The loss of organic material from the roots of forage rape (Brassicanapus L.,) was studied by pulse-labelling 25-d-old non-sterilesand-grown plants with 14CO2. The distribution of 14C withinthe plant was measured at 0, 6 and 13 d after labelling whilst14 C accumulating in the root-zone was measured at more frequentintervals. The rates of 14C release into the rhizosphere, andloss of 14CO2 from the rhizosphere were also determined. Thesedata were used to estimate the accumulative loss of 14C fromroots and loss respiratory 14CO2 from both roots and associatedmicro-organisms. Approximately 17-19% of fixed 14CO2 was translocatedto the roots over 2 weeks, of which 30-34% was released intothe rhizosphere, and 23-24% was respired by the roots as 14CO2. Of the 14C released into the rhizosphere, between 35-51%was assimilated and respired by rhizosphere micro-organisms.Copyright1993, 1999 Academic Press Brassica napus L., carbon loss, carbon partitioning, microbial nutrition, microbial respiration, forage rape, pulse-labelling, rhizodeposition, root respiration, sand culture  相似文献

Well-nodulated soya bean (Glycine max L.) plants were allowedto assimilate 13CO2 for 10 h in the light, under steady-stateconditions in which CO2 concentration and 13C abundance wereboth strictly controlled at constant levels. The respiratoryevolution of 13CO2 from roots and nodules and 13C incorporationinto various metabolic fractions were measured during the 13CO2feeding and subsequent 48 h chase period. CO2 respired from nodules was much more rapidly labelled with13C than that from roots. The level of labelling (percentageof carbon currently assimilated during the 13COM2 feeding period)of CO2 respired from nodules reached a maximum of about 87 percent after 4 h of steady-state l3CO2 assimilation and thereafterremained fairly constant. The absolute amount of labelled carbonevolved by the respiration of the nodules during the 10 h 13CO2feeding period was 1·5-fold that of root respiration. These results demonstrated that the currently assimilated (labelled)carbon was preferentially used to support nodule respiration, while root respiration relied considerably on earlier (non-labelled)carbon reserved in the roots. Sucrose pools were mostly composed of currently assimilatedcarbon in all tissues of the plants, since the levels of labellingaccounted for 86–91 per cent at the end of the 13CO2 feeding. In the nodules, the kinetics and levels of sucrose labellingwere in fairly good agreement with those of respired CO2, whilein the roots, the level of labelling of respired CO2 was significantlylower than that of sucrose. Succinate and malate were highly labelled in both roots andnodules but they were labelled much more slowly than sucroseand respired CO2. The kinetics and levels of labelling of theseKrebs cycle intermediates resembled those of major amino acidswhich are derived directly from Krebs cycle intermediates. Itis suggested that large fractions of organic acids in noduleswere physically separate from the respiration site.

