This paper presents the evaluation of genetic diversity in 29populations of wild taxa of the Brassica oleraceaL. group (2 n=18)and two cultivars, using RAPDs as molecular markers. In a previouspaper (Lázaro and Aguinagalde, Annals of Botany82: 000000,1998), 11 isozymes were used for the same purpose. Results obtainedwith the two molecular markers (isozymes and RAPDs) are compared.DNA from ten individuals per population was analysed using sixdifferent primers; the 151 detected bands were polymorphic,11 were common to all species, six to all taxa, only one toevery population; and no bands were shared by every individual.The dendrogram obtained using genetic distances clusters B. oleraceapopulationswith B. bourgeaui, B. alboglabra, B. montanaand B. incana. Genomic in situ hybridization (GISH) has been used to study characteristics of the formation of alloplasmic lines detected among self-pollinated backcrossed progeny (BC1F5-BC1F8) of barley--wheat amphiploids [Hordeum geniculatum All. (2n = 28) x Triticum aestivum L. (2n = 42)] (2n = 70). The chromosome material of the wild barley H. geniculatum has been shown to contribute to these lines. For example, fifth-generation plants (BC1F5) had genotypes (2n = 42w + 2g), (2n = 42w + 1g + 1tg), and (2n = 41w + 1g), where w is common wheat chromosomes, g is barley (H. geniculatum) chromosomes, and tg is the telocentric chromosome of wild barley. Beginning from the BC1F6 generation, alloplasmic telocentric addition lines (2n = 42 + 2tg) and (2n = 42 + 1tg) appear. This lines has been found cytogenetically unstable. The progeny of each of these cytological types include not only the (2n = 42 + 2tg) and (2n = 42 + 2tg) addition plants, but also plants with the monosomic (2n = 41 + 1tg) and the disomic (2n = 40 + 2tg) substitutions, as well as the (2n = 41 + 2tg) plants, which lack one wheat chromosome and have two telocentric barley chromosomes. It has been demonstrated that the selection for well-filled grains favors the segregation of telocentric addition lines (2n = 42 = 2tg) and (2n = = 42 + 1tg). 相似文献

Of the 15 perennial species of the subgenus Glycine Willd., G. tomentella Hayata is unique in that it has four cytotypes (2n = 38, 40, 78, and 80) and a wide range of geographical distributions. The objective of this study was to uncover the genomic diversity among accessions of aneudiploid (2n = 38) and diploid (2n = 40) G. tomentella based on crossability rate, hybrid seed and seedling viability, meiotic chromosome pairing of F1 hybrids, and seed protein and protease inhibitor profiles. Aneudiploid and diploid G. tomentella accessions were divided into two (D1 and D2) and three [D3(A,B,C), D4, and D5] groups, respectively, based on previous isozyme studies. Crossability rate, intergenomic hybrid viability, degree of chromosome pairing, total seed protein profiles, and trypsin and chymotrypsin inhibitor banding patterns confirmed the isozyme grouping with minor disagreements. A consistent variation was not observed among the aneudiploid accessions in any method of analysis used in this study. Similarly, cytogenetic analysis and the total seed protein profiles did not show dissimilarity among the accessions from Papua New Guinea (PNG; the D3 group) and north of Mitchell River in Northern Queensland [N.Qld(n)]. However, trypsin and chymotrypsin inhibitor analysis revealed that the PNG accessions were distinctly different from N.Qld(n) accessions. The D4 and D5 group accessions were clearly distinguishable by both cytogenetic and biochemical methods. Thus, this study indicates the presence of four genomic groups among G. tomentella (2n = 38, 40) accessions, including the aneudiploids D1 and D2 in one group and diploids in three groups (D3, D4, and D5). These findings will be useful in further genome analysis and add to our present understanding of the biosystematics of the genus Glycine. 相似文献

Summary Crosses between male sterile L. corniculatus (2n=4x=24) and L. tenuis (2n=2x=12) plants were performed in order to verify the presence of 2n gametes in L. tenuis. All but one of the plants from these crosses had 2n=4x=24 and the L. corniculatus phenotype; this plant had 2n=2x=12 and the L. tenuis phenotype. The plants also showed good quantity of pollen at tripping, good pollen fertility and good percentage of seed setting in the backcross to L. corniculatus. On the whole, both cytological and morphological observations, showing that all but one of the plants from L. corniculatus x L. tenuis were normal tetraploids, suggest the existence of diploandrous gametes in L. tenuis. On the other hand, haploid parthenogenesis probably gave origin to the dihaploid plant 2n=2x=12. 相似文献

