Total histones estimated after M. Alfred and I. Geschwind were studied cytophotometrically in nuclei of neurons of different types (layers III and V in the frontal-parietal cortex and in the thalamic lateral nucleus) of the "mirror" epileptiform focus in the rat brain on the 11th and 63d day after cobalt implantation. On the 11th day of the experiment, integral optic density of the histones decreased by 33%, 25% and 37%, and the area of the nuclei -- by 27%, 24% and 35% in the neurons of layers II and V in the frontal-parietal cortex and in the thalamic lateral neucleus, respectively. On the 63d, integral optic density of the histones in the neurons mentioned increased respectively by 42%, 52% and 42%, and the area of the nuclei -- by 21%, 43% and 27%. Concentration of the histones in the nuclei decreased by 3--9% on the 11th day and increased, except layer V, by 9--18% on the 63d of the experiment. Interconnection between changes in histone content and total proteins in cytoplasm of functionally different neurons in the "mirror" epileptiform focus is disscussed, with special reference to histones as a regular of genetic activity for protein synthesis.  相似文献

Protein content in the neurons of layers III and V in the rat brain anterior-parietal cortex and the lateral thalamic nucleus was studied 11 and 63 days after cobalt implantation. In the course of the experiment, protein content increased by 44--49% in the neurons of layer III and decreased by 24--32% in the thalamic neurons. In small and large pyramids of layer V, protein content decreased by 21--28%, 11 days after cobalt implantation, and approached normal content by the 63rd day. It was concluded that according to morpho-functional characteristic of certain neuronal types (associative, in layer III, predominantly efferent, in layer V and associative neurons in subcortical formations of the lateral thalamic nucleus) their protein changes in response to convulsive activity were different.  相似文献

The timetable of cell generation, neuronal death and neuron numbers in the fused proximal glossopharyngeal (IX) and vagal (X) ganglion and distal IX and X ganglia were studied in normal and nerve growth factor (NGF) treated chick embryos. 3H-thymidine was injected between the 3rd and 7th days of incubation and embryos sacrificed on the 11th day. Neurons in the distal IX and X ganglia were generated between the 2nd and 5th days of incubation, the peak mitotic activity occurring on the 4th and 3rd days, respectively. Neurons of the proximal IX and X ganglion were generated between the 4th and 7th days, with maximum neuron generation on the 5th day of incubation. Counts of neurons in the 3 ganglia between the 5th and 18th days of incubation showed a maximum of 22,000 on the 8th day in the proximal IX and X ganglion and this decreased to 12,000 by the 13th day. In the distal IX ganglion, the neuron number decreased by 44% from 4,500 on the 6th day to 2,500 by the 11th day. A similar decrease of 43% was found in the distal X ganglion, the neuron number falling from 11,500 on the 7th day to 6,500 by the 11th day of incubation. Neuronal cell death in these ganglia extended from the 5th to the 12th day of incubation, maximum cell death occurring at or after the cessation of mitotic activity. NGF administration from the 5th to the 11th day of incubation did not have a measurable effect on the neurons of proximal IX and X and distal IX ganglia, but increased neuronal survival by 30% in the distal X ganglion.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The experimental lesion in rat brain was produced by the implantation of cobaltogelatin rod into the right parietalis anterior area. A quantitative measurement of total protein was made by microdensitometer in neurons controlateral to cobalt lesion at 112-115 days after cobalt implantation. In the experimental animals, protein content decreased in neurons of lamina III and V area parietalis anterior and the nucleus lateralis thalami by 6, 60 and 53%, resp. It has been concluded that different type neurons have different protein changes in response to paroxyzmal activity at late stages of epileptogenic mirror focus.  相似文献