Kouchi, H., Yoneyama, T. and Akao, S. 1986. Compartmental analysisof the partitioning of photo-assimilated carbon in nodulatedsoybean plants during the light period.—J. exp. Bot. 37:994–1005. Dynamics of the partitioning of photo-assimilated carbon invegetative nodulated soybean (Glycine max L.) plants in thelight period was investigated by compartmental analysis basedon data from steady-state 13CO2 assimilation experiments. Themodel assumes a total of 18 compartments consisting of activeand temporary storage pools for soluble materials, starch andstructural materials in leaves, stems plus petioles, roots andnodules together with respired carbon from the roots and nodules.Carbon flow between compartments was described by 22 rate parameters. The rate parameters were evaluated by a non-linear least squaresearch method to optimize the fitness of the simulated resultswith the experimental tracer distribution. The compartment model was well applicable to interpret the carbonpartitioning in whole plants. The analysis showed that: (I)The largest carbon flux during the light period was to storagematerials (starch and temporary storage soluble pools) in theabove-ground parts. The total flux to storage pools was considerablylarger than the transporting flux to below-ground parts. (2)The main carbon flux to the nodules was via direct phloem pathwaysfrom the shoot and not via the compartment of root soluble materials. This flux was 72% of the total carbon flux from the shoot tothe nodulated root system. (3) A large amount of carbon wasreturned to the shoot from below-ground parts. The total returnof carbon flux to the shoot (85% from nodules) was equivalentto 54% of the total influx of carbon to below-ground parts.Direct carbon transfers between roots and nodules were relativelysmall. Rising atmospheric CO2 may stimulate future forest productivity, possibly increasing carbon storage in terrestrial ecosystems, but how tropospheric ozone will modify this response is unknown. Because of the importance of fine roots to the belowground C cycle, we monitored fine-root biomass and associated C fluxes in regenerating stands of trembling aspen, and mixed stands of trembling aspen and paper birch at FACTS-II, the Aspen FACE project in Rhinelander, Wisconsin. Free-air CO2 enrichment (FACE) was used to elevate concentrations of CO2 (average enrichment concentration 535 µl l-1) and O3 (53 nl l-1) in developing forest stands in 1998 and 1999. Soil respiration, soil pCO2, and dissolved organic carbon in soil solution (DOC) were monitored biweekly. Soil respiration was measured with a portable infrared gas analyzer. Soil pCO2 and DOC samples were collected from soil gas wells and tension lysimeters, respectively, at depths of 15, 30, and 125 cm. Fine-root biomass averaged 263 g m-2 in control plots and increased 96% under elevated CO2. The increased root biomass was accompanied by a 39% increase in soil respiration and a 27% increase in soil pCO2. Both soil respiration and pCO2 exhibited a strong seasonal signal, which was positively correlated with soil temperature. DOC concentrations in soil solution averaged ~12 mg l-1 in surface horizons, declined with depth, and were little affected by the treatments. A simplified belowground C budget for the site indicated that native soil organic matter still dominated the system, and that soil respiration was by far the largest flux. Ozone decreased the above responses to elevated CO2, but effects were rarely statistically significant. We conclude that regenerating stands of northern hardwoods have the potential for substantially greater C input to soil due to greater fine-root production under elevated CO2. Greater fine-root biomass will be accompanied by greater soil C efflux as soil respiration, but leaching losses of C will probably be unaffected.  相似文献

The fluid exudation rates and the ionic composition of the exudatesof isolated maize roots were compared with those of similarroots from which the epidermal and outer cortical cells hadbeen removed. The results indicate that in low ionic strengthmedia the salt fluxes into the exudation stream are severaltimes larger in the control than in the treated roots. As theionic strength of the bathing medium increases to 10 mM KClor greater, the salt fluxes become equal. A mechanism to accountfor this behaviour is discussed. The values of Lp, the hydraulic conductivity of the roots toradial water flow, are 0.26 x 10–6 cm sec–1 atm–1for treated and 0.14 x 10–6 cm sec–1 atm–1for control roots, but a statistical technicality precludeda significance test on this apparent difference. The kinetics of tritiated water exchange from external mediumto the exudation fluid in both control and treated roots werenot significantly different. The entry of labelled water seemsto be achieved by the fluid exudation rate in both cases; thusthe barrier to water movement to the exudate compartment remainsintact after removal of the epidermis.  相似文献

