本研究通过形态解剖和原位末端标记法(TUNEL),对中华蜜蜂Apis cerana cerana视叶胚后发育过程中的细胞凋亡进行了研究,结果表明:视叶内的细胞程序性死亡开始出现在1龄幼虫末期,随后凋亡细胞数量逐渐增加;在视叶的胚后发育过程中,细胞凋亡经历了3个高峰期,即2龄幼虫、5龄幼虫和蛹发育的第2天;在视叶3个部分的发育中,视髓层中细胞凋亡的数量远远多于视小叶和视神经节层,而视神经节层最少,说明了细胞凋亡的数量和位置与各部分结构发育的时间以及神经投射有关。广泛的细胞凋亡是蜜蜂视叶发育过程中的一个显著特征。  相似文献

【目的】通过研究苯醚威对印鼠客蚤 Xenopsylla cheopis (Rothschild,1903)的早 3 龄幼虫和未吸血新羽化成虫的组织学变化,探讨其灭蚤机理,为鼠疫媒介蚤种的防治提供基础资料。【方法】以微量点滴法将苯醚威施药于印鼠客蚤早3龄幼虫和未吸血新羽化成虫,采用组织学、显微摄影及统计学方法观察组织变化。【结果】经苯醚威作用后,印鼠客蚤的早 3 龄幼虫的表皮增厚、卵巢芽生殖细胞萎缩、睾丸芽精原细胞间质减少;未吸血新羽化成虫的睾丸塞消失快、唾液腺细胞破坏严重、中肠上皮细胞萎缩。【结论】(1)苯醚威通过干扰印鼠客蚤幼虫的变态,引起幼虫表皮、生殖芽异常改变,不能发育为成虫而死亡;(2)苯醚威可加速印鼠客蚤新羽化雄性成虫的睾丸塞吸收;(3)苯醚威可破坏印鼠客蚤新羽化成虫的唾液腺细胞,并引起中肠上皮细胞萎缩。  相似文献

【目的】上颚腺(mandibular gland)是昆虫重要的外分泌腺,它产生的化学物质在昆虫的种内信息交流中起重要的作用。本研究目的在于了解中华蜜蜂Apis cerana cerana上颚腺的组织结构以及胚后发育特点。【方法】本研究通过组织形态学、BrdU免疫组织化学等技术,对中华蜜蜂上颚腺的结构和发育过程进行了比较研究。【结果】中华蜜蜂的上颚腺在不同级型间差异显著,蜂王的面积最大,工蜂较小,而雄蜂退化。上颚腺出现在末龄幼虫到预蛹阶段,细胞分裂活动的高峰期发生在蛹发育的第1天,随后分裂细胞数减少,并一直持续到蛹发育的第6天结束。在上颚腺发育早期,由分泌细胞分化的内膜就已经出现,并一直保持到成虫。【结论】本研究为昆虫上颚腺的发育和功能研究提供了理论基础。  相似文献

The p38 mitogen-activated protein kinase (MAPK) is involved in various processes, including stress responses, development, and differentiation. However, little information on p38 MAPK in insects is available. In this study, a p38 MAPK gene, Accp38b, was isolated from Apis cerana cerana and characterized. The quantitative real-time PCR (Q-PCR) analysis revealed that Accp38b was induced by multiple stressors. Notably, the expression of Accp38b was relatively higher in the pupae phase than in other developmental phases. During the pupae phase, Accp38b expression was higher in the thorax than in the head and abdomen and higher in the fat body than in the muscle and midgut. Immunohistochemisty showed significant positive staining of Accp38b in sections from the brain, eyes, fat body, and midgut of A. cerana cerana. These results suggest that Accp38b may play a crucial role in stress responses and have multiple aspects function during development.  相似文献

In Drosophila gibberosa, differences between midgut and salivary gland chromosomes fall into two categories: tissue-specific band modulations which persist throughout the 90 h developmental period that we studied and tissue-specific puffs. Puffs that are common to both tissues tend to appear earlier in the midgut. Some major early ecdysteroid-induced puffs appear simultaneously in both tissues at the end of the third larval instar; however, the many late puffs that follow in the salivary glands are absent from the midgut. Intense puff activity in the early third larval instar midgut declines at the time of the hormonal pulse that initiates intense gene and secretory activity in salivary glands; the sloughing of midgut cells ensues.  相似文献

