Effect of the Bacterial Phytotoxin Coronatine on Plant Cell Cultures The influence of the bacterial phytotoxin coronatine on cell cultures of Lycopersicon peruvianum and Chenopodium album was studied. It was detectable that the cell culture L. peruvianum more related to the host plant of the phytopathogenic bacteria reacts more sensitive to coronatine. Concentrations of 3.5 ng coronatme/ml culture liquid mduce reductions in growth and the TTC-activity. In the case of the other used cell culture the critical amount of coronatine was higher than 7 ng/ml culture liquid. In further experiments the concentrations of free amino acids and proteins within the cell were followed. After 6–7 days of culture a slightly recovery of the cultures treated with coronatine was detectable.  相似文献

The continuous replication of potato spindle tuber viroid (PSTV) in callus cultures from PSTV-infected wild-type potato (Solanum dem/ssum L.) and tomato (Lycopersicon peruvianum L. Mill) plants and in cell suspensions derived from potato protoplasts (Solanum tuberosum L.) inoculatedin vitro is described. The persistence of PSTV replication in these cell lines through at least 14 subculture passages, which corresponds to a continous replication over a period of more than one year, was demonstrated by infectivity assay and by polyacrylamide-gel electrophoresis of isolated nucleic acids. This continuous synthesis denovo of PSTV was substantiated by the incorporation of [3H]uridine and of [32P]orthophosphate into viroid RNA.  相似文献

Tubers from somatic hybrids produced by protoplast fusion between Solanum brevidens, a diploid, non-tuber-bearing wild species, and a tetraploid S. tuberosum showed resistance to decay caused by soft rot Erwinia. Tubers of the S. tuberosum fusion parent and potato cultivar Russet Burbank are susceptible to bacterial soft rot. Tubers of somatic hybrids indicated higher levels of activities of polyphenol oxidase and peroxidase than tubers of the parental line of S. tuberosum and cultivar Russet Burbank. This is true for intact tubers and also for injured or inoculated tubers. Polish commercial potato cultivars indicated a higher susceptibility to soft rot than somatic hybrids. However, there were some differences in susceptibility to soft rot between Polish commercial potato cultivars, only slight differences were observed in the activities of the polyphenol oxidase and peroxidase between Polish cultivars. A relation between soft rot resistance and the activity of each enzyme was not found for intact, injured or inoculated tissue of commercial cultivars. On the contrary, the activities of both enzymes were significantly higher in the periderm than in the medullary tissue of somatic hybrids, the parental line and the commercial cultivars.  相似文献

Parasitism ofLygus lineolaris (Palisot de Beauvois) eggs by three species ofMymaridae, Anaphes iole Girault,Erythmelus miridiphagus Dozier andPolynema pratensiphagum Walley and one species ofScelionidae, Telenomus sp., was investigated at Ste-Anne-de-Bellevue, Québec. The maximum level of field parasitism ofL. lineolaris eggs by individual species was 15.4, 53.8, 70.0 and 16,7%, respectively. Parasitoids were retrieved from eggs ofL. lineolaris inserted in stems and branches ofAmaranthus retroflexus L.,Chenopodium album L.,Coronilla varia L.,Rumex obtusifolius L. andSolanum tuberosum L. Amaranthus retroflexus andS. tuberosum sustained a large population ofL. lineolaris and egg parasitism was high enough to consider it as a potential control factor.   相似文献

Cells of Chenopodium album and whole plants of the duckweed Wolffia arrhiza were cocultivated. In the presence of Wolffia arrhiza the synthesis of a red-violet pigment (betacyanin) was induced in several cells or cell clusters of Chenopodium album in the light. The exchange of solutes through the liquid phase was necessary for the induction of pigment formation. The red-violet cells could be selected and subcultivated resulting in a red callus. Pseudomonas solanacearum biovar 3 was identified as the causal agent of two disease syndromes in custard apples (Annona spp.), namely sudden death of young trees and the decline of mature trees. Cross inoculation tests using cultures of P. solanacearum from custard apple and tomato on A. squamosa, A. squamosa×A. cherimola hybrids, Capsicum annuum, three cultivars of Lycopersicon esculentum and Solanum tuberosum failed to distinguish differences in virulence among these isolates. Bacterial wilt resistant rootstock clones, particularly of A. cherimola, constitute the most effective control measure available.  相似文献