The specific respiration rates of nodulated root systems, ofnodules and of roots were determined during active nitrogenfixation in soya bean, navy bean, pea, lucerne, red clover andwhite clover, by measurements on whole plants before and afterthe removal of nodule populations. Similar measurements weremade on comparable populations of the six legumes, lacking nodulesbut receiving abundant nitrate-nitrogen, to determine the specificrespiration of their roots. All plants were grown in a controlled-environmentclimate which fostered rapid growth. The specific respiration rates of nodulated root systems ofthe three grain and three forage legumes during a 7–14-dayperiod of vegetative growth varied between 10 and 17 mg CO2g–1 (dry weight) h–1. This mean value consistedof two components: a specific root respiration rate of 6–9mg CO2 g–1 h–1 and a specific nodule respirationrate of 22–46 mg CO2 g–1 h–1. Nodule respirationaccounted for 42–70 per cent of nodulated root respiration;nodule weight accounted for 12–40 per cent of nodulatedroot weight. The specific respiration rates of roots lackingnodules and utilizing nitrate nitrogen were generally 20–30per cent greater than the equivalent rates of roots from nodulatedplants. The measured respiratory effluxes are discussed in thecontext of nitrogen nitrogen fixation, nitrate assimilation. Kouchi, H., Akao, S. and Yoneyama, T. 1986. Respiratory utilizationof 13C-labelled photosynthate in nodulated root systems of soybeanplants.—J. exp. Bot. 37: 985–993. An improved method for the measurement of respiratory utilizationof current photosynthate in the nodulated root system of water-culturedsoybean (Glycine max L.) plants was developed using a steady-state13CO2 labelling technique. Well-nodulated plants at the latevegetative stage were allowed to assimilate 13CO2 for 10 h incontinuous light at a constant CO2 concentration with a constant13C abundance. The respiratory evolution of 13CO2 from rootsand nodules was measured continuously throughout the periodof 13CO2 assimilation and during a subsequent 36 h chase periodby using a differential infrared 13CO2 analyser. The plantswere grown with nitrogen-free or (15 mmol dm–3)-containing culture solution for 3 d before13CO2 assimilation. In plants grown without , nodule respiration averaged 69% of the total respiration of the undergroundparts over the full experimental period and the CO2 respiredreached an apparent isotopic equilibrium at 80–85% labellingafter initiating 13CO2 assimilation. By contrast, the CO2 respiredfrom the roots did not reach an isotopic equilibrium and labellingwas only 56% at the end of exposure to 13CO2 These findingsdemonstrated that nodule respiration is strongly dependent onrecently assimilated carbon compared with root respiration. Plants supplied with in the culture solution showed a decreased rate of nodule respirationand a slightly increased rate of root respiration. The extentsand time courses of labelling of respired CO2 from both theroots and nodules were similar in the presence and absence of except that the maximum level of labelling of CO2 derived from nodule respiration in plantswith was significantly higher (about 91%) than for plants growing without . The amounts of carbon released into soil from roots of wheatand barley seedlings grown under three environmental conditionsfor 3 weeks with shoots in constant specific activity 14CO2are reported. This carbon loss was measured as respired 14CO2from both the root and the accompanying microbial populationand as root derived 14C-labelled organic C compounds in thesoil. With a 16 h photoperiod, growth at 15 ?C constant or 18?C day/14 ?C night gave a loss of 33–40% of the totalnet fixed carbon (defined as 14C retained in the plant plus14C lost from the root). The proportion of 14C translocatedto the roots that was released into the soil did not changewith temperature, so carbon distribution within the plant musthave changed. With a 12 h photoperiod and a temperature regimeof 18 ?C/14 ?C carbon loss from the roots was decreased to 17–25%of the total fixed carbon. Key words: Cereals, Roots, Carbon loss  相似文献

