【目的】探讨葡萄糖作为外加碳源对热带海洋小球藻(Chloralla sp.HN08)生物质生产和脂、光合色素、碳水化合物及可溶性蛋白等细胞主要成份含量的影响。【方法】分析比较小球藻HN08在光合自养和兼养(添加10 g/L葡萄糖)2种营养方式下的生长速率、细胞密度、光合放氧速率、油脂相对含量,以及可溶性总糖、淀粉和可溶性蛋白的含量。【结果】结果表明,在光照条件下葡萄糖(10 g/L)能促进小球藻(Chloralla sp.HN08)生长,提高细胞终密度,而异养条件下藻细胞逐渐衰亡。兼养条件下,细胞相对生长速率及细胞终密度分别是自养条件下的6.8倍和1.3倍。兼养藻细胞中可溶性糖、淀粉、油脂含量显著高于(P0.05)光合自养细胞,然而可溶性蛋白质和光合色素含量显著低于(P0.05)光合自养细胞。添加葡萄糖的小球藻液的光饱和点和呼吸速率均高于光自养条件下的细胞,但2种培养条件下藻液的净光合速率无显著差异(P0.05)。【结论】光照条件下,添加葡萄糖可显著提高小球藻HN08相对生长速率和细胞终密度,促进油脂与淀粉的积累。  相似文献

Summary In vitro plantlets of Phalaenopsis ‘Happy Valentine’, Neofinetia falcate Hu, Cymbidium kanran Makino, and Cymbidium goeringii Reichb. f. were grown under photoautotrophic [high photosynthetic photon flux (PPF), high CO2 concentration, and increased number of air exchanges] and heterotrophic (low PPF, low CO2 concentration, no air exchanges) culture conditions. After 40 d of culture, a significant difference in plantlet growth was observed between the two cultures. Total fresh and dry mass were on average 1.5 times greater in photoautotrophic culture than in heterotrophic culture. Higher net photosynthetic rates were also observed for Phalaenopsis in photoautotrophic culture. In photoautotrophic culture, little difference was observed in air temperature between the inside and outside of the culture vessel, whereas in heterotrophic culture, air temperature inside the culture vessel was 1–2°C higher than that outside the culture vessel. Relative humidity inside the culture vessel was remarkably different between the two cultures: 83–85% in photoautotrophic culture and 97–99% in heterotrophic culture. These results indicated that growth and net photosynthetic rate of in vitro orchid plantlets were susceptible to the culture environments such as PPF, CO2 concentration, relative humidity (RH), and the number of air exchanges, which would allow a more efficient micropropagation system for these orchid plants.  相似文献

When Euglena gracilis was grown in the heterotrophic condition with glucose and (NH4)2SO4 as the carbon and nitrogen source, a high cell yield (4.28–4.48 g l–1) was obtained and the culture pH decreased to 1.6–2. The biomass production in the heterotrophic culture was compared to those in the autotrophic and mixotrophic cultures. Autotrophic growth was 4.7–6.3% of the heterotrophic one, whereas about 15–19% higher growth was obtained in the mixotrophic culture. Moreover, good production of chlorophyll (39.4 mg l–1) and carotenoids (13.8 mg l–1) were attained in the mixotrophic culture, giving the highest fermenter productivity with respect to biomass as well as chlorophyll and carotenoids. Through an energetic analysis in the mixotrophic culture, it was estimated about 25–28% of the total ATP requirement is formed in the photochemical reactions. This resulted in an improved biomass production in the mixotrophic culture of E. gracilis.  相似文献

To investigate the production potential of eicosapentaenoic acid (EPA) by the diatom Nitzschia laevis, the growth characteristics and fatty acid composition of the cells were studied under photoautotrophic, mixotrophic and heterotrophic conditions of growth. The specific growth rate and maximum biomass concentration were respectively 0.466 d–1 and 2.27 g l–1 for mixotrophic culture, 0.344 d–1 and 2.04 g l–1 for heterotrophic culture, and 0.167 d–1 and 0.5 g l–1 for photoautotrophic culture, respectively. As for EPA production, the yield and productivity were respectively 52.32 mg l–1 and 10.46 mg l–1 d for mixotrophic culture, 35.08 mg l–1 and 6.37 mg l–1 d for heterotrophic culture, and 6.78 mg l–1 and 3.39 mg l–1 d for photoautotrophic culture, respectively. Results suggest that mixotrophic culture is the most suitable growth mode for the production of EPA by the diatom Nitzschia laevis. The results are useful for the development of a cost-effective fermentation process for EPA production by Nitzschia laevis.  相似文献