The weekly administration of 1,2-dimethyl-hydrazine (DMH) by subcutaneous injection for a period of 16-20 weeks is a well known procedure for producing colonic tumors in mice and rats. Quantitative histomorphological, histochemical and kinetic studies, as well as investigation of the significance of epithelial cell density were carried out in mice between the 7th and the 91st day after the first DMH injection. These studies showed that between the 28th and the 35th day, several simultaneous alterations in the colonic epithelium involving modification of glandular form, decreased mucus secretion, an increase in epithelial cell density and an increase in the number of S phase cells (BrdU labeling index: LI). Around the 35th day, the glands tended to expand and from the 35th to the 63rd day, they were stretched and displayed compartments of dedifferentiated and non-mucinous crypts (DNMC). In these crypts the cell density became very high, reaching twice the control value on the 91st day. This feature was accompanied by alteration in cell morphology and by an increase in the available basement membrane area. A decrease in mucus secretion was apparent from the 14th day and by the 63rd day, mucus secretion was only about 60% of the control value in all crypts. The LI was increased until the 35th day following which a paradoxical and progressive decrease occurred in all glandular compartments.  相似文献

Synthesis of RNA in neurons of the hypoglossal nerve nucleus after axonal section was studied by means of [5-3H]uridine administration and radioautographic counting techniques in mice. The results of the experiments were evaluated by counts of silver grains over the nucleoplasm and cytoplasm of the neurons. RNA synthesis was greater in neurons after axonal section, and this increase was evident from 12 hr after the operation. The greatest increases in the operated side were observed in the 1st, 2nd and 3rd days after operation. In the 7th and 14th days RNA synthesis was still greater in the hypoglossal nucleus of the sectioned nerve but the difference in the control nucleus was not so striking. In the 30th day synthesis of RNA in left and right hypoglossal nuclei was comparable.  相似文献

Sensory fibers regeneration after dorsal root ganglion (DRG) section was studied in the rat. After middle cross-section of DRG-13 (left side) there were neurons in the proximal part with damage to peripheral processes and in the distal part with damage to central processes. Axonal ionophoresis of cobalt salts was used for the study of sensory fibers regeneration through the scar in DRG during 3, 7, 15, 30, 120 and 180 days after the damage. Peculiarity regeneration of the sensory fibers was shown in spite of damage localization near ganglion cells body, the regeneration of peripheral and central processes of ganglion cells started already on the 3rd day, and sprouting sensory fibers through the scar of DRG--on the 7th day.  相似文献

The present study describes the major aspects of how monoaminergic (serotonin, dopamine) systems change in the course of regeneration of the brain in the earthworm (Eisenia fetida), investigated by immunocytochemistry, HPLC assay, and ligand binding. Following brain extirpation, the total regeneration time is about 80 days at 10 degrees C. On the 3rd postoperative day serotonin, and on the 11th postoperative day tyrosine hydroxylase-immunoreactive neurons can be observed in the wound tissue. Thereafter the number of the immunoreactive cells increases gradually, and by the 76th-80th postoperative days all serotonin- and tyrosine hydroxylase-immunopositive neurons can be found in their final positions, similarly to those observed in the intact brain. Labeled neurons located in the dorsal part of the regenerated brain appear earlier than the cells in lateral and ventral positions. Both serotonin- and tyrosine hydroxylase-immunoreactive neurons of the newly formed brain seem to originate from undifferentiated neuroblasts situated within and around the ventral ganglia and the pleura. Dopaminergic (tyrosine hydroxylase-immunoreactive) elements may additionally derive from the proliferation of neurons localized in the subesophageal ganglion and the pharyngeal nerve plexus. Following brain extirpation, both serotonin and dopamine levels, assayed by HPLC, first increase in the subesophageal ganglion; by the 25th day of regeneration, the monoamine content decreases in it and increases in the brain. Hence it is suggested that monoamines are at least partly transported from this ganglion to the regenerating brain. At the same time, (3)H-LSD binding can be detected in the regenerating brain from the 3rd postoperative day, showing a continuous increase until the 80th postoperative day, suggesting a guiding role of postsynaptic elements in the monoaminergic reinnervation of the newly formed brain.  相似文献