Summary Binding of fluorescein-conjugated wheat-germ agglutinin (F-WGA) and some other lectins to tissues from various plants were examined by epifluorescence microscopy. F-WGA bound specifically to the walls of tracheary elements (TEs) and phloem cells of pea roots. The binding sites in TEs were localized only in the secondary thickening and became evident at very early stages of differentiation. Fluorescein-conjugated derivatives ofSolanum tuberosum lectin,Lycopersicon esculentum lectin, andDatura stramonium lectin, which bind N-acetylglucosamine residues as WGA, also bound to the secondary thickening of TEs of pea roots. The binding sites for F-WGA were not removed by extraction with hot EDTA and proteinase K, but removed by extraction with an alkali solution. The alkali-extracted binding sites from the roots were precipitated together with hemicelluloses by 80% ethanol. These results indicate that the binding sites are not present on pectins, proteins, or cellulose, but hemicelluloses. Triacontanol [CH3(CH2)28CH2OH] increased growth in vitro of cell cultures of haploid tobacco (Nicotiana tabacum). The fresh weight of cell cultures of tomato (Lycopersicon esculentum), potato (Solanum tuberosum), bean (Phaseolus vulgaris), and barley (Hordeum vulgare x H. jubatum) was also increased. The increase in growth of tobacco callus seems to have been due to an increase in cell number. Another long chain alcohol, octocosanol [CH3(CH2)26CH2OH], did not increase the growth of tobacco cell cultures.  相似文献

Changes in the limiting porosity of cell walls, i.e. the size limit for permeation of neutral molecules through the wall, were studied in several higher-plant cell-suspension cultures. For this purpose, samples of biomass fixed at different cultivation times were investigated using a method based on size-exclusion chromatography of polydisperse dextrans before and after equilibration with the extracted cell clusters. In suspension cultures of Chenopodium album L., Dioscorea deltoidea Wall. and Medicago sativa L., the mean size limit (MSL; critical Stokes' radius for exclusion of neutral polymers from half of the intracellular space) was found to vary between 2.4 and 3.8 nm. It decreased significantly during transition from the growth phase to the stationary phase. In the case of the C. album culture this change was found to be irrespective of whether sucrose in the medium was completely depleted at the end of the growth phase or not. The MSL was kept constant for long periods of the stationary phase if cell viability was maintained by repeated sucrose supplement. In a suspension strain of Triticum aestivum L., the MSL of cell wall permeation was comparatively small (1.75 nm) and remained constant during all cultivation phases. Relations between limiting porosity and cell wall growth, loss of pectic compounds to the medium, cross-linking activities and cell wall stiffening are discussed. Potato virus S causes destructive disease on the plants. In this research, 44 weed samples symptomless were collected during 2015 from Fars, Razavi Khorasan and Kerman provinces of Iran. The coat protein (CP) and 11 K genes from eight PVS isolates were amplified, cloned and sequenced. PVS was detected in eight weed samples including: one sample of Solanum nigrum, two samples of Chenopodium botrytis, three samples of Chenopodium album and two samples of Amaranthus hybridus. Phylogenetic analysis showed that seven Iranian isolates fell into group I, II near to European isolates and one Iranian isolate formed a separate group. Comparison of coat protein and 11 k nucleotide indicated that all Iranian isolates belonged to Ordinary strain and there were 79–100% identity among the eight Iranian isolates and the world isolates of PVS. The highest identity was between Iranian and Ukraine isolates. Recombination analysis identified four recombinant isolates among eight new Iranian isolates.  相似文献