The effect of nitrogen (N: low = 2% N or moderate = 10% N) levels and cultivation (photoautotrophic or mixotrophic) modes on the biochemicals in Chlorella minutissima was evaluated using a mass culture system. Moderate N and mixotrophic cultures had higher biomass, protein, carbohydrate content and photosynthetic pigments than the low N and photoautotrophic treatments. In contrast, lipid and fatty acid content of the low N and photoautotrophic treatments were higher than in the moderate N and mixotrophic cultures. More phytochemicals were accumulated in moderate N and mixotrophic cultures which corresponded to better antioxidant capacity in the extracts. The most potent (0.7 mg · mL?1) acetylcholinesterase inhibitory activity was displayed by moderate N and mixotrophic treatment. Approximately 60% of the extracts exhibited a noteworthy antimicrobial activity regardless of the N levels and cultivation modes. Thus, moderate N level enhanced the phytochemicals and biological activities of C. minutissima cultured under a mixotrophic system.  相似文献

In microbial cultures, both cellular growth rate and yield (defined as the degree of substrate conversion into the biomass) are important. Although effect of culture conditions on growth kinetics has been well documented for various microbial strains, there is almost no literature concerning the effect of environmental conditions on growth equilibrium, expressed as biomass yield coefficients from substrate. The present paper discusses the effect of culture conditions: irradiance (physical substrate) and glucose concentration (chemical substrate) on biomass yield coefficients from two chemical substrates: glucose and nitrate-nitrogen in photoautotrophic, heterotrophic and mixotrophic culture of blue-green alga Spirulina (Arthrospira) sp. The efficiency of substrates incorporation into the biomass can be precisely determined only if the elemental composition of the biomass is known. The experimental results showed that culture conditions had a substantial influence on biomass yield coefficients (biomass yield from glucose and nitrate-nitrogen). It was found that, the increase of irradiance favoured increase of biomass yield coefficient from both, glucose and nitrate-nitrogen. However, in the case of yield from nitrogen in mixotrophic culture, the effect was opposite. The effect of glucose concentration was different: the higher the initial glucose concentration, the lower the biomass yield coefficients from chemical substrates.  相似文献

A strain of Nannochloropsis isolated originally from the East China Sea and obtained from Institute of Hydrobiology, Chinese Academy of Sciences was shown to utilize glucose or ethanol for mixotrophic and heterotrophic growth. The highest cell density, 550 mg L− 1 dry weight after culture for 8 days, was obtained during mixotrophic culture with 30 mM glucose. The organic carbon sources had no effect on the net photosynthetic rate, but enhanced the respiratory rate. The addition of an organic carbon source led to an increase in the cell lipid content and a decrease in their eicosapentaenoic acid (EPA) content. The EPA yield was 21.9 mg L− 1 using photoautotrophic culture, and 23.4 mg L− 1 and 23.0 mg L− 1, respectively, in mixotrophic cultivation with glucose or ethanol as the carbon source.  相似文献

Photoautotrophic cultivation of Euglena gracilis results in cells with high α-tocopherol content but the final cell concentration is usually very low due to the difficulty of supplying light efficiently to the photobioreactor. On the other hand, Euglena grows heterotrophically to high cell concentrations, using various organic carbon sources, but the α-tocopherol contents of heterotrophically grown cells are usually very low. Sequential heterotrophic/photoautotrophic cultivation, by which cells are grown heterotrophically to high cell concentrations and then transferred to photoautotrophic culture for accumulation of α-tocopherol was therefore investigated for efficient α-tocopherol production. In batch culture, using glucose as the organic carbon source, the cellular α-tocopherol content increased from 120 μg g−1 at the end of heterotrophic phase to more than 400 μg g−1 at the end of the photoautotrophic phase. By using ethanol as the organic carbon source during the heterotrophic phase, adding corn steep liquor as a nitrogen source and optimizing light supply during the photoautotrophic phase, the α-tocopherol content of the cells at the end of the photoautotrophic phase increased to 1700 μg g−1. A system consisting of a mini-jar fermentor (for the heterotrophic phase) and an internally illuminated photobioreactor (for the photoautotrophic phase) was then constructed for continuous sequential heterotrophic/photoautotrophic cultivation. The cells were continuously cultivated heterotrophically in the mini-jar fermentor and the effluent was continuously passed through the photobioreactor for α-tocopherol accumulation. In this way, it was possible to produce 7 g l−1 cells containing about 1100 μg α-tocopherol per g-cell continuously for more than 420 h. The continuous process resulted in α-tocopherol productivity of 100 μg l−1 h−1 which is about 9.5 and 4.6 times higher than those obtained in batch photoautotrophic culture and batch heterotrophic cultures, respectively.  相似文献