目的应用活体荧光技术,研究血管损伤后初期病变形成的动态变化。方法 112只雄性LDLR-/-小鼠随机分成14组,每组8只。将绿色荧光蛋白表达而低密度脂蛋白受体敲除(GFP+/LDLR-/-)小鼠的骨髓移植到LDLR-/-小鼠中,行血管损伤手术。从术后第1天至14天,麻醉小鼠,在荧光显微镜下直接观察股动脉血管病变变化的动态状况。结果术后第1天即见血管内大量荧光细胞随血液高速循环,术后第3天出现血液中的荧光细胞呈点状粘附于血管内壁,术后第6天,在血管内壁荧光细胞粘附的部位,外膜组织开始明显增生,增生的外膜组织中可见荧光细胞,此时血管内壁的病变呈不规则的片状分布。术后第9天,血管外纤维组织显著增生,并见大量的荧光细胞,同时可见外膜组织中有血液流动的新生营养血管。至病变第14天,受损血管的病变程度在以前的基础上继续增加,病变部位血管内膜上粘附聚集大量的荧光细胞,形成内衬而附着于血管内膜。结论血管损伤后的初期病变存在着由血管内到外的发展趋势。病变的形成与循环血中骨髓来源的干细胞在内膜部位粘附和聚集具有紧密的联系,血管内膜的病变对血管外纤维组织的增生具有明显的影响。  相似文献

目的:研究丙戊酸(VPA)浓度和干预时间对神经干细胞(NSCs)体外分化的影响。方法:以不同浓度的VPA(0.1、0.3、0.5、0.75和1.0mmol/L)处理原代培养的神经干细胞,以NB培养基组做对照,分别于神经干细胞分化后3天、7天、10天和14天用免疫荧光双标鉴定并计数微管蛋白-Ⅲ(β-tubllin III)和胶质原纤维酸性蛋白(GFAP)阳性细胞的比例,并作统计学分析。结果:同一时相点组间比较,3天时各组中神经元分化比例无显著差异;7天时不同浓度VPA组与对照组分化神经元比例开始呈现差异;10天时这种差异继续增大,0.75 mmol/L VPA组中神经元比例为82.15±0.93%;14天时保持这种差异;但各时相点1.0 mmol/L VPA组与0.75 mmol/L VPA组神经元分化的比例无显著差异。不同时相点组内比较发现,3-10天内随着时间的延长,各组神经元分化的比例显著增加,但14天时神经元分化的比例较10天无显著变化。结论:0.75mmol/L VPA在10天时促神经干细胞向神经元分化的作用最佳。  相似文献

The effect of pre-loading and in situ loading of cobalt onto a cobalt-limited granular sludge on the performance of methanol fed bioreactors was investigated. One upflow anaerobic sludge bed (UASB) reactor was inoculated with cobalt pre-loaded sludge (24h; 30 degrees C; 1 mM CoCl2) and a second UASB with unloaded sludge. The UASB reactors (30 degrees C; pH 7) were operated for 77 days at 8 h hydraulic retention time and organic loading rates ranging from 5 to 20 g COD.L reactor(-1).d(-1). Cobalt pre-loading clearly stimulated the methanogenic activity of the sludge with methanol as the substrate, e.g., after 30 days of reactor operation this activity was 5.8 times higher than that of the cobalt unloaded sludge. During the experiment, part of the cobalt leached from the pre-loaded sludge, i.e., 54% of the cobalt content was lost during the 77 days of reactor operation. Sequential metal extraction showed that losses mainly occurred from the exchangeable and carbonate fraction and in the sludge remaining cobalt was mainly present in the organic/sulfide fraction of the sludge. In situ loading of cobalt in the unloaded UASB reactor on day 57 by adding 31 microM cobalt to the influent for a 24-h period (16% of the cobalt present in the loaded sludge at day 11) resulted in a 4 time increase of the methanogenic activity of the sludge with methanol as the substrate at the end of the reactor experiment, while the accumulated amount of cobalt in the sludge only amounted to 6% of the cobalt accumulated in the loaded sludge (on day 11). This study showed that both pre-loading sludge and in situ loading are adequate for achieving an increased reactor performance of methanol fed UASB reactors operating under cobalt limitation. However, the in situ dosing procedure needs substantially lower amounts of cobalt, while it also gives significantly smaller losses of cobalt with the effluent.  相似文献

