Lipids from cultured cells, leaves and seeds of two varieties each of soybean (Glycine max) and oil seed rape (Brassica napus) were separated into neutral lipids, glycolipids and phospholipids and their fatty acids were analysed. Usually, the fatty acid composition differed between corresponding fractions from cultured cells, leaves and seeds. Differences were least marked in (i) the phospholipids from cultured cells and leaves of soybean and (ii) the neutral lipids from cultured cells and seeds of rape. In the cultured cells, the fatty acid composition of the phospholipids differed from that of the glycolipids and neutral lipids, and fatty acids of chain length greater than C18 comprised a large proportion of the fatty acids of the glycolipids.  相似文献

To compare the effects of ozone and peroxyacetyl nitrate (PAN) on leaf lipids, fatty acids and malondialdehyde (MDA), morning glory (Pharbitis nil Choisy cv Scarlet O'Hara) and kidney bean (Phaseolus vulgaris L. cv Gintebo) plants were exposed to either ozone (0.15 microliter per liter for 8 hours) or PAN (0.10 microliter per liter for up to 8 hours). Ozone increased phospholipids in morning glory and decreased in kidney bean at the initial stage (2-4 hours) of exposure, while it scarcely changed glycolipids, the unsaturated fatty acids, and MDA in both plants. A large reduction of glycolipids occurred 1 day after ozone exposure in both plants. PAN caused marked drops in phospholipids and glycolipids in kidney bean at relatively late stage (6-8 hours) of exposure, while it increased phosphatidic acid and decreased the unsaturated fatty acids, an increase which was accompanied by a large increase in MDA. These results suggest that ozone may not directly oxidize unsaturated fatty acids at the initial stage of exposure, but may alter polar lipid metabolism, particularly phospholipids. On the other hand, PAN may abruptly and considerably degrade phospholipids and glycolipids by peroxidation or hydrolysis at the late stage of exposure. The present study shows that ozone and PAN affect polar lipids in different manners.  相似文献

Tall fescue (Festuca arundinacea Schreb. cv S.170) plants were grown in environments differing only in temperature: 6/4, 16/14 or 21/19°. The content of total and individual sugars and amino acids in leaf laminae and roots did not relate closely to the hardiness of the organ. The unsaturation of lipid fatty acids alone was clearly unrelated to hardiness because the difference in unsaturation was greater in the neutral lipids, glycolipids and phospholipids from roots than from leaves but only the latter differed in hardiness. Total amounts of lipids could have been related to hardiness but phospholipids and glycolipids in the roots were not. At least some of these changes may be related to adaptation of growth to temperature.  相似文献

Most of the nonstarch lipids in brown rice (Oryza sativa) of three rices differing in amylose content were contributed by bran, germ, polish and subaleurone layer. Nonstarch lipids consisted of 82–91% neutral lipids (of which 73–82% were triglycerides), 7–10% phospholipids and 2–8% glycolipids. Linoleic, oleic and palmitic acids were the major fatty acids. Nonwaxy (24 and 29% amylose) milled rice had proportionally more starch lipids and less nonstarch lipids than waxy (2% amylose) milled rice. Starch lipids were mainly lysophosphatidyl choline, lysophosphatidyl ethanolamine and free fatty acids. The major fatty acids were palmitic and linoleic acids, followed by oleic acid.  相似文献

Phospholipid fatty acids (PLFAs) are key components of microbial cell membranes. The analysis of PLFAs extracted from soils can provide information about the overall structure of terrestrial microbial communities. PLFA profiling has been extensively used in a range of ecosystems as a biological index of overall soil quality, and as a quantitative indicator of soil response to land management and other environmental stressors.The standard method presented here outlines four key steps: 1. lipid extraction from soil samples with a single-phase chloroform mixture, 2. fractionation using solid phase extraction columns to isolate phospholipids from other extracted lipids, 3. methanolysis of phospholipids to produce fatty acid methyl esters (FAMEs), and 4. FAME analysis by capillary gas chromatography using a flame ionization detector (GC-FID). Two standards are used, including 1,2-dinonadecanoyl-sn-glycero-3-phosphocholine (PC(19:0/19:0)) to assess the overall recovery of the extraction method, and methyl decanoate (MeC10:0) as an internal standard (ISTD) for the GC analysis.  相似文献