Cytogenetical studies were carried out on the successive generations of offsprings from the induced tetraploid hybrid (2n = 32) between Ranunculus silerifolius (2n = 16) and R. chinensis (2n = 16). Aneuploids, 2n = 30 to 35, frequently occurred. In latter subsequent generations the deviation of aneuploids increased, but the proportion of euploids decreased, accompanied by the reduction of fertility of pollen grains and seed sets. F 2 and F 4 PMCs constantly exhibited meiotic abnormality, i.e. formation of quadrivalents and univalents. The speciation process of R. cantoniensis (2n = 32), which was presumed to arise from tetraploid hybrids between the above two species, is discussed on the basis of the above evidences.Former contributions of this series are Okada & Tamura (1977) and Okada (1984). 相似文献

采用SRAP(Sequence-related amplified polymorphism,序列相关扩增多态性)标记对中国13个省24份野生石蒜(Lycoris radiata)资源94个样品进行了检测。10个引物组合共扩增出218条带,其中173条为多态性条带,多态性百分比达79.36%。石蒜的观测等位基因数(na)、有效等位基因数(ne)、基因多样性指数(h)、Shannon信息指数(I)分别为1.7936、1.4131、0.2415和0.3664。石蒜不同种源间的遗传分化系数(Gst)达0.9547、基因流(Nm)仅0.0136,表明种源间遗传分化显著,遗传变异主要存在于种源间。根据Nei′s遗传距离对24份种源进行UPGMA聚类,所有石蒜种源聚成两大类,第I大类由7份种源组成,除江苏连云港的石蒜(JS3)外,均来自我国西南或西北地区;其余的17份种源构成第II大类,它们遍及华南、华中和华东地区;各大类中的分支结果与野生石蒜外部形态及生长发育习性有一定联系。将石蒜种源的遗传多样性与其所处的经度、纬度、海拔、年均降雨量、年均温等进行相关性分析,结果显示它们之间的相关性均不显著,即石蒜对环境依赖性小,能分布在各种生境中。根据以上结果,我们认为野生石蒜具有较丰富的遗传多样性,而种源间遗传分化显著的原因主要是基因流的隔离。研究结果对我国的野生石蒜资源的开发利用与保护有重要意义。 相似文献

To widen the germplasm base for the introgression of economically important traits such as resistance to biotic and abiotic stresses from related species, crosses of cultivated pearl millet were made with pollen from four related species differing in the basic chromosome number (x=5,7,8 and 9). Embryo rescue technique was used to obtain viable progeny. Pollinations of pearl millet with Pennisetum ramosum (2n=2x=10) did not give any viable progeny. Pearl millet interspecific hybrids with P. schwelnfurthii (2n-2x-14), P. mezianum (2n=4x-32) and P. orientale (2n=2x=18) were obtained. The hybrid between P. glaucum and P. mezianum (2n=23) is the first successful report. Interspecific hybrid plants resembled their corresponding pollen parents. Southern blots of Psfl digested DNAs from interspecific hybrids and the parental species were hybridized to a full length rDNA to further confirm their hybridity. This further revealed differential amplification of two rDNA repeats among the F 1 hybrids from the same cross ( P. glaucum X P. orientale). 相似文献

Cowpea (Vigna unguiculata (L.)) is an important crop for food security in Senegal; therefore, understanding the genetic diversity of local germplasm is relevant for crop improvement and genetic maintenance in the era of climate change. For this purpose, 15 microsatellite markers were used to estimate the genetic diversity of Senegalese cowpea germplasm, including 671 accessions grown in eight regions and 66 wild relatives and intermediate forms (weedy). For the cultivated, the main expected heterozygosity (mHe) ranged between 0.317 (Fatick) and 0.439 (South). A narrow genetic variation between accessions from the different regions was observed with genetic similarity ranging from 0.861 to 0.965 and genetic differentiation indices (Fst) between 0.018 and 0.100. The accessions from southern Senegal (Kédougou, Sédhiou, and Kolda regions) are more diverse than the others. However, the accessions from the North (Saint-Louis) are genetically different from other regions. The diversity analysis in wild relatives from Senegal, which had never been performed before, revealed that the wild/weedy forms remain more diverse than the cultivated with genetic diversity values (He) of 0.389 and 0.480, respectively. STRUCTURE software divided the Senegalese germplasm into five subpopulations. Three of them (i, ii, and iii) included only cultivated accessions from several regions, one (v) mainly from Saint-Louis, and one (iv) the wild/weedy with some cultivated accessions. Our results support the hypothesis that Vigna unguiculata var. spontanea is the wild progenitor of cowpea. The accessions from the South, the northern recession accessions, and the wild/weedy could serve as sources of new genes for the genetic improvement of cowpea in Senegal. 相似文献