A binary competition experiment between carrot (Daucus carota L.) and Chenopodium album L. was conducted in a greenhouse at seven population densities and eight nutrient concentrations to investigate the effects of a nutrient gradient on plant competition in both monocultures and mixtures. The patterns of carrot biomass allocation (measured as root : shoot ratio) in monocultures and mixtures with C. album were affected by both nutrient availability and population density. Chenopodium album had a broader response to nutrient concentrations than carrot. The maximum yield of carrot in both monocultures and mixtures occurred at fourfold the standard concentration of nutrients, while C. album in both monocultures and mixtures had the maximum yield at 16-fold the standard nutrient concentration. The yield–density relationship of carrot tended to be increasing or asymptotic at lower nutrient concentrations but parabolic at higher concentrations, whereas that of C. album was little affected by nutrient availability. Nutrient availability had a profound influence on the competitive relationships between the two species: at both low and high nutrient concentrations, C. album tended to be more competitive than carrot, while at intermediate levels of nutrients, carrot was more competitive than C. album. Our results suggest that in relation to competitive performance, the weed has a greater ability to adapt itself to varying environments than does the crop. Additionally, the relative merits of the quantitative measures of competitive ability are briefly discussed.  相似文献

HSP68 — a DnaK-like heat-stress protein of plant mitochondria   总被引:1,自引:0,他引:1  
A 68-kDa heat-stress protein (HSP68) has been purified from cell-suspension cultures of tomato (Lycopersicon peruvianum L.). Antibodies raised against HSP68 cross-react with the Escherichia coli heat-stress protein DnaK. HSP68 was found to be a hydrophilic, ATP-binding protein. Immunological analysis of subcellular fractions and immunogold-labelling of ultrathin sections showed consistently that HSP68 is localized in the mitochondrial matrix. In-vitro translation experiments indicated that HSP68 is synthesized as a precursor protein. Immunoscreening of cDNA libraries from tomato and potato (Solanum tuberosum L.) led to the isolation of corresponding cDNA clones. The deduced amino-acid sequences show strong relationships to the DnaK-like proteins from bacteria and organelles of eukaryotic cells. Large protein crystals were located in the leaf and stem trichomesof Solanum tuberosum L. and Lycopersicon esculentum Mill. Inpotato the crystals ranged from 1.05 to 4.5 µm (average2.3 µm) on a side and in tomato they ranged from 1.16to 3.5 µm (average 2.7 µm) on a side. The proteinnature of the crystals was determined by histochemical stainingwith Coumassie brilliant blue R250 and aniline blue black. Thecrystalline structure of the inclusions was observed in ultrathinsections using electron microscopy. In potato, in cleared areasof the cytoplasm, ribosomes were observed scattered among proteinfilaments. The filaments were approximately 7 nm in diameter.Morphologically similar crystals were observed in the tomatotrichomes but the protein filaments were smaller (approximately4 nm in diameter). Protein crystals were also observed in palisadeand spongy parenchyma and epidermal leaf cells in tomato. Summary Culture-derived plants and cell cultures of potato (Solanum tuberosum L.) respond to the application of the herbicides SYS 67 ME (MCPA) and OMNIDEL (Na-2,2-dichloropropionate) in a comparable fashion. By gradually increasing the herbicide concentration, cell lines were developed which tolerated 50 mg/l of ME or 300 mg/l of OMNIDEL. Any further increase in concentration resulted in the death of all cell cultures. From cell cultures that had been able to grow on media supplemented with 30 mg/l of ME, regenerate plants were obtained that were also tolerant to this concentration. This new trait was retained even after repeated vegetative propagation of the plants.  相似文献

This study investigated the induced defence responses and protective effects on susceptible tomato (Lycopersicon esculentum Mill.) against Xanthomonas vesicatoria (Doidge) by a heat-treated aqueous extract (VLA) from dry necrotic tissue of ‘Lobeira’ (Solanum lycocarpum St. Hil.) branches infected with the fungus Crinipellis perniciosa (Stahel) compared with acibenzolar-S-methyl (ASM), a commercial inducer of resistance. Plantlets were sprayed with VLA and ASM and challenged 4 days later with a virulent strain of X. vesicatoria, under greenhouse conditions. The disease severity, fresh weight of shoots, the activities of phenol peroxidase (POX), polyphenol oxidase (PPO), chitinase (CHI), phenylalanine ammonia-lyase (PAL), lignin deposition, and soluble phenolic contents were evaluated in the leaf tissues. Reduction of the bacterial spot severity was observed in plantlets treated with VLA which conferred 63% of the ASM protection. This protective effect and lesion reduction promoted by VLA were probably associated particularly with POX and PAL activities, lignin deposition on leaf tissues and, to a less extent, CHI activity.  相似文献