The presence of noncovalently associated lipids and covalently bound fatty acids was investigated in preparations of mucus glycoproteins obtained by using density-gradient centrifugation in CsCl/guanidinium chloride. No phospholipids, glycolipids, cholesterol, or triglycerides could be detected. However, small amounts of extractable fatty acids were consistently found, the sum of which ranged from 0.3 to 0.9 micrograms/mg of glycoprotein. The amount of fatty acid released after subsequent treatment with KOH ranged from 0 to 27 ng/mg of glycoprotein. We conclude that density-gradient centrifugation in CsCl/guanidinium chloride is very efficient in removing noncovalently associated lipids from mucus glycoproteins and that covalently bound fatty acids are probably not present in the macromolecules.  相似文献

The inability of silicic acid to completely separate the neutral lipids from phospholipids has been reported by several investigators (1,2). Hornstein et al. (3) increased the polarity of the solvent system and reported a clean separation of the phospholipid fraction by adsorption on activated silicic acid. Studies on bovine lipids by Hood and Allen (2) utilized acid-washed Florisil to separate the lipid fractions claiming that silicic acid incompletely separates the free fatty acids from the phospholipids. Work performed in this laboratory (4) on bovine lipids confirmed that phospholipids could be effectively separated from free fatty acids by adsorption on silicic acid by incorporating the solvent system described by Hornstein et al. (3). The liquid-liquid partition procedure of Hamilton and McDonald (5) was also found to be sensitive enough to partition the extremely small amount of free fatty acids from the esterified fatty acids. This paper provides evidence for the effectiveness of these methods in separating the frec fatty acids by incorporating an internal standard [1-14C]palmitic acid.  相似文献

When heterotrophic cell cultures of red goosefoot (Chenopodium rubrum) turned photoautotrophic, their contents of various glycolipids and phospholipids increased. The total lipids and the individual lipid classes, especially monogalactosyldiacylglycerols, became richer in linolenic and poorer in linoleic acids. When photoautotrophic cell cultures were rendered heterotrophic again a reversal of changes occurred; both the composition of lipids and the patterns of their constituent fatty acids became similar to those of the starting heterotrophic cultures.The results indicate that the biosynthesis of linolenic acid in photoautotrophic cell cultures involves mainly desaturation of linoleic acid and that chain extension of hexadecatrienoic acid is possibly another, though minor pathway. Monogalactosyldiacylglycerols are apparently the substrates preferred for linolenic acid biosynthesis, whereas various phospholipids are the substrates preferred for linoleic acid biosynthesis.During a growth period of 6 weeks, the levels of polyunsaturated fatty acids in the lipids from both heterotrophic and photoautotrophic cell cultures decrease with time, whereas the proportions of palmitic acid increase.  相似文献

The effect of neutral lipids, glycolipids and phospholipids associated with dog gastric mucus glycoprotein, and that of covalently bound fatty acids on the ability of glycoprotein to retard the diffusion of hydrogen ion was investigated. Purified mucus glycoprotein in its native form, placed between equimolar (0.155M) solutions of HCl and NaCl in a specially designed two-compartment chamber, caused a 90% reduction in permeability to hydrogen ion when compared with a layer of NaCl. Extraction of associated lipids lead to a 68% increase in permeability of the glycoprotein to hydrogen ion, while removal of the covalently bound fatty acids increased further the diffusion rate by 6%. Reassociation of the delipidated glycoprotein with its neutral lipids reduced the permeability to hydrogen ion by 34%, an 11% reduction was obtained with glycolipids, and 23% with phospholipids. Since neutral lipids account for 47% of the glycoprotein lipids, glycolipids 41.1% and phospholipids 11.9%, the quantitative decrease in permeability of the delipidated glycoprotein following its reassociation with phospholipids is 2.7 times greater than that of neutral lipids and 7.3 times greater than that of glycolipids.  相似文献