We present the first genetic map of wild rice ( Zizania palustris L., 2n=2 x=30), a native aquatic grain of northern North America. The map is composed principally of previously mapped RFLP (restriction
fragment length polymorphism) genetic markers from rice ( Oryza sativa 2n=2 x=24). The map is important as a foundation for genetic and crop improvement studies, as well as a reference for genome organization
comparisons among Gramineae species. A comparative mapping approach with rice is especially useful because wild rice is grouped
in the same subfamily, Oryzoideae, and no other mapping comparison has yet been made within the subfamily. As rice is the
reference point for mapping and gene cloning in cereals, establishing a consensus map within the subfamily identifies conserved
and unique regions. The genomes of wild rice and rice differ in total DNA content (wild rice has twice that of rice) and chromosome
pairs (wild rice=15 versus rice=12). The wild rice linkage map reported herein consists of 121 RFLP markers on 16 linkage
groups spanning 1805 cM. Two linkage groups consist of only two markers. Colinear markers were found representing all rice
linkage groups except #12. The majority of rice loci mapped to colinearly arranged arrays in wild rice (92 of 118). Features
of the map include duplication of portions of three rice linkage groups and three possible translocations. The map gives basic
information on the composition of the wild rice genome and provides tools to assist in the domestication of this important
food source.

Understanding the genetic diversity and population structure of Hevea is of great importance for managing its conservation and for utilization of rubber genetic resources. In this study, we investigated the genetic diversity and population structure of eight populations of Hevea rubber genotypes from Malaysia (MY), India (IN), Sri Lanka (LK), Indonesia (ID), France (FR), Thailand (TH), Brazil (BR), and China (CN), in addition to individual primary clones, using 10 nuclear and 11 polymorphic novel chloroplast microsatellite markers (nSSRs and cpSSRs, respectively). ) for both nSSR (0.841) and haploid genetic diversity (cpSSR; 0.207) markers. Bayesian analysis was applied to infer genetic structure based on nSSR data using STRUCTURE software, and a neighbor-joining tree was used to construct an unrooted phylogram based on Nei’s genetic distance that clustered these Hevea genotypes into three groups. Six polymorphic cpSSR markers produced 13 alleles and eight chlorotypes. Seven chlorotypes, A, B, C, D, E, F, and H were detected among Brazilian populations from Acre (AC), Rondônia (RO), and Mato Grosso (MT) locations. The G and H chlorotypes were found in Asiatic genotypes or Wickham clones and only one genotype, 15 AC-F-7 38-62 from AC location. These results provide valuable data for in situ or ex situ conservation and utilization of germplasm collections for breeding programs. 相似文献

Summary Starch gel electrophoresis with two different buffer systems and several substrates and inhibitors have been used to study the electrophoretic variability of esterases in leaves of cultivars of Triticum aestivum. Each one of the buffer systems showed different levels of variability, according to the electrophoretic patterns. At the same time green and etiolated leaves showed different patterns in each buffer system. The variability was dependent upon the developmental stage of the leaves. According to the results from chromosomal location, the genes controlling esterases in green leaves were located in homoeology group 3, while the genes controlling esterases in etiolated leaves were in homoeology group 6. But both esterase isozymes showed a similar electrophoretic migration and a similar reponse to substrates and inhibitors. The possible origin of both sets of genes due to an interchromosomal duplication is discussed. 相似文献

The karyotypes of eight taxa of Orchis L. with 2n = 32 and 2n = 36 have been investigated using morphometrical measurements following staining with Feulgen, Giemsa (C-banding) and the DNA specific fluorochrome Hoechst 33258. The karyotypes of O. coriophora subsp. fragrans, and O. papilionacea proved to be the most asymmetrical, while O. morio and O. longicornu exhibited the most symmetrical karyotypes. Using C-banding and the fluorochrome H33258 only the taxa with high asymmetry indices showed the presence of differentially stained chromatin bands. In most chromosomes heterochromatin bands were present at the telomeric position. The present results seem to indicate that the analysed species do not form a homogeneous group and further subdivisions are possible, which, in turn, do not always correlate with divisions based on morphological characters. Both karyomorphology and heterochromatin distribution coincide in indicating a possible evolutionary pathway. 相似文献

