拟南芥中PAP特异磷酸酶(AtAHL)的折叠识别与结构预测

通过现有的序列同源性比较、二级结构预测、三维结构预测和模拟方法,得到了拟南芥中PAP特异磷酸酶的三维结构.这是一种与酵母中的Hal2p蛋白质类似,并且N端为α＋β,C端为α/β结构域的多结构域蛋白质.分析预测所得结构,发现了Mg²⁺等金属离子的结合位点,推测了对Na⁺敏感的结构基础.这些结合位点与其生化功能相关.而且,通过结构与功能分析,讨论了蛋白质数据库(PDB)中同一个酶已有理论结构的不合理性.     

Immunocytochemical localization of host (Apis mellifera) proteins in growing oocytes of a hemophagous mite (Varroa jacobsoni) by the unlabeled antibody—enzyme (PAP) method

The mite, Varroa jacobsoni is a hemophagous ectoparasite of honeybees (Apis spec.). Host hemolymph proteins are resorbed by Varroa without digestive degradation. By immunocytochemical methods, these foreign proteins can be localized in growing Varroa oocytes; they are assimilated into the eggs.     

Synthesis, structure and characterization of new complexes [Fe2(μ-OMe)2(PAP)(X)4] (PAP= 1,4-di(2-pyridyl)aminophthalazine, X=Cl, OAc) and their oxidation catalysis

Two dinuclear iron(III) complexes with tetradentate N-donor ligand 1,4-di(2^′-pyridyl)aminophthalazine (PAP), [Fe₂(μ-OMe)₂(PAP)Cl₄] (2) and [Fe₂(μ-OMe)₂(PAP)(OAc)₄] (3) were prepared and characterized. Single crystal X-ray molecular structure of [Fe₂(μ-OMe)₂(PAP)Cl₄] · 2MeOH have been elucidated. The six-coordinate iron atoms are in distorted octahedral environment bridged by the oxygen atoms of two methoxy groups and the PAP ligand. The Mössbauer spectra of both complexes show one quadrupole doublet and the isomer shift and quadrupole splitting values indicate the presence of octahedral high-spin Fe^III ions. Complex 2 showed catalytic activity for alkane oxidation with hydrogen peroxide.

多重PAP及多重PAP结合ABC免疫细胞化学方法的建立、敏感性分析与应用

作者在传统的 PAP 及 ABC 染色方法的基础上,通过重复使用桥抗及 PAP 的次数或重复使用生物素化桥抗及 PAP 和 ABC 的次数,使 PAP 与 ABC 方法成功地结合,增加连接于抗原部位的酶分子数,建立起两种敏感的免疫细胞化学方法。染色结果经显微分光光度图象分析,方差分析,回归分析及染色效果的直接观察表明,重复染色可增加阳性强度,扩大阳性染色范围,提高信噪比,从而提高方法的敏感性,以生物素化桥抗取代非生物素化桥抗,将 ABC 引入到多重 PAP 方法中,可使方法的敏感性进一步提高。该方法结合单克隆抗体及多克隆抗体成功地检测出用传统双PAP 及 ABC 难以或不能检测出的常规福尔马林固定石蜡包埋的肾综合征出血热尸检组织中的病毒抗原及石蜡包埋肿瘤活检组织中的桥粒抗原。     

The Reg3α (HIP/PAP) Lectin Suppresses Extracellular Oxidative Stress in a Murine Model of Acute Liver Failure

MethodsPrimary hepatocytes stressed with the reactive oxygen species (ROS) inducers TNFα and H₂O₂ were incubated with a recombinant Reg3α protein. ALF was induced in C57BL/6J mice by an anti-CD95 antibody. Livers and primary hepatocytes were harvested for deoxycholate separation of cellular and extracellular fractions, immunostaining, immunoprecipitation and malondialdehyde assays. Fibrin deposition was studied by immunofluorescence in frozen liver explants from patients with ALF.ResultsFibrin deposition occurs during experimental and clinical acute liver injuries. Reg3α bound the resulting transient fibrin network, accumulated in the inflammatory extracellular matrix (ECM), greatly reduced extracellular ROS levels, and improved cell viability. Hepatocyte treatment with ligands of death receptors, e.g. TNFα and Fas, resulted in a twofold increase of malondialdehyde (MDA) level in the deoxycholate-insoluble fractions. Reg3α treatment maintained MDA at a level similar to control cells and thereby increased hepatocyte survival by 35%. No antioxidant effect of Reg3α was noted in the deoxycholate-soluble fractions. Preventing fibrin network formation with heparin suppressed the prosurvival effect of Reg3α.ConclusionsReg3α is an ECM-targeted ROS scavenger that binds the fibrin scaffold resulting from hepatocyte death during ALF. ECM alteration is an important pathogenic factor of ALF and a relevant target for pharmacotherapy.

