相似文献

Effects of N source and media-N and P levels were examined on growth, N uptake, and N2 fixation ofAzolla pinnata withAnabaena azollae association (azolla) at two inoculum-P concentrations. Each expeiment was conducted for 7 days in a growth chamber using azolla at a predetermined inoculum-P concentration and the growth media containing a combination of four levels of P (0, 15, 75, and 200 M) and three levels (0, 1, and 5 mM) of either15N-enriched NH 4 + as ammonium sulfate or15N-enriched NO 3 as potassium nitrate. Nitrogen uptake and N2 fixation were measured by15N isotopic dilution method. Tissue P and N, N uptake, and N2-fixation increased with increasing P concentration in the media regardless of the inoculum-P level of azolla. Increasing P concentration in the media increased growth of azolla at low inoculum P, but the effect on high inoculum-P azolla was either small or absen. High inoculum-P concentration resulted in increased growth, tissue-N and P concentrations, N uptake, and N2 fixation by azolla. Ammonium in the growth media caused larger increase in tissue-N and greater repression of N2 fixation than equimolar concentration of NO 3 . In the presence of NH 4 + or NO 3 , in the growth media, N uptake by azolla exceeded the corresponding decrease in N2 fixation, resulting in an overall increase in tissue-N concentration. Phosphorus in the media tended to negate the inhibitory effect of NH 4 + or NO 3 on N2 fixation. A multiple regression model showed that the effect of tissue-N on N2 fixation was negative while that of tissue-P was positive. Therefore, a relative change in tissue-N and P appeared to regulate N2 fixation. Tissue-N and P had similar effects on relative growth rate of azolla also. The source level in the olive cultivar Leccino was varied by girdling at different stages of fruit growth. Afterwards, the effects on gas exchange, fruit growth, and ripening and blooming were studied. Girdling during fruit growth did not significantly influence net photosynthetic rate (P N) except in the last phase of fruit growth when the P N was reduced. In the girdled branch, P N began to decrease at the onset of starch accumulation because fruit growth ceased. In mid-November stomatal conductance (g s) and transpiration rate (E) were also reduced by girdling, whereas sub-stomatal CO2 concentration (C i) increased in leaves from the girdled branches. The total chlorophyll content (Chl) tended to decrease in parallel with the reduced P N. Girdling did not substantially influence the leaf and shoot water contents. The large availability of assimilates seems to cause an earlier fruit ripening. In general, girdling increased fruit dry mass. Healing before the time when the majority of pulp growth occurs reduced the effect of girdling. June girdling increased the pit dry mass. Girdling at the beginning of August and September, compared to the control, increased the pulp dry mass, but the pit dry mass did not differ with respect to the control. The percentage of oil in the fruit, on a dry mass basis, increased with August and September girdlings, but the percentage of oil in the pulp did not change. Girdling reduced shoot growth, but the internode length was unchanged. Girdling slightly stimulated differentiation of flower buds.  相似文献

The rates of growth, cell size, elemental and biochemical composition ofDunaliella primolecta were monitored during exponential growth after conditioning over three weeks in media containing different concentrations of nitrogen. The rate of growth, measured both as cell density and cell volume, was correlated positively with the N concentration of the medium (P<0.01). N-starved cells did not grow and died within three days. Cell volume and dry weight per cell were significantly higher (P<0.01) in the N-low (0.45 g-at 1–1 NO3-N) condition than in the N-high I condition (3.53 g-at 1–1 NO3-N). In the N-high II condition the addition of 1.87 g-at 1–1 NH4-N to 3.53 g-at 1–1 NO3-N did not significantly enhance growth.The composition was influenced by the availability of nitrogen. The amount of protein per cell decreased significantly from 20.1 to 9.7 pg with reduced nitrate availability between the N-high condition and the N-low condition (P<0.01). The associated carbohydrate content increased significantly from 10.6 to 26.74 pg per cell (P<0.001). These findings confirm the potential for significant variability in the composition ofD. primolecta, both in the natural environment and following experimental manipulation.  相似文献