Current culture methods based on monocultures under phototrophic regimes are prone to contamination, predation, and collapse. Native cultures of multiple species are adapted to the local conditions and are more robust against contamination and predation. Growth, lipid and biomass productivity of a Louisiana native coculture of microalgae (Chlorella vulgaris) and cyanobacteria (Leptolyngbya sp.) in heterotrophic and mixotrophic regimes were investigated. Dextrose and sodium acetate at C:N ratios of 15:1 and 30:1 under heterotrophic (dark) and mixotrophic (400 μmol m?2 s?1) regimes were compared with autotrophic controls. The carbon source and C:N ratio impacted growth and biomass productivity. Mixotrophic cultures with sodium acetate (C:N 15:1) resulted in the highest mean biomass productivity (156 g m?3 d?1) and neutral lipid productivity (24.07 g m?3 d?1). The maximum net specific growth rate (U) was higher (0.97 d?1) in mixotrophic cultures with dextrose (C:N 15:1) but could not be sustained resulting in lower total biomass than in mixotrophic cultures with acetate (C:N 15:1), with a U of 0.67 d?1. The ability of the Louisiana coculture to use organic carbon for biomass and lipid production makes it a viable feedstock for biofuels and bioproducts.  相似文献

The phototrophic capacity of aerobic anoxygenic phototrophic bacteria endows them with a selective advantage over other heterotrophic bacteria in the oligotrophic ocean. Here, we reported the phototrophic features and proteome of an aerobic phototrophic bacterium Roseobacter denitrificans under starvation stress. The fluorescence induction and relaxation measurements suggested that the photosynthetic capacity in R. denitrificans was preserved but was lower than in the photoautotrophic bacterium Rhodobacter sphaeroides. The existence of light-harvesting complexes (LH1 and LH2) and the reaction center (RC) in the native membrane were demonstrated through atomic force microscopy image analysis as direct evidence of their phototrophy. The homology-based LH1–RC complex structure was proposed in which RC was the Rb. sphaeroides homolog structure surrounded by the LH1. Moreover, the protein expression profiles of cells in the stationary phase under heterotrophic and mixotrophic conditions show that light enhanced or activated some proteins such as carbon monoxide dehydrogenase and NifU to cope with the low levels of amino acids and carbon sources under starvation conditions.  相似文献

Synechocystis sp. PCC 6803 was grown in a 2.5 l enclosed photobioreactor on medium with or without glucose. The incident light intensities ranged from 1.5 klux to 7 klux. The highest average specific growth rates of mixotrophic culture and photoautotrophic culture were, respectively, 1.3 h–1 at a light intensity of 7 klux on 3.2 g l–1 glucose and 0.3 h–1 at both light intensities of 5 klux and 7 klux. The highest cell density 2.5 g l –1 was obtained at both of light intensities 5 klux and 7 klux on 3.2 g glucose l–1. Glucose consumption decreased with decreasing light intensity. The energy yields of mixotrophic cultures were 4 to 6 times higher than that of photoautotrophic cultures. Light favored mixotrophic growth of Synechocystis sp. PCC 6803, especially at higher light intensities (5–7 klux).  相似文献

We attempted to enhance the growth and total lipid production of three microalgal species, Isochrysis galbana LB987, Nannochloropsis oculata CCAP849/1, and Dunaliella salina, which are capable of accumulating high content of lipid in cells. Low nitrogen concentration under photoautotrophic conditions stimulated total lipid production, but a decreasing total lipid content and an increasing biomass were observed with increasing nitrogen concentration. Among the different carbon sources tested for heterotrophic cultivation, glucose improved the growth of all three strains. The optimal glucose concentration for growth of I. galbana LB987 and N. oculata CCAP849/1 was 0.02 M, and that of D. salina was 0.05 M. Enhanced growth occurred when they were cultivated under heterotrophic or mixotrophic conditions compared with photoautotrophic conditions. Meanwhile, high total lipid accumulation in cells occurred when they were cultivated under photoautotrophic or mixotrophic conditions. During mixotrophic cultivation, biomass production was not affected significantly by light intensity; however, both chlorophyll concentration and total lipid content increased dramatically with increasing light intensity up to 150 µmol/m2/s. The amount and composition ratio of saturated and unsaturated fatty acids in cells were different from each other depending on both species and light intensity. The highest accumulation of total fatty acid (C16–C18) among the three strains was found from cells of N. oculata CCAP849/1, which indicates that this species can be used as a source for production of biodiesel.  相似文献