【目的】囊状幼虫病病毒(sacbrood virus, SBV)是严重危害中华蜜蜂Apis cerana cerana蜂群健康和种群数量的病原微生物,但其对蜜蜂的致死机制不明。本研究旨在探究SBV对不同发育阶段中华蜜蜂营养代谢和免疫的影响。【方法】分别给中华蜜蜂2日龄幼虫和新羽化成虫饲喂SBV,逐日统计死亡蜜蜂数量,检测病毒对蜜蜂存活的影响;利用qPCR检测中华蜜蜂4日龄幼虫、预蛹以及10和20日龄成虫体内SBV RNA、营养代谢基因(ilp1, ilp2, hex110, hex70b, hex70c和vg)、先天性病毒免疫基因(rel, toll, apidaecin, abaecin, defensin, hymenoptaecin, jra, key和state92e)、细胞凋亡基因(atg7和LOC100577876)和抗RNA病毒基因(dis3和dicer)的表达水平。【结果】 SBV感染显著降低了中华蜜蜂幼虫的存活率,但对成虫的生存影响不明显。SBV RNA在中华蜜蜂4日龄幼虫和预蛹体内的表达量显著高于其在10和20日龄成虫体内的表达量。SBV显著降低了中华蜜蜂幼虫营养代谢基因ilp1, ilp2, hex110, hex70b和hex70c以及成虫营养代谢基因vg和hex110的表达量,但显著提高了4日龄幼虫rel, toll, apidaecin, abaecin, defensin, hymenoptaecin和jra以及成虫key和 state92e等先天性病毒免疫基因的表达量,还引起预蛹体内的细胞凋亡基因atg7和LOC100577876的表达量显著增加。【结论】SBV在中华蜜蜂幼虫和预蛹体内的感染水平远高于在成虫体内的,其对幼虫的危害也大于对成虫。SBV显著影响了中华蜜蜂正常的营养代谢,染毒中华蜜蜂能够提高自身的免疫水平来应对;预蛹期中华蜜蜂幼虫细胞凋亡水平的显著增加可能与化蛹异常及死亡有关。  相似文献

中华蜜蜂(Apis cerana cerana)的脑由前脑、中脑和后脑三部分构成,蕈形体位于前脑的背侧,是其重要的学习及其他复杂行为的整合中心。通过对中华蜜蜂工蜂的幼虫、蛹及成虫的蕈形体形态发育的观察研究,发现中华蜜蜂的蕈形体包含约1000个成神经细胞,它们最终形成了蕈形体的所有Kenyon细胞。这些成神经细胞来自于在新孵化的幼虫脑中已存在的四丛成神经细胞,每一丛细胞的数量不多于45个。蕈形体柄区的出现约在3龄幼虫,而α叶和β叶在5龄幼虫已可明显辨认。冠区出现较晚,大约在蛹期的第二天以后。由于社会性昆虫复杂的学习、记忆和认知需求,其蕈形体的体积和复杂程度都优于其他昆虫。  相似文献

【目的】本研究旨在利用位点特异性重组技术(FullCoV)将中华蜜蜂 Apis cerana cerana 幼虫膜蛋白cDNA连接到pPR3-N载体上,构建中华蜜蜂幼虫膜蛋白酵母双杂交cDNA文库。【方法】提取2-3日龄中华蜜蜂工蜂幼虫总RNA;分离mRNA后,在反转录酶的作用下合成幼虫膜蛋白cDNA第1链,并合成双链cDNA。在双链cDNA的5′端加上带有重组序列的接头后,通过FullCoV技术与载体pPR3-N进行连接,然后将连接产物电转化到DH10B感受态细胞,构建中华蜜蜂幼虫膜蛋白酵母双杂交cDNA文库,并对该文库插入片段大小和文库滴度进行检测。【结果】通过FullCoV技术成功构建了中华蜜蜂幼虫膜蛋白酵母双杂交cDNA文库,经检测,中华蜜蜂幼虫膜蛋白酵母cDNA文库的总库容量为1.5×10^7 cfu,文库滴度为3×10^6 cfu/mL,重组率达到100%。【结论】本研究利用FullCoV技术成功构建了中华蜜蜂幼虫膜蛋白酵母cDNA文库,为进一步探究感染中华蜜蜂的病原微生物与宿主蛋白互作研究奠定了基础。  相似文献

In the anterior part of the midgut and in the Malpighian tubules of the stick insect Bacillus rossius, about 10% of the epithelial cells develop endonuclear bodies which appear as DNA-RNA masses; in these cells the usual nucleoli are no longer evident. The DNA-RNA bodies are first formed in third instar larvae, become numerous in the fourth instar and persist in adults. In all larval instars and adults a different kind of DNA-body has been noticed in the epithelial cells of the posterior midgut. The DNA-RNA bodies of the anterior midgut and of the Malpighian tubules have been interpreted as the result of somatic gene amplification, whereas the DNA masses of the posterior midgut are likely due to a virus infection.  相似文献