目的:探讨超声乳化白内障手术患者角膜内皮细胞的损伤情况。方法:收集我院确诊为白内障的患者121例,随机分配为微切口组与常规切口组。常规切口组采用3.0 mm切口超声乳化白内障手术方案,微切口组采用1.8 mm小切口超声乳化白内障手术。手术前、手术后1日、7日、1个月、3个月监测患者角膜内皮细胞密度、六角形细胞比例、角膜内皮细胞变异系数及中央角膜厚度。结果:与治疗前相比,两组患者手术后1日、7日、1个月、3个月的角膜内皮细胞密度、六角形细胞比例及角膜内皮细胞变异系数均较治疗前显著降低(P0.05);手术后1日、7日时,两组患者的中央角膜厚度均较手术前明显变薄,有统计学差异(P0.05);手术后1个月、3个月,两组患者的中央角膜厚度均呈降低趋势,最终与手术前相似。微切口组患者不同时点六角形细胞比例与同期常规切口组比较均显著升高,角膜内皮细胞变异系数与同期常规切口组比较均明显降低有统计学差异(P0.05)。两组患者手术前后不同时间点角膜内皮细胞密度、中央角膜厚度组间比较均无统计学差异(P0.05)。结论:超声乳化白内障手术后患者角膜内皮细胞损伤与手术切口有相关性,缩小手术面积的小切口手术使术后修复增快,安全有效,适宜临床推广。  相似文献

Cerebrospinal ganglia of 11-, 13-, 15-, 18- and 20-day-old embryos have been studied light- and electron-microscopically. On the 11th day of anlage the ganglia are presented exclusively as neuroblasts: the satellite-cells are formed later from lemmoblasts that install into the ganglion on the 13th-15th days both from the surrounding mesenchyme and from the developing roots and trunklets of the cerebrospinal nerves. In the ganglion they undergo proliferation and further differentiation. On the 15th day simple satellite-neuron interrelationships are formed: one satellite-cell is tightly connected with perikaryons of some neuroblasts and young neurons. On the 18th day around some neurons a double layered capsule is formed from the satellite-cells. Multilayered tunics are formed only on the 10th-15th day after birth. During the process of the capsule formation an unusual reactive character of the satellite-neuron interrelationships is revealed, it is manifested as, performed by the satellite-cells, an active microphagocytosis of the perikaryon cytoplasmic processes and the initial segment of the sensitive neuron main process. In embryogenesis the lemmocyte-axon interrelationships develop earlier than the satellite-neuronal ones.  相似文献

本文收集了一只14岁大熊猫产后17d内的乳汁样品10个,包括产后第2,3,5,6,7,8,9,11,12和17天,并测定了这些样品的营养成分的含量。即粗蛋白(CP)、粗脂肪(EE)、乳糖(Lac)、17种氨基酸(AA)含量的总和、5种常量元素(Ca,P,K,Na,Mg)和6种微量元素(Fe,Cu,Mn,Zn,Co,Se)。结果如下:(1)每个样品的粗蛋白和17种氨基酸含量的总和十分接近,分别在5.20%~7.50%和5.17%~7.62%的范围内波动;乳糖含量的变化范围为0.87%~1.51%;粗脂肪为0.6%~3.2%;(2)粗蛋白和总氨基酸含量第3天明显低于第2天,从第3天到第6天逐步升高,第7天有些下降,从第7到17天逐渐上升;乳糖含量变化不大,在1%左右,第2,3和5天接近,第6天明显升高。之后逐渐降低直至第17天;粗脂肪含量波动较大,没有明显变化趋势;绝大多数矿物元素含量随产后时间的延长都有不同程度的升高趋势;(3)产后6~7天的乳汁样品营养物质含量与一周后有明显差别。作者建议在幼大熊猫人工乳的配制方面,应依据大熊猫乳汁的营养物质含量,充分保证蛋白质水平。降低乳糖含量,同时供给充足的矿物质元素;必须根据幼大熊猫的日龄及其生长发育情况,调整人工乳的营养水平。  相似文献