Production of biomass and phycocyanin (PC) were investigated in highly pigmented variants of the unicellular rhodophyte Galdieria sulphuraria, which maintained high specific pigment concentrations when grown heterotrophically in darkness. The parental culture, G. sulphuraria 074G was grown on solidified growth media, and intensely coloured colonies were isolated and grown in high-cell-density fed-batch and continuous-flow cultures. These cultures contained 80–110 g L−1 biomass and 1.4–2.9 g L−1 PC. The volumetric PC production rates were 0.5–0.9 g L−1 day−1. The PC production rates were 11–21 times higher than previously reported for heterotrophic G. sulphuraria 074G grown on glucose and 20–287 times higher than found in phototrophic cultures of Spirulina platensis, the organism presently used for commercial production of PC.  相似文献

The ability of Acidithiobacillus caldus to grow aerobically using pyruvate, acetate, citrate, 2-ketoglutarate, succinate, and malate as either an electron donor and carbon source (heterotrophic growth), or as a carbon source when potassium tetrathionate was added as an electron donor (mixotrophic growth), was tested in chemostat cultures. Under both heterotrophic and mixotrophic conditions, organic acids were added to a sub-lethal concentration (50 μM). Under mixotrophic conditions, potassium tetrathionate was added to an excess concentration (10 mM). No cell growth was observed under heterotrophic conditions; however, effluent cell concentrations increased over threefold when pyruvate was coupled with potassium tetrathionate. Under these conditions, the effluent pyruvate concentration was reduced to below the detection limit (2 μM), and oxygen consumption increased by approximately 100%. Although pyruvate provided a carbon source in these experiments, ambient carbon dioxide was also available to the cells. To test whether At. caldus could grow mixotrophically using pyruvate as a sole carbon source and potassium tetrathionate as an electron donor, cells were batch cultured in a medium free of dissolved inorganic carbon, and with no carbon dioxide in the headspace. These experiments showed that At. caldus was able to convert between 65 ± 8 and 82 ± 15% of the pyruvate carbon to cellular biomass, depending on the initial pyruvate concentrations. This work is the first to identify a defined organic-carbon source, other than glucose, that At. caldus can assimilate. This has important implications, as mixotrophic and heterotrophic activity has been shown to increase mineral leaching in acidic systems.  相似文献

Combined effect of light intensity and glucose concentration on Arthrospira platensis growth and photosynthetic response was evaluated using a 32 factorial design. This design was carried out with light levels of 50, 100, and 150 μmol photons m−2 s−1 and glucose concentrations of 0.5, 1.5, and 2.5 g L−1. Results from the response surface methodology were that the highest level of light intensity and glucose concentration improved biomass (1.33 g L−1), maximum specific growth rate (0.49 day−1), and net photosynthetic rate (139.89 μmol O2 mg Chl−1 h−1). Furthermore, the interaction of both factors showed that at low light, glucose had a low effect on maximum biomass and maximal net photosynthetic rate. However, at the highest light levels, the effect of glucose was more sensitive and the increase of glucose concentration increased the levels of all responses. The rates of the instantaneous relative growth, net photosynthesis, and dark respiration of growth cultures showed two different phases in mixotrophic condition. The first was distinguished by the preponderance of the photoautotrophic mode; the second was based mainly on photoheterotrophy.  相似文献

Chlorella strain (UTEX 27) maintains optimal photosynthetic capacity when growing photoautotrophically in the presence of ammonium. Nitrate-grown photoautotrophic cells, however, show a drastic loss of chlorophyll content and ribulose-1,6-bisphosphate carboxylase/oxygenase activity, resulting in a greater than 10-fold decrease in photosynthetic capacity and growth rate. Nitrate-grown cells are not deficient in protein content, and under mixotrophic and heterotrophic conditions, the alga can utilize nitrate as well as it does ammonium. The alga metabolizes both glucose and acetate in the dark with a doubling time of 5 to 6 hours. However, its growth on acetate is inhibited by light. Ribulose-1,6-biphosphate carboxylase/oxygenase activity correlates well with photosynthetic capacity, and glucose 6-phosphate dehydrogenase and hexokinase activities are altered in a manner consistent with the availability of glucose in growing cells. The alga appears to assimilate ammonium under photoautotrophic conditions primarily via the glutamine synthetase pathway, and shows an induction of both NADH and NADPH dependent glutamate dehydrogenase pathways under mixotrophic and heterotrophic conditions. Multiple isoforms are present only for hexokinase and glucose 6-phosphate dehydrogenase. Etiolated nitrate-grown cells resume greening and increase their photosynthetic capacity after about 6 hours of incubation in the presence of ammonium under photoautotrophic conditions. Similarly, the loss of photosynthetic capacity in ammonium-grown photoautotrophic cells commence about 9 hours after their transfer to heterotrophic nitrate containing media.  相似文献