咽下神经节是昆虫腹神经索的第一个复合神经节, 主要调节口器附肢的活动。本研究通过形态解剖、 BrdU免疫组织化学等技术, 对中华蜜蜂Apis cerana cerana咽下神经节的组织结构和胚后发育过程进行了比较研究。结果表明: 中华蜜蜂的咽下神经节由上颚、 下颚和下唇3个神经节组成。在胚后发育过程中, 细胞增殖的活跃期主要集中在预蛹和蛹发育的第1天, 增殖活动一直持续到蛹发育的第4天结束。根据神经胶质细胞的位置和形态, 咽下神经节中的神经胶质可分为3种类型--表面神经胶质、 皮层神经胶质和神经纤维网神经胶质。本研究为蜜蜂神经系统的发育和功能研究提供了理论基础。  相似文献

为研究蜜蜂幼虫信息素中3种酯类成分(甲基棕榈酸酯、乙基棕榈酸酯和乙基油酸酯)对中华蜜蜂Apis cerana cerana和意大利蜜蜂Apis mellifera ligustica工蜂哺育行为、封盖行为以及蜂王发育影响, 在人造蜂蜡王台中加入1%和0.1%(w/w)的3种酯类作为实验组, 以不添加酯类(0%)的为对照组, 移入1日龄工蜂幼虫, 测定王台接受率、单个王台中幼虫和王浆重量; 另将分别添加1%和0.1%(w/w)3种酯类的石蜡假幼虫放入工蜂巢房中, 同样设对照组, 测定假幼虫的封盖率; 在新鲜王浆中以1%和0.1%(w/w)分别加入3种酯类作为实验组, 以不添加酯类(0%)的作为对照组, 再分别在1日龄、2日龄和3日龄幼虫王台中加入0.01 mL含有酯类的蜂王浆, 并测定蜂王初生重和卵巢管数量。结果表明: 0.1%甲基棕榈酸酯可以显著提高中蜂和意蜂幼虫重量; 意蜂的甲基棕榈酸酯和乙基油酸酯两个实验组(1.0%, 0.1%)假幼虫封盖率都极显著高于对照组; 乙基油酸酯两个实验组(1.0%, 0.1%)都显著降低了中蜂和意蜂蜂王初生重和卵巢管数量。这说明不同蜜蜂幼虫信息素具有不同的生物学效应。  相似文献

为揭示褐飞虱Niloparvata lugens Stl若虫在发育过程中中肠的凋亡细胞,使用末端脱氧核苷酸转移酶介导的dUTP-生物素断端标记法(TUNEL)进行中肠组织切片检测,结果表明,1~5龄若虫中肠分别存在2%~5%的凋亡细胞。利用4′,6-二脒基-2-苯基吲哚二盐酸(DAPI)染色法检测表明,存在Ⅰ,Ⅱa和Ⅱb期凋亡的细胞核,其特征包括染色体浓缩、边缘化及细胞核碎裂。透射电子显微镜检测结果表明,早期凋亡的细胞呈现染色质浓缩、边缘化特征,晚期凋亡的细胞出现细胞核碎裂、形成凋亡小体及细胞质空泡化等。本研究揭示了在正常发育过程中褐飞虱若虫中肠有少量的细胞发生了凋亡。通过人工干预的方式调控中肠细胞的凋亡进程有可能使之成为防治该水稻害虫的新靶标。  相似文献

神经胶质作为视觉系统的重要成分之一, 对视觉系统的发育及功能起着重要的作用。本研究通过组织解剖观察、 免疫组织化学等技术, 对中华蜜蜂Apis cerana cerana幼虫和蛹的视觉系统中神经胶质的类型和发育过程进行了比较研究。研究表明: 在中华蜜蜂视觉系统中, 根据神经胶质的位置和形态主要分为表面神经胶质、 皮层神经胶质和神经纤维网神经胶质3种类型; 神经胶质主要来源于视柄和视叶中的神经胶质前体中心; 神经胶质细胞数量的增加一方面来自于细胞的迁移, 另一方面来自于神经胶质细胞自身的分裂增殖。本研究为昆虫神经胶质的发育以及功能研究提供理论基础。  相似文献