Various plant and environmental factors influence the hydraulicproperties for roots, which were examined using negative hydrostaticpressures applied to the proximal ends of individual excisedroots of a common succulent perennial from the Sonoran Desert,Agave deserti Engelm. The root hydraulic conductivity, Lp, increasedsubstantially with temperature, the approximately 4-fold increasefrom 0.5°C to 40°C representing a Q10 of 1.45. Suchvariations in Lp with temperature must be taken into accountwhen modelling water uptake, as soil temperatures in the rootzone of such a shallow-rooted species vary substantially bothdaily and seasonally. At 20°C, Lp was 2.3 x 10–7 ms{macron}1MPa{macron}1for 3-week-old roots, decreasing to abouthalf this value at 10 weeks and then becoming approximatelyhalved again at 6 months. For a given root age, Lp for rainroots that are induced by watering as lateral branches on theestablished roots (which arise from the stem base) was aboutthe same as Lp for established roots. Hence, the conventionalbelief that rain roots have a higher Lp than do establishedroots is more a reflection of root age, as the rain roots tendto be shed following drought and thus on average are much youngerthan are established roots. Unlike previous measurements onroot respiration, lowering the gas-phase oxygen concentrationfrom 21% to 0% or raising the carbon dioxide concentration from0.1% to 2% had no detectable effect on Lp for rain roots andestablished roots. Lp for rain roots and established roots wasdecreased by an average of 11% and 35% by lowering the soilwater potential from wet conditions (soil=0 kPa) to {macron}40kPa and {macron}80 kPa, respectively. Such decreases in Lp mayreflect reduced water contact between soil particles and theroot surface and should be taken into account when predictingwater uptake by A. deserti. The effects of three ranges of CO2 concentration on growth, carbon distribution and loss of carbon from the roots of maizegrown for 14 d and 28 d with shoots in constant specific activity14CO2 are described. Increasing concentrations of CO2 led toenhancement of plant growth with the relative growth rate (RGR)of the roots affected more than the RGR of the shoots. Between16% and 21% of total net fixed carbon (defined as 14C retainedin the plant plus 14C lost from the root) was lost from theroots at all CO2 concentrations at all times but the amountsof carbon lost per unit weight of plant decreased with time.Possible mechanisms to account for these observations are discussed. In hydroponically grown Lycopersicon esculentum (L.) Mill. cv.F144 the site of NO3 reduction and assimilation withinthe plant was shifted from the shoot to the root by salinity.Uptake of NO3 from the root solution was strongly inhibitedby salinization. Consequently, NO3 concentrations inthe leaf, stem and root tissues as well as the nitrate reductaseactivities of the leaves were lower in salinized than in controlplants. Lower NO3, but higher reduced-N, concentrationswere observed in the xylem sap as a result of the enhanced participationof the root in NO3 reduction in salinized plants. Lowerstem K+ concentrations and leaf malate concentrations were foundin salinized compared to control plants which indicates reducedfunctioning of the K+–shuttle in the salinized plants. Incorporation of inorganic carbon by the root was determinedby supplying a pulse of NaH14CO3 followed by extraction andseparation of the labelled products on ion exchange resins. The rate of H14CO3 incorporation was c. 2-fold higherin control than in salinized plants. In salinized plants theproducts of H14CO3 incorporation within the roots werediverted into amino acids, while the control plants divertedrelatively more 14C into organic acids. Products of inorganiccarbon incorporation in the roots of salinized plants providean anaplerotic source of carbon for assimilation of reducedNO3 into amino acids, while in control plants the productswere predominantly organic acids as part of mechanisms to maintainionic balance in the cells and in the xylem sap. During a period of sulphate deprivation, roots of Macroptiliumatropurpureum responded by increasing their uptake capacityat the plasma membrane. This effect was apparent both in intactplants and in tissues excised prior to uptake. In experiments using excised root systems previousy labelledwith 35SO42- the rate of tracer transport to the xylem was muchgreater in roots subsequently deprived of external sulphatethan in those