Vestibular compensation is the process of behavioral recovery following peripheral vestibular lesion. In clinics, the histaminergic medicine is the most widely prescribed for the treatment of vertigo and motion sickness, however, the molecular mechanisms by which histamine modulates vestibular function remain unclear. During recovery from the lesion, the modulation of histamine receptors in the medial vestibular nucleus (MVN) and the flocculus may play an important role. Here with the means of quantitative real-time PCR, western blotting and immunohistochemistry, we studied the expression of histamine receptors (H1, H2, and H3) in the bilateral MVN and the flocculus of rats on the 1st, 3rd, and 7th day following unilateral labyrinthectomy (UL). Our results have shown that on the ipsi-lesional flocculus the H1, H2 and H3 receptors mRNA and the protein increased significantly on the 1st and 3rd day, with compare of sham controls and as well the contralateral side of UL. However, on the 7th day after UL, this expression returned to basal levels. Furthermore, elevated mRNA and protein levels of H1, H2 and H3 receptors were observed in the ipsi-lesional MVN on the 1st day after UL compared with sham controls and as well the contralateral side of UL. However, this asymmetric expression was absent by the 3rd post-UL. Our findings suggest that the upregulation of histamine receptors in the MVN and the flocculus may contribute to rebalancing the spontaneous discharge in bilateral MVN neurons during vestibular compensation.  相似文献

Correlation between activities of neurons in the right and left lateral hypothalamus of a rabbit recorded during quiet wakefulness, after 24-h food deprivation, and after satiation was studies by plotting cross- and autocorrelation histograms. A predominant order in correlated bilateral discharges of hypothalamic neurons was revealed in hungry animals: in a significantly greater number of cases (59%) the left-side discharges led the right-side ones with time delays to 200 ms (peaks at 30 and 160 ms). The opposite order of discharges was less common (21%). In the state of hunger, the probability of appearance of theta-range frequencies in correlated neuronal discharges increased (from 11 to 29%).  相似文献

To determine whether behavioral sensitization produced by prolonged D-amphetamine administration affects susceptibility of nigrostriatal dopaminergic neurons to the neurotoxic actions of 6-hydroxydopamine (6-OHDA), rats were treated daily from the 23 rd day after birth for 11 consecutive days with D-amphetamine (1.0 mg/kg s.c.) or saline. On the last day of treatment, one group primed with D-amphetamine and one control group of rats were tested to confirm behavioral sensitization development. The remaining animals were additionally treated on the 34 th day (one day after the last D-amphetamine injection) with 6-OHDA HBr (300 microg in 10 microl i.c.v., salt form, half in each lateral ventricle) or its vehicle. Four weeks later the levels of dopamine (DA) and its metabolites 3,4-dihydroxyphenylacetic acid (DOPAC), homovanillic acid (HVA), 3-metoxytyramine (3-MT), as well as 5-hydroxytrypatmine (5-HT) and its metabolite 5-hydroxyindoleacteic acid (5-HIAA) were assayed in the striatum, by HPLC/ED. In rats with behavioral sensitization, 6-OHDA reduced endogenous dopamine and its metabolites content to a comparable degree in comparison to controls. This finding indicates that presumed up-regulation of the dopamine transporter in the behaviorially sensitized rats did not increase the neurotoxicity of a high dose of 6-OHDA.  相似文献

After the extirpation of the brain reorganization of the peptidergic (FMRFamide, neuropeptide Y, proctolin) systems was studied in the newly forming cerebral ganglion of the annelid Eisenia fetida. During regeneration, all immunoreactive fibres appear on the 1st-2nd postoperative day. At the beginning of regeneration, immunoreactive neurons and fibres form a mixed structure in the wound tissue. On the 3rd postoperative day, FMRFamide positive and neuropeptide Y-immunoreactive, while on the 7th postoperative day proctolin-immunoreactive neurons appear in the loose wound tissue. From the 25th postoperative day a capsule gradually develops around it. The neurons of the preganglion move to the surface of the newly appearing preganglion. The number of these cells gradually increase, and by the 72th-80th postoperative days the localization and number of peptide-immunoreactive neurons is similar to that in the intact one. The neurons of all examined peptidergic systems may originate from the neuroblasts, situated on the inner and outer surface of the intact ganglia (e.g. suboesophageal and ventral cord ganglia). In addition FMRFamide and proctolin immunoreactive neurons may take their derive by mitotic proliferation from the pharyngeal neurons, too.  相似文献