The effect of glycerol on xylose-to-xylitol bioconversion by Candida guilliermondii was evaluated by its addition (0.7 and 6.5 g/l) to semidefined media (xylose as a substrate). The glycerol concentrations were chosen based on the amounts produced during previous studies on xylitol production by C. guilliermondii. Medium without glycerol addition (control) and medium containing glycerol (53 g/l) in substitution to xylose were also evaluated. According to the results, the addition of 0.7 g/l glycerol to the fermentation medium favored not only the yield (Y P/S = 0.78 g/g) but also the xylitol productivity (Q P = 1.13 g/l/h). During the xylose-to-xylitol bioconversion, the formation of byproducts (glycerol and ethanol) was observed for all conditions employed. In relation to the cellular growth, glycerol as the only carbon source for C. guilliermondii was better than xylose or xylose and glycerol mixtures, resulting in a maximum cellular concentration (5.34 g/l).  相似文献

At high concentration (98% or higher, v/v), glycerol induces collapse of acid-denatured cytochrome c into a compact state, the GU state, showing a molten globule character. The GU state possesses a nativelike -helix structure but a tertiary conformation less packed with respect to the native state. The spectroscopic properties of the GU state closely resemble those of the molten globule stabilized by the organic solvent from the native protein (called the GN state), indicating that glycerol can stabilize the molten globule of cytochrome c either from the native or the acid-denatured protein. The GU and the GN states show spectroscopic (and, thus, structural) properties and stabilities comparable to those of molten globules stabilized by different effectors, despite the fact that the mechanisms involved in the molten globule formation may significantly differ. This implies in cytochrome c a hierarchy for the rupture (native-to-molten globule) or the formation (unfolded-to-molten globule) of intramolecular interactions leading to the stabilization of the molten globule state of the protein, independently from the effector responsible for the structural transition, in accord with the sequential model proposed by Englander and collaborators.  相似文献

Trichodesmium sp., isolated from the Great Barrier Reef lagoon, was cultured in artificial seawater media containing a range of Fe concentration. Fe additions stimulated growth, N2 fixation, cellular chlorophyll a content, light-saturated chlorophyll a-specific gross photosynthetic capacity (Pm chla) and the dark respiration rate (Rd chla). Cell yields only doubled for 9 nM Fe relative to zero added Fe, whereas N2 fixation increased 11-fold considerably for 450 nM Fe. The results suggest that N2 fixation of Trichodesmium is more sensitive to Fe limitation than are the cell yields.  相似文献

Ferric ethylenediamine di-(o-hydroxyphenylacetate) (FeEDDHA) and ferric hydroxyethylethylenediaminetriacetic acid (FeHEDTA) were evaluated as Fe sources for hydroponic growth of alfalfa (Medicago sativa L., cv. Mesilla), either dependent on N2 fixation or supplied with NO3. The hydroponic medium was maintained at pH 7.5 by addition of CaCO3. Nitrogen-fixing cultures were inoculated with Rhizobium meliloti 102 F51 and grown in medium without added nitrogen. After five to seven weeks of growth under greenhouse conditions, plants were harvested. Nitrogen fixation was measured by the acetylene reduction method.When FeEDDHA was supplied, growth of alfalfa, whether dependent on N2 fixation or supplied with NO3, was severely limited at concentrations typically used in hydroponic medium (10 or 20 M). Maximum yield of NO3-supplied alfalfa was obtained at 100 M while maximum yield of N2-fixing alfalfa was obtained in the range of 33 to 200 M FeEDDHA. Nodule fresh weights and N2 fixation rates increased with FeEDDHA concentration up to 33 M and remained essentially constant up to 200 M. With FeHEDTA, maximum yields of both NO3-grown and N2-fixing alfalfa were obtained at 10 M. Growth of NO3-supplied plants was inhibited at 200 M FeHEDTA while growth of N2-fixing plants was inhibited at 100 M FeHEDTA. The numbers of nodules per plant increased between 3.3 and 10 M FeHEDTA; however, inhibition of nodule formation occurred at a concentration of 33 M or higher. Nodule weights per plant and N2 fixation rates were depressed at 3.3 M as well as at 100 M FeHEDTA. The results suggest that alfalfa dependent on N2 fixation is more sensitive to limited Fe availability than alfalfa supplied with NO3.  相似文献