supplied with unlabelled sulphate. Cultivated Agave mapisaga and A. salmiana can have an extremelyhigh above-ground dry-weight productivity of 40 Mg ha–1yr–1. To help understand the below-ground capabilitiesthat support the high above-ground productivity of these Crassulaceanacid metabolism plants, roots were studied in the laboratoryand in plantations near Mexico City. For approximately 15-year-oldplants, the lateral spread of roots from the plant base averaged1.3 m and the maximal root depth was 0.8 m, both considerablygreater than for desert succulents of the same age. Root andshoot growth occurred all year, although the increase in shootgrowth at the beginning of the wet season preceded the increasein growth of main roots. New lateral roots branching from themain roots were more common at the beginning of the wet season, which favoured water uptake with a minimal biomass investment, whereas growth of new main roots occurred later in the growingseason. The root: shoot dry weight ratio was extremely low, less than 0.07 for 6-year-old plants of both species, and decreasedwith plant age. The elongation rates of main roots and lateralroots were 10 to 17 mm d–1, higher than for various desertsucculents but similar to elongation rates for roots of highlyproductive C3 and C4 agronomic species. The respiration rateof attached main roots was 32 µmol CO2 evolved kg–1dry weight s–1 at 4 weeks of age, that of lateral rootswas about 70% higher, and both rates decreased with root age.Such respiration rates are 4- to 5-fold higher than for Agavedeserti, but similar to rates for C3 and C4 agronomic species. The root hydraulic conductivity had a maximal value of 3 x 10–7ms–1 MPa–1 at 4 weeks of age, similar to A. deserti. The radial hydraulic conductivity from the root surface to thexylem decreased and the axial conductivity along the xylem increasedwith root age, again similar to A. deserti. Thus, although rootsof A. mapisaga and A. salmiana had hydraulic properties perunit length similar to those of a desert agave, their highergrowth rates, their higher respiration rates, and the greatersoil volume explored by their roots than for various desertsucculents apparently helped support their high above-groundbiomass productivity Key words: Crassulacean acid metabolism, productivity, root elongation rate, root system, water uptake  相似文献

Fully symbiotic or nitrate treated (3 d, 4·0 mol m–3)soybean (Glycine max [L.] Merr.) cv. Bragg and a nitrate tolerantsupernodulating soybean mutant nts 1007 were exposed to 13Cenriched CO2 for a period of 10 h. During this period and forthe subsequent 24 h, continuous measurements of 13CO2 and 12CO2evolution of their root systems were undertaken. Three harvestsduring the experiment allowed determinations of the distributionof recently fixed carbon in different plant organs. These measurementsindicated higher dependence of N2 fixation in nts 1007 on recentlyfixed carbon (RFC) by showing elevated RFC concentrations innodules as well as their augmented respiration. Root respirationof both genotypes was generally more reliant on stored carbon. Nitrate induced in all measured parameters a clear responsein the mutant analogous to the wild type, but quantitative differencesremained throughout. Nodule respiratory activity, the relativespecific activity (RSA), and the utilization of RFC were substantiallyreduced, but remained higher in nts 1007 than in Bragg, whilethe demand of roots for RFC increased in Bragg more than inthe supernodulator. The elevated carbon requirement of the nodulecomplement of the mutant and a high dependence on recently fixedcarbon could be attributed to higher nodule growth and maintenancecosts of the supernodulating genotype and were not associatedwith augmented nitrogen fixation activity. This less efficientutilization of carbon and the associated almost parasitic characterof the nodule complement of nts 1007 is considered to be thecause of reduced growth of the mutant. No evidence was foundfor a physiologically based nitrate tolerance in terms of nitrogenfixation. Root growth in chickpea (Cicer arietinum) has been studied fromthe early vegetative phase to the reproductive stage in orderto elucidate its growth and maintenance respiration and to quantifythe translocation of assimilates from shoot to root. A carbonbalance has been drawn for this purpose using the growth andrespiration data. The increase in the sieve tube cross-sectionalarea was also followed simultaneously. Plants growing in a nutrient culture medium were studied