In this study, the production of extracellular enzymes by fungi associated with southern pine beetle was investigated for the first time. Cellulase and polyphenol oxidase production were analyzed for three beetle associated fungi. Only the mutualistic symbiont Entomocorticium sp. A was found to produce cellulases and polyphenol oxidase. In time course analyses of cellulase production in batch cultures, Entomocorticium sp. A showed maximum activity of 0.109 U/ml and 0.141 U/ml for total cellulase and endoglucanase activity respectively. Polyphenol oxidase production was simultaneous with fungal growth. Characterization of polyphenol oxidase by activity staining suggests that the enzyme is a tyrosinase/catechol oxidase. Enzyme assays in the presence of polyphenol oxidase inhibitors support the results of the activity staining.  相似文献

 Somatic hybrids between three dihaploid Solanum tuberosum (2n=2x=24) genotypes and the frost-tolerant, diploid, relative Solanum commersonii (2n=2x=24) were analysed for variation in morphological traits, fertility and frost tolerance. The somatic hybrids were more vigorous than their parents and in many ways resembled a tetraploid S. tuberosum. All of the hybrids flowered profusely, although the male fertility was largely dependent on the S. tuberosum genotype used. In one hybrid combination all plants were both male- and female-fertile, while in the other two combinations the majority of plants were male-sterile but female-fertile. The somatic hybrids showed an increase in direct frost tolerance when compared with the dihaploid S. tuberosum parents, and to a varying extent had gained the capacity to cold acclimate. Linum album accumulates anti-tumor podophyllotoxin (PTOX) and its related lignans, which were originally isolated from an endangered species Podophyllum. In the present study, we examined the effects of five fungal extracts on the production of lignans in L. album cell cultures. Fusarium graminearum extract induced the highest increase of PTOX [143 μg g−1 dry weight (DW) of the L. album cell culture], while Rhizopus stolonifer extract enhanced the accumulation of lariciresinol up to 364 μg g−1 DW, instead of PTOX. Typical elicitors, such as chitin, chitosan, or methyl jasmonate (MeJA), were shown to be less effective in lignan production in L. album cell cultures. These results verified the advantages of fungal extracts to increase lignan production in L. album cell culture, and suggested potential on-demand metabolic engineering of lignan biosynthesis using differential fungal extracts.  相似文献

Cell cultures of Solanum chacoense (monohaploid) and Solanum tuberosum (tetraploid cultivars and parthenogenetically derived dihaploid clones) were found to be highly mixoploid.Relative stabilization of chromosome number at the ploidy level of the original plant material was achieved in microcalli obtained from single cells or small cell colonies (up to about 5 cells) of stock callus lines. This relative stabilization was maintained over three subcultures, which is sufficient for selection procedures. It has been shown that the stabilization can be maintained during a number of further subcultures. Division centers were repeatedly observed in calli characterized by high mitotic activity. As has been shown for the first time there exist significant differences in the ploidy levels of several division centers within one and the same callus. Genotypes of Solanum tuberosum, assessed for their differential response to a protoplast regeneration protocol, were identified. This enabled comparisons to be made in relation to their abilities to regenerate into dividing cells and colonies. Protoplasts from genotypes with contrasting regeneration responses were plated as single or mixed genotype cultures at various plating densities in replicated, randomised experiments, and the effect on the regeneration of particular cell types observed. Protoplasts from single genotype cultures showed intergenotypic variation in the extent of cell regeneration and there were significant effects of genotype mixing and density × genotype mixing interactions. Overall, the effect of mixing was beneficial to a regenerating culture but significant density genotype mixing interactions showed only a positive benefit at the lowest plating density. The protoplast mixing phenomenon was not correlated with the behaviour of the same genotypes at the plantlet level in experiments with in vitro meristem cultures.  相似文献