Gas exchange in Gossypium hirsutum L. cv. H-777 as affected by water deficit and growth regulators (IAA, GA3, BAP, ABA, ethrel) was examined. Sixty days after sowing, growth regulators in concentration 50 µM were applied as foliar spray and irrigation was withheld to get desired (moderate and severe) water deficit. All the parameters were measured on the third leaf from the top between 10:00 and 11:00. Net photosynthetic rate (PN), transpiration rate (E), stomatal conductance (gs), carboxylation efficiency (CE), and water potential (w) decreased significantly with the increasing water stress, however, water use efficiency (WUE) was unaffected. Foliar spray with IAA, GA3 and BAP partially counteracted the effect of water deficit on the above parameters except w, which became more negative. ABA and up to some extent ethrel increased WUE and maintained higher w, however, caused further decrease in PN, E, and gs.  相似文献

不同施氮水平对巨桉幼树耐旱生理特征的影响   总被引:1,自引:0,他引:1  
采用盆栽方法,研究了巨桉幼树在N0(不施氮)、N1(1.4g尿素·盆-1)、N2(2.8g尿素·盆-1)3个氮处理水平下,连续干旱不同时间[分别停水0(D0)、3、6、9、12、15、18d]时巨桉的生理响应。结果显示:(1)除D0外,试验期内N1和N2处理的巨桉叶片含水量(LWC)、叶片相对含水量(LRWC)和叶片保水力(LWHC)基本低于N0水平,尤其在干旱中期最为明显,表明在干旱胁迫前施氮可能对巨桉叶片水分生理产生负面作用。(2)干旱初期,氮处理间的可溶性蛋白(SP)和可溶性糖(SS)含量的差异不大,而干旱处理后期(9~18d),N0处理的SP和SS较初期明显增加,但N1和N2处理相对于N0变化较为平缓,表明施氮不利于SP和SS积累;N1和N2处理下脯氨酸(Pro)含量的增幅随着干旱胁迫时间的延长明显大于N0处理。(3)随干旱时间延长干旱程度加重,N1、N2处理巨桉叶片过氧化氢(H2O2)和丙二醛(MDA)含量明显高于N0处理,表明施氮使得巨桉在干旱条件下水分缺乏更为严重,产生更多的活性氧(ROS)。(4)整个干旱处理期内,施氮并未显著改变巨桉的超氧化物歧化酶(SOD)活性和抗坏血酸(AsA)含量,但N1和N2的过氧化物酶(POD)活性明显高于N0。(5)施氮增加了巨桉叶片的色素含量并在干旱初期和中期保持较高水平,在干旱初期(0~3d)增加了巨桉叶片的净光合速率(Pn),但随着干旱时间的延长而迅速下降;施氮(N1、N2)的蒸腾速率(Tr)和气孔导度(Gs)在干旱初期均显著小于N0,但在干旱中后期(6d以后)各处理间差异不显著且均处于极低水平。研究表明,水分充足时施氮有助于增强巨桉的光合同化能力,促进其生长,但遇到持续干旱时施氮更易面临水分亏缺,降低其抵抗干旱的能力,因此在巨桉人工林的经营管理过程中,不应在干旱或季节性干旱即将到来之前施氮,若干旱过程中需施氮则应采取灌溉等途径保证其充足的水分供应。  相似文献