The midgut structure of 4-7 day old fourth instar Chironomus thummi larvae was investigated with the light arid electron microscopes. Four regions are present which may be identified by the following major features: (1) Anterior I: this is the region under the esophageal invagination. (2) Anterior II: short microviili characterize these cells. Long narrow, basal plasma membrane infoldings associated with mitochondria are conspicuous. This region is hypothesized to be important in ion and fluid transport. (3) Anterior III: numerous crystals are seen in these cells. Storage is proposed to be a major function of this region. (4) Posterior: these cells have long microviili, extensive RER, many Golgi, and short basal plasma membrane infoldings. Posterior cells probably function in secretion of digestive enzymes and absorption of nutrients. Cadmium is sequestered by the midgut, almost exclusively in the posterior cells. 中华蜜蜂Apis cerana cerana是一种真社会性昆虫,也是我国重要的经济昆虫。本实验目的是为了检测精子是否可以作为载体将外源egfp基因介导转入中华蜜蜂。首先将雄蜂精子与线性化的质粒DNA共浴,然后通过人工授精技术将精子导入处女王,再对实验蜂群后代进行分析。结果显示EGFP蛋白在一群实验组蜂的1~2日龄小幼虫中表达较强,能检测到0.01%~0.02%荧光阳性小幼虫个体;通过PCR和RT-PCR技术分析,证实转入的外源egfp基因获得表达。实验结果表明精子载体法能够用于中华蜜蜂外源基因的转移和表达。  相似文献

The degradation of haemoglobin (Hb) during the metamorphosis of Chironomus pallidivitatus was studied by means of spectrophotometry, disc electrophoresis, and isotopic tracer methods. Hb concentration was maximal during the late fourth instar larva and exhibited a continual decline throughout the pharate adult periods; in the mature adult Hb is almost entirely absent. Coupled with this reduction in Hb is an increase in the quantity of bile pigment (tentatively identified as bililatrenes) most probably representing products of the Hb degradation. These pigments are found in considerable quantity within the meconium at the time of adult emergence.Larval, 59Fe-labelled, Hb injected into late fourth instar larvae was found to be most concentrated in the midgut portion of the early pharate adult. The quantity of 59Fe-haemoglobin in the midgut of early pharate adult was at least five times the concentration found in late pharate pupae. Spectrophotometric and electrophoretic analysis of the midgut contents of untreated larvae showed that Hb was one of the major soluble proteins in the midgut at the time of pupation. This rapid intestinal uptake of Hb suggests that the intestine is involved in the mechanism for the degradation and removal of the Hb, no longer required in the adult stage, a seemingly important step in the metamorphosis of this insect.  相似文献

The mosquito Stegomyia aegypti (=Aedes aegypti) (Diptera: Culicidae) is a vector for the dengue and yellow fever viruses. As blood digestion occurs in the midgut, this organ constitutes the route of entry of many pathogens. The effects of the insecticide imidacloprid on the survival of St. aegypti were investigated and the sub‐lethal effects of the insecticide on midgut development were determined. Third instar larvae were exposed to different concentrations of imidacloprid (0.15, 1.5, 3.0, 6.0 and 15.0 p.p.m.) and survival was monitored every 24 h for 10 days. Midguts from imidacloprid‐treated insects at different stages of development were dissected and processed for analyses by transmission electron microscopy, immunofluorescence microscopy and terminal deoxynucleotidyl transferase dUTP nick‐end labelling (TUNEL) assays. Imidacloprid concentrations of 3.0 and 15.0 p.p.m. were found to affect midgut development similarly. Digestive cells of the fourth instar larvae (L4) midgut exposed to imidacloprid had more multilamellar bodies, abundantly found in the cell apex, and more electron‐lucent vacuoles in the basal region compared with those from untreated insects. Moreover, imidacloprid interfered with the differentiation of regenerative cells, dramatically reducing the number of digestive and endocrine cells and leading to malformation of the midgut epithelium in adults. The data demonstrate that imidacloprid can reduce the survival of mosquitoes and thus indicate its potentially high efficacy in the control of St. aegypti populations.  相似文献

Ribosomal protein L17 (RPL17) is a core protein of the large (60S) ribosomal subunit and plays an important role in protein synthesis. In this report, a RPL17 gene was isolated from Apis cerana cerana, designated as AccRPL17. Alignment analysis showed that AccRPL17 exhibits high homology to other known RPL17s. Moreover, genomic sequence analysis revealed that five exons are splitted by four introns, and the position of the first intron is comparatively conservative, being localized in the 5′ untranslated region. Partial putative cis‐acting elements related to development were also examined. Quantitative real‐time PCR showed that the highest mRNA level was detected in larvae on the fifth day. Simultaneously, immunohistochemical localization showed that AccRPL17 is primarily concentrated in muscular tissues, stigma, body wall, and the surrounding of the eye in the fifth‐instar larvae. Further studies suggested that AccRPL17 might be involved in responses to abiotic stresses. This is a report attempting to analyze the expression and distribution of RPL17 in A. cerana cerana. These results indicated that AccRPL17 might play an important role in insect development, and the importance of AccRPL17 in participating in abiotic stresses is discussed. © 2010 Wiley Periodicals, Inc.  相似文献