Summary The frequency of aborted fruits and the changes and abnormalities that occur during the embryo development in intraspecific crosses of sweet-potato Ipomoea batatas (2n=6x=90) and interspecific crosses between I. batatas and I. trifida (2n=2x=30) were investigated in order to study the causes of the low seed production. Three genotypes of I. batatas and 18 genotypes of I. trifida were intermated. The frequency of aborted fruits was below 25% in the intraspecific crosses and over 90% in the interspecific crosses. Paraffin sections were used to examine the developmental stages of fruits and seeds. Embryos in different developmental stages were observed to determine the stage of abortion. These observations permitted the identification of developmental stages of embryo rescue in interspecific crosses. There were no significant differences in the frequency of embryo abortion before the early globular stage among female sweet-potato progenitors for the intraspecific and interspecific crosses. The frequency of the late occurrence of embryo abortion (when embryo abortion occurs after the pre-globular stage) was higher in interspecific crosses (19.1%) than in intraspecific crosses (5.5%). The frequency of the late occurrence of embryo abortion in interspecific crosses was higher at the globular stage (9.6%) than at the heart stage (4.3%). Providing that embryo rescue is conducted in interspecific crosses, the estimated number of potentially viable embryos could be increased: 30 times with embryos at the globular stage; 20 times with embryos at the heart stage; and 11 times if embryos at the torpedo stage were used for the rescue with respect to the seed set. The results suggested that the appropriate time for embryo rescue in interspecific crosses is at the globular stage. If embryos could be rescued at the globular stage, it would be possible to increase the number of surviving embryos up to 30 times in interspecific crosses and 0.02 times in intraspecific crosses with respect to natural conditions without embryo rescue.This research was initiated during sabbatical of M.I. at the Asian Vegetable Research and Development Center (AVRDC) in Taiwan 相似文献

Individual plants from the BC1F5 and BC1F6 backcross progenies of barley--wheat (= H. geniculatum All.) (2n = 28) x T. aestivum L. (2n = 42)] and the BC1F6 progeny of their amphiploids were used to obtain alloplasmic euploid (2n = 42) lines L-28, L-29, and L-49 and alloplasmic telocentric addition (2n = 42 + 2t) lines L-37, L-38, and L-50. The lines were examined by genomic in situ hybridization (GISH), microsatellite analysis, chromosome C-banding, and PCR analysis of the mitochondrial 18S/5S repeat. Lines L-29 and L-49 were characterized by substitution of wild barley chromosome 7H1 for common wheat chromosome 7D. In line L-49, common wheat chromosomes 1B, 5D, and 7D were substituted with homeologous barley chromosomes. Lines L-37, L-38, and L-50 each contained a pair of telocentric chromosomes, which corresponded to barley chromosome arm 7H'L. All lines displayed heteroplasmy for the mitochondrial 18S/5S locus; i.e., both barley and wheat sequences were found. 相似文献

We studied some features of the development of self-fertile 42-chromosome lines on the base of self-pollination progeny of 46-chromosome plants obtained by backcrossing of barley--wheat hybrids Hordeum marinum subsp. gussoneanum Hudson (= H. geniculatum All.) (2n = 28) x Triticum aestivum L. (2n = 42). The stabilization of karyotypes, resulting in 42-chromosome plants of the wheat type was generally completed by generation BC1F10. The plants of all self-pollination progenies, including BC1F10, showed some phenotypic traits characteristic of wild barley. Plants of BC1F10 with the chromosome sets 2n = 42 and 2n = 42 + t were analyzed by RAPD with a set of 115 primers. Fragments of the wild barley genome were detected in RAPD patterns with 19 primers. Cross-hybridization confirmed that these fragments belonged to the wild barley genome. We raised four phenotypically different 42-chromosome lines from grains obtained from plants of generation BC1F10, and these lines proved to be cytogenetically stable and self-fertile when grown in the field. 相似文献

