Oral squamous cell carcinoma (SCC) represents more than 90% of all head and neck cancers as reported by Hermans (Cancer Imaging, 5(Spec No A), S52–S57, 2005), which draws attention of investigative research for novel predictive metabolic biomarkers to understand the malignancy induced biochemical perturbations occurring at molecular level. In the present work, proton HR-MAS NMR spectroscopic studies have been performed on resected human oral SCC tumor tissues, its neighboring margins and bed tissues (n = 159), obtained from 36 patients (n = 27 training set; n = 9 unknown test set), for the identification of metabolic fingerprints. The proton NMR spectra were then subjected to chemometric unsupervised PCA and supervised OSC-filtered PCA and PLS-DA multivariate analysis. Application of PLS-DA on orthogonally signal corrected training data-set (n = 120 tissue specimens; 27 patients) allowed >95% correct classification of malignant tissues from benign samples with >98% specificity and sensitivity. The OSC-PLS-DA model thus constructed was used to predict the class membership of unknown tissue specimens (n = 39) obtained from nine patients. These tissue samples were correctly predicted in its respective histological categories with 97.4% diagnostic accuracy. The regression coefficients obtained from OSC-filtered PLS-DA model indicated that malignant tissues had higher levels of glutamate, choline, phosphocholine, lactate, acetate, taurine, glycine, leucine, lysine, isoleucine and alanine, and lower levels of creatine and PUFA, representing altered metabolic processes (lipidogenesis, protein synthesis, and volume regulation) during tumor progression. Thus proton HR-MAS MR spectroscopy could efficiently identify the metabolic perturbations of malignant tumor from non-malignant bed and margins tissue specimens, which may be helpful in understanding the extent of tumor penetration in neighboring tissues.  相似文献

A cDNA clone encoding the photosystem I subunit, PSI-G was isolated from barley using an oligonucleotide specifying a partial amino acid sequence from a 9 kDa polypeptide of barley photosystem I. The 724 bp sequence contains an open reading frame encoding a precursor polypeptide of 15 107 kDa. Import studies using the in vitro expressed barley PsaG cDNA clone demonstrate that PSI-G migrates with an apparent molecular mass of 9 kDa on SDS-polyacrylamide gels together with PSI-C (subunit-VII). The previous assignment of the gene product of PsaG from spinach as subunit V (Steppuhn J, Hermans J, Nechushtai R, Ljungberg U, Thümmler F, Lottspeich F, Herrmann RG, FEBS Lett 237: 218–224, 1988) needs to be re-examined. The expression of the psaG gene is light-induced similar to other barley photosystem I genes. A significant sequence similarity to PSI-K from Chlamydomonas reinhardtii was discovered when a gene database was searched with the barley PSI-G amino acid sequence. Extensive sequence similarity between the nuclear-encoded photosystem I subunits has not previously been found. The observed sequence similarity between PSI-G and PSI-K suggests a symmetric location of these subunits in the photosystem I complex. The hydropathy plot of the barley PSI-G polypeptide indicates two membrane-spanning regions which are also found at the corresponding locations in the PSI-K polypeptide. PSI-G and PSI-K probably have evolved from a gene duplication of an ancestral gene.  相似文献

We have used refractive index matching to determine the concentration of protein in the fibers in fibrin clots and of needlelike crystals of native fibrinogen. Our results are in agreement with those of Carr and Hermans [(1978) Macromolecules 11 , 46–50], as determined by light scattering—namely, that protein makes up about 20% of the volume of the fiber. However, we have found that the protein concentration is strongly dependent on ionic strength. An increase in ionic strength caused a substantial drop in the protein concentration. In a buffer containing 100 mM NaCl, the protein concentration was 26.6–29.8 g of protein per 100 cm3 of polymer, and at 200 mM NaCl it was reduced to 22.1–23.1 g/100 cm3.  相似文献