Intact magnetosomes of Aquaspirillum magnetotacticum were purified from broken cells by a magnetic separation technique. Electron microscopic and chemical analyses revealed the magnetite to be enclosed by a lipid bilayer admixed with proteins. Lipids were recovered in fractions expected to contain (i) neutral lipids and free fatty acids, (ii) glycolipids and sulfolipids, and (iii) phospholipids (in a weight ratio of 1:4:6). Phospholipids included phosphatidylserine and phosphatidylethanolamine. Two of the numerous proteins detected in the magnetosome membrane were not found in other cell membranes or soluble fractions.  相似文献

Normal phase, isocratic high-performance liquid chromatography methods are described for the separation of neutral lipid and fatty acid classes using low wavelength detection. Prior to high-performance liquid chromatography, methods were developed and are described for the separation of phospholipids from neutral lipids and fatty acids using small (600 mg) silica Sep-PaksTM. Recoveries of cholesteryl esters, triglycerides, fatty acids, and phospholipids from the silica columns were greater than 95%. Two mobile phases are described for lipid class separation by high-performance liquid chromatography. The first mobile phase, hexane-2-propanol-acetic acid 100:0.5:01, resulted in incomplete separation of cholesteryl ester and triglyceride but excellent separations of fatty acids and cholesterol. The second mobile phase, hexane-n-butyl chloride-acetonitrile-acetic acid 90:10:1.5:0.01, resulted in complete separation of the four lipid classes. This mobile phase also separated individual triglycerides and fatty acids based on the number of double bonds. Recoveries of radiolabeled lipids for the four lipid classes from high-performance liquid chromatography was greater than 95% with both mobile phases.  相似文献

The effect of the synthetic analogue of natural cytokinin 6-benzylaminopurine (BAP) on the fatty-acid composition of membrane lipids in shoots of Karelian (Curly) birch Betula pendula Roth var. carelica (Mercklin) Hämet-Ahti under in vitro conditions was studied. It was found that the addition of BAP (in a concentration of 0.25, 0.5, 1.0, or 2 mg/L) to the nutrient medium leads to an increase in the proportion of saturated fatty acids in phospholipids, whereas in the control variant unsaturated fatty acids predominated in phospholipids. In glycolipids, on the contrary, BAP caused an increase in the synthesis of unsaturated fatty acids (predominantly, linolenic), but the degree of its effect depended on the concentration of the hormone. In particular, when the maximum amount of the studied concentration (2.0 mg/L) was used, the use of BAP was shown to decrease the proportion of linolenic acid. It is suggested that the revealed changes in the composition of glycolipids are indicators of the stimulating effect of cytokinin (up to a certain concentration) on the structure and function of chloroplasts. Due to the photomixotrophy of shoot cultures, chloroplasts are an important factor determining not only their photosynthetic activity, but also cell proliferation and the direction of in vitro morphogenesis.  相似文献

The objective of this study was to document and compare the lipid class and fatty acid composition of the green microalga Chlorella zofingiensis cultivated under photoautotrophic and heterotrophic conditions. Compared with photoautotrophic cells, a 900% increase in lipid yield was achieved in heterotrophic cells fed with 30 g L−1 of glucose. Furthermore heterotrophic cells accumulated predominantly neutral lipids (NL) that accounted for 79.5% of total lipids with 88.7% being triacylglycerol (TAG); whereas photoautotrophic cells contained mainly the membrane lipids glycolipids (GL) and phospholipids (PL). Together with the much higher content of oleic acid (C18:1) (35.2% of total fatty acids), oils from heterotrophic C. zofingiensis appear to be more feasible for biodiesel production. Our study highlights the possibility of using heterotrophic algae for producing high quality biodiesel.  相似文献