Summary Data are presented on the potential of gynogenesis for the production of monohaploids and on factors affecting their frequency and relative vigour. Diploid Solanum tuberosum L. and S. tuberosumx S. phureja Juz et Buk hybrids were used as maternal parents and selected S. phureja clones as prickle pollinators with embryo-spot and nodal band as dominant seed and plant marker. About 2 million seeds were screened for absence of embryo-spot. After raising plants from phenotypically spotless seeds, further screening for absence of nodal bands and for ploidy level was carried out. Finally more than 500 monohaploid plants from three genetically different groups of maternal parents were obtained. Frequency and vigour of the monohaploids were clearly dependent on their maternal genotypes. The data also indicated an effect of the pollinator genotype, the physiological stage of the maternal plant and the environment on monohaploid frequency. On the basis of these results the possibility of breeding for a higher monohaploid production rate and for more stable and vigorous monohaploids is discussed. Furthermore, gynogenesis and androgenesis are compared. It is suggested that both should be used in order to obtain monohaploids from sufficiently various diploid breeding material. 相似文献

Contrary to the generally held view, M. murex Willd. was found to be of 2 n=14 constitution in the great majority of accessions. The 2 n= 16 type, though distributed sparsely over a wide area, is most likely a relic. Chromosome studies at pachytene indicate that the 2 n=14 type probably arose from the 2 n=16 type by translocation of the chromosomal material of the smallest chromosome in the 2 n=16 complement to one of the longer chromosomes, with loss of the centromere of the small chromosome. The total chromosome lengths of the two types thus remained not significantly different, which is in line with the few small morphological differences which were detectable between the two types. Of considerably phylogenetic interest was the finding of complete inter-sterility of the two M. murex types. Neither M. aculeata 2 n= 16 nor M. constricta 2 n=14 could be crossed with M. murex of the same chromosome number. Inviable hybrids were obtained on crossing M. murex 2 n= 16 with M. turbinata 2 n=16. The spiny vs. spineless pod character was investigated in the 2 n=16 type and found to be inherited on a monofactorial basis. 相似文献

Crop wild relatives have a long history of use in potato breeding, particularly for pest and disease resistance, and are expected to be increasingly used in the search for tolerance to biotic and abiotic stresses. Their current and future use in crop improvement depends on their availability in ex situ germplasm collections. As these plants are impacted in the wild by habitat destruction and climate change, actions to ensure their conservation ex situ become ever more urgent. We analyzed the state of ex situ conservation of 73 of the closest wild relatives of potato ( Solanum section Petota) with the aim of establishing priorities for further collecting to fill important gaps in germplasm collections. A total of 32 species (43.8%), were assigned high priority for further collecting due to severe gaps in their ex situ collections. Such gaps are most pronounced in the geographic center of diversity of the wild relatives in Peru. A total of 20 and 18 species were assessed as medium and low priority for further collecting, respectively, with only three species determined to be sufficiently represented currently. Priorities for further collecting include: (i) species completely lacking representation in germplasm collections; (ii) other high priority taxa, with geographic emphasis on the center of species diversity; (iii) medium priority species. Such collecting efforts combined with further emphasis on improving ex situ conservation technologies and methods, performing genotypic and phenotypic characterization of wild relative diversity, monitoring wild populations in situ, and making conserved wild relatives and their associated data accessible to the global research community, represent key steps in ensuring the long-term availability of the wild genetic resources of this important crop. 相似文献

The expansion of trinucleotide repeats has been implicated in 17 neurological diseases to date. Factors leading to the instability of trinucleotide repeat sequences have thus been an area of intense interest. Certain genes involved in mismatch repair, recombination, nucleotide excision repair, and replication influence the instability of trinucleotide repeats in both Escherichia coli and yeast. Using a genetic assay for repeat deletion in E. coli, the effect of mutations in the recA, recB, and lexA genes on the rate of deletion of (CTG)n.(CAG)n repeats of varying lengths were examined. The results indicate that mutations in recA and recB, which decrease the rate of recombination, had a stabilizing effect on (CAG)n.(CTG)n repeats decreasing the high rates of deletion seen in recombination proficient cells. Thus, recombination proficiency correlates with high rates of genetic instability in triplet repeats. Induction of the SOS system, however, did not appear to play a significant role in repeat instability, nor did the presence of triplet repeats in cells turn on the SOS response. A model is suggested where deletion during exponential growth may result from attempts to restart replication when paused at triplet repeats. 相似文献