本文介绍一种碱性磷酸酶标Ａ蛋白加强的ＰＡＰ技术。采用ＰＡＰ技术、碱性磷酸酶标Ａ蛋白（ＰＡＡＰ）技术ＰＡＡＰ和加强的ＰＡＰ（ＰＡＰ－ＰＡＡＰ）技术显示下丘脑室旁核催产素（ＯＴ）能神经元。结果发现,其中使用ＰＡＰ－ＰＡＡＰ技术免疫反应产物的显色最深。此技术的原理可能是,由于Ａ蛋白分子至少有四个位点能与ＩｇＧ分子的Ｆｃ段高亲合性地结合,故在该技术中,先经过ＰＡＰ程序的三步免疫反应并显色后,每个与一抗结合的二抗分子上和每个与二抗结合的ＰＡＰ复合物分子上各暴露一个能与Ａ蛋白分子结合的Ｆｃ段,在随后经过ＰＡＡＰ技术处理时,部分ＰＡＡＰ复合物分子就结合在这些Ｆｃ段上,经显色后,ＰＡＡＰ技术显示的浅紫兰色与ＰＡＰ技术显示的浅棕褐色重叠,变成更深的反差明显的深棕褐色。     

Syntheses of (±)-Epoformin (Desoxyepoxydon) and (±)-Epiepoformin (Desoxyepiepoxydon)

Boron is an essential nutrient for plants, but it is toxic in excess. Transgenic rice plants expressing an Arabidopsis thaliana borate efflux transporter gene, AtBOR4, at a low level exhibited increased tolerance to excess boron. Those lines with high levels of expression exhibited reduced growth. These findings suggest a potential of the borate transporter BOR4 for the generation of high-boron tolerant rice.     

光对商陆发状根生长和PAP基因表达的影响

利用发根农杆菌菌株Ar1334与美洲商陆(Phytolacca americana)叶片外植体共培养转化体系,共获得58个发状根无性系(SL-1~58).以发根农杆菌Ri质粒TL-DNA上的rol C基因设计特异引物,对发状根进行PCR检测,得到了预期的560 bp目的片段,表明Ri质粒T-DNA整合到发状根基因组中.将筛选出的株系SL-7接种在MS培养基上分别置于光、暗条件下进行培养.结果发现:SL-7在暗培养条件下呈乳白色,具有多分枝、多根毛、无向地性等典型的发状根特性;在光培养条件下,发根呈粉红色,少分支且生长缓慢;以商陆抗病毒蛋白(pokeweed antiviral protein,PAP)cDNA片段为探针,分别对光、暗条件下的发状根进行Northern blot检测,发现光对PAP基因的转录具有一定抑制作用;将发状根粗蛋白提取液与TMV病毒液混合后,摩擦接种于心叶烟(Nicotiana glutinosa)离体叶片,发现暗培养的发状根粗蛋白提取液对TMV抗性明显提高.表明商陆发状根的生长及PAP基因的表达都受到光的负向调控.该结果为商陆发状根的规模化培养和PAP蛋白的离体合成优化体系的建立奠定了基础.

Structure of Poly (dT)·Poly (dA)·Poly (dT)

Abstract

The molecular structure of poly (dT)·poly (dA)·poly (dT) has been determined and refined using the continuous x-ray intensity data on layer lines in the diffraction pattern obtained from an oriented fiber of the DNA. The final R-value for the preferred structure is 0.29 significantly lower than that for plausible alternatives. The molecule forms a 12-fold right- handed triple-helix of pitch 38.4 Å and each base triplet is stabilized by a set of four Crick-Watson-Hoogsteen hydrogen bonds. The deoxyribose rings in all the three strands have C2′-endo conformations. The grooveless cylindrical shape of the triple-helix is consistent with the lack of lateral organization in the fiber.

Structures for the polynucleotide complexes poly(dA) · poly(dT) and poly(dT) · poly(dA) · poly(dT)

X-ray diffraction analyses of fibers of polydeoxyadenylic acid · polydeoxythymidylic acid show that this molecule exists as a 10-fold double-helix with axial rise per nucleotide $\text{h = 3.24}\text{to}\text{3.29}\text{A}\text{?}$. The structure is very similar to B-DNA ($\text{h = 3.37}\text{A}\text{?}$) in having C3-exo furanose rings and base-pairs positioned centrally on the helix axis, but distinctive enough to have two packing modes, neither of which has been observed for B-DNA. Although the triple-stranded poly(dT) · poly(dA) · poly(dT) also has a large value of h(3.26 Å), each of the chains is a 12-fold helix of the A-genus with C3-endo furanose rings and bases displaced several Angstrom units from the helix axis.