todetermine the relative growth rate (RGR) 5–60 d aftergermination. RGR declined from 113 to 41 mg d–1 g–1during the measurement period. Simultaneous with the RGR analysis, respiration rate was also measured using an oxygen electrode. The respiration rate declined as the plants aged and a drasticreduction was recorded following anthesis. The relationshipbetween RGR and respiration rate was used to extrapolate themaintenance respiration (m) and growth respiration (1/YEG).The respiration quotient (r.q.) of the roots was 1.2 and theQ10 in the range 20–25 °C was 2·2. A carbon balance for the roots was constructed by subtractingthe carbon lost during respiration from that gained during growth. The roots were found to respire no less than 80% of the carbontranslocated. The increase in the cross-sectional area composed of sieve tubeswas measured near the root-shoot junction as the plants grew.Chickpea has storied sieve plates which simplifies these measurements.Their cross-sectional area increased during growth mainly becauseof an increase in sieve tube number. The diameter of individualsieve tubes remained constant. Specific mass transfer (SMT) values for seive tubes into theroots have been computed during various stages of growth. SMTvalues were relatively constant before anthesis (approx. 6·5g h–1 cm–2), but decreased following anthesis. Wedid not evaluate possible retranslocation from roots: any suchretranslocation would have the effect of increasing our SMTvalues. Respiratory oxygen consumption by roots was 1·4- and1·6-fold larger in NH+4-fed than in NO-3-fed wheat (Triticumaestivum L.) and maize (Zea mays L.) plants respectively. Higherroot oxygen consumption in NH+4-fed plants than in NO-3-fedplants was associated with higher total nitrogen contents inNH+4-fed plants. Root oxygen consumption was, however, not correlatedwith growth rates or shoot:root ratios. Carbon dioxide releasewas 1·4- and 1·2-fold larger in NO+3-fed thanin NH+4-fed wheat and maize plants respectively. Differencesin oxygen and carbon dioxide gas exchange rates resulted inthe gas exchange quotients of NH-4-fed plants (wheat, 0·5;maize, 0·6) being greatly reduced compared with thoseof NO-3-fed plants (wheat, 1·0; maize, 1·1). Measuredrates of HCO-3 assimilation by PEPc in roots were considerablylarger in 4 mM NH+4-fed than in 4 NO-3 plants (wheat, 2·6-fold;maize, 8·3-fold). These differences were, however, insufficientto account for the observed differences in root carbon dioxideflux and it is probable that HCO-3 uptake is also importantin determining carbon dioxide fluxes. Thus reduced root extension in NH+4-fed compared with NO-3-fedwheat plants could not be ascribed to differences in carbondioxide losses from roots.Copyright 1993, 1999 Academic Press Triticum aestivum, wheat, Zea mays, maize assimilation, ammonium assimilation, root respiration  相似文献

Seedlings of Ricinus communis L. cultivated in quartz sand weresupplied with a nutrient solution containing either 1 mol m–3NO3 or 1 mol m–3 NH+4 as the nitrogen source. Duringthe period between 41 and 51 d after sowing, the flows of N,C and inorganic ions between root and shoot were modelled andexpressed on a fresh weight basis. Plant growth was clearlyinhibited in the presence of NH+4. In the xylem sap the majornitrogenous solutes were nitrate (74%) or glutamine (78%) innitrate or ammonium-fed plants, respectively. The pattern ofamino acids was not markedly influenced by nitrogen nutrition;glutamine was the dominant compound in both cases. NH+4 wasnot transported in significant amounts in both treatments. Inthe phloem, nitrogen was transported almost exclusively in organicform, glutamine being the dominant nitrogenous solute, but theN-source affected the amino acids transported. Uptake of nitrogenand carbon per unit fresh weight was only slightly decreasedby ammonium. The partitioning of nitrogen was independent ofthe form of N-nutrition, although the flow of nitrogen and carbonin the phloem was enhanced in ammonium-fed plants. Cation uptakerates were halved in the presence of ammonium and lower quantitiesof K+, Na+ and Ca2+ but not of Mg2+ were transported to theshoot. As NH+4 was balanced by a 30-fold increase in chloride in thesolution, chloride uptake was increased 6-fold under ammoniumnutrition. We concluded that ammonium was predominantly assimilated inthe root. Nitrate reduction and assimilation occurred in bothshoot and root. The assimilation of ammonium in roots of ammonium-fedplants was associated with a higher respiration rate.