Live bacteria in modifiedDiamond’s axenic medium did not support growth ofEntamoeba histolytica. Cysteine hydrochloride, required for the multiplication of amoeba, was broken down by live bacteria and toxic substances were produced which were lethal for amoebae. Monoxenic and xenic cultures ofaxenically grownE. histolytica could be established in Boeck and Drbohlav medium with bacteria and rice starch. Bacterial lipids prepared from 15 human intestinal bacteria supported growth and multiplication ofE. histolytica in axenic medium. In a pilot experiment using lipids ofStreptococcus faecalis, free fatty acids did stimulate the multiplication of amoebae. When total lipids of this bacteria were fractionated into neutral lipids and phospholipids by chromatography and used, neither fraction was found to stimulate growth. Free fatty acids prepared by chemical hydrolysis of the total lipids, neutral lipids and phospholipids stimulated growth ofE. histolytica, The sterols present in the bacterial lipids (neutral lipids or non-saponifiable fractions) stimulated growth of amoebae. It was found thatE. histolytica is incapable of liberating fatty acids from di- or triglyceridesof phospholipids and the multiplication of the organism is stimulated by the presence of free fatty acids and sterols (cholesterol).  相似文献

The neutral fraction of nonstarch lipids in developing brown rice (Oryza sativa L., cv IR42) was accumulated up to 16 days after flowering (DAF), but phospholipids and glycolipids increased only up to 8 DAF. Fatty acids accumulated in nonstarch lipids until 12 DAF. However, the proportion of linolenic acid in the lipid fraction decreased and that of oleic acid increased during this period. Accumulation of fat-by-hydrolysis in the brown rice occurred until 20 DAF and followed closely that of starch. The proportion of linolenic acid decreased and that of linoleic acid increased until 16 DAF. The fatty acid composition of fat-by-hydrolysis and starch lipids were identical and fat-by-hydrolysis accounted for 48% by weight of starch lipids. Nonstarch lipids were mainly composed of triglycerides and were located in the bran and embryo of mature brown rice. Starch lipids were mainly composed of lysophosphatidyl choline, free fatty acids and lysophosphatidyl ethanolamine, and were located in the endosperm.  相似文献

The principal lipids associated with the electron transport membrane of Haemophilus parainfluenzae are phosphatidylethanolamine (78%), phosphatidylmonomethylethanolamine (0.4%), phosphatidylglycerol (18%), phosphatidylcholine (0.4%), phosphatidylserine (0.4%), phosphatidic acid (0.2%), and cardiolipin (3.0%). Phospholipids account for 98.4% of the extractible fatty acids. There are no glycolipids, plasmalogens, alkyl ethers, or lipo amino acid esters in the membrane lipids. Glycerol phosphate esters derived from the phospholipids by mild alkaline methanolysis were identified by their staining reactions, mobility on paper and ion-exchange column chromatography, and by the molar glycerol to phosphate ratios. Eleven diacyl phospholipids can be separated by two-dimensional thin-layer chromatography. Each lipid served as a substrate for phospholipase D, and had a fatty acid to phosphate ratio of 2:1. Each separated diacyl phospholipid was deacylated and the glycerol phosphate ester was identified by paper chromatography in four solvent systems. Of the 11 separated phospholipids, 3 were phosphatidylethanolamines, 2 were phosphatidylserines, and 2 were phosphatidylglycerols. Phosphatidylcholine, cardiolipin, and phosphatidic acid were found at a single location. Phosphatidylmonomethylethanolamine was found with the major phosphatidylethanolamine. Three distinct classes of phospholipids are separable according to their relative fatty acid compositions. (i) The trace lipids consist of two phosphatidylethanolamines, two phosphatidylserines, phosphatidylcholine, phosphatidic acid, and a phosphatidylglycerol. Each lipid represents less than 0.3% of the total lipid phosphate. These lipids are characterized by high proportions of the short (C(10) to C(14)) and long (C(19) to C(22)) fatty acids with practically no palmitoleic acid. (ii) The major phospholipids (93% of the lipid phosphate) are phosphatidylethanolamine, phosphatidylmonomethylethanolamine, and phosphatidylglycerol. These lipids contain a low proportion of the short (C(19)) fatty acids. Palmitic and palmitoleic acids represent over 80% of the total fatty acids. (iii) The fatty acid composition of the cardiolipin is intermediate between the other two classes. Both palmitoleic and the longer fatty acids represent a significant proportion of the total fatty acid.  相似文献