Microsomes were prepared from cultured neonatal rat cardiomyocytes. Incubation of microsomes in buffer containing 5µM CaCl2, 5 mM cholate and 100 nM [3H-]Phosphatidylinosito14,5-bisphosphate (PtdIns(4,5) P2) resulted in the formation of [3H-]InsP 3. GTP-gamma-S (125 µM) stimulated the production of [3H-]InsP 3. Microsomes prepared from phorbol ester-treated (100 nM phorbol 12-myristate 13-acetate, PMA) cardiomyocytes showed decreased activities of basal as well as GTP-gamma-S-stimulated [3H-]Ptdlns(4,5)P 2 hydrolysis. In the microsomes a 15 kD protein was demonstrated to be the major substrate phosphorylated by intrinsic protein kinase C, which was activated by 0.5 mM Ca2+. Addition of phorbol ester (100 nM PMA) enhanced the 32P-incorporation into the 15 kD protein. Protein kinase C, purified from rat brain, in the presence of Ca2+, diglyceride, and phosphatidylserine did not change the phosphorylation pattern any further. In conclusion, it was shown that phorbol ester pretreatment of neonatal rat cardiomyocytes reduces microsomel GTP-gamma-S-stimulated Ptdlns(4,5)P 2-specific phospholipase C activity, as estimated with exogenous substrate, and that in cardiomyocyte microsomes phorbol ester activates protein kinase C-induced 15 kD protein phosphorylation. Fractional rates (%/day) of muscle protein synthesis and degradation of the genotypes Dw/Dw and dw/dw of male White Plymouth Rock chickens were determined by measuring the output of N-methylhistidine (N-MH) in the excreta at 2, 4, and 8 weeks of age. The fractional growth rate of dw/dw was significantly lower (P<0.05) than that of Dw/Dw at 2 weeks of age but not at 4 and 8 weeks of age. No significant differences in the degradation rate (K d; %/day) were found at any age. A significant difference (P<0.05) between genotypes in the rate of synthesis (K s; %/day) was found at 2 weeks of age (Dw/Dw=11.8, dw/dw=9.9) but not at 4 and 8 weeks of age. These results suggest that the dw gene has a depressing effect on the synthesis rate of muscle protein, and the difference between genotypes in the growth rate at the early stage is a reflection of this effect.  相似文献

Plant growth, contents of photosynthetic pigments, photosynthetic gas exchange, and chlorophyll (Chl) fluorescence in soybean [Glycine max (L.) Merr. cv. Heinong37] were investigated after it was inoculated with Sinorhizobium fredii USDA191 or treated with 5 mM (NH4)2SO4 (N5) and 30 mM (NH4)2SO4 (N30), respectively. In the plants following N5 fertilization, not only plant biomass, leaf area, and Chl content, but also net photosynthetic rate (P N), stomatal conductance (g s), carboxylation efficiency (CE), maximum photochemical efficiency (Fv/Fm) of photosystem 2 (PS2), and quantum yield of PS2 (ΦPS2) were markedly improved as compared with the control plants. There were also positive effects on plant growth and plant photosynthesis after rhizobia inoculation, but the effects were much less than those of N5 fertilization. For N30 plants there were no significant positive effects on plant growth and photosynthetic capacity. Plant biomass, P N, and g s were similar to those of N-limited (control) plants. ΦPS2 and photochemical quenching (qP) were obviously declined while content of carotenoids and non-photochemical quenching (qN) were significantly enhanced in N30 treated plants. This indicated that excess N supply may cause some negative effects on soybean plants.  相似文献

