Cross-linked homopolymers and copolymers of 2-hydroxyethyl methacrylate, HEMA, and ethylene glycol methacrylate phosphate, MOEP, have been synthesized, and the diffusion of water into these systems has been investigated. Only polymers with 0-20 mol % MOEP exhibited ideal swelling behavior as extensive fracturing occurred in the systems with greater than 20 mol % MOEP as the polymers began to swell during water sorption. Gravimetric studies were used in conjunction with magnetic resonance imaging of the diffusion front to elucidate the diffusion mechanism for these systems. In the case of the cross-linked HEMA homopolymer gels, the water transport mechanism was determined to be concentration-independent Fickian diffusion. However, as the fraction of MOEP in the network increased, the transport mechanism became increasingly exponentially concentration-dependent but remained Fickian until the polymer consisted of 30 mol % MOEP where the water transport could no longer been described by Fickian diffusion.  相似文献

The peptides secreted by Tetrahymena cells into inorganic medium were chromatographed. Six fractions showing a marked enzyme-like activity were examined for influence on certain physiological parameters of Tetrahymena. The enzymatically active fractions increased the phagocytic activity of Tetrahymena and decreased its binding capacity for lectins and hormone (insulin), but enhanced insulin imprinting at primary interaction. It remains to be clarified whether these effects were due to the enzymatic or other components of the fractions investigated, or to lack of the compensatory influence of the fractions not studied.  相似文献

Cellulose-derived glucose generated using the supported ruthenium catalyst was applied to poly(3-hydroxybutyrate) [P(3HB)] production in recombinant Escherichia coli. By the reaction with the catalyst at 220°C, 15-20 carbon mol% of cellulose was converted into glucose. The hydrolysate also contained byproducts such as fructose, mannose, levoglucosan, oligomeric cellulose, 5-hydroxymethylfurfural (5-HMF), and furfural together with unidentified compounds. Setting the reaction temperature lower (215°C) improved the ratio of glucose to 5-HMF, which was a main inhibiting factor for the cell growth. Indeed, the recombinant E. coli exhibited better performance on the hydrolysate generated at 215°C and accumulated P(3HB) up to 42 wt%, which was the same as the case of the same concentration of analytical grade glucose. The result indicated that the ruthenium-mediated cellulose hydrolysis has a potency as a useful biorefinery process for production of bio-based plastic from cellulosic biomass.  相似文献

Six transmembrane protein of prostate 2 (STAMP2) is a protein that has been extensively studied due to its association with prostate cancer. Currently, STAMP2 is well known for its critical role in metabolism and modulating inflammatory signals. Even so, the molecular mechanism of STAMP2 activity and its downstream effectors are still largely unknown. Here, we review the current knowledge of STAMP2, and suggest possible explanations for some of its less well-understood features. A few studies suggest that STAMP2 may interact with mitochondria. Considering STAMP2 functions as a potential component of mitochondrial biology may yield valuable insight into this protein.  相似文献

Most chloroplastic proteins are synthesized in the cytoplasm and are transported to their proper location as a posttranslational event. In the present paper we briefly review some aspects of this transport process. Because chloroplasts contain six different locations, one interesting aspect of protein targeting into chloroplasts that we consider is how precursor proteins are targeted to these various locations. One step shared by many proteins is transport across the envelope membranes. Although this process has been well studied, the components of the apparatus that mediate this transport step are mostly unidentified. Strategies to identify components of this transport apparatus are considered.  相似文献

The AMP-activated protein kinase (AMPK), a sensor of cellular energy status found in all eukaryotes, responds to changes in intracellular adenosine nucleotide levels resulting from metabolic stresses. Here we describe crystal structures of a heterotrimeric regulatory core fragment from Schizosaccharomyces pombe AMPK in complex with ADP, ADP/AMP, ADP/ATP, and 5-aminoimidazole-4-carboxamide 1-beta-D-ribofuranotide (AICAR phosphate, or ZMP), a well-characterized AMPK activator. Prior crystallographic studies had revealed a single site in the gamma subunit that binds either ATP or AMP within Bateman domain B. Here we show that ZMP binds at this site, mimicking the binding of AMP. An analogous site in Bateman domain A selectively accommodates ADP, which binds in a distinct manner that also involves direct ligation to elements from the beta subunit. These observations suggest a possible role for ADP in regulating AMPK response to changes in cellular energy status.  相似文献

Abstract. Transport of gases between the intercellular spaces of plant leaves and the surrounding air is analysed in terms of multicomponent collision processes through an isothermal, porous septum. Interaction of diffusing species with each other and with the pore walls is described using a modified Stefan–Maxwell equation and an equation relating the pressure gradient to the sum of the diffusive fluxes, weighted by their appropriate Knudsen diffusivities. Viscous How arising from an excess pressure within the leaf is also considered.
Equations are derived which describe the flux densities of water vapour and CO2 through the stomata. The analysis is general and is applicable to trace gases other than CO2. A simple conductance is defined for water vapour to relate the flux and mol fraction difference across the stomata, viz. Nw =− gw , δ xw / xa . A simple conductance cannot be defined for CO2 because the flux of water vapour has a significant influence on the CO2 gradient. The equation derived for the intercellular mol fraction of CO2 is in terms of the fluxes of CO2 and water vapour and represents a 'large-pore' ( d > μm) approximation which requires no information about stomalal geometry. Analogous equations are developed for transfer of gases through the leaf boundary layer. Sample calculations are presented to illustrate the effect of neglecting the interaction of water vapour and CO2 on the calculated intercellular and surface concentrations of CO2. Equations for computing water vapour and CO2 flux densities from leaf chamber measurements are also presented.  相似文献

Reprogramming involves multiple layers of molecular regulation, yet it remains relatively unknown how the cell's metabolism is changing and/or contributing to this process. In this issue of Cell Metabolism, Folmes et?al. It is not surprising that past predictions of the extent of adoption of new technology by the food and agricultural industries have only been partly realized. This is a result of the difficulty of forecasting the take-up of technology that is capable of being transferred from other industries, for example, process control methods and the advent of new packaging materials. Most technology that is adopted is normally incremental over the existing technology and forecasting of this type is best done by specific experts within the industry. Factors which influence uptake of technology in the food industry include not only the available technology, but also the supply of raw materials, economics and disposable income, food habits, health and nutrition and market requirements. In addition to these there is legislation, which imposes compositional, labelling and trade requirements on grower, processor and retailer. New products and processes are determined by all these factors, the overriding influence being the consumer's requirement for palatable and nutritious foods that provide value for money. In the cereals-processing industry significant developments have taken place in the provision of U.K. wheat varieties for breadmaking. The U.K. is now in the position where the bulk of its breadmaking wheats are homegrown. Further advances can be make by investing in applied research into those characteristics of bread wheats which are determinants of good bread flours, that in turn will help in the provision of suitable bread wheat varieties.