Fractions of polar lipids have been isolated from bifidobacteria, and the immunoreactivity and serological specificity of glycolipids and phospholipids have been studied. Enzyme immunoassay (dot-EIA) of polar lipids demonstrates that the fractions of glycolipids and phospholipids of bifidobacteria are highly immunoreactive. Pronounced reactions of major glycolipids and phospholipids with a homologous polyvalent antiserum against B. adolescentis 94-BIM have been observed at antigen concentrations of approximately 100 ng. The antiserum contained high titers of specific antibodies against glycolipids and phospholipids of bifidobacteria, as demonstrated by heterogeneous solid-phase enzyme immunoassay (ELISA). Experimental data confirming the presence of subpopulations of specific antibodies (antiglycolipid and antiphospholipid) in the blood sera of immunized animals lead to the conclusion that the major glycolipids and phospholipids of bifidobacteria are specific markers appropriate for serological diagnostics.  相似文献

A comparative study of the mycelial lipid composition of a wild strain (V35) and one unsaturated fatty acid auxotroph (UFA2) of Aspergillus niger has been performed. The lipid composition of both strains are qualitatively the same but quantitatively different. All the strains contain the following phospholipids: cardiolipin, phosphatidylethanolamine, phosphatidylcholine, lysophosphatidylethanolamine, lysophosphatidylcholine, and phosphatidylserine; and triglycerides, diglycerides, monoglycerides, ergosterol, and sterol esters as the neutral lipids; mono- and di-galactosyl diglyceride as the major glycolipids along with small amounts of the corresponding mannose analogs. Phosphatidylethanolamine and phosphatidylcholine constitute the bulk of the phospholipids. The mutant (UFA2) contains a higher level of glycerides and lower levels of sterol (both free and esterified form), phospholipids, and glycolipids than the wild type. Aspergillus niger contains C16 to C18 saturated and unsaturated fatty acids. Small amounts of long-chain (C20 to C24) and short-chain (C10 to C14) saturated and unsaturated acids are also present. Linoleic, oleic, and palmitic are the major acids, stearic and linolenic acids being minor ones. UFA2 grows only in the presence of unsaturated fatty acid (C16 or C18) and accumulates a higher concentration of supplemented acid which influences its fatty acid profile.  相似文献

All major classes of lipids were found in the young adults in brain (22 days post-infection) and gravid Angiostrongylus cantonensis in lung of rats (34 days post-infection) comprising approximately 60% of phospholipids, 30% of neutral lipids and the rest, glycolipids. 2. The relative composition of phospholipids were quite similar between worms from the two different habitats, with phosphatidylcholine predominating. The glycolipid profiles were also similar. 3. More neutral lipids in the worms from brain existed as cholesterol and cholesterol esters than those from the lung. More than 20% of the fatty acids in these lipids of the brain were found as C10-C14 acids while oleic acid was the main component in the lung worm.  相似文献

Triglycerides, free fatty acids, free and esterified ergosterol, Q9, phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, lysophosphatidylethanolamine, and three different acylglycoses were identified in the soluble lipids of Sporendonema epizoum mycelium. The same compounds as well as a sterol glycoside were also found in conidia. The mycelium is richer than the conidia in phospholipids, Q9 and free and esterified ergosterol but contains less glycolipids. The most abundant fatty acid in all non-polar fractions is C18:2. The prevalent fatty acid of the phospholipids is C18:1, except for conidial phosphatidylethanolamine and mycelial lysophosphatidylethanolamine.  相似文献