以豫西旱地玉米农田为研究对象,设置不同生物炭施用量处理(T0:不施用生物炭;T1:施用生物炭20 t/hm2;T2:施用生物炭40 t/hm2),采用密闭式静态箱法测定N2O排放通量和荧光定量PCR法分析丛枝菌根(arbuscular mycorrhizal,AM)真菌、氨单加氧酶(amoA)、亚硝酸盐还原酶(nirSnirK)以及氧化亚氮还原酶(nosZ)的基因丰度,同时测定土壤理化性状的变化。研究结果表明,随着生物炭施用量的增加,土壤pH和含水量呈增加趋势,土壤有机碳、全氮和铵态氮含量显著提高,土壤容重和硝态氮含量显著降低。T1和T2处理土壤有机碳含量分别较T0显著提高38.44%和71.01%;T1和T2处理土壤铵态氮含量分别较T0显著增加15.89%和30.46%;T2处理土壤全氮含量较T0处理显著提高14.87%;T1和T2处理土壤硝态氮含量分别较T0减少10.57%和21.40%。随着生物炭施用量的增加,AM真菌侵染率显著增加,T1和T2处理分别较T0处理提高71.88%和115.88%;AOA、AOB、nirKnirS基因丰度显著降低;nosZ基因丰度增加。施加生物炭处理的N2O排放通量和累积排放量均低于不施生物炭处理,具体表现为:T0 > T1 > T2。相关分析表明,生物炭施用量与AM真菌基因丰度呈显著正相关;与nosZ基因丰度呈正相关;与AOA、AOB、nirKnirS基因丰度呈极显著负相关。N2O排放通量与AOA、nirKnirS基因丰度呈极显著正相关;与土壤含水量和土壤硝态氮含量呈显著正相关;与AM真菌、nosZ基因丰度、易提取球囊霉素含量、铵态氮含量呈极显著负相关。集成推进树(ABT)分析表明,AOA对N2O排放的影响最大,其次是AM真菌和nirS。总之,生物炭处理改善土壤理化性质、提高土壤AM真菌侵染率、调节硝化、反硝化相关功能基因的丰度,减少N2O气体排放,为旱地农田合理施用生物炭减少N2O气体排放提供理论依据。  相似文献

Nitrogen fixation has been proposed as a mechanism that allows the diazotrophic cyanobacterium, Cylindrospermopsis raciborskii, to bloom in nitrogen‐limited freshwater systems. However, it is unclear whether dinitrogen fixation (N2 fixation) can supplement available dissolved inorganic nitrogen (DIN) for growth, or only provides minimum nitrogen (N) for cell maintenance under DIN deplete conditions. Additionally, the rate at which cells can switch between DIN use and N2 fixation is unknown. This study investigated N2 fixation under a range of nitrate concentrations. Cultures were grown with pretreatments of nitrate replete (single dose 941 μmol  · L?1) and N‐free conditions and then either received a single dose of 941 μmol  · L?1 (N941), 118 μmol  · L?1 (N118) or 0 N. Heterocysts appeared from days 3 to 5 when treatments of high were transferred to N free media (N941:N0), and from day 5 in N941 transferred to N118 treatments. Conversely, transferring cells from N0 to N941 resulted in heterocysts being discarded from day 3 and day 5 for N0:N118. Heterocyst appearance correlated with a detectable rate of N2 fixation and up‐regulation of nifH gene expression, the discard of heterocysts occurred after sequential reduction of nifH expression and N2 fixation. Nitrate uptake rates were not affected by pretreatment, suggesting no regulation or saturation of this uptake pathway. These data demonstrate that for C. raciborskii, N2 fixation is regulated by the production or discard of heterocysts. In conclusion, this study has shown that N2 fixation only provides enough N to support relatively low growth under N‐limited conditions, and does not supplement available nitrate to increase growth rates.  相似文献

Pure singlet molecular oxygen (1O2) generated by thermal decomposition of the 3,3-(1,4-naphthylidene) dipropionate endoperoxide (NDPO2), inhibited respiration of isolated rat liver mitochondria supported by NADH-linked substrates or succinate, but not by N,N,N,N-tetramehyl-p-phenylene-diamine (TMPD)/ascorbate. Under the latter conditions, mitochondria treated with 2.7 mM NDPO2 exhibited a decrease in transmembrane potential () in manner dependent on NDPO2 exposure time. This process was sensitive to the mitochondrial permeability transition inhibitors EGTA, dithiothreitol, ADP, and cyclosporin A. The presence of deuterium oxide (D2O), that increases 1O2 lifetime, significantly enhanced NDPO2-promoted mitochondrial permeabilization. In addition, NDPO2-induced mitochondrial permeabilization was accompanied by DTT or ADP-sensitive membrane protein thiol oxidation. Taken together, these results provide evidence that mitochondrial permeability transition induced by chemically generated singlet oxygen is mediated by the oxidation of membrane protein thiols.  相似文献