The floral morphology of the southern African genera of Orchideae-Habenariinae (Bonatea, Cynorkis, Habenaria, Platycoryne, Stenoglottis, Centrostigma and Roe-perocharis) is surveyed paying special attention to the gynostemium. Ontogenetic data are provided for the species from which adequate material was available. It is shown that the floral architecture is essentially an elaboration and complication of that found in the better known Orchidinae. The structural similarities are particularly evident in the early ontogeny. Although the tribe Orchideae is commonly said to have gynostemia with erect anthers, a few Habenariinae are reported here to have reflexed anthers. In most cases both 'auricles' (filament excrescences) and 'basal bulges' (staminodes) are united to form the lateral gynostemium appendages. The primordia of both structures are clearly recognizable in the early ontogeny in all species studied. In Habenaria dregeana the basal bulges are only basally fused to the auricles, but in their main portion become adnate to the lip and petal bases: the auricles then solely form the lateral gynostemium appendages. It is suspected that this occurs also in other species not studied here. Systematic and phylogenetic aspects of the southern African representatives of the Habenariinae are discussed: the generic separation of Bonatea, Platycoryne and Centrostigma from Habenaria does not appear justified. Cynorkis, Roeperocharis and Stenoglottis are morphologically dissimilar to Habenaria. Based on the findings in the southern African taxa the status of the Habenariinae, Orchidinae, Orchideae and Diseae is discussed: there is no clear distinction between Habenariinae and Orchidinae; while the Diseae seem to represent a monophyletic group, the Orchideae are possibly polyphyletic.  相似文献

Anthobothrium laciniatum Linton, 1890 is redescribed based on specimens taken from the dusky shark Carcharhinus obscurus (Lesueur) collected from the Northwestern Atlantic Ocean, and a neotype is designated. A. laciniatum differs from A. cornucopia van Beneden, 1850, A. altavelae Euzet & Ben Hassine, 2002, A. lesteri Williams, Burt & Caira, 2004 and A. spinosum Subhapradha, 1955 in total length. It further differs from A. cornucopia, A. altavelae and A. spinosum in proglottid number, and differs from A. galeorhini Suriano, 2002, A. cornucopia, and A. spinosum in testis number. A. lyndoni n. sp. is described from the sandbar shark C. plumbeus (Nardo). This new species differs from A. laciniatum in ovarian width and from A. cornucopia, A. altavelae, A. galeorhini and A. spinosum in the total number of proglottids. It further differs from A. cornucopia, A. galeorhini, and A. spinosum in total length, and from A. cornucopia and A. galeorhini in the number of testes. A. lyndoni n. sp. differs from A. lesteri in bothridial muscle and ovarian morphology. Anthobothrium caseyi n. sp. is described from Prionace glauca (Linnaeus). This new species differs conspicuously from the other six species of Anthobothrium van Beneden, 1850 (sensu stricto) in the shape of its proglottid laciniations. The taxonomic status of 43 species that have been associated with Anthobothrium is addressed. Taxonomic actions regarding Anthobothrium during the past century have resulted in a polyphyletic taxon.  相似文献

A diagnosis is given for the lecithasterid genus Hysterolecithoides Yamaguti, 1934, which is now found to have two to six (possibly seven) vitelline masses. The species H. frontilatus (Manter, 1969) is returned to the genus, having been considered a member of the bunocotylid genus Neotheletrum by recent authors. It is redescribed from Siganus nebulosus, Moreton Bay, and S. doliatus, Lizard Island, Great Barrier Reef and New Caledonia, with emphasis on the presence of Juel's organ, a uterine seminal receptacle and the blind sac associated with the genital atrium. It differs from its congeners in the trajectory of the pars prostatica which recurves dorsally to the sinus-sac. Oligolecithoides Shen, 1982 is synonymised with Hysterolecithoides and O. trilobatus Shen, 1982 is synomised with H. epinepheli Yamaguti, 1934. Machidatrema León-Règagnon, 1998 is diagnosed, and found to be close to Hysterolecithoides, but differs in the lack of a blind-sac projecting from the dorsal genital atrium, by its tandem testes, the coiling of the uterus between the testes and the ovary, and the ventral excretory pore. M. leonae n. sp. is described from Siganus fuscescens, S. lineatus, S. doliatus, S. corallinus, S. vulpinus and Scarus globiceps at Heron Island, Queensland. It differs from its closest congener, M. akeh, in the muscular and tegumental flap over the genital pore and details of the terminal genitalia. M. chilostoma (Machida, 1980) and M. kyphosi (Yamaguti, 1970) are redescribed from Kyphosus vaigiensis from Heron Island. Neotheletrum Gibson & Bray, 1979 is diagnosed: it differs from Hysterolecithoides in its confluent excretory arms, blind seminal receptacle (no Juel's organ) and uniformly tripartite vitellarium. A cladistic analysis suggests that M. chilostoma and M. kyphosi are not best accommodated in Machidatrema, that Machidatrema (sensu stricto) is monophyletic and that Hysterolecithoides is paraphyletic. Hysterolecithoides and Machidatrema are considered hysterolecithine lecithasterids, whilst Neotheletrum is retained as an opisthadenine bunocotylid.  相似文献