Beggiatoa alba strain B18LD was grown in continuous culture under heterotrophic conditions on acetate or acetate and asparagine and under mixotrophic conditions on acetate plus either 1 mM sodium sulfide or 1 mM sodium thiosulfate. Considerable differences were observed between the yields and the cell compositions of heterotrophic and mixotrophic cultures at all dilution rates tested. The dry weight yield per gram acetate utilized was approximately three times higher in the acetate-sulfide mixotrophic culture than in the acetate heterotrophic culture, whereas the poly--hydroxybutyric acid and carbohydrate contents were much higher in the heterotrophic cultures. The high yields (0.52–0.75, corrected for the weight of the sulfur) obtained with the mixotrophic cultures imply that the acetate was utilized mainly for biosynthesis. Thus, the oxidation of sulfide supplied energy. The addition of catalase to the chemostat cultures increased yields slightly, but it was insufficient to explain the differences between the heterotrophic and the mixotrophic cultures.  相似文献

Spirulina (Arthrospira) platensis (Nordstedt) Geitler cells grown under mixotrophic conditions exhibit a modified response to light. The maximal photosynthetic rate and the light saturation value of mixotrophic cultures were higher than those of the photoautotrophic cultures. Dark respiration and light compensation point were also significantly higher in the mixotrophically grown cells. As expected, the mixotrophic cultures grew faster and achieved a higher biomass concentration than the photoautotrophic cultures. In contrast, the growth rate of the photoautotrophic cultures was more sensitive to light. The differences between the two cultures were also apparent in their responses to exposure to high photon flux density of 3000 μmol·m 2·s 1. The light-dependent O2 evolution rate and the maximal efficiency of photosystem II photochemistry declined more rapidly in photoautotrophically grown than in mixotrophically grown cells as a result of exposure to high photon flux density. Although both cultures recovered from the high photon flux density stress, the mixotrophic culture recovered faster and to a higher extent. Based on the above results, growth of S. platensis with a fixed carbon source has a significant effect on photosynthetic activity.  相似文献

Chlorella sorokiniana was cultured in heterotrophic or mixotrophic mode in outdoor enclosed tubular photobioreactor. The culture temperature was maintained at 32–35 °C. At night, theChlorella culture grew heterotrophically, and 0.1 M glucose was completely consumed. The biomass growth yield of glucose was 0.35 ± 0.001 g-biomass g-glucose–1. During the day, the algal culture grew mixotrophically and the biomass growth yield was 0.49 g-biomass g-glucose–1 in low density culture (initial biomass concentration, Xo = 2 g l–1), 0.56 g-biomass g-glucose–1 in medium density culture (Xo = 4 g l–1) and 0.46 g-biomass g-glucose–1 in high density culture (Xo = 7 g l–1). The daily area productivity of the culture, with Xo = 4 g l–1 corresponded to 127 g-biomass m–2 d–1 during the day and 79 g-biomass m–2 d–1 during the night. In all the cultures, the dissolved O2 concentration increased in the morning, reached the maximum value at noon, and then decreased in the afternoon. The dissolved CO2 concentration remained at 3 mBar in the morning and increased in the afternoon. Glycolate was not found to accumulate in culture medium.  相似文献

The growth, photosynthesis, and respiration of the marine diatom Phaeodactylum tricornutum were examined under photoautotrophic and mixotrophic conditions. 100 mM glycerol, acetate, and glucose significantly increased specific growth rate, and mixotrophic growth achieved higher biomass concentrations. Under mixotrophic conditions, respiration rate (R d) and light compensation irradiance (I c) were significantly higher, but net maximum photosynthetic O2 evolution rate (P m) and saturation irradiance (I k) were depressed. Organic carbon sources decreased the cell photosynthetic pigment content and chlorophyll a to c ratio, but with a higher carotenoid to chlorophyll a ratio. Ratios of variable to maximum chlorophyll fluorescence (F v/F m) and 77 K fluorescence spectra of mixotrophic cells indicated a reduced photochemical efficiency of photosystem II. The results were accompanied by lower electron transport rate. Therefore, organic carbon sources reduced the photosynthesis efficiency, and the enhancement of biomass of P. tricornutum implied that organic carbon sources had more pronounced effects on respiration than on photosynthesis.  相似文献