Dunaliella tertiolecta (marine) and D. viridis (halophilic) were each trained by serial transfer to growth at salt concentrations previously regarded as the other's domain. D. viridis then had a salt optimum at 1.0–1.5 M sodium chloride whereas that for D. tertiolecta was less than 0–2 M. Nevertheless D. tertiolecta grew faster than the halophil at all salt concentrations up to 3.5 M, the highest at which they were compared.Both species accumulate glycerol, which is necessary for growth at elevated salinities and which responds in its content to water activity (a w ) rather than specifically to salt concentration. Variation in glycerol content is a metabolic process which occurs in the dark from accumulated starch as well as photosynthetically. Regulation of glycerol content by a w does not require protein synthesis. The NADP-specific glycerol dehydrogenase of each of the algae is likely to be directly involved in the regulation of glycerol content. Kinetic studies, together with those described in an earlier publication, show that the enzyme has regulatory properties, and that both glycerol and dihydroxyacetone act as effectors as well as reactants. A mechanism of the reaction is tentatively proposed.  相似文献

Thirty-days-old plants of two cultivars of okra (Hibiscus esculentus L.), Sabzpari and Chinese-red, were subjected for 30 d to two water regimes (100 and 60 % field capacity). Leaf water potential and osmotic potential of both lines decreased significantly with the imposition of drought. Both the leaf pressure potential and osmotic adjustment were much lower in Chinese-red than those in Sabzpari. Chlorophyll (Chl) b content increased, whereas Chl a content remained unchanged and thus Chl a/b ratios were reduced in both lines. Drought stress also caused a significant reduction in net photosynthetic rate (P N), transpiration rate (E), stomatal conductance (g s), and water use efficiency (WUE) especially in cv. Sabzpari. The lines did not differ in intrinsic WUE (P Ngs) or intercellular/ambient CO2 ratio. Overall, the growth of two okra cultivars was positively correlated with P N, but not with g s or P N/E, and negatively correlated with osmotic adjustment.  相似文献

Gas exchanges and leaf water potential (w) of six-years-old trees of fourteen Prunus amygdalus cultivars, grafted on GF-677, were studied in May, when fruits were in active growing period, and in October, after harvesting. The trees were grown in the field under rain fed conditions. Predawn w showed lower water availability in October compared with May. The lowest w values at midday in May increased gradually afterwards, while in October they decreased progressively until night, suggesting a higher difficulty to compensate the water lost by transpiration. However, relative water content (RWC) measured in the morning was similar in both periods, most likely due to some rainfall that occurred in September and first days of October that could be enough to re-hydrate canopy without significantly increasing soil water availability. The highest net photosynthetic rate (P N) was found in both periods early in the morning (08:00–11:00). Reductions in P N from May to October occurred in most cultivars except in José Dias and Ferrastar. In all cultivars a decrease in stomatal conductance (g s) was observed. Photosynthetic capacity (P max) did not significantly change from spring to autumn in nine cultivars, revealing a high resistance of photosynthetic machinery of this species to environmental stresses, namely high temperature and drought. Osmotic adjustment was observed in some cultivars, which showed reductions of ca. 23 % (Duro d' Estrada, José Dias) and 15 % (Tuono) in leaf osmotic potential (). Such decreases were accompanied by soluble sugars accumulation. The Portuguese cultivar José Dias had a higher photosynthetic performance than the remaining genotypes.  相似文献

Photosynthetic characteristics of two hybrid rice combinations, Peiai 64S/E32 and Shanyou 63, were compared at the panicle differentiation stage. As compared with Shanyou 63, the new combination Peiai 64S/E32 showed a significantly higher net photosynthetic rate (P N), apparent quantum yield of carbon assimilation (c), carboxylation efficiency (CE), and photorespiratory rate (R P) as well as leaf chlorophyll content, but a significantly lower dark respiration rate (R D) and compensation irradiance (I c). It also showed a slightly higher photochemical efficiency (Fv/Fm and F/Fm) of photosystem 2, a lower non-photochemical quenching (qN), and a similar CO2 compensation concentration () as compared to Shanyou 63.  相似文献