Keratinase from Pseudomonas aeruginosa KS-1 was expressed constitutively as an extracellular protein in Escherichia coli with high specific activity of 3.7 kU/mg. It was purified fourfold as a 33 kDa monomeric protein by Q-Sepharose ion exchange chromatography with a recovery of 95%. It is a serine protease with optimal activity at pH 9 and 50°C. It was stable from pH 4 to 12 for 1 h with a t1/2 of 12 min at 70°C. It hydrolyzed haemoglobin > fibrin > feather keratin > azo-casein > casein > meat protein > gelatin. Among synthetic substrates, it efficiently hydrolyzed N-Suc-ala-ala-pro-phe-pNA, N-Suc-ala-ala-ala-pNA, N-Suc-ala-ala-pro-leu-pNA and also plasmin substrate, d-Val-Leu-Lys-pNA  相似文献

Summary The structure of the ocelli of representatives of four of the five families of archiannelids (Polychaeta: Annelida) was studied by light and electron microscopy. The apparent photoreceptoral organelle in each species is an array of microvilli (rhabdomere). Cilia were observed in the eyes of only a couple of specimens in one species of archiannelid (Nerilla antennata). They were unassociated with the rhabdomeres; we regard them as adventitious. Support is given by this study to the theory that the photoreceptoral organelle of the ancestral annelid was a rhabdomere. Fragilidium fissile is a new species of this rare genus. It somewhat resemblesF. subglobosum. It differs from the latter in having a slot and a pore in the first apical plate 1′ (the nomenclature of dinoflagellate plate designation follows the Kofoid system). Both species are also distinguishable by noticeable differences in Po, 1″″ and 1‴.Peridinium tyrrhenicum is a small species differing from all the other known species ofPeridinium in its shape, apical channel and several plates, especially some of the sulcal components.Alexandrium foedum somewhat resemblesA. balechii, but it differs from the latter in that its width is greater than its length, and in the characters of all the main sulcal plates. The above listed species were found in a sample from the Tyrrhenian Sea. The fourth species,Alexandrium andersoni, is a small dinoflagellate obtained in coastal waters off Cape Cod. It differs from all the other species of the minutum group in the very typical shapes of both the 6″ and the S. s.Pentapharsodinium daleii Indelicato and Loeblich is transferred toPeridinium.  相似文献

Eleocharis cryptica (Cyperaceae), known from pine-oak forest in Durango, Mexico, is described and illustrated. It belongs to Eleocharis subg. Eleocharis ser. Tenuissimae and can be distinguished by its diminutive size, with culms 2–13 mm long (including spikelets 1.3–1.7 mm long), and the folliform prolongations of its upper sheaths. It is the smallest species of Eleocharis thus far known.  相似文献

A new species of the genus Paracobitis, Paracobitis nanpanjiangensis is described from tributaries of the Nanpanjiang River drainage in China. It is distinguished from its congeners, except P. oligolepis and P. wujiangensis, by body scaleless or with rudimentary scales (caudal peduncle with several deeply embedded scales). It can be differentiated from P. wujiangensis by the complete lateral line (vs. incomplete), lower dorsal crest reaching the vertical of origin of anal fin (vs. shorter and higher dorsal crest not reaching the base of anal fin). It is distinguished from P. oligolepis by the following characters: branched dorsal fin with 81/2 (a few 91/2) rays (vs. 91/2), interspaces between bars in front of dorsal fin conspicuously thinner than those behind (vs. vermiform markings), dorsal head without vermiform markings or obscure (vs. clearly vermiform markings on dorsal head), vertebrae 4 + 36–38 (vs. 4 + 39–41).  相似文献
